A concentric bipolar electrode (CBE) (consisting of a central disc and an outer annulus) has been proposed as an approximate method of measuring the surface Laplacian of the body surface electrical potential distribution. The derivation of the surface Laplacian approximation, in terms of the potential difference measured with the bipolar electrode, contained an unspecified parameter which has been dubbed the 'effective radius' of the concentric bipolar electrode. This paper presents an asymptotic analysis to derive an expression for the effective radius in terms of the physical dimensions of the electrode (the radius of the central disc and inner and outer radii of the annulus). Also studied is the way in which the value of the effective radius affects the behaviour of the relative error in the surface Laplacian measurement at various dipole source locations within a conducting medium.  相似文献

This study tests the capacity of 50 Hz magnetic and electric fields to stimulate neurite outgrowth in PC-12D cells, a cell line which originated from a pheochromocytoma in rat adrenal medulla. The cells were plated on collagen-coated, plastic petri dishes and exposed to sinusoidal 50 Hz magnetic fields for 22 h in a 5% CO2 incubator at 37°C. Two 1,000 turn coils, 20 cm in diameter, were assembled in a Helmholtz configuration to generate a magnetic field in a vertical orientation, thereby inducing a companion electric field in the dish with intensity proportional to radius. A magnetic-field shield housed the control samples in the same incubator. Total cells and number of cells with neurites at least as long as one cell diameter or having a growth cone were counted within a radius of 0.3 cm of the dish center and within an annulus of 1.7–1.8 cm radii in 60 mm dishes, at 3.6 cm radius in 100 mm dishes, and between 1.9 and 2.1 cm radii in the outer well of organ culture dishes, which are physically separated into two concentric wells. Sham exposure demonstrated no difference in percentage of cells with neurites between the exposed and control locations in the incubator. Exposures were done at 4.0. 8.9, 22, 29, 40, 120, 236, and 400 milliGauss (mG). At dish radii of 1.7–1.8 cm in the 60 mm dishes these magnetic flux densities induced electric fields of 1.1, 2.5, 5.9, 8.1, 11, 33, 65, and 110 μV/m, respectively, while within a radius of 0.3 cm, the induced electric fields were less than 0.2, 0.4, 1.0, 1.5, 1.9, 6.0, 11, and 19 μV/m, respectively. For other dishes, the larger radii produced proportionally larger induced electric fields. At each field strength, there were two control dishes and four to nine exposed dishes: 100 or more cells were counted at each location on the dishes. The results demonstrate that magnetic fields stimulate neurite outgrowth in a flux-density-dependent manner between 22 and 40 mG, reaching an apparent stimulation plateau between 40 and 400 mG; no effects were seen at 8.9 mG or lower. There was no apparent neurite stimulation due to the electric field. Although relatively low intensity (?22mG) magnetic fields alone can stimulate a morphological response in a cell which is normally stimulated by nerve growth factor molecules binding to membrane receptors, the chemical basis of this response is unknown. © 1993 Wiley-Liss. Cylindrical segments from mesenteric veins of 8 cats were prepared and mounted in a Krebs-Ringer tissue bath. The oxygenized solution was bubbled with 95% O2 and 5% CO2. For lowered oxygen tension 95% N2 and 5% CO2 was used. Intraluminal pressure was changed between 0-20-0 mm Hg in consecutive cycles at a rate of 0.93-1.33 mm Hg/sec. Outer radii on the upward routes were recorded. Norepinephrine was added in doses of 6 X 10(-8) -6 X 10(-5) M, first in the oxygenized medium then under hypoxic conditions, and then in oxygenized medium again. Maximally relaxed curves were taken with 1.5 X 10(-4) M papaverine at the end of the experiment. Outer radius of relaxed segments at 20 mm Hg intraluminal pressure was 2.03 +/- 0.12 mm which slowly dilated to 2.09-0.12 mm toward the end of the experiment, and reached 2.11 +/- 0.11 mm with papaverine. Maximum active contractions of the outer radii were found at 6 mm Hg intraluminal pressure and with 6 X 10(-5) M norepinephrine in the bath: 23.1 +/- 3.2% in oxygenized, 20.3 +/- 3.4% in hypoxic and 19.0 +/- 3.4% again in oxygenized media. The observations showed that acute hypoxia had no or had only a limited effect on the contraction of the feline mesenteric vein.  相似文献

Purpose: In catheter-based intravascular brachytherapy, either photon or beta emitters are often used in a linear arrangement so that blood vessels of 10-30 mm lengths can be treated. With a line source, the dose gradient in the radial direction and longitudinal direction depend on the type of radionuclides used in the treatment. The purpose of this study was to investigate the dose fall-off at the edges of a linear source in a blood vessel for different types of photon and beta emitters.Materials/Methods: Dose distributions were calculated on cylindrical blood vessels of various radii. Radioactive sources of 192Ir, 125I, 103Pd, 188Re, 32P, and 90Y/Sr were studied. All the sources were assumed to be in the form of a line. The dose rate at a point in space produced by a radioactive source was computed by integrating the point dose rate kernel of the corresponding radionuclide over the radioactive line. The point dose rate kernel was computed with Monte Carlo simulation of radiation transport. The edge effects were characterized with three newly defined quantities: longitudinal dose uniformity (LDU), effective coverage length (ECL), and margin length (ML). LDU was defined as the ratio of dose at a distance along the long axis of the vessel to the dose at center. ECL was defined as the length over which the LDU was greater than 0.95. ML was defined as half of the length difference between source length L and ECL, which is essentially the length segment at each edge that is covered by the source physical length but is being underdosed.Results: All beta emitters provided more uniform dose distributions and covered a larger portion of blood vessels longitudinally than photon emitters. Typical MLs were 2-3 mm for beta emitters and 4-6 mm for gamma emitters. As the radial depth of the point of interest increased, both the LDU and ECL decreased and ML increased. The ML increased from 2 to 3 mm for beta emitters and from 4 to 6 mm for photon emitters when the radial depth of the point of interest increased from 1.5 to 2.5 mm (typical proximal and distal media points for a 3-mm diameter lumen). The ML increased with increasing source length for all radionuclides. For beta emitters the ML increased initially from 1.5 mm to more than 2.5 mm as source length increased from 5 to 10 mm. When the source length was longer than 15 mm, the ML remains nearly constant, about 3 mm. For photon emitters, ML increased continuously from 1.5 mm to more than 6.0 mm, as source length increased from 5 to 50 mm.Conclusions: A formalism to quantify the dose uniformity along the length of a blood vessel undergoing catheter-based intravascular brachytherapy has been developed. This formalism was used to study the edge effects at the ends of several beta and photon sources. The results indicated that for a centered source the ML at each end due to penumbra effects was about 2 to 3 mm for beta emitters; about 4-6 mm for photon emitters. The ML increases as the radial depth of point of interest in the vessel increases. The ML increases also with increasing source length, especially for photon sources.  相似文献

Intraluminal irradiation of coronary and peripheral arteries has been shown to reduce neointimal hyperplasia following balloon angioplasty, thereby inhibiting restenosis. Several irradiation techniques are being investigated, including temporary intravascular insertion of high activity gamma- or beta-emitting seeds and wires; inflation of dilatation balloon catheter with radioactive liquid or gas; insertion of miniature x-ray tubes via coronary catheters; permanent implantation of radioactive stents; and postangioplasty fractionated external beam irradiation. Unlike conventional brachytherapy, intravascular treatment of restenosis requires accurate knowledge of dose at distances of 0.5-5 mm from the radioactive source. This requirement presents special problems with regard to source calibration and dose specification, because dose gradients at such close distances from a radioactive source are extremely large. This makes it virtually impossible to define the characteristics of an ideal radiation source without some knowledge of the location and radiosensitivity of the target tissues, plus the radiotolerance of normal tissues. Hence, the current debate over whether beta or gamma sources are to be preferred. Imprecise knowledge of dose-volume effects for coronary arteries, plus uncertainties in the biological time sequencing of restenosis fuel a second debate on whether external beam treatments may be efficacious, and whether or not permanent radioactive stents may prove superior to high dose, single fraction brachytherapy. We review here the dosimetric properties of the various irradiation techniques and isotopes that have been proposed, including aspects of radiation safety, dose homogeneity, and practical aspects of source delivery.  相似文献

四面山阔叶林土壤大孔隙特征与优先流的关系   总被引:9,自引:0,他引:9  
为研究土壤大孔隙数量、分布特征与优先流发生之间的关系,在使用亮蓝染色法划分林地优先流发生区域基础上,利用穿透曲线理论方法,对重庆四面山典型亚热带阔叶林土壤剖面染色和未染色区域内的土壤大孔隙进行了量化分析.结果表明:研究林地土壤剖面内大孔隙半径多在0.3~3.0 mm,大孔隙率为6.3%~10.5%,随着土壤深度的增加,大孔隙呈现出聚集态的分布特征.各孔径范围内,染色区域的土壤大孔隙数量较未染色区域高出约1个数量级.半径>0.3 mm,尤其是半径>1.5 mm的大孔隙数量,是影响林地优先流发生的主要通道.森林土壤0.3~3.0 mm孔径范围内,大孔隙数量与其对应的土壤水分稳定出流速率呈显著的正相关关系,其中在0.7~1.5 mm和1.5~3.0 mm孔径范围内大孔隙数量与稳定出流速率相关程度最大,相关系数分别为0.842和0.879.发生优先流的染色区内大孔隙联通状况优于未染色区,两区中1.5~3.0 mm孔径范围内的联通大孔隙数量差异最大,相差78.3%.染色区内大孔隙数量随土壤深度的增加逐渐减少,“漏斗”状的孔隙分布特征可以形成有效的水压梯度,有利于水分优先运移.  相似文献

Reorganization and activation energies for charge transfer reactions occurring inside a dielectric sphere have been calculated by solving the problem of polar medium reorganization within and outside a dielectric sphere placed in another infinite dielectric. The dielectric sphere is assumed to simulate a protein globule, i.e. an enzyme molecule. It has been shown that for some reaction types the activation energy tends to decrease as the globule radius increases and that for each of the reaction types considered there is an optimal globule radius an increase of which does not bring about any tangible activation energy reduction. The calculated optimal radii for different processes are in good agreement with the increasing molecular sizes in the series: ribonuclease less than or equal to lysozyme less than serine proteinases approximately equal to cysteine proteinases less than NAD-dependent dehydrogenases. The calculated radii are usually about 1.5 to 1.7 times (and molecular masses about 4-5 times) smaller than the experimental ones. The reasons for this discrepancy are discussed and it has been suggested that the approximate nature of the treatment of a protein globule as a structureless dielectric is the main reason. It is shown that charge transfer at an acute angle to the globule surface is the optimum process. For endoergonic reaction stages it is the net charge transfer towards the periphery and for exoergonic ones that in the reverse direction which are advantageous. These conclusions are consistent with the data about the structure of the above-mentioned enzymes.  相似文献

Intact roots of Lepidium sativum were photographed at 1.5- to 6-min intervals before and after being tilted from a vertical to a horizontal position. Relative extension rates were calculated for segments of the root by measuring the rate of movement of charcoal particles on the surface of the root. Curvature was measured at points separated by 0.8 mm, from the tip to the root hair zone. For the first hour after the root was tilted to a horizontal position, the relative extension rate for the side of the root producing the outer side of the curve is higher than that during straight growth. The relative rate for the inner side falls to near zero during the first hour in the horizontal position. There is thus a positive gradient in extension rate from the inner to the outer side of the curve. For most of the second hour, the two sides have approximately equal relative rates of extension. In the last 12 min of the second hour, as the root tip is moving the final distance to a vertical position, the rate on the inner side shows a high extension rate. The gradient in extension rate from the inner to the outer side of the curve has thus become negative during the second hour. This reversal in the extension gradient is necessary to prevent the root from continuing to grow in a circle past the point where the root tip is vertical. Curvature reaches maximal values of 0.7 mm-1 to 1.2 mm-1 for the inner side and 0.5 mm-1 to 0.7 mm-1 for the outer side. (Units express reciprocals of the radius of curvature in mm.) Two locations where curvature is highest are separated by a flatter region. The charcoal particles marking specific locations on the root surface move away from the tip at the same rate as the features of the curvature pattern.  相似文献

Dielectrophoretic velocities of human red blood cells in an axisymmetric field were measured as a function of the applied voltage and the distance from the axis of symmetry. The voltage of the alternating electric field (frequency 2 MHz), applied between two concentric cylindrical metal electrodes (outer and inner radii 0.24 and 1 mm, respectively), was varied up to 19 V. Two kinds of mediums were used: (a) 90% of 2.1% glycine solution and 10% of 5.5% glucose solution and (b) 5.4% sorbitol solution. The results have shown that in both mediums the cell velocities are proportional to the square of the applied voltage and inversely proportional to the cube of the distance from the axis of symmetry, as predicted by the theory. The coefficient of proportionality (dielectrophoretic coefficient) is on the order of 10(-25) A2S4kg-1. It depends on the donor of red blood cells and might be used for diagnostic purposes. These results will be used in future investigations of membrane adhesion, stability and fusion.  相似文献

Ion channels contain narrow columns of water molecules. It is of interest to compare the structure and dynamics of such intrapore water with those of the bulk solvent. Molecular dynamics simulations of modified TIP3P water molecules confined within channel-like cavities have been performed and the orientation and dynamics of the water molecules analyzed. Channels were modeled as cylindrical cavities with lengths ranging from 15 to 60 A and radii from 3 to 12 A. At the end of the molecular dynamics simulations water molecules were observed to be ordered into approximately concentric cylindrical shells. The waters of the outermost shell were oriented such that their dipoles were on average perpendicular to the normal of the wall of the cavity. Water dynamics were analyzed in terms of self-diffusion coefficients and rotational reorientation rates. For cavities of radii 3 and 6 A, water mobility was reduced relative to that of simulated bulk water. For 9- and 12-A radii confined water molecules exhibited mobilities comparable with that of the bulk solvent. If water molecules were confined within an hourglass-shaped cavity (with a central radius of 3 A increasing to 12 A at either end) a gradient of water mobility was observed along the cavity axis. Thus, water within simple models of transbilayer channels exhibits perturbations of structure and dynamics relative to bulk water. In particular the reduction of rotational reorientation rate is expected to alter the local dielectric constant within a transbilayer pore.  相似文献

A novel concentric design of double-barrelled Ca2+-selective microelectrode, with an inner pipette tip that protrudes beyond an outer one, has recently been developed and is described. This configuration of pipettes was produced from concentric capillaries in one step using a horizontal pipette puller. For the tip of the inner barrel to protrude, Corning 1724 aluminosilicate glass was selected, as it has a higher melting point than the 1723 glass which is used for the outer barrel. To reduce electrode resistance the inner capillary was best made with a triangular shape. It was preferentially silanized in a dry box by injection of methyltrichlorosilane into only the inner barrel. The Ca2+ neutral carrier-based liquid membrane (ETH 1001) was back-filled from the tip to the shank of the inner pipette and above this CaCl2 solution was added. KCl, which contained EGTA and was buffered to pCa 7, was used to fill the reference barrel. These Ca2+ electrodes showed linear response with slope approximately equal to 30 mV for changes in Ca2+ concentration between 10(-3) and 10(-7) M in the presence of constant [K+]. They offer a number of advantages including a low noise level achieved by the presence of the external concentric KCl electrode, and a simple mechanical structure that allows applications to a variety of small cells.  相似文献

Angiotensin-converting enzyme inhibitors have been shown to inhibit intimal thickening following balloon catheterization of rat carotid arteries. To assess the role of the renin-angiotensin pathway and the angiotensin type-I (AT1) receptor in this effect, the nonpeptide Ang II antagonist losartan (DuP 753) or vehicle was infused continuously i.v. in rats from two days before to two weeks after balloon injury to the left common carotid artery; drug effects upon intimal thickening were examined histologically. Losartan produced a dose-dependent reduction in cross-sectional area of intimal lesions determined two weeks post balloon injury. At 5 mg/kg/day a nonsignificant 23% reduction of intimal area was observed. At the higher dose of 15 mg/kg/day, losartan produced a 48% reduction in intimal area (P less than 0.05) compared to the vehicle-infused group. The cellular density of the neointima was not affected by losartan, indicating a probable effect of the drug upon migration and/or proliferation of smooth muscle cells. In separate groups of non-ballooned rats, losartan infusions of 5 and 15 mg/kg/day produced significant rightward shifts (averaging 6.4- and 55-fold, respectively) in curves relating increases in blood pressure to intravenous Ang II in pithed rats determined between 2 and 16 days following initiation of losartan infusion. Mean arterial blood pressure (determined under alpha-chloralose anesthesia) was reduced following continuous losartan infusion for 6 days from 128 +/- 8 mm Hg (vehicle) to 105 +/- 8 mm Hg at 5 mg/kg/day (P less than 0.05), and 106 +/- 4 mm Hg at 15 mg/kg/day (P less than 0.05). Thus, losartan attenuated the vascular response to balloon catheter injury, and this effect was associated with functional block of vascular AT1 receptors. The results support a role for Ang II, acting via AT1 receptors, in myointimal thickening subsequent to balloon injury of rat carotid arteries.  相似文献

Allometric analysis was employed to compare linear dimensions of forelimb and hindlimb bones (humeri, radii, third and fifth metacarpals, third and fifth manual phalanges, femora, and tibiae) of 227 species of bats and 105 species of nonvolant mammals of varying degrees of phylogenetic affinity to bats. After accounting for body size, all forelimb bones are longer in bats than in nonvolant species, with the exception of humeri and radii of a few highly arboreal primates. Hindlimb bones are generally, but not uniformly, shorter in bats than in other mammals. For the humerus, radius, and metacarpals, midshaft diameters are greater in bats than in their comparably sized relatives. Proximal phalangeal midshaft diameters are statistically indistinguishable from those of other mammals, and distal phalanges show significantly reduced outer diameters. The pattern of relative reduction in wing bone diameters along the wing's proximodistal axis parallels the reduction in bone mineralization along the same axis, and a similar pattern of change in cortical thickness from the smallest wall thicknesses among mammals in the humerus and radius to the greatest wall thicknesses among mammals in the phalanges. The combination of altered cross-sectional geometry and mineralization appears significantly to reduce the mass moment of inertia of the bat wing relative to a theoretical condition in which elongated bones preserve primitive mammalian mineralization levels and patterns of scaling of long bone diameters. This intercorrelated suite of skeletal specializations may significantly reduce the inertial power of flight, contributing significant energetic savings to the total energy budgets of the only flying mammals. During spot tests using Salmonella TA98 derivatives (YG1021, YG1021) and TA100 derivatives (YG1026, YG1029), a unique response of O-acetyltransferase (OAT)-enhanced strains YG1024 and YG1029 to arylamines was observed. On plates containing rat-liver S9, these strains yielded revertant colonies induced in two separate concentric rings around the site of application, while the parent (TA98, TA100) and nitroreductase-enhanced strains (YG1021, YG1026) did not exhibit this response. The inner ring of revertants was accompanied by cytotoxicity and microcolony formation, with the outer ring in a region without background lawn toxicity. Addition of tetracycline to the top agar eliminated formation of the inner ring of YG1024 revertants in spot tests and reduced the revertant count in preincubation assays at cytotoxic dose levels of 2-aminoanthracene, 2-aminofluorene, 2-amino-6-methyldipyrido[1,2-a:3′,2′-d]imidazole and 2-amino-3,4-dimethylimidazo[4,5-f]quinoline. Tetracycline sensitivity indicates that mutant colonies developing at high concentration/toxicity arose, in effect, from TA98 regenerated by functional loss of the tetracycline-resistance plasmid (pYG219) from YG1024. Mutant colonies found at low concentration/toxicity arose from normal plasmid-bearing YG1024. These results indicate the need to consider coincidental toxicity-induced instability in YG1024 during quantitative mutagenicity assays of arylamines and uncharacterized complex mixtures.  相似文献

In aged animal brains, a variety of holes are formed in the neuropil. One type of hole, here designated as the myelin balloon, is an abnormality of the myelin sheath and is found in a number of diverse sites in the brain. Profiles of myelin balloons display rather smoothly rounded peripheral contours and typically range up to 10 μm in diameter, although exceptionally large examples may be twice this size. The balloons are bounded by lamellae of myelin, and to accommodate the contents of the balloon, the myelin sheath becomes split at the intraperiod line. Since the intraperiod line is formed by the apposition of the outer faces of the myelin-forming plasma membrane, the contents of the myelin balloons are, in effect, in continuity with the extracellular space, and it is suggested that the contents of the balloons are fluid, with the fluid exerting an outward pressure on the walls of the balloons to produce their spherical shapes. Myelin balloons are not only produced during aging but also occur in a number of genetic strains of mice and in a number of human disease states. They thus represent a non-specific, though distinctive and common, alteration of the myelin sheath and are a reflection of the fact that under a variety of conditions, including normal aging, oligodendrocytes are unable to maintain the integrity of their sheaths.  相似文献

An apparatus was developed to prevent the freezing of catheter contents during blood sampling or liquid infusion into animals maintained at sub-freezing temperatures. The apparatus consisted of a larger, outer set of plastic and copper tubes and a smaller, inner set of tubes. The catheter was passed inside the inner set. Water circulated through the outer set kept the contents of the catheter inside the inner set from freezing. The apparatus was also useful for the rapid cooling of blood to a predetermined temperature while a sample was being withdrawn from an animal.  相似文献

The transbilayer fatty acid distribution of diacylglycerophosphoethanolamine and the translocation of newly synthesized phosphatidylethanolamine molecules labelled with different fatty acids has been investigated in chick brain microsomes using trinitrobenzensulfonic acid. The determination of the fatty acid composition of diacylglycerophosphoethanolamine in both the outer and the inner leaflet of the microsomal vesicles revealed a similar distribution indicating that both leaflets share the same molecular species. The in vitro incorporation of radioactive fatty acids (16:0, 18:1 and 20:4(n-6] into ethanolamine phospholipids, known to be catalyzed by the lyosphosphatidylethanolamine acyl transferase, showed that the radioactive diacylglycerophosphoethanolamine molecules appeared first in the outer leaflet and were thereafter transferred to the inner leaflet. The apparent rate of translocation of the newly synthesized ethanolamine phospholipid molecules was the highest for those labelled with 16:0 and the lowest for those labelled with 20:4(n-6). The results indicate that the active site of the acyl-CoA:lysophosphatidylethanolamine acyltransferases is located on the outer leaflet of the microsomal vesicles and that the different newly synthesized molecular species of diacylglycerophosphoethanolamine may be translocated from the outer to the inner leaflet at different rates.  相似文献

Despite the increasing importance of airlift fermentors, very little published information is available on how the geometric configurations of the draft tubes and the air-sparging system affect the mixing and oxygen transfer characteristics of the fermentor. A 14-L air-lift fermentor was designed and build with a fixed liquid height to diameter ratio of 1.5 utilizing four equally spaced air jets at the bottom. Two jet orifice sizes were used, 1.27 and 3.81 mm i.d., and for each jet size the following four geometric configurations were used: Single inner concentric draft tube, single outer concentric draft tube, two concentric draft tubes, and no draft tubes where the fermentor was operated as a shallow bubble column. It was found that the presence of draft tubes stabilized liquid circulation patterns and gave systemically higher mixing times than those obtained in the absence of draft tubes. In addition, the double draft tube geometry resulted in higher mixing times than the single draft tubes. For the power unit volume range 20 to about 250 W/m3 the larger 3.81-mm orifices gave systemically higher kL a values than the smaller 1.27-mm i.d. orifices. At 200 W/m3 the use of a single outer draft tube with the 3.81-mm orifices resulted in 94% increase in kL a values over that obtained with no draft tubes. However, the effect of draft tube geometry on kL a values when the 1.27-mm orifices were used was not significant. The air bubble formation characteristics at the jet orifices were found to be different, which reflected the differences observed in mass transfer and mixing characteristics. The power economy for oxygen transfer was found to be depend strongly on the orifice size and less on the geometric configuration of draft tubes.  相似文献
