目的:克隆B19病毒XA株VP1u基因,构建真核重组表达载体.方法:从已构建好的B19病毒XA株原核表达载体中获得VP1u基因,将其克隆入真核表达载体plRES2-EGFP中,经酶切鉴定并测序验证后,获得真核表达载体plRES2-EGFP-VP1u.将其转染至HeLa细胞,提取细胞总蛋白,用Western blot技术检测VP1u蛋白的表达.结果:成功构建了携带人B19病毒VP1u基因的真核表达载体plRES2-EGFP-VP1u,荧光显微镜下可见pIRES2-EGFP-VP1u转染HeLa细胞后表达EGFP蛋白而发出绿色荧光,Western blot证明VP1u蛋白在HeLa细胞中表达.结论:成功构建了携带人B19病毒VP1u基因的真核表达载体plRES2-EGFP-VP1u并在HeLa细胞中正确表达,为今后B19病毒VP1u基因疫苗的研究奠定基础.  相似文献

构建表达质粒pcDNA3 VP2,将其转染CHO细胞建立了稳定表达的细胞系;用间接免疫荧光法和Western印迹证明了表达的VP2蛋白的特异性。对昆虫杆状病毒系统表达的VP2蛋白作初步纯化。分别用由大肠杆菌、CHO细胞和昆虫杆状病毒表达系统表达的VP2蛋白,以间接免疫荧光法和ELISA法检测人群血清中的VP2抗体,结果表明,间接免疫荧光法的敏感性高于ELISA法。  相似文献

VP2 8是对虾白斑综合症病毒 (Whitespotsyndromevirus ,WSSV)的一个重要的囊膜蛋白。为了便于研究VP2 8在和宿主细胞相互作用过程中扮演的角色 ,将VP2 8基因克隆到一个原核表达系统 ,对原核表达的VP2 8的特性进行了研究 ,并制备了抗VP2 8的多克隆抗体和单链抗体 ,对原核表达的和天然病毒的VP2 8蛋白免疫原性进行了比较。结果表示 ,原核表达的VP2 8与天然蛋白具有相似的免疫原性。  相似文献

犬细小病毒VP2蛋白在真核细胞中的分泌表达及特性   总被引:1,自引:1,他引:0  
摘要:【目的】利用真核细胞分泌表达犬细小病毒VP2蛋白和研究其特性。【方法】为构建犬细小病毒(Canine parvovirus, CPV)VP2基因的真核分泌型表达载体,首先通过酶切从含有人CD5信号肽序列的质粒中将CD5信号肽基因片段切出,将其连接到真核表达载体pcDNA3.1A的多克隆位点上,构建成pcDNA3.1-CD5sp质粒。然后再通过PCR方法从含有犬细小病毒VP2基因的质粒中扩增VP2基因,并将其插入到pcDNA3.1- CD5sp载体中CD5信号肽的下游,构建成VP2基因的真核分泌型表达载体pcDNA-CD5sp-VP2。经磷酸钙介导转染293T细胞,使其在真核细胞中进行分泌表达,并通过ELISA检测表达的VP2蛋白与犬转铁蛋白受体(TfR)结合的活性。【结果】序列分析结果表明,本实验构建的犬细小病毒VP2基因真核分泌型表达载体结构正确,将该表达载体转染的293T细胞,在培养基中通过Western-blot检测到有VP2重组蛋白的存在。经ELISA检测表明表达的重组VP2蛋白具有与犬转铁蛋白受体结合的活性。【结论】 利用人的CD5信号肽实现了犬细小病毒VP2蛋白在真核细胞中的分泌表达,表达的VP2蛋白具有与犬转铁蛋白受体结合的活性。  相似文献

目的:分别克隆人细小病毒B19三个主要蛋白VP1、VP2、NS1全长基因,构建真核表达载体。方法:利用PCR和分子克隆技术,分别将B19病毒vp1、vp2、ns1基因全长片段扩增后,构建带荧光标签的真核表达载体;在人体细胞中表达并通过荧光、RT-PCR和Western Blot、测序等方法鉴定。结果:成功构建了包含B19病毒vp1、vp2、ns1全长基因,并在人体细胞中表达了VP1、VP2、NS1蛋白。结论:人微小病毒B19三个主要蛋白基因得到克隆和表达,为进行相关的研究奠定了基础。  相似文献

为构建能表达FPV VP2蛋白的重组犬2型腺病毒(CAV-2)载体。首先用PCR方法从FPV GT-2株细胞培 养物中扩增出了VP2蛋白基因,将其克隆到真核表达质粒pVAX1中构建了含有FPV vp2基因的表达盒(CMV- VP2-PolyA),将该表达盒酶切后定向克隆到含有CAV-2 E3区的穿梭质粒pVAX△E3中,构建出pVAX △E3VP2。用Sal I Nru I双酶切pVAX△E3VP2,回收含有目的基因表达盒部分,将其定向克隆入含有CAV-2 全基因组的骨架质粒pPoly2-CAV-2中,构建了重组质粒pCAV-2-FPV-VP2。Cla I Asc I酶切pCAV-2-FPV- VP2释放出重组基因组,以此转染MDCK细胞,获得了重组病毒CAV-2-VP2。该重组病毒能使MDCK细胞产生 腺病毒样细胞病变。Western blot检测证实,该重组病毒能表达具有免疫学活性的VP2蛋白。该重组病毒可以有 效地诱导免疫猫产生抗FPV和CAV-2抗体。本实验表明该重组病毒有可能成为一个FPV的疫苗株。  相似文献

摘要:【目的】本研究旨在构建在鸡原代骨骼肌细胞中表达IBDV病毒VP2基因的重组杆状病毒。【方法】从IBDV适应细胞毒中提取RNA,用RT-PCR技术扩增VP2基因,将其克隆到自主构建的杆状病毒转移载体的CMV启动子之下,通过Bac-to-Bac系统获得VP2重组Bacmid,并将其转染Sf9昆虫 细胞,获得了VP2重组杆状病毒。重组病毒经扩增后以50个MOI感染鸡原代骨骼肌细胞,接种72h后裂解细胞收获蛋白。【结果】蛋白样品经SDS-PAGE和Western blot证实VP2蛋白获得表达,分子量约48kDa,与预测蛋白大小一致,且能被IBDV阳性血清所识别。【结论】重组杆状病毒可以有效地将VP2基因导入鸡原代细胞,并在CMV的启动下表达具有抗原性的VP2蛋白,本研究为研制IBDV及其他重要禽类传染病的杆状病毒载体疫苗奠定了基础。  相似文献

将猪细小病毒(Porcine Parvovirus, PPV)vp2基因重组到杆状病毒BacToBac表达系统的pFastBacⅠ质粒中,构建了pFastvp2质粒。在DH10Bac大肠杆菌中,pFastvp2与改造过的苜蓿夜蛾核型多角体病毒(AcNPV)基因组(Bacmid)发生同源重组,从而获得重组穿梭载体Bacmidvp2,转染Sf9细胞得到重组病毒AcNPVvp2。SDSPAGE和Westernblotting分析可见大小约为64kD的特异性带,表明AcNPVvp2在Sf9细胞中成功地表达了PPV VP2蛋白。红细胞凝集试验和间接ELISA进一步证实,表达的VP2蛋白具有与全病毒相同的血凝活性和相似的抗原性。电镜观察VP2蛋白的粗提物,发现VP2蛋白可自行装配成许多病毒样粒子(VLPs)。  相似文献

目的:用免疫荧光法快速检测原核和真核细胞中表达的轮状病毒(RV)外壳蛋白VP4。方法:以抗VP4的抗体为一抗、FITC标记的羊抗豚鼠IgG为二抗,用免疫荧光方法检测在大肠杆菌BL21(DE3)中重组表达的同源RVVP4;检测SA11或Wa株RV感染MA104细胞后不同时间段病毒VP4的合成及其在感染细胞中的分布情况。结果:用免疫荧光法可直接检测到原核细胞中表达的外源蛋白,也可检测到病毒蛋白在真核细胞中的分布情况。结论:免疫荧光法可特异、方便、快速地检测RV VP4在原核和真核细胞中的表达;来源于RV TB—Chen株的VP4抗体可特异性识别同源病毒VP4,交叉识别SA11或Wa株的VP4。  相似文献

A novel DNA sequence has been isolated from a subtraction cDNA library of P19 embryonal carcinoma cells treated with retinoic acid which induces neural differentiation of the stem cells. The cDNA insert (4B) hybridized with a single 1.7 kb mRNA, whose abundance was markedly increased in P19 cells after retinoic acid treatment. The 1.7 kb mRNA was also expressed in the brain, but not in other non-neuronal tissues. A 1.6 kb cDNA insert (4BFL), which was cloned by screening another cDNA library with the 4B probe, encodes a novel protein sequence of 325 amino acids (Mr 36,831). The protein expressed in 4BFL-transfected COS cells was translocated into the nuclei as detected with antibodies against subsequences of the predicted protein. The antibodies stained the nuclei of neurally differentiated P19 cells but not of the undifferentiated stem cells. This novel mRNA encoding the nuclear protein, termed necdin, may represent a useful marker for the differentiation and development of brain cells.  相似文献

MMP19 and MMP23B belong to the Matrix metalloproteases (MMPs) family, which are zinc-binding endopeptidases that are capable of degrading various components of the extracellular matrix. They are thought to play important roles in embryonic development, reproduction and tissue remodeling, as well as in cell proliferation, differentiation, migration, angiogenesis, apoptosis and host defense. However, they are poorly understood in pigs. Here, we obtained the full length coding region sequence and genomic sequence of the porcine MMP19 and MMP23B genes and analyzed their genomic structures. The deduced amino acid sequence shares similar precursor protein domains with human and mouse MMP19 and MMP23B protein, respectively. Using IMpRH panel, MMP19 was mapped to SSC5p12-q11 (closely linked to microsatellite DK) and MMP23B was mapped to SSC8q11-q12 (linked to microsatellite Sw2521). Quantitative real-time PCR showed that MMP19 was abundantly expressed in the liver, while MMP23B was strongly expressed in the ovarian and heart. Furthermore, both genes were all expressed increasingly in prenatal skeletal muscle during development. Three SNPs were detected by sequencing and PCR-RFLP methods, and association analysis indicated that C203T at exon 5 of MMP19 has a significant association with the blood parameters WBC (G/L) and IgG2 (mg/mL) (P<0.05), SNP C131T at exon 3 of MMP23B is significantly associated with the blood parameters HGB (g/L) and MCH (P<0.05), and A150G in exon 4 has no significant association with the economic traits in pigs.  相似文献

The ZAP-70 protein is a member of the Syk/ZAP protein tyrosine kinase family, normally expressed in T cells and NK cells but not found in normal, mature B cells. The protein plays a critical role in the initiation of T-cell signaling. Leukemic cells from patients with B-cell chronic lymphocytic leukemia (B-CLL) that expressed nonmutated immunoglobulin V genes were found to express levels of ZAP-70 protein that were comparable to those detected in T cells of healthy adults. The ZAP-70 protein expression can be evaluated by flow cytometry and may be used as a prognostic marker in B-CLL patients. We modified the method of immunocytochemical assessment of ZAP-70 expression. The traditional two-step method with monoclonal anti-ZAP-70 antibody in the first step followed by FITC-conjugated goat anti-mouse IgG was changed for one-step method with monoclonal anti-ZAP-70 antibody labeled by Zenon Alexa Fluor 488. The method is simple and fast. The major advantage of Zenon labeling technique is its compatibility with simultaneous staining of surface antigens. The cells may be earlier immunostained for CD3, CD19 and/or CD5 to compare of the ZAP-70 kinase expression in B and T cells.  相似文献

采用RT-PCR方法从分泌抗人类白细胞表面分化抗原CD19单克隆抗体的杂交瘤细胞中克隆出VH和VL可变区基因,再通过重叠延伸拼接(spliceoverlap extension)PCR方法在VH和VL可变区基因之间引入连接肽(Gly4Ser)3,体外构建抗人CD19单链抗体(抗CD19-ScFv)基因。将其克隆至表达载体PET28a并在大肠杆菌中表达。SDS-PAGE和Westernblot分析结果表明,抗CD19-ScFv在BL21(DE3)菌中获得表达,重组蛋白的相对分子量为27kD,表达产物以不溶性包涵体形式存在,经过溶解包涵体,镍柱亲和层析纯化和体外复性过程,获得了高纯度的单链抗体片段。流式细胞分析结果证实抗CD19ScFv可与人类白细胞表面的分化抗原CD19结合,保留了鼠源性单抗与CD19结合活性。抗人CD19-ScFv的构建与表达,为下一步针对B淋巴系统恶性肿瘤的靶向治疗奠定了基础。  相似文献

Results from our cloning studies on toxin A indicated that the gene for toxin B resided approximately 1 kb upstream of the toxin A gene. Clone pCD19, which contains the 5-end of the toxin A gene and a small open reading frame, was found to contain 1.2 kb of DNA which, when subcloned, expressed a nontoxic peptide that reacted with toxin B antibodies. The rest of the toxin B gene was located on the 6.8 kb cloned fragment of plasmid pCD19L. The two fragments overlapped 0.8 kb. Lysates containing protein expressed by the 6.8 fragment were cytotoxic and lethal, and were neutralized by toxin B antibody. The two fragments were ligated to give the complete toxin B gene. The protein expressed by the complete gene was cytotoxic and lethal, and showed complete immunological identity with toxin B. Further analysis of the expressed protein and the toxin B gene confirmed our earlier findings showing that toxin B has a molecular weight of 240,000 or greater.  相似文献

CYP2B19 is an arachidonic acid monooxygenase highly expressed in the outer, differentiated cell layers of mouse epidermis. We aimed to establish whether CYP2B19 is the source of epidermal epoxyeicosatrienoic acids (EETs), which are implicated in mechanisms regulating epidermal cornification. We show that primary cultures of mouse epidermal keratinocytes expressed native CYP2B19, as determined by mass spectrometry. Differentiation upregulated CYP2B19 mRNA levels ( approximately 39-fold) detected by real-time PCR, CYP2B19 immunoreactivity detected by Western blotting, and cellular levels of the CYP2B19 product 11,12-EET. Cellular 11,12-EET formed from endogenous arachidonic acid increased preferentially (4- to 12-fold) at Day 4 or 5 of differentiation, compared with undifferentiated (Day 0) keratinocyte cultures. Temporally, these results concur with the maximal levels of CYP2B19 mRNA measured at Day 2 and CYP2B19 immunoreactivity at Day 4. We conclude that while mouse epidermis likely expresses multiple cytochrome P450 enzymes, existing evidence supports native CYP2B19 as being the major source of epidermal EET formation.  相似文献

目的构建结核分枝杆菌Ag85B-Esat6-HspX融合基因,并对其在体外真核细胞中进行表达。方法用PCR法从结核分枝杆菌H37Rv株基因组中分别扩增Ag85B、Esat6、HspX基因,插入到pUC19-T载体,序列测定正确后,将融合基因再次克隆到真核表达载体pcDNA3.1(-)。重组质粒经酶切鉴定并测序正确后,用MegaTran1.0转染293T细胞,并用Western-Blot检测目的蛋白的表达。结果 Western-blot检测到分子量大小约65 kDa的目的蛋白。结论成功地构建了结核分枝杆菌Ag85B-Esat6-HspX融合基因的真核表达载体,且该重组载体可在体外真核细胞中获得特异性的表达。  相似文献

The B19 parvovirus is a cause of bone marrow failure in humans. B19 is toxic to erythroid progenitor cells in vitro. Viral products possibly responsible for toxicity were explored by transfection of cloned B19 genome into HeLa cells. The nonstructural (NS) protein was detected in cells 30 h after transfection. Plasmids containing the B19 genome were transfected with selectable marker genes in stable transformation assays. Plasmids that contained the left side of the B19 genome, which encodes the NS protein of the virus, inhibited antibiotic-resistant colony formation. Transformation occurred when NS protein expression was blocked by mutation. Suppression of transformation by NS protein was not tissue specific, suggesting a role for NS protein in toxicity for nonpermissive cells without parvovirus replication or virion accumulation.  相似文献

经RTPCR从人新鲜扁桃体组织中扩增人高迁移率族蛋白B1(HMGB1)中的Bbox(88~162残基)的cDNA,构建于载体pUC19,经测序后与GenBank中报道的已知序列完全一致,再构建于原核表达载体pQE80LDHFR中,表达并鉴定目的蛋白.经Ni2+亲和层析柱、多粘菌素B层析柱纯化获得高纯度的DHFRHMGB1Bbox蛋白,然后将此重组HMGB1Bbox加入到人外周血单核细胞(PBMCs)中,37℃,5%CO2下,刺激PBMCs6h,用ELISA检测PBMCs释放TNFα、IL6的量,经检测后证明纯化后的重组HMGB1Bbox能显著地刺激PBMCs释放致炎因子TNFα、IL6.HMGB1Bbox的表达及其生物活性的初步研究,为进一步研究HMGB1Bbox的作用机制以及新型抗炎制剂的研发奠定基础.  相似文献
