Olfactory neuropathology is a cause of olfactory loss in Alzheimer’s disease (AD). Olfactory dysfunction is also associated with memory and cognitive dysfunction and is an incidental finding of AD dementia. Here we review neuropathological research on the olfactory system in AD, considering both structural and functional evidence. Experimental and clinical findings identify olfactory dysfunction as an early indicator of AD. In keeping with this, amyloid-β production and neuroinflammation are related to underlying causes of impaired olfaction. Notably, physiological features of the spatial map in the olfactory system suggest the evidence of ongoing neurodegeneration. Our aim in this review is to examine olfactory pathology findings essential to identifying mechanisms of olfactory dysfunction in the development of AD in hopes of supporting investigations leading towards revealing potential diagnostic methods and causes of early pathogenesis in the olfactory system.  相似文献

Summary The terminal nerve has been described in all vertebrate classes, with the exception of cyclostomes and birds. With regard to this question, we have examined representatives of these two classes using tracer techniques, and found a terminal nerve in larval lampreys and young domestic mallards. Horseradish peroxidase or cobaltous lysine was injected into the olfactory mucosa, which is known to be innervated by peripheral branches of the terminal nerve. The brains were then searched for labeled, centrally directed fibers of the terminal nerve that project further caudally than the glomerular layer of the olfactory bulb. In larval lampreys, centrally projecting fibers of the terminal nerve were found in the tel-, diand mesencephalon. Termination of labeled fibers was observed in the hypothalamus. Some fibers of the terminal nerve cross to the contralateral side via the commissure of the posterior tuberculum. In young ducks, the terminal nerve projects ipsilaterally along the medial edge of the telencephalon.  相似文献

Recent studies have indicated that avian social behavior is influenced by olfactory cues. During the reproductive season a change in the chemical composition of uropygial gland secretion has been reported in some species and the hypothesis that olfactory signals may be produced by this gland has been proposed. To examine this hypothesis we performed two behavioral experiments to determine whether a female’s uropygial gland produce chemical signals that stimulate mating behaviors in domestic chickens. In Experiment 1 the role of the female’s uropygial gland in male mating behavior was examined by removing and examining the female’s uropygial gland. The frequency of mounts and copulations of intact male birds with sham-operated female birds was significantly higher than with uropygial glandectomized female birds. With respect to the number of waltzing that is one of the courtship displays intact males showed no significant difference between sham-operated female birds and uropygial glandectomized female birds. In Experiment 2 the relationship between male olfaction and the female’s uropygial gland was investigated using olfactory bulbectomized male birds. The number of mounts and copulations of sham-operated male birds with sham-operated female bird was significantly higher than with uropygial glandectomized female birds. In contrast olfactory bulbectomized male birds showed no significant differences in the number of mounts and copulations between sham-operated female birds and uropygial glandectomized female birds. These results indicate that intact and sham-operated male birds prefer to mate with female birds with the uropygial gland. The number of courtship waltzing of sham-operated male birds showed no significant difference. However olfactory bulbectomized male birds significantly courted to uropygial glandectomized female birds. Summarizing our results show that while anosmic males did not have any preference both intact and sham-operated male birds chose to mate with female birds having an intact uropygial gland suggesting that mate preference involves in male olfaction and that the female’s uropygial gland acts as a source of social odor cues in domestic chickens.  相似文献

Summary Primary olfactory projections in king salmon fry, Oncorhynchus tshawytscha, were studied with the cobaltlysine technique and after sectioning the entire head in a frozen state. The labeled axons can be traced from the olfactory epithelium, where cobalt was applied, into the olfactory bulb and to the ventral and lateral regions of the ventral telencephalon. The latter projection has not previously been reported, and may in actuality represent a transneuronal transport of cobalt. The terminations in the glomerular layer and in the external cellular layer of the bulb appear to be distributed differently in different parts of the bulb, suggesting regional specializations. A few neurons in the bulb were also always labeled suggesting that they may project to the olfactory epithelium.  相似文献

Summary The olfactory tract of the African catfish, Clarias gariepinus, consists of two tracts, the medial and lateral olfactory tract. Ovulated female catfish are attracted by male steroidal pheromones. Attraction tests with catfish in which the medial and lateral olfactory tract have been selectively lesioned show that the effects of these pheromones are mediated by the medial olfactory tract. The central connections of the medial and lateral olfactory tract have been studied by retro- and anterograde transport techniques using horseradish peroxidase as a tracer. Upon entering the forebrain, the medial olfactory tract innervates the posterior pars ventralis and pars supracommissuralis of the area ventralis telencephali and the nucleus preopticus periventricularis, the nucleus preopticus and the nucleus recessus posterioris. Application of horseradish peroxidase to the olfactory epithelium shows that part of the innervation of the area ventralis telencephali and the nucleus preopticus periventricularis can be attributed to the nervus terminalis, which appears to be embedded in the medial olfactory tract. The lateral olfactory tract sends projections to the same brain areas but also innervates the nucleus habenularis and a large terminal field in the area dorsalis telencephali pars lateralis ventralis. Furthermore, the medial olfactory tract carries numerous axons from groups of perikarya localized in the area dorsalis telencephali. Contralateral connections have been observed in the olfactory bulb, telencephalon, diencephalon and mesencephalon. It is suggested that processes of the medial olfactory tract innervating the preoptic region may influence the gonadotropin-releasing hormone system and in doing so may lead to behavioral and physiological changes related to spawning.  相似文献

Little is known about the olfactory capabilities of extinct basal (non-neornithine) birds or the evolutionary changes in olfaction that occurred from non-avian theropods through modern birds. Although modern birds are known to have diverse olfactory capabilities, olfaction is generally considered to have declined during avian evolution as visual and vestibular sensory enhancements occurred in association with flight. To test the hypothesis that olfaction diminished through avian evolution, we assessed relative olfactory bulb size, here used as a neuroanatomical proxy for olfactory capabilities, in 157 species of non-avian theropods, fossil birds and living birds. We show that relative olfactory bulb size increased during non-avian maniraptoriform evolution, remained stable across the non-avian theropod/bird transition, and increased during basal bird and early neornithine evolution. From early neornithines through a major part of neornithine evolution, the relative size of the olfactory bulbs remained stable before decreasing in derived neoavian clades. Our results show that, rather than decreasing, the importance of olfaction actually increased during early bird evolution, representing a previously unrecognized sensory enhancement. The relatively larger olfactory bulbs of earliest neornithines, compared with those of basal birds, may have endowed neornithines with improved olfaction for more effective foraging or navigation skills, which in turn may have been a factor allowing them to survive the end-Cretaceous mass extinction.  相似文献

Summary Central connections of the olfactory bulb of Polypterus palmas were studied with the use of horseradish peroxidase and cobalt-tracing techniques. The olfactory bulb projects to subpallial and palliai areas in the ipsilateral telencephalon; a projection to the contralateral subpallium is noted via the habenular commissure. A further target of secondary olfactory fibers is a caudal olfactory projection area in the ipsilateral hypothalamus. No labeling was seen in the anterior commissure and in the contralateral olfactory bulb. The medial and the lateral pallium receive secondary olfactory fibers in distinct areas. Neurons projecting to the bulb are found in the ipsilateral subpallium, mainly in one dorsal longitudinal nucleus. The main connection with the tel- and diencephalon is mediated via the medial olfactory tract. This tract also contains fibers to the contralateral telencephalon, and to the hypothalamus. The smaller lateral olfactory tract mediates fibers to the lateral pallium. The organization of pathways of secondary olfactory fibers in the telencephalon is described. Several workers have investigated the effect of anosmia on pigeon navigation in different geographical locations because it has been suggested that homing behavior is based on different cues, such as olfactory cues, the Earth's magnetic field or infrasound, and that in the absence of one cue another would be used. In this situation, no cue is universally indispensable, including olfactory ones. In order to extend such observations to a novel biome, we observed the behaviour of 192 young inexperienced birds raised in southeastern Brazil, a tropical area where olfactory tests had never been run before. The birds were released from eight symmetrically distributed sites 17 to 44 km from the loft. Half of these birds (experimentals) had been made temporarily anosmic by washing their olfactory mucosae with 4% solution of ZnSO4 the day before release, while controls were treated with Ringer solution. The results of release tests showed that anosmia totally impaired the navigational performance of experimental birds, which were unable to home from sites at relatively short distances from home (34-44 km) and whose pooled initial bearings produced a (negative) homeward component not significantly different from 0. Homing performance of controls was significantly better, and their pooled vanishing bearings had a significant homeward component, in spite of much scatter in individual releases. We conclude that pigeon homing in the study area depends on olfactory information, even though local environmental conditions in the interior of the State of Sao Paulo, as in several other parts of the world, do not appear to be as favorable as Italy for the development of efficient olfactory navigation.  相似文献

Summary Neurons containing luteinizing hormone-releasing hormone (LHRH) are first detected in newt embryos (Cynops pyrrhogaster) in the olfactory epithelium and ventromedial portion of the olfactory nerve, after which they sequentially appear in the intracerebral course of the terminal nerve at prometamorphosis, and in the septo-preoptic area at postmetamorphosis. In adults, however, LHRH-immunoreactive cells are rarely seen in the nasal region, and their distribution shifts into the brain, suggesting their migration. In order to ascertain the origin and possible migration route of these neurons in newt larvae, the effect of unilateral or bilateral olfactory placodectomy on the LHRH neuronal system has been studied. Removal of the olfactory placode results in the absence of LHRH-immunoreactive cells in the nasal and brain regions of the operated side, whereas the subsequent growth and the LHRH-immunoreactive cellular distribution in the contralateral side are identical to those of normal larvae. Following bilateral placodectomy, no LHRH immunoreactivity is detected on either side of the olfactory-brain axis. These results suggest that LHRH neurons of the newt, Cynops pyrrhogaster, originate in the olfactory placode and then migrate into the brain during embryonic development.  相似文献

Summary We have investigated the central connections of the classical olfactory system in the weakly electric fish Gnathonemus petersii using HRP and cobalt labelling techniques. The olfactory bulb projects bilaterally via the medial and lateral olfactory tracts to restricted areas of the telencephalon, namely to its rostromedial, lateral and posterior medial parts. The most extensive telencephalic target is the posterior terminal field, which arcs around the lateral forebrain bundle at levels posterior to the anterior commissure. Projections to the contralateral hemisphere cross in the ventral telencephalon rostral to the anterior commissure and via the posterior dorsal part of the anterior commissure; endings are also present within the anterior commissure. Bilateral projections to the preoptic area, to the nucleus posterior tuberis and to an area in the thalamus are apparent. In all cases, contralateral projections are less extensive than those on the side ipsilateral to the injected bulb. A projection via the medial olfactory tract can be followed to the contralateral bulb. Following injections into the olfactory bulb, retrogradely labelled neurons are found in the contralateral bulb and in six telencephalic areas; they are also present in the periventricular diencephalon and in an area lateral to the nucleus posterior tuberis. The present results support the suggestion that a reduction in olfactory input to the telencephalon occurs together with increased telencephalic differentiation in actinopterygian fishes.  相似文献

A prominent hypothesis for the function of the glomerular structures in the primary olfactory neuropil of many groups of vertebrate and invertebrate animals is that they enable the processing and coding of information about the chemical compounds that compose complex odors. Previous studies have indicated that various degrees of glomerulus formation in the antennal lobes of the brain of the moth Manduca sexta can be effected by reducing the number of olfactory sensory axons that grow from the antenna into the antennal lobe during metamorphosis. To test the hypothesis that the presence of glomerular structure is necessary to process and identify odors, we substantially reduced, by surgery, the number of antennal segments in developing moths and upon metamorphosis we observed and quantified behavioral responses known to be elicited by odors. Intact and lesioned adult female moths were challenged to fly upwind to the source of an attractive host-plant odor in a wind tunnel. Some of the moths that had developed with reduced olfactory input flew upwind to the odor source. The flight behavior of these individuals was similar to the odor-mediated flight typically observed in moths that had developed normally. Histological analysis of the moths antennal lobes revealed that the lobes of more than half of the respondents that had been lesioned during development lacked normal glomerular organization. The neuropil of these abnormally developed antennal lobes was mostly aglomerular, but with a few isolated, clearly abnormal glomerulus-like structures. This suggests either that even a few abnormal glomeruli are sufficient to mediate this specific behavior or that canonical glomerular organization per se is not necessary for this odor-mediated behavior.  相似文献

The detection of odorant signals from the environment and the generation of appropriate behavioral outputs in response to these signals rely on the olfactory system. Olfactory sensory neurons (OSNs) of the olfactory epithelium are located in the nasal cavity and project axons that synapse onto dendrites of second-order neurons in the olfactory bulb (OB) that in turn relay the information gathered to higher order regions of the brain. The connections formed are remarkably accurate such that axons of OSNs expressing the same olfactory receptor innervate specific glomeruli within the complex three-dimensional structure that represents the OB. The molecular determinants that control this complex process are beginning to be identified. In this review, we discuss the role of various families of axon guidance cues and of recently characterized families of adhesion molecules in the formation of stereotypic connections in the olfactory system of mice. In dealing with the spatial and temporal variability of prey species, predators may be able to forage optimally if they have flexible and rapid behavioral plasticity rather than predetermined responses. For predators that learn to focus attention on the cryptic prey type most frequently encountered during recent searching (termed a “search image”), rare prey types may be overlooked because of a focus on more common prey. Search imaging reflects biased searching for one of a number of available prey types, and has been studied widely in birds and mammals. Here we discuss the significant implications of this phenomenon for insect predator–prey systems, particularly with respect to parasitic wasps searching for host species using learned olfactory cues. We (1) review studies about perceptual development through individual ontogeny, (2) define the term “search image” and discuss the cognitive mechanisms involved in search-image formation, (3) discuss the role of search images and frequency-dependent predation as a proximate mechanism in the maintenance of prey diversity, (4) examine data on host–parasitoid olfactory search imaging, and (5) conclude by identifying important research areas for future studies in the field of olfactory search images.  相似文献

哺乳动物的嗅觉系统拥有惊人的能力,它可以识别和分辨成千上万种分子结构各异的气味分子。这种识别能力是由基因决定的。近年来,分子生物学和神经生理学的研究使得我们对嗅觉识别的分子基础和嗅觉系统神经连接的认识有了质的飞跃。气味分子的识别是由一千多种气味受体完成的,鼻腔中的嗅觉感觉神经元表达这些气味受体基因。每个感觉神经元只表达一种气味受体基因。表达同种气味受体的感觉神经元投射到嗅球表面的一个或几个嗅小球中,从而在嗅球中形成一个精确的二维连接图谱。了解嗅球对气味信息的加工和处理方式是我们研究嗅觉系统信号编码的一个重要环节。文章概述并总结了有关嗅球信号处理的最新研究成果。  相似文献

A central nervous system isolate from an acquired immunodeficiency syndrome (AIDS) patient of 103 Cryptococcus neoformans cells was instilled intranasally into guinea pigs. These were killed to evaluate the fate of the organisms and the response of the olfactory mucosa. Olfactory epithelium prevented the penetration ofCryptococcus neoformans and showed focal hyperplastic responses. The organisms, which manifested an affinity for the olfactory portion of the nasal cavities, were cleared from the olfactory space with no intervention from the immune system cells. By the end of the fifth week almost no organisms could be found and there was no histological evidence of dissemination. In contrast, destruction of the olfactory epithelium with zinc sulfate allowed the invasion of the subepithelial tissues, demonstrating the role of the olfactory mucosa in preventing infection withCryptococcus neoformans through the nasal route. The results and the model described in this report may be useful in clarifying the pathogenic mechanisms of cryptococcosis and the non immune mediated host responses toCryptococcus neoformans.  相似文献

Olfaction provides chemical information to an animal about its environment. When environmental conditions change, individuals should be able to adequately maintain function. Temperature may influence olfaction in a double manner, as it modifies the concentrations of gaseous compounds and affects biological processes. Here, we address acclimatization to environmental temperature in the olfactory system of Drosophila melanogaster using heat and cold treatments. Because the consequences of temperature shifts persist for some time after the treatment's end, comparison of olfactory behaviors at the same temperature in treated and untreated flies allows us to infer the biological effects of temperature in olfaction.At intermediate odorant concentrations heat always generates a reduction of olfactory sensitivity, as they would be expected to compensate for the increase of volatiles in the air. Cold produces the opposite effect. These changes are observed in both sexes and in natural populations as well as in standard laboratory stocks.Short applications suffice to cause detectable olfactory perception changes, but even prolonged temperature treatments have only a transitory effect. Together, these results suggest that olfaction in Drosophila underlies acclimatization to environmental temperature. However, sensitivity changes are not immediate and may cause imperfect adjustment of olfactory function for short time periods.  相似文献

Summary Neurons displaying FMRFamide(Phe-Met-Arg-Phe-NH2)-like immunoreactivity have recently been implicated in neural plasticity in salmon. We now extend these findings by describing the extent of the FMRF-like immunoreactive (FMRF-IR) system in the brain, retina and olfactory system of sockeye salmon parr using the indirect peroxidase anti-peroxidase technique. FMRF-IR perikarya were found in the periventricular hypothalamus, mesencephalic laminar nucleus, nucleus nervi terminalis and retina (presumed amacrine cells), and along the olfactory nerves. FMRF-IR fibers were distributed throughout the brain with highest densities in the ventral area of the telencephalon, in the medial forebrain bundle, and at the borders between layers III/IV and IV/V in the optic tectum. High densities of immunoreactive fibers were also observed in the area around the torus semicircularis, in the medial hypothalamus, median raphe, ventromedial tegmentum, and central gray. In the retina, immunopositive fibers were localized to the inner plexiform layer, but several fiber elements were also found in the outer plexiform layer. The olfactory system displayed FMRF-IR fibers in the epithelium and along the olfactory nerves. These findings differ from those reported in other species as follows: (i) FMRF-IR cells in the retina have not previously been reported in teleosts; (ii) the presence of FMRF-IR fibers in the outer plexiform layer of the retina is a new finding for any species; (iii) the occurrence of immunopositive cells in the mesencephalic laminar nucleus has to our knowledge not been demonstrated previously.  相似文献

Immunocytochemical and histochemical methods have been used to describe the neuronal population migrating from the rat olfactory placode and to analyze the spatio-temporal evolution of this neuronal migration during development. Several neuronal markers, such as binding to the lectin Ulex europaeus (UEA I) and the presence of neuron-specific enolase (NSE), olfactory marker protein (OMP), and luteinizing hormone-releasing hormone (LHRH), have been tested in order to determine whether migrating neurons originate from both the medial and the lateral parts of the placode and whether they all express LHRH. Our data show that a large population of differentiated migrating neurons can be identified with an antibody against NSE from the 14th day of gestation and with UEA I one day later. Migrating neurons are closely associated with both the vomeronasal axon fascicles emerging from the medial pit and the olfactory axons originating from the lateral pit. However, the neuron migration from the lateral pit appears to be more discrete than that from the medial pit. No LHRH immunoreactivity has been detected among neurons migrating from the lateral pit. Some neurons accompanying the olfactory axon fascicles exhibit a high level of maturation as shown by their OMP-positivity. Numerous neurons positive for both NSE and UEA I have also been observed within the presumptive olfactory nerve layer in early embryonic stages.  相似文献

培养的嗅鞘细胞的最终纯度受到多种因素的影响,如嗅鞘细胞的取材来源、分离方法等等;对培养的嗅鞘细胞进行纯化可获得高纯度的嗅鞘细胞。纯化嗅鞘细胞的方法有许多种,主要有单纯差速贴壁法、免疫吸附法、化学药物抑制法、无血清饥饿法等,现在的实验研究更趋向于以上2-3种方法联合应用对嗅鞘细胞进行纯化,这些联合纯化方案主要是在采用单纯差速贴壁方法的基础上再次运用其他一种或几种方法进行嗅鞘细胞的纯化。就获取的嗅鞘细胞的最终纯度而言,许多方法取得了可观的效果。但不同的纯化方法各有利弊,除了价格不同外,不同的纯化方法对嗅鞘细胞的生物活性造成不同程度的影响。因此在选择纯化方法时,应综合考虑各方面因素,根据研究目的和实际需要选择合理的方案进行纯化。本文通过查阅各数据库中与嗅鞘细胞的分离培养及纯化有关的文献和其他相关书籍,来探讨纯化嗅鞘细胞的不同方法以及这些纯化方法对嗅鞘细胞最终纯度的影响。  相似文献
