As the demand for biofuels for transportation is increasing, it is necessary to acquire technologies that will allow affordable production of biodiesel. Conventional biodiesel is mainly produced from vegetable oil by chemical transesterification, but this product has relatively low yield and is competing with agricultural land that can be used for food production. In the present study, 14 filamentous fungi were isolated from different types of soil from Al-Hassa, Saudi Arabia. Nile red staining revealed that lipid bodies were present in 6 of the 14 fungal isolates. Lipid extraction showed that 3 fungi were able to accumulate lipid >20% (wt/wt) of their dry cell mass (0.59–2.40 g/L). The profile of fatty acids revealed a high content of oleic (C18:1, n9), palmitic (C16:0) and linoleic (C18:2) acids similar to conventional vegetable oils used for biodiesel production. Isolate RY4, with the highest lipid yield was identified as Mucor circinelloides based on morphological characteristics confirmed with 18S rRNA gene sequencing. Sequence data of nucleotides were obtained from DNA sequencing submitted to GenBank [GenBank: AB916546.1]. Certain oil compounds were determined by FTIR spectroscopy.  相似文献

Siberian apricot (Prunus sibirica L.), an excellent woody oil plant unique to Asia, is well known for its ability to produce high‐oil seeds for use as a promising feedstock of biodiesel. Based on the investigation of natural Siberian apricot resources in China in the early stage, seeds of Siberian apricot from 74 geographic provenances which can fully reflect the overall information were collected. In this research, seeds oil content, fatty acid composition and biodiesel properties were evaluated, and the key environmental factors that caused the variation of these in different geographic provenance were analyzed. The oil content of Siberian apricot seeds is 45.48%–61.07%, and the average was 50.95% for all provenances. The characteristics of oil can identify and quantify eight fatty acids. The most abundant fatty acids were oleic acid (C18:1; 54.02%–76.54%), followed by linoleic acid (C18:2; 16.78%–38.49%) and erucic acid (C16:0; from 3.27% to 6.12%). Monounsaturated fatty acids are the most abundant in 54.75%–77.03% compared with saturated fatty acids and polyunsaturated fatty acids. The biodiesel properties of most provenance seeds meet the standards of the ASTM D6751 and GB/T 20828, and a few meet the standards of the EN14214. Through the clustering of oil content and fatty acid composition and the analysis of biodiesel properties indexes, it is concluded that KSK provenance is the most suitable for biodiesel production. The XBZ, HHE, AES, ZLQ and LD provenances may be preserved as potential biodiesel. RDA and VPA showed that the effects of environmental factors on the oil properties of Siberian apricot were ranked as terrain factor > climate factor > soil factor, among which longitude, latitude and altitude are the main terrain indicators. These evaluations can provide reference for the effective utilization and further development of Siberian apricot as a bioenergy feedstock.  相似文献

Background

The enzymatic production of biodiesel through alcoholysis of triglycerides has become more attractive because it shows potential in overcoming the drawbacks of chemical processes. In this study, we investigate the production of biodiesel from crude, non-edible Jatropha oil and methanol to characterize Burkholderia cepacia lipase immobilized in an n-butyl-substituted hydrophobic silica monolith. We also evaluate the performance of a lipase-immobilized silica monolith bioreactor in the continuous production of biodiesel.

Results

The Jatropha oil used contained 18% free fatty acids, which is problematic in a base-catalyzed process. In the lipase-catalyzed reaction, the presence of free fatty acids made the reaction mixture homogeneous and allowed bioconversion to proceed to 90% biodiesel yield after a 12 hour reaction time. The optimal molar ratio of methanol to oil was 3.3 to 3.5 parts methanol to one part oil, with water content of 0.6% (w/w). Further experiments revealed that B. cepacia lipase immobilized in hydrophobic silicates was sufficiently tolerant to methanol, and glycerol adsorbed on the support disturbed the reaction to some extent in the present reaction system. The continuous production of biodiesel was performed at steady state using a lipase-immobilized silica monolith bioreactor loaded with 1.67 g of lipase. The yield of 95% was reached at a flow rate of 0.6 mL/h, although the performance of the continuous bioreactor was somewhat below that predicted from the batch reactor. The bioreactor was operated successfully for almost 50 days with 80% retention of the initial yield.

Conclusions

The presence of free fatty acids originally contained in Jatropha oil improved the reaction efficiency of the biodiesel production. A combination of B. cepacia lipase and its immobilization support, n-butyl-substituted silica monolith, was effective in the production of biodiesel. This procedure is easily applicable to the design of a continuous flow-through bioreactor system.  相似文献

Increased costs and limited availability of traditional lipid sources for biodiesel production encourage researchers to find more sustainable feedstock at low prices. Microbial lipid stands out as feedstock replacement for vegetable oil to convert fatty acid esters. In this study, the potential of three isolates of filamentous fungi (Mucor circinelloides URM 4140, M. hiemalis URM 4144, and Penicillium citrinum URM 4126) has been assessed as single-cell oil (SCO) producers. M. circinelloides 4140 had the highest biomass concentration with lipid accumulation of up to 28?wt% at 120?hr of cultivation. The profile of fatty acids revealed a high content of saturated (SFA) and monounsaturated fatty acids (MUFA), including palmitic (C16:0, 33.2–44.1?wt%) and oleic (C18:1, 20.7–31.2?wt%) acids, with the absence of polyunsaturated fatty acids (PUFA) having more than four double bonds. Furthermore, the predicted properties of biodiesel generated from synthesized SCOs have been estimated by using empirical models which were in accordance with the limits imposed by the USA (ASTM D6715), European Union (EN 14214), and Brazilian (ANP 45/2014) standards. These results suggest that the assessed filamentous fungus strains can be considered as alternative feedstock sources for high-quality biofuel production.  相似文献

Phenotypic characterization of soybean event 335-13, which possesses oil with an increased oleic acid content (> 85%) and reduced palmitic acid content (< 5%), was conducted across multiple environments during 2004 and 2005. Under these conditions, the stability of the novel fatty acid profile of the oil was not influenced by environment. Importantly, the novel soybean event 335-13 was not compromised in yield in both irrigated and non-irrigated production schemes. Moreover, seed characteristics, including total oil and protein, as well as amino acid profile, were not altered as a result of the large shift in the fatty acid profile. The novel oil trait was inherited in a simple Mendelian fashion. The event 335-13 was also evaluated as a feedstock for biodiesel. Extruded oil from event 335-13 produced a biodiesel with improved cold flow and enhanced oxidative stability, two critical fuel parameters that can limit the utility of this renewable transportation fuel.  相似文献

Marine microalgae have emerged as important feedstock for liquid biofuel production. The identification of lipid-rich native microalgal species with high growth rate and optimal fatty acid profile and biodiesel properties is the most challenging step in microalgae-based biodiesel production. In this study, attempts have been made to bio-prospect the biodiesel production potential of marine and brackish water microalgal isolates from the west coast of India. A total of 14 microalgal species were isolated, identified using specific molecular markers and based on the lipid content; seven species with total lipid content above 20% of dry cell weight were selected for assessing biodiesel production potential in terms of lipid and biomass productivities, nile red fluorescence, fatty acid profile and biodiesel properties. On comparative analysis, the diatoms were proven to be promising based on the overall desirable properties for biodiesel production. The most potential strain Navicula phyllepta MACC8 with a total lipid content of 26.54 % of dry weight of biomass, the highest growth rate (0.58 day?1) and lipid and biomass productivities of 114 and 431 mgL?1 day?1, respectively, was rich in fatty acids mainly of C16:0, C16:1 and C18:0 in the neutral lipid fraction, the most favoured fatty acids for ideal biodiesel properties. The biodiesel properties met the requirements of fuel quality standards based on empirical estimation. The marine diatoms hold a great promise as feedstock for large-scale biodiesel production along with valuable by-products in a biorefinery perspective, after augmenting lipid and biomass production through biochemical and genetic engineering approaches.  相似文献

A technique to produce biodiesel from crude Zanthoxylum bungeanum seed oil (ZSO) with high free fatty acids (FFA) was developed. The acid value of ZSO was reduced to 1.16mg KOH/g from 45.51mg KOH/g by only one-step acid-catalyzed esterification with methanol-to-oil molar ratio 24:1, H(2)SO(4) 2%, temperature 60 degrees C and reaction time 80min, which was selected as optimum for the acid-catalyzed esterification. During the acid-catalyzed esterification, FFA was converted into fatty acid methyl esters, which was confirmed by (1)H NMR spectrum. Compared with the other two-step pretreatment procedure, this one-step pretreatment can reduce the production cost of ZSO biodiesel. Alkaline-catalyzed transesterification converted the pretreated ZSO into ZSO biodiesel. The yield of ZSO biodiesel was above 98% determined by (1)H NMR spectrum. This study supports the use of crude ZSO as a viable and valuable raw feedstock for biodiesel production.  相似文献

本文根据德国、欧盟和美国制定的生物柴油标准制定了以碘值、十六烷值和脂肪酸组成等参数作为植物油质量的评价体系。通过四条标准,即51<十六烷值<65、碘值<115、亚麻酸<12%和十八碳四烯酸<1%、碳链长度为C12-C22,对国产118种种子含油量超过30%的木本油料植物进行评估,共筛选出53种木本油料植物的种子油可作为发展生物柴油最适合的原料。其中油茶、杏、无患子、臭椿、白檀、海州常山分布广,是值得推广种植的生物柴油植物。富油大科山茶科和无患子科植物的种子油一般都适合发展生物柴油,而富油大科樟科、松科和卫矛科植物的种子油不适合作生物柴油原料。我国各省区可因地制宜选择合适的能源树种,发展生物柴油产业。  相似文献

当前国内外致力于发展生物柴油,因其性能优良,成为化石柴油的替代品。由于以植物油脂生产生物柴油原料成本占总成本的70%-85%,所以亟待开发廉价油脂资源。微生物油脂主要是微生物利用碳水化合物合成的甘油脂,其脂肪酸组成和植物油相近。产油微生物具有资源丰富、油脂含量高、碳源利用谱广等特点,开发潜力大。然而,目前微生物油脂生产成本偏高,研究工作仍以富含多不饱和脂肪酸的高附加值菌油为目标。随着现代分子生物学和生物化工技术的发展,对产油微生物菌种筛选、改良、代谢调控和发酵工程的研究日趋深入,将降低微生物油脂生产成本,为未来生物柴油产业提供廉价原料。  相似文献

Algal biofuels are a growing interest worldwide due to their potential in terms of sustainable greenhouse gas displacement and energy production. This article describes a comparative survey of biodiesel production and conversion yields of biodiesel via alkaline transesterification of acylglycerols extracted from the microalgae Thalassiosira pseudonana and Phaeodactylum tricornutum, grown under silicate or nitrate limitation, and that of model vegetable oils: soybean, and rapeseed oil. Acylglycerols were extracted with n-hexane and the total yield per biomass was determined by gravimetric assay. Under our conditions, the total acylglycerol yield from the microalgae studied was 13-18% of total dry weight. The biodiesel samples were analyzed using gas chromatography-flame ionization detector to determine quantitative information of residual glycerol, mono-, di-, and tri-acylglycerol concentrations in the biodiesel. All of the algal-based biodiesel demonstrated less mono-, di-, and tri-acylglycerol concentrations than the vegetable-based biodiesel under identical transesterification conditions. The fatty acid compositions of all the feedstock oils and their resultant biodiesel were also analyzed and reported. Based on the fatty acid methyl ester compositions of our samples we qualitatively assessed the suitability of the algal-derived biodiesel in terms of cetane number (CN), cold-flow properties, and oxidative stability.  相似文献

Since the biodiesel program has been started in Brazil, the investigation of alternative sources of triacylglycerides from species adapted at semi-arid lands became a very important task for Brazilian researchers. Thus we initiated studies with the fruits of the Terminalia catappa L (TC), popularly known in Brazil as "castanhola", evaluating selected properties and chemical composition of the oil, as well any potential application in biodiesel production. The oil was obtained from the kernels of the fruit, with yields around 49% (% mass). Also, its fatty acid composition was quite similar to that of conventional oils. The crude oil of the TC was transesterified, using a conventional catalyst and methanol to form biodiesel. The studied physicochemical properties of the TC biodiesel are in acceptable range for use as biodiesel in diesel engines.  相似文献

We investigated the potential of seaweeds as feedstock for oil‐based products, and our results support macroalgae (seaweeds) as a biomass source for oil‐based bioproducts including biodiesel. Not only do several seaweeds have high total lipid content above 10% dry weight, but in the brown alga Spatoglossum macrodontum 50% of these lipids are in the form of extractable fatty acids. S. macrodontum had the highest fatty acid content (57.40 mg g?1 dw) and a fatty acid profile rich in saturated fatty acids with a high content of C18:1, which is suitable as a biofuel feedstock. Similarly, the green seaweed Derbesia tenuissima has high levels of fatty acids (39.58 mg g?1 dw), however, with a high proportion of PUFA (n‐3) (31% of total lipid) which are suitable as nutraceuticals or fish oil replacements. Across all species of algae the critical parameter of fatty acid content (measured as fatty acid methyl esters, FAME) was positively correlated (R2 = 0.67) with total lipid content. However, the proportion of fatty acids to total lipid decreased markedly with total lipid content, generally between 30% and 50%, making it an inaccurate measure of the potential to identify seaweeds suitable for oil‐based bioproducts. Finally, we quantified within species variation of fatty acids across locations and sampling periods supporting either environmental effects on quantitative fatty acid profiles, or genotypes with specific quantitative fatty acid profiles, thereby opening the possibility to optimize the fatty acid content and quality for oil production through specific culture conditions and selective breeding.  相似文献

The physicochemical characteristics, fatty acid (FA) profile, and triacylglyceride (TAG) composition of seed oils from fresh Bokbunja (Rubus coreanus Miq.) fruits and traditional Bokbunja wine processing waste were determined in this study. Oil contents of the fresh seeds and the seeds from wine processing waste were similar, accounting for about 18% of dry weight. The free fatty acid (FFA) content between the two seed oils was significantly different (0.50% for fresh seed oil and 73.14% for wine seed oil). Iodine, conjugated diene, saponification values, and unsaponifiable matter were very similar in the oil samples, but the specific extinction coefficients at 232 and 270 nm of wine seed oil were higher than those of fresh seed oil. Linoleic (C18:2, 50.45-53.18%, L) and linolenic (C18:3, 29.36-33.25%, Ln) acids were the dominant FAs in the two seed oils, whereas oleic (C18:1, 7.32-8.04%, O), palmitic (C16:0, 1.55-1.65%, P), and stearic (C18:0, 0.65-0.68%, S) acids were the minor FAs. LLL, OLL, LLLn, OOL, LLnLn, and OOO were the abundant TAGs, representing >90% of the oils.  相似文献

The need to develop biomass-based domestic production of high-energy liquid fuels (biodiesel) for transportation can potentially be addressed by exploring microalgae with high lipid content. Selecting the strains with adequate oil yield and quality is of fundamental importance for a cost-efficient biofuel feedstock production based on microalgae. This work evaluated 29 strains of Chlorella isolated from Malaysia as feedstock for biodiesel based on volumetric lipid productivity and fatty acid profiles. Phylogenetic studies based on 18S rRNA gene revealed that majority of the strains belong to true Chlorella followed by Parachlorella. The strains were similarly separated into two groups based on fatty acid composition. Of the 18 true Chlorella strains, Chlorella UMACC187 had the highest palmitic acid (C16:0) content (71.3?±?4.2 % total fatty acids, TFA) followed by UMACC84 (70.1?±?0.7 %TFA), UMACC283 (63.8?±?0.7 %TFA), and UMACC001 (60.3?±?4.0 %TFA). Lipid productivity of the strains at exponential phase ranged from 34.53 to 230.38 mg L?1 day?1, with Chlorella UMACC050 attaining the highest lipid productivity. This study demonstrated that Chlorella UMACC050 is a promising candidate for biodiesel feedstock production.  相似文献

The study documented the potential of isolated filamentous fungus Aspergillus sp. as whole cell biocatalyst for biodiesel production using Sabourauds dextrose broth medium (SDBM) and corncob waste liquor (CWL) as substrates. SDBM showed improvement in both biomass production (13.6 g dry weight/1000 ml) and lipid productivity (23.3%) with time. Lipid extraction was performed by direct (DTE) and indirect (IDTE) transesterification methods. DTE showed higher transesterification efficiency with broad spectrum of fatty acids profile over IDTE. CWL as substrate showed good lipid productivity (22.1%; 2g dry biomass; 48 h) along with efficient substrate degradation. Lipids derived from both substrates depicted high fraction of saturated fatty acids than unsaturated ones. Physical characteristics of fungal based biodiesel correlated well with prescribed standards. CWL derived biodiesel showed relatively good fuel properties (acid number, 0.40 mg KOH/g of acid; iodine value, 11 g I?/100 g oil; density, 0.8342 g/cm3) than SDBM derived biodiesel.  相似文献

Background

Jatropha curcas is recognized as a new energy crop due to the presence of the high amount of oil in its seeds that can be converted into biodiesel. The quality and performance of the biodiesel depends on the chemical composition of the fatty acids present in the oil. The fatty acids profile of the oil has a direct impact on ignition quality, heat of combustion and oxidative stability. An ideal biodiesel composition should have more monounsaturated fatty acids and less polyunsaturated acids. Jatropha seed oil contains 30% to 50% polyunsaturated fatty acids (mainly linoleic acid) which negatively impacts the oxidative stability and causes high rate of nitrogen oxides emission.

Results

The enzyme 1-acyl-2-oleoyl-sn-glycero-3-phosphocholine delta 12-desaturase (FAD2) is the key enzyme responsible for the production of linoleic acid in plants. We identified three putative delta 12 fatty acid desaturase genes in Jatropha (JcFAD2s) through genome-wide analysis and downregulated the expression of one of these genes, JcFAD2-1, in a seed-specific manner by RNA interference technology. The resulting JcFAD2-1 RNA interference transgenic plants showed a dramatic increase of oleic acid (> 78%) and a corresponding reduction in polyunsaturated fatty acids (< 3%) in its seed oil. The control Jatropha had around 37% oleic acid and 41% polyunsaturated fatty acids. This indicates that FAD2-1 is the major enzyme responsible for converting oleic acid to linoleic acid in Jatropha. Due to the changes in the fatty acids profile, the oil of the JcFAD2-1 RNA interference seed was estimated to yield a cetane number as high as 60.2, which is similar to the required cetane number for conventional premium diesel fuels (60) in Europe. The presence of high seed oleic acid did not have a negative impact on other Jatropha agronomic traits based on our preliminary data of the original plants under greenhouse conditions. Further, we developed a marker-free system to generate the transgenic Jatropha that will help reduce public concerns for environmental issues surrounding genetically modified plants.

Conclusion

In this study we produced seed-specific JcFAD2-1 RNA interference transgenic Jatropha without a selectable marker. We successfully increased the proportion of oleic acid versus linoleic in Jatropha through genetic engineering, enhancing the quality of its oil.  相似文献

In this study, the non-catalytic supercritical method has been studied in utilizing dimethyl carbonate. It was demonstrated that, the supercritical dimethyl carbonate process without any catalysts applied, converted triglycerides to fatty acid methyl esters with glycerol carbonate and citramalic acid as by-products, while free fatty acids were converted to fatty acid methyl esters with glyoxal. After 12 min of reaction at 350 degrees C/20 MPa, rapeseed oil treated with supercritical dimethyl carbonate reached 94% (w/w) yield of fatty acid methyl ester. The by-products from this process which are glycerol carbonate and citramalic acid are much higher in value than glycerol produced by the conventional process. In addition, the yield of the fatty acid methyl esters as biodiesel was almost at par with supercritical methanol method. Therefore, supercritical dimethyl carbonate process can be a good candidate as an alternative biodiesel production process.  相似文献

Lipase-catalyzed alcoholysis of soybean oil deodorizer distillate (SODD) for biodiesel production was studied. During this system both free fatty acids and glycerides could be converted to biodiesel simultaneously. tert-Butanol has been adopted as the reaction medium, in which both the negative effects caused by excessive methanol and by-product glycerol could be eliminated completely. There was no obvious loss in lipase activity even after being repeatedly used for 120 cycles. Fine-pored silica gel and 3 Å molecular were found to be effective to control by-product water concentration and much higher biodiesel yield could be achieved with those adsorbents present in the reaction system. The highest biodiesel yield of 97% could be achieved with 3 Å molecular sieve as the adsorbent.  相似文献

Whole cell Rhizopus oryzae (R. oryzae) IFO4697 immobilized within biomass support particles (BSPs) was used as catalyst for biodiesel production in tert-butanol, in which the stability of the catalyst could be enhanced significantly. Different feedstocks (refined, crude and acidified rapeseed oils) were adopted further for biodiesel production in tert-butanol system and it was found that when acidified rapeseed oil was used as feedstocks, the reaction rate and final methyl ester (ME) yield were significantly higher than that of refined and crude rapeseed oil. Major differences among the aforementioned oils were found to be the contents of free fatty acid (FFA), water and phospholipids, which showed varied influences on whole cell mediated methanolysis for biodiesel production. The reaction rate increased with the increase of free fatty acid content in oils; water content had varied influence on reaction rate and biodiesel yield; using adsorbent to remove excessive water could increase biodiesel yield significantly (from 73 to 84%); it was also found interestingly that phospholipids contained in oils could increase the reaction rate to a certain extent.  相似文献

测定了羊奶果(Elaeagnus conferta Roxb.)种子脂肪酸组成以及矿质元素含量。结果表明:脂肪酸含量为1.98%,主要是油酸(C18:1)34.15%、亚油酸(C18:2)31.51%、软脂酸(C16:0)13.83%、硬脂酸(C18:0)2.88%。饱和脂肪酸:单不饱和脂肪酸:多不饱和脂肪酸含量比为1:2.23:2.75。矿质元素K高达7837.69mgkg-1,Fe为30.99mgkg-1、Zn为10.13mgkg-1,Na为259.5mgkg-1。  相似文献
