The fragment A of diphtheria toxin was linked to the fragment Fab' of Ig from rabbit antiserum against L1210 cells by the reaction of Fab' with the S-sulfonated fragment A and purified by chromatography on Sephadex G 150. The hybrid thus prepared showed a high cytotoxicity in vitro to L1210 cells which are insensitive to diphtheria toxin itself.  相似文献

A hybrid protein was prepared by coupling the A-chain of diphtheria toxin with the Fab′ fragment of immunoglobulin with N,N′-o-phenylenedimaleimide (PDM). Although in this hybrid, the two components were linked with each other with bonds which could not be reductively cleaved with 2-mercaptoethanol as in a hybrid cross-linked with a disulfide bond (e.g. Fab′-S-S-A-chain), it exhibited a potent cytotoxicity invitro, one-third of that of Fab′-S-S-A-chain, against the target L1210 cells.  相似文献

Cytotoxic actions of various prostaglandins were examined on L1210 mouse leukemia and several human leukemia cell lines, and prostaglandin D2 (PGD2) was found most active. PGD2 exerted a dose dependent inhibition of L1210 cell growth over 3.6 μM. At 14.3 μM growth was completely inhibited, and the number of viable cells remarkably decreased during culture. Microscopically the remaining cells showed degenerative changes with many vacuoles in their cytoplasm. The IC50 value of PGD2 on L1210 cell growth was calculated to be 6.9 μM (2.4 μg/ml), and at this concentration the DNA synthesis in 24 hr cultured cells was also decreased to a half of the level in the control cells. Such growth inhibition by PGD2 was also found at similar concentrations with several human leukemia cell lines such as NALL-1, RPMI-8226, RPMI-8402, and Sk-Ly-16. Among other prostaglandins tested, PGA2 showed a comparable, and PGE2 a less but significant growth inhibitory activity, while PGB2, PGF and PGI2 had no such effects on cell proliferation at 14.3 μM concentration. These results suggest a potential antineoplastic activity of PGD2.  相似文献

Superior antitumor activity of 1-β-D-arabinofuranosylcytosine (ara-C) conjugates of prednisolone and prednisone against L1210 leukemic mice, based on ara-C content, has encouraged us to synthesize 5′-(cortisone-21-phosphoryl)-1-β-D-arabinofuranosylcytosine (I) and 5′-(cortisone-21-phosphoryl)-1-β-d-arabinofuranosylcytosine (II) by condensation of N4,2′,3′-triacetyl-1-β-d-arabinofuranosylcytosine 5′-monophosphate with cortisol and cortisone in the presence of N,N′-dicyclohexylcarbodiimide at room temperature followed by removing the acetyl groups in 2 N methanolic ammonia in 20% yield. The conjugates I and II inhibited the invitro growth of L1210 by 50% (ED50) at 0.25 μM and 0.07 μM, respectively, while ara-C showed ED50 0.1 μM. However, the conjugates I and II exhibited 287% and 238% of TC at 50 mg/kg/day × 5 doses against L1210 leukemic mice, respectively, while ara-C at 25 mg and 50 mg/kg/day × 5 gave the respective 127% and 110% of TC.  相似文献

Summary In studies of antitumor antibody-cytotoxic agent conjugates as potential antitumor agents with improved tumor specificity, the toxic subunit A-chain of ricin was conjugated with a monoclonal antibody to a tumor-associated antigen expressed weakly on murine leukemia L1210 cells and strongly on L1210/GZL cells, a guanazole-resistant subline of L1210, employing N-succinimidyl 3-(2-pyridyldithio)propionate as cross-linking agent. The conjugate (anti-L1210 conjugate) exhibited a potent concentration-dependent cytotoxicity against cultured L1210/GZL cells, and inhibited cell growth at concentrations over 0.8 g/ml. The conjugate killed all L1210/GZL cells at a concentration of 100 g/ml. Neither nonimmune conjugate similarly prepared from mouse nonimmune IgG nor unconjugated anti-L1210 IgG alone showed cytotoxicity against L1210/GZL cells. When (BALB/c×DBA/2)F1 mice inoculated with 1 × 105 L1210/GZL cells were treated with IP injections of 27 g anti-L1210 conjugate 1 h and 5 days after tumor cell inoculation, a life-prolonging effect was observed. [Lifespan in treated animals as percentage of that in controls (T/C)=146%]. However, when the dose per injection was increased to 50 g per mouse, survival was the same as in the control group. Postmortem examination of mice that had been treated with 50 g anti-L1210 conjugate revealed lesions with necrosis and hemorrhage in the liver parenchyma and the intestinal epithelium, respectively. A similar toxic effect on the host mice was also observed with nonimmune conjugate.  相似文献

The turnover of dihydrofolate reductase has been studied in rapidly dividing cells of mouse lymphoma L1210 and Lactobacillus casei. Cells in culture were exposed to [14C]leucine for 24 hr and the subsequent decrease in radioactivity of the enzyme was followed as a function of time. The L1210 enzyme was isolated in pure form by subjecting the cell sonicate to affinity chromatography on amethopterin-Sepharose. The L. casei cells were processed by a multistep procedure which yielded the pure enzyme in both of its principal forms: (I), without TPNH; and (II), containing an equimolal amount of noncovalently bound TPNH. The half-lives (t12>) of dihydrofolate reductase in the amethopterin-sensitive L1210 cells (L1210/S) and in the cells of a partially resistant subline (L1210/R2), characterized by an 8-fold increase in enzyme level, were 18 and 19 hr. When these cells were grown in the presence of sublethal concentrations of amethopterin, t12 values were increased to 39 and 90 hr. These results suggest that the transient increase in dihydrofolate reductase activity, observed when cells are exposed to amethopterin, is due largely to a decreased susceptibility of the enzyme-inhibitor complex to degradation. Bound TPNH also increases the half-life of dihydrofolate reductase as shown by the fact that forms (I) and (II) of the L. casei enzyme had tx12 values of approximately 3 and 9 hr.  相似文献

Complexes of the formula cis-[Pt(HN+N)(L)Cl2], where (HN+N) are the protonated diamines including 3-aminoquinuclidine, N-aminopiperidine, piperazine, N-methylpiperazine, 1,1,4-trimethylpiperazine, and N-methyl-1,4-diazabicyclo [2,2,2] octane (N-methyl-dabco) and L = SCN?, NO2?, Br?, and F?, were synthesized from the protonated diamine complexes, [Pt(HN+N)Cl3]. The antitumor activities of the complexes were evaluated in vitro against L1210 murine leukemia cells, and ID50 values for the L-substituted complexes were compared to values of the parent complexes. In each case it was found that replacement of a chloride ion by SCN?, NO2?, Br?, or F?, either reduced or completely eliminated antitumor activity. This effect is explained in terms of the trans-directing ability of the ligand, L, compared to chloride. The NO2-substituted complex of 3- aminoquinuclidine was tested in vivo and found to exhibit little or no antitumor activity.  相似文献

L1210/R81 lymphoma cells are resistant to methotrexate (MTX) by virtue of a 35-fold elevation in dihydrofolate reductase and an inability to transport the folate antagonist drug effectively. In a phosphate-containing buffer there was little or no influx into the resistant cells at either 1 or 50 μm MTX. Replacement of this buffer with a 4-(2-hydroxyethyl)-1-piperazine-N′-2-ethanesulfonic acid-Mg2+ system resulted in an apparent influx of MTX into the resistant cells. Under these conditions, L1210/R81 cells achieved an apparent steady state at an extracellular MTX concentration of 50 μm. The apparent steady-state level of 5 nmol [3H]MTX109 cells was well below the intracellular level of dihydrofolate reductase (45 nmol/109 cells). Efflux experiments at the apparent steady state indicated that 60% of the MTX was very rapidly removed from the cells by washing. Over the range of the experiment a further 20% of the MTX effluxed more slowly (t12 = 12 min). The apparent influx into the resistant cells at 5 μm MTX was inhibited 13% by sodium azide (100 μm) and initially stimulated, then inhibited, by p-chloromercuriphenylsulfonic acid (100 μm). 5-Methyltetrahydrofolate (100 μm) had little effect on the process while aminopterin (100 μm) was inhibitory (68%). Kt and V values of 2 × 10?5m and 0.31 nmol [3H]MTX109 cells/min, respectively, were determined for the apparent influx in L1210R81 cells. Comparison of apparent MTX influx in the resistant cells with MTX transport in the sensitive cells indicates profound differences in the two processes. The evidence suggests that the apparent influx in the former cell line may consist of MTX binding to the cell membrane together with a small degree of MTX influx into the intracellular compartment.  相似文献

The ability of the high mobility group (HMG) proteins to be phosphorylated was examined in Ehrlich ascites and L1210 cells incubated invitro. HMG proteins were selectively extracted from isolated nuclei with 2% trichloroacetic acid, and electrophoretically separated on acid-urea or SDS polyacrylamide gels. Autoradiography of the gels revealed that among the HMG proteins, only HMG 14 and 17 were labeled. The specific activities of these two proteins were approximately equal to that of histone H1. Phosphorylation of HMG 14 and 17 reached a maximum in 2–3 hr and had turnover rates in pulse-chase experiments similar to that of phosphorylated histone H1.  相似文献

Insulin has been cross-linked via a disulfide bond to the diphtheria toxin fragment A which is catalytically active in ADP-ribosylating elongation factor-2 but does not retain binding sites for toxin receptors. The purified conjugate proved to be cytotoxic to mouse Swiss/3T3 cells which are toxin resistant but express insulin receptors. This cytotoxicity coincided with a decrease in protein synthesis and with drastic morphology changes. In contrast, IN-2 cells, which are insulin-nonresponsive variants derived from mouse BALBc3T3 cells, were resistant to the conjugate. Thus, the conjugate (a chimeric insulin) appears to mediate entry of the toxic fragment A into 3T3 cells through insulin receptors.  相似文献

Superior antitumor activity and reduced toxicity of 1-β-arabinofuranosylcytosine (ara-C) conjugates of corticosteroids through a phosphodiester linkage have prompted us to synthesize the new ara-C conjugates of corticosteroids through a pyrophosphate diester linkage. Condensation of ara-CMP morpholidate with the steroid-21-monophosphates in pyridine at room temperature for 6 days and the subsequent separation on a DE-52 (formate) column using a linear gradient of 0?0.5 M triethylammonium formate (pH 7.0) gave ara-CDP-prednisolone (I) and ara-CDP-cortisol (II) in 37–55% yield. The conjugates were hydrolyzed to ara-CMP and the corresponding steroid-21-monophosphate by phosphodiesterase I. Conjugates I and II inhibited the in, vitro growth of L1210 lymphoid leukemia by 50% (ED50) at 0.03 and 0.08 μM, respectively, while ara-C and ara-CMP showed the respective ED50 of 0.1 μM and 0.05 μM. Conjugates I and II exhibited ILS values of 116 and 86%, respectively, at 40 mg (53.7 μmole)/kg/day × 5 against L1210 leukemic mice, while that of ara-C at the nearly same dose (49 μmole/kg/day × 5) was 65%.  相似文献

Thirteen diversified antimetabolites of coenzyme Q10 which have antitumor activity in vivo were tested for inhibition of uptake of tritiated thymidine and uridine into DNA and RNA, respectively, of L1210 cells grown in tissue culture. Eight of these antimetabolites have inhibitory activities of the same order of magnitude as the used anticancer drugs, rubidazone and ellipticine. 5-ω-Phenylpropylmercapto-2,3-dimethoxy-1,4-benzoquinone was particularly potent to inhibit nucleic acid synthesis; ED50 for DNA = 2.1 μM and ED50 for RNA = 4.0 μM.  相似文献

The effect of the combination of 1-methyl-2-p-chlorophenylaziridine and 1,2-dimethyl-3-p-chlorophenyldiaziridine on cytotoxic activities against L-1210 mouse leukemia cells was studied. Both compounds clearly showed an antagonistic interaction. The results supported our hypothesis that nitrosomethane formed by the fragmentation of aziridine via the N-oxide in a cellular system causes the cytotoxic behavior of the N-methyl aziridines.  相似文献

Both d-glucose and its nonmetabolized analog 3-O-methyl-d-glucose are known to protect the pancreatic B-cell against the toxic action of alloxan, as if the protective action of hexoses were to involve a membrane-associated glucoreceptor site. In the present study, the protective actions of the two hexoses were found to differ from one another in several respects. Using the process of glucose-stimulated insulin release by rat pancreatic islets as an index of alloxan cytotoxicity, we observed that the protective action of d-glucose was suppressed by d-mannoheptulose and menadione, impaired by NH4Cl, and little affected by aminooxyacetate. These findings and the fact that d-glucose failed to decrease [2-14C]alloxan uptake by the islets suggest that the protective action of d-glucose depends on an increase in the generation rate of reducing equivalents (NADH and NADPH). The latter view is supported by the observation that the protective action of a noncarbohydrate nutrient, 2-ketoisocaproate, was also abolished by menadione. Incidentally, the protective action of 2-ketoisocaproate was apparently a mitochondrial phenomenon, it not being suppressed by aminooxyacetate. In contrast to that of glucose, the protective action of 3-O-methyl-d-glucose was unaffected by d-mannoheptulose, failed to be totally suppressed by menadione, and coincided with a decreased uptake of [2-14C]-alloxan by the islets. It is concluded that the protective action of d-glucose in linked to the metabolism of the sugar in islet cells, whereas that of 3-O-methyl-d-glucose results from inhibition of alloxan uptake. This conclusion reinforces our opinion that the presence in the B-cell of an alleged stereospecific membrane glucoreceptor represents a mythical concept.  相似文献

Recently, 1-β-D-arabinofuranosylcytosine-5′-diphosphate-DL-1,2-dipalmitin (VIa) was reported to inhibit the growth of L51784 cells in mice and of human colon carcinoma HCT-15 cells, also in mice. This paper describes the synthesis of a single diastereomer by conversion of 1-β-D-arabinofuranosylcytosine 5′-monophosphate (II) to the nucleoside 5′-phosphomorpholidate (III), followed by reaction with L-α-dipalmitoylphosphatidic acid (IV) to give 1-β-D-arabinofuranosylcytosine-5′-diphosphate-L-1,2-dipalmitin (V) in good yield. The separation of the product is described and its characterization by chromatography, elemental analysis, and spectroscopic methods. The lipophilic nature of V renders it insoluble in aqueous media and a method of sample preparation utilizing sonication techniques is described which provides a clear solution suitable for biological evaluation. In addition, the ability of V to inhibit the invitro growth of L1210 cells and of mouse myeloma MPC 11 cells is desscribed and compared with 1-β-D-arabinofuranosylcytosine (I) and other lipophilic prodrugs of I.  相似文献

A soluble macromolecule-peptide conjugate, [(Met)3-OPEG] inhibited the uptake of Met-Met-[14C] Met into S.cerevisiae. Uptake of leucine into this strain was not affected by Met3-OPEG under identical conditions. Inhibition by the macromolecular inhibitor was competitive (KI = 5.1 × 10?5M)and followed the structural requirments of the peptide transport systems in S.cerevisiae and C.albicans. These findings give the first example of inhibition of metabolite transport by a synthetic macromolecular competitor.  相似文献

A rapid isolation method was developed for plasma membranes from mouse lymphoid cells such as lymph node lymphocytes, thymocytes, radiation-induced thymoma cells and L1210 cells. Lysates of these lymphoid cells were prepared by Dounce homogenization under hypotonic conditions and directly layered on sucrose step density gradients containing 2 mM CaCl2 and 5 mM MgCl2, and centrifuged at 52 000 × g for 1 h. Plasma membrane fractions appeared at the interface between 20 and 42% sucrose in the gradients. The procedure permitted purified membranes from cells to be obtained within 3 h, and the preparations appeared to be uniform by electron microscopy. Specific activities of (Na+ + K+)-ATpase, Mg2+-ATPase and 5′-nucleotidase of the isolated plasma membranes were enriched 23- to 61-fold, 12- to 15-fold and 18- to 34-fold, respectively, in comparison with those of the corresponding cell homogenates. Cholesterol content of the malignant cell membranes was lower than that of the normal membranes and the molar ratio of cholesterol to phospholipid of the malignant cell membranes was also lower than that of the normal membranes. A decreased plasmalogen content was observed in the malignant plasma membranes, together with a higher percentage of phosphatidylethanolamine and a lower percentage of phosphatidylserine. In the normal cell membranes, thymocytes contained a higher percentage of phosphatidylcholine and a lower percentage of sphingomyelin than those of the lymph node lymphocytes. At all temperature ranges (5 to 40°C) the plasma membranes of the malignant cells had lower microviscosity than those of the normal cells.  相似文献

[3 H] Estradiol-17β-succinyl bovine serum albumin conjugate ([3H]-E2-BSA) was synthesized with a specific activity of 1.92 × 107 cts/min/mg. The conjugate was administered iv to ovariectomized rats and the quantity of free [3H] steroid in the uterus was determined. Radioactive material was detected in all of the subcellular fractions of the uterus and identified as estradlol-17β. Similar subcellular distribution of the radioactivity was observed when [3H]E2-BSA was added in vitro to uterine homogenate. Free estradlo1-17β was released when the conjugate was Incubated with rat uterine homogenate or with serum. The results of the present study suggest that E2-BSA is hydrolyzed in vivo and under in vitro conditions. It is recommended that the stability of a hormone-protein conjugate be established before use.  相似文献

d1-Prostaglandin E1 and d1-11-deoxyprostaglandin E1 are conveniently synthesized via the copper (I) catalyzed conjugate addition of the Grignard reagent prepared from 3-trityloxy-trans-1-octenyl bromide to the appropriate cyclopentenone precursor. OKY-1581 is an effective inhibitor of thromboxane synthesis invivo and invitro. The generation of thromboxane B2 (TxB2), prostaglandin E (PGE) and prostaglandin F (PGF) was measured following clotting and during platelet aggregation induced by collagen. The presence of OKY 1581 either invivo or invitro caused a reduction in TxB2 generation during clotting and platelet aggregation with a concomitant increase in PGE and PGF. The effect could be observed two hours after oral or subcutaneous administration of 5 to 100 mg per rabbit and lasted for 24 to 48 hours. The reduction in TxB2 was not accompanied by an inhibition of clotting or platelet aggregation. OKY-1581 appears to be a suitable agent for studying the role of TxB2 in atherosclerosis.  相似文献
