Changes in fibre diameters of extraocular muscles of the rabbit were studied at different times after denervation. The whole inferior oblique muscle hypertrophied, while some of the muscle fibres hypertrophied and others showed atrophy, depending on the fibre type. Fibre types have been determined by their histochemical enzyme profile. In the central layer of the muscle the phasic muscle fibres, which are rich in mitochondria, exhibited a transient hypertrophy being maximal 4-5 weeks after denervation and afterwards they atrophied; other phasic muscle fibres, which are poor in mitochondria, atrophied without having shown any sign of hypertrophy. Special, putatively slow tonic muscle fibres, which have low enzyme activities, underwent small long-lasting increases of their diameters. In the superficial layer of extraocular muscle there are two types of extremely thin muscle fibres rich in mitochondira. Both these fibre types hypertrophied to the greatest degree and for a very long time. Comparable changes in fibre diameters as described here for the muscle fibre types of an extraocular muscle are known from special muscle fibres in other vertebrate  相似文献

The microscopic organization and ultrastructure of the submandibular muscle of 10 species of Amphibia were compared. Among other fibre features the diameter of fibres, their content of mitochondria and fat, organization of sarcomeres: morphology of Z-line, M-band and sarcoplasmic reticulum were taken into consideration and 4 main types of muscle fibres were distinguished. They correspond to tonic (slow) and phasic (red, white and intermediate) ones. Slight variety of fibre morphology and of fibre elements among the examined species was found. Special attention to the variety of fibre morphology among the established types has been paid and the existence of continuous "spectrum" of fibres was suggested. The correlation of frequency of fibres of particular types with the body size, gular oscillation frequency, and some other characteristics of the submandibular muscle in the examined species was discussed. Also the zonal arrangement of muscle according to the fibre types, as well as possible dynamic nature of muscle fibres were emphasised.  相似文献

Continuous activity of isolated frog gastrocnemius muscle fibres provoked by repetitive stimulation of 5 Hz was used as an experimental model for fatigue development in different fibre types. Parameter changes of the elicited intracellular action potentials and mechanical twitches during the period of uninterrupted activity were used as criteria for fatigue evaluation. Slow fatigable muscle fibre (SMF) and fast fatigable muscle fibre (FMF) types were distinguished depending on the duration of their uninterrupted activity, which was significantly longer in SMFs than in FMFs. The normalized changes of action potential amplitude and duration were significantly smaller in FMFs than in SMFs. The average twitch force and velocity of contraction and relaxation were significantly higher in FMFs than in SMFs. Myosin ATPase (mATPase) and succinate dehydrogenase activity were studied by histochemical assessment in order to validate the fibre type classification based on their electrophysiological characteristics. Based on the relative mATPase reactivity, the fibres of the studied muscle were classified as one of five different types (1-2, 2, 2-3, 3 and tonic). Smaller sized fibres (tonic and type 3) expressed higher succinate dehydrogenase activity than larger sized fibres (type 1-2, 2), which is related to the fatigue resistance. The differences between fatigue development in SMFs and FMFs during continuous activity were associated with fibre-type specific mATPase and succinate dehydrogenase activity.  相似文献

The bundle of tonic fibres situated at the proximal end of the locust metathoracic extensor tibialis muscle is innervated by the dorsal unpaired median neurone (DUMETi) as well as by the slow excitatory (SETi)) and common inhibitor (CI) neurones. It is not innervated by the fast excitatory neurone (FETi).These fibres contract spontaneously and rhythmically. The myogenic rhythm can be modified by neural stimulation.Spontaneous slow depolarizing potentials resembling the pacemaker potentials of insect cardiac muscle were demonstrated in these fibres.The actions of glutamate on the tonic muscle fibres are not compatible with its being a specific excitatory transmitter. Glutamate can stimulate weak contractions of the muscle, but this action is inhibited when chloride ions are removed from the saline.10?6 M Octapamine hyperpolarizes the tonic fibre membrane. Octopamine, GABA and glutamate all inhibit the myogenic contractions and reduce the force of the neurally evoked contractions.The tonic muscle is very responsive to proctolin. At 5 × 10?11 M proctolin enhances the force and increases the frequency of myogenic contractions. At 10?9 M it depolarizes the muscle membrane potential, and at that and higher concentrations it causes the muscle to contract. At 2 × 10?7 M proctolin induces contractures which resemble those evoked by sustained high-frequency neural stimulation. Iontophoretic experiments show that proctolin receptors occur at localized sites on the tonic fibre membrane.  相似文献

The morphology of the peroneus longus muscle was compared in three Galliformes and five Passeriformes in relation to partial behavioral characteristics. In the quail two fibre types are found, while the muscle of the other species is composed of three fibre types. The frequencies of these fibres are different, especially between Galliformes and Passeriformes. The peroneus longus muscle in the quail is innervated only from the phasic system. The other species show phasic and tonic innervation. There is a correlation between the muscle fibre calibre and the extent of the synaptic gutter.  相似文献

Ionic selectivity of sodium channels was examined under voltage clamp conditions in normal and denervated twitch fibres and denervated tonic fibres isolated from m. ileofibularis of the frog (R. temporaria). Membrane currents were recorded by means of the Hille-Campbell vaseline-gap voltage clamp method from muscle fibre segments exposed to a potassium-free artificial internal solution. Permeability ratio (PS/PNa) were determined from changes in the reversal potential after replacing all Na ions in the solution bathing the voltage clamped external membrane area with sodium substituting ions (S). The permeability sequence was: Na+ greater than Li+ greater than NH4+ greater than K+. No inward currents were observed for Ca2+. The permeability ratios were as follows. Denervated tonic fibres: 1:0.88:0.23:0.012; control twitch fibres: 1:0.94:0.22:0.076; denervated twitch fibres: 1:0.91:0.14:0.082. The permeability to Li+ ions deviates from independence to a greater extent in tonic than in phasic fibres. Our results are consistent with the Hille model of sodium channel selectivity, and they support the hypothesis that sodium channels formed in denervated tonic muscle fibres of the frog are of the same genetic origin as Na channels expressed under physiological conditions.  相似文献

Several studies have reported estimations of the total number of fibres in a muscle, e.g. before and after training or before and after inactivity. In those investigations a combination of computed tomographic estimations of muscle size and morphological studies of fibre size has most often been used. There have been doubts about the reliability of those studies on pennate muscles, since changes in muscle fibre size have been said to alter fibre angulation and thus the number of fibres that will cross a section. If such an alteration in fibre angulation takes place with an increase in fibre size, there ought to be some correlation between fibre size and fibre angulation. The present study was designed to test whether repetitive estimations of muscle fibre angulation could be performed in vivo and if any such correlation could be found between fibre size and fibre angulation. A group of 15 women volunteered to take part in the study. Repeated ultrasonographic recordings were made on five subjects on 3 consecutive days to test the repeatability of ultrasonographic measurement of fibre angulation. Both muscle morphological analyses and ultrasonographic measurements of fibre angulation were performed on the other 10 subjects. Ultrasonographic measurement of fibre angulation was found to be reproducible since no variation between measurements made on different days was found. When trying to correlate muscle fibre size to the muscle fibre angulation, measured ultrasonographically, no significant correlation was found.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

Six extraocular muscles of the grass snake, Natrix natrix L. together with their motor end plates were examined in the light and electron microscope, and the measurements of the diameter of muscle fibres and the area of their motor end plates were performed. Morphologically, two types of muscle fibres: tonic and red phase ones were distinguished. The former fibres, 2,3 to 14,5 mum in diameter possess single or multiple (up to five on a single fibre) "en grappe" motor end plates, without postsynaptic junctional folds. The latter fibres, 10...40 mum in diameter have single, "en plaque" motor end plates, with numerous postsynaptic junctional infoldings. The morphological features of muscle fibres and motor end plates as well as the correlation between the diameter of muscle fibres and the area of motor end plates are discussed.  相似文献

The effect of muscle hypertrophy on the relationship between magnetic resonance (MR) relaxation time and muscle fibre composition was investigated. Relaxation time and muscle fibre composition were measured in five subjects before and after a 20-week period of strength training. Muscle fibre composition in all subjects exhibited a significant shift to a predominance of fast-twitch (FT) fibres as a result of 20-week strength training (% area FT fibres: mean values from 49.8%, SD 17.9% to 57%, SD 5.6%; P less than 0.05). Longitudinal relaxation time (T1) and transverse relaxation time (T2) were prolonged significantly after strength training (T1 mean values from 334.9 ms, SD 13.6 to 359.0 ms, SD 9.0, P less than 0.001; T2 from 27.5 ms, SD 0.9 to 30.8 ms, SD 2.3, P less than 0.05). A constant relationship was observed in changes caused by strength training in muscle fibre composition (% area FT) and relaxation time, with a high correlation obtained between both parameters. These results indicate that MR relaxation time can be used for non-invasive estimation of muscle fibre composition.  相似文献

Based on previous observations that capsaicin can selectively damage group III and IV afferents and induce muscle fibre transformation, we hypothesized that eliminating, by means of capsaicin, the group III and IV afferents of a peripheral territory it could lead to a fibre transformation in a muscle involved in the flexor reflexes of the same peripheral territory. Therefore, capsaicin was injected into the palmar nerves of the forelimb of the horse to investigate if eliminating group III and IV afferents from the hand of the horse a muscle fibre transition would occur in the flexor carpi radialis (FCR) muscle, which is involved in the flexor reflexes of the finger itself. 120 days after capsaicin injection, type I slow fibres increased and type IIA fast fibres decreased. We presume that the long lasting deafferentation of the ergo-nociceptive fibres causes a plastic remodelling in the central nervous system and indirectly influences the motoneuron excitability via short or long loop-pathways enhancing their tonic discharge.  相似文献

The aims of this study were (1) to determine the relationship between muscle fibre cross-sectional area and cytoplasmic density of myonuclei in high- and low-oxidative Xenopus muscle fibres and (2) to test whether insulin and long-term high fibre length caused an increase in the number of myonuclei and in the expression of α-skeletal actin and of myogenic regulatory factors (myogenin and MyoD) in these muscle fibres. In high- and low-oxidative muscle fibres from freshly frozen iliofibularis muscles, the number of myonuclei per millimetre fibre length was proportional to muscle fibre cross-sectional area. The in vivo myonuclear density thus seemed to be strictly regulated, suggesting that the induction of hypertrophy required the activation of satellite cells. The effects of muscle fibre length and insulin on myonuclear density and myonuclear mRNA content were investigated on high-oxidative single muscle fibres cultured for 4–5 days. Muscle fibres were kept at a low length (~15% below passive slack length) in culture medium with a high insulin concentration (~6 nmol/l: “high insulin medium”) or without insulin, and at a high length (~5% above passive slack length) in high insulin medium. High fibre length and high insulin medium did not change the myonuclear density of isolated muscle fibres during culture. High insulin increased the myonuclear α-skeletal actin mRNA content, whereas fibre length had no effect on α-skeletal actin mRNA content. After culture at high fibre length in high insulin medium, the myonuclear myogenin mRNA content was 2.5-fold higher than that of fibres cultured at low length in high insulin medium or in medium without insulin. Myonuclear MyoD mRNA content was not affected by fibre length or insulin. These in vitro experiments indicate that high muscle fibre length and insulin enhance muscle gene expression but that other critical factors are required to induce adaptation of muscle fibre size and performance.This work was partially supported by a research grant from the Haak Bastiaanse Kuneman Stichting.  相似文献

Cellular adaptation of the trapezius muscle in strength-trained athletes   总被引:16,自引:4,他引:12  
 The aim of this study was to elucidate the cellular events that occur in the trapezius muscle following several years of strength training. In muscle biopsies from ten elite power lifters (PL) and six control subjects (C), several parameters were studied: cross-sectional area of muscle fibres, myosin heavy chain composition (MHC) and capillary supply [capillaries around fibres (CAF) and CAF/fibre area]. A method was also developed for counting the number of myonuclei and satellite cell nuclei. The proportion of fibres expressing MHC IIA, the cross-sectional area of each fibre type and the number of myonuclei, satellite cells and fibres expressing markers for early myogenesis were significantly higher in PL than in C (P<0.05). A significant correlation between the myonuclear number and the cross-sectional area was observed. Since myonuclei in mature muscle fibres are not able to divide, we suggest that the incorporation of satellite cell nuclei into muscle fibres resulted in the maintenance of a constant nuclear to cytoplasmic ratio. The presence of small diameter fibres expressing markers for early myogenesis indicates the formation of new muscle fibres. Accepted: 17 November 1998  相似文献

A high proportion of the "intermediate" muscle fibres, which are semi-dark after both acidic and alkaline preincubation in ATP-ase staining, was found in the thigh with heart failure. Characteristic patterns of fibre splitting were found mostly in jumpers' muscles. Possibilities of fibre subgroup transformation and fibre hyperplasia due to training are discussed.  相似文献

Summary Modern histochemical and immunohistochemical techniques have been used to type skeletal muscle fibres from threeRana species andXenopus laevis.Differing myosin properties and metabolic capacities (representing various contractile properties) define a minimum of four fibre types inRana and five inXenopus. TheRana andXenopus types are sufficiently similar so that a single nomencclature can be applied to them. This nomenclature uses an initial letter indicating the probable contractile performance (F=fast-twitch, S=slow-twitch and T=tonic), and a number indicating rank order of presumed shortening velocity.The largest, fastest fibres-F1-have low oxidative and, at best, moderate glycolytic capacities. Commonly adjacent to them are smaller, F2 fibres with variable but at least moderate metabolic capacities. F3 fibres are rarer and have on average the highest oxidative capacity, and at least moderate glycolytic capacity. They usually occur in the reddest parts of the muscle and, inRana, only in the vicinity of tonic fibres.Metabolically weak, classical amphibian tonic fibres (T5) occur in bothXenopus andRana, but onlyXenopus also has an S4 fibre type. This has moderate metabolic capacity and myosin properties suggesting it is probably capable of slow shortening as well as tonic hold. Immunohistochemically, S4 fibres are most similar to avian slow-twitch fibres.  相似文献

An analysis has been performed of the native myosin isoenzyme composition of isolated skeletal muscle fibres from Xenopus laevis with well-defined isotonic contraction properties. Fast twitch 'white' (type 1) fibres contained three isomyosins; fast twitch 'red' (type 2) fibres showed two major myosin bands with migration velocities very similar to those of the two slower bands in type 1. Slow twitch (type 3) fibres yielded a single, slowly migrating band as did slow tonic (type 5) fibres, whereas the myosin from type 4 (very slow twitch, 'intermediate') fibres migrated with a somewhat higher mobility. The results suggest that amphibian skeletal muscle may possess the principal fibre types found in mammals and birds.  相似文献

A skeletal muscle fibre maintains its cytoplasmic volume by means of hundreds of myonuclei distributed along its entire length. Therefore it is hypothesised that changes in fibre size would involve modifications in myonuclear number. In this study, we have examined whether 10 weeks of strength training can induce changes in the number of myonuclei and satellite cells in female trapezius muscles. Biopsies were taken pre- and posttraining from the upper part of the descending trapezius muscle of nine subjects. Muscle samples were analysed for fibre area and myonuclear and satellite cell number using immunohistochemistry. There was a 36% increase in the cross-sectional area of muscle fibres. The hypertrophy of muscle fibres was accompanied by an approximately 70% increase in myonuclear number and a 46% increase in the number of satellite cells. Myonuclei number was positively correlated to satellite cell number indicating that a muscle with an increased concentration of myonuclei will contain a correspondingly higher number of satellite cells. The acquisition of additional myonuclei appears to be required to support the enlargement of multinucleated muscle cells following 10 weeks of strength training. Increased satellite cell content suggests that mitotic divisions of satellite cells produced daughter cells that became satellite cells. Accepted: 30 November 1999  相似文献

Abstract. The myofibrillar ATPase (mATPase) activity and the pattern of expression of several myosin heavy chain (MHC) isoforms and of M-protein (Mr 165000) were studied in serial cross sections of neonatally deefferented 5- to 8-week-old rat hindlimb muscle spindles with supernumerary intrafusal fibres. In a sample of 5- to 6-week-old neonatally deefferented muscle spindles cut through the A region, the average number of intrafusal fibres per spindle was 8.4 in comparison to 4.2 in control spindles. Parent fibres extended throughout the whole encapsulated portion of the spindle, whereas supernumerary fibres were found only in the A region. The diameters of the supernumerary intrafusal fibres varied from less than 1 μ up to 10 μ approximately. On the basis of the mATPase activity and the pattern of expression of MHC isoforms and of M-protein, the vast majority of the supernumerary fibres could be classified as nuclear bag2, bag1 or chain fibres. However, some supernumerary fibres with small diameters exhibited features that did not fit any of the three known intrafusal fibre types. Two major processes, namely fibre splitting versus activation and fusion of satellite cells, might account for the formation of supernumerary fibres. The data presented suggest the existence of at least two types of intrafusal satellite cells. One type of satellite cell is related to the nuclear bag fibres and gives rise to myotubes which, if they have sensory innervation, can express slow tonic MHC and, therefore, differentiate into a phenotype similar to that seen in nuclear bag fibres. The other type of satellite cells form myotubes which attain a fast phenotype similar to that seen in nuclear chain fibres irrespective of the presence or absence of sensory innervation.  相似文献

The effect of an endurance training program lasting 17 weeks was studied in two cyprinid species, Chondrostoma nasus (L.) and Leuciscus cephalus (L.). Red, intermediate and white axial muscle were investigated. Morphometrical analysis revealed that training induced, in both species, increased red and intermediate muscle mass, fibre diameter and capillarization. Differences between species in the response to training were observed for volume densities of mitochondria and lipid. In contrast to C. nasus, L. cephalus show higher values for these compartments in red and intermediate fibres. The results are considered adaptational changes which increase the aerobic capacity of red and intermediate muscle fibres to meet higher sustained swimming activities.  相似文献

In frog twitch muscle fibres, Na-octanoate (NaC8) shifted the relation between potassium induced tension and membrane potential to the right. The present study has been carried out to investigate the effect of this fatty acid on frog tonic fibres. Potassium contractures measured on bundles of 30-40 fibres of ileofibularis muscles were less decreased by NaC8 (2.5-10 mmol/l) than those of twitch fibre bundles. In denervated muscles the sensitivity to NaC8 was increased, probably due to the development of sodium channels in the membranes. Experiments with mixed fibre bundles also showed a lower influence of NaC8 on potassium contracture of tonic fibres. On the other hand, tonic fibres showed a lower threshold of the potassium induced tension as well as a lower K+ concentration for maximal activation. This lower threshold was further lowered by NaC8, corresponding to a shift of the relation between potassium concentration and tension to the left. The membrane resting potentials were -58 +/- 9 mV in tonic fibres and -83 +/- 5 mV in twitch fibres. Five mmol/l NaC8 only induced depolarization of the membrane of tonic fibres. This depolarization (by about 20 mV) may be responsible for the threshold shift to lower K+ concentration in NaC8-exposed tonic fibres. In addition to the effects of NaC8 on sodium channels, interactions with Ca2+ binding sites are discussed.  相似文献

Mantle muscle tissue of Idiosepius pygmaeus was examined to describe changes in structure and organization associated with growth. Growth in I. pygmaeus|DD was a function of both an increase in muscle fibre number and fibre size within muscle blocks. Continuous fibre production over the observed life span of I. pygmaeus was indicated by the presence of very small muscle fibres (< 1.0 μm in diameter) in substantial proportions in all sizes of individuals. Muscle blocks became larger as animals increased in size, although new muscle blocks were generated in all sizes of individuals. Mantle muscle fibres had a maximum size of 11 μm. Therefore, for an individual to continue increasing muscle block sizes, new fibres must be produced. This is further evidence of continuous fibre production throughout the size range of I. pygmaeus examined. The relative rates of muscle fibre generation and fibre growth depended on the size of the animal and position along the mantle (anterior, mid or posterior mantle). The predominance of small fibres and blocks at the anterior end of the mantle suggested that this was the primary growth region. Mitochondriapoor and mitochondria-rich muscle fibres from small individuals had much larger mitochondrial cores than muscle fibres from larger animals. Changes in the muscle structure are discussed with respect to the metabolic and energetic requirements of I. pygmaeus , and how these may change with growth.  相似文献
