The khapra beetle, Trogoderma granarium, is one of the most important stored product pests worldwide. A study of digestive proteinases in T. granarium was performed to identify potential targets for proteinaceous biopesticides, such as proteinase inhibitors. The pH of guts was determined by addition of pH indicator solutions to broken open gut regions. The last instar larvae were dissected in cold distilled water and the whole guts were cleaned from adhering unwanted tissues. The pooled gut homogenates were centrifuged and the supernatants were used in the subsequent enzyme assay. Total proteinases activity of the gut homogenates was determined using the protein substrate azocasein. Optimal azocasein hydrolysis by luminal proteinases of the larvae of T. granarium was highly alkaline in pH 10-10.5, although the pH of luminal contents was slightly acidic (pH 6.5). The extract showed the highest activity at 55 degrees C (pH 6.5), 45 degrees C (pH 8) and 30 degrees C (pH 10). The proteolytic activity was strongly inhibited in the presence of phenylmethylsulphonyl fluoride (82.33+/-4.37% inhibition). This inhibition was decreased with increasing of the pH of assay incubating medium. N-p-tosyl-L-lysine chloromethyl ketone (51.6+/-3.3% inhibition) and N-tosyl-L-phenylalanine chloromethyl ketone (27.23+/-4.37 % inhibition) showed inhibitory effect on proteolysis. Addition of thiol activators dithiothreitol and L-cysteine had not enhanced azocaseinolytic activity. The data suggest that protein digestion in the larvae of T. granarium is primarily dependent on serine proteinases; trypsin- and chymotrypsin-like proteinases.  相似文献

Brief extraction of the virgin females of Trogoderma granarium yielded a mixture of fatty acids, the major ones being palmitic and oleic acids; these were accompanied by minor quantities of linoleic, palmitoleic, margaric and stearic acids. The aggregation effect of these cuticular fatty acids on the females of Trogoderma granarium is recorded.

On obtient un mélange d'acides gras dont les plus importants sont les acides palmitique et oléique, accompagnés de petites quantités d'acides linoléique, palmitoléique, margarique et stéarique. Ces divers acidès gras cuticulaires ont un effet attractif plus ou moins marqué sur les femelles de Trogoderma.

To date, a molecular phylogenetic approach has not been used to investigate the evolutionary structure of Trogoderma and closely related genera. Using two mitochondrial genes, Cytochrome Oxidase I and Cytochrome B, and the nuclear gene, 18S, the reported polyphyletic positioning of Trogoderma was examined. Paraphyly in Trogoderma was observed, with one Australian Trogoderma species reconciled as sister to all Dermestidae and the Anthrenocerus genus deeply nested within the Australian Trogoderma clade. In addition, time to most recent common ancestor for a number of Dermestidae was calculated. Based on these estimations, the Dermestidae origin exceeded 175 million years, placing the origins of this family in Pangaea.  相似文献

With efficient sequencing techniques, full mitochondrial genomes are rapidly replacing other widely used markers, such as the nuclear rRNA genes, for phylogenetic analysis but their power to resolve deep levels of the tree remains controversial. We studied phylogenetic relationships of leaf beetles (Chrysomelidae) in the tribes Galerucini and Alticini (root worms and flea beetles) based on full mitochondrial genomes (103 newly sequenced), and compared their performance to the widely sequenced nuclear rRNA genes (full 18S, partial 28S). Our results show that: (i) the mitogenome is phylogenetically informative from subtribe to family level, and the per‐nucleotide contribution to nodal support is higher than that of rRNA genes, (ii) the Galerucini and Alticini are reciprocally monophyletic sister groups, if the classification is adjusted to accommodate several ‘problematic genera’ that do not fit the dichotomy of lineages based on the presence (Alticini) or absence (Galerucini) of the jumping apparatus, and (iii) the phylogenetic results suggest a new classification system of Galerucini with eight subtribes: Oidina, Galerucina, Hylaspina, Metacyclina, Luperina, Aulacophorina, Diabroticina and Monoleptina.  相似文献

【目的】从线粒体基因组水平上探讨枣食芽象甲Scythropus yasumatsui与近缘种的系统发育关系。【方法】利用Illumina MiSeq测序平台对枣食芽象甲线粒体基因组进行测序,对基因组序列进行拼装、注释和特征分析;利用贝叶斯法和最大似然法构建基于象甲科13个物种的线粒体基因组13个蛋白质编码基因核苷酸序列的系统发育树。【结果】结果表明,枣食芽象甲线粒体基因组全长为16 472 bp (GenBank登录号: MF807224),包含13个蛋白质编码基因、22个tRNA基因、2个rRNA基因和2个非编码控制区,37个基因的排列顺序与祖先昆虫的线粒体基因排列顺序一致。13个蛋白质编码基因的起始密码子为ATN,其中除了cob和nad1基因的完全终止密码子为TAG外,其余11个基因的完全终止密码子为TA(A)。22个tRNA基因中除了trnS1缺少DHU臂,反密码子由GCT变为TCT外,其余均能形成典型的三叶草结构。基于13个蛋白质编码基因序列构建的系统发育树结果显示,象甲科8个亚科系统发育关系为:(((隐喙象亚科(Cryptorhynchinae)+(象虫亚科(Curculioninae)+魔喙象亚科(Molytinae)))+长小蠹亚科(Platypodinae))+(粗喙象亚科(Entiminae)+Cyclominae亚科))+隐颏象亚科(Dryophthorinae)+小蠹亚科(Scolytinae))。【结论】在13种象甲科昆虫物种中,同属于粗喙象亚科的枣食芽象甲与南美果树象甲Naupactus xanthographus在系统发育树中聚为同一分支,表明基于线粒体基因组全序列的分子系统发育结果与传统的形态分类结果是一致的。  相似文献

Haemulidae species are morphologically diversified fishes with wondrous and changeable coloration. These species are prevalent in global tropical, subtropical, and temperate inshore reef areas. Previous morphological classification within Haemulidae and some related families was problematic, and no comprehensive molecular evaluation was conducted on these groups. In this study, we revealed the first molecular phylogenetic analysis involving representative species within Haemulidae and the relevant families (Nemipteridae and Teraponidae). The analysis was performed using both mitochondrial DNA (16S rRNA) and nuclear DNA (TMO-4c4) genes to construct maximum parsimony, maximum likelihood, and Bayesian analysis tree topologies. The molecular phylogeny recovered well-resolved relationships within Haemulidae species and the problematic taxa. In the trees, the Haemulidae species (except for genus Hapalogenys) were divided into two distinct sister lineages, including grunts (Haemulinae) and sweetlips (Plectorhynchinae). Hapalogenys was positioned outside the major Haemulidae tribe and formed an independent group, which challenged the traditional taxonomy that Hapalogenys was classified into Haemulidae. The results did agree with most current studies that Hapalogenys was only distantly related to Haemulidae and could potentially be removed from this family. Additionally, the genus Diagramma was observed to be tightly grouped inside the clade Plectorhinchus, indicating its highly close affinity within Plectorhinchus. Regarding interfamily relationships, the phylogenetic constructions suggested distant relationships within two pairs: between Scolopsis and Haemulidae and between Teraponidae and Haemulidae. Scolopsis was revealed to be highly close with Nemipteridae, and Teraponidae was clustered in an independent group, which was in accordance with most current taxonomic studies.  相似文献

Laboratory studies were carried out to investigate bioactivity of Piper guineense seeds and Moringa oleifera leaf powders applied singly or in a mixture against larvae and adult Trogoderma granarium Everts in airtight containers. Three levels (0.0 g, 0.5 g, and 1.0 g/20 g groundnut seeds) of the plant powders were used and pirimiphos-methyl was applied at 0.01 g/20 g seeds (recommended dose). Another control consisting of untreated seeds with aerated lids was included in the bioassay. Both larvae and adults were not killed in control with aerated lids throughout the experimental period and larvae were also tolerant to airtight storage conditions. Adults were more susceptible to plant powders than larvae and adult mortality recorded in P. guineense at 1.0 g, 0.5 g and M. oleifera at 1.0 g/20 g seeds were not significantly different from the mortality observed with the recommended dose of Pirimiphos methyl at five days after treatment (DAT). Larval mortality observed in a mixture of both plants (1:1; w/w) caused significantly higher mortality (77.5%) than other treatments at 5 DAT. All treatments (P. guineense and M. oleifera applied singly or in a mixture) were repellant to larvae T. granarium with 60% repellency recorded in the mixture of plants, 50% repellency in P. guineense and 30% repellency in M. oleifera slurry. The water absorption capacity of treated seeds was not affected by treatment with plant powders and ranged from 31.98% to 37.59%.  相似文献

To assess the accuracy of species delimitation and phylogenetic relationships of the Aleochara fucicola species complex, we performed molecular phylogenetic analyses. Detailed micromorphological characters were also examined using scanning electron microscopy (SEM). The molecular data set included two mitochondrial (COI and COII) and three nuclear protein‐coding genes (CAD, EF1α and wg) for 19 populations of five species. Significant discordance was found between mitochondrial and nuclear gene trees. Two species, A. puetzi (Assing) and A. segregata Yamamoto and Maruyama, were not separated in the mitochondrial gene trees, but clearly were differentiated in the nuclear and concatenated gene trees. The SEM data also supported the separation of these two species, but an analysis of genetic divergence data did not. Adaptation to extremely harsh environments might reduce morphological variation in the A. fucicola species complex during the colonization of seashores. We present a multilocus phylogeny of the species complex. It suggests that the ancestor of the A. fucicola species complex occurred along the southern coasts of Northeast Asia, followed by dispersals to northern coasts.  相似文献

Six new species of the Australian myrmecophilous ptinid genus Polyplocotes are described from South Australia. Three are from the deserts of central Australia, one from the Franklin Islands in the Great Australian Bight, one from Eyre Peninsula and one from the Riverland region. Morphologically, the majority of these new species are conventional Polyplocotes , but two are less typical. The characters uniting the genus are explored in the discussion, and comparisons are made to related genera. Although the six new species described here have not been observed in the field, the species of this genus are known to be myrmecophilous, and ant – beetle interactions similar to those seen in other spider beetles might occur between these new species and their host ants.  相似文献

A phylogenetic analysis of 6.4 kb of nucleotide sequence data from seven genes (mitochondrial cox1-cox2 and tRNA(leu), and nuclear Ef-1alpha C0, Ef-1alpha C1, 28S, and 18S) was done to reconstruct the phylogenetic relationships of the ground-beetle tribe Sphodrini. Gene regions of variable nucleotide length were aligned using both a secondary structure model, Clustal W, and a combination of the two. Sensitivity analysis was performed in order to explore the effect of alignment methods. The ribosomal and protein-coding genes were largely congruent based on the ILD test and partitioned Bremer support measures. MtDNA analysis provided high resolution and high support for most clades. The tribe Sphodrini and the related tribes Platynini, Pterostichini and Zabrini made up monophyletic clades, but the relationship between them was weakly resolved and sensitive to alignment strategy. Previously suggested relationships between subtribes of Sphodrini were not corroborated, and only the subtribe Atranopsina revealed high support as the sister clade to the other subtribes. The analyses clearly demonstrated the importance of exploring effects of alignment methods that may become particularly important in resolving polytomies and nodes with low support.  相似文献

The family Theridiidae is one of the most diverse assemblages of spiders, from both a morphological and ecological point of view. The family includes some of the very few cases of sociality reported in spiders, in addition to bizarre foraging behaviors such as kleptoparasitism and araneophagy, and highly diverse web architecture. Theridiids are one of the seven largest families in the Araneae, with about 2200 species described. However, this species diversity is currently grouped in half the number of genera described for other spider families of similar species richness. Recent cladistic analyses of morphological data have provided an undeniable advance in identifying the closest relatives of the theridiids as well as establishing the family's monophyly. Nevertheless, the comb-footed spiders remain an assemblage of poorly defined genera, among which hypothesized relationships have yet to be examined thoroughly. Providing a robust cladistic structure for the Theridiidae is an essential step towards the clarification of the taxonomy of the group and the interpretation of the evolution of the diverse traits found in the family. Here we present results of a molecular phylogenetic analysis of a broad taxonomic sample of the family (40 taxa in 33 of the 79 currently recognized genera) and representatives of nine additional araneoid families, using approximately 2.5kb corresponding to fragments of three nuclear genes (Histone 3, 18SrDNA, and 28SrDNA) and two mitochondrial genes (16SrDNA and CoI). Several methods for incorporating indel information into the phylogenetic analysis are explored, and partition support for the different clades and sensitivity of the results to different assumptions of the analysis are examined as well. Our results marginally support theridiid monophyly, although the phylogenetic structure of the outgroup is unstable and largely contradicts current phylogenetic hypotheses based on morphological data. Several groups of theridiids receive strong support in most of the analyses: latrodectines, argyrodines, hadrotarsines, a revised version of spintharines and two clades including all theridiids without trace of a colulus and those without colular setae. However, the interrelationships of these clades are sensitive to data perturbations and changes in the analysis assumptions.  相似文献

The 32 species of the Centrarchidae are ecologically important components of the diverse fish communities that characterize North American freshwater ecosystems. In spite of a rich history of systematic investigations of centrarchid fishes there is extensive conflict among previous hypotheses that may be due to restricted taxon or character sampling. We present the first phylogenetic analysis of the Centrarchidae that combines DNA sequence data from both the mitochondrial and nuclear genomes and includes all described species. Gene sequence data were collected from a complete mtDNA protein coding gene (NADH subunit 2), a nuclear DNA intron (S7 ribosomal protein intron 1), and a portion of a nuclear DNA protein-coding region (Tmo-4C4). Phylogenetic trees generated from analysis of the three-gene dataset were used to test alternative hypotheses of centrarchid relationships that were gathered from the literature. Four major centrarchid lineages are present in trees generated in maximum parsimony (MP) and Bayesian maximum likelihood analyses (BML). These lineages are Acantharchus pomotis, Micropterus, Lepomis, and a clade containing Ambloplites, Archoplites, Centrarchus, Enneacanthus, and Pomoxis. Phylogenetic trees resulting from MP and BML analyses are highly consistent but differ with regard to the placement of A. pomotis. Significant phylogenetic incongruence between mtDNA and nuclear genes appears to result from different placement of Micropterus treculi, and is not characteristic of relationships in all other parts of the centrarchid phylogeny. Slightly more than half of the 27 previously proposed hypotheses of centrarchid relationships were rejected based on the Shomodaira-Hasegawa test.  相似文献
