According to the automatic tracing of the movement of ticks in a Petri dish, motivational variability of the spontaneous activity and behavioral response of the taiga tick to olfactory stimuli was analyzed. In the studied sample, two groups of ticks that differ in the movement trajectory in the absence of stimulus were isolated, including ticks that mainly moved on the edge of the dish at maximum accessible height (group 1) and ticks that mainly moved at the bottom and wall of the dish (group 2). It was registered that ticks of group 1 (as opposed to ticks of group 2) demonstrated a pronounced behavioral response to olfactory stimuli (human synthetic pheromones and ammonia) and negative geotaxis. It was established that belonging to these groups depended on the time of day when the testing was performed and did not depend on the physiological age and infectious status.  相似文献

The CD147 receptor is a cell-surface glycoprotein in the IgG family that plays pivotal roles in intercellular interactions involved with numerous physiological and pathological processes such as extracellular matrix remodeling. We previously found an Amblyomma americanum (Aam) tick CD147 receptor homolog among genes that were up regulated in response to tick feeding stimuli. This study characterizes an AamCD147 receptor protein that is 72–83% conserved in other tick species and possess characteristic CD147 receptor sequence features: an extracellular (EC) region containing two IgG domains, a transmembrane and the cytoplasmic domains. Likewise, the AamCD147 EC domain folds into secondary structures that are consistent to the human homolog: an amino-terminus β-barrel that is linked to 2-carboxy-terminus β-sheets with consensus disulfide bonds conserved in each of the 2 domains. CD147 receptor signaling and regulatory mechanisms are putatively conserved in ticks as revealed by in silico analysis that show presence in the tick genome of CD147 receptor signaling protein homologs, cyclophilin (CyP) A and B, and chaperones that transport it to the plasma membrane, caveolin-1 and CyP60. The AamCD147 receptor has a dichotomous expression pattern of where it is up regulated in response to feeding in the salivary gland but remains constant at the midgut and ovary levels suggesting that it may regulate different functions in different tick organs. We speculate that biological functions of the AamCD147 receptor are essential to tick feeding success as revealed by RNAi-mediated silencing that caused ticks to obtain smaller blood meals, of which ~69% were below threshold to trigger spontaneous detachment of ticks from the host. These ticks showed unusual cuticle tenderness and assumed a reddish coloration, a phenomenon that has been attributed to tick midgut damage allowing red blood cells to leak into tick hemolymph. On the basis of the CD147 receptor being linked to tissue growth regulation in mammals, we speculate that silencing of the AamCD147 receptor blocked progression of the tick intermolt growth, a process that precedes tick engorgement and their spontaneous detachment of from the host to end feeding. The results are discussed in context of advances in tick molecular physiology.  相似文献

Spores of microsporidia have been recovered in 5 specimens of 13 ixodid ticks Ixodes ricinus from various populations of the Republic Moldova collected in spring of 2004. Microsporidia were detected by means of fluorescent microscopy. Intensity of infection was 3-6 spores per a micropreparate from one mite. Based on spore size, character of staining and the presence of diplocarion, these spores are referred to the Nosema-like type. Low intensity of infection probably is caused by that fact that ticks were collected in spring period and were unfed.  相似文献

When the cattle tick Boophilus microplus, after dropping from its host, was maintained at 27 C, the digestion of hemoglobins in the gut proceeded at a steady rate and was virtually complete by the 13th day. The rate was essentially the same whether the ticks were strains susceptible (Yeerongpilly) or resistant (Biarra) to organophosphorous compounds or whether the hosts were British breed, Brahman or banteng cattle, or buffalo. Ferrihemoglobin appeared in the gut contents and hematin was deposited. About 10% of the hematin released from the hemoglobin was transferred to the eggs and from them to the larvae. Translucent ticks, sometimes found on heavily infested hosts and referred to as “serum” ticks, had about half the normal hemoglobin content.  相似文献

THE MIGRATION OF LYMPHOCYTES ACROSS SPECIALIZED VASCULAR ENDOTHELIUM   总被引:1,自引:0,他引:1  
Lymphocytes were exposed in vitro to either trypsin or neuraminidase. The ability of the treated cells to migrate into tissues was measured (a) by i.v. injection into intact recipients and (b) by vascular perfusion through an isolated lymph-node preparation. The localization of trypsinized cells in the lymph-nodes of recipients was deficient when compared to untreated lymphocytes and there was a surplus of trypsinized cells in the blood. Trypsinized cells migrated into the isolated nodes in reduced numbers. By contrast, neuraminidase treated lymphocytes were markedly deficient in the blood of recipients early after injection; their localization in the spleen and lymph-nodes was also deficient but they were in surplus in the liver. Moreover they migrated into the isolated nodes in slightly increased numbers. By 24 hr after injection the perturbed localization pattern produced by either enzyme was partly restored to normal. In conclusion, trypsin interfered with the capacity of lymphocytes to migrate into lymph-nodes but neuraminidase did not; the latter promoted the hepatic sequestration of cells and the reduced localization in the blood and tissues was a consequence of this. The hypothesis that lymphocytes adhere to specialized endothelia in lymph-nodes because of specific glycoside sequences on their surface lacks experimental support.  相似文献

Histological features of Rhipicephalus sanguineus ticks fed on dog, a non resistant host, and on guinea pig, a resistant host, were compared. Unfed ticks and ticks from each host species were collected during first and third infestation and processed for histology. Many ticks from guinea pigs, especially during third infestation, were unattached, dehydrated and small. Only the midgut of ticks fed on guinea pigs had host leukocytes. Vacuolization of midgut cells was observed in all ticks, with exception of those fed on dogs for more than 96 h. Ticks of guinea pigs, particularly from third infestation, had vacuolated tracheae and swelling of malpighian tubules. Solely ticks from third infestation of guinea pigs displayed vacuolization of oocytes. Ticks fed on guinea pigs also had an increased number of guanine spherules. Observed alterations in ticks from guinea pigs are discussed.  相似文献

Some features of three-host exophilic ticks important for their survival differ at the generic level in relation to the types of their habitat. Ixodes and Haemaphysalis are typical forest genera, Hyalomma is an open country genus, whereas Dermacentor and Rhipicephalus, having some features of forest ticks, are closer to the ticks of open country. Forest ticks encounter rather stable and favorable temperature, humidity and illumination conditions as compared with open country ticks. A few differences determining every-day survival, host-seeking, and reproduction of ticks are considered. (1) Tolerance to desiccation is very low in forest ticks and much greater in ticks of open country. (2) Lack of eyes in forest ticks (having, however, epithelial photoreceptor cells) and formation of eyes as special morphological structures in ticks of open country. (3) Capability of mating and insemination of unfed specimens both on and off hosts in forest ticks (Ixodes) and insemination of partially engorged females only on hosts by fed males in open country ticks. (4) Reciprocal sexual dimorphism where in forest ticks females have a larger body than males and in open country ticks this correlation is opposite. (5) Linear dependence between female weight and number of eggs laid is established in forest ticks after the female weight becomes higher than 50% of mean engorgement weight whereas in open country ticks linear dependence begins to reveal itself much earlier. (6) In forest ticks the compensatory growth occurs only during adult feeding whereas in open country ticks such a growth occurs both during nymphal and adult feeding. Three-way PCA has been applied to proteomic pattern images to identify the classes of samples present in the dataset. The developed method has been applied to two different datasets: a rat sera dataset, constituted by five samples of healthy Wistar rat sera and five samples of nicotine-treated Wistar rat sera; a human lymph-node dataset constituted by four healthy lymph-nodes and four lymph-nodes affected by a non-Hodgkin's lymphoma. The method proved to be successful in the identification of the classes of samples present in both of the groups of 2D-PAGE images, and it allowed us to identify the regions of the two-dimensional maps responsible for the differences occurring between the classes for both rat sera and human lymph-nodes datasets.  相似文献

In the present study, PCR has been applied to detect and analyze DNA of Babesia spp. extracted from Ixodes ricinus ticks. Collection of I. ricinus was made in 6 forested areas of Zachodniopomorskie Voivodship, Poland, during 2 seasonal peaks of tick activity, i.e., spring and autumn, 2001. In total, 1,328 I. ricinus were collected and processed for PCR with F34 and R323 primers. Babesia spp. was detected in 28 (2% of 1,328 tested) ticks; 26 were identified as B. divergens. The other 2 were identified as B. microti. PCR was conducted with 18S rRNA specific primers and sequencing was processed to precisely identify and compare these isolates with B. microti and B. divergens sequences from Europe, North America, and Asia obtained from the GenBank. Analysis revealed that sequences of B. microti from northwestern Poland are almost identical (99.94%) with those referred to as "Munich strain"; both form a clade different from other European strains, as well as those from Asia and North America (called B. microti, sensu stricto). An investigation performed with B. divergens sequences showed that the sequence from northwestern Poland is 99.94% homologous to an isolate from Ireland ("Purnel"), and differs in just a few nucleotides from other European sequences. Phylogenetic analysis revealed that the sequence of B divergens isolated from Polish ticks form a group that comprise 4 European sequences from Great Britain and Ireland and is, therefore, closely related to other European and North American B. divergnens sequences.  相似文献

Idiosyncratic markers are features of genes and genomes that are so unusual that it is unlikely that they evolved more than once in a lineage of organisms. Here we explore further the potential of idiosyncratic markers and changes to typically conserved tRNA sequences for phylogenetic inference. Hard ticks were chosen as the model group because their phylogeny has been studied extensively. Fifty-eight candidate markers from hard ticks (family Ixodidae) and 22 markers from the subfamily Rhipicephalinae sensu lato were mapped onto phylogenies of these groups. Two of the most interesting markers, features of the secondary structure of two different tRNAs, gave strong support to the hypothesis that species of the Prostriata (Ixodes spp.) are monophyletic. Previous analyses of genes and morphology did not strongly support this relationship, instead suggesting that the Prostriata is paraphyletic with respect to the Metastriata (the rest of the hard ticks). Parallel or convergent evolution was not found in the arrangements of mitochondrial genes in ticks nor were there any reversals to the ancestral arthropod character state. Many of the markers identified were phylogenetically informative, whereas others should be informative with study of additional taxa. Idiosyncratic markers and changes to typically conserved nucleotides in tRNAs that are phylogenetically informative were common in this data set, and thus these types of markers might be found in other organisms.  相似文献

In recent years, in urban areas of Novi Sad, unique ecological conditions, specific floristic and faunistic composition and poor habits of citizens in sense of public health, facilitate the development and maintenance of ticks. Regarding the importance of ticks as vectors of severe human and animal diseases, complex and detailed studies are conducted with an aim to find the most efficient methods for tick control. Two tick species, Ixodes ricinus and Dermacentor marginatus, were identified during a 3-year period on the territory of Municipality of Novi Sad. During 2006, the efficacy of the pyrethroid lambda-cyhalothrin in tick control varied from 60.7 to 100%. The highest efficacy recorded in 2007 was 92.3%. The efficacy of lambda-cyhalothrin in 2008 varied from 39.1 to 100%. Lambda-cyhalothrin showed high efficacy in tick control at localities which were improved before the application (mowed, litter removed, abundance control and euthanasia of abandoned cats and dogs). The results of this research indicate that lambda-cyhalothrin has a toxic effect on ticks and could be used as efficient acaricide for tick control, although its efficacy depends on formulation, terrain features and methods of application.  相似文献

In cardiac myocytes, calcium (Ca) can be released from the sarcoplasmic reticulum independently of Ca influx from voltage-dependent membrane channels. This efflux of Ca, referred to as spontaneous Ca release (SCR), is due to Ryanodine receptor fluctuations, which can induce spontaneous Ca sparks, which propagate to form Ca waves. This release of Ca can then induce delayed after-depolarizations (DADs), which can lead to arrhythmogenic-triggered activity in the heart. However, despite its importance, to date there is no mathematical model of SCR that accounts for experimentally observed features of subcellular Ca. In this article, we present an experimentally based model of SCR that reproduces the timing distribution of spontaneous Ca sparks and key features of the propagation of Ca waves emanating from these spontaneous sparks. We have coupled this model to an ionic model for the rabbit ventricular action potential to simulate SCR within several thousand cells in cardiac tissue. We implement this model to study the formation of an ectopic beat on a cable of cells that exhibit SCR-induced DADs.  相似文献

A rapid method is described for preparing and staining salivary glands of Rhipicephalus appendiculatus ticks infected with Theileria parva. The technique, involving the use of a modified methyl green pyronin stained minimizes the risk of losing material and allows examination of stained glands within minutes of preparation. The technique was applied in a series of studies in which ticks were either infected with T. parva under different conditions, or maturation of parasites in adult ticks was stimulated by different means. When nymphal ticks were fed on the ears of cattle the subsequent infection rate of the adult ticks showed no correlation with the parasitaemia of the cattle at the time of nymphal engorgement. There was no difference in infection rates between adult ticks in which parasite maturation had been stimulated either by incubation at 37 degree C or by feeding on rabbits. However, parasite maturation took about 1 day longer in incubated ticks than in rabbit-fed ticks. Female ticks were consistently more highly infected than males, both in terms of the percentage of ticks infected and the mean number of infected acini/tick. Ticks were infected with T. parva by injection of nymphs with parasitaemic bovine blood, but the resultant adult infection was lower than that in ticks which had been infected naturally by feeding on cattle.  相似文献

Laboratory-reared and field-collected Amblyomma americanum ticks were hosts of a Coxiella sp. and a Rickettsia sp. While the Coxiella sp. was detected in 50 of 50 field-collected ticks, the Rickettsia sp. was absent from 32% of ticks. The Coxiella sp. showed evidence of a reduced genome and may be an obligate endosymbiont.  相似文献

Understanding the factors that influence patterns of ectoparasite infestation within wildlife populations involves knowledge of the mechanisms that influence host infestation. For ectoparasitic ticks, knowing where ticks might occur in the off-host environment and how they locate their hosts is essential to understanding patterns of ectoparasite infestation. The tuatara tick (Amblyomma sphenodonti) parasitises the tuatara (Sphenodon punctatus) on Stephens Island, New?Zealand. We completed a series of laboratory microcosm experiments to examine daily activity patterns, microhabitat preferences and host-seeking behaviour by Amblyomma sphenodonti. Firstly, to determine the diel activity pattern of ticks, we observed the behaviour of ticks every 2?h over a 48-h period. We then tested the preferences of ticks for soil moisture, soil texture and shade by offering different pairs of substrate conditions. Last, to determine what cues ticks used to locate their hosts, we tested the response of ticks to filter paper infused with host scent or excrement. Ticks were most active at night. They also showed a significant preference for moister, coarser and shaded substrates 12 h after the start of the experiment. Ticks did not show an immediate response to either of the two host stimuli, but after 12 h showed a significant preference for host scent and avoided host excreta. We suggest that the microhabitat preferences of ticks reflect conditions within host refuges (burrows), and that the delayed response to host odour suggests ticks could use host scent to identify substrates frequented by hosts.  相似文献

Tick-borne encephalitis is an important zoonosis in many parts of north-western, central and eastern Europe, Russia and the Far East, with considerable altitudinal and latitudinal shifts described during recent decades. The reported routes of transmission for TBE virus include the saliva-activated non-viraemic transmission between co-feeding ticks taking place on rodent hosts. During the period 2001–2014, a population of the yellow-necked mouse (Apodemus flavicollis), which is considered among the most efficient TBE competent host, especially in central and western Europe, was intensively live-trapped in a known TBE focus in the Province of Trento, Italy. Individual live-trapped mice were checked for the number and position of feeding ticks and serologically screened for TBEv antibodies. A combined effect of climatic conditions and density of both roe deer and mice on the number of co-feeding tick groups was observed. Specifically, the occurrence of co-feeding ticks on mice during the questing season was affected by autumnal cooling in the previous season. On the other hand, co-feeding occurrence was also positively associated with roe deer abundance, while mouse density showed a hump-shaped pattern. Individual features of A. flavicollis such as weight and sex also affected co-feeding occurrence with the heaviest (breeding adult) males carrying more co-feeding ticks. We also found that the overall number of co-feeding ticks on mice positively affected TBEv antibody detection in this species the following year. In conclusion, a specific combination of climatic conditions in conjunction with certain rodent and roe deer densities are the principal determinants of the number of co-feeding ticks on A. flavicollis and, consequently, TBEv circulation. These variables can be used to provide an early warning signal for a TBE hazard, thus representing a useful tool for Public Health authorities to prepare action for prevention and control within TBEv circulation areas.  相似文献

为了解发热伴血小板减少综合征布尼亚病毒(SFTSV)的传播机制,采集了山东疫区家养牛、羊和狗等动物体表蜱,分类鉴定后,通过Real-time PCR筛查、病毒分离培养和基因组序列分析等方法分离鉴定蜱中的病毒。所采集的蜱,以长角血蜱为主,占91.4%。其中3头SFTSV核酸检测阳性,阳性率为2.14%,并在其中一份羊体表蜱标本中分离到SFTSV病毒,命名为SDLZTick12。序列分析显示与我国在不同省份患者标本中分离的病毒全基因序列具有高度同源性,且病毒的抗原性和生长特性与人源病毒相同。本研究首次在山东疫区蜱中分离到新型布尼亚病毒,并与人源病毒进行了系统比较研究,提示蜱可能为该新病原体的传播媒介,对疾病的防控具有重要的指导意义。  相似文献

THE MIGRATION OF LYMPHOCYTES ACROSS SPECIALIZED VASCULAR ENDOTHELIUM   总被引:1,自引:0,他引:1  
The chain of lymph-nodes in the rat mesentery was isolated and the preparation was perfused via cannulae in the superior mesenteric vessels. The perfusate consisted of serum to which labelled lymphocytes had usually been added. The entry of radioactively labelled lymphocytes from the blood vessels into the lymph-nodes was studied by scintillation counting and autoradiography. It was observed that: (1) In the perfused node labelled lymphocytes localized in and around post-capillary venules in the paracortex as they do early after i.v. injection. (2) The number of lymphocytes which entered the node was directly proportional to the concentration in the perfusate over the range studied. The proportion of cells retained in the node varied considerably around a mean of 11 % of lymphocytes reaching it. (3) The isolated lymph-node released few if any lymphocytes into the effluent (venous) perfusate. (4) Large lymphocytes migrated into isolated lymph-nodes slightly more readily than did small lymphocytes. (5) Unlike intact cells isolated lymphocyte membranes did not adhere to specialized vascular endothelium.  相似文献

Ticks were collected from 8 houses in Mvumi Mission village, near Dodoma, Tanzania. All ticks were examined for Borrelia infestation by flagellin gene-based nested polymerase chain reaction. All houses were highly infested with ticks, and all ticks collected were of the Ornithodoros porcinus species. Fifty-one out of 120 ticks were infected with spirochetes, and a flagellin gene sequence comparison showed that most of the spirochetes belonged to Borrelia duttonii, which is the causative agent of tick-borne relapsing fever in East Africa. The rest of the spirochetes were quite different from B. duttonii and instead resembled the New World tick-borne relapsing fever borreliae. Phylogenetic analysis using 16S ribosomal RNA gene sequences also supported the interpretation that the spirochete was a Borrelia species distinct from previously described members of the genus.  相似文献
