The aldehyde dehydrogenase activity of the sulfate-reducing bacterium Desulfovibrio simplex strain DSM 4141 was characterized in cell-free extracts. Oxygen-sensitive, constitutive aldehyde dehydrogenase activity was found in cells grown on l(+)-lactate, hydrogen, or vanillin with sulfate as the electron acceptor. A 1.83- to 2.6-fold higher specific activity was obtained in cells grown in media supplemented with 1 μM WO4 2–. The aldehyde dehydrogenase in cell-free extracts catalyzed the oxidation of aliphatic (K m < 20 μM) and aromatic aldehydes (K m < 0.32 mM) using methyl viologen as the electron acceptor. Flavins (FMN and FAD) were also active and are proposed to be the natural cofactors, while no activity was obtained with NAD+ or NADP+. 185WO4 2– was incorporated in vivo into D. simplex; it was found exclusively in the soluble fraction (≥ 98%). Anionic-exchange chromatography demonstrated coelution of 185W with two distinct peaks, the first one containing hydrogenase and formate dehydrogenase activities, and the second one aldehyde dehydrogenase activity. Extracts from Cleome rosea were investigated for their activity against acyclovir-resistant strains of Herpes simplex type 1 (ACVr-HSV-1) and type 2 (ACVr-HSV-2). Methanolic and acidified (1% (v/v) HCl) methanolic extracts were prepared from field-grown plants and in vitro propagated plants, as well as from calli and cell suspension cultures. The extracts presented low cytotoxicity and caused virus titer reduction above 70%, with different mechanisms of action. Extracts from leaves of field-grown plants inhibited viral infection mainly by affecting the virus particle itself (virucidal effect), while extracts from calli acted mainly on cell receptors. On the other hand, all extracts evaluated affected the virus entry across the cell membrane and the intracellular viral replication at similar percentages, causing reduction on titers in the range of 68–90%. This study validated the potential use of in vitro materials as sources of antiherpetic agents from C. rosea.  相似文献

Purpose of the present study was to evaluate antioxidant, antibacterial, antifungal, and antiviral activities of the petroleum ether, chloroform, ethyl acetate and methanol extracts as well as the alkaloid fraction of Lycopodium clavatum L. (LC) from Lycopodiaceae growing in Turkey. Antioxidant activity of the LC extracts was evaluated by 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical-scavenging method at 0.2 mg/ml using microplate-reader assay. Antiviral assessment of LC extracts was evaluated towards the DNA virus Herpes simplex (HSV) and the RNA virus Parainfluenza (PI-3) using Madin-Darby Bovine Kidney (MDBK) and Vero cell lines. Antibacterial and antifungal activities of the extracts were tested against standard and isolated strains of the following bacteria; Escherichia coli, Pseudomonas aeruginosa, Proteus mirabilis, Acinobacter baumannii, Klebsiella pneumoniae, Staphylococcus aureus, Bacillus subtilis as well as the fungi; Candida albicans and C. parapsilosis. All of the extracts possessed noteworthy activity against ATCC strain of S. aureus (4 μg/ml), while the LC extracts showed reasonable antifungal effect. On the other hand, we found that only the chloroform extract was active against HSV (16–8 μg/ml), while petroleum ether and alkaloid extracts inhibited potently PI-3 (16–4 μg/ml and 32–4 μg/ml, respectively). However, all of the extracts had insignificant antiradical effect on DPPH. In addition, we also analyzed the content of the alkaloid fraction of the plant by capillary gas chromatography-mass spectrometry (GC-MS) and identified lycopodine as the major alkaloid.  相似文献

A vitamin B12-dependent N5-methyltetrahydrofoIate-homocysteine methyltransferase was found in cell-free extracts of Corynebacterium simplex ATCC 6946 grown aerobically in a medium containing hydrocarbon as a sole carbon source and the enzyme was partially purified. Absolute requirements for S-adenosylmethionine and an appropriate reducing system were observed for the transmethylation from N5-methyltetrahydrofolate. The same preparation catalyzed also the formation of methionine from homocysteine and methyl-B12 under both aerobic and anaerobic conditions. The concentration of cobalt ion in the growth medium had a pronounced effect on the intracellular vitamin B12 level and the activity of the vitamin-dependent methionine-synthesizing system in the bacterium. The relationship between the methionine synthesis and the methyl branched-chain fatty acid formation was discussed.  相似文献

Novel 1-(1-aryl-4,5dihydro-1H-imidazoline)-3-chlorosulfonylourea derivatives 3a3f were synthesized in the reaction of 1-aryl-4,5-dihydro-1H-imidazol-2-amines with chlorosulfonyl isocyanate. The second series of compounds 4a4f was prepared from the respective 1-(1-aryl-4,5-dihydro-1H-imidazoline)-3-chlorsulfonylureas 3a3f and 1,1′-carbonyldiimidazole (CDI). The selected compounds were tested for their activity against Herpes simplex virus and coxsackievirus B3 (CVB3). It was determined that three derivatives, i.e 3d, 4a and 4d are active against Herpes simplex virus (HSV-1). Compounds 3d and 4c are active against CVB3. Their favorable activity can be primarily attributed to their low lipophilicity values. Moreover, the lack of substituent in the phenyl moiety or 4-methoxy substitution can be considered as the most beneficial for the antiviral activity.  相似文献

This paper describes the activity against Anisakis simplex s.l. L3 larvae of six monoterpenic derivatives obtained from different essential oils, (α-pinene, β-pinene, ocimene, myrcene, geranyl acetate, and cineole). In in vitro assays, α-pinene, ocimene and cineole showed high activity at a concentration of 125 μg/mL (48 h) but only α-pinene and ocimene were active at 62.5 μg/mL. In in vivo assays, L3 larvae and study compounds were simultaneously administered per os to Wistar rats. The most active compound was α-pinene, finding lesions in only 20% of treated rats versus 98% of controls. Further in vivo studies are required to investigate whether addition of these compounds to food could have a prophylactic effect, reducing the pathogenicity of A. simplex s.l. L3 in humans, and to explore any possible synergy among compounds.  相似文献

Sedum album and Pelargonium zonale extracts do not show any peroxidase activity. Both extracts provoke a lag phase in the horse-radish peroxidase-catalyzed oxidation of guaiacol by H2O2. Preincubation of Sedum album extract with ascorbate oxidase eliminated completely the lag phase. Ascorbic acid has been identified as the substance responsible for this lag phase by reacting with a coloured intermediary product of the analytical reaction. In the Pelargonium zonale extract, the lag phase seems to be due to competitive inhibitors of peroxidase, which are of a phenolic nature.  相似文献

Presence of a non-acidic, growth-promoting indole compound, identical in Rf and colour reaction and closely similar in UV spectrum to synthetic indoleacetaldehyde (IAAId) has been demonstrated in extracts of etiolated shoots of Pisum and Helianthus. On chemical as well as enzymic oxidation of this neutral extract, activity passes over into the acidic fraction. This neutral substance is believed to be IAAId and occurs in the free state as short-term, low temperature extractions indicate. On the basis of Rf, colour reaction and UV spectrum, tryptophol has also been identified in Helianthus extracts. Its identification in pea extracts, however, rests only on Rf, colour reaction and inactivity in Avena bioassays.  相似文献

Aims: To investigate the in vitro antiviral activity of Distictella elongata (Vahl) Urb. ethanol extracts from leaves (LEE), fruits (FEE), stems and their main components. Methods and Results: The antiviral activity was evaluated against human herpesvirus type 1 (HSV‐1), murine encephalomyocarditis virus (EMCV), vaccinia virus Western Reserve (VACV‐WR) and dengue virus 2 (DENV‐2) by the 3‐(4, 5‐dimethylthiazol‐2‐yl)‐2,5‐diphenyltetrazolium bromide (MTT) colorimetric assay. LEE presented anti‐HSV‐1 [EC50 142·8 ± 5·3 μg ml?1; selectivity index (SI) 2·0] and anti‐DENV‐2 activity (EC50 9·8 ± 1·3 μg ml?1; SI 1·5). The pectolinarin ( 1 ) isolated from LEE was less active against HSV‐1 and DENV‐2. A mixture of the triterpenoids ursolic, pomolic and oleanolic acids was also obtained. Ursolic and oleanolic acids have shown antiviral activity against HSV‐1. A mixture of pectolinarin ( 1 ) and acacetin‐7‐O‐rutinoside ( 2 ) was isolated from FEE and has presented anti‐DENV‐2 activity (EC50 11·1 ± 1·6 μg ml?1; SI > 45). Besides the antiviral activity, D. elongata has disclosed antioxidant effect. Conclusions: These data shows that D. elongata has antiviral activity mainly against HSV‐1 and DENV‐2, besides antioxidant activity. These effects might be principally attributed to flavonoids isolated. Significance and Impact of the Study: Distictella elongata might be considered a promising source of anti‐dengue fever phytochemicals.  相似文献

The main aim of this work was to assign the cuticular lipids identified in a parasitic nematode and to distinguish those originating from its host. The hypothesis that long-chained fatty acids and sterols are imported by the parasite in the absence of certain enzymes was also tested. The organisms (Anisakis simplex and Gadus morhua) were extracted in petroleum ether and dichloromethane. Matrix-assisted laser desorption time-of-flight mass spectrometry (MALDI-TOF) was used to identify unknown components, and electrospray ionization mass spectrometry (ESI/MS) to verify recognized groups of lipids. The lipid classes identified in the surface layer were free saturated and unsaturated fatty acids, triacylglycerols, sterols and non-polar sphingolipids (ceramides, sphingoid bases). The most abundant fraction consisted of fatty acids. The predominant saturated acids were tetradecanoic acid in the petroleum ether extract of A. simplex, hexadecanoic acid in the dichloromethane extract of A. simplex, and also the polyunsaturated octadecahexaenoic and octadecatrienoic acids in both extracts of the parasitic nematode. The mass spectrum revealed the presence of fatty acids with different numbers of carbons, and with odd and even numbers of unsaturated bonds. The MALDI-TOF mass spectrum also identified triacylglycerols (TAGs). The dominant short-chain TAGs were CoCoCy:1, CoCoPg and Bu0:0B:6. The majority of TAGs were found in the ether and dichloromethane extracts of A. simplex. Sterols were the least common class of lipids found in the nematode extracts; most likely, this is the fraction that is entirely incorporated from the host organism because of the parasite’s inability to synthesize them. MALDI-TOF also identified non-polar sphingolipids - ceramides and sphingoid bases. The signals due to N-octanoyl-d-erythro-octasphinganine (m/z 288.3) and N-tetranoyl-d-erythro-tetradecasphinganine (m/z 316.4) were dominant on the mass spectra; quite a large number of short-chain non-polar sphingolipids were also identified.  相似文献

A preparation of living Arthrobacter simplex cells immobilized in polyacrylamide gel, which showed steroid-Δ1-dehydrogenase activity, was studied. The entrapped microorganisms catalyzed the transformation of cortisol to prednisolone and this reaction was followed spectrophotometrically or with the aid of thin layer chromatography (TLC) and high pressure liquid chromatography (HPLC). About 40% of the original activity found with free bacteria was retained after immobilization. The steroid dehydrogenase activity of polyacrylamide-entrapped A. simplex could be raised to a minor extent in alcoholic solvents or by addition of a cofactor such as menadione. On incubation in various nutrient media, on the other hand, the activity could be increased considerablyl, usually 7–10 times. Possible causes for the observed increase in activity have been investigated, and microbial growth of the original entrapped microorganisms appears to be the major reason. Frozen activated preparations of immobilized A. simplex showed only a small loss of activity on storage for at least four months. A semicontinuous batch wise operation with immobilized A. simplex in different nutrient media was carried out. At the end of the experiment the steroid transformation capacity was 0.5 g steroid per day per g gel (wet weight).  相似文献

Synthetic substance P stimulated adenylate cyclase activity in particulate preparations from rat and human brain.The concentration of substance P for half maximal stimulation in rat brain was 1.8 · 10−7 M.The stimulatory effect of substance P on the rat brain adenylate cyclase activity was 88% compared with 48% by noradrenalin, 163% by prostaglandin E1 and 184% by prostaglandin E2.Both the basal and substance P-stimulated adenylate cyclase activity in rat brain were inhibited by concentration of Ca2+ above 10−6 M.The chelating agent ethyleneglycol-bis-(β-aminoethylether)-N,N′-tetraacetic acid at a concentration of 0.1 mM reduced the basal adenylate cyclase activity by 64% and eliminated the substance P-stimulated activity.The inhibition by ethyleneglycol-bis-(β-aminoethylether)-N,N′-tetraacetic acid was completely reversed by increasing concentrations of Ca2+.  相似文献

Three species, Zygophyllum album L., Z. coccineum L. and Z. simplex L., from family Zygophyllaceae were collected from two locations in Egypt to study their response to environmental conditions. Organic solutes (amino acids, soluble proteins and soluble sugars) and inorganic solutes (Na+, K+, Ca2+, Mg2+, Cl?, PO43? and SO42?) were estimated to study their role in osmotic adjustment under the effect of drought and salinity. The study showed that Z. coccineum is most tolerant for drought and salinity than Z. simplex. Z. coccineum was dependent on soluble proteins and soluble sugars, to increase its content of bound water, to undergo water deficit in desert. Z. simplex accumulated inorganic solutes more than Z. coccineum and less organic solutes. Amino acids content increased in Z. coccineum and Z. simplex survived in saline conditions to play a role in osmotic adjustment. Under the effect of salinity, all the studied species showed a tendency and high capacity to accumulate inorganic solutes. The main inorganic salutes were Ca2+, Mg2+ and Cl?. The role of Na+ was less than Ca2+ and Mg2+. Z. album and Z. simplex preferred Mg2+ more than Z. coccineum which preferred Ca2+.  相似文献

The bacterium formerly known asArthrobacter globiformis 193 has high 1(2)-dehydrogenase activity toward pharmaceutically important steroids, 9(11)-dehydrocortexolone in particular. The complex analysis of the morphostructural, physiological, biochemical, and phylogenetic properties of this bacterium allowed us to reclassify it into Nocardioides simplex (N. simplex VKM Ac-2033D).  相似文献

Cell-free extracts of Lactobacillus plantarum contain non-proteinaceous compounds which mimic superoxide dismutase activity. Using the test system in which O 2 is generated by xanthine oxidase, superoxide dismutase activity is found in cell-free extracts, where proteins are removed by precipitation. This activity is strongly decreased after dialysis of cell-free extracts. Superoxide dismutase activity was also investigated by means of pulse radiolysis. Cell-free extracts of Escherichia coli were also investigated as a comparison, which were known to contain superoxide dismutase. With cell-free extracts of both L. plantarum and E. coli the decay of O 2 was markedly increased. However, the type of reaction of the O 2 decay was of first order in the presence of E. coli extracts due to superoxide dismutase(s), and of second order in the presence of L. plantarum extracts, indicating that O 2 elimination is not an enzymic reaction. Mn2+ phosphate(s) might be responsible for the observed elimination of O 2 . The production of O 2 is not detectable during NADH-, lactate- or pyruvate oxidase reactions in L. plantarum extracts.  相似文献

A substance inducing teliospore production inPuccinia racondita f. sp.tritici was found in water and methanol extracts of wheat leaves with telia of the wheat leaf rust just before harvest time. Methanol (MeOH) and water extracts from uninfected wheat leaves also showed telia-inducing activity. However, the MeOH and water extracts from wheat leaves covered with telia showed much stronger activity than those from uninfected wheat leaves. We obtained a fraction (0.2 mg) showing activity at 2 ng/ml by purification of the water extract.  相似文献

In the present study, we investigated the protective effect of Quercus aliena acorn extracts against CCl4-induced hepatotoxicity in rats, and the mechanism underlying the protective effects. Aqueous extracts of Quercus aliena acorn had higher superoxide radical scavenging activity than other types of extracts. The Quercus aliena acorn extracts displayed dose-dependent superoxide radical scavenging activity (IC50 = 4.92 μg/ml), as assayed by the electron spin resonance (ESR) spin-trapping technique. Pretreatment with Quercus aliena acorn extracts reduced the increase in serum aspartate aminotransferase (AST) and serum alanine aminotransferase (ALT) levels. The hepatoprotective action was confirmed by histological observation. The aqueous extracts reversed CCl4-induced liver injury and had an antioxidant action in assays of FeCl2- ascorbic acid induced lipid peroxidation in rats. Expression of cytochrome P450 2E1 (CYP2E1) mRNA, as measured by RT-PCR, was significantly decreased in the livers of Quercus aliena acorn-pretreated rats compared with the livers of the control group. These results suggest that the hepatoprotective effects of Quercus aliena acorn extract are related to its antioxidative activity and effect on the expression of CYP2E1.  相似文献

Virus-free individuals of the plant-hopper Javesella pellucida (Fabr.) infected plants with European wheat striate mosaic virus (EWSMV) after being injected at 5° C. with extracts of either plants or hoppers, but extracts of hoppers provided a better inoculum. Hoppers were unable to infect plants until at least 8 days at 20–25° C. after they were injected, and nymphs fed on infected plants similarly required 8 days before they gave infective extracts. Few hoppers survived more than a week after injection with untreated extracts of hoppers or with material sedimented from them by centrifuging the extracts at 8000g, but 60–70% survived injection with purer virus preparations. Injection of the virus seemed harmless, because as many hoppers survived CO2 anaesthesis + injection, whether or not they later infected plants, as survived anaesthesis without injection. Attempts to determine the properties of the virus in vitro gave inconsistent results, but virus from hoppers was still infective after 10 min. at 30° C, 36 hr. at 5° C, precipitation at pH 4.0, storage for several months at -15° C, or at a dilution equivalent to 0.0014 g. hopper/ml. The best extraction medium contained 0.2 M-Na2HPO4+ ascorbic acid + 0.01 M-DIECA at pH 7.0–7.3. In sucrose density-gradients, EWSMV sedimented more slowly than tobacco mosaic virus. No specific particle with which infectivity could be correlated was seen by electron microscopy.  相似文献

Antimicrobial, antioxidant, and antiviral activities of flower extracts of Retama raetam Forssk. Webb (Fabaceae) were screened both from standard and isolated Gram-positive and Gram-negative bacteria by solid medium technique. Oxacillin, Amoxicillin, Ticarcillin, Cefotaxim, and Amphotericin were used as the control agents. The antiviral activity was evaluated against human cytomegalovirus (HCMV) strain AD-169 (ATCC Ref. VR 538) and coxsackie B virus type 3 (CoxB-3) using a cytopathic effect (CPE) reduction assay. The antioxidant activity was evaluated using two tests: 1,1-diphenyl-2-picrylhydrazyl (DPPH) free-radical scavenging and the ammonium thiocyanate methods. All extracts were characterized quantitatively for the presence of polyphenols, flavonoids, and tannins. Of the extracts tested, butanol and ethyl acetate extracts showed important antibacterial activity against Gram-positive and Gram-negative bacteria but only moderate antifungal activity. Methanol extract exhibited moderate antiviral activity against HCMV with IC50 of 250 μg/ml. Ethyl acetate, chloroform, and methanol fractions were found to cause significant free-radical-scavenging effects in both assays. These results may suggest that R. raetam flowers could be used as a natural preservative ingredient in the food and/or pharmaceutical industries.  相似文献

Aims: To evaluate the antiviral activity of Bignoniaceae species occurring in the state of Minas Gerais, Brazil. Methods and Results: Ethanol extracts of different anatomical parts of bignoniaceous plant species have been evaluated in vitro against human herpesvirus type 1 (HSV‐1), vaccinia virus (VACV) and murine encephalomyocarditis virus (EMCV) by the 3‐(4,5‐dimethylthiazol‐2‐yl)‐2,5‐diphenyltetrazolium bromide assay. A total of 34 extracts from 18 plant species selected according to ethnopharmacological and taxonomic criteria were screened. Fifteen of the 34 extracts (44·1%) have disclosed antiviral activity against one or more of the viruses assayed with EC50 values in the range of 23·2 ± 2·5–422·7 ± 10·9 μg ml?1. Conclusions: Twelve of the 34 extracts (35·3%) might be considered promising sources of antiviral natural products, as they have shown EC50 ≤ 100 μg ml?1. The present screening discloses the high potential of the Bignoniaceae family as source of antiviral agents. Significance and Impact of the Study: Active extracts were identified and deserve bioguided studies for the isolation of antiviral compounds and studies on mechanism of action.  相似文献
