We report a straightforward and widely applicable cryopreservation method for Lilium shoot tips. This method uses adventitious shoots that were induced from leaf segments cultured for 4 weeks on a shoot regeneration medium containing 1 mg/l α-naphthaleneacetic acid and 0.5 mg/l thidiazuron. Shoot tips (1.5–2 mm in length) including 2–3 leaf primordia were precultured on Murashige and Skoog (MS; 1962) medium with 0.5 M sucrose for 1 day and then treated with a loading solution containing 0.4 M sucrose and 2 M glycerol for 20 min, followed by a Plant Vitrification Solution 2 (PVS2) treatment for 4 h at 0 °C. Dehydrated shoot tips were transferred onto 2.5 µl PVS2 droplets on aluminum foil strips, prior to a direct immersion into liquid nitrogen for 1 h. Frozen shoot tips were re-warmed in MS medium containing 1.2 M sucrose for 20 min at room temperature, followed by post-thaw culture for shoot regrowth. Shoot regrowth levels ranged from 42.5 % for L. longiflorum × Oriental ‘Triumphator’ to 87.5 % for L. Oriental hybrid ‘Siberia’, with a mean shoot regrowth level of 67.1 % across the six diverse Lilium genotypes tested. Histological observations found that the survival patterns were similar in cryopreserved shoot tips of ‘Triumphator’ and ‘Siberia’. Assessments using inter-simple sequence repeat markers found no differences in regenerants recovered from the control stock cultures and from cryopreserved shoot tips in ‘Triumphator’ and ‘Siberia’. This Lilium droplet-vitrification cryopreservation method is efficient, simple and widely applicable for the long-term conservation of lily genetic resources.  相似文献

ABSTRACT

Individuals with an evening chronotype are at increased risk of experiencing emotional problems, including depressive symptoms. However, the mechanisms underlying these associations remain unclear. The present study aimed to determine whether poor sleep quality, substance use and cognitive emotion regulation difficulties – which have been implicated in the etiology of depression – mediate the relationship between chronotype and depressive symptoms in a student sample, which was assessed cross-sectionally and after 1 year. A total of 742 Dutch students (75% women, mean age 21.4 ± 2.9 years) completed the Quick Inventory of Depressive Symptomatology, the Morningness-Eveningness Questionnaire, the Pittsburgh Sleep Quality Index, a questionnaire assessing alcohol, caffeine, tobacco and cannabis use, the Cognitive Emotion Regulation Questionnaire and the Behavioral Inhibition/Activation Scale. A subsample (n = 115) was assessed 1 year later with the same questionnaires. Cross-sectional analyses showed that evening chronotype was associated with more depressive symptoms, adjusted for age and gender (β = ?0.082, p = 0.028). The relationship between eveningness and depressive symptoms was mediated by sleep quality, alcohol consumption and the cognitive emotion regulation strategies of self-blame and positive reappraisal. In longitudinal analyses, eveningness at baseline predicted more depressive symptoms at follow-up, adjusted for age and gender (β = ?0.29, p = 0.002); after additional adjustment for baseline depressive symptoms, chronotype remained a significant predictor of depressive symptoms at T2 (β = ?0.16, t = ?2.01, p = 0.047). Only poor sleep quality at follow-up was a significant mediator of this relationship. Even though the effect is small in terms of explained variance, eveningness is related to depressive symptoms and this relationship is mediated by poor sleep quality, also in a prospective design. Self-blame and reduced positive reappraisal are correlated with eveningness. Further research is needed to assess the efficacy of chronotherapeutic interventions for the prevention of depression, in addition to sleep education and cognitive approaches.  相似文献

An aminopeptidase was purified from Aspergillus sojae X–816. The molecular weight of the enzyme was estimated to be 220,000. The isoelectric point was at pH 5.3. The optimum pH for l-leucylglycylglycine was 7.5. The enzyme was stable up to 37°C against temperature treatment for 15 min. Some chelating agents inhibited the enzyme activity. The Km value for l-leucylglycylglycine at pH 7.5 and 37°C was 45 mm. The Km value for l-leucyl-β-naphthylamide at pH 7.0 and 37°C was 2.2 mm.  相似文献

The laboratory rat, Rattus novegicus, is a major model system for physiological and pathophysiological studies, and since 1966 more than 422,000 publications describe biological studies on the rat (NCBI/Medline). The rat is becoming an increasingly important genetic model for the study of specific diseases, as well as retaining its role as a major preclinical model system for pharmaceutical development. The initial genetic linkage map of the rat contained 432 genetic markers (Jacob et al. 1995) out of 1171 developed due to the relatively low polymorphism rate of the mapping cross used (SHR × BN) when compared to the interspecific crosses in the mouse. While the rat genome project continues to localize additional markers on the linkage map, and as of 11/97 more than 3,200 loci have been mapped. Current map construction is using two different crosses (SHRSP × BN and FHH × ACI) rather than the initial mapping cross. Consequently there is a need to provide integration among the different maps. We set out to develop an integrated map, as well as increase the number of markers on the rat genetic map. The crosses available for this analysis included the original mapping cross SHR × BN reciprocal F2 intercross (448 markers), a GH × BN intercross (205 markers), a SS/Mcw × BN intercross (235 markers), and a FHH/Eur × ACI/Hsd intercross (276 markers), which is also one of the new mapping crosses. Forty-six animals from each cross were genotyped with markers polymorphic for that cross. The maps appear to cover the vast majority of the rat genome. The availability of these additional markers should facilitate more complete whole genome scans in a greater number of strains and provide additional markers in specific genomic regions of interest. For hydrogel patches, the laboratory tests could not fully reveal the existing problems of full scale of industrial production, and there are few studies about the preparation technique for the industrial manufacturing process of hydrogel patches. So, the purpose of this work was to elucidate the effects of mainly technological operation and its parameters on the performance of hydrogel patches at the industrial-scale production. The results revealed the following: (1) the aqueous phase was obtained by polyvinylpyrrolidone (PVP) along with tartaric acid dissolved in purified water, then feeding this into a vacuum mixer as a whole in one batch, thus extended the crosslinking reaction time of hydrogel paste (matrix) and allowed the operation of coating/cutting-off to be carried out easily, and there was no permeation of backing layer; (2) the gel strength of the hydrogel patches increased with the increase of working temperature, however, once the temperature exceeded 35 ± 2 °C, the hydrogel paste would lose water severely and the resultant physical crosslinking structure which has lower gel/cohesive strength would easily bring gelatinization/residues during application; (3) the relative humidity (RH) of the standing-workshop was dynamically controlled (namely at 35 ± 2 °C, keeping the RH at 55 ± 5% for 4 days, then 65 ± 5% for 2 days), which would make patches with satisfactory characteristics such as better flexibility, higher adhesive force, smooth flat matrix surface, and without gelatinization/residues and warped edge during the using process; (4) the aging of the packaged hydrogel patches was very sensitive to storage temperature, higher temperature, higher gel strength and lower adhesiveness. The storage temperature of 10 ± 2 °C could effectively prevent matrix aging and adhesion losing, which would also facilitate the expiration date of patches extended obviously. In conclusion, this work provides an optimized and feasible preparation technique for the industrial production of the hydrogel patches and establishes the hydrogel patches as a novel carrier for transdermal drug delivery.  相似文献

We performed a Landsat 5-TM derived normalized difference vegetation index (NDVI) analysis in a semi-arid watershed (2700 km2) in the Andes of southern Peru from 1985 to 2010. There, pastoralists rely on wetlands (bofedales) particularly during dry season months and in drought. We calculated annual dry season NDVI for 20 of the 26 years from 1985 to 2010 and used the mean to delineate wetlands in the watershed. To investigate the trends in NDVI, a multiple regression model with the covariates precipitation, temperature, Julian day, and year of image acquisition was performed on each cell (three million individual regressions). Results indicate there is a modest increase in NDVI for the majority of cells (81 %) in the watershed. Approximately 30 % of wetland areas display a decrease in NDVI. Dry season NDVI is moderately correlated with wet season precipitation (R 2 = 0.56, p < 0.05) but absent a trend in precipitation, NDVI trends are not explained by this variable. Changes in land management may result in more intensive use of wetlands, causing the decreasing vegetation trends in some locations.  相似文献

Male, albino, Sprague-Dawley rats were sacrificed by cervical separation. Segments of jejunum were excised, everted and examined with the electron microscope. Examination of tissue fixed immediately after eversion revealed the following changes as compared to non-everted segments fixed insitu and invitro: 1) an increase in the length of microvilli from (mean ± S. E.) 0.991 ± 0.011μ for normal tissue to 1.389 ± 0.023μ for everted tissue, 2) an increase in width of microvilli from (mean ± S. E.) 0.089 ± 0.001μ for normal tissue to 0.097 ± 0.001μ for everted tissue, 3) an increase in length and number of lateral membrane interdigitations, and 4) the appearance of intercellular “lakes” in the lateral spaces. The above changes are in those structures hypothesized to be involved with salt and water transport across epithelia and may reflect altered transport rates invitro as compared to invivo.  相似文献

In order to reduce the socio-economic burden induced by osteoporotic hip fractures, finite element models have been evaluated as an additional diagnostic tool for fracture prediction. For a future clinical application, the challenge is to reach the best compromise between model relevance and computing time. Based on this consideration, the current study focused on the development and validation of a subject-specific FE-model using an original parameterised generic model and a specific personalization method. A total of 39 human femurs were tested to failure under a quasi-static compression in stance configuration. The corresponding FE-models were generated and for each specimen the numerical fracture load (F FEM) was compared with the experimental value (F EXP), resulting in a significant correlation (F EXP = 1.006 F FEM with r 2 = 0.87 and SEE = 1220 N, p < 0.05) obtained with a reasonable computing time (30 mn). Further in vivo study should confirm the ability of this FE-model to improve the fracture risk prediction.  相似文献

An automated analytical system has been used to determine the amount of oxytetracycline in pharmaceutical products and fermentation broth at a level of 0.1mg/mL. The method is based on the formation of a colour complex between the oxytetracycline and ammonium molybdate in acid media at 80°C. The effect of temperature, concentration of HCl and ammonium molybdate upon the stability of the complex was determined. Standard error of difference is 0.43% for pharmaceutical products and the fermentation broth.  相似文献

An empirical model of carbon fluxes in Russian tundra   总被引:1,自引:0,他引:1  
This study presents an empirical model based on a GIS approach, which was constructed to estimate the large‐scale carbon fluxes over the entire Russian tundra zone. The model has four main blocks: (i) the computer map of tundra landscapes; (ii) data base of long‐term weather records; (iii) the submodel of phytomass seasonal dynamics; and (iv) the submodel of carbon fluxes. The model uses exclusively original in situ diurnal CO2 flux chamber measurements (423 sample plots) conducted during six field seasons (1993–98). The research sites represent the main tundra biome landscapes (arctic, typical, south shrub and mountain tundras) in the latitudinal diapason of 65–74°N and longitudinal profile of 63°E?172°W. The greatest possible diversity of major ecosystem types within the different landscapes was investigated. The majority of the phytomass data used was obtained from the same sample plots. The submodel of carbon fluxes has two dependent [GPP, Gross Respiration (GR)] and several input variables (air temperature, PAR, aboveground phytomass components). The model demonstrates a good correspondence with other independent regional and biome estimates and carbon flux seasonal patterns. The annual GPP of Russian tundra zone for the area of 235 × 106 ha was estimated as ?485.8 ± 34.6 × 106 tC, GR as +474.2 ± 35.0 × 106 tC, and NF as ?11.6 ± 40.8 × 106 tC, which possibly corresponds to an equilibrium state of carbon balance during the climatic period studied (the first half of the 20th century). The results advocate that simple regression‐based models are useful for extrapolating carbon fluxes from small to large spatial scales.  相似文献

An X-ray study has been made of the synthetic polypeptide poly-L -ornithine hydrobromide to investigate whether, like the chemically related polypeptides poly-L -lysine and poly-L -arginine hydrochlorides, it can undergo conformational changes merely from variations in its degree of hydration. X-ray powder and fiber photographs of specimens with from half up to about three molecules of water per ornithine residue show features that suggest a “cross-β-pleated-sheet” structure. Each pleated sheet is formed from parallel chains and the sheets are piled up along the b axis. The spacings, which do not vary appreciably with hydration, can be satisfactorily indexed in terms of an orthogonal unit cell with a = 4.60 Å, b = 30.2 Å, and c = 6.64 Å. These dimensions are shown by models to be compatible with the proposed structure. Removal of the last half molecule of water results in a very diffuse pattern but on rehydration the sharp pattern reappears. Specimens containing four to nine molecules of water per residue show a quite different pattern. Reflections other than equatorial are absent in oriented diagrams except for a 5.4 Å diffuse streak across the meridian which is suggestive of an α-helical structure. Increasing the relative humidity from 86% to about 100% causes the a axis of the hexagonal unit cell to increase from 14.7 Å to 15.3 Å. On drying, the β structure reappears once again. These conformational changes are very similar to those observed in poly-L -lysine hydrochloride except that the latter shows a more stable α-helical form. This difference may be explained in terms of stabilizing hydrophobic interactions between side chains, since ornithine has a shorter side chain than lysine.  相似文献

Variation in the amount of nuclear DNA, the C-value, does not correlate with differences in morphological complexity. There are two classes of explanations for this observation, which is known as the ''C-value paradox''. The quantity of DNA may serve a ''nucleotypic'' function that is positively selected. Alternatively, large genomes may consist of junk DNA, which increases until it negatively affects fitness. Attempts to resolve the C-value paradox focus on the link between genome size and fitness. This link is usually sought in life history traits, particularly developmental rates. I examined the relationship among two life history traits, egg size and embryonic developmental time and genome size, in 15 species of plethodontid salamanders. Surprisingly, there is no correlation between egg size and developmental time, a relationship included in models of life history evolution. However, genome size is positively correlated with embryonic developmental time, a result that is robust with respect to many sources of variation in the data. Without information on the targets of natural selection it is not possible with these data to distinguish between nucleotypic and junk DNA explanations for the C-value paradox.  相似文献

An x-ray study has been made of polyarginine hydrochloride to investigate whether, like polylysine hydrochloride, it can undergo conformational changes merely from variations in the degree of hydration. X-ray powder and fiber photographs of specimens containing up to about five molecules of water per arginine residue show features characteristic of α-helical structures including a 5.4-Å layer line and a meridional 1.5-Å reflection. Increasing the water content from 1/2 to 61/2 molecules per residue causes the a axis of the hexagonal unit cell to increase from 14.4 Å to 15.8 Å, with no appreciable change in the 27.0 Å c axis. Removal of the last half molecule of water results in a very diffuse α pattern, but on rehydration the sharp pattern reappears. Specimens containing five to twenty water molecules per residue show quite a different pattern, the spacing of which do not vary appreciably with hydration. This pattern includes a meridional 3.4-Å reflection, a feature commonly shown by β structures, and indeed all the reflections can be satisfactorily indexed in terms of a monoclinic unit cell with a = 9.26 Å, b = 22.05 Å, c = 6.76 Å, and γ = 108.9°. These dimensions are shown by models to be compatible with a β pleated-sheet structure.  相似文献

The “attached cultivation” method of microalgae in which the wet paste of algal biomass is attached onto supporting materials to form an immobilized biofilm layer, and the culture medium is supplied to this layer to provide nutrients and moisture for growth was highly efficient in biomass production and represents a promising technology to improve the biofuel industry. To optimize the nitrogen supply strategy for this attached cultivation method, the growth and total lipids accumulation properties for the green alga Aucutodesmus obliquus with this method were studied under different quantities of nitrogen source and different volumes of aqueous medium that continuously circulated inside the photobioreactor. Results showed that, compared with medium volume, the nitrogen quantity was a stronger factor affecting the growth and total lipid accumulation. An optimized nitrogen supply strategy for the attached cultivation of A. obliquus is proposed as circulating ca. 60 L of BG-11 medium containing 1/10 of nitrate concentration for 1 m2 of cultivation surface. With this strategy, the attached A. obliquus accumulated biomass and total lipids simultaneously and obtained a high triacylglyceride productivity of 2.53 g m?2 day?1 in 7 days under subsaturated illumination of 100 μmol photons m?2 s?1. The water usage of 60 L m?2 was potentially decreased to <2 L m?2 if the nutrient supply was further improved. Dissolving the nitrogen source in small volume was the best way to efficiently utilize the nitrogen source with minimum of waste.  相似文献

An exact test based on the unconditional distribution of a test statistic for the Hardy-Weinberg equilibrium is introduced. This test is usually more powerful and requires less extensive tabulations compared with the (ordinary) exact conditional test. Several methods for the isolation of Micromonospora from soil samples have been developed; however, it is unclear whether these methods are optimal for estuarine samples. In this study, we optimized the conditions of a wet-heat method for the selective isolation of Micromonospora from estuarine sediments. Sediments were collected from the Arakawa River (estuarine sediments) and Tokyo Bay (marine sediments). Sediment samples were wet-heated at 45, 55, or 65 °C for 30 min and then incubated at 27 °C for 3 weeks. After incubation, most of the actinomycete colonies were macroscopically determined to be of the genus Micromonospora or Streptomyces. In contrast to the treatment at 55 °C, treatment at 65 °C drastically reduced the number of Streptomyces colonies but increased the number of Micromonospora colonies from the estuarine sediments. This procedure allowed us to grow cultures that were composed of more than 90 % Micromonospora. In addition, treatment at 65 °C did not affect the diversity of Micromonospora species compared with treatment at 55 °C. These results indicate that the wet-heat method, which involves pre-treating the sediment at 65 °C for 30 min, is a very simple and effective method for the selective enrichment of a large number of diverse Micromonospora from estuarine sediments. Our results may lead to the isolation of new Micromonospora species, which produce novel bioactive compounds, from different estuarine sediments.  相似文献

Enzymes hydrolysing the exopolysaccharides of Xanthomonas campestris and related species (xanthan) have been obtained from a Bacillus species isolated by enrichment culture. Growth on xanthan induced a number of enzymes acting on the xanthan molecule. These included one or more β-glucanohydrolases and β-glucosidases, together with mannosidases. The former activities were also present in cultures grown in the presence of laminaran or scleroglucan, but not in simple synthetic media with glucose as substrate. Partial purification of the enzymes active on glucans was achieved by ammonium sulphate precipitation and chromatography on DEAE-sepharose and CM-sepharose. The specificity of the β-glucosidase and the β-glucanohydrolase were investigated. Several β-glucans were hydrolysed to glucose and disaccharides, but there was no activity against β→ 6 linked polymers, cellulose azure or microcrystalline cellulose. Carboxymethylcellulose was hydrolysed, as were laminaran, scleroglucan and pachyman. Activity was greater against the β→ 4 linked glucans than against the β→ 3 linked glucans tested. As periodate-oxidized laminarin was also hydrolysed, it was concluded that the glucanohydrolase acted as an endo enzyme. The β-glucosidase had a pH optimum at about 8–2 and a temperature optimum at 45°C; it showed higher activity against o -nitrophenyl-D-glucopyranoside, cellobiose, trehalose and sophorose than against gentibiose.  相似文献

Microalgae have vast potential as a sustainable and scalable source of biofuels and bioproducts. However, algae dewatering is a critical challenge that must be addressed. Ultrasonic settling has already been exploited for concentrating various biological cells at relatively small batch volumes and/or low throughput. Typically, these designs are operated in batch or semicontinuous mode, wherein the flow is interrupted and the cells are subsequently harvested. These batch techniques are not well suited for scaleup to the throughput levels required for harvesting microalgae from the large‐scale cultivation operations necessary for a viable algal biofuel industry. This article introduces a novel device for the acoustic harvesting of microalgae. The design is based on the coupling of the acoustophoretic force, acoustic transparent materials, and inclined settling. A filtration efficiency of 70 ± 5% and a concentration factor of 11.6 ± 2.2 were achieved at a flow rate of 25 mL·min?1 and an energy consumption of 3.6 ± 0.9 kWh·m?3. The effects of the applied power, flow rate, inlet cell concentration, and inclination were explored. It was found that the filtration efficiency of the device is proportional to the power applied. However, the filtration efficiency experienced a plateau at 100 W L?1 of power density applied. The filtration efficiency also increased with increasing inlet cell concentration and was inversely proportional to the flow rate. It was also found that the optimum settling angle for maximum concentration factor occurred at an angle of 50 ± 5°. At these optimum conditions, the device had higher filtration efficiency in comparison to other similar devices reported in the previous literature. © 2014 American Institute of Chemical Engineers Biotechnol. A method is described for the analysis of α-tocopherol in sheep liver, employing a direct attack on the tissue with alcoholic alkali in the presence of pyrogallol and hydroquinone. The alkali treatment is rapid and avoids drying, grinding, and solvent extraction. Combined with two-dimensional paper chromatography, it eliminates time-consuming freezing and column chromatographic techniques. Mean ± SE recoveries of α-tocopherol alone, or mixed with liver tissue, were 97.1 ± 0.05% and 87.1 ± 4.1%, respectively. The mean ± SE levels of α-tocopherol in the liver of sheep fed dry rations were 1.0 ± 0.1 μg/g, and in that of sheep fed green pasture, 18.5 ± 1.4 μg/g.  相似文献
