In S. holoschista, the vas deferens is differentiated into 3 regions based on the functional morphology of the epithelial lining. The proximal part with a highly secretory epithelial layer may be responsible for the secretion which is rich in protein, and moderately so in carbohydrate and lipid substances. The mid-region serves for storage of the spermatozoa. The distal part has specialized epithelial cells of a secretory nature. In addition, there are 4 typhlosole-like ridges fringed with cilia which probably aid in the conduction of the sperm cord during copulation. Secretions of the accessory reproductive glands contain a large number of discrete granules rich in mucoprotein. The principal function of this secretion appears to be the digestion of the sperm cord in the oviduct.  相似文献

We described the ultrastructure and histochemistry of the reproductive system of five Callinectes species, and evaluate the seasonal variation in weight of the reproductive system and hepatopancreas by comparing annual changes of somatic indices. The somatic indices changed little throughout the year. In Callinectes, spermatogenesis occurs inside the lobular testes and, within each lobule, the cells are at the same developmental stage. Spermatogenesis and spermiogenesis follow the same development pattern in all Callinectes studied. Mature spermatozoa are released into the seminiferous ducts through the collecting ducts. Cells of the vas deferens are secretory as evidenced by rough endoplasmic reticulum, Golgi complex, and secretory vesicles that produce the seminal fluid. The anterior vas deferens shows two portions: proximal and distal. In proximal portion (AVDp), spermatozoa are clustered and embedded in an electron-dense, basophilic glycoproteinaceous secretion Type I. In the distal portion (AVDd), the spermatophore wall is formed by incorporation of a less electron-dense glycoproteinaceous secretion Type II. The secretion Type I change to an acid polysaccharide-rich matrix that separates the spermatophores from each other. The median vas deferens (MVD) stores the spermatophores and produces the granular glycoproteinaceous seminal fluid. The posterior vas deferens (PVD) has few spermatophores. Its epithelium has many mitochondria and the PVD seminal fluid changes into a liquid and homogeneous glycoprotein. Many outpocketings in the PVD and MVD help to increase the fluid production. Overall, the reproductive pattern of Callinectes is similar to other species that produce sperm plugs. The secretions of AVD, MVD, and PVD are responsible for the polymerization that forms the solid, waxy plug in the seminal receptacle. The traits identified here are common to all Portunidae species studied so far.  相似文献

Accessory gland secretions of male insects have many important functions including the formation of spermatophores. We used light and electron microscopy to investigate the structure of the accessory glands and posterior vasa deferentia of the carabid beetle Pterostichus nigrita to try to determine where spermatophore material is produced. Each accessory gland and posterior vas deferens had an outer layer of longitudinal muscle, beneath which was a layer of connective tissue and a thin band of circular muscle, all of which surrounded a layer of epithelial cells lining the lumen of the ducts. Based on the ultrastructure of the epithelial cells, and their secretory products, we identified two epithelial cell types in each region (distal and proximal) of the accessory glands and four types in the posterior vas deferens. Most secretory products, which stained positively for proteins and some mucins, were released into the lumen of the ducts by apocrine secretion. The accessory glands produced one type of secretory product whereas in posterior vasa deferentia, four types of secretory products were found layered in the lumen. Our results suggest that most of the structural material used to construct a spermatophore is produced by the cells of the posterior vasa deferentia.  相似文献

This study aims to analyze the functional anatomy of the male reproductive system in Neocaridina davidi, a very popular ornamental species of caridean shrimp. Mature males were cold‐anaesthetized and their reproductive systems were dissected for histological and histochemical analysis, while the spermatozoa and spermatophore wall ultrastructure were analyzed under transmission electron microscopy. The male reproductive system consisted of two coiled testes, which were continuous with the vasa deferentia. Testes were positioned on the dorsal side of the cephalothorax above the hepatopancreas, and comprised seminiferous tubules where spermatogenesis occurred. Each vas deferens (VD) was a long tube dorsolaterally positioned with respect to the hepatopancreas, and increased in diameter at the distal end. Three regions could be recognized in the VD: proximal, middle, and distal. The proximal region had a cylindrical epithelium with secretory cells. The middle region (or typhlosole) had a dorsal fold (or typhlosole) with a thick columnar epithelium and high secretory activity. The spermatophore was a continuous cord with three acellular layers, which were mainly characterized by the presence of neutral glycoconjugates and proteins. The sperm morphology was distinct from the inverted cup‐shaped spermatozoa observed in the majority of caridean shrimps. The spermatozoa in specimens of N. davidi were spherical in shape, with a cross‐striated, single, short spike, and arranged in clusters of three or four sperm cells. The composition of the spermatophore, and the arrangement and form of the spermatozoa, seem to be unique in comparison to other species of Caridea.  相似文献

Vas deferens is a conduit for sperm and fluid from the epididymis to the urethra. The duct is surrounded by a thick smooth muscle layer. To map the actin cytoskeleton of the duct and its epithelium, we reacted sections of the proximal and distal regions with fluorescent phalloidin. Confocal microscopic imaging showed that the cylinder‐shaped epithelium of the proximal region has a thick apical border of actin filaments that form microvilli. The epithelium of the distal region is covered with tall stereocilia (13–18 µm) that extend from the apical border into the lumen. In both regions, the lateral and basal cell borders showed a thin lining of actin cytoskeleton. The vas deferens epithelium contains various channels to regulate the fluid composition in the lumen. We mapped the localization of the epithelial sodium channel (ENaC), aquaporin‐9 (AQP9), and cystic fibrosis transmembrane conductance regulator (CFTR) in the rat and mouse vas deferens. ENaC and AQP9 immunofluorescence were localized on the luminal surface and stereocilia and also in the basal and smooth muscle layers. CFTR immunofluorescence appeared only on the luminal surface and in smooth muscle layers. The localization of all three channels on the apical surface of the columnar epithelial cells provides clear evidence that these channels are involved concurrently in the regulation of fluid and electrolyte balance in the lumen of the vas deferens. ENaC allows the flow of Na+ ions from the lumen into the cytoplasm, and the osmotic gradient generated provides the driving force for the passive flow of water through AQP channels.  相似文献

An attempt was made here to study the structure of the male reproductive system of Portunus pelagicus, which would improve the knowledge base on the reproductive biology of the species and also help in the maintenance of broodstock under controlled conditions. Male P. pelagicus of different sizes were collected from the Palk Bay off Mandapam (9°17′ N, 79°9′ E) and maintained under controlled conditions for the study. Tissues from testis, anterior vas deferens (AVD), median vas deferens (MVD), posterior vas deferens (PVD), ejaculatory duct and penis were fixed in Bouin's fluid and 2.5% buffered glutaraldehyde separately and processed for light and electron microscopic studies, respectively. The reproductive system consisted of testis, commissure, vas deferens, ejaculatory duct and penis. The vas deferens was divided based on the morphology and/or histology into AVD, MVD and PVD. The AVD was further divided based on histology into proximal and distal regions, and the MVD, based on diameter into major and minor coils. The testicular lobe had several lobules with a central seminiferous tubule, which continued till the penis. The seminiferous tubule was lined by a layer of cuboidal or columnar epithelium. The lining of the central tubule of the vas deferens formed several ‘folds’, which at times formed ‘pouches’. High incidence of cell organelles in the columnar epithelial cells, aggregations of vesicles and occurrence of blebs at the luminal periphery and the projection of numerous microvilli containing electron‐dense materials into the lumen from the cell lining denoted high secretory activity of the epithelial cells.  相似文献

应用光镜和透射电镜技术研究了日本沼虾输精管的结构及其在精荚形成中的作用。结果表明,日本沼虾输精管从形态结构上可分为近端输精管、卷曲输精管、远端输精管和膨大的远端输精管四部分。各部分的管壁皆由分泌上皮、基膜、肌肉层和结缔组织构成,其中分泌上皮包括高度明显不同的低柱状上皮和高拄状上皮两部分。输精管各部分管腔内含有处于不同形成阶段的精荚。进入近端输精管内的精子被支撑在一种嗜酸性基质中。近端输精管的分泌物主要帮助形成精子团,同时形成精荚壁的极小部分。卷曲和远端输精管分泌形成精荚壁的绝大部分,其分泌物由细胞顶端通过外排作用和顶泌机制分泌产生。膨大的远端输精管具有贮存精荚的作用,其分泌上皮也通过外排作用和顶泌机制产生分泌物包裹在已基本形成的精荚外侧,管壁肌肉层在雌雄交配时将管腔内的精荚切割成适宜长度并排出体外。    相似文献

Extragonadal sperm reserves in male rats were measured in different regions of the genital tract before and subsequent to normal ejaculation. In sexually rested rats, the sperm count (million spermatozoa for the paired organs) in different regions was: distal vas, 18; proximal vas, 9.8; cauda epididymidis, 229; caput + corpus epididymidis, 154. Following mating, the sperm count was reduced in the proximal and distal vas deferens and in the cauda epididymidis. The reproductive tract of mated females was found to contain 29% (no copulatory plug) or 59% (with copulatory plug) of the estimated mean ejaculate, which was estimated from the difference between the sperm counts in the sexually rested rat and following ejaculation. It is concluded that in the rat the immediate source of spermatozoa for ejaculation is the cauda epididymidis, with a smaller contribution arising from the vas deferens.  相似文献

The organization of the dwarf male of Bonellia viridis was studied by electron microscopy. The epidermis is formed by two types of epithelial cells: the majority are multiciliated cells; highly vacuolated, non-ciliated cells are less abundant. The body wall musculature consists of an outer circular, a diagonal, and a longitudinal layer. As a unique feature in coelomate spiralians it was found that the perikarya of all muscle cells are located internal to the entire contractile muscular layer. The muscles are solitary myocytes embedded in extracellular matrix. Masses of secretory and indifferent cells occur inside the muscles. Two types of secretory cells were distinguished. Both of them apparently undergo holocrine secretion. A complete lining of thin peritoneal cells delimits the body cavity. Also, the gut and sperm sac have a complete peritoneal lining. The coelomic lining of the gut is a single-layered myoepithelium, that of the sperm sac a pseudo-stratified myoepithelium. The vas deferens was seen to be ciliated. The entrance of the sperm sac is formed by a ciliated funnel that leads into the reservoir by means of a thin, ciliated canal. The existence of repeated transverse nerves and of four longitudinal nerve cords is described for the first time.  相似文献

Each vas deferens of the Mediterranean flour moth, Anagasta kuehniella (Zeller), consists of a short swollen portion immediately below the testis, another swollen portion that forms a seminal vesicle, and an elongate lower portion that empties into one arm of the ductus ejaculatoris duplex. Three types of epithelial cells occur sequentially. Phagocytic cells that engulf debris from the testis form the anterior two-thirds of the first swollen portion. Tall secretory cells form the distal third of the first swollen region and extend to the seminal vesicles. The secretory cells surround a slit-like lumen and appear to function as a valve between the two swollen regions. Many membrane-enclosed secretory granules are stored at the apical ends of the cells and are released into the lumen together with small amounts of the surrounding cytoplasm. The granules remain intact while they are in the male tract. A second type of secretory cell forms the walls of the seminal vesicles and the lower vasa deferentia. These cells produce secretory granules whose contents become dispersed through the semen. PTA-chromic acid staining indicates that the seminal plasma has a high glycoprotein content. A thin muscle layer is basal to the epithelial cells. Both apyrene and eupyrene sperm undergo some development in the vasa deferentia. The epithelial cells, muscle, and stored sperm all undergo extensive changes with age.  相似文献

This paper details for the first time the gonad development characteristics and sex ratio of triploid shrimp (Fenneropenaeus chinensis). In triploid shrimp the development of gonad is apparently impaired, especially in females. In the ovary of triploids, germ cells mainly remain at oogonia stage during September through December. From January to February of the next year, partial primary oocytes developed in the ovary lobes. Spermatocytes and spermatids could be observed in the testes of triploids, and a few sperm were observed in the vas deferens and spermatophores. The morphology of sperms in triploid shrimp was abnormal. Flow cytometry was used to detect the ploidy of sperm in the vas deferens. The data showed that triploidy could affect the sex ratio in Chinese shrimp. The female-to-male ratio in triploids of about 4:1 will favor triploid shrimp aquaculture.  相似文献

Callichirus major inhabits the intertidal region of marine ecosystems and it is frequently used as live bait for fishing. This study aimed to describe the functional anatomy of the male reproductive system by microscopic techniques. The animals were collected along the Corujão beach, Piuma—ES, Brazil, and, in laboratory, the males were classified into two phases: immature (IM) and developed (DE) based on the macroscopic characteristics of the gonads. The gonad and vas deferens were dissected for histological routine and histochemical tests. Histologically, it was noted that in both phases, the more distal region of gonads has ovarian characteristics, showing developing oocytes. Also, different male germ cells were identified: spermatogonium (SPG), spermatocytes I and II (SPTCI, SPTCII), initial and final spermatid (IS, FS) and sperm (SPZ). Accessory cells with spherical or pyramidal nuclei were also present inside the testicular lobules. According to the vas deferens structure, three regions can be characterized: proximal (PVD), middle (MVD) and distal (DVD). In the lumen of the vas deferens, a spermatophoric matrix highly reactive for histochemical tests was observed. The presence of female germ cells in males suggests the occurrence of intersexuality or hermaphroditism in this species.  相似文献

The fertilizing capacity was compared between testicular and vas deferens sperm in Cynops pyrrhogaster. The testicular sperm was not capable of fertilizing jelly eggs. In contrast, the vas deferens sperm was already capable of fertilizing the newt jelly eggs. There was no inhibitory factor for fertilizing jelly eggs in the testis. These results suggest that the testicular sperm is immature as to the fertilizing capacity. The testicular sperm gained the fertilizing capacity for the jelly eggs by treatment with Holtfreter's solution or 1/20 strength Holtfreter's solution. The treatment may promote the step of maturation to achieve the fertilizing capacity. The treated testicular sperm did not fertilize dejellied eggs, although vas deferens sperm fertilized dejellied eggs. Therefore, the maturation state of the treated testicular sperm is different from that of vas deferens sperm. Newt sperm may be matured within the vas deferens, as the newt does not have an organ like the mammalian epididymis.  相似文献

Summary

The male reproductive tract of Scyllarus chacei consists of paired testes and vasa deferentia that conduct sperm containing spermatophores to the genital pores at the base of each fifth walking leg. The testis is joined to the vas deferens which can be divided into four regions: (1) the anterior vas deferens can be further divided into three regions. It is highly convoluted and is the region in which the sperm become encapsulated in ovoid spermatophores of approximately 100 sperm as well as produces seminal fluids. (2) The middle vas deferens is the primary site of sperm storage and adds to seminal fluids which formed in the anterior region. (3) The posterior region is highly muscularized and may serve for limited sperm storage. (4) The most distal portion is the ejaculatory duct which is highly muscularized for extruding the spermatophoric mass for transfer to the female. A final seminar product is added here.  相似文献

We have examined the anatomy of the vas deferens of the lobster Homarus americanus and have described the structure of the proximal vas deferens (segments one and two). The two tubes of segment one descend from the testes and gradually merge into segment two. The epithelium of segment one has synthetic activity and appears to contribute to the sperm-supporting matrix by exocytotic release of granules through its apical surface. The epithelium of segment two is also highly synthetic and secretes the primary spermatophore layer and part of the intermediate layer that surround the sperm mass. The trifoil shape of the extruded spermatophore is established through a change in height of some of the cells lining the lumen in segment two. Connective tissue and circular bands of striated muscle surround the epithelium of both segments.  相似文献

Pea crabs of the subfamily Pinnotherinae (Pinnotheridae) have a high investment in reproduction and an outstanding reproductive output, probably as an adaptation to the required increase in reproductive rate due to the pinnotherids small size and their parasitic, host‐dependant way of life. In the present study, we investigate the male internal reproductive structures and the ultrastructure of spermatozoa of Pinnotheres pisum and Nepinnotheres pinnotheres by histological methods and both scanning‐ and transmission electron microscopy. In the Brachyura, the male internal reproductive systems generally consist of paired testes and corresponding vasa deferentia where spermatozoa develop and mature. Spermatozoal ultrastructure of the investigated pinnotherids conforms to the thoracotreme type, however, N. pinnotheres has an accessory opercular ring and a periopercular rim, neither of which are present in spermatozoa of P. pisum. Spermatozoa are enclosed within spermatophores in the secretory proximal vas deferens. Two types of secretions were observed in P. pisum and N. pinnotheres: an electron dense substance secreted in the proximal vas deferens involved in spermatophore formation, and large electron‐luscent vesicles constituting the seminal plasma in the medial and distal vas deferens. The medial vas deferens is strongly widened compared to other brachyurans to purpose storing spermatophores embedded in seminal plasma. Tubular appendices, which produce and store large amounts of seminal plasma, arise from the distal region of the vas deferens. The appendices extend into the ventral cephalothorax and also in the first pleomere. The latter being an exceptional location for reproductive structures among male brachyurans. J. Release of mature bundles of spermatozoa from the testis into the vas deferens is a critical but poorly understood step in male insect reproduction. In moths, the release of sperm bundles is controlled by a circadian clock which imposes a temporal gate on the daily exit of bundles through the terminal epithelium-a layer of specialized epithelial cells separating testis follicles from the vas deferens. The sequence of cellular events associated with the daily cycle of sperm release was investigated by scanning and transmission electron microscopy. In the hours preceding sperm release, there is a solid barrier between the testis and the vas deferens formed by the interdigitation of cytoplasmic processes of adjacent terminal epithelial cells. At the beginning of the sperm release cycle, sperm bundles protrude through this barrier while the terminal epithelial cells change their shape and position relative to the bundles. Subsequently, the cyst cells enveloping the sperm bundles break down and spermatozoa move out of the testis through the exit channels formed between the epithelial cells. Afterwards, cyst cell remnants and other cellular debris are released into the vas deferens lumen, and the epithelial barrier is reconstructed due to phagocytic activity of its cells. These data provide a foundation on which to build an understanding of the cellular mechanisms of clock-controlled sperm release in insects.  相似文献

The objective was to examine, in vitro, the motility, acrosomal proteolytic activity (APA), and penetrating ability of fowl sperm recovered from the testis and epididymis, as well as the proximal, middle, and distal vas deferens, to assess the potential fertilizing ability of sperm as a function of maturation. A motile sperm separation technique was used to estimate sperm motility with Accudenz, a gelatin slide technique was used to measure the diameter of the halo around the acrosome of individual sperm as an indication of APA, and a sperm-inner perivitelline layer (IPL) interaction assay was done to estimate the number of hole formations as an indication of sperm penetration into the IPL. Sperm in the testis exhibited the least motility, produced the smallest halos, and created the least number of holes per 0.25 mm2. Motility, diameter of the halo, and number of holes increased gradually (P < 0.05) from the epididymis to the distal vas deferens and were markedly different (P < 0.05) between testicular and deferent duct sperm. Based on these in vitro experimental findings, we inferred that fowl sperm undergo a gradual process of maturational changes in motility, APA, and penetrability as a means of acquiring potential fertility during their passage throughout the male genital tract.  相似文献
