Abstract— Phenylethanolamine N -methyltransferase (PNMT) and adrenaline (A) have been studied in organ cultures of neonatal rat sympathetic ganglia. Organ culture for 2 days without added nerve growth factor (NGF) caused a fall in noradrenaline (NA) and PNMT contents but there was no change in dopamine (DA) or A contents compared to controls. However, in the presence of dexamethasone, there was a marked increase in both PNMT activity and A content, but no change in NA or DA content. Addition of NGF to cultures stimulated with dexamethasone caused no further significant change in PNMT activity or A content, whereas both NA and DA were increased. Prolonged culture without NGF, in the presence of dexamethasone resulted in reductions in both NA and DA content, but the high levels of PNMT activity and A content were sustained. The results are consistent with the hypothesis that both PNMT and A are not contained in the noradrenergic cell bodies but are located chiefly within the small intensely fluorescent cells in sympathetic ganglia.  相似文献

The objective of this study was to determine if adenosine alters growth of mammary epithelium. Mouse mammary epithelial cells (NMuMG) were cultured in DMEM supplemented with 10% fetal calf serum. After serum starvation for 24h, EGF (0–100ng/ml) and/or adenosine (0–100μm ) was added. Adenosine at concentrations of 1, 10 or 100μm increased DNA synthesis significantly, when compared to control. Addition of epidermal growth factor (EGF) (10ng/ml) into 1 or 10μm adenosine showed the interaction in DNA synthesis between EGF and adenosine. A similar result was observed when 100μm adenosine added to various concentrations of EGF (0–100ng/ml). In the second mammary gland (thoracic) organ culture studies, mammary development scores were increased by adenosine (100μm ), EGF (100ng/ml) and adenosine plus EGF. These results indicate that the purine nucleoside adenosine stimulates mammary epithelial cell growth and interacts with EGF in DNA synthesis of mouse mammary epithelial cells.  相似文献

NGF,BDNF和NT-3在培养鸡胚背根节神经元的表达   总被引:1,自引:0,他引:1  
目的 探讨NGF ,BDNF ,NT - 3在体外培养鸡胚背根节神经元中的表达变化。方法 采用NGF ,BDNF ,NT - 3的兔抗血清分别对培养前后的鸡胚背根节神经元以免疫组化ABC法染色。观察NGF、BDNF和NT - 3在培养前、后鸡胚背根节神经元的表达情况 ,计数并比较培养前、后三种因子免疫阳性神经元百分数。结果 未培养的神经元 ,NGF ,BDNF ,NT - 3的阳性神经元百分数分别是 :10 %± 3%,2 7%± 5 %,2 9%± 7%。培养 48小时后 ,NGF ,BDNF ,NT - 3的阳性神经元百分数分别是 :77%± 6 %,6 4%± 7%,2 4%± 7%。结论 培养后NGF ,BDNF的表达较未培养者增加 (P <0 0 1) ,而NT - 3者则有减少 (P <0 0 5 )。提示在体外培养的鸡胚背根节神经元NT - 3的表达有不同于NGF和BDNF的调节方式。  相似文献

The leading tips of elongating nerve fibers are enlarged into "growth cones" which are seen in tissue culture to continually undergo changes in conformation and to foster numerous transitory slender extensions (filopodia) and/or a veillike ruffling sheet. After explantation of 1-day-old rat superior cervical ganglia (as pieces or as individual neurons), nerve fibers and tips were photographed during growth and through the initial stages of aldehyde fixation and then relocated after embedding in plastic. Electron microscopy of serially sectioned tips revealed the following. The moving parts of the cone, the peripheral flange and filopodia, contained a distinctive apparently filamentous feltwork from which all organelles except membranous structures were excluded; microtubules were notably absent from these areas. The cone interior contained varied forms of agranular endoplasmic reticulum, vacuoles, vesicles, coated vesicles, mitochondria, microtubules, and occasional neurofilaments and polysomes. Dense-cored vesicles and lysosomal structures were also present and appeared to be formed locally, at least in part from reticulum. The possible roles of the various forms of agranular membranous components are discussed and it is suggested that structures involved in both the assembly and degradation of membrane are present in the cone. The content of these growing tips resembles that in sensory neuron growth cones studied by others.  相似文献

Small amounts of nerve growth factor (NGF) were present in the superior cervical ganglion and the iris of the rat. The observations that NGF content in each of the tissues was depleted during organ culture and that more NGF appeared in the media than was originally present in the tissues indicated that synthesis or activation of NGF had occurred in organ culture. Antibody to NGF or the depletion of endogenous NGF retarded growth of new sympathetic axons into irides in organ culture. Exogenously added NGF appeared to enhance the initiation of axonal sprouting and the rate of the ramification of nerve fibres.  相似文献

Abstract— The developmental increases of tyrosine hydroxylase, dopa decarboxylase and doparnine-β-hydroxylase activities in neonatal mouse superior cervical ganglion were prevented by administration of the ganglionic blocking agent chlorisondamine. In addition, ganglionic blockade prevented the normal post-natal increase in ganglion volume and neuron numbers. Inhibition of development followed nicotinic-receptor blockade, but not muscarinic blockade with atropine.  相似文献

Abstract— Paired iris smooth muscles from rabbits were prelabelled either in vitro by incubation for 30 min at 37°C in an iso-osmotic salt medium containing glucose, inositol, cytidine and 32Pi, or in vivo by administration of the isotope intracamerally into each eye 1 h before death. One of the pair was then incubated at 37°C for 10 min in an unlabelled medium containing 10 mm of 2-deoxyglucose and the other was incubated in the presence of norepinephrine (NE) or other adrenergic agents. Triphosphoinositide (TPI) was found to contain more 32P than any other phospholipid (almost 39% of total lipid radioactivity) in both the in vitro and in vivo experiments. NE (50 μm ) increased the loss of 32P from TPI (the TPI effect') by 28–30% in the 32P-labelled muscle. The TPI effect was accompanied by a significant increase in 32P labelling of phosphatidic acid (PA) and phosphatidylinositol (PI) but not phosphatidylcoholine. In this tissue the TPI effect was found to be mediated through α-adrenergic receptors. At 14 days after surgical sympathetic denervation, incorporation of 32P into phospholipids of the denervated muscle increased by an average of 6% over that of the normal muscle. The increase in TPI, PI and PA was 7%, 4% and 9% of that of the control respectively. There was little change in phospholipid content of the denervated muscle. The increase in sensitivity to NE (12.5 μm ) caused by denervation produced about 18% increase in the TPI effect and a 25% increase in the 32P labelling of PA, but not PI. In view of our previous findings on the requirement of the TPI effect for Ca2+, this observation could suggest that an increase in Ca2+ influx, following the interaction between the neurotransmitter and its receptor could stimulate TPI-phosphodiesterase, thus leading to increased PA via increased diglyceride. This denervation-induced supersensitivity to NE appears to be postsynaptic in nature. 32Pi was injected intracamerally into each eye 1 h before electrical stimulation of one of the sympathetic trunks. After stimulation for 30 min there was a significant loss of 32P from TPI and a significant increase in the labelling of PI and PA of the stimulated muscle. It is concluded that TPI and its enzymes could play an important role in neurotransmission at the neuromuscular junction of smooth muscle.  相似文献

The activity of RNA polymerase in the superior cervical ganglia of the neonatal rat has been studied. The characteristics of the activity with respect to enzyme concentration, temperature, time, and ion concentrations are presented. Ionic conditions of the assay favoring polymerase I and polymerase II were each studied in the presence of specific inhibitors of DNA-dependent RNA synthesis. The Km for GTP under either polymerase I or polymerase II conditions was found to be 10?5M, and the relative amounts of poly(A)-containing RNA synthesized under both conditions was 4-5%. A decrease in the activity of RNA polymerase with age has been observed. The time course of increased activity following treatment of the animals in vivo or treatment of the isolated ganglia in organ culture with nerve growth factor is presented. The increase in activity observed after the administration of nerve growth factor is discussed in terms of possible mechanisms of action of nerve growth factor in this tissue.  相似文献

本研究以成人正中神经切割伤后2~3个月的神经干为材料,冰冻切片,用免疫双重染色技术显示了神经生长因子与睫状节神经营养(诱向)因子在再生的周围神经组织中的表达与分布。神经生长因子选用APAAP法.其阳性产物呈红色;睫状节神经营养(诱向)因子选用ABC系统,4氯-1-萘酚显色,阳性产物为褐色。光镜下观察:神经生长因子的阳性反应产物出现在正中神经切割伤后再生的神经纤维中,高倍镜下可见其阳性产物分布在轴索,而在雪旺氏细胞中没能见到呈红色的阳性反应产物;睫状节神经营养(诱向)因子分布在一些细胞体积大、核大呈增生活跃状态的雪旺氏细胞中。红与褐双色反应产物色调清晰,效果较好。研究结果提示:睫状节神经营养(诱向)因子与神经生长因子在人周围神经再生过程中起着十分重要的作用。  相似文献

Abstract— Neonatal rats were treated with testosterone propionate (TP) or isoproterenol (ISO) to determine whether (1) increases in salivary gland mass are associated with alteration of developing sympathetic neurons and (2) whether effects on neuron growth are secondary to altered target mass itself or to increases in salivary growth factors. TP treatment is known to result in salivary tubule hypertrophy and elevated nerve growth factor (NGF) content whereas IS0 treatment results in acinar hypertrophy and no known alteration in NGF. TP treatment increased submaxillary gland weight as well as tyrosine hydroxylase (T-OH) activity, adrenergic neuron numbers and total protein in the innervating superior cervical ganglion (SCG). Unilateral sialectomy prevented the increase in T-OH activity in the SCG, suggesting that the salivary glands were necessary for this effect. T-OH activity and total protein were elevated in the distant sixth lumbar sympathetic ganglion after TP treatment, suggesting that sympathetic development as a whole was affected and that humoral factors may be involved. Salivary gland weight was also elevated following ISO administration, but T-OH activity in the SCG was not affected. These observations suggest that TP treatment increases T-OH activity and sympathetic neuron numbers by alteration of specific salivary humoral growth factor(s).  相似文献

在38只麻醉及人工呼吸的猫,观察到电刺激小脑顶核嘴侧部能引起动脉血压显著升高;肾交感神经放电于刺激期间显著增加。去缓冲神经对刺激顶核所引起的血压反应的幅度和肾交感神经放电均无明显影响,但可明显延长血压反应升高相以及血压恢复期的时间。静脉注射氯庄定引起血压降低、心率减慢及肾交感神经放电的抑制,并能减弱刺激顶核引起的血压反应,但增强了刺激顶核引起的肾神经放电的变化。电解损毁延髓腹外侧面引起血压降低及肾交感神经放电的抑制,然而无论单侧还是双侧损毁延髓腹外侧面都不能阻断刺激顶核所引起的血压和肾交感神经放电的反应。以上结果表明,电刺激顶核能引起明显的心血管反应,其反应的下行性通路可能不通过延髓腹外侧面。  相似文献

研究前列腺组织中神经生长因子(NGF) 的生理学意义。采用原位杂交和免疫组化法, 检测43 例前列腺增生组织, 8 例腺癌组织和8 例正常组织中β-NGFm RNA及其蛋白的表达及分布。结果显示β-NGFm RNA 在正常组织及增生组织中定位于间质细胞, 偶见于上皮细胞中; 而在癌组织中, 上皮细胞和间质细胞有同样强度的β-NGFm RNA染色。其蛋白在良性组织中表达主要着色在间质细胞中,上皮细胞呈弱表达,而癌组织中上皮细胞见着色明显增强(P< 0.05)。NGF的自分泌异常可见是前列腺组织由良性向恶性转变的原因之一。  相似文献

给ICR/JCL品系的成年小鼠1次注射利血平(2mg/kg体重)后24小时,其颌下腺(SMG)的去甲肾上腺素(NA)贮存能够被耗竭。在注射后第12天,NA 含量恢复到利血平处理以前的水平。在恢复过程中,雄性腺体的 NA 含量均为雌性的2倍左右。切断颈上神经节(SCG)的节前神经,雌雄腺体的 NA 含量都下降至对照侧的80%左右。消除节前神经的影响以后,在 NA 的恢复过程中,雄性腺体的 NA 含量仍然是雌性的2倍左右。利用神经生长因子(NGF)生物测定法,我们测得本品系成年雄性小鼠 SMG 的 NGF 含量为雌性小鼠的94倍左右。正如外源性 NGF 能选择性地提高交感神经元内的酪氨酸羟化酶活力与 SMG 的 NA 含量一样,在利血平处理后的恢复过程中,雄性腺体比雌性腺体具有较高的 NA 含量可能是由于内源性 NGF 对交感神经元内酪氨酸羟化酶产生效应的结果。  相似文献

外源性神经生长因子(NGF)对周边交感系统的营养性影响已有广泛的研究(Black,1978;Purves等,1978)。但是,内源性NGF是否同样行使这种影响,至今还没有被人证实(Nja等,1978)。1979年,Carstairs等人利用丙酸睾酮慢性处理,提高小鼠颌下腺内NGF含量以后,观察到颈上神经节(SCG)中有部分神经元肥大,但其它交感神经节不受影响。作者认为SCG内肥大的神经元就是支配颌下腺的那部分交感神经元。由于雄性小鼠颌下腺内的NGF含量是雌性小鼠的92倍(Hirata等,1979),我们实验室直接检查了小鼠颌下腺和虹膜的交感神经支配,发现成年雄性小鼠颌下腺的荧光基丛索远较雌性小鼠的  相似文献

Nerve growth factor (NGF) at 10 B.U./ml produced a 5-fold increase in the concentration of cyclic AMP 10min after addition to freshly excised embryonic chick dorsal root ganglia (DRG). The presence of NGF at 10 B.U./ml, but not 1 B.U./ml, over a 6 h incubation in nutrient-free medium prevented this cyclic AMP increase when the DRG were again challenged with NGF (10 B.U./ml) after the 6 h. Incubation of DRG for 6 h at 37°C without NGF did not prevent the cyclic AMP response from occurring when NGF was presented at this time. The basal cyclic AMP concentration, however, decreased by 65–75% during the course of the 6 h incubation, and the continuous presence of NGF (10 B.U./ml) was unable to prevent this. When NGF was added to 6-h NGF-deprived DRG, the time for maximum cyclic AMP increase decreased from 15min to 6min with increasing NGF concentrations from 1 to 50 B.U./ml, although the relative magnitude of the cyclic AMP increase was essentially the same (approx 3-fold) at these NGF concentrations. Attempts were made to correlate the cyclic AMP response in NGF-deprived DRG with another rapid response resulting from administration of NGF to NGF-deprived DRG, namely, the reactivation of hexose uptake. The cyclic AMP and permeation responses both occurred on the same time scale (5-15 min), and both responded to increasing concentrations of NGF with a decreasing time to achieve maximal effect. A temporal relationship between cyclic AMP and membrane permeability was noted, but it could not be ruled out that it might reflect difficulties in methodology and/or inadequacy in current knowledge of the underlying mechanisms.  相似文献

目的研究对兔右坐骨神经损伤后局部给予蛇毒神经生长因子(NGF),观察坐骨神经酶活性变化和超微结构的恢复情况,探讨NGF对神经再生的影响.方法乙酰胆碱酯酶(AChE)、酸性磷酸酶(ACPase)的酶组织化学技术和电镜技术.结果神经损伤后:AChE活性明显下降,NGF组的AChE活性恢复快于盐水对照组;ACPase活性逐渐增高,NGF组的ACPase活性恢复时间短于盐水对照组.坐骨神经的超微结构在神经损伤后也发生变化,NGF组的变化程度小于盐水对照组,恢复时间短于对照组.结论NGF可通过影响酶物质的代谢而起到加快受损神经恢复的作用.为临床上应用蛇毒NGF治疗周围神经损伤提供形态学依据.  相似文献

尖吻蝮蛇毒对蟾蜍交感神经节细胞电活动的影响   总被引:1,自引:0,他引:1  
用细胞内电位记录技术,以离体蟾蜍交感链为标本,观察了尖吻蝮蛇毒(AAV)的神经毒性作用。结果表明,该蛇毒(10-200μg/ml)对交感神经元的静息电位、膜电阻和膜电容没有显著的作用,对动作电位形状也无可测出的影响,但能使阈电位轻皮升高,即使神经元兴奋性稍降低。AAV(>25μg/ml)对胆碱能性的快兴奋性突触后电位有剂量依从性的,部分可逆性的抑制作用。其作用机制至少应部分归之于AAV对突触后膜上的N型胆碱能受体的阻断作用。本研究发现的AAV神经毒性作用可能有一定实践意义。  相似文献
