Summary Methods for the removal of exine from mature, ungerminatedLilium longiflorum pollen and release of intact gametophytes (sporoplasts) have been developed. These methods rely on the low temperature solvolytic activity of 4-methylmorpholine N-oxide (MMNO), which allows partial or complete detachment of exine from intine during subsequent washing procedures. These methods are: aqueous MMNO combined with cyclohexylamine (method I), aqueous MMNO at alkaline pH (method II), and aqueous MMNO containing a high Ca2+ concentration with added cellulysin and macerase (method III). Sporoplasts produced by methods I and II are most frequently completely separated from exine and, as shown by histochemical tests, enveloped by the intine layer. Selected enzyme activities in method II sporoplasts are measurable but, as indicated by other tests, considerable damage to the plasma membrane accompanies this treatment. Sporoplasts produced by melhod III largely remain attached to their ruptured exine layer and retain substantial biological competence in terms of extractable enzyme activities, membrane integrity, and respiration.Abbreviations MMNO 4-methylmorpholine N-oxide - SEM scanning electron microscope - TEM transmission electron microscope  相似文献

Summary Investigations were carried out to correlate pollen viability, assessed on the basis of a fluorochromatic reaction (FCR) test, with pollen vigor, assessed on the basis of the time taken for in vitro germination in pollen grains subjected to high humidity (>95% RH) and temperature (38 °C) or storage stress of Nicotiana tabacum, Agave sp., Tradescantia virginiana, and Iris sp. Both high RH and temperature, as well as storage stresses, affected pollen vigor before affecting pollen viability. The results are discussed in the light of available data on the viability and vigor of stressed pollen and of aged seeds. The need for consideration of pollen vigor, particularly in stored pollen, the inadequacy of the methods presently used, and some of the methods suitable to assess pollen vigor are elaborated.  相似文献

50% breaking point pressures of pollen grain walls of eleven species were determined. The breaking point pressures of most pollen grain walls are equivalent to those reported in the literature for other types of living cell walls such as bacterial spore coats, algal cell walls, mold sporophyte cells, and dicot suspension culture cells. The strongest pollen grain walls are two or three orders of magnitude stronger, however. Pollen grain walls are proportionately very thick in comparison to other types of cell walls. It is this thickness, not the construction or physical properties of the pollen grain wall, that most probably accounts for their strength.  相似文献

The sugar composition of pollen grain and pollen tube cell walls was studied for Camellia japonica, C. sasanqua, C. sinensis, Tulipa gesneriana and Lilium longiflorum. In all species, the main components of pollen grain walls were arabinose, galactose, glucose and uronic acid. On the other hand, the pollen tube walls consisted mostly of glucose. The pollen tube wall of C. japonica was fractionated into hemicellulose, α-cellulose and pectic substance fractions in yields of 61, 19 and 3 %, respectively. The hemicellulose fraction was composed essentially of glucose. The sugar composition of the pollen tube wall was not influenced by the nature of exogenously supplied sugars. Rapid growth of the pollen tube seemed to correlate with the synthesis of hemicellulosic glucan.  相似文献

Pollen histochemistry and pollen : ovule ratios in Zingiberaceae   总被引:7,自引:0,他引:7  
BACKGROUND AND AIMS: Pollen grains of 37 species from 11 genera in the family Zingiberaceae were examined to assess qualitatively starch or lipid contents; the pollen grain and ovule numbers per flower and pollen : ovule ratios were also counted and calculated. Pollen : ovule ratios were studied to reveal patterns of variation in the Zingiberaceae. METHODS: Freshly open flowers with dehiscing anthers were collected at random from plants growing in natural habitats or in botanical gardens. Presence of lipids or starch in pollen grains was tested by Sudan solution and IKI solution, respectively, and examined under a microscope. To estimate the pollen and ovule numbers per flower, one anther from each bud was carefully dissected and all pollen grains were counted; ovaries were carefully dissected out of each flower and counted. Whenever possible, at least 10-15 buds were used in the determination. KEY RESULTS: Thirty-three of all the 37 species examined had starchy pollen. Starch was not found in only four species and lipid was not found in only one species; among the four tribes in subfamily Zingiberoideae, all species of Zingibereae and Globbeae had pollen with no starch, Alpineae and Hedychieae had pollen with and without starch, whereas, all species of subfamily Costoideae had starchy pollen with abundant lipids. The mean pollen : ovule ratios in the members of the Zingiberaceae investigated range from 3.25 +/- 1.56 to 616.52 +/- 117.83. CONCLUSIONS: The pollen nutrition types seemed not related to mating systems. The pollen : ovule ratios in members of the Zingiberaceae with the same breeding system are noticeably lower than those recorded by previous authors. The lower pollen : ovule ratios in this family are presumed to be related to the highly efficient pollination systems, mediated by pollen which can be quite glutinous and the relatively large stigma area. In most of the Alpinia species the anaflexistylous flowers have much larger numbers of pollen grains and higher pollen : ovule ratios than the cataflexistylous flowers. There are significant differences in mean pollen grain numbers and pollen : ovule ratios between different life forms but ovule numbers are approximately the same.  相似文献

The impermeable callosic wall that surrounds germinating pollen grains and pollen tubes is removed by brief treatment with Zymolyase, an enzyme preparation that hydrolyzes linear glucose polymers with beta-1,3 linkages. Odors of pollen and whole flowers were compared in taxonomically unrelated species that offer pollen as the only food reward to pollinators. Volatiles were collected using headspace adsorption and analyzed by gas chromatography and mass spectrometry. The odor of pollen was found to be chemically distinct from the total flower odor, and this pollen-odor distinctness varied among the three species. In Papaver rhoeas (Papaveraceae), the contrast between pollen and whole-flower odors was most subtle, with differences observed only in the proportions of individual volatiles (almost exclusively aliphatic hydrocarbons). In Filipendula vulgaris (Rosaceae), pollen volatiles were fewer than in the flowers (comprising mainly benzenoids and fatty-acid derivatives) and their relative proportions produced an odor dominated by 2-heptadecanone that contrasted strikingly with the flower odor dominated by 2-phenyl ethanol. In Lupinus polyphyllus (Fabaceae), the pollen odor contained fewer volatiles and in differing proportions than the flower fragrance (comprising almost exclusively isoprenoids). The findings add to earlier chemical evidence of odor contrasts between pollen and other flower parts in two other species. Drawing on information from pollination studies of these various species, it is suggested that pollen odor is used by pollen-foraging insects both to discriminate between plant species and to assess reward availability in individual flowers, and that it might in addition serve a protective function against destructive flower-feeding insects and pathogens.  相似文献

Unlike most other ecosystems, many wetlands preserve an archive of their own history in their accumulating sediments. Reading this archive can be complex and challenging. This paper explores some of the challenges faced when trying to interpret the pollen record preserved in sediment archives from wetlands. It uses simulations of pollen dispersal and deposition in wetland scenarios to demonstrate that relatively simple models can replicate many of the features of the wetland pollen signal, and explores the application of these models to developing better understanding and interpretation of the palaeo-record.  相似文献

Induction of haploid plants from pollen grains on culture of anthers has been possible in a number of angiosperms but it is not yet understood why only a fraction of pollen responds to form haploids. Noteworthy in this connection are the recent experiments on anther culture of barley, tobacco and wheat, where it has been pointed out that pollen populations are basically dimorphic. Pollen capable of forming haploids occur in a low frequency, arc smaller, and different from the majority of pollen destined to form gametes. Particularly in tobacco it has been possible to increase the frequency of pollen capable of forming embryos by subjecting plants to low temperature prior to flowering, and to achieve differentiation of embryogenic pollen by subjecting young buds from such plants to a prolonged cold treatment. On selective isolation, from the rest of the pollen, the embryogenic pollen from such buds readily form embryos at high frequency on a simple mineral-sucrose medium. The embryogenic pollen are repressed for gametophytic differentiation and in culture they differentiate to produce embryos. These experiments provide evidence that only certain pollen are capable of forming haploids.  相似文献

The micromorphology and ultrastructure of in situ pollen from Cycadeoidea dacotensis are described from permineralized specimens collected from the Lower Cretaceous of North America. Pollen grains are ovoid and relatively small, averaging 25 μm in length and 12 μm in width. Grains are monosulcate with the exine typically invaginated in apertural regions. Exine ornamentation ranges from punctate to psilate. The exine averages 0.73 μm in thickness and is composed of a light-staining sexine and a dark-staining nexine. The sexine consists of a thin, homogeneous tectum, typically with a well-defined inner boundary, and a thicker granular infratectum. The infratectal granules are relatively uniform in size, however, variation occurs in the arrangement of granules. In some grains, the sexine appears homogeneous because there is little lacunal space between the individual granules. The granular infratectum is in direct contact with the underlying nexine. The nexine is uniform in thickness in both apertural and nonapertural regions, and it lacks lamellae throughout. Pollen morphology and ultrastructure are compared with those of the bennettitalean genus Leguminanthus and the dispersed genus Monosulcites. In addition, the fine structure of Cycadeoidea pollen is compared to that of the gymnosperm groups with which the Bennettitales are regarded to be most closely related, including Gnetales, Pentoxylales, and Eucommiidites-type pollen-producing plants.  相似文献

Two solvent systems for fully dissolving, and optionally derivatizing, finely ground plant cell wall material at room temperature are described: dimethylsulfoxide (DMSO) and tetrabutylammonium fluoride (TBAF) or N-methylimidazole (NMI). In situ acetylation produces acetylated cell walls (Ac-CWs) that are fully soluble in chloroform. Lignin structures tested remain fully intact. The dispersion of 13C-1H correlations afforded by two-dimensional (2D) nuclear magnetic resonance (NMR) experiments reveals the major lignin units, allowing the whole lignin fraction to be analyzed by high-resolution solution-state NMR methods for the first time. Non-degradative cell wall dissolution offers the potential to analyze polysaccharide components, and improve current cell wall analytical methods by using standard homogeneous solution-state chemistry.  相似文献

With regard to adaptation of green ash (Fraxinus pennsylvanica Marshall) to ecological conditions in Croatia, pollen germination and pollen tube length after 2, 4 and 6 hours were examined in vitro at 10, 15, 20 and 25°C during two years 2001 and 2002. Narrow leaved ash (F. angustifolia Vahl) pollen served as a control in 2002. The year, time and temperature, and the interaction between time and temperature were significant for both germination percentage and pollen tube length. Interactions year × temperature and year × time were significant for pollen tube length only. The highest germination percentage (17.86% in 2001 and 19.40% in 2002) of green ash pollen was at 15°C after 6 hours. The pollen tube length was greatest at 20°C (393.46 μm) in 2001 and 25°C (899.50 μm) in 2002 after 6 hours. Narrow leaved ash pollen had the highest germination percentage (19.22%) at 20°C after 6 hours and was significantly reduced at 25°C. The pollen tube length was greatest at 25°C (518.90 μm) after 6 hours. It can be concluded that green ash pollen has satisfactory germination in ecological conditions in Croatia and that the optimum temperature for pollen germination is higher than 20°C.  相似文献

Summary Mechanisms acting in pattern morphogenesis in the cell walls of two distant groups of plants, pollen of spermatophytes and diatoms, are compared in order to discriminate common principles from plant group- and wall material-specific features. The exinous wall in pollen is sequentially deposited on the exocellular side of the plasmalemma, while the siliceous wall in diatoms is formed intracellularly within an expanding silica deposition vesicle (SDV) which is attached to the internal face of the plasmalemma. Two levels of patterning occur in diatom and pollen walls: the overall pattern stabilises the wall mechanically and is apparently initiated in both groups by the parent cell, and a microtubule-dependent aperture and portula pattern created by the new mitotic (diatoms) or meiotic (pollen) cells. The parent wall in diatoms, and also the callosic wall in microspores, functions as anchor surfaces for transient, species-specific patterned adhesions of the plasmalemma to these walls, involved in pattern and shape creation. Patterned adhesion and exocytosis is blocked in pollen walls where the plasmalemma is shielded by the endoplasmic reticulum at the sites of the future apertures. In diatoms, wall patterning is uncoupled from the formation of a siliceous wall per se when the SDV and its wall is formed without contact to the the plasmalemma. Conversely, a blue-print pattern laid out in advance along the plasmalemma can be found in several diatoms. This highlights the key function of the plasmalemma and its associated membrane skeleton (fibrous lamina), and its orchestrated co-operation with elements of the radial filamentous cytoskeleton (actin?) in pattern formation. The role of microtubules during generation of the overall pattern may be primarily a transport and stabilizing function. Auxiliary organelles (spacer vesicles, endoplasmic reticulum, mitochondria) involved in diatoms for shaping the SDV, and a mechanism adhering and disconnecting this SDV together with spacer organelles in a species-specifically controlled sequence to and from the plasmalemma, are unnecessary for pollen wall patterning. Compositae exhibit some of the most complex and diverse pollen grains in flowering plants. This paper reviews the evolutionary and developmental origins of this diversity in pollen structure using recent models based on the behaviour of colloids and formation of micelles in the differentiating microspore glycocalyx and primexine. The developmental model is consistent with observations of structures recovered by pollen wall dissolution. Pollen wall diversity in Compositae is inferred to result from small changes in the glycocalyx, for example ionic concentration, which trigger the self-assembly of highly diverse structures. Whilst the fine details of exine substructure are, therefore, not under direct genetic control, it is likely that genes establish differences in the glycocalyx which define the conditions for self-assembly. Because the processes described here for Compositae can account for some of the most complex exine structures known, it is likely that they also operate in pollen walls with much simpler organisation.  相似文献

Based on the assumptions that pollen tube length is predetermined by provisions in the pollen and that it is a function of pistil length, I hypothesise that species with longer pistils will have larger pollen grains than species with shorter pistils, and that pistil length and pollen size will be positively correlated in a linear manner. To test this hypothesis, the relationship between pollen grain volume and pistil length was compared in 43 Asteraceae species from Argentina. A positive linear correlation was found between pollen volume and pistil length. This correlation remained significant even after potential effects of phylogenetic relatedness were removed. The maintenance of this correlation suggests that in Asteraceae the association between pistil length and pollen volume may reflect a functional rather than a phyletic relationship. In addition, the pistil length: pollen volume ratio (PPR) was analysed in relation to the phylogenetic position of the species. High values of PPR would imply a reduction of the male gametophyte in relation to the minimal volume that a pollen grain must have to grow and fertilise an ovule. Thus, the general pattern of pollen volume reduction in relation to pistil length previously found among many angiosperm families will be also present within a family, i.e., PPR values of derived Asteraceae would be higher than those of basal species. Results indicated that reduction of pollen volume in derived Asteraceae was three times greater than the concomitant shortening of pistil length. Consequently, PPR increased with the phylogenetic position of the taxa. This work supports the correlation between pistil and pollen characters previously found for other plant families and confirms the influence of post-pollination processes on pollen size evolution. Pollen monitoring has become a standard investigation method for researchers in several disciplines; among them are Quaternary palynologists, who conduct experiments in order to gain insights that will help to interpret the content of pollen in sediments. A review of the literature shows how these experiments diversified during the 1920s and 1930s with an array of different research questions, ranging from pollination biology to hay fever studies. Quaternary palynologists gained renewed interest with the possibility of radiocarbon dating late Quaternary sediments and obtaining accumulation rates. Also, the comprehensive model of pollen deposition and the pollen budget studies by H. Tauber encouraged researchers to conduct similar experiments using the same type of pollen trap, which became the main trapping device for Quaternary palynologists. The high precipitation in the tropics inspired the development of alternative designs. The equipment used to assess the pollen content in the air has evolved from simple gravity devices to different types of apparatus using a vacuum pump or revolving rods that collect the pollen on impact. Silicone impregnated filters exposed perpendicularly to the wind can also yield a volumetric assessment and have proven useful in areas with a low content of pollen in the air. The literature review is followed by a brief account of the developments which established the basis for the formation of a group of scientists monitoring the pollen deposition at a network of sites using standard pollen traps, the Pollen Monitoring Programme (PMP). Over the last 15 years the network has collected a large dataset, which is now available to answer a number of research questions. A summary of selected regions and environments, for which pollen monitoring results are available, is provided to serve as a complement to the investigations mentioned above and to provide an overview that may stimulate new research.  相似文献
