Abstract Growing roots of Nicotiana tabacum var. Havana generate transcellular ion currents which traverse developing and wounded tissues. Positive current of around 10 mA m?2 enters meristematic and elongating cells at the tip of primary roots. The growing tips of first order laterals are also traversed by a similar positive current with a density of around 2.0 mA m?2, as are immature laterals emerging at the primary root surface. These self-generated ion currents flow basipetally through developing tissues and leave from mature non-elongating tissue. A large positive current of around 70 mA m?2 also enters induced wound sites on the primary root surface. Motile zoospores of the fungal pathogen Phytophthora parasitica var. nicotianae have been reported to associate preferentially with these regions of the root. This might suggest that electrotaxis may be part of the mechanism by which zoospores locate root regions susceptable to fungal infection.  相似文献

A novel, three-dimensional recording, vibrating probe was used for measuring the density and direction of the endogenous ionic current of cress roots (Lepidium sativum L.) bathed in low salt media (artificial pond water, APW). Roots submerged in regular APW and growing vertically show the following current pattern. Current of 0.7 microampere/square centimeter density enters or leaves the root cap; the current changes direction frequently. Current of 1.6 microamperes/square centimeter enters the meristem zone most of the time. Maximum current with a density of 2.2 microamperes/square centimeter enters the apical elongating zone, i.e. between 0.8 and 1.2 millimeters behind the root tip. The current density decreases to 1.4 microamperes/square centimeter at 2 millimeters, i.e. in the central elongating zone, and to 1.0 microampere/square centimeter at 3 millimeters, i.e. in the basal elongating zone. The current direction changes from inward to predominantly outward between 1.2 and 3 millimeters behind the tip. Measurements on opposite flanks of the roots indicate that the current pattern is fairly symmetrical. After placing the roots horizontally, the density of the endogenous current remains stable, but the current direction changes at the root cap and in the meristem zone. The current leaves the root on the upper side and enters on the lower side, causing a highly asymmetrical current pattern at the very tip. The current pattern at the upper and lower side further away from the tip remains the same as in vertical roots. Roots submerged in low Ca2+ APW show a very different current pattern, no gravitropism, and no change of the current pattern after horizontal orientation. In these roots current enters the root cap and the basal elongating zone and leaves the apical elongating zone. Three conclusions are drawn from these results: First, plant roots elongate by two different modes of growth that are correlated with different current directions. They grow by cytoplasmic enlargement at sites of inward current and by turgor-driven elongation at sites of outward current. Second, a change in the current pattern at the root cap and in the meristem zone is a clear indicator of later gravitropism. Third, Ca2+ ions are involved in the gravistimulated change in the current pattern, probably affecting the activity of plasmalemma H+-ATPases.  相似文献

The water relations and hydraulic architecture of growing grass tillers (Festuca arundinacea Schreb.) are reported. Evaporative flux density, E (mmol s?1 m?2), of individual leaf blades was measured gravimetrically by covering or excision of entire leaf blades. Values of E were similar for mature and elongating leaf blades, averaging 2·4 mmol s?1 m?2. Measured axial hydraulic conductivity, Kh (mmol s?1 mm MPa?1), of excised leaf segments was three times lower than theoretical hydraulic conductivity (Kt) calculated using the Poiseuille equation and measurements of vessel number and diameter. Kt was corrected (Kt*) to account for the discrepancy between Kh and Kt and for immature xylem in the basal expanding region of elongating leaves. From base to tip of mature leaves the pattern of Kt* was bell‐shaped with a maximum near the sheath–blade joint (≈ 19 mmol s?1 mm MPa?1). In elongating leaves, immature xylem in the basal growing region led to a much lower Kt*. As the first metaxylem matured, Kt* increased by 10‐fold. The hydraulic conductances of the whole root system, (mmol s?1 MPa?1) and leaf blades, (mmol s?1 MPa?1) were measured by a vacuum induced water flow technique. and were linearly related to the leaf area downstream. Approximately 65% of the resistance to water flow within the plant resided in the leaf blade. An electric‐analogue computer model was used to calculate the leaf blade area‐specific radial hydraulic conductivity, (mmol s?1 m?2 MPa?1), using , Kt* and water flux values. values decreased with leaf age, from 21·2 mmol s?1 m?2 MPa?1 in rapidly elongating leaf to 7·2 mmol s?1 m?2 MPa?1 in mature leaf. Comparison of and values showed that ≈ 90% of the resistance to water flow within the blades resided in the liquid extra‐vascular path. The same algorithm was then used to compute the xylem and extravascular water potential drop along the liquid water path in the plant under steady state conditions. Predicted and measured water potentials matched well. The hydraulic design of the mature leaf resulted in low and quite constant xylem water potential gradient (≈ 0·3 MPa m?1) throughout the plant. Much of the water potential drop within mature leaves occurred within a tenth of millimetre in the blade, between the xylem vessels and the site of water evaporation within the mesophyll. In elongating leaves, the low Kt* in the basal growth zone dramatically increased the local xylem water potential gradient (≈ 2·0 MPa m?1) there. In the leaf elongation zone the growth‐induced water potential difference was ≈ 0·2 MPa.  相似文献

In order to parametrize a leaf submodel of a canopy level gas-exchange model, a series of photosynthesis and stomatal conductance measurements were made on leaves of white oak (Quercus alba L.) and red maple (Acer rubrum L.) in a mature deciduous forest near Oak Ridge, TN. Gas-exchange characteristics of sun leaves growing at the top of a 30 m canopy and of shade leaves growing at a depth of 3–4 m from the top of the canopy were determined. Measured rates of net photosynthesis at a leaf temperature of 30°C and saturating photosynthetic photon flux density, expressed on a leaf area basis, were significantly lower (P = 0.01; n = 8) in shade leaves (7.9μmol m?2 s?1) than in sun leaves (11–5μmol m?2 s?1). Specific leaf area increased significantly with depth in the canopy, and when photosynthesis rates were expressed on a dry mass basis, they were not significantly different for shade and sun leaves. The percentage leaf nitrogen did not vary significantly with height in the canopy; thus, rates expressed on a per unit nitrogen basis were also not significantly different in shade and sun leaves. A widely used model integrating photosynthesis and stomatal conductance was parametrized independently for sun and shade leaves, enabling us to model successfully diurnal variations in photosynthesis and evapotranspiration of both classes of leaves. Key photosynthesis model parameters were found to scale with leaf nitrogen levels. The leaf model parametrizations were then incorporated into a canopy-scale gas-exchange model that is discussed and tested in a companion paper (Baldocchi & Harley 1995, Plant, Cell and Environment 18, 1157–1173).  相似文献

Abstract. Seedlings of Zea mays L. were grown in the dark at 27°C. Four-day-old seedlings were then exposed for 3 days to solutions equilibrated with gas mixtures to give O2 concentrations between 0.02 and 0.25 mol m?3. Root growth was impaired just as severely at 0.06 as 0.02 mol O2 m?3 while growth at 0.16 mol O2 m?3 was about the same as in solutions in equilibrium with air (0.25 mol O2 m?3). Growth of young seedlings at low O2 concentrations was inhibited to the same extent in nutrient solution and 0.5 ml m?3 CaCl2, showing that the adverse effect of O2 deficits on growth was not due to less uptake of inorganic nutrients. Furthermore, at low O2 concentrations neither exposure of the shoots to a relative humidity of 100% (26.0 g H2O m?3) nor excision of the entire shoot enhanced root growth relative to that in plants with shoots at a relative humidity of 50% (13.0 g H2O m?3). Therefore, for these seedlings growing in the dark, impairment of root growth at low O2 concentrations was not a consequence of water deficits in the shoot or of other shoot-root interactions. Total soluble sugars and amino acid concentrations were generally greater at low (0.02–0.06 mol O2m?3) than at high O2 concentrations (0.16–0.25 mol O2 m ?3). This applied specifically to the root apices (0–2 mm) and expanding (2–15 mm) tissue except in some experiments where sugar concentrations in expanding tissue were slightly greater at high than at low O2 concentrations. Critical O2 pressures for respiration of excised root segments were approximately 0.117 and 0.065 mol O2 m?3 in the expanding and expanded zones of the roots, respectively. In contrast, the critical O2 pressure exceeded 0.20 mol O2 m?3 in the apex, suggesting that O2 supply for metabolic processes is most likely to be sub-optimal in this zone. Our results show clearly that the adverse effects of low O2 concentrations are unlikely to be a consequence of substrate shortage for either respiration or synthesis of macromolecules; low rates of ATP regeneration in growing root tissues are the logical cause for impaired growth in young seedlings while they are being sustained by seed reserves.  相似文献

Growth measurements of hormone-treated roots from willow cuttings were combined with electrophysiological recordings to study hormone-induced changes in membrane potential and in endogenous ion currents. The mean growth rate of roots was 10 ± 2 μm min?1 in regular nutrient solution. It increased to 13 ± 2 μm min+1 after application of spermine and decreased to 0.07 ± 0.01 μm min?1 after treatment with abscisic acid (ABA). Transient depolarizations were elicited in root cortex cells by spermine, while ABA caused a transient hyperpolarization. All changes in membrane potential were accompanied by transient responses of the endogenous current. These responses suggest that first anions, then cations leave the root during spermine-induced depolarizations. From the changes of the endogenous current an apparent efflux of anions (presumably Cl?) and cations (presumably K+) of 200 to 700 pmol cm?2 per depolarization was calculated. To further investigate a possible relation between endogenous ion currents, growth and the growth regulators ABA and spermine, long-lasting extracellular vibrating-probe measurements were performed. Control roots showed an inward current of about 1.5 μA cm?2 at the apical elongation zone and an outward current with a maximum density of 1.3 μA cm?2 at the central and basal elongation zone. The addition of ABA and spermine (final concentration 0.1 mM) to the bathing medium affected the endogenous current in opposite ways: ABA caused a reduction of inward and outward current, while spermine stimulated both. Since protons are a major component of the endogenous current, and sucrose can be taken up by root cells from the apoplast via symport with H+, a role of the endogenous current in growth regulation is indicated.  相似文献

Abstract Atriplex amnicola, was grown in nutrient solution cultures with concentrations of NaCl up to 750 mol m?3. The growth optimum was at 25–50 mol m?3 NaCl and growth was 10–15% of that value at 750 mol m?3 NaCl. Sodium chloride at 200 mol m?3 and higher reduced the rate of leaf extension and increased the time taken for a leaf to reach its maximal length. Concentrations of Na+, K+ and Mg2+ in leaves of different ages were investigated for plants grown at 25, 200 and 400 mol m?3 NaCl. Although leaves of plants grown at 200 and 400 mol m?3 NaCl had high Na+ concentrations at young developmental stages, much of this Na+ was located in the salt bladders. Leaves excluding bladders had low Na+ concentrations when young, but very high in Na+ when old. In contrast to Na+, K+ concentrations were similar in bladders and leaves excluding bladders. Concentrations of K+ were higher in the rapidly expanding than in the old leaves. At 400 mol m?3 NaCl, the K+:Na+ ratios of the leaves excluding bladders were 0.4–0.6 and 0.1 for rapidly expanding and oldest leaves, respectively. The Na+ content in moles per leaf, excluding bladders, increased linearly with the age of the leaves; concurrent increases in succulence were closely correlated with the Na + concentration in the leaves excluding the bladders. Soluble sugars and starch in leaves, stems and buds were determined at dusk and dawn. There was a pronounced diurnal fluctation in concentrations of carbohydrates. During the night, most plant parts showed large decreases in starch and sugar. Concentrations of carbohydrates in most plant organs were similar for plants grown at 25 and 400 mol m?3 NaCl. One notable exception was buds at dusk, where sugar and starch concentrations were 30–35% less in plants grown at 400 mol m?3 NaCl than in plants grown at 25 mol m?3 NaCl. The data indicate that the growth of A. amnicola at 400 mol m?3 NaCl is not limited by the availability of photosynthate in the plant as a whole. However, there could have been a growth limitation due to inadequate organic solutes for osmotic regulation.  相似文献

In growing leaves, lack of isoprene synthase (IspS) is considered responsible for delayed isoprene emission, but competition for dimethylallyl diphosphate (DMADP), the substrate for both isoprene synthesis and prenyltransferase reactions in photosynthetic pigment and phytohormone synthesis, can also play a role. We used a kinetic approach based on post‐illumination isoprene decay and modelling DMADP consumption to estimate in vivo kinetic characteristics of IspS and prenyltransferase reactions, and to determine the share of DMADP use by different processes through leaf development in Populus tremula. Pigment synthesis rate was also estimated from pigment accumulation data and distribution of DMADP use from isoprene emission changes due to alendronate, a selective inhibitor of prenyltransferases. Development of photosynthetic activity and pigment synthesis occurred with the greatest rate in 1‐ to 5‐day‐old leaves when isoprene emission was absent. Isoprene emission commenced on days 5 and 6 and increased simultaneously with slowing down of pigment synthesis. In vivo Michaelis–Menten constant (Km) values obtained were 265 nmol m?2 (20 μm ) for DMADP‐consuming prenyltransferase reactions and 2560 nmol m?2 (190 μm ) for IspS. Thus, despite decelerating pigment synthesis reactions in maturing leaves, isoprene emission in young leaves was limited by both IspS activity and competition for DMADP by prenyltransferase reactions.  相似文献

Wildfire is a major disturbance in Baikiaea plurijuga Harms woodland savannas. We tested the hypothesis that the timing and intensity of herbivory influence fuel loads. We used three stocking rates namely light (three cows and four goats ha?1), medium (six cows and eight goats ha?1) and heavy (eleven cows and sixteen goats ha?1) and three times of grazing namely early‐, middle‐ and late‐growing seasons. Season of grazing and stocking rate influenced herbaceous phytomass. Phytomass was generally the highest (53.5 g DM m?2) in paddocks grazed during the early growing season and the lowest (27.8 g DM m?2) in those grazed during the late growing season. Phytomass was also generally the highest (40.4 g DM m?2) in lightly stocked paddocks and the lowest (32.7 g DM m?2) in heavily stocked ones. Litter mass was the lowest (160.8 g DM m?2) in paddocks grazed during the early season whereas there were no differences in ungrazed paddocks and those grazed during either mid‐ or late growing seasons (205.4 g DM m?2). There was a negative relationship between litter mass and stocking rate. Baikiaea Benth. woodlands should be grazed during either the mid‐ or late‐growing season at stocking rates greater than 0.1 LU ha?1 to reduce grass fuel loads.  相似文献

Abstract Atriplex amnicola was grown at 25, 200 or 400 mol m3 NaCl. Root tissues at different stages of development were investigated for concentrations of K+, Na+ and Mg2+, and in some cases for Cl?. Sugar and starch concentrations were measured for plants grown at 25 or 400 mol m3 NaCl. In the ‘slightly vaeuolated’ root tips, Na+ was only 40 mol m?3 at an external concentration of 400 mol m?3 NaCl. The concentrations of K+ were not affected substantially by external NaCl between 25 mol m?3 and 400 mol m?3. The ‘highly vacuolated’ root tissues had substantially higher concentrations of K+, Na+ and Cl? in plants grown at 200 and 400 mol m 3 NaCl than in plants grown at 25 mol m?3 NaCl. Concentrations of Cr and of the sum of the cations in recently expanded tissue were similar to those in the bulk of the roots, consisting mainly of old cells. However, the K+: Na+ decreased with age; at 400 mol m?3 external NaCl with a K+: Na+ of 0.012, the K+: Na+ in recently expanded 12 mm root tips was as high as 1.6, compared with 0.7 for the bulk of the roots. These ion data were used to estimate cytoplasmic and vacuolar concentrations of K+ and Na +. Such calculations indicated that between 25 mol m3 and 400 mol m?3 external NaCl the concentration of the sum of (Na++K+) in the cytoplasm was maintained at about 180–200 mol m?3 (cell water basis). In contrast, the (Na++ K+) concentration in the vacuole was 170 mol m?3 for plants grown at 25 mol m?3 NaCl and 420 mol 400 mol m?3 NaCl. The expanding root (issues exhibited greatly decreased soluble sugars and starch between dusk and dawn. Ai both times, sugar and starch concentrations in these tissues were 2.5–4.0 times greater in plants grown at 400 mol m?3 NaCl compared with plants grown at 25 mol m?3 NaCl. In contrast, carbohydrate concentrations in expanded root tissues were very similar at 25 and 400 mol m?3 and showed little diurnal fluctuation. This paper considers the causes for the slower growth of A. amnicola at 400 than at 25 mol m”3 NaCl, using the data for the roots described here, and those for the shoots presented in the preceding paper (Aslam et al., 1986). There is no support for possible adverse effects by high internal ion concentrations. Instead, there may be deficiencies in supply of organic solutes for osmotic regulation; during part of the night a limited supply of such solutes may well restrict the rate of expansion of cells in plants growing at 400 mol m?3 NaCl. There is insufficient evidence to decide whether this limitation in the expanding tissues is particularly prominent for the roots or for the shoots.  相似文献

With the aid of an extracellular vibrating electrode, natural electric fields were detected and measured in the medium near growing roots and root hairs of barley seedlings. An exploration of these fields indicates that both the root as a whole, as well as individual root hairs, drive large steady currents through themselves. Current consistently enters both the main elongation zone of the root as well as the growing tips of elongating root hairs; it leaves the surface of the root beneath the root hairs. These currents enter with a density of about 2 microamperes per square centimeter, leave with a density of about 0.5 to 1 microampere per square centimeter, and total about 30 nanoamperes.  相似文献

The contribution of pre-defoliation reserves and current assimilates to leaf and root growth was examined in Lolium perenne L. during regrowth after defoliation. Differential steady-state labelling with 13C (CO2 with δ13C = -0.0281 and -0.0088) and 15N (NO3? with 1.0 and 0.368 atom percentage, i.e. δ15N = 1.742 and 0.0052, respectively) was applied for 2 weeks after defoliation. Rapidly growing tissues were isolated, i.e. the basal elongation and maturation zones of the most rapidly expanding leaves and young root tips, with a biomass turnover rate > 1 d?1. C and N weights of the elongation zone showed a transient decline. The dry matter and C concentration in fresh biomass of leaf growth zones transiently decreased by up to 25% 2 d after defoliation, while the N concentration remained constant. This ‘dilution’ of growth zone C indicates a decreased net influx of carbohydrates relative to growth-related influx of water and N in expanding cells, immediately after defoliation. Recovery of the total C and N weights of the leaf elongation zone coincided with net incorporation of currently absorbed C and N, as shown by the kinetics of δ13C and atom percentage 15N in the growth zones after defoliation. C isotope discrimination (Δ13C) in leaf growth zones was about 23‰, 1–2‰ higher than the Δ in root tips. Δ15N in the leaf and root growth zones was 10±3‰. The leaf elongation zones (at 0–0.03 m from the tiller base) and the distant root tips (about 0.2 m from the base) exhibited similar kinetics of current C and N incorporation. The amount of pre-defoliation C and N in the growth zones, expressed as a fraction of total C and N, decreased from 1.0 to 0.5 at 3 (C) and 5 (N) d after defoliation, and to 0.1 at 5 (C) and 14 (N) d after defoliation. Thus, the dependence of growth zones on current assimilate supply was significant, and stronger for C than for N. The important roles of current assimilates (as compared to pre-defoliation reserves) and ‘dilution’ of dry matter in regrowth after defoliation are discussed in relation to the method of labelling and the functional and morphological heterogeneity of shoot tissues.  相似文献

The effect of saline stress on physiological and morphological parameters in Callistemon citrinus plants was studied to evaluate their adaptability to irrigation with saline water. C. citrinus plants, grown under greenhouse conditions, were subjected to two irrigation treatments lasting 56 weeks: control (0.8 dS·m?1) and saline (4 dS·m?1). The use of saline water in C. citrinus plants decreased aerial growth, increased the root/shoot ratio and improved the root system (increased root diameter and root density), but flowering and leaf colour were not affected. Salinity caused a decrease in stomatal conductance and evapotranspiration, which may prevent toxic levels being reached in the shoot. Net photosynthesis was reduced in plants subjected to salinity, although this response was evident much later than the decrease in stomatal conductance. Stem water potential was a good indicator of salt stress in C. citrinus. The relative salt tolerance of Callistemon was related to storage of higher levels of Na+ and Cl? in the roots compared with the leaves, especially in the case of Na+, which could have helped to maintain the quality of plants. The results show that saline water (around 4 dS·m?1) could be used for growing C. citrinus commercially. However, the cumulative effect of irrigating with saline water for 11 months was a decrease in photosynthesis and intrinsic water use efficiency, meaning that the interaction of the salinity level and the time of exposure to the salt stress should be considered important in this species.  相似文献

Abstract

Fine roots (<2 mm) are very dynamic and play a key role in forest ecosystem carbon and nutrient cycling and accumulation. We reviewed root biomass data of three main European tree species European beech, (Fagus sylvatica L.), Norway spruce (Picea abies L. Karst.) and Scots pine (Pinus sylvestris L.), in order to identify the differences between species, and within and between vegetation zones, and to show the relationships between root biomass and the climatic, site and stand factors. The collected literature consisted of data from 36 beech, 71 spruce and 43 pine stands. The mean fine root biomass of beech was 389 g m?2, and that of spruce and pine 297 g m?2 and 277 g m?2, respectively. Data from pine stands supported the hypothesis that root biomass is higher in the temperate than in the boreal zone. The results indicated that the root biomass of deciduous trees is higher than that of conifers. The correlations between root biomass and site fertility characteristics seemed to be species specific. There was no correlation between soil acidity and root biomass. Beech fine root biomass decreased with stand age whereas pine root biomass increased with stand age. Fine root biomass at tree level correlated better than stand level root biomass with stand characteristics. The results showed that there exists a strong relationship between the fine root biomass and the above-ground biomass.  相似文献

Aims: To study the bioelectrochemical behaviour of Pseudomonas aeruginosa (MTCC 17702) and Escherichia coli (MTCC 10436) and to assess their potential to act as anodic biocatalyst with the function of anaerobic consortia for microbial (bio) fuel cell (BFC) application. Methods and Results: Three BFCs (single chamber; open‐air cathode; noncatalysed electrodes) were operated simultaneously in acidophilic microenvironments. Pseudomonas aeruginosa (BFCP) showed higher current density (264 mA m?2) followed by mixed culture (BFCM; 166 mA m?2) and E. coli (BFCE; 147 mA m?2). However, total operating period and substrate degradation were relatively found to be effective with mixed culture (58%; 72 h) followed by BFCP (39%; 60 h) and BFCE (31%; 48 h). Higher electron discharge (ED) was observed with Ps. aeruginosa while mixed culture showed the involvement of redox mediators in the ED process. Conclusions: Mixed culture showed to sustain biopotential for longer periods along with a stable ED. The presence of redox signals and high substrate degradation was also evidencing its performance compared to the pure strains studied. This supports the practical utility of mixed culture over the pure cultures for real‐field BFC applications especially while operating with wastewater. Significance and Impact of the Study: This study revealed the efficiency and viability of mixed consortia in comparison with pure strains for microbial (bio) fuel cell applications.  相似文献

Conversion of large areas of agricultural grassland is inevitable if European and UK domestic production of biomass is to play a significant role in meeting demand. Understanding the impact of these land‐use changes on soil carbon cycling and stocks depends on accurate predictions from well‐parameterized models. Key considerations are cultivation disturbance and the effect of autotrophic root input stimulation on soil carbon decomposition under novel biomass crops. This study presents partitioned parameters from the conversion of semi‐improved grassland to Miscanthus bioenergy production and compares the contribution of autotrophic and heterotrophic respiration to overall ecosystem respiration of CO2 in the first and second years of establishment. Repeated measures of respiration from within and without root exclusion collars were used to produce time‐series model integrations separating live root inputs from decomposition of grass residues ploughed in with cultivation of the new crop. These parameters were then compared to total ecosystem respiration derived from eddy covariance sensors. Average soil surface respiration was 13.4% higher in the second growing season, increasing from 2.9 to 3.29 g CO2‐C m?2 day?1. Total ecosystem respiration followed a similar trend, increasing from 4.07 to 5.4 g CO2‐C m?2 day?1. Heterotrophic respiration from the root exclusion collars was 32.2% lower in the second growing season at 1.20 g CO2‐C m?2 day?1 compared to the previous year at 1.77 g CO2‐C m?2 day?1. Of the total respiration flux over the two‐year time period, aboveground autotrophic respiration plus litter decomposition contributed 38.46% to total ecosystem respiration while belowground autotrophic respiration and stimulation by live root inputs contributed 46.44% to soil surface respiration. Treatment of domestic wastewater using microbial fuel cells (MFCs) will require reactors with multiple electrodes, but this presents unique challenges under continuous flow conditions due to large changes in the chemical oxygen demand (COD) concentration within the reactor. Domestic wastewater treatment was examined using a single-chamber MFC (130 mL) with multiple graphite fiber brush anodes wired together and a single air cathode (cathode specific area of 27 m2/m3). In fed-batch operation, where the COD concentration was spatially uniform in the reactor but changed over time, the maximum current density was 148?±?8 mA/m2 (1,000 Ω), the maximum power density was 120 mW/m2, and the overall COD removal was >90 %. However, in continuous flow operation (8 h hydraulic retention time, HRT), there was a 57 % change in the COD concentration across the reactor (influent versus effluent) and the current density was only 20?±?13 mA/m2. Two approaches were used to increase performance under continuous flow conditions. First, the anodes were separately wired to the cathode, which increased the current density to 55?±?15 mA/m2. Second, two MFCs were hydraulically connected in series (each with half the original HRT) to avoid large changes in COD among the anodes in the same reactor. The second approach improved current density to 73?±?13 mA/m2. These results show that current generation from wastewaters in MFCs with multiple anodes, under continuous flow conditions, can be improved using multiple reactors in series, as this minimizes changes in COD in each reactor.  相似文献

Alfalfa (Medicago sativa L.) is a deeply rooted perennial legume which, under field conditions, may be exposed to varying NO3? concentrations with depth. Our objective was to characterize the effect of localized (deep vs shallow) exposure of alfalfa root systems to NO3? on symbiotic N2 fixation and NO3?-N uptake. Cuttings of a single alfalfa plant were grown in vertical split root systems in a controlled environment chamber. The split root system was a rigid acrylic tube (5 cm diam. by 60 cm long) filled with silica sand and divided into upper and lower sections at the 30-cm depth by a 5-mm-thick wax layer. Roots penetrated the wax layer, but mixing of nutrient solutions between the sections was prevented. Nodulation was restricted to the upper section. The plants were subjected for 10 days to the following treatments: both sections of the split root system received nutrient solution containing either 0.5, 5.25, or 10 mM NO3?; the upper section received 0.5 mM NO3? while the lower section received 10 mM NO3?; or the upper section received 10 mM NO3? while the lower section received 0.5 mM NO3?. Increasing supply of NO3? in the nutrient solution to both sections resulted in higher NO3?-N uptake, lower nodule mass and lower specific nitrogenase activity. Although NO3?-N uptake did not differ, plants exposed to 10 mM NO3? for 10 days in the upper, nodulated section of the root system had a 20% lower nodule mass than plants exposed to the same NO3? concentration in the lower, non-nodulated section of the root system. Specific nitrogenase activity was not different between these two treatments. Therefore, we conclude that: (1) nodule mass was dependent on two factors, the amount of NO3?-N taken up and the concentration of NO3? within the nodulated root zone; and (2) specific nitrogenase activity was little affected by the concentration of NO3? surrounding the nodules, but was strongly inhibited by NO3?-N taken up.  相似文献

The hydraulic conductance of the leaf lamina (Klamina) substantially constrains whole‐plant water transport, but little is known of its association with leaf structure and function. Klamina was measured for sun and shade leaves of six woody temperate species growing in moist soil, and tested for correlation with the prevailing leaf irradiance, and with 22 other leaf traits. Klamina varied from 7.40 × 10?5 kg m?2 s?1 MPa?1 for Acer saccharum shade leaves to 2.89 × 10?4 kg m?2 s?1 MPa?1 for Vitis labrusca sun leaves. Tree sun leaves had 15–67% higher Klamina than shade leaves. Klamina was co‐ordinated with traits associated with high water flux, including leaf irradiance, petiole hydraulic conductance, guard cell length, and stomatal pore area per lamina area. Klamina was also co‐ordinated with lamina thickness, water storage capacitance, 1/mesophyll water transfer resistance, and, in five of the six species, with lamina perimeter/area. However, for the six species, Klamina was independent of inter‐related leaf traits including leaf dry mass per area, density, modulus of elasticity, osmotic potential, and cuticular conductance. Klamina was thus co‐ordinated with structural and functional traits relating to liquid‐phase water transport and to maximum rates of gas exchange, but independent of other traits relating to drought tolerance and to aspects of carbon economy.  相似文献

Whereas for extra‐tropical regions model estimates of the emission of volatile organic compounds (VOC) predict strong responses to the strong annual cycles of foliar biomass, light intensity and temperature, the tropical regions stand out as a dominant source year round, with only little variability mainly due to the annual cycle of foliar biomass of drought‐deciduous trees. As part of the Large Scale Biosphere Atmosphere Experiment in Amazônia (LBA‐EUSTACH), a remote secondary tropical forest site was visited in the dry‐to‐wet season transition campaign, and the trace gas exchange of a strong isoprene emitter and a monoterpene emitter are compared to the wet‐to‐dry season transition investigations reported earlier. Strong seasonal differences of the emission capacity were observed. The standard emission factor for isoprene emission of young mature leaves of Hymenaea courbaril was about twofold in the end of the dry season (111.5 μgC g?1 h?1 or 41.2 nmol m?2 s?1) compared to old mature leaves investigated in the end of the wet season (45.4 μgC g?1 h?1 or 24.9 nmol m?2 s?1). Standardized monoterpene emission rate of Apeiba tibourbou were 2.1 and 3.6 μgC g?1 h?1 (or 0.3 and 0.8 nmol m?2 s‐1), respectively. This change in species‐specific VOC emission capacity was mirrored by a concurrent change in the ambient mixing ratios. The growth conditions vary less in tropical areas than in temperate regions of the world, and the seasonal differences in emission strength could not be reconciled solely with meteorological data of instantaneous light intensity and temperature. Hence the inadequacy of using a single standard emission factor to represent an entire seasonal cycle is apparent. Among a host of other potential factors, including the leaf developmental stage, water and nutrient status, and abiotic stresses like the oxidative capacity of the ambient air, predominantly the long‐term growth temperature may be applied to predict the seasonal variability of the isoprene emission capacity. The dry season isoprene emission rates of H. courbaril measured at the canopy top were also compared to isoprene emissions of the shade‐adapted species Sorocea guilleminiana growing in the understory. Despite the difference in VOC emission composition and canopy position, one common algorithm was able to predict the diel emission pattern of all three tree species.  相似文献
