Utilizing genome sequence data from bacterial and fungal pathogens for the discovery of new antimicrobial agents has received considerable attention, both practical and critical, from the pharmaceutical and biotechnological communities. Although no new drugs derived from genomics-based discovery have been reported to be in a development pipeline, the utilization of genomics has revolutionized many aspects of drug discovery. The application, utility, opportunity, and challenges afforded by many of these new approaches are discussed.  相似文献

In this paper we develop a general modeling framework within which many models for systems which produce events at irregular times through a combination of probabilistic and deterministic dynamics can be comprehended. We state and prove new sufficient conditions for the global asymptotic behaviour of the density evolution in these systems, and apply our results to many previously published models for the cell division cycle. Protein complexes perform many important functions in the cell. Large-scale studies of protein-protein interactions have not only revealed new complexes but have also placed many proteins into multiple complexes. Whilst the advocates of hypothesis-free research touted the discovery of these shared components as new links between diverse cellular processes, critical commentators denounced many of the findings as artefacts, thus questioning the usefulness of large-scale approaches. Here, we survey proteins known to be shared between complexes, as established in the literature, and compare them to shared components found in high-throughput screens. We discuss the various challenges to the identification and functional interpretation of bona fide shared components, namely contaminants, variant and megacomplexes, and transient interactions, and suggest that many of the novel shared components found in high-throughput screens are neither the results of contamination nor central components, but appear to be primarily regulatory links in cellular processes.  相似文献

Microorganisms and plants synthesize a diverse array of natural products, many of which have proven indispensable to human health and well-being. Although many thousands of these have been characterized, the space of possible natural products--those that could be made biosynthetically--remains largely unexplored. For decades, this space has largely been the domain of chemists, who have synthesized scores of natural product analogs and have found many with improved or novel functions. New natural products have also been made in recombinant organisms, via engineered biosynthetic pathways. Recently, methods inspired by natural evolution have begun to be applied to the search for new natural products. These methods force pathways to evolve in convenient laboratory organisms, where the products of new pathways can be identified and characterized in high-throughput screening programs. Carotenoid biosynthetic pathways have served as a convenient experimental system with which to demonstrate these ideas. Researchers have mixed, matched, and mutated carotenoid biosynthetic enzymes and screened libraries of these "evolved" pathways for the emergence of new carotenoid products. This has led to dozens of new pathway products not previously known to be made by the assembled enzymes. These new products include whole families of carotenoids built from backbones not found in nature. This review details the strategies and specific methods that have been employed to generate new carotenoid biosynthetic pathways in the laboratory. The potential application of laboratory evolution to other biosynthetic pathways is also discussed.  相似文献

Helical filaments were the first structures to be reconstructed in three dimensions from electron microscopic images, and continue to be extensively studied due to the large number of such helical polymers found in biology. In principle, a single image of a helical polymer provides all of the different projections needed to reconstruct the three-dimensional structure. Unfortunately, many helical filaments have been refractory to the application of traditional (Fourier-Bessel) methods due to variability, heterogeneity, and weak scattering. Over the past several years, many of these problems have been surmounted using single-particle type approaches that can do substantially better than Fourier-Bessel approaches. Applications of these new methods to viruses, actin filaments, pili and many other polymers show the great advantages of the new methods.  相似文献

Although there were many ancient Chinese mathematicians contributed a lot on geometry,Geometric morphometrics (GM) in modern concept was not firstly proposed by Chinese.The super capability of geometric morphometrics in scientific computing and problem solving has gained a lot of attentions in the world.Until early of 21 centuries,geometric morphometrics was introduced into China.Since then,GM was rapidly applied in many research fields.However,it is a pity that GM is still not well-known in China as many works are published out of China.Thus,the special issue "Geometric morphometrics:Current shape and future directions" is organized.The present issue presents a series of contributions in this scientific field.In future,there will be many considerable new developing fields on GM needed to pay more attentions,for instances,3D geometric morphometrics,4D analysis,visualization of amber,new machine developing,new software developing,automatic identification system,etc.Once these technical bottle-necks on 3D data collecting and merging geometric morphometric data from multiple characters could be solved,the automatic identification system and other fields based on Big Data would come true.  相似文献

Rapid increases in taxonomic diversity are generally described as adaptive or evolutionary radiations. Such radiations differ widely in the rate and extent of morphologic innovation, taxonomic diversification and phylogenetic breadth, suggesting that several patterns, and likely processes, are involved. At least four distinct patterns of evolutionary radiation can be identified: novelty events, which generate new morphological complexity (altering the body plan of the group under consideration) but not necessarily with the associated production of many lower taxa; broad diversification events involving many independent lineages that undergo diversification, generate many new species and are driven by new ecological opportunities; economic radiations of a limited group of ecologically (but not necessarily phylogenetically) related clades exploiting a limited new ecologic opportunity; and adaptive radiations that may occur at any taxonomic level, but involve a rapid increase in diversity within a single clade, including “true”; adaptive radiations. Many events produce simple diversity increases with no corresponding increase in genetic/developmental/morphological/behavioral sophistication, but the most evolutionarily interesting events add new levels of complexity.  相似文献

Investigators have used many techniques to understand diet and tooth use in prehistoric species. A promising new addition to the analytical arsenal is dental microwear analysis—the study of microscopic wear patterns on teeth. On-going work is proceeding on a number of fronts. Studies of modern animals are showing some of the limits of resolution of the technique, while studies of prehistoric animals are answering many new questions and raising still more in the process. Much work remains to be done, but one thing is clear: if we proceed cautiously, we can provide new insights into the evolution of diet and tooth use.  相似文献

Discussions that are aimed at understanding the maintenance of sexual reproduction are in a bit of a quagmire owing to the many competing theories that have been proposed. Also, one of the central observations that asexual lineages are typically short lived still needs to be properly quantified. Exciting new results on ancient asexual organisms show that lineages can persist for many millions of generations without recombination. Recent developments in NMR spectroscopy verify that NMR continues to be an exciting area of research. These advances can be placed into three general categories: new hardware; new techniques; and novel applications. The hardware developments include many advances in the area of flow NMR and some new probe designs. The new techniques include several ways to edit the NMR spectra of mixtures without using chromatographic separation. These new NMR tools are now allowing us to analyze complex mixtures, combinatorial-chemistry libraries, bound drugs, unstable compounds, very small samples, and heterogeneous samples.  相似文献

A new phosphorylating agent for nucleosides, 2-O-(4,4'-dimethoxytrityl) ethylsulfonylethan-2'-yl-phosphate (1), has been developed by us. In the many examples studied by us, phosphorylation yields were found to be very high (about 90%). The procedure appears to be remarkably general and can be utilized for the phosphorylation of many biomolecules. Successful application of this phosphorylation method has contributed to the discovery of inhibitors of HIV integrase in our laboratory.  相似文献

The longstanding, successful use of herbal drug combinations in traditional medicine makes it necessary to find a rationale for the pharmacological and therapeutic superiority of many of them in comparison to isolated single constituents. This review describes many examples of how modern molecular–biological methods (including new genomic technologies) can enable us to understand the various synergistic mechanisms underlying these effects. Synergistic effects can be produced if the constituents of an extract affect different targets or interact with one another in order to improve the solubility and thereby enhance the bioavailability of one or several substances of an extract. A special synergy effect can occur when antibiotics are combined with an agent that antagonizes bacterial resistance mechanisms. The verification of real synergy effects can be achieved through detailed pharmacological investigations and by means of controlled clinical studies performed in comparison with synthetic reference drugs. All the new ongoing projects aim at the development of a new generation of phytopharmaceuticals which can be used alone or in combination with synthetic drugs or antibiotics. This new generation of phytopharmaceuticals could lend phytotherapy a new legitimacy and enable their use to treat diseases which have hitherto been treated using synthetic drugs alone.  相似文献

Canopy gaps are important as entry points for new genotypes and new species into many types of vegetation, yet little is known about them in any type of vegetation but forests. Forest gaps are too large for manipulative experiments to be readily undertaken, and hitherto grassland gaps have been too small to be easily mapped. Preliminary results from mapping small (>1 cm) grassland gaps with a new fibre-optic device suggest that experiments need to be performed at a smaller physical scale than has hitherto been achieved.  相似文献
