A new approach to disinfection of water and sewage water, by oxidative destruction of microorganisms by photosensitization, is described. Samples of water and sewage water, to which an inoculum of fecal Escherichia coli had been added, were exposed to solar radiation in the presence of a dye sensitizer, under continuous aeration. The effects of the sensitizer methylene blue at concentrations of 0 to 10 mg/liter, different radiation times from 0 to 2 h, and sunlight intensities of 0, 68, and 2,030 muE/m2 per s were investigated. In laboratory-scale experiments, 1.3 X 10(9) coliforms in 100 ml of oxidation pond municipal sewage water containing 0.5 mg of methylene blue were destroyed in about 30 min. Similar results were obtained with nonchlorinated potable water. These results demonstrate the possibilities available for the disinfection of water and sewage water by this method.  相似文献

A number of chemical disinfectants were evaluated for their bactericidal and virucidal effectiveness in holding-tank sewage. It was found that the disinfection efficiencies of formaldehyde, benzalkonium chloride, cetylpyridinium chloride, and methylene blue were markedly improved if the pH of the sewage was raised from 8.0 to 10.5. When formaldehyde, benzalkonium chloride, and methylene blue were tested with either 2-week holding times with no sewage additions or 10-day holding times with daily sewage additions, disinfection effectiveness was maintained as long as the sewage pH was kept at 10.5 and the disinfectant concentration was kept at 100 mg/liter or more. Calcium hypochlorite, zinc sulfate, and phenol were found to be relatively ineffective disinfectants for holding-tank sewage.  相似文献

The combination of mutagenicity tests and selective extraction methodologies can be useful to indicate the possible classes of genotoxic organic contaminants in water samples. Treated and source water samples from two sites were analyzed: a river under the influence of an azo dye-processing plant discharge and a reservoir not directly impacted with industrial discharges, but contaminated with untreated domestic sewage. Organic extraction was performed in columns packed with XAD4 resin, that adsorbs a broad class of mutagenic compounds like polycyclic aromatic hydrocarbons (PAHs), arylamines, nitrocompounds, quinolines, antraquinones, etc., including the halogenated disinfection by-products; and with blue rayon that selectively adsorbs polycyclic planar structures. The organic extracts were tested for mutagenicity with the Salmonella assay using TA98 and TA100 strains and the potencies were compared. A protocol for cleaning the blue rayon fibers was developed and the efficiency of the reused fibers was analyzed with spiked samples. For the river water samples under the influence of the azo-type dye-processing plant, the mutagenicity was much higher for both blue rayon and XAD4 extracts when compared to the water from the reservoir not directly impacted with industrial discharges. For the drinking water samples, although both sites showed mutagenic responses with XAD4, only samples from the site under the influence of the industrial discharge showed mutagenic activity with the blue rayon extraction, suggesting the presence of polycyclic compounds in those samples. As expected, negative results were found with the blue rayon extracts of the drinking water collected from the reservoir not contaminated with industrial discharges. In this case, it appears that using the blue rayon to extract drinking water samples and comparing the results with the XAD resin extracts we were able to distinguish the mutagenicity caused by industrial contaminants from the halogenated disinfection by-products generated during water treatment.  相似文献

大城市郊区的小型居住区,将生活污水未经处理直接排入附近的绿地是比较常见的.这种直接排放虽然利用了有限水源,但对绿地土壤带来环境影响.为了研究这类排放对周边土壤环境的影响,选择了京郊一处接受附近生活污水直接排放的绿地,取其污水排放口附近的土壤样品为实验对象,以远离该点的土样为对照来研究生活污水排放对周边土壤环境质量的影响.模糊聚类分析结果表明:与对照点相比,生活污水排放提高了排放口附近土壤的养分含量,土壤pH值由8.9降低至7.32,有机质含量由1.7873%提高到5.9455%,钾含量由220 mg/kg提高到1230 mg/kg,磷含量由11.8 mg/kg提高到150.72mg/kg,全氮含量由0.1078%提高到0.5325%,重金属铅含量由52.87 mg/kg提高到129.53mg/kg、镉含量由0.7525mg/kg提高到2.5692mg/kg.铅含量达到了国家二级标准,而镉含量超过了国家三级标准,存在严重的土壤环境污染.  相似文献

TO determine the amount of K2Cr2O7 required to produce optimal Giemsa type staining, six 1 g amounts (corrected for dye content) of zinc methylene blue were oxidized with graded quantities of K2Cr2O7 to produce 4, 8, 12, 16, 20 and 24% conversion of methylene blue to azure B. These were heated with a blank control 15 minutes at 100 C in 60-65 ml 0.4 N HCI. cooled, and adjusted to 50 ml to give 20 mg original dye/ml. Aliquots were then diluted to 1% and stains were made by the “Wet Giemsa” technic (Lillie and Donaldson 1979) using 6 ml 1% polychrome methylene blue, 4 ml 1% cosin (corrected for dye content), 2 ml 0.1 M pH 6.3 phosphate buffer, 5 ml acetone, and 23 ml distilled water. The main is added last and methanol fixed blood films are stained immediately for 20-40 min.

For methylene blue supplied by MCB 12-H-29, optimal stains were obtained with preparations containing 20 and 24% conversion of methylene blue to azure B. With methylene blue supplied by Aldrich (080787), 16% conversion of methylene blue to azure B was optimal. Eosinates prepared from a low azure B/methylene blue preparation selected in this way give good stains when used as a Wright stain in 0.3% methanol solution. However, when the 600 mg eosinate solution in glycerol methanol is supplemented with 160 mg of the same azure B/methylene blue chloride the mixture fails to perform well. The HCI precipitation of the chloride apparently produces the zinc methylene blue chloride salt which is poorly soluble in alcohol. It appears necessary to have a zinc-free azure B/methylene blue chloride to supplement the probably zinc-free eosinate used in the Giemsa mixture.  相似文献

Procedure:Cut paraffin sections and float on a 45-50 C water bath; spread silicone-rubber adhesive (Clear Seal-General Electric) thinly and evenly over 2/3 of the slide; pick up the sections from the floatation water with the coated slide; dry for 1.5 hr at 25 C and at 60 C for 0.5 hr; deparaffinize, and hydrate to water. Place 150 mg of rhodamine B and 150 mg of methylene blue each in separate 100 ml beakers and add 80 ml of 10% HCl to each beaker. Bring both solutions to a boil on a hot plate in a fume hood; immerse tissue sections in the boiling rhodamine B exactly 2 min; rinse in a beaker of 10% HCl 5 sec; immerse in the boiling methylene blue exactly 0.5 min; rinse in distilled water; blot dry; and mount in a silicone-rubber medium (Glass and Ceramic Adhesive—Dow Corning Corp.). Hair shaft keratin stains red; inner root sheath keratin and keratogenous zone of the hair shaft, blue green; epidermal keratin remains unstained. Pilomatrixornas show foci of both red and blue green keratin; epidermal and hair sheath (“sebaceous”) cysts remain unstained.  相似文献

Starting from ancient reports that rare samples of methylene blue were apparently sufficiently contaminated with azures to give red plasmodial and red purple nuclear chromatin in Chenzinsky type methylene blue eosin stains, it was decided to determine how little azure B would suffice for such staining in methylene blue eosin stains. The traditional 1902 Giemsa had an azure:methylene blue: eosin ratio of about 6:3:6.3:10; Lillie's 1943 formula had a 5:7:10 ratio. In the current series of tests 5:7:10 (I), 4:8:10 (II), 3:9:10 (III), 2:10:10 (IV), 1:11:10 (V), and 0:12:10 (VI) were used. Malaria and blood stains were better than the standard 5:7:10 (I) in III, IV and II in that order. Normal and leukemic human blood, mouse blood with Plasmodium berghei, and monkey blood with the CDC strain of Pl. falciparum were used as test materials. The staining mixtures were made from highly purified samples of azure B and methylene blue. Staining mixtures contained 12 ml 0.1% thiazin dye, 10 ml 0.1% eosin, 2 ml each of glycerol, methanol and 0.1 M phosphate buffer pH 6.5, 3 ml acetone as accelerator, and distilled water to make 40 ml; staining times of 10-30 min were used.  相似文献

Aims: To evaluate the reduction of human norovirus (HuNoV) by chlorine disinfection under typical drinking water treatment conditions. Methods and Results: HuNoV, murine norovirus (MNV) and poliovirus type 1 (PV1) were inoculated into treated water before chlorination, collected from a drinking water treatment plant, and bench‐scale free chlorine disinfection experiments were performed for two initial free chlorine concentrations, 0·1 and 0·5 mg l?1. Inactivation of MNV reached more than 4 log10 after 120 and 0·5 min contact time to chlorine at the initial free chlorine concentrations of 0·1 and 0·5 mg l?1, respectively. Conclusions: MNV was inactivated faster than PV1, and there was no significant difference in the viral RNA reduction rate between HuNoV and MNV. The results suggest that appropriate water treatment process with chlorination can manage the risk of HuNoV infection via drinking water supply systems. Significance and Impact of the Study: The data obtained in this study would be useful for assessing or managing the risk of HuNoV infections from drinking water exposure.  相似文献

Upgrading sewage sludges for adsorbent preparation by different treatments   总被引:3,自引:0,他引:3  
Addressing the adequate management of sludges produced at sewage plants is becoming a fundamental need as a consequence of the high production volumes, both current and forecasted, of this byproduct. European waste-treatment policies consider reuse of sludges as one of the preferred actions; along those lines this study proposes using sewage sludges as adsorbents for pollutants contained in wastewaters. As potential adsorbents, sludges dried at 105 degrees C, dried and pyrolyzed, or dried and chemically activated were tried. As adsorbate, methylene blue was used in order to characterize the adsorption capacity of the different materials. Although surface areas corresponding to pyrolyzed and chemically activated sludges were around 80 and 390 m2/g, respectively, both these materials exhibited poor levels of methylene blue adsorption which may have been due to their high proportions of micropores. Sludges only dried, on the contrary, showed significant methylene blue adsorption capacities.  相似文献

A method is offered for he differential diagnosis of cancer cells. It depends on the use of methylene blue decolorized with sodium thiosulfate (denoted here HLM, i.e. “hyposulfite methylene blue”); this is prepared by dissolving 800 mg. sodium thiosulfate in 10 ml. of 0.1% aqueous methylene blue and adding 3-5 drops of dilute (1:3) HCl. Frozen sections are treated with this reagent for 2-3 minutes, rinsed with a large amount of distilled water, then stained 2-3 minutes with 0.05% aqueous acid fuchsin. Staining should be performed in a darkened room. If all due precautions are observed, normal tissue appears blue, malignant tissue red.  相似文献

In order to reduce the risks of Legionnaires' disease, caused by the bacterium Legionella pneumophila, disinfection of tap water systems contaminated with this bacterium is a necessity. This study investigates if electrochemical disinfection is able to eliminate such contamination. Hereto, water spiked with bacteria (10(4)CFU Escherichia coli or L. pneumophila/ml) was passed through an electrolysis cell (direct effect) or bacteria were added to tap water after passage through such disinfection unit (residual effect). The spiked tap water was completely disinfected, during passage through the electrolysis cell, even when only a residual free oxidant concentration of 0.07 mg/l is left (L. pneumophila). The residual effect leads to a complete eradication of cultivable E. coli, if after reaction time at least a free oxidant concentration of 0.08 mg/l is still present. Similar conditions reduce substantially L. pneumophila, but a complete killing is not realised.  相似文献

This article reports the role of indigenous iron-oxidizing microorganisms in bioleaching of heavy metals from anaerobically digested sewage sludge in presence of toxic dyes namely, methylene blue (MB), Remazol black B (RBB) and mixture of both the dyes (DM). To achieve this goal, different concentrations of dyes (0, 500, 10,000, 15,000 and 25,000 mg/L) were added to the sewage sludge (initial pH ~ 7) and subsequently, the solubilization of heavy metals (Cu, Ni, Zn and Cr) was measured at time intervals of 48 h for 16 days. The results showed that an increase in dye concentration adversely affected the population of indigenous iron-oxidizing microorganisms, thereby decreased the bioleaching of metals. The metal solubilization from sludge is more adversely affected in presence of RBB and DM than MB. In majority of cases, the maximum metal solubilization was recorded at dye concentration of 5,000 mg/L. Two factor analyses (ANOVA) suggests the impact of both factors: dye concentration and dye type on metal leaching rate in sludge system.  相似文献

Effects of radiation dose rate, electron energy and oxygen on disinfection of sewage sludge cake were studied to obtain technological information on disinfection by radiation. Sludge films were irradiated with electron beams at different voltages and various dose rates in oxygen-containing and oxygen-free atmospheres. Without oxygen, the surviving fraction of bacteria decreased with increasing irradiation dose, but depended little on the sludge film thickness. No effects of dose rate and energy change were found in the ranges from 20 kGy/h to 65 MGy/h and from 0.5 to 2.0 MeV when the film was thin enough to allow electron beam penetration. Similar results were obtained with γ-ray irradiation. In the oxygen-containing atmosphere, higher disinfection efficiency was obtained with thinner films and lower dose rates.  相似文献

Methylene blue has intrinsic antimalarial activity and it can act as a chloroquine sensitizer. In addition, methylene blue must be considered for preventing methemoglobinemia, a serious complication of malarial anemia. As an antiparasitic agent, methylene blue is pleiotropic: it interferes with hemoglobin and heme metabolism in digestive organelles, and it is a selective inhibitor of Plasmodium falciparum glutathione reductase. The latter effect results in glutathione depletion which sensitizes the parasite for chloroquine action. At the Centre de Recherche en Santé de Nouna in Burkina Faso, we study the combination of chloroquine with methylene blue (BlueCQ) as a possible medication for malaria in endemic regions. A pilot study with glucose-6-phosphate dehydrogenase-sufficient adult patients has been conducted recently.  相似文献

Out of 15 strains ofAzospirillum spp. isolated from the roots of different plants, only 4 (CY, M, CC, and AM) were able to grow autotrophically with H2 and CO2. All of them showed H2 uptake in the presence of oxygen or methylene blue and ribulose-1,5-bisphosphate carboxylase activity. Among the four strains, strain CC isolated from the roots ofCenchrus cilliaris showed maximum H2+O2 uptake (32.5 l/min. mg protein) as well as H2 uptake in the presence of methylene blue (41.4 l/min·mg protein) and also the maximum activity of ribulose-1,5-bisphosphate carboxylase (17 units [U]/g protein). The doubling time of this strain under autotrophic growth conditions and at low oxygen concentration (2.5%, vol/vol) was 10 h. At the same O2 concentration the maximal rates of H2+O2 uptake were reached. The distribution of hydrogenase activity among soluble and particulate protein fractions revealed that the hydrogenase ofAzospirillum strain CC is a membrane-bound enzyme. It showed cross-reaction with antibodies raised against the membrane-bound hydrogenase ofAlcaligenes eutrophus. The hydrogenase in intact cells and crude extracts reacted with methylene blue, phenazine methosulfate, and ferricyanide, but not with NAD or FMN. The specific hydrogenase activity, with methylene blue as an acceptor, was 5.71 U/mg protein in crude extract at 9.38 U/mg protein in the membrane suspension. Hydrogen evolution from reduced viologen dyes could not be demonstrated. The hydrogenase is oxygen sensitive and can be optimally stabilized by addition of dithionite to H2-gased samples.  相似文献

Although frequently less toxic than many colorless effluents, colored effluents are generally considered by the public as an indicator of pollution. The present investigation aimed at identifying the effectiveness of a local desert plant characteristic of Southwest Algeria and known as Salsolavermiculata, which was pyrolyzed and treated chemically with a 50% zinc chloride solution, to remove methylene blue and iodine. The natural plant adsorption capacities were respectively 23mg/g and 272mg/g for methylene blue and iodine. Corresponding results for the pyrolyzed plant uptakes were 53mg/g and 951mg/g, while those for the pyrolyzed plant, chemically treated and activated at 650 degrees C, were 130mg/g and 1178mg/g, respectively. In comparison, the standard Merck activated carbon capacities were 200mg/g for methylene blue and 950mg/g for iodine. Consequently, this low-cost local plant may also prove useful for the removal of large organic molecules as well as potential inorganic contaminants.  相似文献

The accumulation of methylene blue in native and damaged erythrocytes treated by cetyltrimethylammonium bromide at different initial concentrations (c0) of methylene blue and different volume ratios between external solution and cells (Vs/Vc) was studied. It was shown that at low methylene blue concentrations (c0 < 200 microM), the sorption of the dye by cells made the main contribution to its accumulation. As a result, the internal concentration of methylene blue exceeded manifold its external concentration and their ratio Q was 4-6 for native and 6-9 for damaged cells. As Vs/Vc decreased and especially as c0 increased, the diffusion of methylene blue inward the cells increased concentration gradient, and Q sharply fell to 0.9-1.0. The optimal values of c0 and Vs/Vc that provide the maximum sensitivity of Q to cell damage were determined. The advantages of using Q over other parameters of methylene blue accumulation were shown.  相似文献

Varying the parameters such as agitation time, dye concentration, adsorbent dose, pH and temperature carried out the potential feasibility of thermally activated coir pith carbon prepared from coconut husk for removal of methylene blue. Greater percentage of dye was removed with decrease in the initial concentration of dye and increase in amount of adsorbent used. Kinetic study showed that the adsorption of dye on coir pith carbon was a gradual process. Lagergren first-order, second-order, intra particle diffusion model and Bangham were used to fit the experimental data. Equilibrium isotherms were analysed by Langmuir, Freundlich, Dubnin-Radushkevich, and Tempkin isotherm. The adsorption capacity was found to be 5.87 mg/g by Langmuir isotherm for the particle size 250-500 microm. The equilibrium time was found to be 30 and 60 min for 10 and 20 mg/L and 100 min for 30, 40 mg/L dye concentrations, respectively. A maximum removal of 97% was obtained at natural pH 6.9 for an adsorbent dose of 100 mg/50 mL and 100% removal was obtained for an adsorbent dose of 600 mg/50 mL of 10 mg/L dye concentration. The pH effect and desorption studies suggest that chemisorption might be the major mode of the adsorption process. The change in entropy (DeltaS0) and heat of adsorption (DeltaH0) of coir pith carbon was estimated as 117.20 J/mol/K and 30.88 kJ/mol, respectively. The high negative value of change in Gibbs free energy indicates the feasible and spontaneous adsorption of methylene blue on coir pith carbon.  相似文献
