Larval cuticle fromTrichoplusia ni, Helicoverpa (=Heliothis)zea, andHeliothis virescens and a cellulose substrate were used to quantify release of proteolytic, chitinolytic, and lipolytic enzymes by germinating conidia of the entomopathogenic fungus,Nomuraea rileyi. There was no significant difference in conidial viability incubated withT. ni, H. zea or cellulose substrates. Conidial viability onH. virescens cuticle, however, was significantly lower (ca. 19–25%) than the other three substrates. The presence of cuticle substrates, especially cuticle ofT. ni, stimulated germination. The nature of the substrate influenced both the time and quantity of the enzymes expressed. Specific proteases (aminopeptidase, chymoelastase, trypsin) generally were expressed earlier and/or in greater quantities on cuticular than on the cellulose substrate. Although both chitinolytic enzymes (endochitinase, N-acetylglucosaminidase) were detected on all three cuticular substrates, their activity was substantially lower than that of the proteolytic enzymes. Lipase activity was only minimally present. Early concurrent release of both proteases and chitinases suggested that both may be important in the penetration of the larval integument by germinating conidia ofN. rileyi. Expression of proteases and chitinases, especially aminopeptidase and endochitinase was probably a specific response to cuticle, because little or no activity was expressed on the non-host, cellulose substrate.This article reports the results of research only. Larval cuticle ofHelicoverpa (Heliothis)zea and yeast extract added to a minimal medium (MM) induced germination of conidia ofNomuraea rileyi whereas sterile distilled water or MM alone did not. Yeast extract increased mycelial yield, but when cuticle was added, mycelial yield significantly decreased. The action of plant cysteine proteases on the midgut peritrophic membrane (PM) of a polyphagous herbivorous lepidopteran, Trichoplusia ni, was studied. Proteins in PMs isolated from T. ni larvae were confirmed to be highly resistant to the serine proteinases trypsin and chymotrypsin, but were susceptible to degradation by plant cysteine proteases, which is consistent with the known molecular and biochemical characteristics of the T. ni PM proteins. However, the PM proteins were not degraded by plant cysteine proteases in larvae or in the presence of larval midgut fluid in vitro. With further biochemical analysis, cysteine protease-inhibiting activity was identified in the midgut fluid of T. ni larvae. The cysteine protease-inhibiting activity was heat resistant and active in the tested pH range from 6.0 to 10.0, but could be suppressed by thiol reducing reagents or reduced by treatment with catalase. In addition to T. ni, cysteine protease-inhibiting activity was also identified from two other polyphagous Lepidoptera species, Helicoverpa zea and Heliothis virescens. In conclusion, results from this study uncovered that herbivorous insects may counteract the attack of plant cysteine proteases on the PM by inhibiting the potentially insecticidal cysteine proteases from plants in the digestive tract. However, the biochemical identity of the cysteine protease-inhibiting activity in midgut fluid has yet to be identified.  相似文献

The entomogenous fungusMetarhizium anisopliae attacks a broad range of insects, including the agricultural pestsGalleria mellonella (the Greater Wax Moth) andTrichoplusia ni (the Cabbage Looper). Five strains ofM. anisopliae from widely divergent isolation sources were culturedin vitro on media containing gelatin, glucose plus nitrate, or purified cuticle fromG. mellonella orT. ni larvae. The production of extracellular enzymes such as proteases, chitinases, and esterase was compared. A great deal of natural strain variability was found in enzyme patterns. The highest levels of proteases and endochitinase were produced in cuticle-grown cultures. Three of five strains produced exceptionally high levels of chymoelastase (47,000 to 98,000 IU/mg protein) on cuticle. Surprisingly, the highest levels ofN-acetyl glucosaminidase were produced in gelatin-grown cultures. Most strains produced esterase under all growth conditions. The source of insect cuticle did not strongly influence the production of enzymes.  相似文献

Beauveria bassiana is a promising agent for biological control of agricultural insect pests. Its broad host range includes both the Greater Wax Moth (Galleria mellonella) and the cabbage looper (Trichoplusia ni). Five strains ofB. bassiana from the Agricultural Research Service Culture Collection were obtained and grown on defined media and a medium containing purified cuticle from eitherG. mellonella orT. ni. The production of cuticle-degrading enzymes (such as proteases, chitinases, and esterase) was examined. Natural strain variability in enzyme levels was significant, and there was no evidence of coordinated expression. Further, enzyme expression differed considerably as a function of cuticle source. The results suggest that it may be possible to tailor biological control agents against specific targets. Natural strain variability could be used to rationally develop improved mycoinsecticides.  相似文献

The aim of this work was to compare pupal holotissues (Diatraea saccharalis, Anticarsia gemmatalis,andBombyx mori) and larval hemolymph of several moth species (Helicoverpa zeaandBombyx mori) commonly produced in Brazilian laboratories as components of artificial diets forin vitrorearing ofTrichogramma galloiandTrichogramma pretiosum.Diets based on larval hemolymph ofH. zeaproduced the best development of both natural enemies, although a diet with pupal holotissues ofB. moriallowed full development ofT. pretiosum.Amino acid analysis showed qualitative differences among the insect derivates and eggs of the natural and alternative hosts, but with a quantitative similarity. Electrophoretic analysis resulting in a diverse protein-stained bands evidenced the qualitative differences among these components.  相似文献

The effects of cuticle from larvae of Trichoplusia ni, Heliothis zea and H. virescens on rate and extent of germination of conidia of a Mississippian isolate (MS) and an Ecuadoran (EC) isolate of Nomuraea rileyi were studied. Solid substrates generally stimulated more germination than submerged substrates. There was little or no effect of cuticle source (H. zea or H. virescens) on germination of either the EC isolate or the MS isolate cultured on a solid substrate, however, differences in patterns of germination were obtained in submerged substrates. Addition of cuticle of H. zea or H. virescens generally increased the germination time for the MS isolate. Germination time for the EC isolate was significantly increased when H. virescens cuticle was used.This article reports the results of research only. Naturally occurring populations of immature and adultGeocoris punctipes,adultColeomegilla maculataand immature coccinellids were monitored on field-grown tomato lines susceptible and resistant toManduca sextaandHelicoverpa zea. Helicoverpa zeaandHeliothis virescenseggs and small larvae that serve as prey for these predators also were monitored. MoreH. zeaandH. virescenseggs and small larvae were found on resistant than on susceptible plant lines. However, similar populations of largeH. zeaandH. virescenslarvae were found on resistant and susceptible plants. The number of adultGeocoris punctipes,adultColeomegilla maculataand immature coccinellids on resistant plants was always as high or higher than the number on susceptible plants. The data demonstrate no incompatibility of host-plant resistance with biological control provided by these predaceous insects, but indicate that the number ofG. punctipesand coccinellids required to provide effective biological control may develop too late in the season to be of practical value. Large populations of stilt bugs (Jalysus wickhami,Hemiptera: Berytidae) and spiders were observed to occur earlier in the growing season than eitherG. punctipesor coccinellids and may be a significant source of mortality forH. zeaeggs and small larvae.  相似文献

TwoApium graveolens var.rapaceum (L.) cultivars that differ in their suitability for the survival and growth ofSpodoptera exigua (Hübner) andTrichoplusia ni (Hübner) were used to examine the effect of genetic and seasonal environmental variation in host plant suitability on the efficacy ofBacillus thuringiensis subsp.kurstaki (Berliner). The effects of host plant genotype andB. thuringiensis were generally independent, so thatB. thuringiensis efficacy was greatest on the resistant host plant cultivar. Host plant suitability varied within growing season for both insect species but, while host plant suitability decreased with increasing plant age forT. ni, the response ofS. exigua to plant age was not as clear. Within season variation in host plant suitability affectedB. thuringiensis efficacy and the interaction betweenB. thuringiensis and host plant cultivar forS. exigua but not forT. ni. Soluble protein and Folin-Denis phenolic concentrations of host plant tissue were not correlated with changes in host plant suitability to either insect species.  相似文献

In order to derive quantitative estimates of predation rate from serological gut analysis data, one must have an estimate of the interval during which a meal can be detected after feeding. In practice this has turned out to be ‘Dmax,’ defined as ‘...the time from finishing a meal until that meal could just no longer be detected in any individuals.’ However Dmax substitutes an absolute limit for what is really a continuous variable with significant variation. We examined this problem in a study of the detectability ofHelicoverpa zea Boddie (Lepidoptera: Noctuidae) fifth instar remains in the guts ofPolistes metricus Say (Hymenoptera: Vespidae). Wasps were maintained onTrichoplusia ni (Hübner) (Lepidoptera: Noctuidae) fifth instars before being fed a singleH. zea fifth instar. They were killed and frozen at 0, 24, 48 and 96 h intervals, with those held for more than 24 h fed a singleT. ni fifth instar at 24 h intervals in order to simulate continued feeding. Wasp abdomens were assayed by immunodot, using a monoclonal antibody toH. zea arylphorin. There was a logarithmic decay in the proportion ofP. metricus positive over time, a singleH. zea fifth instar meal having a detectability half-life of 19.4 h at field temperatures. If prey antigen detectability decays exponentially, then a detectability half-life is a more appropriate unit of detectability than an absolute detectability period.  相似文献

Trichoderma harzianum is a widely distributed soil fungus that antagonizes numerous fungal phytopathogens. The antagonism of T. harzianum usually correlates with the production of antifungal activities including the secretion of fungal cell walls that degrade enzymes such as chitinases. Chitinases Chit42 and Chit33 from T. harzianum CECT 2413, which lack a chitin-binding domain, are considered to play an important role in the biocontrol activity of this strain against plant pathogens. By adding a cellulose-binding domain (CBD) from cellobiohydrolase II of Trichoderma reesei to these enzymes, hybrid chitinases Chit33-CBD and Chit42-CBD with stronger chitin-binding capacity than the native chitinases have been engineered. Transformants that overexpressed the native chitinases displayed higher levels of chitinase specific activity and were more effective at inhibiting the growth of Rhizoctonia solani, Botrytis cinerea and Phytophthora citrophthora than the wild type. Transformants that overexpressed the chimeric chitinases possessed the highest specific chitinase and antifungal activities. The results confirm the importance of these endochitinases in the antagonistic activity of T. harzianum strains, and demonstrate the effectiveness of adding a CBD to increase hydrolytic activity towards insoluble substrates such as chitin-rich fungal cell walls.  相似文献

Insect cell cultures have been extensively utilised for means of production for heterologous proteins and biopesticides. Spodoptera frugiperda (Sf9) and Trichoplusia ni (High Five™) cell lines have been widely used for the production of recombinant proteins, thus metabolism of these cell lines have been investigated thoroughly over recent years. The Helicoverpa zea cell line has potential use for the production of a biopesticide, specifically the Helicoverpa armigera single-nucleocapsid nucleopolyhedrovirus (HaSNPV). The growth, virus production, nutrient consumption and waste production of this cell line was investigated under serum-free culture conditions, using SF900II and a low cost medium prototype (LCM). The cell growth (growth rates and population doubling time) was comparable in SF900II and LCM, however, lower biomass and cell specific virus yields were obtained in LCM. H. zea cells showed a preference for asparagine over glutamine, similar to the High Five™ cells. Ammonia was accumulated to significantly high levels (16 mM) in SF900II, which is an asparagine and glutamine rich medium. However, given the absence of asparagine and glutamine in the medium (LCM), H. zea cells adapted and grew well in the absence of these substrates and no accumulation of ammonia was observed. The adverse effect of ammonia on H. zea cells is unknown since good production of biologically active HaSNPV was achieved in the presence of high ammonia levels. H. zea cells showed a preference for maltose even given an abundance supply of free glucose. Accumulation of lactate was observed in H. zea cell cultures. We examined longevity, fecundity, and oviposition strategies ofEucelatoria bryani Sabrosky (Diptera: Tachinidae), a gregarious endoparasitoid ofHelicoverpa zea (Boddie) andHeliothis virescens (F.) (Lepidoptera: Noctuidae). Longevity of adult femaleE. bryani was not related to body size. In contrast to longevity, largerE. bryani females had greater potential fecundity than smaller females, as determined by the number of embryonated eggs present in the common oviduct. However, female parasitoid size did not affect primary clutch size (number of eggs deposited in a host). Because embryos in eggs located in the ovisac were larger than those located elsewhere in the common oviduct, maximum primary clutch size may be physiologically limited by the number of fully mature eggs a female has available at one time.E. bryani females adjusted primary clutch size in response to host size, for bothH. zea andH. virescens. This adjustment appears to be adaptive because females did not overexploit hosts by depositing more larvae than a host could support. Adult emergence was not related to host size. Although host weight positively influencedE. bryani progeny weight, increases in progeny size with host size were counterbalanced by increases in primary clutch size with host size.  相似文献

The effect of leaf shape variation on plant-herbivore interactions has primarily been studied from the perspective of host seeking behavior. Yet for leaf shape to affect plant-herbivore coevolution, there must be reciprocal effects of leaf shape variation on herbivore consumption and performance. We investigated whether alternative leaf morphs affected the performance of three generalist insect herbivores by taking advantage of a genetic polymorphism and developmental plasticity in leaf shape in the Ivyleaf morning glory, Ipomoea hederacea. Across four experiments, we found variable support for an effect of leaf shape genotype on insects. For cabbage loopers (Trichoplusia ni) and corn earworms (Helicoverpa zea) we found opposing, non-significant trends: T. ni gained more biomass on lobed genotypes, while H. zea gained more biomass on heart-shaped genotypes. For army beetworms (Spodoptera exigua), the effects of leaf shape genotype differed depending on the age of the plants and photoperiod of growing conditions. Caterpillars feeding on tissue from older plants (95 days) grown under long day photoperiods had significantly greater consumption, dry biomass, and digestive efficiency on lobed genotypes. In contrast, there were no significant differences between heart-shaped and lobed genotypes for caterpillars feeding on tissue from younger plants (50 days) grown under short day photoperiods. For plants grown under short days, we found that S. exigua consumed significantly less leaf area when feeding on mature leaves than juvenile leaves, regardless of leaf shape genotype. Taken together, our results suggest that the effects of leaf shape variation on insect performance are likely to vary between insect species, growth conditions of the plant, and the developmental stage and age of leaves sampled. Populations ofHelicoverpa (=Heliothis) zea (Boddie),Heliothis virescens (F.),Manduca sexta (L.) andM. quinquemaculata (Haw.) and their egg and larval parasitoids were sampled in field plots of the: insect-resistant wild tomato,Lycopersicon hirsutum f. glabratum C. H. Mull, accession PI 134417; susceptible commercial tomato cultivar ‘Better Boy’; F1 hybrid; and selected, moderately resistant backcross genotype. Densities ofH. zea andH. virescens eggs and small larvae were higher on resistant genotypes than on susceptible genotypes, but densities of large larvae were similar on all genotypes. Densities ofManduca spp. larvae were too low to permit similar analyses of the effects of plant genotype. Rates of egg parasitism byTrichogramma spp. andTelenomus sphingis (Ashmead) were reduced on insect-resistant genotypes. Rates of parasitism by the larval parasitoidsCampoletis sonorensis (Cameron) andCotesia congregata (Say) were reduced on resistant genotypes. No consistent effects on parasitism rates byCotesia marginiventris (Cresson) were observed and parasitism rates byCardiochiles nigriceps Viereck were unaffected.  相似文献

The extracellular proteases ofAspergillus nidulans are known to be regulated by carbon, nitrogen and sulphur metabolite repression. In this study, a mutant with reduced levels of extracellular protease was isolated by screening for loss of halo production on milk plates. Genetic analysis of the mutant showed that it contains a single, recessive mutation, in a gene which we have designatedxprE, located on chromosome VI. ThexprE1 mutation affected the production of extracellular proteases in response to carbon, nitrogen and, to a lesser extent, sulphur limitation. Three reversion mutations,xprF1, xprF2 andxprG1, which suppressxprE1, were characterised. BothxprF andxprG map to chromosome VII but the two genes are unlinked. ThexprF1, xprF2 andxprG1 mutants showed high levels of milk-clearing activity on medium containing milk as a carbon source but reduced growth on a number of nitrogen sources. Evidence is presented that thexprE1 andxprG1 mutations alter expression of more than one protease and affect levels of alkaline protease gene mRNA.  相似文献

We have used total genomic DNA as a probe to size-fractionated restriction enzyme digests of genomic DNA from a range ofTriticeae species from the generaLeymus Hochst.,Psathyrostachys Nevski, andHordeum L., and hybrids betweenHordeum andLeymus to investigate their taxonomic relationships. Genomic Southern hybridization was found to be an effective and simple way to assess the distribution and diversity of essentially species-specific and common, repetitive DNA sequences, and is hence especially useful in evolutionary studies. The DNA sequences ofH. vulgare seem to diverge substantially from those ofH. brachyantherum, H. lechleri, H. procerum, andH. depressum. The genome ofThinopyron bessarabicum shows little homology to those of theLeymus species investigated, confirming thatT. bessarabicum is not an ancestral genome inLeymus. Although the genomes ofLeymus andPsathyrostachys share substantial proportions of DNA sequences, they include divergent repeated sequences as well. Hybridization with a ribosomal DNA probe (pTa 71) showed that the coding regions containing structural genes encoding the 18 S, 5.8 S, and 26 S ribosomal RNA were conserved among the species investigated, whereas the intergenic spacer region was more variable, presenting different sizes of restriction fragments and enabling a classification of the species. The rye heterochromatin probe pSc 119.2 hybridized to DNA fromH. lechleri andT. bessarabicum, but not to DNA from the other species investigated.  相似文献

Accessory gland materials from Spodoptera frugiperda (J. E. Smith) and Heliothis zea (Boddie) (Lepidoptera: Noctuidae) contained kairomones that influenced the host recognition or acceptance behavior of Telenomus remus Nixon (Hymenoptera: Scelonidae) and Trichogramma pretiosum Riley (Hymeoptera: Trichogrammatidae), respectively. Material from S. frugiperda accessory glands stimulated ovipositor probing and drilling by female T. remus but not T. pretiosum. Accessory gland material from H. zea stimulated ovipositor probing and drilling by female T. pretiosum but not T. remus. An active material for T. remus is found in the 700 K protein fraction of accessory gland material from S. frugiperda. The oviposition behavior of the parasitoids is also discussed.
Résumé Cette étude a utilisé une technique voisine de celle employée par Strand & Vinson (1982), pour examiner les réactions de parasitoïdes à des billes de verre enduites de substances, afin de déterminer l'influence des sécrétions des glandes annexes (AGM) de Spodoptera frugiperda et Heliothis zea dans le repérage des hôtes par Telenomus remus et Trichogramma pretiosum.Les femelles de T. remus réagissaient plus aux billes enduites d'AGM de S. frugiperda qu'aux billes témois non enduites; celles de T. pretiosum réagissaient plus aux billes enduites d'AGM de H. zea qu'aux témoins. T. remus n'a pas réagi aux billes enduites d'AGM de H. zea, ni T. pretiosum à celles enduites d'AGM de S. frugiperda.Les poids moléculaires des protéines des glandes annexes de H. zea sont 1.100 K et 50 K, et ceux de S. frugiperda de 700 K et 30 K (Strand & Vinson, 1983a). T. remus a mieux réagi aux billes enduites de la fraction 700 K de S. frugiperda qu'a celles enduites de la fraction 30 K, aux deux fractions de H. zea et aux billes témoins. Les billes enduites de la fraction 1.100 K de H. zea ont provoqué aussi une réaction significative de T. remus. Les sécrétions des glandes annexes de S. frugiperda et de H. zea contiennent des kairomones qui interviennent dans le repérage des hôtes respectifs de T. remus et de T. pretiosum. Mais, il semble peu probable que ces substances soient seules responsables de la reconnaissance de l'hôte.

Microplitis demolitor Wilkinson, a solitary larval parasitoid of several species of Noctuidae, has been imported into the United States from Australia. A closely related species, Microplitis croceipes (Cresson), is native to the United States. The host-selection behavior of M. demolitor females is similar to that of M. croceipes in that it is stimulated by kairomones (including 13-methylhentriacontane) in the frass of Heliothis zea (Boddie) larvae, larval diet affects the kairomonal activity of the frass, and exposure to frass prior to release stimulates host-selection behavior. M. demolitor females also respond to frass from Trichoplusia ni (Hübner) larvae that had fed on Pink Eye Purple Hull Cowpea cotyledons.

Résumé Microplitis demolitor, un parasitoïde solitaire de plusieurs espèces de chenilles de noctuelles, a été introduit d'Australie aux Etats-Unis comme agent potentiel de contrôle biologique. Les fèces des chenilles d'Heliothis zea et Trichoplusia ni élevées sur des cotyledons de pois (Pink Eye Purple Hull Cowpea) stimulent les femelles à sélectionner un hôte, mais les fèces des chenilles des deux espèces élevées sur diète CSM ne possèdent pas cette activité. Un extrait hexanique des fèces des chenilles de H. zea élevées sur cotyledons de pois stimule également ce comportement, de même que le 13-methylhentriacontane, substance présente dans les fèces des chenilles de H. zea (Jones et al., 1971). Les femelles de M. demolitor exposées immédiatement avant leur lâcher en serre aux fèces des chenilles de H. zea, nourries de cotyledons de pois, parasitent un plus fort pourcentage (41.7%) de chenilles de H. zea que cells qui n'ont pas été exposées aux fèces (13.9%).Le comportement de sélection de l'hôte des femelles de M. demolitor est donc semblable à celui des femelles de Microplitis croceipes, un parasitoïde étroitement apparenté indigène des Etats-U-nis. La femelle, par exemple, est stimulée par une kairomone des fèces des larves de H. zea nourries sur des cotyledons de pois (incluant le 13-methyl-hentriacontane), la diète affecte l'activité kairomonale des fèces, et une exposition préalable aux fèces avant le lâcher stimule le comportement de sélection de l'hôte.La stimulation du comportement de sélection de l'hôte, au moment du lâcher peut être utilisée pour améliorer l'efficacité des lâchers des parasitoïdes dans les programmes de contrôle biologique.

Sequences of the internal transcribed spacer region 1 (ITS1) of the ribosomal DNA were used to determine the phylogenetic relationships of species ofTrichodermasect.Pachybasium.To this end, 85 strains—including all the availableex-type strains—were analyzed. Parsimony analysis demonstrated that the section is nonmonophyletic, distributing the 85 strains among three main groups that were supported by bootstrap values. Group A comprises two clades (A1 and A2), with A1 includingT. polysporum, T. piluliferum,andT. minutisporum,while A2 includedT. hamatum, T. pubescens,andT. strigosumin addition to species previously included in sect.Trichoderma(i.e.,T. viride, T. atroviride,andT. koningii). Theex-type strain ofT. fasciculatumformed a separate branch basal to clade A. Clade B contained the sect.PachybasiummembersT. harzianum, T. fertile, T. croceum, T. longipile, T. strictipile, T. tomentosum, T. oblongisporum, T. flavofuscum, T. spirale,and the anamorphs ofHypocrea semiorbisandH. cf. gelatinosa.Sequence differences among clades A1, A2, and B were in the same order of magnitude as between each of them andT. longibrachiatum,which was used as an outgroup in these analyses. Sequence differences within clades A1, A2, and B were considerably smaller: in some cases (i.e.,T. virensandT. flavofuscum; T. strictipileandH. cf. gelatinosa), the ITS1-sequences were identical, suggesting conspecifity. In other cases (e.g.,T. crassumandT. longipile; T. harzianum, T. inhamatum, T. croceum, T. fertile,andH. semiorbis; T. hamatumandT. pubescens;andT. viride, T. atroviride,andT. koningii) differences were in the range of 1–3 nt only, suggesting a very close phylogenetic relationship.
