Populations of sugarbeet (Beta vulgaris L.) plants that differed in taproot/leaf weight ratio and in photosynthate partitioning between taproots and fibrous roots did not differ in root/shoot ratio as indicated by relative dry weight distribution. Based on the hypothesis that dry weight distribution is influenced by the metabolism of imported sucrose, we examined the relationships between the activity of the enzymes of sucrose metabolism and dry weight distribution as a function of genotype and ontogeny. A decreased specific activity of acid invertase in taproots was associated with increased taproot/fibrous root weight ratio at 21 and at 28 days post-emergence. Alkaline invertase activity was negatively correlated with taproot/fibrous root weight ratio at 28 days. Sucrose synthetase specific activities of taproots were not correlated with dry matter distribution. Acid invertase may influence photosynthate partitioning between the taproot and fibrous roots via regulation of sucrose levels in the region of fibrous root initiation.  相似文献

Activities of acid and alkaline invertases and sucrose synthase were determined in roots and nodules of lentil at various stages of development. Alkaline invertase and sucrose synthase were both involved in sucrose metabolism in the nodule cytosol, but there was only a small amount of acid invertase present. Activity of sucrose metabolizing enzymes in roots was significantly less than that observed in the nodules. Amongst sugars, sucrose was found to be the main component in the host cytosol. Lentil neutral invertase (LNI) was partially purified from nodules at 50 days after sowing (DAS). Two forms of invertase were identified, i.e., a major form of 71 kDa which was taken for enzyme characterization and a minor form of 270 kDa which was not used for further studies. The purified enzyme exhibited typical hyperbolic saturation kinetics for sucrose hydrolysis. It had a Km of 11.0 to 14.0 mM for sucrose depending upon the temperature, a pH optimum of 6.8 and an optimum temperature of 40 °C. Compared with raffinose and stachyose, sucrose was better substrate for LNI. The enzyme showed no significant hydrolysis of maltose and p-nitrophenyl--D-glucopyranoside, showing its true -fructosidase nature. LNI is completely inhibited by HgCl2, MnCl2 and iodoacetamide but not by CaCl2, MgCl2 or BaCl2.  相似文献

Sugar Absorption and Sucrose Inversion by Excised Tomato Roots   总被引:3,自引:0,他引:3  
The growth of excised tomato roots in sucrose is accompaniedby the appearance in the medium of glucose and fructose. Thequantitative relations between this appearance of glucose andfructose in the medium, total sugar absorption by the roots,and the decrease in the sucrose concentration of the mediumdo not suggest any causal relationship between sucrose uptakeand glucose and fructose appearance in the medium. Excised tomato roots exhibit surface invertase activity witha pH optimum at 4.0–4.4. Alterations of external pH, whichdid not affect sucrose absorption, drastically altered the levelsof glucose and fructose appearing in the medium. Glucose is preferentially absorbed from mixtures of glucoseand fructose, and by adjustment of the ratio of the two sugars,a mixture can be obtained from which equimolar absorption ofthe two sugars occurs. Root growth in this mixture is, however,very poor compared with that occurring in presence of sucrose. The results are discussed in the light of earlier studies onsucrose uptake by cultured tomato roots.  相似文献

Data from both controlled environment and field experimentson sugarbeet are used to support the concept that dry matterdistribution between top and storage root growth can be adequatelyquantified by over a large part of the growing season. Very young plants,prior to cambial ring development, had higher slopes for thelog-log relationship at each harvest. Estimates of the slopesbecame relatively constant about 30 d after emergence indicatingthat the pattern for dry matter distribution was determinedat an earlier stage of growth than is apparent from considerationof linear growth scales. Estimates of and are similar to thosereported for other storage root crops with differences in reflectingrelative size differences. Differences in applied N in the fieldwere detected as a shift in the intercept of the relationshipfor dry matter distribution. In general, the results with sugarbeetare agreement with the Barnes' model, which relates assimilatepartitioning to relative sink activities, respiration rates,and initial weights of plant parts. Total plate counts were determined on boneless cooked, cubed chicken meat obtained from a commercial processor. Survival of the natural flora was determined after the meat was freeze-dehydrated and rehydrated at room temperature for 30 min and 50, 85, and 100 C for 10 min. Total counts of bacteria in the rehydrated samples were determined during storage of the meat at 4, 22, and 37 C until spoilage odor was detectable. Meat samples were inoculated with Staphylococcus aureus, then dried, rehydrated, and stored at the same temperatures. Numbers of surviving organisms in the inoculated samples were determined with use of both selective and nonselective media. Representative genera surviving the various rehydration treatments were determined. Approximately 32% of the bacteria in the meat survived during dehydration and rehydration at room temperature. Many numbers and types of vegetative bacteria also survived rehydration at 50 C. When meat was rehydrated at 85 or 100 C, the initial count was less than one per gram. The only organisms isolated from samples rehydrated at 85 or 100 C were of the genus Bacillus. S. aureus in inoculated samples survived dehydration and rehydration at 60 C. Storage of all rehydrated samples at 4 C gave a good shelf life (18 or more days). The study indicates that freeze-dehydrated meat should be produced with adequate microbiological control and that such meat should be rehydrated in very hot water.  相似文献

We developed a technique to map the availability of sugars and amino acids along live roots in an intact soil-root matrix with native microbial soil flora and fauna present. It will allow us to study interactions between root exudates and soil microorganisms at the fine spatial scale necessary to evaluate mechanisms of nitrogen cycling in the rhizosphere. Erwinia herbicola 299R harboring a promoterless ice nucleation reporter gene, driven by either of two nutrient-responsive promoters, was used as a biosensor. Strain 299RTice exhibits tryptophan-dependent ice nucleation activity, while strain 299R(p61RYice) expresses ice nucleation activity proportional to sucrose concentration in its environment. Both biosensors exhibited up to 100-fold differences in ice nucleation activity in response to varying substrate abundance in culture. The biosensors were introduced into the rhizosphere of the annual grass Avena barbata and, as a control, into bulk soil. Neither strain exhibited significant ice nucleation activity in the bulk soil. Both tryptophan and sucrose were detected in the rhizosphere, but they showed different spatial patterns. Tryptophan was apparently most abundant in soil around roots 12 to 16 cm from the tip, while sucrose was most abundant in soil near the root tip. The largest numbers of bacteria (determined by acridine orange staining and direct microscopy) occurred near root sections with the highest apparent sucrose or tryptophan exudation. High sucrose availability at the root tip is consistent with leakage of photosynthate from immature, rapidly growing root tissues, while tryptophan loss from older root sections may result from lateral root perforation of the root epidermis.  相似文献

Changes of Sugar Levels in Cucumber Leaves during Ammonium Toxicity   总被引:2,自引:0,他引:2  
Toxic effects of high concentrations of ammonia were studied in the cucumber plant (Cucumis sativus cv. Suisei No. 2). When the cucumber plant was cultured with 200 mg/l NH3-N (as NH4Cl), some characteristic symptoms, probably due to ammonium toxicity, appeared in the leaves after about 1 week, while no such symptoms were observed in the plants cultured with 20 mg/1 NH3-N. The level of free sugars in 20 mg/l NH3-N treated plants decreased with time and was lower than that in plants treated with 200 mg/l NH3-N. Specially distinct differences were found as regards the levels of fructose and glucose. After 9 days' culture the content of glucose in 200 mg/l NH3-N plants was 17 times higher than that of 20 mg/l NH3-N plants. From the results of an incorporation of photosynthesized 14CO2 for 3 hours into newly synthesized glucose it is evident that this accumulation of glucose can not be the degradative product of a glucose polymer such as starch. The levels of starch were also studied, and it was found that the starch level decreased due to ammonium toxicity. These results suggest that the translocation of glucose after its synthesis is inhibited by ammonium toxicity, at least up to starch synthesis.  相似文献

Sucrose in the tissues of the sugarbeet (Beta vulgaris L.) can be shown as follows. Fresh or stored roots are cut into pieces having a block face of about 1 × 2 cm, and sections of about 150 μ thickness prepared from these. The sections are rinsed 15-30 sec in iced distilled water, placed in Ba(OH)2-saturated methanol for 3 min and then rinsed twice in methanol for 1 and 5 min respectively. They are then transferred to ethanol through a graded series consisting of 80, 60, 40, and 20% methanol in ethanol, 5 min in each, with evacuation as necessary to remove bubbles. Temporary mounting is in ethanol and examination made by incident light or darkfield illumination. Gradual replacement of ethanol with xylene permits mounting in a resinous medium. An opaque granular precipitate of barium saccharate shows the location of the sucrose within the cells.  相似文献

Total and anaerobic counts were ascertained on boneless, cooked, cubed, frozen chicken meat. We determined survival of aerobes and anaerobes in the natural flora after the meat was freeze-dehydrated and rehydrated at room temperature for 30 min and at 50, 85, and 100 C for 10 min. Total and anaerobic counts of bacteria in the rehydrated meat were established during storage of samples at 4, 22, and 37 C-until a spoilage odor was detected. Samples were also inoculated with Clostridium sporogenes and were dried and rehydrated at 100 C and stored at 37 C. Approximately 21% of the aerobes and 37% of the anaerobes survived drying and rehydration at room temperature. Many genera of aerobes, anaerobes, and facultative anaerobes survived drying and rehydration at 50 C; only sporeformers survived rehydration at 85 or 100 C. Low-temperature (4 C) storage of rehydrated meat produced ample shelf life (over 20 days), whereas storage at the higher temperature resulted in a shelf life of less than 30 hr. Approximately 81% of the C. sporogenes cells survived rehydration at 100 C and grew to over 10(7) cells within 40 hr. Our study presents additional data for adequate microbiological control in processing of freeze-dehydrated meat. Also, it points out the natural selection for sporeformers at high temperature of rehydration, stressing the need for consumer education in product handling for safety purposes.  相似文献

The activity of sucrose-phosphate synthase (SPS) in sugar beet (Beta vulgaris L.) leaves was shown to exceed considerably the synthesizing activity of sucrose synthase (SS). The rise in SPS activity was related to the daylight period; i.e., it was associated with the rate of photosynthesis. The highest SPS activity was characteristic of fully expanded source leaves. In young developing leaves (leaves expanded to less than half of their final size), which represent the sink organs, the SPS activity was 2.5 times lower. At all stages of leaf development, the synthesizing SS activity was rather low. The diurnal change of SS activity was independent of photosynthesis and showed a slight rise from 6:00–8:00 p.m. Under field conditions, the highest SPS activity was found in leaves in the terminal stage of their development (105-day-old plants); the synthesizing activity of SS showed little changes during this period. The activity of soluble acid invertase was characteristic of young leaves. In mature leaves, the activity of this enzyme correlated with the daylight period. These changes occurred on the background of low sucrose content in leaves. The regulation of SPS, SS, and invertase activity is discussed. It is supposed that compartmentation of these enzymes in the photosynthesizing cell is important for transport, metabolism, and the osmotic function of sucrose in leaves.  相似文献

We studied the effects of synthetic analogs of phytohormones (benzyladenine, IAA, and GA) on the activities of the enzymes catalyzing sucrose synthesis and metabolism, sucrose phosphate synthase (SPS, EC 2.4.1.14) and sucrose synthase (SS, EC 2.4.1.13), and on the content of chlorophyll and protein during the sugar-beet (Beta vulgaris L.) ontogeny. Plant spraying with phytohormonal preparations activated SPS in leaves; direct interaction between phytohormones and the enzyme also increased its activity. The degree of this activation differed during the ontogeny and in dependence on the compound used for treatment. Analogs of phytohormones maintained high protein level in leaves, retarded chlorophyll breakdown, and, thus, prolonged leaf functional activity during development. Phytohormonal preparations practically did not affect the SS activity both after plant treatment and at their direct interaction with the enzyme. It is supposed that the SS activity in sugar-beet roots is controlled by sucrose synthesized in leaves rather than by phytohormones. The effects of hormones on leaf metabolism were mainly manifested in growth activation.  相似文献

In vivo spectrophotometric assays were conducted with sections of plant material cut from modified stems and storage roots. Phytochrome levels are highest in sections associated with growth such as buds and cambial regions. Interior tissues of tubers and the scale leaves of bulbs which are storage tissue contain low or no detectable phytochrome. However, in the storage root of Pastinaca saliva L. phytochrome is distributed in both the phloem and central regions, though it is highest in the cambial region. Total phytochrome in Gladiolus corm buds exposed to 5 minutes of red light followed by approximately 24 hours of darkness decreases more than 60 per cent. This rapid loss of detectable phytochrome in Gladiolus corm buds resembles phytochrome destruction in etiolated seedlings.  相似文献

对三个葡萄品种浆果发育过程中可溶性糖累积和内源IAA、GA3 、ABA含量的动态变化等进行比较研究。结果表明 ,巨峰和木纳格葡萄浆果生长呈双S型曲线 ,无核白葡萄果实生长呈S型曲线 ;浆果内可溶性糖的累积主要发生在果实发育的后期。浆果发育初期 ,无核白葡萄IAA含量远高于两个有核品种的含量 ,有利于幼果坐果 ;浆果发育后期 ,无核白葡萄IAA和GA3 的含量低于两个有核品种 ,而ABA的含量比两个有核品种高。GA3 与ABA比值的变化对浆果的发育起着关键的作用。  相似文献

The Influence of Shoots, Roots, and Hormones on Sucrose Distribution   总被引:1,自引:0,他引:1  
The effects of hormones on sucrose distribution were studiedin relation to the control of the relative growth of the shootand the root. The roots and the shoots above the cotyledonsof bean seedlings were removed and the remaining explants allowedto regenerate so that they consisted of growing buds, a segmentof the hypocotyl, and short growing roots. [14C]Sucrose wasapplied to the middle of the hypocotyl and its distributionwas measured in relation to the presence of the shoots and rootsand to their replacement by auxin and a cytokinin. It was foundthat radioactivity moved towards both the shoot and the root,and that removing one growing region greatly reduced the transportin its direction and increased the relative transport in theopposite direction. Both hormones restored a large part of themovement of radioactive sucrose when they replaced any one ofthe growing regions. However, there was some preferential actionof benzyladenine when it replaced the buds and of auxin whenit replaced the roots. The results indicate that in the intactplant the development of the shoot and the root is correlatedby a positive feedback involving auxin and cytokinins, and themovement of sucrose is controlled indirectly by the sink activityof these growing regions. This control of the movement of sucrose,and presumably other metabolites, accounts for the short-termcompetitive relations between the shoot and the root.  相似文献

Acetic acid bacteria were present at all stages of wine making, from the mature grape through vinification to conservation. A succession of Gluconobacter oxydans, Acetobacter pasteurianus, and Acetobacter aceti during the course of these stages was noted. Low levels of A. aceti remained in the wine; they exhibited rapid proliferation on short exposure of the wine to air and caused significant increases in the concentration of acetic acid. Higher temperature of wine storage and higher wine pH favored the development and metabolism of these species.  相似文献

Changes in the numbers and types of lactate-producing and lactate-utilizing bacteria in the rumen of sheep were followed during stepwise adaptation from a low- to a high-concentrate diet. The mean numbers of bacteria increased after each change in diet when increasing amounts of maize grain were substituted for maize stover. A surge in number of amylolytic bacteria always preceded an increase in lactate-utilizing bacteria, and with the final diet containing 71% grain and molasses the two groups tended to balance each other, which resulted in low lactic acid accumulation. The lactate utilizers thus played a key role in controlling the fermentation. Orderly shifts occurred among the predominating amylolytic and lactate-utilizing bacteria in response to the gradual decrease in ruminal pH as the amount of maize meal in the diet increased. Among the lactate utilizers, the succession began with acid-sensitive Veillonella and Selenomonas, which were superseded by more acid-tolerant Anaerovibrio and Propionibacterium. Among the amylolytic bacteria, Bacteroides was superseded by more acid-tolerant Lactobacillus and Eubacterium. The ecological succession of predominating genera was shown to be correlated significantly with ruminal pH and, more specifically, with the length of time as well as the extent to which the pH remained below a certain critical undefined value in the rumen, arbitrarily set at pH 6.00.  相似文献
