Brassinosteroids (Brs) have drawn wide attention due to their protective role against toxicity of heavy metals in plants. To better understand the role of Br in arsenic (As) and cadmium (Cd) uptake by rice plants, a hydroponic experiment was conducted to investigate the combined effect of 24-epibrassinolide (Br24) or 28-homobrassinolide (Br28) and iron plaque (IP) on As and Cd uptake and accumulation in rice seedlings. Six-week-old seedlings were sprayed with 0.2 or 0.02 μM Br24 or Br28 and grown in nutrient solution for 3 d, and then 20 or 60 mg Fe2+ dm-3 (Fe20 and Fe60) was used to induce root IP formation for 3 d. These seedlings with or without Br and with or without IP were exposed to solution containing 0.5 mg dm-3 AsIII or Cd for 9 d. The results showed that rice growth decreased when Br24 were applied, but it increased when combination of Br24 and IP was applied. Fe concentrations in dithionite-citratebicarbonate (DCB) extracts were increased after 0.2 or 0.02 μM Br24 application in the absence of IP, but decreased by Br24 in the presence of IP. In the absence of IP, As and Cd content in leaves was significantly reduced by 0.02 μM Br24 and 0.2 μM Br28, respectively. The As content in leaves was also reduced by the combination of 0.02 and 0.2 μM Br28 and IP, and the Cd content in leaves was reduced by the combined effect of 0.2 μM Br24 and IP. These results indicate that Br24 and Br28 could impede As and Cd accumulation, and the interactions between Br and IP may have a potential in restricting the transport of As and Cd into rice shoots.  相似文献

Background and aims

Rice (Oryza sativa) is a main source of human exposure to inorganic arsenic and mitigation measures are needed to decrease As accumulation in this staple crop. It has been shown that silicon decreases the accumulation of arsenite but, unexpectedly, increases the accumulation of dimethylarsinic acid (DMA) in rice grain. The aim of this study was to investigate why Si increases DMA accumulation.

Methods

Pot and incubation experiments were conducted to investigate how the addition of sparingly soluble silicate gel affected As speciation in the soil solution and the accumulation of different As species in rice tissues.

Results

Silicon addition significantly decreased the concentration of inorganic As (mainly arsenite) but increased the concentration of DMA in both the vegetative and reproductive tissues of rice. Silicon increased the concentration of DMA in the soil solution, whereas autoclaving soil decreased DMA concentration. Less DMA was adsorbed by the soil than arsenate and Si addition significantly inhibited DMA adsorption.

Conclusions

Silicon increased DMA accumulation and decreased arsenite accumulation in rice through different mechanisms. Silicic acid released from the silicate gel increased the availability of DMA for rice uptake by inhibiting DMA adsorption on the soil solid phase or by displacing adsorbed DMA. Although silicic acid also increased the concentration of inorganic As in the soil solution, this effect was much smaller than the inhibitory effect of Si on arsenite uptake by rice roots.  相似文献

Both solution culture and pot experiments were performed to investigate (a) the effects of external Fe (II) concentrations and forms on the formation of iron plaque on the roots of rice (Oryza sativa) and subsequent P adsorption on iron plaque and shoot P concentrations and (b) the effects of soil moisture regimes on the formation of iron plaque and P adsorption on root surfaces and P accumulation in shoots. The results showed that iron plaque was significantly increased with increasing Fe2+ concentrations in the solution culture. The amounts of P adsorbed on the iron plaque were increased significantly with external Fe2+ concentrations. Although shoot P concentration was not significantly affected by Fe2+ treatment after incubation for 2 days, it was significantly increased in the Fe‐treated plants compared with Fe‐deprived ones after incubation for 4 days. Soil culture experiment showed that the formation of iron plaque on root surfaces was promoted by exogenous iron, with greater amount of iron plaque being formed by addition of ferric hydroxide than of ferric oxide. Phosphorus adsorption on iron plaque also increased with the addition of iron oxides, and increasing soil P increased the amounts of P associated with the iron plaque and shoot P concentration. The amounts of iron plaque were almost sixfold higher under flooding condition than under field capacity condition. Plants pretreated under flooding condition generally had higher shoot P concentrations when they were transplanted to solutions with varying P levels, and this was most pronounced in the treatment with highest solution P concentration. The results suggest that iron plaque acts as a nutrient reservoir for phosphorus in the rhizosphere and helps enhance P acquisition by rice.  相似文献

The influence of sulphur on the accumulation and metabolism of arsenic in rice was investigated. Rice seedlings were grown in nutrient solutions with low sulphate (1.8 μM SO42−) or high sulphate (0.7 mM SO42−) for 12 or 14 d, before being exposed to 10 μM arsenite or arsenate for 2 or 1 d, respectively. In the arsenite exposure treatment, low sulphate-pretreated rice accumulated less arsenite than high sulphate pretreated plants, but the arsenite concentrations in shoots of low sulphate pretreated rice were higher than those of high sulphate pretreated. In the arsenate exposure treatment, the low sulphate pre-treatments also resulted in less arsenite accumulation in rice roots. Sulphur deprivation in nutrient solution decreased the concentrations of non-protein thiols in rice roots exposed to either arsenite or arsenate. The low sulphate-pretreated plants had a higher arsenic transfer factor than the high sulphate-pretreated plants. The results suggest that rice sulphate nutrition plays an important role in regulating arsenic translocation from roots to shoots, possibly through the complexation of arsenite-phytochelatins.  相似文献

Background and aims

Efficient accumulation of arsenic (As) in rice (Oryza sativa L.) poses a potential health risk to rice consumers. The aim of this study was to investigate the mechanisms of uptake, transport and distribution of inorganic arsenic (Asi) and dimethylarsinic acid (DMA) in rice plants.

Methods

Rice was exposed to Asi (As(V)) and DMA in hydroponics. High-performance liquid chromatography inductively coupled plasma mass spectrometry (HPLC-ICP-MS) and synchrotron X-ray fluorescence (SXRF) microprobe were used to determine As concentration and the in situ As distribution.

Results

DMA induced abnormal florets before flowering and caused a sharp decline in the seed setting rate after flowering compared to Asi. Rice grains accumulated 2-fold higher DMA than Asi. The distribution of Asi concentration (root?>?leaf?>?husk?>?caryopsis) in As(V) treatments was different from that of the DMA concentration (caryopsis?>?husk?>?root?≥?leaf) in DMA treatments. SXRF showed that Asi mainly accumulated in the vascular trace of caryopsis with limited distribution to the endosperm, whereas DMA was observed in both tissues.

Conclusions

DMA tended to accumulate in caryopsis and induced higher toxicity to the reproductive tissues resulting in markedly reduced grain yield, whereas Asi mainly remained in the vegetative tissues and had no significant effect on yield. DMA is more toxic than Asi to the reproductive tissues when both of them are at similar concentrations in nutrient solution.  相似文献

采用营养液培养方法研究了水稻根表形成的铁氧化物胶膜对水稻吸收Zn的影响.结果表明,在有Fe2+的嫌气环境中,由于根际氧化作用水稻根表会形成红色的铁氧化物胶膜,根表的铁氧化物胶膜影响水稻对Zn的吸收.铁膜数量较少时,由于对Zn的富集作用有限,其对水稻Zn的吸收虽有促进作用,但不明显.随着根表铁膜数量的增加,这种促进作用也相应增加,并且在铁膜数量增加到一定值时,对水稻吸收Zn的促进作用达到最大.而后,随着铁膜数量的进一步增加,铁膜反而阻碍水稻对Zn的吸收,成为水稻吸收Zn的障碍层.在此过程中,水稻的根分泌物,特别是其中的植物铁载体对覆有铁膜水稻根系吸收Zn有促进作用.这种促进作用随铁膜数量的增加而逐渐减弱.因此,根表铁氧化物胶膜对水稻吸收Zn并不总是起促进作用,其作用的方向和程度取决于铁膜的数量.  相似文献

Summary The effects of nitrogen and phosphorus on seed yield and seed nutrient content inPisum sativum L. cv Sprite were studied. In the first experiment (spring-sown) seed yield was increased by 55% due to the combined effect of high nitrogen and phosphorus, although in subsequent experiments (during the summer) nitrogen had no effect on seed yield, and a response to phosphorus was obtained in only one of two experiments. Human activities have resulted in arsenic (As) and heavy metals accumulation in paddy soils in China. Phytoremediation has been suggested as an effective and low-cost method to clean up contaminated soils. A combined soil-sand pot experiment was conducted to investigate the influence of red mud (RM) supply on iron plaque formation and As and heavy metal accumulation in two wetland plant species (Cyperus alternifolius Rottb., Echinodorus amazonicus Rataj), using As and heavy metals polluted paddy soil combined with three rates of RM application (0, 2%, 5%). The results showed that RM supply significantly decreased As and heavy metals accumulation in shoots of the two plants due to the decrease of As and heavy metal availability and the enhancement of the formation of iron plaque on the root surface and in the rhizosphere. Both wetland plants supplied with RM tended to have more Fe plaque, higher As and heavy metals on roots and in their rhizospheres, and were more tolerant of As and heavy metal toxicity. The results suggest that RM-induced enhancement of the formation of iron plaque on the root surface and in the rhizosphere of wetland plants may be significant for remediation of soils contaminated with As and heavy metals.  相似文献

The effects of phosphorus (P) status on arsenate reductase gene (OsACR2.1) expression, arsenate reductase activity, hydrogen peroxide (H2O2) content, and arsenic (As) species in rice seedlings which were exposed to arsenate after ?P or +P pretreatments were investigated in a series of hydroponic experiments. OsACR2.1 expression increased significantly with decreasing internal P concentrations; more than 2-fold and 10-fold increases were found after P starvation for 30 h and 14 days, respectively. OsACR2.1 expression exhibited a significant positive correlation with internal root H2O2 accumulation, which increased upon P starvation or exposure to H2O2 without P starvation. Characterization of internal and effluxed As species showed the predominant form of As was arsenate in P-starved rice root, which contrasted with the +P pretreated plants. Additionally, more As was effluxed from P-starved rice roots than from non-starved roots. In summary, an interesting relationship was observed between P-starvation induced H2O2 and OsACR2.1 gene expression. However, the up-regulation of OsACR2.1 did not increase arsenate reduction in P-starved rice seedlings when exposed to arsenate.  相似文献

Arsenic (As) accumulation in rice grain poses a serious health risk to populations with high rice consumption. Extrusion of arsenite [As(III)] by ScAcr3p is the major arsenic detoxification mechanism in Saccharomyces cerevisiae. However, ScAcr3p homolog is absent in higher plants, including rice. In this study, ScACR3 was introduced into rice and expressed under the control of the Cauliflower mosaic virus (CaMV) 35S promoter. In the transgenic lines, As concentrations in shoots and roots were about 30% lower than in the wild type, while the As translocation factors were similar between transgenic lines and the wild type. The roots of transgenic plants exhibited significantly higher As efflux activities than those of the wild type. Within 24 h exposure to 10 μM arsenate [As(V)], roots of ScACR3-expressing plants extruded 80% of absorbed As(V) to the external solution as As(III), while roots of the wild type extruded 50% of absorbed As(V). Additionally, by exposing the As-containing rice plants to an As-lacking solution for 24 h, about 30% of the total As derived from pre-treatment was extruded to the external solution by ScACR3-expressing plants, while about 15% of As was extruded by wild-type plants. Importantly, ScACR3 expression significantly reduced As accumulation in rice straws and grains. When grown in flooded soil irrigated with As(III)-containing water, the As concentration in husk and brown rice of the transgenic lines was reduced by 30 and 20%, respectively, compared with the wild type. This study reports a potential strategy to reduce As accumulation in the food chain by expressing heterologous genes in crops.  相似文献

Cadmium (Cd) is a heavy metal toxic to humans and the accumulation of Cd in the rice grain is a major agricultural problem, particularly in Asia. The role of the iron transporter OsNRAMP1 in Cd uptake and transport in rice was investigated here. An OsNRAMP1:GFP fusion protein was localized to the plasma membrane in onion epidermal cells. The growth of yeast expressing OsNRAMP1 was impaired in the presence of Cd compared with yeast transformed with an empty vector. Moreover, the Cd content of OsNRAMP1-expressing yeast exceeded that of the vector control. The expression of OsNRAMP1 in the roots was higher in a high Cd-accumulating cultivar (Habataki) than a low Cd-accumulating cultivar (Sasanishiki) regardless of the presence of Cd, and the amino acid sequence of OsNRAMP1 showed 100% identity between Sasanishiki and Habataki. Over-expression of OsNRAMP1 in rice increased Cd accumulation in the leaves. These results suggest that OsNRAMP1 participates in cellular Cd uptake and Cd transport within plants, and the higher expression of OsNRAMP1 in the roots could lead to an increase in Cd accumulation in the shoots. Our results indicated that OsNRAMP1 is an important protein in high-level Cd accumulation in rice.  相似文献

Background and aims

Iron plaque on roots has been hypothesized to be an effective restraint on the uptake of arsenic (As) by rice plants. Evaluating the formation of iron plaque and its effect on As uptake by various rice cultivars is valuable because selecting low As uptake rice cultivars results in reduced risks associated with rice consumption. This study examines iron plaque formation and its effect on As uptake by different genotypes of rice cultivars.

Methods

Hydroponic cultures were conducted in phytotron at day 25/night 20°C and the rice seedlings in fifth-leaf age were treated with Fe (II) at the levels of 0 and 100 mg L?1 in the Kimura B nutrient solutions for 14 days. The amount of iron plaque formation of 28 rice cultivars was determined by using the DCB extractable Fe of roots. Four cultivars representing high and low iron plaque formation capability, from indica and japonica respectively, were selected out of the 28 cultivars and processed for Fe and As treatments. After Fe treatments for 4 days, the seedlings were fed with As (III) at levels of 0, 0.5, and 1 mg L?1 for another 10 days. We were thus able to determine the amounts of iron plaque formation and the As content in iron plaque, roots, and shoots of the four tested cultivars.

Results

Iron plaque formation capability differed among tested twenty-eight rice cultivars. Feeding As to four tested cultivars enhanced iron plaque formation on roots; the As uptake by roots and shoots was decreased by the addition of Fe. Both the retention of As on iron plaque and the decrease of As uptake by the addition of Fe varied among tested cultivars and were not correlated with the iron plaque formation capability.

Conclusions

Iron plaque can sequestrate As on the roots and reduce rice’s As uptake. However, other factors also influence the As uptake, namely the differences in binding affinity of iron plaque to As, the existent As species in the rhizosphere, and the uptake capability of various As species by rice plants. These factors should also be considered when selecting low As uptake rice cultivars.  相似文献

Rice seedlings were grown in hydroponic culture to determine the effects of external Zn and P supply on plant uptake of Cd in the presence or absence of iron plaque on the root surfaces. Iron plaque was induced by supplying 50 mg l−1 Fe2+ in the nutrient solution for 2 day. Then 43-day-old seedlings were exposed to 10 μmol l−1 Cd together with 10 μmol l−1 Zn or without Zn (Zn–Cd experiment), or to 10 μmol l−1 Cd with 1.0 mmol l−1 P or without P (P–Cd experiment) for another 2 day. The seedlings were then harvested and the concentrations of Fe, Zn, P and Cd in dithionite–citrate–bicarbonate (DCB) extracts and in roots and shoots were determined. The dry weights of roots and shoots of seedlings treated with 50 mg l−1 Fe were significantly lower than when no Fe was supplied. Adsorption of Cd, Zn and P on the iron plaque increased when Fe was supplied but Cd concentrations in DCB extracts were unaffected by external Zn or P supply levels. Cd concentrations in shoots and roots were lower when Fe was supplied. Zn additions decreased Cd concentrations in roots but increased Cd concentrations in shoots, whereas P additions significantly increased shoot and root Cd concentrations and this effect diminished when Fe was supplied. The percentage of Cd in DCB extracts was significantly lower than in roots or shoots, accounting for up to 1.8–3.8% of the plant total Cd, while root and shoot Cd were within the ranges 57–76% and 21–40% respectively in the two experiments. Thus, the main barrier to Cd uptake seemed to be the root tissue and the contribution of iron plaque on root surfaces to plant Cd uptake was minor. The changes in plant Cd uptake were not due to Zn or P additions altering Cd adsorption on iron plaque, but more likely because Zn or P interfered with Cd uptake by the roots and translocation to the shoots.  相似文献

A greenhouse experiment evaluated the effect of phytoextraction of arsenic from a contaminated soil by Chinese Brake Fern (Pteris vittata L.) and its subsequent effects on growth and uptake of arsenic by rice (Oryza sativa L.) crop. Pteris vittata was grown for one or two growing cycles of four months each with two phosphate sources, using single super phosphate (SSP) and di-ammonium phosphate (DAP). Rice was grown on phytoextracted soils followed by measurements of biomass yield (grain, straw, and root), arsenic concentration and, uptake by individual plant parts. The biomass yield (grain, straw and rice) of rice was highest in soil phytoextracted with Pteris vittata grown for two cycles and fertilized with diammonium phosphate (DAP). Total arsenic uptake in contaminated soil ranged from 8.2 to 16.9 mg pot(-1) in first growing cycle and 5.5 to 12.0 mg pot(-1) in second growing cycle of Pteris vittata. There was thus a mean reduction of 52% in arsenic content of rice grain after two growing cycle of Pteris vittata and 29% after the one growing cycle. The phytoextraction of arsenic contaminated soil by Pteris vittata was beneficial for growing rice resulted in decreased arsenic content in rice grain of <1 ppm. There was a mean improvement in rice grain yield 14% after two growing cycle and 8% after the one growing cycle of brake fern.  相似文献
