The short field-life of Bacillus thuringiensis (Bt) insecticidal crystal protein has limited its use. When the Bt toxin is produced in Pseudomonas fluorescens it can be encapsulated and retain its effectiveness for two to three times longer than other Bt formulations. In order to improve Bt expression, we have synthesized cryIA(c) Bt -endotoxin encoding region (GenBank AF537267) according to the usage codon of P. fluorescens and transformed the Bt toxin expression cassette into P. fluorescens strains. T7 RNA polymerase and the T7 promoter system were used to control expression of Bt toxin. SDS-PAGE and Western blotting assay revealed that the -endotoxin was expressed as 8% of the total protein in P. fluorescens. In in vitro tests, release of toxin from dead bacteria was demonstrated. Supplementation of diets with Bt toxin-containing Pseudomonas bacterium resulted in high mortality of cabbage butterfly (Pieris brassicae) larvae.  相似文献

The production of crystals and spores ofBacillus thuringiensis var.israelensis was studied under different aeration conditions. The results with 4 l batch cultures showed that for O2 non-limited, cultures cell yield, toxin production and spore count were constant for all oxygen transfer rates (OTR). Under O2 limitation, °-endotoxin concentrations and spore counts were lower than those obtained in non-limited cultures. In addition, -endotoxin yields diminished under O2 limitation, suggesting that the toxin synthesis mechanism could have been affected.  相似文献

A -endotoxin gene previously cloned from Bacillus thuringiensis subsp. galleriae has been shown by a combination of restriction mapping and DNA sequence analysis to be a cryIIB clone; in common with other cryIIB genes it was found to lack a functional promoter. Addition of a promoter resulted in expression of the gene in Bacillus thuringiensis but did not result in the formation of the crystalline inclusions normally associated with such toxins. Inclusion formation was only observed when the gene was incorporated into an operon containing a gene known to be involved in the crystallisation of another -endotoxin.  相似文献

This report describes a method for quantifying -endotoxin contents in spray-dried preparations ofBacillus thuringiensis strain GC-91. -Endotoxin is solubilized in the pro-toxin form and separated from other soluble compounds by ion exchange chromatography. The method does not discriminate between the different -endotoxins produced by strain GC-91. It is suitable for quality control, since -endotoxin concentrations found in different preparations correlate inversely with the LC 50 in bioassays on artificial diet against freshly hatched larvae ofSpodoptera littoralis. A typical batch of spray-dried material contains 1.22%±0.08% -endotoxin. The method is most accurate with preparations containing 0.5 to 2.0% toxin, for which triplicate estimations give standard errors close to±0.2%.  相似文献

A crystal -endotoxin gene ofBacillus thuringiensis subsp.tenebrionis (B.t.t.) encoding a coleopteran insect-specific toxin was used to construct a chimeric gene which expressed the toxin in plant cells. Via anAgrobacterium tumefaciens binary vector system, the toxin gene was transferred into tomato cells. From leaf disks recombinant plants were regenerated. Hybridization experiments demonstrated that these plants synthesized toxin-specific mRNA of the expected size. Transgenic tomato plants with the chimericB.t.t. toxin gene contained a 74 kDa protein which cross-reacted with toxin antibodies. The expression caused a significant insecticidal activity of the transgenic tomato plants against Colorado potato beetle larvae.  相似文献

With the recombinant pVB131 plasmid, which encodes the mosquitocidal 130 kilodalton peptide ofBacillus thuringiensis var.israelensis as a probe, DNA homology between crystal toxin genes of several dipteran-toxic strains was tested. Results from this study indicate that, while the crystal toxin genes ofB. thuringiensis var.kyushuensis and var.morrisoni isolate PG-14 share homology to the crystal toxin gene of var.israelensis, the -endotoxin genes of other dipteran-active strains tested (i.e., var.colmeri and var.kurstaki) do not exhibit any homology. The crystal toxin genes of vars.kyushuensis andmorrisoni isolate PG-14 were found to be located on plasmids of 60 and 94 megadaltons, respectively.  相似文献

Bioassays with Bacillus thuringiensis var. galleriae Berliner -endotoxin and plant phenolics on Heliothis armigera Hübner enhanced the activity of B.t. var. galleriae endotoxin. The presence of plant phenolics with B.t var. galleria endotoxin not only reduced feeding potential and weight gain by the larvae, but also enhanced the LC50 value of the toxin. Our study demonstrates the effect of phytochemicals from resistant crop plants on the biocidal activity of B. thuringiensis strains in laboratory conditions.  相似文献

Several medium ingredients locally available in Egypt were investigated for their ability to support -endotoxin production by two mosquito-toxic strains ofBacillus thuringiensis H-14 usingCulex pipiens for the bioassay of -endotoxin. Soya beans, black-eyed beans, common peas and lentils supported good production of toxin whereas peanuts, fodder yeast, cheese whey and corn steep liquor gave only low amounts of toxin. Molasses as the sole carbon source at 2 and 3% (w/v) with soya (or Proflo) as the sole nitrogen source at 3% gave the best yields of toxin for strain M1 and S128 respectively.

Les fèves de soja, les haricots à taches noires, les pois communs et les lentilles favorisent la production de toxine, tandis que les arachides, la levure fourragère, le petit lait et la liqueur de trempage de maïs ne produisent que de faibles quantités de toxines. La mélasse comme seule source de carbone à 2% (p/v) avec la soja (ou le Proflo) comme seule source d'azote de 2 à 3% donnent les meilleurs rendements en toxine.

Transgenic sweet pepper plants fromAgrobacterium mediated transformation   总被引:2,自引:0,他引:2  
Fertile transgenic sweet pepper (Capsicum annuum var. grossum) plants were regenerated at relatively high rate from various explants that were cocultivated withAgrobacterium tumefaciens strain GV3111-SE harbouring a plasmid that contains the cucumber mosaic virus coat protein (CMV-CP) gene. The rate of plant regeneration was found to depend on the types of explants cultured and the media used. Young leaves were most effective for bud induction and subsequent plant elongation while hypocotyls were the most inefficient. Southern analysis of DNA isolated from putative transgenic plants revealed that 3 out of 5 R1 plant lines reacted positively with the CMV-CP gene. Western blot analysis of CMV-CP containing R1 plants showed that two of them accumulated significant levels of the foreign gene product while the other two expressed it only, to low levels. Summary The toxicity and the spore count of batch and fed batch cultures of Bacillus thuringiensis var. israelensis were studied. Spore counts reached in both batch and fed batch cultures were as high as those reported in the literature, but the levels of toxicity found in the latter were about one order of magnitude lower than those attained in batch cultures. Avoiding restricted cultures might be necessary to reach high titres of -endotoxin, which are essential if a good product is intended. Furthermore, spore count might not be a good parameter to predict insecticidal activity of Bacillus thuringiensis cultures.  相似文献

Morphology of the parasporal -endotoxin crystals of Bacillus thuringiensis subsp. kurstaki isolate A3-4, a native (Taiwan) strain was studied by scanning (SEM) and transmission (TEM) electron microscopy. The typical bipyramidal crystals and an unusual parasporal inclusion with an embedded body were observed. The parasporal -endotoxin crystal with an embedded body, which was in different size and shape was well demonstrated in thin sections by transmission electron microscopy. The sporulated cells had multiple individually separated inclusions up to four crystals in one cell, which was unique to this isolate, and has not been reported before. The -endotoxin crystals included in the cell or released from the cell after batch fermentation or fed-batch fermentation did not show any altered morphological characteristics. However, judging from thin-sections of TEM, cells and the included parasporal crystals from fed-batch fermentation appeared larger than those from batch fermentation. It was observed that release of spore and parasporal crystals from the bacterial cell produced from batch cultures was earlier than that of fed-batch cultures. Preliminary bioassay results showed that the isolate cultures from both types of culture were equally effective against Plutella xylostella larvae. Based on the morphological observations, this strain may have a multiple insecticidal activities toward different insect species.  相似文献

A hybrid -endotoxin protein was designed against a polyphagous lepidopteran insect pest Spodoptera litura, which is tolerant to most of the known -endotoxins. The hybrid -endotoxin was created by replacing amino acid residues 530–587 in a poorly active natural Cry1Ea protein, with a highly homologous 70 amino acid region of Cry1Ca in domain III. The truncated -endotoxins Cry1Ea, Cry1Ca and the hybrid protein Cry1EC accumulated in Escherichia coli to form inclusion bodies. The solubilised Cry1EC made from E. coli was 4- fold more toxic to the larvae of S. litura than Cry1Ca, the best known -endotoxin against Spodoptera sp. None of the two truncated toxins, solubilised from E. coli caused larval mortality. However, trypsinised Cry1Ca protoxin obtained from E. coli and solubilised from inclusion bodies caused mortality of S. litura with LC50 513 ng/ml semi synthetic diet. A synthetic gene coding for the hybrid$-endotoxin Cry1EC was designed for high level expression in plants, taking into consideration several features found in the highly expressed plant genes. Transgenic, single copy plants of tobacco as well as cotton were developed. The selected lines expressed Cry1EC at 0.1–0.7% of soluble leaf protein. Such plants were completely resistant to S. litura and caused 100% mortality in all stages of larval development. Hence, unlike in E. coli, the hybrid -endotoxin folded into a functionally active conformation in both tobacco and cotton leaves. The truncated Cry1EC expressed in tobacco leaves was about 8-fold more toxic (LC50 58 ng/ml diet) compared to expression in E. coli.  相似文献

Bacillus thuringiensis produces insecticidal crystalline -endotoxins during sporulation. Leakage of radiolabeled markers from insect tissue culture cells and erythrocytes supports a mechanism of colloid osmotic lysis for the 23K and 27K polypeptides from the -endotoxins ofBacillus thuringiensis var.darmstadiensis 73-E10-2 and var.israelensis, respectively. We investigated the in vitro effects of ions, carbohydrates, lectins, and charged compounds on the cytolytic and hemolytic action of the purified -endotoxin ofBacillus thuringiensis var.darmstadiensis 73-E10-2, and other -endotoxins. Cytotoxicity was inhibited by preincubating the toxin with N-acetylneuraminic, glucuronic, galacturonic, and N-acetylglutamic acids and ATP. Lectins were unable to inhibit toxicity. Pretreatment of sheep erythrocytes with neuraminidase enhanced hemolysis. These results suggested a nonspecific inhibition of cytotoxicity based on the presence of a negative charge. Supraphysiological concentrations of divalent cations such as Ca2+, Mg2+, and Zn2+ in the medium yielded a reduced toxicity, whereas EDTA and EGTA enhanced cytotoxicity.  相似文献

On the energy hypersurfaces of the anions HP4 - and CH2P3 - at the RMP2(fc) /6-31+G(d) level, the isomers with triphosphaallyl moiety are the lowest energy structures. For these free 1-X-2,4-(PB)2-3-PA - anions characteristic 31P NMR chemical shifts, are predicted to be (for X = PH, 1, 31P(PA) = 517, 31P(PB) = 424, and 31P(PX) = 50; for X = CH2, 4, 31P(PA) = 611, 31P(PB) = 450). The observed exp 31P values for HP4 - (Na/K, DME) completely disagree with the 31P calculated at GIAO/MP2/6-311+G(d) //RMP2(fc) /6-31+G(d) for structure 1. The rotational average of the phosphinidyltriphosphirene structures (P3-PH-, 3) agree better with the exp 31P than those with a bicyclo[1.1.0]hydrogentetraphosphanide backbone, 2. MO analysis can rationalize the extreme endo/exo effect (31P = 455 ppm) on the chemical shift in the exocyclic PH group of 3. The lowest energy geometry of the anion 3 has Erel of 31 kJ mol-1 relative to 1. The most favored 3 + Na+ structure is only 15 kJ mol-1 above the lowest energy HP4Na minimum, 2 + Na+ with Na+ in endo and H in exo orientation of the bicyclo-P4 framework (Erel of 1 + Na+ is 13 kJ mol-1). In most HP4Na structures the Na+ changes the 31P NMR chemical shifts towards higher field with respect to the bare anions.Electronic Supplementary Material available.  相似文献

The presence of cyanobacteria generally decreased the effectiveness of Bacillus thuringiensis H-14 (BTI) as a mosquito larvicide. The effect was more pronounced when the mosquito larvae were exposed to BTI in the presence of several cyanobacterial strains. No synergistic or antagonistic effect between the -endotoxin from BTI and the hepatotoxin from cyanobacteria was seen. Neurotoxic cyanobacterial strains caused very fast paralysis in mosquito larvae; the decreases in the effectiveness of BTI when tested in combination with a neurotoxic strain might be due to the effect of this paralytic action on the feeding rate of the mosquito larvae.  相似文献

Bacillus thuringiensis subsp. tenebrionis produces a 65 kilodalton polypeptide toxin which is lethal to various coleopteran insect larvae. The gene encoding this toxin was cloned in E. coli in the broad host range vector pKT230 and subsequently transferred to Rhizobium leguminosarum by conjugation. Western blot analysis showed that the toxin gene was expressed in the free living state of Rhizobium producing two major polypeptides of 73 and 68 kilodalton in size. The level of expression of the toxin gene in Rhizobium varied from strain to strain. Cell extracts from toxin-producing rhizobia were toxic to larvae of Gasterophysa viridula. Bioassays also showed that the -endotoxin was toxic to larvae of the clover weevil Sitona lepidus. Furthermore, pea (Pisum sativum) and white clover (Trifolium repens) plants suffered less root and nodule damage by Sitona larvae when they were inoculated with Rhizobium strains containing the toxin gene. This suggests that such rhizobia could be useful in the biological control of this important legume pest.Abbreviations B.t.t. The novel finding of this study is that the δ-endotoxin present in the spore coat of Bacillus thuringiensis strain 1.1 (Bt1.1), plays a central role in spore germination by generation of germinant via its β-glucosidase activity and is based on the following: (i) the crystals of Bt1.1 consist of the 140 kDa δ-endotoxin which exhibits β-glucosidase enzymatic activity. Besides crystals, δ-endotoxin is also located in the spore coat and at this site displays β-glucosidase activity, resulting in glucose production; (ii) glucose is an efficient germinant of both Bt1.1 and acrystalliferous Bt4.1 strain; (iii) substrates of β-glucosidase can activate the germination of Bt1.1 spores, but not those of the acrystalliferous Bt4.1 sister strain that do not contain the 140 kDa δ-endotoxin; (iv) Reduction or enhancement of enzymatic activity of δ-endotoxin, results in retardation or acceleration of germination and outgrowth, respectively. Bt1.1 cells secrete a 60 kDa polypeptide which displays β-glucosidase activity as indicated by zymogram analysis and which is immunologically related to the 140 kDa δ-endotoxin.  相似文献

Biolistic transformation was used to introduce genes encoding the insecticidal proteins snowdrop lectin (Galanthus nivalis agglutinin; GNA) and cry1Ac Bt toxin (-endotoxin from Bacillus thuringiensis) into elite rice (Oryza sativa) cultivars. Plant transformation was carried out in parallel experiments simultaneously by using either whole plasmids containing suitable gene constructs, or the corresponding minimal gene cassettes, which were linear DNA fragments lacking vector sequences excised from the plasmids. Both transformation methods generated similar numbers of independent transformation events. Selected R0 clonal plant lines were further characterised for presence and expression of transgenes. Co-transformation of the unselected genes (cry1Ac and gna) with the selectable marker (hpt) was at least as efficient for transformation with minimal gene cassettes as with whole plasmid DNA, and higher levels of accumulation of the insecticidal gene products GNA and cry1Ac were observed in plants resulting from minimal gene cassette transformation. Insect bioassays with major pests of rice showed that transgenic plants expressing gna showed enhanced resistance to brown planthopper (Nilaparvata lugens), and plants expressing cry1Ac were protected against attack by striped stem borer (Chilo suppressalis). Expression of both transgenes gave protection against both pests, but did not increase protection against either pest significantly over the levels observed in plants containing a single insecticidal transgene.  相似文献
