Malondialdehyde(MDA)is a well known inducer of carbonyl stress in a variety of human cells,however,its effects on human bone marrow mesenchymal stem cells(hMSCs)have not been documented.In this study,the effects of MDA concentration on the growth rate and proliferation of hMSCs in vitro were assessed.Under high concentrations of MDA,the cell count was decreased and the population doubling time(PDT)was lengthened.Flow cytometry(FCM)demonstrated that MDA triggered cells to undergo apoptosis.in parallel with the findings in MTT[3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide]assay which showed that it can also impair cellular viability.Surprisingly,FCM also determined that the percentage of hMSCs in G2/M- and S-phases also increased in a dose-dependent manner with respect to MDA concentration.These results strongly suggest that even though hMSCs were severely impaired by high concentrations of MDA,they were still able to send signals that resulted in accelerated cellular proliferation process.This study provided important insights on how carbonyl stress affects cell cycle and proliferation of hMSCs. 相似文献

The germination of laser-irradiated Chinese pine seeds was carried out under drought stress. The activities of superoxide
dismutase (SOD) and peroxidase (POD), and the content of malondialdehyde (MDA) were determined. Results showed notably increased
germination percentage, root length, vitality index and fresh weight. The SOD and POD protective enzyme system activity of
the Chinese pine seedlings obviously rose. It can be concluded that the germination and juvenile resistance of Chinese pine
seeds under drought stress are enhanced after laser processing.

Five ethanolic extracts from the mycelia of Ganoderma lucidum, G. tsugae, G. oerstedii, G. subamboinense, and G. resinaceum were respectively studied on their anticancerous activities against leukemic HL-60 cell line in vitro. Results showed that
all five extracts potently inhibited HL-60 proliferation. The extract from G. lucidum mycelia exerted the highest activity. Annexin V/PI bivariate flow cytometric analysis further revealed that the five extracts
significantly induced early apoptosis in HL-60 cells. The results illustrate that not only G. lucidum but also other Ganoderma species can inhibit cancer cells, and their mechanisms are related to induction of apoptosis.

The effects of hematopoietic stem/progenitor cells (HSPCs) expanded in the two step coculture with human bone marrow mesenchymal
stem cells (hMSCs) on the hematopoietic reconstruction of irradiated NOD/SCID mice were studied. Mononuclear cells (MNCs)
were isolated from human umbilical cord blood (UCB) and cultured in the non-coculture scheme of rhSCF + rhG-CSF + rhMDGF combination
and the coculture scheme of rhSCF + rhG-CSF + rhMDGF + hMSCs. Sublethally-irradiated NOD/SCID mice were transplanted with
ex vivo expanded HSPCs with the dose of 8.5 × 106 cells per mouse. After transplantation, the dynamics of WBC in the transplanted mice was measured periodically, and the Alu
sequence fragment special for human in the transplanted mice was inspected by PCR. Results showed that the coculture scheme
increased proliferation of UCB-derived HSPCs. After transplantation with expanded HSPCs, the population of WBC in the transplanted
mice increased in 12 d and reached the first peak in 25 d, then showed the second increasing of WBC in 45∼55 d. Expanded cells
from the coculture scheme appeared to be favorable for the second increasing of WBC in the transplanted mice. After 85 d,
the Alu sequence fragment was detected in the probability of 87.5% (7/8) for the non-coculture scheme and 88.9% (8/9) for
the coculture scheme.

Abstract We evaluated the inhibitory effect of 3,7-dinitrodibenzobromonium salts (cBr) on the proliferation of human chronic
myelogenous leukemia K562 cell by trypan blue exclusion test and MTT colorimetric assay. The degree of DNA damage in K562 cells treated with cBr, was detected by isotopic tracer method (3H-TdR). The morphological changes of these K562 cells were examined by fluorescence and electron microscopy. Biochemical characteristics of K562 cells were detected by flow cytometry and 3H-thymidine incorporation assay. Findings indicated that cBr could significantly inhibit cell proliferation and result in
DNA damage of K562 cells. cBr is a new type of immunostimulant and can induce cell apoptosis.

Carboxymethytl pachymaram (CMP) was administered to HBV transgenic mice through abdominal injection. Lymphocytes were extracted
from the spleens. MTT method was used to detect cytotoxicity of CMP. Dendritic cells (DCs) were separated from lymphocytes
and incubated with granulocyte-macrophage colony-stimulating factor (GM-CSF) and interleukin-4 (IL-4). Phenotypes of DC’s
were assayed by flow cytometry (FCM). IL-12 released by DCs and IL-10 and IFN-γ produced by T cells in mixed lymphocyte reaction (MLR) were measured using ELISA. Results showed that CMP within the concentration
of 0–500 μg/mL did not produce cytotoxicity to lymphocytes and could even increase DC phenotypes, and IL-12 level in HBV transgenic
mice. It could also increase the secretion of IFN-γ, and inhibit the secretion of IL-10 inMLR. Thus it can up-regulate DC function.

Foamy viruses (FVs) have broad cellular tropism infecting vertebrates from fish to human being, which indicates that Env protein
has a high capability for membrane fusion. Conservative features in all FV transmembrane (TM) proteins include a region of
hydrophobic domain called membrane-spanning domain (MSD), which contains several stretches of hydrophobic amino acids. To
investigate whether these features were associated with the cytotoxicity effect of TM on Escherichia coli, a series of mutants were constructed and expressed in the E. coli BL21 (DE3) using pET-32a (+) as expressing vector. The results showed that only TM3 without MSD was expressed in E. coli, whereas the other two containing full or part of the MSD (TM1 and TM2) could not be expressed. Furthermore, the bacterial
amount and living bacteria analysis revealed that the cytotoxicity of TM was dependent on its MSD, especially on the stretches
of hydrophobic amino acids. Western blotting analysis showed that TM3 protein was purified with affinity purification.

Acute-phase response is documented to be a significant mechanism of innate immunity in vertebrates and invertebrates. In this
study, proteomic methodologies were applied for different protein expressions in hemolymph of Scylla serrata challenged by Vibrio parahaemolyticus after immunization, and in muscles of the crabs separately challenged by V. parahaemolyticus, V. anguillarum and Aeromonas hydrophila. Up-regulated cryptocyanin is documented in the hemolymph and up-regulated calexcitin, wingless (fragment) and tachykinin-related
peptide in the muscle as acute-phase proteins. All the four altered proteins were responsible for bacterial stress, but cryptocyanin
seemed to be a memory response protein against the challenge by a live bacterium after immunization of the live cells. These
up-regulated proteins can be indicative of an understanding of immunity of a crab.

Seven alkaloids were isolated from the seeds of Ammopiptanthus mongolica by thin layer chromatography and silica gel column chromatography, and the chemical structures of five alkaloids, 17-oxosparteine,
β-isosparteine, 3α-hydroxysparteine, sparteine, and 3β-hydroxysparteine were identified by 1H nuclear magnetic resonance (NMR) and electron ionization mass spectrum (EIMS).

Recent collections from six sections in Lanongla area, Tethyan Himalaya allow the establishment of four buchia assemblages. In ascending order, they are Buchia-Buchia spitiensis, Buchia masquensis-Buchia rugasa, Buchia blanfordiana, Buchia piochii and Buchia subokensis assemblages. These Buchia assemblages first demonstrate that not only the Upper Jurassic strata but also the highest Buchia assemblage-Buchia subokensis, which appeared in Lower Cretaceous strata all over the world are present in Lanongla area. This first records the highest
Buchia assemblage in Lanongla area.

To obtain an anti-tumor peptide of Tumstatin and detect its biological activity, the nucleotide sequence encoding 185–203
amino acids (19peptide) of Tumstatin was synthesized and inserted into the fusion protein vector pTYB2. After identification
by sequencing and restriction endonucleases, the recombined vector was transformed into BL-21 (DE3) E. coli competent cells. Transformed E. coli BL-21 (DE3) were induced by isopropyl-β-thiogalactopyranoside (IPTG), and then expressed. By 1,4-dithiothreitol (DTT) reduction,
the soluble 19peptide was obtained from a chitin affinity chromatograph. The biological activity of 19peptide was determined
by 3-[4,5-dimethylthiazol-2-y1]-2,5-diphenytetrazolium bromide (MTT) assay, cell growth curve, the effect of the ascitic fluid
transfevent H22 hepatoma on mice and via histopathological slices. The purified 19peptide directly inhibited proliferation
and migration of murine B16 melanoma cells, SMMC-7721hepatoma carcinoma cells and human umbilical vein endothelial cells (HUVEC).
The tumor inhibition rate of mice ascitic fluid transfevent H22 hepatoma was 48.46%. Histopathological slices showed that
it could promote tumor tissue necrosis and decrease the density of blood vessels. With higher anti-tumor activity, 19peptide
has the potential to become a novel, potent anti-tumor agent.

After 20 years of dedicated research, Jiafuzhan has been successfully developed under the new technologies in breeding high-quality
early indica rice cultivars. Its rice quality has almost reached the A-level Editable Rice of Agriculture Department of China, and its
average production reaches 400–500 kg/(666.7 m2). This new cultivar also has other characteristics such as enhanced resistance of blast and fallen, steady productivity,
and strong adaptability. Jiafuzhan has been put into production of over 11.4 × 104 hm2 in Fujian Province and has been introduced and extended in other Provinces like Jiangxi, Guangdong, and Guangxi, China. The
successes of breeding Jiafuzhan is a solution to the existing perennial problems in the rice industry, such as poor grain
quality of big-grain rice and early indica rice, low productivity, and poor blast resistance of elite rice.

Proteins are fundamental components of all living cells and the protein-protein interaction plays an important role in vital
movement. This paper briefly introduced the original Resonant Recognition Model (RRM), and then modified it by using the wavelet
transform to acquire the Modified Resonant Recognition Model (MRRM). The key characteristic of the new model is that it can
predict directly the protein-protein interaction from the primary sequence, and the MRRM is more suitable than the RRM for
this prediction. The results of numerical experiments show that the MRRM is effective for predicting the protein-protein interaction.

This paper is based on the fieldwork in Xishuangbanna Natioanl Nature Reserve in Yunnan Province of China. GPS data of Asian
elephants were collected and analyzed with the remote sensing satellite photos of the region to estimate the landform physiognomy
of different colors. We also analyzed a series of ecological factors including altitude, landform, relief, villages and roads
which affected the distribution and movement of Asian elephants. The results suggested the possibility of designing and establishing
corridors in Xishuangbanna National Nature Reserve to protect the population of wild elephants in the region.

The rhizosphere environment of tea (Camellia sinensis Kuntze) intercropped with persimmon (Diospyros kaki) differs from monocultures of tea. A trial was conducted to determine the effects of intercropping with persimmon on root
exudates and soil nutrient condition of tea. Amino acid exuded in intercropping was three times higher than that in monoculture.
Phenol, phenol/amino acid ration, dissolved sugar, and total organic acid were also lower in intercropping. The value of pH
in soil was higher, and soil nutrient condition of rhizosphere, especially available nutrient, was not as well in intercropping
as that in tea grown alone. While soil nutrient of non-rhizosphere was better than that in monoculture, tea quality and soil
nutrient condition were better in intercropping ecosystem.

We induced 3 cell lines that were in vitro cultured from Lepidoptera with heat inactivated Escherichia coli DH5α to stimulate the antibacterial peptide followed by antibacterial activity assay, induction dynamic research and Tricine sodium
dodecyl sulfate-polyacrylamide gel electrophoresis (Tricine SDS-PAGE) experiment. The antibacterial activity of the induced
BTI-Tn-5B1 cell line was the highest, and the antibacterial activity increased gradually to the highest level in 16 hours
after stimulation. A new antibacterial peptide with a molecular weight of about 8000 Da was preferentially induced in Trichoplusia ni BTI-Tn-5B1 cells in 16 hours after stimulation. Antibacterial activity assays indicated that it had inhibition against Staphylococcus aureus, Escherichia coli K12D31 and Salmonella derby. It has especially strong inhibition against Gram-negative bacteria such as Escherichia coli K12D31 and Salmonella derby.

Chinese Cupressus L. includes five species. The molecular phylogenetic relationship of the Cupressus species and Chamaecyparis L. were determined by comparing 417–479 bp of chloroplast petG-trnP intergenic spacer sequence. In PAUP* analysis, Platycladus orientalis was used as the functional out group. By using the maximum likelihood method 1 077 trees were examined and the result showed
that one tree had a best score of -Ln=2 232.47. The phylogenetic tree clearly showed that Chamaecyparis nootkatensis was diverged from other Chamaecyparis species. Based on the results, together with evidences from other aspects, we consider that Cupressus funebris and Chamaecyparis nootkatensis should be placed in the genus Cupressus. The use of cpDNA intergenic spacer petG-trnP in Cupressus was also discussed.
