The gills of Paragnetina media function as true respiratory organs. The animals with all the thoracic gill removed did not respire more than a few minutes. The respiratory rates showed a correlation to the body weight.  相似文献

New rudimentary (r) mutants have been isolated following mutagenesis with ethyl methanesulfonate (rLE), ICR–170 (rLI) and X rays (rLX). From wing phenotype measurements on homoallelic females, it has been shown that the rLE mutant series includes several leaky alleles, as well as alleles that produce moderate and strong r phenotypes. All of the tested rLI alleles yielded strong r phenotypes in homoallelic females, whereas the rLX series was found to include both moderate and strong alleles. Based on allele complementation for the wing phenotype, it was found that all three mutant series include both complementing and noncomplementing alleles, but the relative frequencies of these two types of alleles differ considerably among the three series. Complementing alleles comprise most of the rLE mutant series (19 of 25) and almost one-half of the rLX series (five of 12), while only one of 16 rLI mutants is a complementing allele. Data from enzyme assays of mutants mostly support the direct correlation of genetic complementation units with the activities of the first three enzymes in the de novo pyrimidine biosynthetic pathway. All of these findings are discussed in light of evidence that these three enzymes are contained within a trienzyme complex in animals. We conclude that the available genetic evidence supports the contention that the trienzyme complex is encoded by a single mRNA.  相似文献

We measured gill slit fluctuating asymmetry (FA), a measure of developmental noise, in adults of three invertebrate deuterostomes with different feeding modes: the cephalochordate Branchiostoma floridae (an obligate filter feeder), the enteropneusts Protoglossus graveolens (a facultative filter feeder/deposit feeder) and Saccoglossus bromophenolosus (a deposit feeder). FA was substantially and significantly low in B. floridae and P. graveolens and high in S. bromophenolosus. Our results suggest that the gills of species that have experienced a relaxation of the filter feeding trait exhibit elevated FA. We found that the timing of development of the secondary collagenous gill bars, compared to the primary gill bars, was highly variable in P. graveolens but not the other two species, demonstrating an independence of gill FA from gill bar heterochrony. We also discovered the occasional ectopic expression of a second set of paired gills posterior to the first set of gills in the enteropneusts and that these were more common in S. bromophenolosus. Moreover, our finding that gill slits in enteropneusts exhibit bilateral symmetry suggests that the left‐sidedness of larval cephalochordate gills, and the directional asymmetry of Cambrian stylophoran echinoderm fossil gills, evolved independently from a bilaterally symmetrical ancestor.  相似文献

Gill morphology, traditionally, has played an important role in attempts to reconstruct the phylogenetic history of the Crustacea Decapoda. We examined the gills of dendrobranchiate shrimps (Crustacea, Decapoda, Dendrobranchiata) to test the assumption that all members of the clade have gills that are “dendrobranchiate” (highly branching) in form, from whence the taxon name Dendrobranchiata comes. Currently, the Dendrobranchiata consists of two superfamilies and seven families. Specimens from two genera in each of the known families were examined using light and scanning electron microscopy. Members of the family Luciferidae, all of which lack gills as adults, were not examined. Only one genus was examined for the Penaeidae (because they have been the subject of numerous previous studies) and Sicyoniidae (a monogeneric family). All gills examined have secondary branches that are further subdivided, conforming to existing and rather broad definitions of dendrobranchiate gills. Families with “typical” dendrobranchiate gills, which consist of curved secondary branches that in turn bear branched (dendritic) tubular tertiary elements on their distal surfaces, include the Penaeidae, Aristeidae, and Solenoceridae. In other families, secondary and tertiary gill elements are sometimes quite flattened, and the tertiary elements are not dendritic, giving the gill a distinctly non-dendrobranchiate appearance. Flattened biserial secondary branches and their flattened tertiary elements are particularly obvious in gills of the monogeneric family Sicyoniidae (Sicyonia). Within the family Sergestidae, gills of the genus Sergestes are unusual in having secondary branches that arise from the main gill axis in an alternating pattern; these gills also have distinctly oval tertiary elements that are not further subdivided and are directed basally rather than distally. Another sergestid genus, Petalidium, displays gills that differ from those of Sergestes; in Petalidium the secondary branches also come off the main gill axis in an alternating pattern, but these branches are more widely spaced and have relatively larger and broader tertiary elements when compared with gills of Sergestes. The family Benthesicyemidae also contains species with different gill types; Gennadas is shown to have flattened, plate-like tertiary elements, whereas Benthesicyemus has more typical dendrobranchiate gills. The significance of this variation in gill morphology within families and within the Dendrobranchiata as a whole is unclear at this point; rearrangements of the currently accepted phylogeny and resulting classification based solely on gill morphology are not recommended at this time.  相似文献

The gills of the air-breathing estuarine goby,Pseudapocryptes lanceolatus, are reduced owing to the development of a specialized organ of O2 uptake from air. In the first gill arch, the filaments of the outer hemibranch are reduced to nearly one-half in comparison to those of its inner hemibranch. A smaller number of secondary lamellae per mm (27.6) occurring on one side of the gill filament reduces the gill surface area. A bilogarithmic plot of the gill area and the body weight indicates a curve with two significantly different components, one (b = 0.924) related to the fish weighing up to 6 g and the other (b = 0.405) to the fish weighing 8 g and above.  相似文献

Mutants of simian virus 40 (SV40) with base substitutions at or near the origin of replication of the viral genome have been constructed by bisulfite mutagenesis at the BglI restriction site of SV40 DNA, followed by transfection of cells with the BglI-resistant (BglIr) DNA so generated. Based on plaque morphology at different temperatures, the resulting BglIr mutants could be classified into four-groups. Class I mutants (designated ar for “altered restriction”) were indistinguishable from wild-type SV40; class II mutants (designated shp for “sharp plaque”) produced small, sharp-edged plaques; class III mutants (designated sp for “small plaque”) produced small plaques at 32 °C, 37 °C and 40 °C; and class IV mutants (designated cs for “cold sensitive”) produced small plaques at 32 °C and wild-type plaques at 37 °C and 40 °C. That the altered plaque morphology of sp and cs mutants was related to mutation at the BglI restriction site was demonstrated by co-reversion to wild-type of the plaque phenotype and BglI sensitivity. The nucleotide sequence around the original BglI site was determined in the DNA from one mutant of each class. In each case a different base-pair substitution was found, at a site outside sequences coding for SV40 proteins. When rates of replication of mutant DNAs were measured during productive infection, ar mutant DNA was synthesized at a rate comparable to that of wild-type SV40 DNA, shp mutant DNA was made at a rate exceeding that of wild-type, sp mutant DNA was synthesized at a lower rate than that of wild type. and cs mutant DNA synthesis was reduced at 32 °C, but about the same as the wild-type rate at 40 °C. These patterns of mutant DNA synthesis were unaltered in cells co-infected with mutant and wild-type virus, i.e. the defects in DNA synthesis were not trans-complementable. We conclude that the defective mutants have single base-pair changes in a cis element that determines the rate of viral DNA replication, presumably within the origin signal itself.  相似文献

Summary The levels of Na+, K+ ATPase were measured in gills fromPlatichthys flesus adapted to seawater and freshwater using a variety of experimental techniques. Na+, K+ ATPase was assayed directly in crude gill homogenates,3H-ouabain binding was determined in isolated, perfused gills and ouabainsensitive oxygen consumption measured in sliced gill filaments. These experimental approaches all failed to show any difference in Na+, K+ ATPase activity or in enzyme turnover rate in gills from seawateradapted and freshwater-adaptedPlatichthys. The results are discussed in terms of the marine origin of the flounder and the energetic demands of ion regulation in euryhaline fish.  相似文献

The mutation wingless produces a homeotic transformation in which the distal structures (appendages) of both the wing and haltere discs are replaced by a duplication of the proximal structures (thorax). However, not all of the mutant discs show mutant phenotype; some of them differentiate normal appendages. Gynandromorph and clonal analyses suggest that the phenotype does not result from massive cell death followed by regeneration and/or duplication. We conjecture that the mutant phenotype is caused by a specific failure in the process of compartmentalization. In contrast to other homeotic mutants, wingless is not cell autonomous; that is, mutant clones show wildtype phenotype when produced in wildtype wings.  相似文献

Programmed cell death often depends on generation of reactive oxygen species, which can be detoxified by antioxidative enzymes, including catalases. We previously isolated catalase-deficient mutants (cat2) in a screen for resistance to hydroxyurea-induced cell death. Here, we identify an Arabidopsis thaliana hydroxyurea-resistant autophagy mutant, atg2, which also shows reduced sensitivity to cell death triggered by the bacterial effector avrRpm1. To test if catalase deficiency likewise affected both hydroxyurea and avrRpm1 sensitivity, we selected mutants with extremely low catalase activities and showed that they carried mutations in a gene that we named NO CATALASE ACTIVITY1 (NCA1). nca1 mutants showed severely reduced activities of all three catalase isoforms in Arabidopsis, and loss of NCA1 function led to strong suppression of RPM1-triggered cell death. Basal and starvation-induced autophagy appeared normal in the nca1 and cat2 mutants. By contrast, autophagic degradation induced by avrRpm1 challenge was compromised, indicating that catalase acted upstream of immunity-triggered autophagy. The direct interaction of catalase with reactive oxygen species could allow catalase to act as a molecular link between reactive oxygen species and the promotion of autophagy-dependent cell death.  相似文献

SYNOPSIS Bimodal breathing crustaceans, while representing astage in the transition from the aquatic to terrestrial habitat,also constitute a distinct group that can be characterized bymorphological, behavioral, and physiological traits. Morphologically,this group displays reduced gill surface area and enlarged branchialchambers. The lining of the branchial chambers, the branchiostegites,also has increased surface area and is highly vascularized.The branchiostegites can be smooth, cutaneous epithelia, orthey can have more complex evaginations or invaginations tofurther increase surface area. In addition, the thickness ofthe branchiostegal epithelium is greatly reduced, compared tothat in the gills, thus minimizing the diffusion distance betweenair and hemolymph. These animals maintain a store of water inthe branchial chamber that covers the gills and allows for simultaneousgas exchange with two media (air and water). There is also apartitioning of gas exchange, with oxygen uptake occurring preferentiallyfrom air across the branchiostegites, and carbon dioxide excretionoccurring across the gills into the branchial water. One criticalfactor that appears to separate bimodal breathing crustaceansfrom fully terrestrial, exclusively air-breathers is the abilityof the latter group to excrete CO2directly into air across thegills and branchiostegites. It is suggested that the incorporationof the enzyme carbonic anhydrase into the membrane fractionof the branchiostegites may have been one of the key molecularevents which allowed for pulmonary CO2 excretion into air.  相似文献

Individual variations in the phospholipid compositions of the organs of marine invertebrates were investigated in the representatives of three phyla: Arthropoda [decapods: Paralithodes camtschaticus (Tilesius, 1815) and Erimacrus isenbeckii (Brandt, 1848)]; Echinodermata [starfishes: Distolasterias nipon (Doderlein, 1902) and Evasterias echinosoma Fisher, 1926]; and Tunicata [ascidians: Halocynthia aurantium (Pallas, 1787), H. roretzi (Drasche, 1884), and Styela clava Herdman, 1881]. The specificity of individual phospholipid variation was shown to be related to the systematic status and functional properties of the organs of marine invertebrates. Phosphatidylcholine and phosphatidylethanolamine were the least variable. Low variation of phospholipid composition was found in the liver, legs, and gills of decapods; in the liver of starfishes; and in the gonads and skin-muscular and gill sacs of ascidians. The contents of ceramide aminoethylphosphonate (CAEP) and phosphatidylglycerol (PG) showed the highest variation. These phospholipids were found but not in all individuals or organs studied. After 7 days of starvation, CAEP completely disappeared from the organs of the animals; thus, we suggest its exogenous origin.  相似文献

1. Gene ? is a recessive lethal factor found in the white strain of axolotls. Animals heterozygous for the gene are phenotypically normal. When mated with each other they give offspring 25% of which exhibit the lethal effects of the gene. 2. The ?/? homozygotes develop normally to an advanced embryonic stage (Harrison stage 40) before the effects of the gene are first manifested. They then come to display a characteristic combination of abnormalities, including a disproportionately small head, small and poorly developed eyes, abnormal poorly developed gills, undifferentiated limb buds, and reduced overall growth rate. They may feed briefly, but soon stop and invariably die within a few weeks of the time of hatching. 3. The action of gene ? has been analyzed by parabiosing mutant and normal embryos, and by grafting various organ primordia reciprocally between mutant and normal embryos. Parabiosis to normal embryos fails to correct the abnormalities of the mutants, although their survival may be somewhat prolonged. Grafts of mutant organ primordia (eye, limb, gill, pronephros, gonad, head) also invariably fail to show improved development or to survive on normal hosts; normal organ primordia develop normally on mutant hosts so long as the mutant survives. These experiments indicate that gene ? is a recessive autonomous cell lethal affecting all of the organ systems during late embryonic and early larval development.  相似文献

A direct screening technique was devised in which applications of a juvenile hormone analogue (JHA) (RO-20-3600; ENT No. 70357; 6,7-Epoxy-3-methyl-7-ethyl-1-(3,4-[methyl-enedioxy]-(phenoxy)-2-cis/trans-octene), Hoffman-LaRoche) to the food resulted in the death of all the F-1 descendants of mutagenized Drosophila melanogaster males with the exception of those animals which possessed sexlinked mutations affecting the metabolism of JHA. These exceptions would comprise (a), those mutant animals which possess any resistance to the lethal effects of the JHA (JHR mutants) and/or (b), those mutant animals which have a defective JH metabolism and would normally die but are rescued by the exogenous application of JHA (JH? mutants). Approximately 95, 850 mutagenized chromosomes were tested in this screen, 99,6% of which were killed by the JHA. Of the surviving 181 fertile males, 6 were found to be JHR mutants. The statistically significant resistance factors ranged from 1.4X to 21.5X. The analysis of the most resistant mutant strain indicates that it may possibly be a behavioral mutant. The implications of such JHR mutants are discussed in terms of ‘third generation pesticides’.  相似文献

We have analyzed five Mod-1 (malic enzyme) mutants at the molecular and biochemical level. Four of these mutants, three electrophoretic variants and one null mutant, were induced by ethylnitrosourea (ENU). Another null mutant was the result of a spontaneous mutation. All of these mutations were heritable in a Mendelian fashion and viable in the homozygous condition. Restriction endonuclease and Southern blot analysis revealed that the spontaneous null mutant possessed an altered restriction fragment banding pattern. All of the ENU-induced mutants possessed normal restriction fragment banding patterns. All 5 mutants produced normal levels of Mod-1-specific mRNA. Only the spontaneous null mutant produced mRNA with altered size, which was consistent with the altered DNA-banding pattern. MOD-1 enzyme activity levels were normal in the three ENU-induced mutants with altered electrophoretic mobility. Enzyme activity was significantly lower than normal in tissues from animals homozygous for the null alleles, however, using Western blot analysis, low but significant levels of MOD-1 protein in Mod-1 null homozygotes were detected.  相似文献

Parallel adaptive radiation events provide a powerful framework for investigations of ecology's contribution to phenotypic diversification. Ecologically driven divergence has been invoked to explain the repeated evolution of sympatric dwarf and normal lake whitefish (Coregonus clupeaformis) species in multiple lakes in eastern North America. Nevertheless, links between most putatively adaptive traits and ecological variation remain poorly defined within and among whitefish species pairs. Here, we examine four species pairs for variation in gill, heart, and brain size; three traits predicted to show strong phenotypic responses to ecological divergence. In each of the species pairs, normals exhibited larger body size standardized gills compared to dwarfs – a pattern that is suggestive of a common ecological driver of gill size divergence. Within lakes, the seasonal hypoxia experienced in the benthic environment is a likely factor leading to the requirement for larger gills in normals. Interestingly, the morphological pathways used to achieve larger gills varied between species pairs from Québec and Maine, which may imply subtle non‐parallelism in gill size divergence related to differences in genetic background. There was also a non‐significant trend toward larger hearts in dwarfs, the more active species of the two, whereas brain size varied exclusively among the lake populations. Taken together, our results suggest that the diversification of whitefish has been driven by parallel and non‐parallel ecological conditions across lakes. Furthermore, the phenotypic response to ecological variation may depend on genetic background of each population.  相似文献

The induction of lethal sectoring and pure mutant clones by ultraviolet light has been studied in a homogeneous G1 population of Saccharomyces cerevisiae grown in a normal growth medium. At the lowest UV dose of 250 ergs, which corresponds to a shoulder in the survival curve, all mutants appeared as pure clones. At higher doses the frequency of mosaic mutants progressively increased. These results indicate a relationship between the highest frequency of complete mutants and the maximum repair activity. In addition, the frequency of lethal sectoring at all doses tested was too low to account for the origin of pure mutant clones.  相似文献

Because of its synchrony and relative homogeneity, the subcutaneous model of the resorption of mineral-containing, devitalized bone particles (BPs) is useful to evaluate the recruitment, differentiation, and activity of bone-resorbing, osteoclastic cells. Bone particles were prepared from normal rats or mice and were implanted in normal and osteopetrotic rats (ia, tl, op strains) or mice (mi strain). In addition, particles of microcrystalline hydroxyapatite or polymethylmethacrylate were implanted into tl and op mutants and their unaffected littermates.Non-decalcified histomorphometry of elicited tissues after 12 days revealed significantly less resorption in in each mutant. Enzyme histochemical assays revealed that only normal animals showed tartrate-resistant acid phosphatase-positive cells around the BPs. In agreement with this, only normal animals showed ruffled borders against the BPs. op and tl strains were tested for generation of foreign body giant cells in response to particulate hydroxyapatite or polymethylmethacrylate and no differences were found between mutant and normal animals. These mutants appear to have intact fusion of mononuclear progenitors.These data show impaired recruitment of osteoclasts by BP implants in several rodent strains of osteopetrotic mutants.  相似文献

The accumulation of cadmium, its affinity for metallothioneins (MTs), and its relation to copper, zinc, and selenium were investigated in the experimental mudpuppy Necturus maculosus and the common toad Bufo bufo captured in nature. Specimens of N. maculosus were exposed to waterborne Cd (85???g/L) for up to 40?days. Exposure resulted in tissue-dependent accumulation of Cd in the order kidney, gills > intestine, liver, brain > pancreas, skin, spleen, and gonads. During the 40-day exposure, concentrations increased close to 1???g/g in kidneys and gills (0.64?C0.95 and 0.52?C0.76; n?=?4), whereas the levels stayed below 0.5 in liver (0.14?C0.29; n?=?4) and other organs. Cd exposure was accompanied by an increase of Zn and Cu in kidneys and Zn in skin, while a decrease of Cu was observed in muscles and skin. Cytosol metallothioneins (MTs) were detected as Cu,Zn?Cthioneins in liver and Zn,Cu?Cthioneins in gills and kidney, with the presence of Se in all cases. After exposure, Cd binding to MTs was clearly observed in cytosol of gills as Zn,Cu,Cd?Cthionein and in pellet extract of kidneys as Zn,Cu,Cd?Cthioneins. The results indicate low Cd storage in liver with almost undetectable Cd in liver MT fractions. In field trapped Bufo bufo (spring and autumn animals), Cd levels were followed in four organs and found to be in the order kidney > liver (0.56?C5.0???g/g >0.03?C0.72???g/g; n?=?11, spring and autumn animals), with no detectable Cd in muscle and skin. At the tissue level, high positive correlations between Cd, Cu, and Se were found in liver (all r?>?0.80; ???=?0.05, n?=?5), and between Cd and Se in kidney (r?=?0.76; n?=?5) of autumn animals, possibly connected with the storage of excess elements in biologically inert forms. In the liver of spring animals, having higher tissue level of Cd than autumn ones, part of the Cd was identified as Cu,Zn,Cd?Cthioneins with traces of Se. As both species are special in having liver Cu levels higher than Zn, the observed highly preferential Cd load in kidney seems reasonable. The relatively low Cd found in liver can be attributed to its excretion through bile and its inability to displace Cu from MTs. The associations of selenium observed with Cd and/or Cu (on the tissue and cell level) point to selenium involvement in the detoxification of excessive cadmium and copper through immobilization.  相似文献
