In SO2-fumigated spinach leaves under light, chloroplast SHenzymes, glyceraldehyde-3-phosphate dehydrogenase (NADP-GAPD)(EC 1.2.1.13 [EC] ), ribulose-5-phosphate kinase (Ru5PK) (EC 2.7.1.19 [EC] )and fructose-1,6-bisphosphatase (FBPase) (EC 3.1.3.11 [EC] ) weremore remarkably inactivated than other chloroplast enzymes.Their activities recovered after removal of SO2. The inactivationparalleled light-dependent CO2-fixation in spinach leaves. Inilluminated chloroplasts isolated from SO2-fumigated spinachleaves, NADP-GAPD and Ru5PK were more specifically in activatedthan other chloroplast enzymes. These two enzymes could be protectedfrom the inactivation by adding catalase. The NADP-GAPD inactivationwas suppressed by DCMU, cytochrome c or anaerobic conditions.By adding thiol compounds, the NADP-GAPD inactivation was dischargedand the activity increased. In chloroplasts or crude extractsfrom non-fumigated spinach leaves, NADP-GAPD and Ru5PK weremore strongly inhibited by externally added H2O2 than otherchloroplast enzymes. All results supported the idea that thesuppression of photosynthesis at the beginning of SO2 fumigationwas caused by the reversible inhibition of chloroplast SH enzymewith H2O2. Chlorophyll a and carotenoids of spinach began to be destroyed2 to 3 hr after fumigation with 2 ppm SO2 under light, whereaschlorophyll b was undamaged during 8 hr of exposure to SO2.Pheophytin a was not affected by the fumigation. Chlorophylla destruction was inhibited by phenazine methosulfate but stimulatedby methyl viologen. Addition of superoxide dismutase (SOD) tothe homogenate of SO2-fumigated leaves inhibited the chlorophylla destruction. The activity of endogenous SOD was reduced to40% by 2-hr fumigation before the loss of chlorophyll was observed.These results suggest that chlorophyll a destruction by SO2was due to superoxide radicals (O2). Moreover, malondialdehyde (MDA), a product of lipid peroxidation,was formed in SO2-fumigated leaves. MDA formation was inhibitedby tiron, hydroquinone and DABCO but not by benzoate and formate.MDA formation was increased by D2O. Distribution of iron-containing oxidases in aging nodal rootsof rice and wheat was studied. Activities of cytochrome c oxidase(1.9.3.1 [EC] , cytochrome c : O2 oxidoreductase), catalase (1.11.1.6 [EC] ,H2O2: H2O2 oxidoreductase) and peroxidase (1.11.1.7 [EC] , donor:H2O2 oxidoreductase) in wheat roots were comparatively higherthan were those in rice roots at corresponding stages. Cytochromec oxidase in roots remained active throughout the lives of therice and wheat crops. In rice roots, catalase seemed to playa distinct role around the panicle formation stage. Decay ofcatalase activity took place earlier than did that of peroxidaseand cytochrome c oxidase activities. In wheat roots similarenzyme activity changes were not observed. Changes in the metabolic activities of peroxide-producing systemsand peroxide-scavenging systems after freezing and thawing inflower buds of the apple, Malus pumila Mill., were studied withspecial reference to freezing injury. In flower buds of the‘McIntosh’ apple that were frozen below lethal temperatures,the activity of NADH-Cyt c reductase (EC 1.6.99.3 [EC] ), one of theenzymes in the electron-transport chains that are related tothe peroxide-producing systems, decreased slightly, while thatof Cyt c oxidase (EC 1.9.3.1 [EC] ) hardly changed. By contrast, theactivities of glucose-6-phosphate dehydrogenase (EC 1.1.1.49 [EC] ),dehydroascorbate reductase (EC 1.8.5.1 [EC] ) and ascorbate peroxidase(EC 1.11.1.11 [EC] ), which are involved in the peroxide-scavengingsystems, decreased to very low levels. The activity of glyceraldehyde-3-phosphatedehydrogenase (EC 1.2.1.12 [EC] ) also decreased markedly. However,little change was observed in the activities of hexokinase (EC2.7.1.1 [EC] ), glucosephosphate isomerase (EC 5.3.1.9 [EC] ), glutathionereductase (EC 1.6.4.2 [EC] ) and glutathione peroxidase (EC 1.11.1.9 [EC] ).Examination of substrates involved in the peroxide-scavengingsystems revealed that the levels of glucose-6-phosphate andfructoses-phosphate decreased to approximately 10–4 to10–5 M and 10–5 M, respectively, and the levelsof GSH decreased to about 10–5 M or became barely detectable.A decrease in the levels of GSSG also occurred while levelsof ascorbate rose slightly. Similar results were observed withflower buds from ‘Starking Delicious’ and ‘Jonathan’apple trees. These results suggest that the freezing injury to apple flower-budsis closely related to the collapse of the peroxide-scavengingsystems that are coupled with the pentose phosphate cycle. Theresults also suggest that the dysfunction of these peroxide-scavengingsystems is caused by H2O2, which may be produced during freezingand thawing. A sulfite-dependent ATPase [EC 3.6.1.3 [EC] ] of Thiobacillus thiooxidanswas activated and solubilized by treatment with trypsin [EC3.4.4.4 [EC] ], and purified 84-fold with a 32% recovery. It requiredboth Mg2+ and SO32– for full activity, and its optimumpH was found at 7.5–8.0. Mn2+, Co2+, and Ca2+ could partiallysubstitute for Mg2+, while SeO32– and CrO42– couldpartially substitute for SO32–. The enzyme hydrolyzed ATP and deoxy-ATP most rapidly and otherphosphate esters were poorer substrates. The apparent Km valuefor ATP was 0.33 mM. The fat-storing endosperm of Ricinus communis L. was found tocontain an ascorbate peroxidase (EC 1.11.1.11 [EC] ), which is nearlyas active as catalase (EC 1.11.1.6 [EC] ) in degradation of hydrogenperoxide (H2O2) at its physiological concentrations. This ascorbateperoxidase probably functions together with monodehydroascorbatereductase (EC 1.6.5.4 [EC] ) or dehydroascorbate reductase (EC 1.8.5.1 [EC] )and glutathione reductase (EC 1.6.4.2 [EC] ) to remove the H2O2 producedduring the transformation of fat to carbohydrate in the glyoxysomes.The activities of these enzymes as well as the content of ascorbateand glutathione increase parallel to the activities of glyoxysomalmarker enzymes during the course of germination. Inhibitionof catalase by aminotriazole results in increases of the ascorbateperoxidase activity and of the glutathione content. All fourenzymes are predominantly localized in the cytosol of the Ricinusendosperm with low activities found in the plastids and themitochondria. The results suggest, that the ascorbate-dependentH2O2 scavenging pathway, which has been shown to be responsiblefor the reduction of photosynthetically derived H2O2 in thechloroplasts, operates also in the Ricinus endosperm. Lysates of chloroplasts isolated from wheat (Triticum aestivumL. cv. Aoba) leaves were incubated on ice (pH 5.7) for 0 to60 min in light (15 µmol quanta m–2 s–1),and degradation of the large subunit (LSU) of ribulose-l,5-bis-phosphatecarboxylase/oxygenase (Rubisco: EC 4.1.1.39 [EC] ) was analyzed byapplying immunoblotting with site-specific antibodies againstthe N-terminal, internal, and C-terminal amino acid sequencesof the LSU of wheat Rubisco. The most dominant product of thebreakdown of the LSU and that which was first to appear wasan apparent molecular mass of 37-kDa fragment containing theN-terminal region of the LSU. A 16-kDa fragment containing theC-terminal region of the LSU was concomitantly seen. This fragmentationof the LSU was inhibited in the presence of EDTA or 1,10-phenanthroline.The addition of active oxygen scavengers, catalase (for H2O2)and n-propyl gallate (for hydroxyl radical) to the lysates alsoinhibited the fragmentation. When the purified Rubisco fromwheat leaves was exposed to a hydroxyl radical-generating systemcomprising H2O2, FeSO4 and ascorbic acid, the LSU was degradedin the same manner as observed in the chloroplast lysates. Theresults suggest that the large subunit of Rubisco was directlydegraded to the 37-kDa fragment containing the N-terminal regionand the 16-kDa fragment containing the C-terminal region ofthe LSU by active oxygen, probably the hydroxyl radical, generatedin the lysates of chloroplasts. N-Acetyl-D-[2-3H]glucosamine was synthesized from N-acetyl-D-mannosamineby alkaline 2-epimerization in pyridine containing 3H2O andnickelous acetate. The reaction involves reversible formationof an enol intermediate and therefore also resulted in incorporationof tritium into N-acetylmannosamine. After completed reaction,the two N-acetylhexosamines were separated from other radioactiveproducts and Morgan-Elson chromogens by chromatography on acolumn of Sephadex G-10, which was eluted with 10% ethanol,and were then separated from each other by chromatography onSephadex G-15 in 0·27 M sodium borate (pH 7·8).The location of the incorporated tritium was established bytreatment of the N-acetylhexosamines with borate under the conditionsof the Morgan-Elson reaction, which converts the sugars to Kuhn'schromogen I with concomitant loss of the C-2 hydrogen. As expected,this treatment resulted in the formation of 3H2O, indicatingthat the tritium was located at C-2. [2-3H]Glucosamine was preparedby acid hydrolysis of the labelled N-acetylglucosamine and wasconverted to [2-3H]glucosamine 6-phosphate by incubation withhexokinase and ATP. The sugar phosphate was used as a substratefor glucosamine 6-phosphate deaminase (isomerase, EC 5.3.1.10 [EC] )in a simple 3H2O release assay. A 50-year-old Pinus sylvestris L. stand was exposed for 2 yearsto low concentrations of SO2 and NO2 in an open-air exposureexperiment in northern Sweden. The mean SO2 concentrations inthe centre of the exposed plot during the 1988 exposure from14 June to 25 September, and during the 1989 exposure from 6June to 30 September were 15 nl 1–1 and 12 nl 1–1,respectively. The corresponding values for NO2 were 15 nl 1–1and 10 nl 1–1, respectively. The concentration in thecontrol plot was never higher than a few ppb, and mostly below1 nl 1–1. Needles sampled from the SO2 and NO2-exposed area showed reducedactivities of glutathione reductase (GR; EC 1.6.4.2 [EC] ) and superoxidedismutase (SOD; EC 1.15.1.1 [EC] ) compared with controls. The GRactivity showed decreased levels in autumn and winter, whilethe exposure had ceased, and SOD showed decreased activity duringthe second summer of exposure. Neither membrane-bound nor water-solubleanti-oxidants such as -tocopherol, carotenoids or glutathionechanged due to the exposure. The sulphur/nitrogen ratio wasincreased in needles that were exposed to SO2 and NO2 implyinga changed nutrition balance. The results suggest that the capacityof SOD and GR in the ascorbate-glutathione pathway was reduceddue to the exposure to air pollutants. The effect of Ca2+ and ammonia on mitochondrial NADH-glutamatedehydrogenase (GDH: EC 1.4.1.2 [EC] ) isolated from turnip root (Brassicarapa L.) activity was examined. Increasing the ammonia [(NH4)2SO4]concentration led to significant substrate inhibition whichcould be reversed by micromolar levels of Ca2+. The sensitivityof the enzyme to ammonia inhibition and its reversal by Ca2+was affected by proteolysis. After treatment with various proteases,lower concentrations of Ca2+ were capable of fully activatingthe enzyme or overcoming the inhibitory effects of high ammonium,compared to non-treated enzyme. However, the protease-treatedenzyme was still sensitive to ethylene glycol-bis(ß-aminoethylether) N,N,N',N'-tetraacetate (EGTA). In contrast, NADH-GDHactivity was inhibited approx. 30% by organic mercurials (200µm), but the residual activity was not affected by thesubsequent additions of EGTA. NADH-GDH activity could also bestimulated by additions of high concentrations of NaCl (300mM) in the absence of added Ca2+. At concentrations of CO2 less than saturating, carbonic anhydrase(EC 4.2.1.1 [EC] ) stimulates the carboxylation of ribulose bisphosphatecatalysed by ribulose bisphosphale carboxylase (EC 4.1.1.3 [EC] .9)in vitro. This is not through any beneficial association ofthe two enzymes but is a consequence of the increased rate ofconversion of HCO3 ion to CO2, the substrate for thecarboxylation. Carbonic anhydrase should always be includedin reaction mixtures used to determine the Michaelis constantof ribulose bisphosphate carboxylase for CO2 where fixationof radioactive CO2 into phosphoglycerate is the basis of rateestimation. The effect is to decrease the value obtained forthe Michaelis constant.  相似文献

Methionine sulfoximine (MSO) greatly reduced the carbon dioxideexchange rate (CER) of detached wheat (Triticum aestivvm L.cv Roland) leaves in 21% O2, but only slightly reduced it in2% O2. A supply of 50 mM NH4Cl had little effect on the CERirrespective of the O2 concentration. A simultaneous additionof glutamine and MSO protected against the inhibition of photosynthesisto a considerable extent and caused the accumulation of moreNH3 than did the addition of MSO alone. Fixation of 14CO2 in wheat leaves was inhibited by MSO treatmentin 22% O2, and there was decreased incorporation of 14G intoamino acids and sugars and increased label into acid fractions.The addition of MSO and glutamine together eliminated the effectof MSO on the photosynthetic 14CO2 fixation pattern. NH4Cl stimulatedthe synthesis of amino acids from 14CO2, especially the synthesisof serine in 22% O2. The charophyte Lamprothamnium papulosum (Wallr.) J. Gr. is foundat salinities varying from nearly fresh water to twice thatof sea water. It can maintain its turgor constant at 302 mosmolkg–1 (0.73 MPa) when exposed to external osmotic pressuresof 550 to 1350 mosmol kg–1 (1.3–3.3 MPa). Turgorshows a tendency to rise slightly at lower osmotic pressure(388 mosmol kg–1 of turgor at 150 mosmol kg–1 externalosmolality). K+ and Cl are the main solutes in the vacuole,and are most important in controlling internal osmotic pressure.Mg2+, Ca2+, and SO2–4 are present in significant amountsbut their concentrations do not change with changes in externalsalinity. Na+ is present in lower concentration than K+, andplays a minor role in regulating turgor. Sucrose is presentin significant concentrations, but changes little with changesin salinity. Two enzymes involved in sucrose metabolism, sucrosephosphate synthetase (EC 2.4.1.14 [EC] ), and sucrose synthetase (EC2.4.1.13 [EC] ) are active in whole cell extracts of Lamprothamnium.As in the fresh water charophytes, Lamprothamnium membrane potentialmay be depolarized (close to EK) or hyperpolarized, and presumablyof electrogenic origin. Carbonic anhydrase (CA, EC. 4.2.1.1 [EC] ) activity in air-grown Characorallina was detected mainly in the intracellular fraction,most of which composed of chloroplasts and cytoplasmic gel,and not on the cell surface. Only minor levels of CA activity,on the basis of equivalent volumes, were detected in the cellsap and the cytoplasmic sol. The maximum rate of photosynthetic O2 evolution by air-grownChara corallina at pH 6.0 was twice that at pH 7.6, while theapparent Km for external inorganic carbon (Ci) at pH 7.6 wasabout three times that at pH 6.0. However, the apparent Km(CO2)was about three times larger at pH 6.0 than at pH 7.6. The Km(Ci)-valueat pH 7.6 increased severalfold in the presence of acetazolamide(AZA), an inhibitor of CA, but no inhibition was observed atpH 6.0. The pH-dependence may be due to differences in the permeabilityof AZA at the given pH values. Fixation of 14CO2 at 20 µMand of H14CO3 at 200 µM over the course of 5 swas very similar at pH 7.4. Addition of CA significantly suppressedthe photosynthetic 14CO2-fixation but it stimulated the H14CO3-fixation.This result indicates that free CO2 is an active species ofCi that is incorporated into the cell during photosynthesis. The effect of the intracellular concentration of ATP ([ATP]1)on the light-induced potential change (LPC) in tonoplast-freeChara cells was studied. The LPC was hardly affected by loweringthe [ATP]1 by about 1/10 or by raising it to about 10 timesthe normal cytoplasmic concentration (0.5–1.3 mM). Theinsensitivity of LPC to [ATP]1 excludes the possibility thatan increase in [ATP]1 due to photosynthesis may induce the LPC.However, extreme lowering of the [ATP]1 to about 1–2 µMcompletely inhibited LPC, although photosynthetic O2 evolutionwas not significantly inhibited. This fact supports the hypothesisthat light stimulates the putative H+pump fueled by ATP. Theuncoupling agents DNP and CCCP greatly depolarized the membrane,and inhibited LPC strongly, but they did not decrease [ATP]1.Photosynthetic O2 evolution was inhibited to some extent by2 µM CCCP and strongly inhibited by 0.1 mM DNP. Sincethe membrane resistance increased significantly, these chemicalsare believed to act on the membrane as an inhibitor of the electrogenicH+ pump not as an H+conductor. The results of our previous study [Sawada et al. (1989) PlantCell Physiol. 30: 691] implied that, under sink-limited conditions,a decrease in the activity of ribulose-l,5-bisphosphate carboxylase(EC 4.1.1.39 [EC] ) caused a reduction in the rate of photosyntheticfixation of CO2 in single-rooted leaves of soybean (Glycinemax L. Merr. cv. Tsurunoko). This reduction in the rate of photosynthesisin source leaves seemed to correspond to a decrease in the demandby sink tissues for photoassimilates. In the present study,the activity of RuBPcase in vivo was estimated by measuringthe "initial" activity immediately after extraction from standardleaves (grown under a regime of 10 h of light and 14 h of darkness)and from sink-limited leaves (exposed for 6 or 7 d to continuouslight to alter the source/sink balance). The rate of photosynthesisin the sink-limited leaves decreased to 47% of that in the standardleaves. The "initial" activity of RuBPcase was 4.3 in the standardleaves but only 1.6 µmole CO2.(mg Chl)–1.min–1in the sink-limited leaves. These results appear to indicatethat the reduction in photosynthetic activity under sink-limitedconditions was mostly due to a deactivation of RuBPcase. Theactivity of deactivated RuBPcase in the sink-limited leaveswas restored to 4.1 µmole CO2.(mg Chl)–1.min–1by incubation of the enzyme in a medium that contained onlyNa2HPO4. This result suggests that free Pi in chloro-plastsplays an important role in the activation of the enzyme. Thelevel of Pi in the sink-limited leaves was 62% of that in thestandard leaves. On the basis of these various results, it appearsthat the deactivation of RuBPcase in the sink-limited leavesis the result of a decrease in the level of Pi. In chloroplasts isolated from SO2-fumigated leaves at 2.0 ppm,electron flow from water to 2,6-dichloroindophenol (DCIP) wasinhibited, but the electron flow from reduced DCIP to methylviologen was not affected. Neither diphenylcarbazide nor MnCl2could restore the activity of the DCIP-Hill reaction of SO2-injuredchloroplasts. Electron flows, from water to ferricyanide orto silicomolybdic acid, were inhibited in a degree similar tothat of the DCIP-Hill reaction. The rate of carotenoid photobleaching in the presence of carbonylcyanide-m-chlorophenylhydrazone was suppressed and paralleledthe inhibition of the DCIP-Hill reaction. In SO2-injured chloroplasts, the variable part of the fluorescencetransient was diminished, and the fluorescence yield loweredby SO2 was increased with 3-(3', 4'-dichlorophenyl)-l, l-dimethylurea(DCMU) or more pronouncedly by incubating the sample with sodiumdithionite. However, the yield could not recover to the levelfound in non-fumigated chloroplasts. With SO2 fumigation, thetime required to reach steady-state level of fluorescence becamelonger in the absence of DCMU, but was not altered in the presenceof DCMU. The pool size of the primary electron acceptors decreasedwith SO2 fumigation. We concluded that SO2 inactivated the primaryelectron donor or the reaction center itself. Photosynthetic and photorespiratoryenzymes in widely divergent plant species with special referenceto the moss Ceratodon purpureus: Properties of ribulose bisphosphatecarboxylase/oxygenase, phosphoenolpyruvate carboxylase and glycolateoxidase.—J. exp. Bot. 36: 1677–1684. Km(CO2) values and maximal velocities of ribulose bisphosphatecarboxylase/oxygenase (E.C. 4.1.1.39 [EC] ) were determined for sixplant species growing in the wild, consisting of a moss, a fernand four angiosperms. The maximum velocities of the RuBP carboxylasesvaried from 0.13 to 0.;62 µmol CO2 fixed min–1 mg–1soluble protein and the Km(CO2) values from 15 to 22 mmol m–3CO2. The highest Km(CO2) values found were for the moss, Ceratodonpurpureus, and the grass, Deschampsia flexuosa. These plantsalso had the highest ratios of the activities of RuBP carboxylaseto RuBP oxygenase. Glycolate oxidase (E.C. 1.1.3.1 [EC] ) activitieswere slightly lower in D.flexuosa, but not in C. purpureus,than for typical C3 species. Phosphoenolpyruvate carboxylase(E.C. 4.1.1.31 [EC] ) was not involved in the photosynthetic carboxylationby these two plants. However, another grass, Phragmites australis,was intermediate in PEP carboxylase activity between C3 andC4 plants The properties of RuBP carboxylase/oxygenase are discussedin relation to the activities of PEP carboxylase and glycolateoxidase and to the internal CO2 concentration. NADP malic enzyme (EC 1.1.1.40 [EC] ) from leaves of two C4 speciesof Cyperus (C. rotundus and C. brevifolius var leiolepis) exihibiteda low level of activity in an assay mixture that contained lowconcentrations of Cl. This low level of activity wasmarkedly enhanced by increases in the concentration of NaClup to 200 mM. Since the activity of NADP malic enzyme was inhibitedby Na2SO4 and stimulated by relatively high concentration ofTris-HCl (50–100 mM, pH 7–8), the activation ofthe enzyme by NaCl appears to be due to Cl. Variationsin the concentration of Mg2+ affected the KA (the concentrationof activator giving half-maximal activation) for Cl,which decreased from 500 mM to 80 mM with increasing concentrationsof Mg2+ from 0.5 mM to 7 mM. The Km for Mg2+ was decreased from7.7 mM to 1.3 mM with increases in the concentration of NaClfrom zero to 200 mM, although the increase of Vmax was not remarkable.NADP malic enzyme from Cyperus, being similar to that from otherC4 species, was able to utilize Mn2+. The Km for Mn2+ was 5mM, a value similar to that for Mg2+. The addition of 91 mMNaCl markedly decreased the Km for Mn2+ to 20 +M. NADP malicenzyme from Setaria glauca, which contains rather less Clthan other C4 species, was inactivated by concentrations ofNaCl above 20 mM, although slight activation of the enzyme wasobserved at low concentrations of NaCl at pH7.6.
