Thirty-six neonatal pigs were randomly assigned to the following treatment groups: sham implanted gonadally intact males (B), sham-implanted castrated males (C), or castrated males implanted with testosterone propionate (C + TP). Four pigs from each group were sacrificed at 7, 14, or 21 days of age after a 6-hr continuous infusion of [3H]thymidine. Myofibers isolated from the triceps brachii were prepared for satellite cell enumeration by light microscope autoradiography. A developmental decline in labeled myofiber nuclei occurred in all groups, however, the greatest decline occurred in C (P less than 0.01). A treatment-by-age interaction was observed for percentage of labeled nuclei. Castration reduced total and labeled nuclei per millimeter myofiber (P less than 0.05), and C + TP had a higher percentage of labeled nuclei than C (2.8 vs 2.2%; P less than 0.05). Since triceps brachii muscles from 21 day B and C + TP were 120% (P less than 0.05) of C, the results indicate that postnatal growth of skeletal muscle is dependent on satellite cell mitotic activity and that testosterone enhances this activity in neonatal pigs. 相似文献

By means of light, electron microscopy and morphometry methods contents and structural organization of endocrinocytes have been investigated in gastric glands of rats, to whom testosterone propionate has been injected for 5, 10, 15, 20 days. The hormone produces certain stimulating effect on the endocrine apparatus function. During the first stages of the experiment most of the endocrinocytes are at the state of active secretion; this is testified as a decreasing composition of histochemically revealed cells in epithelium and their degranulation. During the following stages the process of synthesis is activated in the cells and their amount increases. Appearance of numerous agranular, poorly differentiated endocrinocytes and exo-endocrine cells demonstrates certain intensification of cytogenesis of endocrinocytes in the gastric epithelium. The changes observed are considered as adaptive, directed to stabilize homeostasis, when the hormonal balance is disturbed. 相似文献

The effects of graded doses of testosterone propionate administered to female rats on Day 4 of postnatal life have been determined. The incidence of failure of ovulation as adults was related to the dose. With increasing dosage over the range 1–100 μg no significant evidence of a progressive decrease in immunoassayable luteinizing hormone concentration in the plasma or anterior pituitary was obtained. The reported protective action of sodium phenobarbitone when administered with testosterone propionate could not be confirmed. Single injections of 100 μg of testosterone, dihydrotestosterone, 5 α-androstanediol, or a combination of the two latter compounds had no masculinizing effect. When the 17-β propionate derivatives of these compounds were administered at the same dose level only testosterone propionate had a masculinizing effect. 相似文献

Two gilts were administered testosterone propionate and their subsequent plasma testosterone concentrations and male sex behavior were recorded. These were compared to testosterone concentrations and male sex behavior in boars. Testosterone propionate (75 mg) was administered to the gilts every other day for 20 days (induction scheme) and every 10 days there-after (maintenance scheme). Concentrations of testosterone in plasma were elevated to concentrations detected in the boars during the induction scheme. During the maintenance scheme, concentrations of testosterone appeared to be lower than in boars. At 20, 30 and 40 days following the first injection, sniffing, nosing and mating song behaviors were exhibited by the testosterone treated gilts similar in frequency to the boars. Mounting behavior was first detected 30 days following the first testosterone propionate injection, and by day 40, the frequency of mounting was greater than observed in boars. 相似文献

A sensitive and specific liquid chromatography-tandem mass spectrometry (LC-MS/MS) assay for the detection of several synthetic glucocorticoids in kidney, muscle and hair samples of cattle after a single intramuscular injection is described. After a dichloromethane wash of the hair samples, analytes were released from the hair matrix by enzymatic digestion. Muscle samples were also digested enzymatically using proteinase, while kidney samples were deconjugated by Helix pomatia juice. These preliminary steps were followed by a methanol extraction and a solid phase extraction (SPE) clean up step for all matrices. Chromatographic separation was achieved on a Hypersil Hypercarb column and MS/MS data were obtained in the multiple reaction monitoring mode using negative electrospray ionization. The developed protocols were evaluated by assessing residue concentrations in muscle, kidney and hair samples of thirteen calves, treated with a particular intramuscular injection of glucocorticoid. The lowest residue levels were found in muscle samples (approximately 5% of the residue levels in kidney), while high residue levels were obtained in hair samples. Hair is an interesting matrix since the sampling is non-invasive and the drugs may stay incorporated for a longer period of time. 相似文献

Serum estradiol (E2), testosterone (T) and dihydrotestosterone (DHT) were measured in juvenile (pre-pubertal) male rhesus monkeys injected with either 8 mg or 80 mg of testosterone propionate (TP). After one week, the three steroids were elevated and remained essentially unchanged for the duration of the study. There was little difference in serum E2 or DHT when comparing the two groups of steroid-treated monkeys. In contrast, T levels were consistently greater in the animals given the high dosage of TP. 相似文献

Two ewes were administered testosterone propionate and subsequent plasma testosterone concentrations determined and male sex behavior recorded. Initially ewes were administered 50 mg of testosterone propionate every other day for 20 days. Within 6 days following the first injection, concentrations of testosterone in plasma increased to 8.0 to 10.0 ng/ml. A 50 mg injection of testosterone propionate administered every 10 days thereafter maintained concentrations of plasma testosterone at 1.0 to 3.0 ng/ml. Sex behavior tests conducted with non-estrus and estrus ewes showed that both testosterone treated ewes developed male sex behavior similar to a ram. Ewes in estrus were mounted by testosterone treated ewes an average of 6.7 ± 1.2 times during a 10 minute test whereas none of the non-estrus ewes were ever mounted. Silastic implants containing testosterone propionate placed in the ewes 83 days following the first injection maintained concentrations of plasma testosterone at 6.0 to 8.0 ng/ml for a 20 day period. Therefore, administration of testosterone propionate to ewes effectively stimulates male sex behavior and would obviate the necessity for vasectomized rams for estrus detection. 相似文献
