Using various photostationary state light sources to obtain reproducible phytochrome conversion of from 5 to 88% PFR, assayed by 2 wavelength in vivo spectrophotometry, relationships between initial percent PFR and elongation of apical Avena coleoptile segments over the succeeding 20 hours in darkness were studied. With material grown in total darkness, all PFR levels promote elongation, and maximal promotion requires roughly 50% PFR. The promotion caused by an initial 5 minute red (88% PFR) treatment at hour 0 is partially reversible at hour 5 by sources forming less than 48% PFR, but totally irreversible at hour 8, though less than 50% of the growth has been accomplished by this time. Direct photometric assays at hour 5 indicate a phytochrome state of roughly 45% PFR, consistent with the reversal data. At hour 8, however, 11 to 22% of the phytochrome still assays as PFR, an inconsistency suggesting simply that the elongation process has proceeded beyond photochemical control. Thus, in contrast with results previously reported for Pisum and Phaseolus, there is no contradiction between photometric and physiological assays of phytochrome state in Avena coleoptile segments.

Attempts to expand this study by using segments from seedlings pretreated with red light showed that such pretreatment as little as 1 to 2 hours before drastically reduces subsequent elongation and photoresponse on the medium employed. This decline in growth potential can be halted at any time before its completion by either excision of the segment or far-red treatment of the intact seedling.

The effects of 3-deoxygibberellin C (DGC) on the growth-promoting actions of gibberellins A1, A2, A3, A4, A5, A7, A8, A9, A13, A18, A19, A20, and A23 (GAn) as well as 13-deoxygibberellin A5 (deoxy-GA5) were tested with seedlings of gibberellin-deficient dwarf mutants (d2 and d5) of maize (Zea mays L.). It was found that DGC promoted the actions of gibberellins having both C-1 double bond and C-3 axial hydroxyl group, and it inhibited the action of gibberellins having the saturated ring A and lacking the C-3 axial hydroxyl group, whereas it did not affect that of the ones having the hydroxyl group. The presence of C-2 double bond, as in GA5 and deoxy-GA5, diminished the inhibitory action of DGC. The DGC inhibition was alleviated by raising the doses of the relevant GAs, suggesting that it is a competitive inhibition. These results and the finding that the growth of normal maize and rice seedlings are inhibited by DGC indicate that GA9, GA19, GA20 or other gibberellins having ring A of the same structure are involved in the growth of these plants as active form(s) or as intermediate(s) leading to the active form(s).  相似文献

Summary In henbit (Lamium amplexicaule L.) seeds, micromolar levels of gibberellic acid (GA3) introduced at the onset of imbibition markedly stimulated germination. Sensitivity to GA3 stimulation is rapidly lost if the seeds are pretreated in darkness at 15° for 1 day or more on a water substrate. Loss in responsiveness to GA3 is hastened by pretreatment in far-red or by dark pretreatment at higher (25°) temperatures and is reduced by pretreatments at 5°. Spectral characteristics of partially purified undegraded peaphytochrome were investigated in different ionic conditions.At the red-light-induced photostationary state in low ionicstrength buffer phytochrome had reduced absorbance in its far-redpeak as reported previously. Elevation of the ionic strengthof the buffer reversibly increased the absorbance in the far-redregion at the photostationary state. It was found that the effectof increase of ionic strength was strengthened secondarily bychaotropicity of salts. It was confirmed that phytochrome preparations of low ionicstrength contained a photosensitive component(s) other thanthe red-light-absorbing form (PR) and farred- light-absorbingform (PFR) during photochemical transformation, as well as duringthe first several min in the dark after phototransformation.At high ionic strength, phytochrome became a two-component systemcomposed of only PR and PFR at the redlight-induced photostationarystate though a significant accumulation of another component(s)occurred during phototransformations. Increasing ionic strengthalso enhanced A723 of phytochrome at the red-light-induced photostationarystate. Phytochrome was partially purified from etiolated seedlings of Avena sativa L. Several properties of the red-absorbing (PR) and far-red absorbing (PFR) forms of the pigment were compared. The 2 forms could not be shown to differ with respect to their sedimentation velocity in sucrose density gradients, elution volume from Sephadex G-200 columns, binding properties on calcium phosphate, or electrophoretic mobility. PFR, however, was more labile than PR during precipitation with 50% ammonium sulfate. Sephadex G-200 elution diagrams obtained with fresh phytochrome preparations revealed 2 components of different molecular weights, 1 roughly 180,000, and 1 roughly 80,000. Native phytochrome had an absorption spectrum in vivo showing an absorption maximum for PR of 667 nm. Both the large and small forms of phytochrome mentioned above can be maintained with an absorption maximum for PR of 667 nm. However, allowing them to remain for several hours as PFR, even at 4°, shifted this peak to 660 nm. The protein conformational change during phytochrome transformation may be quite small, though the various comparative techniques used do not strictly rule out a fairly large one. The need for maintaining the pigment as PR during all steps of purification, but particularly during ammonium sulfate precipitation is underscored.  相似文献

Endogenous gibberellins were analyzed from a parasitic plant, clover broomrape (Orobanche minor Smith), and its host, clover (Trifolium repens L.). Members of both the early-13- and the early-non-hydroxylation pathways were identified from both the parasite and the host (GA12, GA24, GA9 GA4, GA44, GA19, GA20, and GA1 from clover broomrape; GA9, GA4, GA44, GA19, GA20, and GA1 from clover). Quantitative analyses showed that GA44 was present at high levels in both host and parasite. The similarity in the gibberellins suggests the possibility that the major gibberellins in clover broomrape are transported from clover. However gibberellins such as GA58, GA38, and notably GA47 which was identified from a plant for the first time were detected only from clover broomrape, suggesting that the parasite may have the ability to produce at least those gibberellins  相似文献

The fern Asplenium nidus L. is in great demand as an ornamental plant. The aim of this work was to investigate the influence of phytohormones in promoting a gametophytic and sporophytic growth in homogenized sporophytes tissue. Exogenous application of 0.5 and 5 μM N 6-benzyladenine, 0.05 and 0.5 μM indole-3-acetic acid (IAA), and 0.3 and 3 μM gibberellic acid (GA3) favoured sporophyte regeneration, whereas gametophyte regeneration took place when plant material was cultured in a hormone-free liquid MS medium. The endogenous contents of the auxin IAA, the cytokinins trans-zeatin, trans-zeatin riboside, dihydrozeatin, dihydrozeatin riboside, isopentenyladenine and isopentenyladenosine, and the gibberellins GA1, GA3, GA4, GA7, GA9 and GA20 in growing gametophytes and sporophytes were evaluated. Similar levels of the auxin and cytokinins and qualitative differences in the gibberellins were found between both generations.  相似文献

Germination of Kalanchoé blossfeldiana seeds is absolutely light-requiring and needs repeated daily light periods. With increasing length of the photoperiod there was a gradual escape from the far-red inhibition. This escape depended also upon the duration of the far-red exposure: 10-second far-red caused a strong inhibition after a 10- to 30-minute photoperiod and did not inhibit after a 4-hour day, although the effect of the latter was completely suppressed by 5 minutes far-red. The action of a 12-hour photoperiod was not reversed by 10 minutes far-red but it was by 12 hours far-red. Light intensity and temperature during the photoperiod were two other important factors influencing the escape from far-red inhibition. The common features of this escape displayed in very different photomorphological responses are stressed. In order to explain our results in terms of phytochrome action, we distinguish two effects of white light: 1) on the initial photoconversion of the inactive to the active PFR form 2) on the much slower transformation of PFR to a reacted form P*FR; the latter reaction can also proceed in darkness, but is enhanced by light and is dependent upon light intensity and temperature; this reacted phytochrome is not reversible by a brief far-red illumination.  相似文献

Eight gibberellins (GAs) were identified from vegetative shoots of navel orange trees (Citrus sinensis L. Osbeck cv Washington) after sequential purification by reverse-phase C18 high performance liquid chromatography, Nucleosil 5N(CH3)2 high performance liquid chromatography, and capillary gas chromatography-mass spectrometry. GA1, GA17, GA19, GA20, GA29, and iso-GA3 were identified based on the full scan mass spectra and Kovats retention indices. GA8 was tentatively identified based on the comparison of the full scan mass spectra with the published spectra. GA44 was tentatively identified from the characteristic masses at the correct Kovats retention index.  相似文献

Summary The effect of gibberellins A1 through A9 on stem elongation and flower formation in five plants was tested. The plants wereMyosotis alpestris and a biennial strain ofCentaurium minus (cold-requiring plants),Silene armeria andCrepis parviflora (long-day plants), andBryophyllum crenatum (a long-short-day plant). The two former plants were maintained on non-inductive temperatures and long days, the three latter on short days, InMyosotis, flower formation was only obtained with GA7 and GA1, the latter being relatively less active. InCentaurium GA3 was the most effective, followed by GA1, GA4 and GA7 and perhaps GA5 and GA9. InSilene, flower formations was induced only by GA7. InCrepis, the most effective gibberellins were GA4 and GA7, inBryophyllum, GA3, GA4 and GA7. Thus, the different gibberellins exhibited considerable differences in their activity with respect to flower induction, and different plants exhibited in this respect certain specific differences in their sensitivity to the various gibberellins. Except inCrepis, flower initiation as a result of gibberellin treatment was always preceded by substantial stem or internode elongation; however, the correlation between the effect of the different gibberellins on stem elongation and flower induction was not in all cases complete. Dark germination of Amaranthus retroflexus L. seeds at 35° increased after several days of prechilling at 20° or lower. Irradiation with far-red light for short periods during the early hours of a prechilling period at 10° inhibited subsequent dark germination at 35°. The inhibition was completely reversible with red light. Far-red irradiation in the latter part of the prechilling period was less effective. Increased dark germination of A. retroflexus seeds following a prechilling period at 20° or less is attributed to action of preexistent PFR, the far-red absorbing form of phytochrome, within the seeds. Inactivation of PFR was found to proceed ca. 4 times more rapidly at 25° than at 20°. Failure of imbibition temperatures above 20° to increase dark germination of A. retroflexus seeds is attributed to the rapid thermal reversion of pre-existent PFR. We suggest that the action of prechilling (layering) on many other seed kinds arises in a similar way.  相似文献

Summary Action spectra were determined in the UV region of the spectrum for the first phase of the phototactic response (stop response) and for the phytochrome pigment associated with this response in the dinoflagellate Gyrodinium dorsum Kofoid. Differences between these action spectra indicate the participation of two pigments in phototaxis. Following R (620 nm) irradiation of the phytochrome, the stop response maxima occur at 470 and 280-nm; after FR irradiation they shift to 490 and 300–310 nm. These maxima suggest that the photoreceptor pigment for phototaxis is a carotenoprotein. The action spectrum shift following the different phytochrome conversions may represent a trans to cis isomer change by the carotenoid. The absorption maximum of PR in the UV appears to be at 320 nm, which is consistent with the shift of the R absorption maximum to shorter wavelengths (620 nm) as compared to higher plants. The PFR absorption maximum appears as a broad band between 360 and 390 nm. Comparison of PR to PFR conversions by different intensities of 620-nm and 320-nm light indicates that at lower intensities the logarithm of the threshold for the stop response is inversely proportional to the logarithm of the intensity of the sensitizing light. A comparison has been made of the relative effectiveness of light quality and quantity and gibberellic acid (GA3) treatment on the elongation growth of the coleoptile and the first foliage leaf in durum wheat (Triticum durum Desf. cvs. Cappelli and Creso). The cultivar Creso is a shortstrawed variety carrying the Gai 1 gene on chromosome 4A, which influences both plant height and insensitivity to applied gibberellins. The main conclusions are as follows: 1) coleoptile elongation growth appears to be modulated via the fluencerate-dependent action of a blue-light receptor and via a low energy response of phytochrome; 2) the inhibition of first-foliage-leaf growth depends on the operation of a single blue-light-responsive photoreceptor; 3) high energy blue light produces the same inhibitory effect on the two wheat cultivars, whereas at relatively low fluences of white and blue light, the cultivar Creso is more sensitive; 4) the insensitivity to applied GA3 exerted by the gene Gai 1 in Creso is independent of light; 5) in Cappelli, the action of light on coleoptiles appears to be independent of the applied GA3, whereas the hormone is able to change the pattern of growth inhibition of the first-foliage-leaf.Abbreviations BL blue light - FR far-red light - GA gibberellin - GA3 gibberellic acid - R red light - WL white light  相似文献

The radio-labeled gibberellins GA1, GA3,GA4, and GA7 were applied to intact developing applefruits (Malus domestica Borkh. cv. Jonagold) during theperiod when GAs are suggested to inhibit flower bud induction for the followingyear. Radioactivity from these compounds was found to be transported intoadjacent tissues as there are pedicels and bourses (4%). Application topedicels, after removal of the fruits, enhanced the transport into adjacentbourses up to 11%. The bud-carrying lateral bourse shoots contained onlyminor amounts of radioactivity on average 0.4% in both cases. Theseexport rates were identical, 1 or 5 days after application.After application of the corresponding deuterium-labeled GAs and analyses bymass spectrometry the specific metabolization of GA1 toGA1 13-O-glucoside and of GA3 to GA313-O-glucoside was demonstrated. Additional metabolites of GA1 andGA3 were not detected. After fruit application of GA3 theratio of GA3 to GA3 13-O-glucoside was found to be 1:2 inthe fruit. Pedicel application led to ratios of 1:4 and 1:5, respectively, inthe pedicel and in the adjacent bourse. After the application of GA4and GA7, neither glucosylation products nor other GA-like metabolitescould be identified.This is the first report of the metabolism of GAs to GA 13-O-glucosides indeveloping apple fruits. The possible function of the GAs as a signal in flowerbud formation for the following year is discussed.  相似文献

Four novel gibberellins GA30, GA31, GA33, GA34, five known gibberellins GA8, GA17 (free acid and its monomethyl ester), GA19, GA27, GA29 and the gibberellin-like substances were isolated from immature seeds of evening-glory (Calonyction aculeatum). Summary The rate of total phytochrome decay in the dicotyledons Amaranthus caudatus, Mirabilis jalapa and Pisum sativum under continuous illumination with red, incandescent, and blue light depends on the PFR/Ptotal maintained by each source. Amaranthus is an exception to this in that there is a deviation from firstorder decay kinetics under continuous illumination with incancdescent light. This deviation is probably not related to the chlorophyll present in the Amaranthus sample since chlorophyll-rich Pisum buds have the same phytochrome decay rate as epicotyl tissue under continuous incandescent light. Reports of a prolonged lag phase before the onset of first-order decay kinetics of phytochrome in Pisum have not been confirmed and the small lag phase observed in the present work can be accounted for by the time required to attain the PFR/Ptotal ratio characteristic of blue light in a carotenoid rich tissue. In the monocotyledon, Avena sativa, and perhaps monocotyledons in general, decay rate is maximal at a low PFR concentration and the decay curve is the same under continuous red, incandescent and blue light. This dicotyledon/monocotyledon difference with respect to saturation of phytochrome decay does not correlate with the other dicotyledon/monocotyledon difference, the presence or absence of dark reverions of PFR to PR, since the dicotyledons Amaranthus and Mirabilis that lack reversion still show no saturation of decay. Possible growth control by the PFR/Ptotal ratio is discussed in relation to environmental changes in light quality.Research carried out at Brookhaven National Laboratory under the auspices of the U. The action of prolonged far-red on seed germination was studied in Lactuca sativa L. var. Grand Rapids. Exposure of imbibed seeds to 6 hours far-red before the application of gibberellic acid (GA3) and thiourea completely prevented germination. Using GA3, this far-red was effective after the sixth hour of imbibition. At 6, 12, and 18 hours of imbibition equal durations of far-red had equal effects. The kinetics of far-red action was investigated: it was found that although far-red for several hours, irrespective of the energy level, was needed for maximum inhibition, shorter durations (15 and 30 mins) were also appreciably effective provided they were followed by several hours darkness before the supply of GA3. This is taken to indicate the existence of labile product(s) of the action of a far-red sensitive pigment. Evidence is provided for the existence of promotive dark processes controlled by this pigment, which are essential for germination whether triggered by GA3, thiourea or red-light. A model for the operation of the pigment system is proposed and its role in the germination mechanism of this seed is discussed.  相似文献

The germination of Empress tree (Paulownia tomentosa Steud.) seeds is phytochrome-controlled. Liquid smoke could not induce germination in darkness but red light irradiation of liquid smoke imbibed seeds induced a high percentage of germination. Maximum germination was achieved at liquid smoke concentration of 0.1% (v/v) when present during the imbibition phase or during the phase of phytochrome activity. The light requirement of these seeds could be completely substituted by exogenously applied gibberellins. In the presence of liquid smoke, optimal concentrations of GA3, GA4, and GA9 necessary for inducing germination were several times lower than in the controls, while that of GA7 was equally active when applied at a concentration one order of magnitude lower. The inhibitory effect of the applied growth retardants was strongly reduced and liquid smoke, in the presence of retardants, allowed light-induced germination, if applied simultaneously or after retardants treatment.  相似文献

The effects of various chemically pure gibberellins and cytokinins on leaf yellowing of Alstroemeria were described. The loss of chlorophyll was measured both in leaves of cut flowering stems and in a model system consisting of detached leaf tips. It was demonstrated that plant growth substances affected chlorophyll loss in both systems to the same extent. Leaf senescence was delayed by various gibberellins and cytokinins. The results demonstrated that some of the gibberellins (GA4 and GA7) are far more effective in delaying chlorophyll loss than GA3, which is commonly used as a postharvest treatment for Alstroemeria cut flowering stems. This work showed for the first time the relationship between the effect of exogenous auxins and gibberellins on apogamy in Dryopteris affinis (Lowe) Fraser-Jenkins sp. affinis and its endogenous contents during early apogamic events. The addition of NAA (0.53 and 5.37 μM) or GA3 2.8 μM to an MS solid medium significantly increased apogamous sporophyte formation. BA induced brown callus that regenerated sporophytes in a hormone-free medium. The endogenous contents of GA1, GA3, GA4, GA7, GA9 and IAA were determined by GC–MS in gametophytes cultured on MS solid medium, before and during early stages of apogamous embryo development. The accumulation of both GA9 and IAA before embryo development was evident as high levels of GA4 in the earliest analysed stage of embryo development and high levels of GA3 in elongating shoots were found. The role of gibberellins on apogamy was also supported by data showing a decrease in the percentage of gametophytes developing embryos because of the addition of flurprimidol to the culture medium.  相似文献
