Sporelings of the monoecious red alga Gelidium vagum were placed into 4500 individual cultures after treatment with the chemical mutagen nitrosoguanidine and raised to sexual maturity to search for reproductive mutants. Isolates undergoing normal self-fertilization were discarded, leaving approximately 250 self-sterile plants and mutants with abnormal reproduction or reproductive structures. Self-sterile mutants were tested further in crosses to a fertile green marker stock. From the results, many mutants appeared to be either male-sterile, female-sterile or unable to form carposporophytes. Although sufficient data on the inheritance pattern of the self-sterile mutants are available for only a few of the isolates, some apparently stable sterility mutations with simple Mendelian transmission were identified. Preliminary testing of one of the male-sterile mutations confirmed that it effectively eliminated self-fertilization and facilitated the formation of hybrid plants in crosses.  相似文献

Seed resulting from self-fertilization of Nicotiana tabacum plants was tested for germination in uninterrupted darkness at monthly intervals beginning at time of harvest. All seed lots were light-requiring immediately after harvest. Some lots continued to be light-requiring, but others gradually became light-indifferent during storage under laboratory conditions. One light-requiring and one light-indifferent selection were self-pollinated, and reciprocally crossed. The seed resulting from self-fertilization was light-requiring or light-indifferent, respectively, according to the parentage. Seed resulting from the reciprocal crosses differed in dark-germinability. Further, dark-germination of seed from each of the reciprocal crosses differed from that of either parent. Both parents contributed toward light sensitivity of the seed; however, contribution of the maternal parent was greater than that of the pollen parent.  相似文献

Natural clones and clones from laboratory crossings of Daphnia pulex have been tested in their response to stimuli for male and ephippia formation. The clones are from ponds in Illinois, USA, an area with both obligate and cyclical clones of D. pulex. The progeny from two types of crosses were studied: 1) crosses between an obligate clone producing males and a cyclical clone producing only females, and 2) self-fertilization within a cyclical clone producing both sexes. Both the natural and the artificial clones showed great variation in response to environmental stimuli for males. Ephippia response seems more equal among the clones, but two of the artificial clones could hardly be stimulated to produce ephippia in the induction experiments. In crowded cultures, however, they produced a few. Crossings between cyclicals and obligates yielded mostly cyclical progeny. Progeny from the selfed clone had low survival and fecundity.  相似文献

The relation between inbreeding depression and rate of self-fertilization was studied in nine natural populations of the annual genus Amsinckia. The study included two clades (phylogenetic lineages) in which small-flowered, homostylous populations or species are believed to have evolved from large-flowered, heterostylous, self-compatible ones. In one lineage the small-flowered species is tetraploid with disomic inheritance. Rates of self-fertilization were 25% to 55% in the four large-flowered, heterostylous populations; 72% in a large-flowered but homostylous population; and greater than 99.5% in the four small-flowered, homostylous populations, which produce seed autonomously. When present, inbreeding depression occurred in the fertility but not the survival components of fitness. Using a cumulative fitness measure incorporating both survival and fertility (flower number), we found inbreeding depression to be lower in the four very highly self-fertilizing populations than in the five intermediate ones. The Spearman rank correlation between inbreeding depression and selfing rate for the nine populations was –0.50, but was not statistically significant (P = 0.12). Inbreeding depression was greater in the two tetraploid populations than in the very highly self-fertilizing, diploid ones. Phenotypic stability of progeny from self-fertilization tended to be higher in populations with lower inbreeding depression. We conclude that levels of self-fertilization and inbreeding depression in Amsinckia are determined more by other factors than by each other. Estimates of mutation rates and dominance coefficients of deleterious alleles, obtained from a companion study of the four highly self-fertilizing populations, suggest that a strong relationship may not be expected. We discuss the relationship of the present results to current theory of the coevolution of self-fertilization and inbreeding depression.  相似文献

RFLPs (restriction fragment length polymorphisms) of PCR (polymerase chain reaction) -amplified fragments were used to trace the pattern of plastid DNA inheritance in the genus Actinidia. A total of 51 progeny originating from interspecific crosses between three A. arguta cultivars and A. deliciosa, the kiwifruit, and 12 progeny originating from the cross between A. kolomikta and A. chinensis were analysed together with their parents. No reciprocal crosses could be tested since they all failed to set viable seeds. Attempts to rescue immature embryos failed in all cases as well. The A. arguta×A. deliciosa crosses were checked for the RFLP patterns of a sequence encoding part of the Rubisco large subunit (rbcL), using either AluI or MseI, and for a sequence encoding part of the photosystem II D1 protein (psbA), using HinfI. The A. kolomikta×A. chinensis cross was checked for the RFLP patterns of sequences encoding the spacers between trnT and the 5-trnL exon (a-b spacer DNA) and the trnL 3 exon and trnF (e-f spacer DNA), respectively. The first spacer revealed a natural polymorphism between the two parent species due to a large deletion occurring in A. kolomikta detectable without further restriction enzyme treatment. The e-f spacer DNA was digested with HinfI. The comparison of the RFLP patterns in the parents and their progeny showed a strictly paternal inheritance of chloroplast DNA in Actinidia, with no exception found in any of the crosses examined. As the reciprocal crosses were not available, we do not know whether paternal inheritance of plastids is restricted to the crosses we analysed or if this is the general rule for plastid inheritance in the genus Actinidia. Actinidia is dioecious and is the first purely outbreeding species for which a paternal plastid inheritance has so far been documented.  相似文献

Inheritance of chloroplast DNA (cpDNA) was examined in F1 progenies derived from three crosses and three corresponding reciprocal crosses betweenStellaria porsildii andS. longifolia. Chloroplast DNA restriction fragments were analyzed using methods of nonradioactive digoxigenin-11-dUTP labeling and chemiluminescent detection with Lumi-Phos 530. Distinct interspecific restriction fragment polymorphisms were identified and used to demonstrate the mode of cpDNA inheritance. Mode of cpDNA inheritance differed among crosses. Two crosses in whichS. porsildii, SP2920-21, was the maternal parent exhibited three different types of plastids, maternal, paternal and biparental, among the F1 hybrids, suggesting a biparental cpDNA inheritance and plastid sorting-out inStellaria.  相似文献

 Previous studies have established that chloroplasts are inherited paternally in Actinidia interspecific crosses. However, fertilisation problems in interspecific crosses may affect the transmission of organelles. Six female clones, i.e. ‘Abbott’, ‘Bruno’, ‘Greensill’, ‘Hayward’, ‘Jones’, ‘Monty’, and four male clones were used to identify cpDNA polymorphisms within the cultivated kiwifruit species A. deliciosa. The restriction patterns by HpaII of a chloroplast fragment amplified by PCR with a pair of universal primers revealed a polymorphism at the intraspecific level. The inheritance of cpDNA in 143 seedlings from three intraspecific crosses in kiwifruit (Actinidia deliciosa) was studied. All offspring displayed the restriction pattern of the paternal parent, indicating that maternal inheritance of cpDNA in kiwifruit is rare at best. Strict maternal inheritance of mtDNA was confirmed in the same crosses used to investigate cpDNA transmission. Studies of cytoplasmic inheritance in the Actinidia genus represent to date the best documented report of differential organelle inheritance of cpDNA and mtDNA in angiosperms. The genus Microseris contains species with disporangiate stamens and species with tetrasporangiate stamens. We determined the number of microsporangia per stamen in serial sections of heads from all 13 species of Microseris, its close relative Uropappus lindleyi, and the two allopolyploid species of Slebbinsoseris. Four Microseris species, three diploid and one tetraploid, have two microsporangia per stamen; all other species investigated have four. The most parsimonious assumption is that the disporangiate condition is derived and arose once in the evolution of Microseris. The inheritance of the number of microsporangia per stamen in crosses between M. bigelovii (disporangiate) and M. douglasii (tetrasporangiate) was determined. Segregation of microsporangium number per stamen in F2s derived from these crosses is quantitative rather than Mendelian. The average number of microsporangia per stamen in the F2 plants ranges from 2.0 to 4.0. There is a predominance of tetrasporangiate stamens in the F1 and in most F2 plants. The observed pattern of inheritance suggests a major gene with dominance and quantitative modifiers.  相似文献

Summary Nucleocytoplasmic determination of male sterility in Thymus vulgaris L. has been assumed in all papers attempting to explain the remarkably high frequencies of male steriles found in natural populations of this species. This paper provides strong evidence that both nuclear and cytoplasmic genes are involved in the determination of male sterility of this species, giving a complex inheritance. Interpopulation and intrapopulation crosses have shown that the ratio of females versus hermaphrodites among offsprings varied widely from 10 to 11. Furthermore, interpopulation crosses consistently yielded a higher frequency of females than intrapopulation crosses. Nucleocytoplasmic inheritance was demonstrated by an absence of male fertiles in backcrosses and asymmetrical segregation in reciprocal crosses. Molecular analysis of the mitochondrial DNA of some of the parents used in crosses suggested the involvement of different cytoplasms in the inheritance of male steriliy.  相似文献

Restriction enzyme analysis was used to determine the inheritance of chloroplast DNA in conifers. The plant material studied included five individual trees of European larch (Larix decidua Mill.) and Japanese larch (Larix leptolepis Sieb. & Zucc.) and six hybrids from controlled crosses between these species. The chloroplast DNA fragment patterns generated by Bam-HI and Bcl-I were species-specific. Paternal inheritance of chloroplast DNA patterns was found in most Larix crosses. One hybrid showed maternal chloroplast DNA patterns. In addition, two other hybrids had mixed Bam-HI patterns suggesting recombination between maternal and paternal chloroplast DNA. The mechanisms favoring paternal inheritance in conifers are not known. Paternal inheritance of chloroplast DNA is suggested it to be a general phenomenon in conifers.  相似文献

The inheritance of mitochondrial (mt) and chloroplast (ct) DNA in the progeny from interspecific crosses between the cultivated carrot (Daucus carota sativus) and wild forms of the genus Daucus was investigated by analysis of mt and ct RFLPs in single plants of the parental and filial generations. We observed a strict maternal inheritance of the organellar DNAs in all interspecific crosses examined. Previous studies on putative F2 plants from a cross between Daucus muricatus x D. carota sativus suggested paternal inheritance of ctDNA. Our reinvestigation of this material revealed that the mtDNA of the putative F2 plants differed from the mtDNA of both putative parents. Therefore, our data suggest that the investigated material originated from other, not yet identified, parents. Consequently, the analysis of this material cannot provide evidence for a paternal inheritance of ctDNA.  相似文献

The karyotypes of 18 samples assigned to 13 agamospecies in Taraxacum are described. The species are classified among six sections; Eiythrosperma Lindb. f., Vulgaria Dahlst., Alpestria v. Soest, Alpina Hagl., Fontana v. Soest and Cucullata v. Soest, all from alpine localities in Europe. No previous cytological work is known for agamospecies in the last four sections. Two small chromosome types and the occurrence of accessory chromosomes are described for the first time in Taraxacum. Sibling karyotypes were found to be invariable, but extensive karyotypic variation was found between samples, even intraspecifically. Karyotypes were compared by means of a “karyotype similarity index”. Samples with a high overall similarity tended to have smaller chromosomes. Eight species exhibited regular chromosome sets, the remainder being irregular. The nature of the origin of the latter is discussed in relation to the origin of agamospecies in the genus.  相似文献

Summary The inheritance of seed coat color, pericarp color, polyphenoloxidase activity and bristle, glume, collar, and leaf-base anthocyanic colorations was investigated using intra- and interspecific crosses between Setaria italica and S. viridis. The results were compared to inheritance results obtained by previous authors. In most cases, the inheritance is simple (one or two loci) and data from different crosses (intra- and interspecific) and from different authors can be compared. Two sets of two characters were found to share common loci: the polyphenoloxidase locus is one of the loci responsible for seed coat color, and bristle and glume color are determined by the same two loci. The evolutionary significance of these results is discussed.  相似文献

Algebraic models of gynodioecy show that the effects on the equilibrium sex ratio of the relative survival and seed production of the sexes and of inbreeding of male-fertile plants are identical for all genic modes of inheritance, provided that different genotypes among male-fertile plants (or among females) do not differ in average fitness. The effects of three modes of inbreeding on equilibrium sex ratios are examined. If there is competition between self- and cross-fertilization of male-fertile individuals, a stable sexual dimorphism can be maintained by an outbreeding advantage of females if both the proportion of cross-fertilized seeds among those borne on male-fertile individuals,t, and the inbreeding depression (fitness inbred/outbred seeds),i, are less than one half. A lower frequency of females is obtained for the same values oft andi if self-fertilization precedes cross-fertilization. If self-fertilization follows cross-fertilization, gynodioecy cannot be maintained by an outbreeding advantage of females. When the sex phenotypes of gynodioecious populations are determined by cytoplasmic inheritance, females need only a slight advantage over males in survival, ovule production or outbreeding to persist at equilibrium. When determined by nuclear genes, androdioecy can be maintained by greater fecundity or a higher survival rate of males than of female-fertile plants, but not by an outbreeding advantage. Androdioecy cannot be maintained with cytoplasmic inheritance of sex. The models suggest explanations for the more frequent occurrence of gynodioecy than of andrdioecy, the high frequency of gynodioecy in Hawaii and New Zealand, and the origin of gynodioecy from hermaphrodite but not from monoecious ancestors.  相似文献

We present nine polymorphic di- and tri-nucleotide repeat nuclear microsatellite markers selected specifically for their use in high throughput studies concerning the dioecious allotetraploid Salix albaSalix fragilis willow complex. These taxa and their hybrids are difficult to discriminate using morphological characters. Thus, multiplex reactions were developed for these microsatellite loci and their effectiveness to distinguish individuals, especially hybrids, and their inheritance patterns in controlled crosses were determined. All loci displayed disomic–monogenic inheritance which allowed for the genotype data to be analysed as for a diploid organism. The nine loci produced a total of 67 alleles (mean, 7.4 alleles per locus; range, 3–11 alleles) in a reference panel of 57 individuals from two germplasm collections and natural populations. Gene diversity values (as measured by the expected heterozygosity) ranged from 0.000–0.820. A total of 53 distinct multilocus genotypes were observed, and ordination analysis revealed three separate clusters corresponding to S. alba, S. fragilis and hybrids. The microsatellite loci described here will be used in population genetic studies to investigate genetic variation, gene flow, levels of hybridisation and the extent of introgression in natural populations of the S. albaS. fragilis complex. They are also useful for clonal identification, conservation and sustainable management of germplasm collections, genetic mapping and the selection of individuals and/or certification of controlled crosses for breeding programmes.  相似文献

Summary The inheritance of avenin components, the prolamins (or alcohol soluble proteins) of Avena, is studied by means of gel electrophoresis. Avenin is composed of rather similar proteins which appear as a polymorphic group from a biochemical point of view. After a first preliminary investigation it showed a surprisingly high interspecific variability. The average number of its constituents increases with the ploidy level but it still is much lower than that of wheat gliadin.The avenin electrophoretic patterns of 47 samples (F4, F5 or F6 seeds) resulting from 3 hexaploid crosses are compared with the parental patterns. Four kinds of inheritance are observed. Roughly 50% of progeny profiles are identical to those of one of the parents. They are composed occasionally of partial sections of parental patterns. Complete additiveness occurs rather seldom. However, in one of the crosses a significant number of progeny samples show a band, one of the very slow moving constituents, which was not present in either of the parents.The study of avenin in F1 seeds, arising from reciprocal crosses between two homozygous parent plants, shows a significant effect of maternal gene dose in the triploid endosperm.Because of both the variability and the relatively small number of avenin constituents, these results show that typical endosperm proteins such as oat prolamin constitute a useful tool for phylogenetic studies of the genus Avena.  相似文献

Summary The cloning of white spruce (Picea glauca) mitochondrial DNA homologous to the cytochrome oxidase II and ATPase genes of maize is described. These probes were used to define restriction fragment length polymorphisms which distinguish the white, Engelmann (P. engelmannii) and Sitka spruce (P. sitchensis) populations that occur in British Columbia. Analysis of progeny from crosses between the species revealed that mitochondrial DNA was maternally inherited in all cases (32 progeny from five independent crosses). The inheritance of chloroplast DNA was determined using a probe described previously; in this case, all progeny exhibited paternal inheritance (27 progeny from four crosses). Mitochondrial and chloroplast probes were used to test trees from zones of introgression between coastal (Sitka) and interior spruces (white and Engelmann). In most cases mitochondria and chloroplasts within individuals were contributed by different species. The data shows that there is a significant Sitka spruce component in trees east of the coastal watershed in British Columbia.  相似文献

Summary The inheritance of chloroplast (cp) DNA was examined in F1 hybrid progenies of two Populus deltoides intraspecific controlled crosses and three P. deltoides × P. nigra and two P. deltoides × P. maximowiczii interspecific controlled crosses by restriction fragment analysis. Southern blots of restriction digests of parental and progeny DNAs were hybridized to cloned cpDNA fragments of Petunia hybrida. Sixteen enzymes and five heterologous cpDNA probes were used to screen restriction fragment polymorphisms among the parents. The mode of cpDNA inheritance was demonstrated in progenies of P. deltoides × P. nigra crosses with 26 restriction fragment polymorphisms of cpDNA differentiating P. deltoides from P. nigra, as revealed by 12 enzyme-probe combinations, and in progenies of P. deltoides × P. maximowiczii crosses with 12 restriction fragment polymorphisms separating P. deltoides from P. maximowiczii, as revealed by 7 restriction enzyme-probe combinations. In all cases, F1 offspring of P. deltoides × P. nigra and P. deltoides × P. maximowiczii crosses had cpDNA restriction fragments of only their maternal P. deltoides parent. The results clearly demonstrated uniparental-maternal inheritance of the chloroplast genome in interspecific hybrids of P. deltoides with P. nigra and P. maximowiczii. Intraspecific P. deltoides hybrids also had the same cpDNA restriction fragments as their maternal parent. Maternal inheritance of the chloroplast genome in Populus is in agreement with what has been observed for most other angiosperms.  相似文献

To discriminate between the roles of differential pollen tube growth and inbreeding depression in causing reduced seed production following self-pollination of distylous Amsinckia grandiflora (Boraginaceae), callose plug and pollen tube production were compared following cross- and self-pollinations of A. grandiflora. Pin (long-styled morph) × thrum (short-styled morph) crosses yielded more pollen tubes than pin × pin crosses or pin self-pollinations; no difference could be detected for the pin × pin or pin self-pollination categories. Using thrums as the female parent, no significant differences were detected in any pollination category. These results indicate that reduced seed production following self-fertilization of A. grandiflora is likely to be the result of inbreeding depression rather than inhibition of self pollen tubes.  相似文献

Plastidial (pt) and mitochondrial (mt) genes usually show maternal inheritance. Non-Mendelian, biparental inheritance of plastids was first described by Baur (Z Indukt Abstamm Vererbungslehre 1:330–351, 1909) for crosses between Pelargonium cultivars. We have analyzed the inheritance of pt and mtDNA by examining the progeny from reciprocal crosses of Pelargonium zonale and P. inquinans using nucleotide sequence polymorphisms of selected pt and mt genes. Sequence analysis of the progeny revealed biparental inheritance of both pt and mtDNA. Hybrid plants exhibited variegation: our data demonstrate that the inquinans chloroplasts, but not the zonale chloroplasts bleach out, presumably due to incompatibility of the former with the hybrid nuclear genome. Different distribution of maternal and paternal sequences could be observed in different sectors of the same leaf, in different leaves of the same plant, and in different plants indicating random segregation and sorting-out of maternal and paternal plastids and mitochondria in the hybrids. The substantial transmission of both maternal and paternal mitochondria to the progeny turns Pelargonium into a particular interesting subject for studies on the inheritance, segregation and recombination of mt genes.  相似文献
