Summary We have analyzed 18 kb of DNA in and upstream of thedefective chorion-1 (dec-1) locus of the eight known species of themelanogaster species subgroup ofDrosophila. The restriction maps ofD. simulans, D. mauritiana, D. sechellia, D. erecta, andD. orena are shown to have basically the restriction map ofD. melanogaster, whereas the maps ofD. teissieri andD. yakuba were more difficult to align. However, the basic amount of DNA and sequence arrangement appear to have been conserved in these species. A small deletion of varying length (65–200 bp) is found in a repeated sequence of the central transcribed region ofD. melanogaster, D. simulans, andD. erecta. Restriction site mapping indicated that thedec-1 gene is highly conserved in themelanogaster species subgroup. However, sequence comparison revealed that the amount of nucleotide and amino acid substitution in the repeated region is much larger than in the 5 translated region. The 5 flanking region showed noticeable restriction site polymorphisms between species. Based on calculations from the restriction maps a dendrogram was derived that supports earlier published phylogenetic relationships within themelanogaster species subgroup except that theerecta-orena pair is placed closer to themelanogaster complex than toD. teissieri andD. yakuba.  相似文献

Summary Globin mRNAs ofXenopus borealis andXenopus tropicalis have been cloned and sequenced. The nucleotide and derived amino acid sequences were compared with each other and with already available data fromXenopus laevis. This analysis rendered clear evidence that the common ancestor ofX. laevis andX. borealis, but not ofX. tropicalis, had lost one amino acid of the -globins prior to a genome duplication event that preceded the segregation of the former two species. Replacement-site substitutions were used to calculate a rough time scale of genome duplication and species segregation. The results suggest an ancient separation between theX. laevis and theX. tropicalis groups occurring approximately 110–120 million years ago. Analysis of the amino acid chains demonstrated various alterations. However, some functional domains, like heme-binding sites and12 contact sites, were subject to a high degree of conservation, indicating the existence of functional constraints on them also in the genusXenopus.  相似文献

Parsimony and maximum likelihood analyses of combinedtrnL (UAA) 5 exon —trnF (GAA) andrps4 exon cpDNA, and 18S nrDNA sequences of 60 arthrodontous moss taxa indicate strong support for the monophyly of a clade containing theSplachnineae, Orthotrichineae, and diplolepideous alternate sub-orders. A clade including theSplachnineae, Meesiaceae andLeptobryum (Bryaceae) is similarly well supported and forms the sister group to a clade comprising theOrthotrichineae and the other diplolepideous alternate mosses. Within this latter clade a number of well supported lineages are identified, but relationships among these remain poorly resolved. These analyses indicate that the Splachnaceous and Orthotrichaceous peristomes have been independently derived from an ancestral perfect bryoid peristome.  相似文献

The genusHolarrhena, described byRobert Brown in 1811, has had a problematic taxonomic history, in part due to a suite of characters that does not conform with accepted concepts within theApocynaceae. In a number of important taxonomic charactersHolarrhena is typical of subfam.Apocynoideae. But due to the relatively unspecialized structure of the anthers most recent authors have placedHolarrheng, together withCarruthersia andSpirolobium, as the subtribeHolarrheninae in subfam.Plumerioideae. For the present investigation the floral structure and pollen morphology ofHolarrhena, Carruthersia andSpirolobium were analyzed. From the chemical literature reports of the occurrence of steroidal alkaloids in thePlumerioideae were evaluated. Our results indicate that the three genera belong to subfam.Apocynoideae in the tribeNerieae, but that the Holarrheninae is an unnatural group, and that the three genera should be accommodated individually within the tribe.  相似文献

The -esterase cluster of D. melanogaster comprises two tandemly duplicated genes. Est6 encodes the well-characterized 5 gene, but the product of the second gene, denoted EstP, had not previously been identified. Here we show that the EstP gene encodes the carboxylesterase EST7. Expression of EstP using the Baculovirus system led to production of a carboxylesterase biochemically indistinguishable from EST7. Furthermore, a naturally occurring EstP variant produces greatly reduced amounts of EstP mRNA and no detectable EST7 protein. Finally, introduction of a wild-type copy of EstP by germline transformation into the variant strain confers the wild-type EST7 phenotype. We show that EST7 differs from EST6 in its substrate and inhibitor specificities and tissue distribution. Germline transformation experiments show that EstP expression is controlled by sequences located between 192 bp 5 and 609 bp 3 of the EstP coding region. Data comparisons with other drosophilid esterases suggest that the site of expression, and hence the function, of EST7 has been conserved across lineages in both the subgenera Drosophila and Sophophora.  相似文献

Summary Anthers ofBetula pendula were collected at regular intervals during the dormancy period until anthesis. Ultrathin sections of maturing pollen grains were especially stained for polysaccharides and proteins and examined with TEM to determine whether structural or/and chemical changes in the pollen wall occur during the dormancy period of the plant life cycle. At the beginning of the dormancy period, the microspore wall consists of a well developed tectum, columellae and a foot layer. Spinules and supratectal elements are prominent. Microchannels are present in the tectum but not obvious in the foot layer. Some of the columellae are not clearly connected with the foot layer, but some connections are evident. Pores are filled with a thick fibrillar network flocculent material. The cytoplasm is packed with starch grains and lipid globules. The stainability for acidic and neutral polysaccharides and protein was tested, and variations in the pollen wall are illustrated. As temperature increased towards the end of dormancy and before anthesis there is obvious differentiation in the morphology of the pollen wall. The granular fibrillar layer beneath the pore and the Zwischenkörper are the most variable part of the wall. Different histochemical reactions observed in different layers at the aperture sites indicate different functions of these layers.  相似文献

A 647-bp 5-flanking fragment obtained from genomic clone Sta 44G(2) belonging to a family of polygalacturonase genes expressed inBrassica napus pollen was fused to the-glucuronidase (GUS) marker gene. This fusion construct was introduced intoB. napus plants viaAgrobacterium tumefaciens transformation. Analysis of the transgenicB. napus plants revealed that this promoter fragment is sufficient to direct GUS expression specifically in the anther and that GUS activity increases in pollen during maturation.Abbreviation GUS -Glucuronidase  相似文献

Many members ofRanunculaceae contain unusual fatty acids in their seed oils. This leads to rather typical genus-specific fatty acid patterns or fingerprints in these seed oils. The members of theDelphinioideae and/orHelleboroideae, however, do not contain highly unusual fatty acids. Nevertheless, their seed oil fatty acid fingerprints are also fairly typical and genus-specific, and the patterns found are rather consistent throughout several species of one genus. It was found that species ofAconitum do not contain fatty acids with 20 carbon atoms.Delphinium, Consolida, Helleborus, Nigella and others do contain C20 fatty acids. In allHelleborus species, for example, there was a consistent C20 fatty acid pattern of 20:020:120:2>20:3. Species ofNigella andGaridella contain high levels,Helleborus low levels, of 20:2n-6 in their seed oils.Delphinium andAconitum both contain low levels of 18:3n-3, whereasHelleborus spp. consistently show high levels of this fatty acid. The genus-specific fatty acid patterns found are discussed, and a correlation with the subfamily and tribe affiliation of the genera investigated here is attempted.  相似文献

Summary The sequence of a segment of theDrosophila virilis mitochondrial DNA (mtDNA) molecule that contains the A+T-rich region, the small rRNA gene, the tRNAf-met, tRNAgln, and tRNAile genes, and portions of the ND2 and tRNAval genes is presented and compared with the corresponding segment of theD. yakuba mtDNA molecule. The A+T-rich regions ofD. virilis andD. yakuba contain two correspondingly located sequences of 49 and 276/274 nucleotides that appear to have been conserved during evolution. In each species the replication origin of the mtDNA molecule is calculated to lie within a region that overlaps the larger conserved sequence, and within this overlap is found a potential hairpin structure. Substitutions between the larger conserved sequences of the A+T-rich regions, the small mt-rRNA genes, and the ND2 genes are biased in favor of transversions, 71–97% of which are AT changes. There is a 13.8 times higher frequency of nucleotide differences between the 5 halves than between the 3 halves of theD. virilis andD. yakuba small mt-rRNA genes. Considerations of the effects of observed substitutions and deletion/insertions on possible nucleotide pairing within the small mt-rRNA genes ofD. virilis andD. yakuba strongly support the secondary structure model for theDrosophila small mt-rRNA that we previously proposed.  相似文献

The GAS1 -related genes of fungi encode GPI-anchored proteins with -1,3-glucanosyltransferase activity. Loss of this activity results in defects in the assembly of the cell wall. We isolated mutants that show a synthetic defect when combined with a gas1 allele in Saccharomyces cerevisiae, and identified nine wild-type genes that rescue this defect. The indispensability of BIG1 and KRE6 for the viability of gas1 cells confirmed the important role of -1,6-glucan in cells that are defective in the processing of -1,3-glucan. The identification of the Wsc1p hypo-osmotic stress sensor and components of the PKC signal transduction pathway in our screen also confirmed that the cell wall integrity response attenuates the otherwise lethal gas1 defect. Unexpectedly, we found that the KEX2 gene is also required for the viability of the gas1 mutant. Kex2p is a Golgi/endosome-membrane-anchored protease that processes secretory preproteins. A cell wall defect was also found in the kex2 mutant, which was suppressible by multiple copies of the MKC7 or YAP3 gene, both of which encode other GPI-anchored proteases. Therefore, normal cell wall assembly requires proteolytic processing of secretory preproteins. Furthermore, the genes CSG2 and IPT1 were found to be required for normal growth of gas1 cells in the presence of 1 M sorbitol. This finding suggests that complex sphingolipids play a role in the hyper-osmotic response.Communicated by C. P. Hollenberg  相似文献

In direct contrast to mostHibbertia spp., the flowers ofH. fasciculata R. Br. ex D. C. bear only a single whorl of stamens and these stamens are arranged separately (not in typical bundles). The short filaments are appressed to the three carpels so that the inflated, porose and introrsive anthers form a centralized cluster obscuring the three ovaries. The three slender styles emerge at right angles from between the filaments. These styles curve upward and the stigmas form the three points of a triangle; each stigma is approximately one millimeter outside the centralized cluster of anthers. The flowers are nectarless and bear a bright yellow corolla. A pungent and unpleasant fragrance appears to be concentrated within the pollenkitt. When native bees attempt to forage for the pollen, within the cluster of anthers, the ventrally deposited loads of pollen, on the bees' abdomens, contact the outer triangle of stigmas. The major pollinators ofH. fasciculata are female bees in the polylectic genera,Lasioglossum (subgenusChilalictus, Halictidae) andLeioproctus (Colletidae). These bees carry an average of more than two pollen taxa when they are caught foraging onH. fasciculata. 78% of the 47 bees, captured onH. fasciculata carried the pollen from at least one sympatric taxon bearing nectariferous flowers (e.g., genera in theMyrtaceae, Compositae, andEpacridaceae). The pollination biology ofH. fasciculata is assessed in relation to the known radiation of bee-pollinated flowers in the genusHibbertia, and within theDilleniaceae s. l.  相似文献

Summary The tryptic peptides from the A and B chains of cat hemoglobins A and B have been isolated and the amino acid compositions determined. Differences between the two chains were found in two peptides,T-1 (GlySer) andT-14 (AsnSer and LysArg). The GlySer and LysArg substitutions are placed at-1 and-144 respectively from earlier work, and the third substitution, AsnSer at-139 is suggested from this work. In addition, the presence of a blocked amino terminus in B has been confirmed. Tentative sequences constructed by homology with known-chain structures suggest the occurrence of substitutions at 1 1 contacts in A and B that may be functionally significant. There are at least 18 differences in amino acid composition between cat A and dog-chains and 22 differences between cat A and normal adult human-chains.  相似文献

The anther wall layers ofCypripedium cordigerum are six to eight. The glandular tapetum is 2- or 3-layered and its cells are uninucleate. Simultaneous cytokinesis results in decussate, isobilateral and tetrahedral pollen tetrads. Ripe pollen grains are 2-celled. The mature ovules are anatropous, bitegmic and tenuinucellate. Both the integuments are dermal in origin and 2-layered. The inner integument alone forms the micropyle. The female gametophyte is 6-nucleate and bisporic. The reduction of nuclei is due to the strike phenomenon. Double fertilization occurs. The primary endosperm nucleus divides to form two free endosperm nuclei. The mature embryo is undifferentiated. The cells ca, m and n contribute to the embryo. The suspensor is single-celled. The seed coat is formed entirely by the outer layer of the outer integument. There are three sterile and three fertile valves in the ovary. In the prefertilization stages these valves consist of parenchymatous cells with starch and raphides. After fertilization, the sterile valves develop sclerotic cells whereas the fertile valves remain parenchymatous. The pericarp structure and embryological features support the retention of tribeCypripedieae within theOrchidaceae.  相似文献

The four genera investigated show solitary sulcate pollen grains. The structure and sculpture of the sporoderm is very similar inBocageopsis andUnonopsis, and supports the close relationship of both genera; their flower morphology also is very similar. In contrast, the sporoderm ofEphedranthus, and especially that ofMalmea, is different in some characters and suggests more remote relationships; this is also confirmed by differences in flower morphology. Within this group of genera a distinct exine progression can be recognized from non-columellate (Malmea) to granular (Unonopsis guatterioides), further to somewhat irregularly (Bocageopsis, someUnonopsis species), and finally to very regularly columellate (Ephedranthus). The sculpture of the tectum varies from a reticulum with large lumina (Malmea) to one with small performations (Ephedranthus). Within theAnnonaceae the genusMalmea is among the most primitive in respect to pollen structure. The sulcus of the four genera is very large and runs over 1/3 of the pollen grain. It is characterized by a reduction of the exine and a bulgy thickening of the intine.

A study of the phenolic compounds of the closely related papilionoid tribes,Podalyrieae andLiparieae, proved that the flavonoid patterns of hydrolysed seed extracts are remarkably conservative. Butin (7, 3, 4-trihydroxyflavanone), 3-hydroxydaidzein (7, 3, 4-trihydroxyisoflavone), vicenin-2 (6, 8-di--D-glucopyranosyl-5, 7, 4-trihydroxyflavone) and orobol (5, 7, 3, 4-tetrahydroxyisoflavone) were isolated and identified as the major flavonoids. The seeds ofAmphithalea, Coelidium, Liparia, Xiphotheca, Calpurnia, Stirtonanthus andPodalyria accumulated three isoflavone O-glycosides that yielded 3-hydroxydaidzein on hydrolysis. In contrast,Virgilia contained a unique combination of vicenin-2 and orobol. Vicenin-2 was also present inCalpurnia as a major compound, butStirtonanthus insignis was the only other species studied that contained orobol (in trace amounts only). Butein, a chalcone, was reported byHarborne from the seed ofCyclopia subternata. This compound's flavanone analog, butin, was the principal component inCyclopia. A cladistic analysis, using flavonoid, alkaloid and morphological data, showed that the seed flavonoids of thePodalyrieae andLiparieae behave rather poorly as cladistic characters. They are, however, of considerable taxonomic value at the tribal level favouring the opinion that the two tribes should be combined. The apparent absence of flavonoids in the seed ofHypocalyptus supports the suggestion that it should be excluded from theLiparieae. Flavonoids also show that theArgyrolobium-group is very different from the tribeCrotalarieae and support the recent transfer of this group to the tribeGenisteae.  相似文献

Summary Species of the generaAshbya, Eremothecium, Holleya, andNematospora were compared from extent of divergence in a 580-nucleotide region near the 5 end of the large subunit (26S) ribosomal DNA gene. The four genera are closely related and comprise a subclade of the hemiascomycetes. Because the taxa show little divergence, it is proposed that all be placed in the genusEremothecium. The family Eremotheciaceae, fam. nov., is proposed.This paper is dedicated to Professor Herman Jan Phaff in honor of his 50 years of active research which still continues.  相似文献

All genera ofAnnonaceae endemic in Australia (Ancana, Fitzalania, Haplostichanthus) show almost exactly the same type of disulcate (disulculate) pollen with intact exine extending over the sulci. Tetrad stages inHaplostichanthus andAncana reveal a latudinal subequatiorial orientation of the two sulci at the proximal hemisphere. Sometimes they fuse into a ±zonosulcate aperture.Fissistigma pollen grains are ±globose and have a flattened pole with a central elevation and a concentric groove, covered by a somewhat reduced exine. This palynological characters give further support for separating the generaAncana andFissistigma. Germination was observed inHaplostichanthus where the pollen tube emerges at one of the two sulci and inFissistigma where the flattened part breaks up during germination. The aperture types described here are obviously transitional stages between aperturate and inaperturate pollen grains and are discussed in regard to pollen evolution.  相似文献

New investigations on the flower and fruit structure of extantHamamelidaceae and other LowerHamamelididae together with new finds of fossil flowers and seeds from the Upper and Lower Cretaceous provide the outline of an increasingly more differentiated picture of the early evolution of the subclass. Three patterns of valvate anther dehiscence are recognized in the subfamilyHamamelidoideae (and the subclassHamamelididae). The basic (plesiomorphic) type within theHamamelididae has 2 valves per theca. The type with 1 valve but 2 pollen sacs per theca is both consistent and exclusive for the 5 southern genera of theHamamelidaceae. They seem to be the remnants of a homogeneous group that originated before the Upper Cretaceous. This is supported by fossil hamamelidaceous flowers from the Upper Cretaceous that have thecae with 1 valve. Since several-seededHamamelidaceae predate one-seeded forms in the fossil seed record (in Europe) and the systematic structure of the one-seeded group is relatively more homogeneous, several-seeded groups are considered to be more ancient. Several parallel evolutionary trends are recognized within theHamamelidaceae as well as within the LowerHamamelididae: anther dehiscence with 2 valves per theca 1 slit or 1 valve; pollen sacs per theca 2 1; pollen tricolpate polyforate; exine coarsely reticulate finely reticulate; loss of perianth (tepals or petals and sepals) and concomitant loss of fixed number of floral organs; differentiation of exposed nectaries.  相似文献

This article summarizes some of the evidence for the existence of light-driven structural changes in the and subunits of the chlorplast ATP synthase. Formation of a transmembrane proton gradient results in: (1) a change in the position of the subunit such that it becomes exposed to polyclonal antibodies and to reagents which selectively modifyLys109; (2) enhanced solvent accessibility of several sulfhydryl residues on the subunit; and (3) release/ exchange of tightly bound ADP from the enzyme. These and related experimental observations can, at least partially, be explained in terms of two different bound conformational states of the subunit. Evidence for structural changes in the enzyme which are driven by light or nucleotide binding is discussed with special reference to the popular rotational model for catalysis.  相似文献

Summary A heterogeneous distribution of H+-ATPase was visualized in germinated pollen ofLilium longiflorum using monoclonal antibodies raised against plasma membrane H+-ATPase. Immunolocalization studies of protoplasts and subprotoplasts derived from pollen tubes and sectioned pollen grains and pollen tubes show that H+-ATPases are abundant in the plasma membrane of pollen grains but are absent or sparsely distributed in the plasma membrane of pollen tubes.
