Pattern formation mechanisms in developing organisms determine cellular differentiation and function. However, the components that interact during the manifestation of a spatial pattern are in general unknown. Characean algae represent a model system to study pattern formation. These algae develop alternating acid and alkaline transport domains that influence the pattern of growth. In the present study, it will be demonstrated that a diffusion mechanism is implicated in acid and alkaline domain formation and this growth pattern. Experiments on the characean growth pattern were performed that resulted in pronounced, however, unpredictable modifications in the original pattern. A major component involved in this pattern-forming mechanism emerged from the nonlinear kinetics of the H+-ATPase that is located in the plasma membrane of these algae. Based on these kinetics, a mathematical model was developed and numerically analyzed. As a result, the contribution of a diffusional component to the characean acid/alkaline pattern appeared most likely.This work was supported by the Deutsche Forschungsgemeinschaft (grant #571 1/1) to JF.  相似文献

探索加热回流法提取薏苡仁油的最佳工艺条件,并对提取过程进行深入探讨。采用单因素法考察溶剂、料液比、药材粒径、提取时间等对薏苡仁油提取率的影响,在此基础上研究其提取动力学。结果表明薏苡仁油较佳的提取条件为采用丙酮作为溶剂,液固比7倍,药材平均粒径0.25 mm,提取3 h。动力学研究表明,粒径较小时(0.25~0.83 mm),薏苡仁油的提取动力学符合二阶溶出动力学模型,即减小药材粒径,可明显增加有效成分的提取率。  相似文献

The biosorption of Pb(II), Cd(II), and Co(II), respectively, from aqueous solution on green algae waste biomass was investigated. The green algae waste biomass was obtained from marine green algae after extraction of oil, and was used as low-cost biosorbent. Batch shaking experiments were performed to examine the effects of initial solution pH, contact time and temperature. The equilibrium biosorption data were analyzed using two isotherm models (Langmuir and Freundlich) and two kinetics models (pseudo-first order and pseudo-second order). The results indicate that Langmuir model provide best correlation of experimental data, and the pseudo-second order kinetic equation could best describe the biosorption kinetics of considered heavy metals.  相似文献

Coated vesicles, essential organelles of intracellular membrane traffic, have been extensively studied in animal and higher plant cells. In the algae, cytological studies only have been performed which demonstrate the presence of such coated vesicles with their surrounding clathrin lattice. The present work has been carried out on coated vesicles isolated for the first time from the brown algae Laminaria digitata. For comparison of the antigenic characteristics of clathrin prepared from the Bovine brain or adrenocortical cells and the clathrin prepared from algae, polyclonal antibodies have been raised to a purified Bovine brain clathrin in Goat and to Bovine adrenocortical clathrin in Rabbit. The positive immunological responses of the coated vesicles and the clathrin from Algae to these antibodies, evidence an homology between antigenic determinants of clathrin from animal and vegetal cells.  相似文献

A simple 60-min boiling water extraction of algae known to contain surplus P separates essential P compounds and surplus-stored P compounds. Surplus P compounds can be measured in the extract as orthophosphate. Extracts of algae limited in their growth by the amount of available P contain little or no orthophosphate. Limitation of algal growth by P supply induces the enzyme alkaline phosphatase. The activity of this enzyme can be measured at pH 9 using p-nitro-phenylphosphate as substrate. Algae which were P-limited and contained no extractable orthophosphate have as much as 25 times more alkaline phosphatase activity than algae with surplus available P .  相似文献

FLOW CYTOMETRY AND THE SINGLE CELL IN PHYCOLOGY   总被引:1,自引:0,他引:1  
Flow cytometers measure light scattering and fluorescence characteristics from individual particles in a fluid stream as they cross one or more light beams at rates of up to thousands of events per second. Flow cytometrically detectable optical signals may arise naturally from algae, reflecting cell size, structure, and endogenous pigmentation, or may be generated by fluorescent stains that report the presence of otherwise undetected cellular constituents. Some flow cytometers can physically sort particles with desired optical characteristics out of the flow stream and collect them for subsequent culture or other analyses. The statistically rigorous, cell‐level perspective provided by flow cytometry has been advantageous in experimental investigations of phycological problems, such as the regulation of cell cycle progression. The capacity of flow cytometry to measure large numbers of cells in large numbers of samples rapidly and quantitatively has been used extensively by biological oceanographers to define the distributions and dynamics of marine picophytoplankton. Recent work has shown that flow cytometry can be used to elucidate relationships between the optical properties of individual cells and the bulk optical properties of the water they live in, and thereby may provide an explicit link between algal physiology and global biogeochemistry. Unfortunately, commercially available flow cytometers that are optimized for biomedical applications have a limited capacity to analyze larger phytoplankton. To circumvent these limitations, many investigators are developing flow cytometers specifically designed for analyzing the broad range of sizes, shapes, and pigments found among algae. These new instruments can perform some novel measurements, including simple fluorescence excitation spectra, detailed angular scattering measurements, and in‐flow digital imaging. The growing accessibility and power of flow cytometers may allow the technology to be applied to a wider array of problems in phycology, including investigations of nonplanktonic and multicellular algae, but also presents new challenges for effectively analyzing the large quantity of multiparameter data produced. Ultimately, the detection of molecular probes by flow cytometry may allow single‐cell taxonomic and physiological information to be garnered for a variety of algae, both in culture and in nature.  相似文献

The anti-inflammatory, antiangiogenic, anticoagulant, and antiadhesive properties of fucoidans obtained from nine species of brown algae were studied in order to examine the influence of fucoidan origin and composition on their biological activities. All fucoidans inhibited leucocyte recruitment in an inflammation model in rats, and neither the content of fucose and sulfate nor other structural features of their polysaccharide backbones significantly affected the efficacy of fucoidans in this model. In vitro evaluation of P-selectin-mediated neutrophil adhesion to platelets under flow conditions revealed that only polysaccharides from Laminaria saccharina, L. digitata, Fucus evanescens, F. serratus, F. distichus, F. spiralis, and Ascophyllum nodosum could serve as P-selectin inhibitors. All fucoidans, except that from Cladosiphon okamuranus carrying substantial levels of 2-O-alpha-D-glucuronopyranosyl branches in the linear (1-->3)-linked poly-alpha-fucopyranoside chain, exhibited anticoagulant activity as measured by activated partial thromboplastin time whereas only fucoidans from L. saccharina, L. digitata, F. serratus, F. distichus, and F. evanescens displayed strong antithrombin activity in a platelet aggregation test. The last fucoidans potently inhibited human umbilical vein endothelial cell (HUVEC) tubulogenesis in vitro and this property correlated with decreased levels of plasminogen-activator inhibitor-1 in HUVEC supernatants, suggesting a possible mechanism of fucoidan-induced inhibition of tubulogenesis. Finally, fucoidans from L. saccharina, L. digitata, F. serratus, F. distichus, and F. vesiculosus strongly blocked MDA-MB-231 breast carcinoma cell adhesion to platelets, an effect which might have critical implications in tumor metastasis. The data presented herein provide a new rationale for the development of potential drugs for thrombosis, inflammation, and tumor progression.  相似文献

Intertidal macroalgae Fucus and Laminaria experience seasonally fluctuating inorganic N supply. This study examined the effects of long‐term N deprivation, recovery following N resupply, and effects of elevated ammonium and nitrate exposure on N acquisition in intertidal algae using manipulations of N supply in tank culture. Over 15 weeks of N deprivation, internal N and nitrate reductase activity (NRA) declined, but maximum quantum yield of PSII was unaffected in Fucus serratus and Fucus vesiculosus. Low NRA was maintained despite no external nitrate availability and depletion of internal pools, suggesting a constitutive NRA, insensitive to N supply. Nitrate resupplied to N‐starved thalli was rapidly taken up and internal nitrate pools and NRA increased. Exposure to elevated (50 μM) nitrate over 4 days stimulated nitrate uptake and NRA in Laminaria digitata and F. serratus. Exposure to elevated ammonium suppressed NRA in L. digitata but not in F. serratus. This novel insensitivity of NRA to ammonium in Fucus contrasts with regulation of NRA in other algae and higher plants. Ammonium suppression of NRA in L. digitata was not via inhibition of nitrate uptake and was independent of nitrate availability. L. digitata showed a higher capacity for internal nitrate storage when exposed to elevated ambient nitrate, but NRA was lower than in Fucus. All species maintained nitrate assimilation capacity in excess of nitrate uptake capacity. N uptake and storage strategies of these intertidal macroalgae are adaptive to life in fluctuating N supply, and distinct regulation of N metabolism in Fucus vs Laminaria may relate to position in the intertidal zone.  相似文献

Extraction parameters (temperature, pH, duration) and alkaline pre-treatment duration have been systematically varied in the aim of exploring their impact on both chemical structure and gelling properties of carrageenan biopolymers obtained from Mastocarpus stellatus seaweeds, collected on the Northern coast of Portugal. Increasing the alkaline pre-treatment duration PT leads to kappa/iota-hybrid carrageenans containing less sulphate groups and biological precursor monomers. As a result, gel properties in the presence of KCl are improved as demonstrated by the increase in the Young's modulus with parameter PT. Increasing the extraction duration t ameliorates the biopolymer yield with no significant change in the complex kappa/iota-hybrid carrageenan chemical structure. However, smaller molecular weights are obtained and gel properties are seen to be negatively affected. Extraction temperature and pH have dramatic effects on the biopolymer gel strength, and a set of extraction parameters optimized with respect to extraction yield and gel properties is reported. In addition, kappa/iota-hybrid carrageenans obtained throughout this study exhibit a wide range of gel strengths in KCl, and allow us to present correlations between gel thermal properties and the kappa/iota-hybrid carrageenans chemical structure.  相似文献

We investigated the ability of the water flea, Daphnia, to adaptthe size and structure of its filtering apparatus as a responseto experimentally increased biomass of inedible filamentousand colonial cyanobacteria in a large in situ enclosure experiment.Predator-induced phenotypic plasticity in Daphnia has been extensivelydocumented, but only a small number of studies have focusedon morphological changes induced by food quantity and quality.Here we show that Daphnia responded to increased biomass ofinedible phytoplankton in its environment by enlarging the areaand the mesh size of its filtering apparatus. These observationssuggest that Daphnia responds to increased concentrations ofinedible particles in the same fashion as it does in a verylow food environment. In our study, daphnids exposed to a highbiomass of inedible algae, in fertilized enclosures, had significantlylarger (12–15%) filter screens attached to their thirdand fourth limbs in comparison to daphnids exposed to a lowbiomass of inedible algae. The mesh size also increased in thesame conditions. These results suggest that daphnids used theirphenotypic plasticity to respond to changes in their food qualityand quantity. By using this strategy, daphnids can maximizetheir food uptake and hence compensate for the scarcity of suitablefood encountered in very oligotrophic conditions or even ineutrophic conditions when phytoplankton communities are dominatedby large inedible species.  相似文献

The kinetics of phosphate limited growth of two green algae Chorella pyrenoidosa and Selenastrum capricornutum have been studied in chemostats. Several kinetic models which express the specific growth rate as a function of the intracellular phosphours content have been examined, and one of the models was found to be significantly better than the other models. The principles of this model were described in a recent paper by Nyholm. The kinetics of phosphate uptake have been investigated by adding pulses of phosphate to the chemostats. The uptake by phosphours deficient cells could be described by Michaelis–Menten kinetics for phosphate concentrations below approximately 500 μg P/liter. Further, with the assumption of a discontinuous adjustment of the uptake rate at the onset of phosphours deficiency, a complete kinetic model for growth and phosphate removal is proposed. The mean cell size and the contents of chlorophyll and RNA per unit dry weight have been measured for C. pyrenoidosa as a function of the dilution rate.  相似文献

Summary The minimal size of active membrane fragments from sonic homogenates of a Nitrobacter cell suspension is about 22 m. Nitrite oxidizing activity is absent from particles smaller than that although the distribution of cytochrome a, a link in the respiratory chain responsible for nitrite oxidation, does not show any abnormality below the critical size indicated.Two independent estimates of the parameters of the cumulative distribution function for particle size in sonic homogenates have been made using either statistics from electron micrographs or sedimentation pattern observed after density gradient centrifugation. The distribution function was found to be of the log-normal type which was considered to be a consequence of the observed kinetics of sonic comminution. Another estimate of the parameters of the distribution function was made by fixed angle centrifugation, a method much less accurate with respect to size determination but which allowed a rapid estimate of nitrite oxidizing capacity remaining in particles smaller than the given size.Results from the first two determinations of the distribution function for particle size and of its parameters were then used for correcting the error in the estimate of the critical size from the activity found in supernatants from centrifugations in fixed angle rotors.  相似文献

An experimental investigation was carried out, and a mathematical model of interaction between invertebrates (infusoria Paramecium caudatum and rotifera Brachionus plicatilis) and algae (Chlorella vulgaris and Scenedesmus quadricauda) in the "producer-consumer" aquatic biotic cycle with spatially divided links was constructed. The model describes the dynamics of a mixed culture of infusoria and rotifera in the "consumer" link, when they consume a mixed culture of algae coming from the "producer" link. A negative influence of products of algae Scenedesmus metabolism upon the reproduction of infusoria P. caudatum was revealed. Taking this into account, a qualitative coincidence of the results of mathematical modeling with experimental data was obtained. It was shown that the co-existence of mixed algae culture in the "producer" link with invertebrates in the "consumer" link in the "producer-consumer" aquatic biotic cycle is impossible because of the displacement of infusoria P. caudatum by rotifera Brachionus plicatilis.  相似文献

Wild populations of brown marine algae (Phaeophyta) provide extensive surfaces to bacteria and epiphytic eukaryotes for colonization. On one hand, various strategies allow kelps prevent frond surface fouling which would retard growth by reducing photosynthesis and increasing pathogenesis. On the other hand, production and release of organic exudates of high energy value, sometimes in association with more or less selective control of settlement of epiphytic strains, allow bacteria to establish surface consortia not leading to macrofouling. Here, we present the analysis of adhesion and biofilm formation of bacterial isolates from the kelp Laminaria digitata and of characterized and referenced marine isolates. When they were grown in flow cell under standard nutrient regimes, all used bacteria, except one, were able to adhere on glass and then develop as biofilms, with different architecture. Then, we evaluated the effect of extracts from undisturbed young Laminaria thalli and from young thalli subjected to oxidative stress elicitation; this latter condition induced the production of defense molecules. We observed increasing or decreasing adhesion depending on the referenced strains, but no effects were observed against strains isolated from L. digitata. Such effects were less observed on biofilms. Our results suggested that L. digitata is able to modulate its bacterial colonization. Finally, mannitol, a regular surface active component of Laminaria exudates was tested individually, and it showed a pronounced increased on one biofilm strain. Results of these experiments are original and can be usefully linked to what we already know on the oxidative halogen metabolism peculiar to Laminaria. Hopefully, we will be able to understand more about the unique relationship that bacteria have been sharing with Laminaria for an estimated one billion years.  相似文献

The effect of alkaline treatment on the ultrastructure of C-type starch granules was investigated during the alkaline extraction of Araucaria angustifolia (pinhao) starch. The efficiency in protein removal was evaluated using intrinsic fluorescence and Kjeldahl's method. In parallel, morphological changes of starch granules were observed using scanning electron microscopy and atomic force microscopy. The starch crystallinity was monitored by wide-angle X-ray scattering and the lamellar structure was studied by small-angle X-ray scattering (SAXS). The paracrystalline model was employed to interpret the SAXS curves. It was found that the granular organization was significantly altered when alkaline solutions were used during the extraction. A partial degradation of B-type allomorph of starch and a significant compression of semicrystalline growth rings were observed.  相似文献

Biosorption of copper ions by an industrial algal waste, from agar extraction industry has been studied in a batch system. This biosorbent was compared with the algae Gelidium itself, which is the raw material for agar extraction, and the industrial waste immobilized with polyacrylonitrile (composite material). The effects of contact time, pH, ionic strength (IS) and temperature on the biosorption process have been studied. Equilibrium data follow both Langmuir and Langmuir-Freundlich models. The parameters of Langmuir equilibrium model were: q(max)=33.0mgg(-1), K(L)=0.015mgl(-1); q(max)=16.7mgg(-1), K(L)=0.028mgl(-1) and q(max)=10.3mgg(-1), K(L)=0.160mgl(-1) respectively for Gelidium, algal waste and composite material at pH=5.3, T=20 degrees C and IS=0.001M. Increasing the pH, the number of deprotonated active sites increases and so the uptake capacity of copper ions. In the case of high ionic strengths, the contribution of the electrostatic component to the overall binding decreases, and so the uptake capacity. The temperature has little influence on the uptake capacity principally for low equilibrium copper concentrations. Changes in standard enthalpy, Gibbs energy and entropy during biosorption were determined. Kinetic data at different solution pH (3, 4 and 5.3) were fitted to pseudo-first-order and pseudo-second-order models. The adsorptive behaviour of biosorbent particles was modelled using a batch reactor mass transfer kinetic model, which successfully predicts Cu(II) concentration profiles.  相似文献

Release of macromolecules by S. digitata, in 9 different media under in vitro condition have been studied. A direct relationship between microfilariae (mf) release and associated folin positive materials was seen in majority of the cases. High activities of hydrolytic enzymes such as protease, collagenase, alkaline phosphatase and lipase were detected in the excretary-secretary products and worm preparations. Activity of collagenase could not be detected in the male worm under experimental conditions.  相似文献

The potential of surface phytoplankton to withstand photostress was investigated in August 1998 along a transect from the mouth of the Peene‐Strom (Pomeranian Bight) to the open Arkona Sea (Baltic). Photosynthetic efficiency, algal class composition and pigment pattern were determined. Algae were photoinhibited by artificial illumination and the kinetics of recovery were recorded. Under photoinhibitory treatment, algae from the estuary showed a low effective quantum yield but a high potential to recover their maximum photosynthetic efficiency. Contrary to this, the relatively high effective quantum yield of open sea algae under photoinhibitory treatment is accompanied by a low final recovery of maximum photosynthetic efficiency. These phenomena are discussed with respect to nutrient supply, algal class composition and to different strategies of algae to react to light stress. Literature data of summer primary productivity of open sea and coastal algae are compared with our data on electron transport rates. This revealed a low influence of photoinhibitory effects on productivity.  相似文献
