The parasitoid Aphidius colemani developed normally (approximately 90% adult emergence) when its cotton aphid (Aphis gossypii) host was treated with Verticillum lecanii conidia 5 or 7 days after parasitization. Fungus exposure 1 day before or up to 3 days after parasitization, however, reduced A. colemani emergence from 0 to 10%. Also, numbers of spores and mycelial fragments in aphid homogenates were much higher in aphids exposed to the fungus up to 3 days after parasitization than in aphids treated after 5 or 7 days. Our results suggest that the parasitoid and fungus may be used together for aphid biocontrol as long as fungus applications are timed to allow late-instar development of the parasitoid.  相似文献

The new series Elatostema section Elatostema series Albopilosoides Q.Lin & L.D.Duan (Urticaceae) is described, and two new species of Elatostema, namely Elatostema albopilosoides Q.Lin & L.D.Duan and Elatostema purpureum Q.Lin & L.D.Duan. from south Guizhou province, south‐west China are described and illustrated. Both species were found growing only at the base of a large limestone chamber at an altitude of c. 800 m. Elatostema albopilosoides is morphologically similar to E. albopilosum W.T.Wang, but differs by having female inflorescences with peduncles 10–60 mm long (0–1.5 mm long in E. albopilosum) and receptacles 7–27 mm long, 7–24 mm wide (E. albopilosum: 1–4 mm long and 1–3 mm wide). Elatostema purpureum is also morphologically similar to E. albopilosum, but has stipules 5–6 mm long, 0.8–1.5 mm wide (4–7 mm long, 1.5–2.8 mm wide in E. albopilosum) and leaf blades obliquely elliptical to obliquely oblong‐obovate, 3.5–7.5 mm long and 1.5–2.5 mm wide (obliquely narrowly oblanceolate‐oblong, 12–17 cm long, 3–5 cm wide in E. purpureum:). © 2008 The Linnean Society of London, Botanical Journal of the Linnean Society, 2008, 158 , 674–680.  相似文献

When a three-step roller culture system was used, second-stage larvae of Ascaris suum, artificially hatched from eggs, developed in high numbers to the fourth stage, and a few to young and mature adults. The culture system consisted of (1) Medium KW-2 supplemented with 10 mM L-cysteine for the first 4 days, and with 5 mM L-cysteine for the following 7 days; (2) followed by Medium API-18 for 7 days; and (3) thereafter, by Medium API-1 supplemented with hemin (bovine) at a concentration of 24 micrograms/m1. Cultures were gassed with 95% nitrogen-5% carbon dioxide for the first 4 days and 85% nitrogen-5% oxygen-10% carbon dioxide thereafter, and incubated at 39 C. Two mature females that produced unfertilized eggs and a mature male with spermatozoa were the most advanced stages attained. The mature females were obtained in 67 and 73 days; and the largest female measured 110 mm. The latter produced 1,356,000 unfertilized eggs, from days 67 to 125. The mature male was obtained in 80 days; it measured 77 mm long and had paired spicules that were 1.5 mm long. Development of A. suum in three other culture systems showed that deletion of Medium API-18 or its substitution by Medium KW-2 limited development to late fourth stage and early, young adults, respectively; and the use of Medium API-1 without hemin limited development to early fourth stage.  相似文献

The aims of this study were to investigate steady‐state level of Kit Ligand (KL) mRNA and its effects on in vitro survival and growth of caprine preantral follicles. RT‐PCR was used to analyze caprine steady‐state level of KL mRNA in primordial, primary, and secondary follicles, and in small (1–3 mm) and large (3–6 mm) antral follicles. Furthermore, ovarian fragments were cultured for 1 or 7 days in Minimal Essential Medium (MEM+) supplemented with KL (0, 1, 10, 50, 100, or 200 ng/ml). Noncultured (control) and cultured fragments were processed for histology and transmission electron microscopy (TEM). RT‐PCR demonstrated an increase in steady‐state level of KL mRNA during the transition from primary to secondary follicles. Small antral follicles had higher steady‐state levels of KL mRNA in granulosa and theca cells than large follicles. After 7 days, only 50 ng/ml of KL had maintained the percentage of normal follicles similar to control. After 1 day, all KL concentrations reduced the percentage of primordial follicles and increased the percentage of growing follicles. KL at 10, 50, 100, or 200 ng/ml increased primary follicles, compared to MEM+ after 7 days. An increase in oocyte and follicular diameter was observed at 50 ng/ml of KL. TEM confirmed ultrastructural integrity of follicles after 7 days at 50 ng/ml of KL. In conclusion, the KL mRNAs were detected in all follicular categories. Furthermore, 50 ng/ml of KL maintained the integrity of caprine preantral follicle cultured for 7 days and stimulated primordial follicle activation and follicle growth. Mol. Reprod. 绿盲蝽对不同生长期棉花的刺吸危害特性   总被引:1,自引:0,他引:1  
【目的】近年绿盲蝽Apolygus lucorum (Meyer-Dür)对棉花的危害成为农业中热议的话题和重点研究的课题。为更加深入了解此种害虫对棉花各组织的危害特性及量化危害,为早期预测预报提供依据,我们研究了绿盲蝽对棉花不同生长阶段(各组织)早期危害表征与后期成铃情况的关系。【方法】通过大田接虫法研究了绿盲蝽对苗期、蕾期、花期和铃期棉花植株的危害特性。【结果】绿盲蝽危害24 h会造成棉花子叶期和2叶期产生无头苗,2叶期后受绿盲蝽危害不会形成无头苗。建立了无头苗率和生长点被害级别的关系模型y=0.1906x-0.0439 (1≤x≤5)和棉花苗期(不同叶期)被害10 d后的叶片被害指数与刺吸点数和生长点被害级别回归模型:4叶期:y=0.532+0.202x1+0.005x2; 6叶期:y=-0.063+0.339x1+0.002x2; 8叶期:y=-0.150+0.087x1+0.001x2 (0≤x1≤5, 1≤x2≤5)。绿盲蝽对蕾和花的刺吸危害没有对后期棉铃的脱落率和大小产生显著影响。10 mm和15 mm直径棉铃被害后的校正脱落率分别为55.00%和26.60%,而直径大于15 mm的棉铃校正脱落率均为0;绿盲蝽的刺吸危害对棉铃的成铃大小无显著影响;建立了幼铃(10 mm、10~15 mm直径棉铃)刺吸点数和脱落率的回归模型[10 mm: y=0.1857x-0.081; 10~15 mm: y=0.0522x-0.0068(1≤x≤5)]。【结论】结果表明,幼苗阶段,棉花的子叶期和2叶期是受绿盲蝽危害的敏感时期;成株阶段,棉花嫩铃期是受绿盲蝽危害的敏感时期;建立的绿盲蝽刺吸危害的回归模型可用于绿盲蝽危害损失短期预测。因此,应在棉花受害敏感时期对绿盲蝽重点防治,提高防治效率。  相似文献

Micropropagation is currently the most popular method for orchid propagation through the production of protocorm-like bodies (PLBs). It is suggested that converting the PLBs into artificial seeds by encapsulation with sodium alginate can be useful for short-term preservation and distribution to the laboratories and commercial nurseries. Prior to the production of artificial seeds, the best developmental stage of PLBs based on sizes for increased conversion to plantlet was determined. PLBs were categorized based on size and presence of shoot namely ≤2 mm (S1), >2–4 mm (S2), >4–6 mm (S3), >2–4 mm with shoot (S4) and >4–6 mm with shoot (S5). S4 and S5 gave significantly higher conversion percentage (85 and 90 %, respectively) as compared to the PLBs without shoot (S1, S2 and S3). Thus, for uniformity PLBs of 3–5 mm with shoot were used for encapsulation with sodium alginate to form artificial seeds. The feasibility of germinating artificial seeds of Dendrobium Shavin White in different substrates namely; M1 (semi-solid ½ Murashige and Skoog (1962) basal medium), M2 (cotton irrigated with sterilized liquid ½ MS basal medium), M3 (cotton irrigated with sterilized distilled water) and M4 (cotton irrigated with non-sterilized distilled water) was tested. The encapsulated PLBs regenerated well in M1 where 96 % of encapsulated PLBs germinated after 12 days of inoculation and 76 % of them converted into plantlet after 37 days of inoculation while PLBs subjected to sterile distilled water gave 56 % germination and 44 % conversion after 42 and 167 days of inoculation respectively. The ability to store encapsulated PLBs would be advantageous for transport of planting materials. Encapsulated PLBs survived longer when stored at 25 ± 2 °C compared to 4 °C, 10 °C and 30 ± 2 °C whereby storage up to 75 days retained 80–92 % survival. Further storage up to 135 days retained 52 % survival. All plantlets survived after acclimatization when transferred to charcoal media under shade.  相似文献

Walnut somatic embryos were multiplied by repetitive embryogenesis on a solid basal DKW medium at 25°C in the dark. When the embryos were isolated at early cotyledonary stage (1–2 mm long) from the primary embryos and cultured on the medium for 3 weeks, they developed into mature embryos showing white, enlarged cotyledons and shoot and root apex. After transfer to light on solid germination medium, however, few mature embryos (0–5%) germinated. Germination percentage increased to about 10% when the mature embryos were pretreated by a storage at 4°C in the dark for 2 months, or by desiccation at 25°C in the dark for 3 or 5 days under an air-humidity conditioned by saturated salt solutions (Mg(NO3)2.6H2O, or ZnSO4.7H2O). Similar results were obtained by the addition of gibberellic acid (GA3) to the germination medium. When mature embryos were desiccated and then placed on medical cotton compresses in liquid germination medium, 45% of the embryos germinated into complete plantlets. The relatively rapid and extensive corolla expansion in Ipomoea nil (L.) Roth cv. ‘Scarlett O'Hara’ appears to be initiated by a shift in the balance between a growth promoter (gibberellin, GA) and a growth inhibitor (ethylene) two days before anthesis. The effects of applied growth regulators in vitro, were measured as a change in area of segments from whole young corollas 16–17 mm long. Applied gibberellic acid (GA3) strongly promoted growth, while a GA action inhibitor (ancymidol) reduced growth. Inhibitors of GA biosynthesis (AMO-1618, paclobutrazol, tetcyclasis, and chlorocholine chloride) had little effect, implying that the GAs were not being synthesized within the corolla segments, at least at this stage of corolla growth. Both the level of endogenous GAs and response of the segments to sucrose increased as the corolla size also increased from 15 to 20 mm in length. Applied 1-aminocyclopropane-l-carboxylic acid (ACC), an ethylene precursor, inhibited the response of corollas to applied GA3. Applications of ethylene biosynthesis inhibitors (AVG and cobalt ions) promoted growth of the corolla segments. Rapid ethylene production by corollas 15–17 mm long (48 hr before flower opening) appeared to inhibit the growth that may have been induced by low levels of endogenous GAs. Older corollas (longer than 18 mm) had very low levels of ethylene production and much higher levels of GA-like substances; these changes apparently allow the corollas to begin to expand in vivo and to strongly respond to sucrose applications alone in vitro. Acid-induced growth does not seem to be an important component of corolla expansion; applied fusicoccin (a proton efflux promoter) and sodium orthovanadate (a proton efflux inhibitor) had no significant effect on growth of the 16–17 mm long corollas. Buffers (Good's and phosphate) over a wide pH range had no effect on corolla expansion. Taken together, our results indicate that the regulation of corolla expansion depends (at least in part) on compensatory shifts in the levels of two plant growth regulators [PGRs], ethylene and GA.  相似文献

Employing a different culture strategy, we obtained a greatly improved frequency of embryo rescue in intersubgeneric soybean hybrids. Successful crosses were obtained in 31 different genotype combinations between nine Brazilian soybean lines as the female parents and 12 accessions from Glycine canescens, G. microphylla, G. tabacina and G. tomentella. The hybrid pod retention rate dropped to about 10% during the first 8 days after pollination and stayed largely unchanged up to the 20th day. Immature harvested seeds fell into three size groups: Group 1, smaller than 1.3 mm (mostly empty seed coats); Group 2, 1.9–5.0 mm; Group 3, larger than 5 mm (from selfing). A total of 90 putative hybrid embryos were rescued using a highly enriched B5 medium to nourish the newly dissected embryos. The growing embryos were then placed in a high osmotic, modified B5 medium to induce maturation and dormancy. Schenk and Hildebrandt medium was used to germinate the dormant, partially dehydrated, physiologically mature embryos. Approximately 37% of the rescued embryos developed into plantlets in vitro, and approximately 8% grew into mature plants in the greenhouse. Morphological, cytological and isoenzyme patterns confirmed the hybrid status of all seven mature plants, all of which were generated using G. tomentella G 9943 as the paternal parent. It was observed that all soybean lines crossed with G 9943 were capable of producing mature hybrid plants. There was no correlation between the initial size of Group 2 seeds and plant survival rate. The hybrids were cloned by grafting and treated with colchicine. One of the treated plants displayed chromosome doubling.  相似文献

The relation of ovarian follicle size to pituitary and ovarian responses to copulation was studied in domesticated South American camelids (llamas and alpacas). Females from each species were divided into four groups according to follicle size: small (4-5 mm), growing (6-7 mm), mature (8-12 mm), and regressing (10-7 mm). The pituitary response to copulation was determined by analysis of LH and FSH concentrations in plasma. The ovarian response to copulation was determined by ultrasonography and by analysis of estrone sulfate (follicular status) and pregnanediol glucuronide (luteal status) concentrations in urine. Females with small follicles (4-5 mm) released less LH after copulation than did those with larger follicles, and ovulation was not induced. Females with growing and mature follicles (7-12 mm) released LH in response to copulation that was adequate to induce ovulation and to initiate normal luteal activity. While copulation-induced LH release in females with regressing follicles was similar to that released in animals with growing and mature follicles, regressing follicles were luteinized instead of being ovulated. The luteal structure formed as a result of luteinization of follicles had a short life span, i.e., 5.1 days. Copulation-induced LH release was significantly higher in llamas vs. alpacas in animals with mature or regressing follicles, but not in those with small or growing follicles. Urinary estrone sulfate and pregnanediol glucuronide concentrations correlated positively with the presence of follicles and corpora lutea, respectively.  相似文献

Copper content in muscle and the presence of bullet fragments were assessed in samples from red deer (Cervus elaphus), roe deer (Capreolus capreolus), fallow deer (Dama dama) and wild boar (Sus scrofa) (total of 46 animals) which had been hunted with two types of solid copper bullets. Also, the release of copper from bullets or fragments remaining in muscle was tested. For bullet type “B”, a fragment was detected in only 1 out of 34 carcasses whereas for type “A”, fragments were detected in all 12 carcasses, with up to 24 fragments (maximum size, 5?×?7?×?2 mm). Median copper concentrations around the shot wound (0–30 mm distance) were 1.25 and 1.77 mg/kg fresh weight for bullet types B and A, respectively, and thus in the expected range for venison. Around bullet fragments that had remained in muscles, the copper content increased significantly. In roe deer longissimus muscle that had been exposed for 7 days at +7 °C to bullets of type B, up to 1,000 mg/kg copper (fresh weight) was found in a distance of 0–2 mm. However, in a distance of 10–20 mm, maximum copper contents were <10 mg/kg fresh weight. Bullet fragments can constitute physical hazards and will release copper under acidic conditions as those prevailing in meat. Removal of bullet fragments prior to culinary preparation should ensure that a recommended dietary copper intake of 1.25 mg per adult consumer per day is not exceeded. From a food hygiene viewpoint, non-fragmenting bullets seem to be preferable.  相似文献

Shirleya grahamae Pigg & DeVore gen. et sp. nov. (Lythraceae) is established for silicified fruits from the middle Miocene Yakima Canyon of central Washington State, USA. The capsules are 10 mm long × 11.5-12.5 mm wide, enclosed in a persistent floral tube and contain 5-7 locules. They are loculicidally dehiscent, fracturing into fragments and leaving the central axis free. Placentation is axile. Five to seven mature seeds are tightly packed per locule, often with several smaller seeds. Seeds are winged, anatropous, and narrowly attached subapically to the central axis, curving basally and radially within the fruit. They are up to ~4.6 mm long × 1.9 mm wide, with a small, triangular embryo cavity and a prominent distal wing. The inflated wing is filled with a bilobed parenchymatous pad of tissue with a central cavity. Shirleya grahamae is assigned to the Lythraceae, and is most similar to Lagerstroemia, based on the synapomorphies of distally winged seeds and revolute cotyledons. Shirleya differs from Lagerstroemia in seed arrangement, and pericarp and wing anatomy. This study provides the first anatomical information about a Miocene Lagerstroemia-like fruit and documents further diversity of the Lythraceae in the Neogene of northwestern North America.  相似文献

The subtidal brown algal species Sporochnus dotyi Brostoff (Sporochnales, Phaeophyceae), which has been regarded as a Hawaiian endemic, is reported from Kushimoto, Kii Peninsula, Pacific coast of central Honshu, Japan, for the first time outside Hawai'i. The species grew on subtidal rocks ca. 5–20 m deep attached by a small conical holdfast. The erect thalli were 5–30 cm high, terete, robust and alternately branched in 1–2 orders. When mature, pedicellate receptacles developed on the branches, and formed elliptical sori 1 mm long with a pedicel 3–5 mm long. The apical parts of the thalli and the receptacles were terminated with a tuft of simple assimilatory filaments of up to 4 mm long and showed prominent green to yellow underwater iridescence. Reproductive filaments (paraphyses) were densely packed, simple, up to 200 μm long and bore 4–6 mostly unilateral unilocular zoidangia 20–22 μm long and 5–6 m in diameter. In the genetic analyses, the Sporochnus alga from Kushimoto had partial rbcL sequence identical to S. dotyi from Hawai'i. The cox3 phylogeny revealed that this alga formed a fully supported clade with S. dotyi. Therefore, we identified the alga from Kushimoto as S. dotyi. This finding of S. dotyi from Japan, together with the recent reports of the mesophotic macroalgae Ryuguphycus kuaweuweu (=Umbraulva kuaweuweu), Ulva iliohaha and Newhousia imbricata from various localities in the Pacific Ocean including Japan, suggest closer biogeographical connections of subtidal/mesophotic macroalgae in the Pacific than previously recognized.  相似文献

通过两年的田间试验,研究了滴水量和滴水频率对膜下滴灌棉田土壤水分分布及棉花水分利用效率的影响.结果表明: 从整个生育期来看,当滴水量(375 mm)相同时,高频滴灌(每3天1次)处理0~20 cm土层含水率较高而深层土壤湿润不够;低频滴灌(每10天1次)处理有利于水分的下渗和侧渗,深层土壤含水率较高,但水分补给不及时,表层土壤偏低;总体上中频滴灌(每7天1次)处理有利于水分在土壤剖面中的均匀分配.当滴水频率相同时,滴水量越大,土壤含水率越高,40 cm以下土层含水率也越高.不同处理的棉田耗水规律基本一致,苗期较低,平均不高于1.7 mm·d-1,蕾期开始上升至花铃期达到最高,日均耗水量可达8.7 mm·d-1,吐絮期回落到1.0 mm·d-1左右.总耗水量与降水和滴水量密切相关,而与滴水频率无关;滴水频率对棉花水分利用效率无显著影响,但水分利用效率随滴水量的增大而显著降低.少量滴灌(300 mm)虽然可以获得较高的水分利用效率,但减产严重,过量滴灌(450 mm)无显著增产效应,水分浪费严重.在当地棉田自然条件下,采用中量(375 mm)+中低频(每7天或10天1次)的滴灌模式为宜.  相似文献

Meiotic maturation of mammalian oocytes is under the control of cell cycle molecules Cdc2 kinase and MAP kinase (mitogen-activated protein kinase). In the present study, we investigated the relationship between the ability to activate Cdc2 kinase and MAP kinase and the acquisition of meiotic competence during pig oocyte growth. Growing and fully grown pig oocytes were collected from four groups of antral follicles of various diameters (A, 0.5-0.7 mm; B, 1.0-1.5 mm; C, 2.0-2.5 mm; D, 4.0-6.0 mm) and cultured in vitro. Fully grown oocytes from class D follicles, which have full competence to mature to metaphase II, had the ability to activate both Cdc2 kinase and MAP kinase. In contrast, growing oocytes from class A follicles, which have limited competence to resume meiosis, had no such ability. Cyclin B1 molecules did accumulate, however, with phosphorylated 35 and 36 kDa bands of p34cdc2 appearing in the cultured oocytes. Of the growing oocytes from class B follicles, 60% resumed meiosis but arrested at metaphase I. Some of the oocytes in this class were capable of activating Cdc2 kinase, although they did not appear to have established a MAP kinase-activating pathway or the ability to activate MEK. These results suggest that limited meiotic competence in growing oocytes from class A follicles is due to their inability to activate Cdc2 kinase and their incomplete MEK-MAP-kinase pathway, although the oocytes are capable of accumulating cyclin B1 molecules. During the final growth phase, pig oocytes acquire the ability to activate Cdc2 kinase and then establish the MEK-MAP-kinase pathway for full meiotic competence.  相似文献

We compared bird community responses to the habitat transitions of rainforest‐to‐pasture conversion, consequent habitat fragmentation, and post‐agricultural regeneration, across a landscape mosaic of about 600 km2 in the eastern Australian subtropics. Birds were surveyed in seven habitats: continuous mature rainforest; two size classes of mature rainforest fragment (4–21 ha and 1–3 ha); regrowth forest patches dominated by a non‐native tree (2–20 ha, 30–50 years old); two types of isolated mature trees in pasture; and treeless pasture, with six sites per habitat. We compared the avifauna among habitats and among sites, at the levels of species, functional guilds, and community‐wide. Community‐wide species richness and abundance of birds in pasture sites were about one‐fifth and one‐third, respectively, of their values in mature rainforest (irrespective of patch size). Many measured attributes changed progressively across a gradient of increased habitat simplification. Rainforest specialists became less common and less diverse with decreased habitat patch size and vegetation maturity. However, even rainforest fragments of 1–3 ha supported about half of these species. Forest generalist species were largely insensitive to patch size and successional stage. Few species reached their greatest abundance in either small rainforest fragments or regrowth. All pastures were dominated by bird species whose typical native habitats were grassland, wetland, and open eucalypt forest, while pasture trees modestly enhanced local bird communities. Overall, even small scattered patches of mature and regrowth forest contributed substantial bird diversity to local landscapes. Therefore, maximizing the aggregate rainforest area is a useful regional conservation strategy.  相似文献

研究基于新疆地区54个国家基本气象站1963-2012年的逐日气象资料,采用联合国粮食与农业组织FAO推荐的Penman-Monteith公式及作物系数法,计算了近50年来新疆棉花全育期和各生长阶段需水量。并利用线性趋势估计、MK检验、空间插值法对农作物全育期及各生长阶段需水量年际变化趋势和空间分布特征进行分析。结果表明:1)1963-2012年,新疆54个气象站棉花全育期平均需水量为759 mm,各站变化范围为555-1205 mm,差异明显。其空间分布整体呈现西北向东南递增、南疆大于北疆、东部大于西部的趋势。2)1963-2012年,在棉花的生长初期与发育期,全疆需水量空间分布总体由北向南增大并呈带状。在棉花的生长中期与生长末期,需水量空间分布表现为由西向东增大的格局。3)近50年棉花不同生长阶段需水量总体呈现下降趋势,并且除了生长中期下降趋势较显著外其他都不太明显。其中,生长中期下降速率最大为6.1 mm/10a,发育期及生长末期下降速率次之分别为2 mm/10a和1.3 mm/10a,生长初期下降速率最小,需水量以0.6 mm/10a的速率递减。4)新疆1963-2012年棉花全育期需水量总体呈下降趋势,递减速率平均约为5.3 mm/10a,并在1981年开始发生突变。5)全育期年需水量下降倾向率总体呈现与全育期平均需水量相同的空间分布特征,总体呈现北疆小于南疆、东部大于西部、由西北向东南递增的空间分布特征。  相似文献

Observations were made on the maturation of stamens and ovaries from cultured maize (Zea mays L.) ear inflorescences. Immature ears (5.1–10.0 mm long) of maize were cultured in kinetin medium to study microsporogenesis and pollen maturation in developing stamens. Male spikelets developed on ears cultured in kinetin medium. Meiosis-I began by 7 days of culture in the developing anthers and the mature tri-nucleate pollen grains were developed by 20 days of culture. Further, kinetin was required in the culture medium for at least initial 5 days to obtain the microspores in differentiated stamens.To observe the embryosac formation in developed ovaries, ears were cultured in control, kinetin (10.1–15.0 mm long ears) medium, and kinetin + gibberellic acid (5.1–10.0 mm long ears) medium. Formation of embryosacs was noticed in the developed ovaries which were sampled after 20 days of culture. This differential flower development using two growth regulators provides an opportunity to uncover the biochemistry and physiology of micro- and mega-gametophyte development in maize.  相似文献

Growth parameters of vegetative hyphae and isolated tip fragments of the mycelial fungus N. crassa were studied after complete substitution of an easily metabolized carbon source (glucose) for a non-metabolized one (sorbitol). The images of growing tips were recorded at 20–30-min intervals. Using original image processing software, geometrical parameters of the hyphal trees (length and number of branches, area of convex polygons circumscribed about the hyphal trees, etc.) were determined and growth characteristics, such as rate of tip elongation (V) and the ratio of the total hyphal length to the number of growing tips (termed “hyphal growth unit”, HGU), were calculated. It is shown that after 4–5-h growth in sorbitol-enriched media growth characteristics of intact hyphae did not differ significantly from the corresponding parameters of hyphae growing in glucose-enriched media. In isolated tip fragments (about 800-μ m long), the values of V were lower than those in intact hyphae but did not depend on the carbon source in the nutrient media. However, in such fragments growing in sorbitol-enriched media the number of branches decreased, while the HGU value and the number of large intracellular vacuoles increased. Staining of cells with a standard chitin probe, Calcofluor White (10 μg/ml), did not reveal any considerable differences in hyphal cell walls and septa in tip fragments grown in the presence of different carbon sources. Possible mechanisms of the dependence of the tip growth parameters on the glucose deficiency are discussed.  相似文献

In vitro polyoma DNA synthesis: discontinuous chain growth   总被引:9,自引:0,他引:9  
Using an in vitro system for polyoma DNA synthesis from polyoma-infected mouse BALB/3T3 cells, we have shown that short pulses of radioactively labeled deoxynucleoside triphosphates are incorporated into viral replicative intermediates. Upon denaturation, the pulse-labeled replicative intermediates yield two size classes of growing DNA chains, namely a heterogeneous long class with S values up to unit viral DNA length (16 S) and a rather discrete short class of 5 S pieces. We have shown that these short fragments are involved as precursors in viral DNA chain elongation and that they can be chased into mature viral DNA. The fragments are found in replicative intermediates at all stages of replication and are therefore presumably not involved in specialized initiation or termination processes. Kinetic analysis of the appearance of radioactivity in short and long chains shows that initially approximately equal amounts are incorporated at a linear rate into the two classes. Subsequently, the rate of incorporation into long chains approximately doubles, while the amount of radioactivity in short chains reaches a plateau. This not only suggests that short chains are precursors to long chains, but that the synthesis of long chains occurs as a separate event and is not simply a result of joining of short fragments. Under the in vitro labeling conditions the time taken for radioactivity in short chains to reach a constant level can be used as a measure of the average lifetime of a 5 S piece. Our analysis indicates that there may be a considerable lag between the completion of a 5 S piece and its joining into longer DNA. Estimates of the self-annealing of the short chains showed approximately 20% self-complementarity. Thus, polyoma synthesis in vitro appears to proceed predominantly by a semi-discontinuous mechanism in which the nascent DNA on one side of the growing fork is elongated continuously, while on the other side of the fork DNA is synthesized discontinuously as 5 S fragments, which are subsequently joined. Both the short and the long chains are synthesized in the 5′ to 3′ direction.A fraction of the pulse-labeled material is found as free 3 to 5 S single-stranded DNA. These pieces are of both viral and cellular origin. A majority of them appear to be involved as precursors in DNA chain elongation.  相似文献
