Synopsis Hippocampus comes is presently exploited at significant rates in the central portion of – and possibly throughout – its range. The species is traded in dried form for traditional medicine and alive for the aquarium trade. Here we provide the most current life history, abundance and distribution information available for H. comes. These data should be helpful to researchers, and to management agencies in countries signatory to the Convention on International Trade in Endangered Species of Wild Fauna and Flora (CITES), tasked to implement strategies ensuring that trade is not detrimental to wild populations of H. comes (listed on Appendix II).  相似文献

Behaviour of morphocycline (H5R) and its complex with beryllium ions in acqueous solutions was studied fluorimetrically. The ionization constants of H5R were estimated at pH 1.5-15 according to the data of fluorimetric determination with respect to OH-group: pK1 6.40, pK2 8.25, pK3 10.65, pK4 11.30. Two constants characterizing the deprotonization process with respect to the carbonylic group and nitrogen were also estimated: pK01--1.0 (greater than C = 0) and pK02 4.75 (--N=). Formation of an intensively fluorescing compound less than [Be3(OH)3(H2O2)5]2HR greater than 2+ was observed at pH 6.0-7.0. The cause of such intensive fluorescence was deformation of ion [Be3(OH)3(H2O)6]3+ because of its exclusion from the coordinating sphere of one molecule of water during the complex formation and decreasing of level H comes from II as compared to the morphocycline level II comes from n. A procedure for detecting morphocycline in the blood of humans and animals was developed.  相似文献

Nucleosomal semihistone A24 (uH2A) is composed of histone H2A and ubiquitin peptides. The kinetics of incorporation of nascent H2A and ubiquitin into A24 of transformed chicken lymphocytes (MSB cells) have been examined by peptide mapping, COOH-terminus analysis, density labeling and isopycnic centrifugation of chromosomal proteins. We find that newly synthesized H2A is rapidly conjugated to ubiquitin. SDS-PAGE analysis of fractionated density gradients suggests, however, that newly synthesized ubiquitin becomes bound nonselectively to both new and preexisting H2A molecules.  相似文献

Exogenous tritiated -aminobutiric acid ([3H]GABA) is retained in two compartments in sheep cortex synaptosomes, corresponding to cytoplasmic and vesicular spaces, assuming that freeze-thawing the synaptosomes loaded with [3H]GABA releases the cytoplasmic [3H]GABA (81±3.9%), and that subsequent solubilization of the synaptosomes with 1% sodium cholate releases the vesicular [3H]GABA (19±3.9%). Depolarization of synaptosomes with 40 mM K+ in a Na+-medium, in the absence of Ca2+, releases 20.3±2.7% of the [3H]GABA retained in the synaptosomes. The [3H]GABA released under these conditions comes predominantly from the cytoplasm. The presence of 1 mM Ca2+ during depolarization releases and additional 13% (a total of about 33.5±9.9%) of the releasable [3H]GABA, and the [3H]GABA release which is Ca2+-dependent also comes mostly from the cytoplasmic compartment. When choline replaces external Na+, the [3H]GABA release is absolutely Ca2+-dependent, and the [3H]GABA released also comes mostly from the cytoplasmic pool. Therefore, it appears that [3H]GABA taken up by synaptosomes is accumulated mostly in the cytoplasmic compartment from which it is released upon depolarization. The technique described permits distinguishing the effect of different factors on the two pools of accumulated [3H]GABA.  相似文献

The hipparions of the Vallesian locality «Ravin dela Pluie in the lower Axios valley are studied. Two different species were found, a large one, H. primigenium and another small one, H. macedonicum. Their characteristics and dimensions as well as their comparison to the eurasiatic Vallesian samples allow us to find that they belong to Upper Vallesian. H. primigenium of Ravin de la Pluie is more evolved than the typical one of Eppelsheim, while H. macedonicum represents to oldest small hipparion which probably comes from a large form of Lower Vallesian. From the former took rise the later small hipparions. Some primitive features of hipparions are also discussed.  相似文献

The present study revealed the mechanism underlying the nuclear trafficking of metallothionein (MT). Nuclear localization of MT in digitonin-permeabilized BALB 3T3 cells was enhanced in the presence of a cytosolic factor added as a rat red blood cell lysate by oxidation with H2O2 in a dose-dependent manner, but inhibited with excess glutathione. A cytosolic partner was assumed to bind MT and retain it in the cytoplasm, and its oxidation can mobilize MT to the nuclei on cellular oxidation. Pre-treatment of nuclei with H2O2 did not enhance the localization, and MT that had been localized in the nuclei was washed out, indicating that MT is in the nuclei as a result of a higher rate of uptake by the nuclei than the rate of diffusion from the nuclei. Nuclear localization of lysozyme and nuclear localization signal (NLS)-bearing allophycocyanin were not enhanced by the oxidation in the presence of cytosolic factor, suggesting that the nuclear traffic occurring on oxidation is specific to MT. Moreover, when cells were arrested the cell cycle at the S phase, MT was localized in the nuclei in response to coincidental generation of a feeble reactive oxygen species (ROS). These observations suggest that MT comes localized in the nuclei on the sensing of intracellular oxidation, whereby a cytosolic partner specific to MT comes oxidized as a cargo system, MT being localized as a result of enhanced uptake in the nuclei and re-localized in the cytoplasm diffusely. Nuclear MT was proposed to protect the nuclei from the oxidation occurring with progression of the cell cycle.  相似文献

Histone modifications have great importance in epigenetic regulation. JMJD2A is a histone demethylase which is selective for di- and trimethyl forms of residues Lys9 and Lys36 of Histone 3 tail (H3K9 and H3K36). We present a molecular dynamics simulations of mono-, di- and trimethylated histone tails in complex with JMJD2A catalytic domain to gain insight into how JMJD2A discriminates between the methylation states of H3K9. The methyl groups are located at specific distances and orientations with respect to Fe(II) in methylammonium binding pocket. For the trimethyllysine the mechanism which provides the effectual orientation of methyl groups is the symmetry, whereas for the dimethyllysine case the determining factors are the interactions between methyllysine head and its environment and subsequently the restriction on angular motion. The occurrence frequency of methyl groups in a certain proximity of Fe(II) comes out as the explanation of the enzyme activity difference on di- and tri-methylated peptides. Energy analysis suggests that recognition is mostly driven by van der Waals and followed by Coulombic interactions in the enzyme-substrate interface. The number (mono, di or tri) and orientations of methyl groups and water molecules significantly affect the extent of van der Waals interaction strengths. Hydrogen bonding analysis suggests that the interaction between JMJD2A and its substrates mainly comes from main chain-side chain interactions. Binding free energy analysis points out Arg8 as an important residue forming an intra-substrate hydrogen bond with tri and dimethylated Lys9 of the H3 chain. Our study provides new insights into how JMJD2A discriminates between its substrates from both a structural and dynamical point of view.  相似文献

Plant pathogen effectors encoded by Avirulence (Avr) genes benefit the pathogen by promoting colonization and benefit plants that have a matching resistance (R) gene by constituting a signal that triggers resistance. The Hessian fly, Mayetiola destructor (Say) (Diptera: Cecidomyiidae), resembles a plant pathogen in showing R/Avr interactions. Because of these interactions, a wheat plant with the H13 resistance gene can be resistant or susceptible depending on the genotype of the larva that attacks the plant, being resistant if attack comes from a larva with a functional vH13 gene, but susceptible if attack comes from a larva with a non‐functional vH13 gene. In this study we asked: does this susceptible interaction involving plants with H13 look like susceptible interactions with plants lacking H13? Possibly, the H13 plant attacked by a larva with a non‐functional vH13 is induced to partial rather than complete resistance. Or the larva, lacking its vH13‐encoded effector, is compromised in its ability to induce susceptibility, which includes forcing the plant to create a gall nutritive tissue. Responses of epidermal cells to larval attack were explored using imaging techniques (light microscopy, scanning and transmission electron microscopy). Whole‐organism responses were investigated by measuring the growth of plants and larvae. No evidence was found for partial resistance responses by H13 plants or for a compromise in the ability of vH13 loss‐of‐function larvae to induce susceptibility. It appears that disrupting vH13 function is sufficient for overcoming the induced resistance mediated by the H13 gene.  相似文献

Summary It has been suggested that the effect of cytokinins in retarding leaf senescence comes about through their incorporation into tRNA. To test this hypothesis, kinetin-8-14C, 6-benzylaminopurine-benzyl-7-14C and adenine-8-3H were applied to detached tobacco leaves, and the nucleic acids were thereafter extracted and chromatographed on MAK columns. Kinetin-8-14C and adenine-8-3H were readily incorporated into RNA in a similar pattern. 6-Benzylaminopurine-benzyl-7-14C was effective in delaying chlorophyll loss but was not incorporated into any nucleic-acid fraction. It is concluded that the possibility of cytokinins retarding leaf senescence by completion of tRNA is not supported.  相似文献

Swiss 3T3 cells were labelled with [3H]glycerol and stimulated with bombesin over a time course of 20 min. The individual 1,2-diacylglycerols produced were quantified by acetylation followed by analysis by HPLC and argentation chromatography. The major phospholipids and phosphatidic acid were acetolysed and then analysed in the same manner. The data show that even at an early time of stimulation (30 s), stimulated diacylglycerol formation comes from at least two sources--phosphoinositides and phosphatidylcholine.  相似文献

Helicobacter pylori is now considered to be the main cause for most stomach diseases including ulcer, MALT lymphoma, adenocarcinoma and gastritis. The infection with this bacterium is chronic despite a local and systemic immune response towards it. Among the cellular infiltrate that arises during H. pylori-mediated gastritis, there is a considerable frequency of CD4+ Th1 cells producing IFNgamma, but not of Th2 cells producing IL-4. Since IFNgamma may induce binding of H. pylori to gastric epithelial cells followed by apoptosis of these cells, one may speculate that H. pylori-mediated diseases are in part autoimmune diseases initiated by H. pylori-specific Th1 cells infiltrating the gastric mucosa. Recent support for this hypothesis comes from an animal model in which mice are infected with H. pylori and display strongly reduced gastritis in the absence of IFNgamma.  相似文献

It has been proposed that the major portion of [3H]GABA released from rat cortical slices upon exposure to high K+ comes from a neuronal pool. Using carrier mediated exchange diffusion of DABA or β-alanine in the superfusion medium for GABA in the slice as a technique for manipulating neuronal and glial pools of GABA, it was found that DABA but not β-alanine substantially reduced the K+ stimulated release of [3H]GABA. The present study using synaptosomes as an in vitro model of the nerve ending was undertaken to ascertain whether this neuronal pool of releasable [3H]GABA was associated with a specific transmitter pool in nerve endings. A continuous superfusion system employing a Ca2+ pulse to produce a calcium coupled release (Levy et al, 1973) was used to study the effect of two concentrations (20 μm , 1 mm ) of DABA and β-alanine on the release of [3H]GABA from synaptosomes. In contrast to the results in slices, DABA at both concentrations had no effect on the release of [3H]GABA from synaptosomes in spite of evidence that exchange diffusion was occurring. With protoveratrine as the releasing agent there was no effect of DABA on the release of [3H]GABA from either slices or synaptosomes. The results suggest that the major portion of [3H]GABA released from cortical slices by high K+ comes from a non-transmitter pool in the neuron. Use of K+ stimulated release of amino acids from cortical slices as a criterion for neurotransmitter function must be viewed with caution.  相似文献

This study examined how habitat associations changed with ontogeny in the tiger tail seahorse Hippocampus comes Cantor, 1850, over four reef zones in a coral reef ecosystem. Hippocampus comes showed ontogenetic differences in their use of habitat at the scale of reef zones (macrohabitat) and holdfasts (microhabitat). Across reef zones, juvenile size classes (25–105 mm standard length, L S) were most abundant in wild macroalgal beds ( Sargassum spp.) (55·7%), while adults (>105 mm L S) occupied both coral reefs (39·7%) and macroalgal beds (42·7%). Microhabitat use also varied with ontogeny. Juveniles generally used macroalgal holdfasts, while adults >135 mm L S used a greater diversity of specialized microhabitats that included branching sponges, branching corals and tall seagrass. Ontogenetic changes in habitat association, as well as size-related shifts in crypsis and aggregation, suggest that H. comes experiences fitness trade-offs that vary with size; juveniles may associate with habitat that reduces predation, while larger individuals may use distinct microhabitat in reef zones to optimize reproductive success. Results are discussed in the context of targeted exploitation, expanding artisanal mariculture, habitat damage from illegal fishing and reserve design.  相似文献

Linoleic acid was used as a model system to study lipid peroxidation initiated by the reaction of ferrous iron with hydrogen peroxide. Low-level chemiluminescence of the peroxidation was measured with a high-sensitivity single-photon counter. It was found that the luminescence primarily comes from the dimol reaction of singlet oxygen and that the peak intensity of emission is a quadratic function of the concentration of either Fe2+ or H2O2, provided that the other Fenton reagent is in great excess. Under the same conditions, analysis on reaction kinetics shows a linear relationship between the maximal level of the initiator formed by the Fenton reaction and the initial concentration of Fe2+ or H2O2. This implies that the peak intensity of the chemiluminescence may be a good index of the maximal level of the initiator.  相似文献

The serotonergic system may play a role during general anesthesia but the effect of the volatile anesthetic halothane on the release of serotonin (5-HT) is not fully understood. Rat brain cortical slices were labeled with [3H]5-HT to investigate the effects of halothane on the release of this neurotransmitter from the central nervous system. Halothane induced an increase on the release of [3H]5-HT that was dependent on incubation time and anesthetic concentration (0.006, 0.012, 0.024, 0.036, 0.048 and 0.072 mM). This effect was independent of extracellular calcium and was not affected by tetrodotoxin (blocker of voltage dependent Na+ channels). In contrast, the halothane-evoked [3H]5-HT release was reduced by BAPTA-AM, a membrane-permeable BAPTA analog that chelates intracellular Ca2+. The anesthetic-induced [3H]5-HT release depends on the ryanodine-sensitive intracellular calcium store since it was blocked by dantrolene and azumolene (inhibitors of the calcium-release through ryanodine receptors) but was not affected by aminoethoxydiphenylborate (2-APB), an inhibitor of inositol 1,4,5-triphosphate receptor. The [3H]5-HT release induced by halothane comes mainly from the vesicular pool since it was reduced in about 70% by reserpine, a blocker of vesicular monoamine transporter. The halothane-evoked release of [3H]5-HT release is reduced by fluoxetine, an inhibitor of 5-HT uptake, and the volatile agent also decreased the uptake of [3H]5-HT into rat brain cortical slices. Moreover, a decrease on halothane-induced release of [3H]5-HT was also observed when the brain cortical slices were incubated at low temperature, which is known to interfere with the carrier-mediated release of the neurotransmitter. Ouabain, a Na+/K+ ATPase pump inhibitor, which induces 5-HT release through reverse transport, also decreased [3H]5-HT release induced by halothane, confirming the involvement of a carrier-mediated release of the neurotransmitter in the presence of halothane. In conclusion, these data suggest that halothane induces vesicular and carrier-mediated release of [3H]5-HT in rat brain cortical slices.  相似文献

In this report, we introduce the use of DNA-cellulose chromatography for evaluating the strength of binding of histones to DNA under a variety of conditions. We have found that histones added directly to DNA-cellulose at physiological salt concentrations bind relatively weakly, with all histones eluting together at about 0.5 M NaCl when a salt gradient is applied. However, much tighter binding of the four nucleosomal histones to DNA-cellulose is obtained if gradual histone-DNA reconstitution conditions are used. In this case, the binding of histones H2A, H2B, H3, and H4 to DNA-cellulose closely resembles their binding to native chromatin. The nativeness of the binding is indicated both by the distinctive sodium chloride elution profile of these histones from DNA-cellulose and by their relative resistance to trypsin digestion when DNA-bound. The binding to DNA-cellulose of histones H2A, H2B, H3, and H4, which have had the first 20 to 30 amino acid residues removed from their NH2 termini, is indistinguishable from the binding to DNA-cellulose of the same intact histones, as judged by their salt elution profile. Thus, even though the NH2 termini contain 40 to 50% of the positively charged amino acid residues (thought to interact with the DNA backbone), a major contribution to the DNA binding comes from the remainder of the histone molecule. Finally, we have discovered that histones can form a "nucleosome-like" complex on single-stranded DNA. The same complex does not appear to form on RNA. Histones H3 and H4 play a predominant role in organizing this histone complex on single-stranded DNA, as they do on double-stranded DNA in normal nucleosomes. We suggest that, in the cell nucleus, nucleosomal structures may form transiently on single strands of DNA, as DNA and RNA polymerases traverse DNA packaged by histones.  相似文献

DNase I was used to probe the higher order chromatin structure in whole nuclei. The digestion profiles obtained were the result of single-stranded cuts and were independent of pH, type of divalent ion and chromatin repeat length. Furthermore, the protection from digestion of the DNA at the entry/exit points on the nucleosome was found to be caused not by the H1/H5 histone tails, but by the compact structure that these proteins support. In order to resolve symmetry ambiguities, DNase I digestion fragments over several nucleosome repeat lengths were analysed quantitatively and compared with computer simulations using combinations of the experimentally obtained rate constants (some of which were converted to 0 to simulate steric protection from DNase I digestion). A clear picture of precisely defined, alternating, asymmetrically protected nucleosomes emerged. The linker DNA is inside the fibre, while the nucleosomes are positioned above and below a helical path and/or with alternating orientation towards the dyad axis. The dinucleosomal modulation of the digestion patterns comes from alternate protection of cutting sites inside the nucleosome and not from alternating exposure to the enzyme of the linker DNA.  相似文献

The trade‐off hypothesis posits that increased performance on a given resource comes at the cost of decreased performance on other resources, and that this trade‐off is a driving force of food specialization in both predators and herbivores. In this study, we examined larval survival and performance in two sibling ladybird species, Harmonia yedoensis Takizawa and Harmonia axyridis Pallas (Coleoptera: Coccinellidae), fed on one of four prey species. Harmonia yedoensis is a specialist predator that preys mostly on pine aphids in the field, whereas H. axyridis is a generalist predator with a broad prey range. We experimentally showed in the laboratory that larval survival and performance were not higher when H. yedoensis was fed on pine aphids, compared with the other prey species. Rather, prey suitability was similar in both ladybird species, and H. yedoensis larvae developed as well or even better on prey species that they never utilize in nature. These results suggest that the host range in H. yedoensis may not be limited by the intrinsic suitability of the aphid species per se. Moreover, as shown by our previous study, the pine aphid is a highly elusive prey that is difficult for small ladybird hatchlings to capture, which means that the cost of utilizing this prey is high. Therefore, we conclude that some factor other than prey suitability is responsible for the observed food specialization in H. yedoensis.  相似文献
