A study was made of the higher alcohols (fusel oils) produced during the Indonesian tapé ketan fermentation using Amylomyces rouxii as the principal mold, alone or in combination with yeasts belonging to genera commonly found in the tapé ketan fermentation (Endomycopsis, Candida, and Hansenula). Total fusel oils increased with length of fermentation. Fusel oils detected in the product distillate included isobutanol and isoamyl and active amyl alcohols. No n-propanol was detected. Isobutanol and isoamyl alcohols were formed in the largest amounts. A. rouxii alone produced nearly the same quantity of fusel oils (total production, 275 mg/liter at 192 h) as it did in combination with Endomycopsis burtonii (total production, 292 mg/liter at 192 h).A. rouxii and Endomycopsis fibuliger produced fusel oils totaling 72 mg/liter at 32 h and 558 mg/liter at 192 h. A. rouxii in combination with Candida yeasts produced somewhat more fusel oils, ranging from 590 to 618 mg/liter at 192 h. A. rouxii in combination with Hansenula yeasts produced the least fusel oils, totaling 143 to 248 mg/liter at 192 h. During the first 36 h, production of fusel oils was higher at 30 and 35°C than at 25°C. At 48 h fusel oil production was slightly higher at 30°C than at 35°C. Beyond 48 h, production of fusel oils was higher at 25°C. A. rouxii in combination with Hansenula anomala and Hansenula subpelliculosa produced considerable ethyl acetate, ranging from 145 to 199 mg/liter at 36 h and 354 to 369 mg/liter at 192 h.  相似文献

Aspergillus flavus ATCC 15517 produced up to 212 mg per liter of total aflatoxin in submerged culture in aerated (3,000, 6,000, 9,000, and 12,000 ml/min) and agitated medium in 14-liter fermentors with 10 liters of medium consisting of 2% yeast extract and 10% sucrose. Aflatoxin production increased with time. A maximum of 212 mg/liter was produced at 9,000 ml/min aeration, whereas the yield decreased substantially at the lower aeration rates. Two other strains of A. flavus synthesized aflatoxin in smaller quantities.  相似文献

Three strains of Caldariomyces fumago were immobilized in 4% k-carrageenan and tested for semicontinuous production of chloroperoxidase (CPO). Over an 80-day period, growing in defined medium, C. fumago strains CMI 89362 and ATCC 11925 produced enzyme concentrations of 99 and 71 mg/liter, respectively, during six production periods of 12 to 14 days, while C. fumago DAOM 137632 produced only 24 mg of CPO per liter during six growth periods of 10 days. CPO production was unaffected by various regimens of washing between transfers. Mycelial growth was primarily restricted to the head surface, and bead size increased linearly with time. Attempts to restrict growth but maintain CPO production were unsuccessful. Pigment production, fructose utilization, and pH change in the immobilized cell cultures compared closely with the growth characteristics of free cell cultures. By using an airlift tower fermentor with an external loop run with continuous medium replacement of 20 ml/h (D = 0.016), strain CMI 89362 in bead form produced CPO at 40 mg/liter for 11 days.  相似文献

The effectiveness of iron compounds on growth inhibition of Sphaerotilus species was compared. In this study, two strains of Sphaerotilus were tested with different iron concentrations in a synthetic sewage (S-medium) as formulated by Lackey and Wattie (Sewage Works J. 12:669-684, 1940). For both strains, >80% inhibition of the maximum respiration rate was obtained by the following levels of soluble iron concentrations at pH 6.0: iron citrate, 20 mg/liter as Fe; iron cysteine, 5 mg/liter as Fe; and ferrous sulfate, 10 mg/liter as Fe. At a pH of 6.7 with iron citrate (20 mg/liter as Fe), inhibition of both strains was in excess of 50%. Insoluble iron compounds, such as iron hydroxides and ferrous carbonate, were found to be much less effective than the soluble iron compounds as inhibitors of these two strains. Aged iron hydroxide (500 mg/liter as Fe) produced a 70% inhibition in the maximum respiration rate while fresh iron hydroxide (52 mg/liter as Fe) and ferrous carbonate (500 mg/liter as Fe) produced a 20% inhibition. Chemical analyses of the iron-inhibited Sphaerotilus strains showed a close relationship between the inhibition of the organism's growth and the amount of iron sorbed by the organism.  相似文献

The effect of methyl jasmonate (JA-Me) on the induction of gum was studied in relation to the action of ethylene in peach (Prunus persica Batsch cv. Benishimizu) shoots. JA-Me applied at concentrations of 0.1–2.5% (w/w) in lanolin paste to current growing or older shoots substantially induced gums 3 days after treatment. The amount of gums exuded increased depending on the dose of JA-Me. Ethephon (2-chloroethyl- phosphonic acid) at 1 or 2% (w/w) in lanolin induced gum and strongly enhanced the promoting effect of JA-Me on gum formation. JA-Me also induced anthocyanin accumulation in current growing shoots, but ethephon did not. Anthocyanin accumulation in response to JA-Me at a concentration of 10 mg/liter or higher was observed also in the cut shoots of peach. Ethephon (100 mg/liter) substantially inhibited anthocyanin accumulation induced by JA-Me. These facts suggest that JA-Me plays an important role in gum formation as well as ethylene and in anthocyanin accumulation and that these processes are not necessarily accompanied by each other in peach shoots. Shake flask and pH-controlled reactor tests were used to determine the mathematical parameters for a mixed-culture bacterial thiosalt treatment pond. Values determined were as follows: Km and Vmax (thiosulfate), 9.83 g/liter and 243.9 mg/liter per h, respectively; Ki (lead), 3.17 mg/liter; Ki (copper), 1.27 mg/liter; Q10 between 10 and 30°C, 1.95. From these parameters, the required bioxidation pond volume and residence time could be calculated. Soluble zinc (0.2 g/liter) and particulate mill products and by-products (0.25 g/liter) were not inhibitory. Correlation with an operating thiosalt biooxidation pond showed the parameters used to be valid for thiosalt concentrations up to at least 2 g/liter, lead concentrations of at least 10 mg/liter, and temperatures of >2°C.  相似文献

Summary Selaginella pilijera consistently displays two rows of microsporangia and two rows of megasporangia. It is therefore possible to detect in longitudinal sections of strobili at least three significant differences in development between both types of sporangia prior to meiosis: size of young sporangia, abortion or development of sporocytes based an RNA staining, and presence or absence of callose walls around sporocytes. The functional significance of these differences is discussed in relation to the heterosporous habit and higher plant sporogenesis.  相似文献

A recombinant protein expression system working at low temperatures is expected to be useful for the production of thermolabile proteins. We constructed a low-temperature expression system using an Antarctic cold-adapted bacterium, Shewanella sp. strain Ac10, as the host. We evaluated the promoters for proteins abundantly produced at 4°C in this bacterium to express foreign proteins. We used 27 promoters and a broad-host-range vector, pJRD215, to produce β-lactamase in Shewanella sp. strain Ac10. The maximum yield was obtained when the promoter for putative alkyl hydroperoxide reductase (AhpC) was used and the recombinant cells were grown to late stationary phase. The yield was 91 mg/liter of culture at 4°C and 139 mg/liter of culture at 18°C. We used this system to produce putative peptidases, PepF, LAP, and PepQ, and a putative glucosidase, BglA, from a psychrophilic bacterium, Desulfotalea psychrophila DSM12343. We obtained 48, 7.1, 28, and 5.4 mg/liter of culture of these proteins, respectively, in a soluble fraction. The amounts of PepF and PepQ produced by this system were greater than those produced by the Escherichia coli T7 promoter system.  相似文献

Chemically synthesized 4-hydroxybenzoate (4HBA) is widely used in the chemical and electrical industries as a material for producing polymers such as those of the liquid crystal type. Its alkyl esters, called parabens, have been the most widely used preservatives by the food and cosmetic industries. We report here for the first time a microorganism, a marine bacterium, which biosynthesizes these petrochemical products. The marine bacterial strain, A4B-17, which was found to belong to the genus Microbulbifer on the basis of its rRNA and gyrB sequences, was isolated from an ascidian in the coastal waters of Palau. Strain A4B-17 was, surprisingly, found to produce 10 mg/liter of 4HBA, together with its butyl (24 mg/liter), heptyl (0.4 mg/liter), and nonyl (6 mg/liter) esters. We therefore characterized 23 other marine bacteria belonging to the genus Microbulbifer, which our institute had previously isolated from various marine environments, and found that these bacteria also produced 4HBA, although with low production levels (less than one-fifth of that produced by A4B-17). We also show that the alkyl esters of 4HBA produced by strain A4B-17 were effective in preventing the growth of yeasts, molds, and gram-positive bacteria.  相似文献

Brucella abortus is a facultative intracellular gram-negative bacterial pathogen that infects humans and animals by entry mainly through the digestive tract. B. abortus causes abortion in pregnant cattle and undulant fever in humans. The immunogenic B. abortus ribosomal protein L7/L12 is a promising candidate antigen for the development of oral live vaccines against brucellosis, using food-grade lactic acid bacteria (LAB) as a carrier. The L7/L12 gene was expressed in Lactococcus lactis, the model LAB, under the nisin-inducible promoter. Using different signals, L7/L12 was produced in cytoplasmic, cell-wall-anchored, and secreted forms. Cytoplasmic production of L7/L12 gave a low yield, estimated at 0.5 mg/liter. Interestingly, a secretable form of this normally cytoplasmic protein via fusion with a signal peptide resulted in increased yield of L7/L12 to 3 mg/liter; secretion efficiency (SE) was 35%. A fusion between the mature moiety of the staphylococcal nuclease (Nuc) and L7/L12 further increased yield to 8 mg/liter. Fusion with a synthetic propeptide (LEISSTCDA) previously described as an enhancer for heterologous protein secretion in L. lactis (Y. Le Loir, A. Gruss, S. D. Ehrlich, and P. Langella, J. Bacteriol. 180:1895-1903, 1998) raised the yield to 8 mg/liter and SE to 50%. A surface-anchored L7/L12 form in L. lactis was obtained by fusing the cell wall anchor of Streptococcus pyogenes M6 protein to the C-terminal end of L7/L12. The fusions described allow the production and targeting of L7/L12 in three different locations in L. lactis. This is the first example of a B. abortus antigen produced in a food-grade bacterium and opens new perspectives for alternative vaccine strategies against brucellosis.  相似文献

The effects of temperature, initial pH, and the concentrations of ammonium, phosphate, and heavy metals on the oxidation of thiosalts by an authentic strain of Thiobacillus thiooxidans (ATCC 8085) and by a mixed culture isolated from a base metal-processing mill effluent pond were studied. The optimum temperature was 30°C and the optimum initial pH was 3.75 for both cultures using thiosulfate and for the mixed culture using tetrathionate. T. thiooxidans ATCC 8085 did not oxidize tetrathionate. For a thiosalt concentration of 2,000 ppm (2,000 mg/liter), maximal rates of destruction occurred at concentrations of ammonium ion above 2 mg/liter and in the presence of 1 mg of phosphate per liter. Under optimal conditions, the rate of thiosulfate oxidation by the pure culture was 55 ± 3 mg/liter per h; the mixed culture oxidized thiosulfate at the rate of 40 ± 1 mg/liter per h and tetrathionate at the rate of 50 ± 2 mg/liter per h. Metal ions caused normal inhibition kinetics in the oxidation of thiosulfate by T. thiooxidans ATCC 8085. Ki values were calculated for cadmium (16 mg/liter), copper (0.46 mg/liter), lead (2 mg/liter), silver (3.1 mg/liter), and zinc (33 mg/liter). Only a slight additive effect was apparent in the presence of all of these metal ions. The mixed culture of thiosalt-oxidizing bacteria was less sensitive to heavy metal inhibition; the order of inhibition of thiosulfate oxidation was Cd < Zn < Pb < Ag < Cu, and that of tetrathionate oxidation was Zn < Cd < Pb < Ag < Cu.  相似文献

Summary Mature seeds of plains Old World bluestem [Bothriochloa ischaemum (L.) Keng.] were used to initiate suspension cultures. The medium contained the major and minor minerals of Murashige and Skoog, Gamborg's B-5 vitamins, 30 g/liter sucrose, and 3 mg/liter 2,4-dichlorophenoxyacetic acid with or without 12 mM proline at pH 5.5. Cultures contained both embryogenic and nonembryogenic (NE) cells. Suspensions that had been filtered through a 40-mesh sieve were plated out on medium with 6 g/liter agar. Two-to-three weeks later, clumps that formed in suspension cultures that had been filtered previously were removed by filtration through a 40-mesh sieve and plated out on agar medium. Colonies were rated on the basis of surface area. of the total area of colonies formed from plated suspensions 70.9% were embryogenic, 19.8% were NE, and 9.3% were mixed colonies. Of the total area from plated clumps, 57.1% were E, 12.9% were NE, and 30% were mixed colonies. Embryoid maturation and germination was accomplished by transferring E or mixed colonies to MS medium with 1 mg/liter zeatin (mixted isomers). Rooting was completed on half-strength basal MS medium. Over 90% of plantlets survived transfer to the greenhouse and 95% of them survived transfer to the field. We reported previously on the function of acyl coenzyme A (acyl-CoA) oxidase isozymes in the yeast Yarrowia lipolytica by investigating strains disrupted in one or several acyl-CoA oxidase-encoding genes (POX1 through POX5) (H. Wang et al., J. Bacteriol. 181:5140–5148, 1999). Here, these mutants were studied for lactone production. Monodisrupted strains produced similar levels of lactone as the wild-type strain (50 mg/liter) except for Δpox3, which produced 220 mg of γ-decalactone per liter after 24 h. The Δpox2 Δpox3 double-disrupted strain, although slightly affected in growth, produced about 150 mg of lactone per liter, indicating that Aox2p was not essential for the biotransformation. The Δpox2 Δpox3 Δpox5 triple-disrupted strain produced and consumed lactone very slowly. On the contrary, the Δpox2 Δpox3 Δpox4 Δpox5 multidisrupted strain did not grow or biotransform methyl ricinoleate into γ-decalactone, demonstrating that Aox4p is essential for the biotransformation.  相似文献

Three selective enrichment broths and four selective agar media were evaluated for their ability to support the growth of Pseudomonas pseudomallei both at 35°C and at ambient temperature (range, 20 to 32°C; mean, 25°C). Colony counts of 50 strains of P. pseudomallei and recovery studies with 1 soil strain in 60 simulated soil samples demonstrated that enrichment with Trypticase soy broth incorporating 5 mg of crystal violet per liter and 20 mg of colistin per liter (CVCB) and subculture to Ashdown medium supported the growth of all 50 strains and produced the highest recovery rates with the greatest suppression of other soil flora. An enrichment broth of MacConkey broth (purple) incorporating 10 mg of crystal violet per liter, 5 mg of bromcresol purple per liter, 25 mg of gentamicin per liter, and 650 mg of streptomycin per liter showed greater suppression of soil bacteria than CVCB, but it failed to support the growth of three strains of P. pseudomallei. Recovery rates were essentially the same irrespective of whether the soil samples were incubated at 35°C or at ambient temperature, provided cultures were incubated in protected shade for an extended period. This is an important feature for field work in large-scale epidemiological surveys in which resources are limited.  相似文献

HD-187, a new isolate of Bacillus thuringiensis serotype 5 (5a,5b), produced higher yields of δ-endotoxin than any other isolate previously tested at the laboratories of the Agricultural Research Service of the U.S. Department of Agriculture at Brownsville, Texas. Recoveries of as high as 2,000 × 106 international units (2,000 mIU)/liter of harvested beer were achieved from fermentations of HD-187, giving products with potencies as high as 200 × 103 (200 kIU)/mg. These contrasted with recoveries of about 200 mIU/liter and products with potencies of about 15 kIU/mg obtained in fermentations of HD-1, the strain of B. thuringiensis serotype 3 (3a,3b), that is used in most commercial production of the δ-endotoxin.  相似文献

Seeds of Cattleya aurantiaca (Orchidaceae) were germinated and grown aseptically on Knudson C medium containing 2.5, 5, 10, 20, and 50 mg l-1 of 2-chloroethylphosphonic acid (Ethephon), ethylphosphonic acid, or the inorganic acid moiety of both, phosphosphorous (phosphonic) acid. 2-Chloroethylphosphonic acid, an ethylene precursor, reduced leaf length at 5–50 mg l-1 proportional to increasing concentrations. Seedlings produced fewer leaves within the range of 20–50 mg l-1 Ethephon compared to lower concentrations (2.5–10 mg l-1). Germination was not affected adversely. Cultures containing the homologous, but not ethylene-generating ethylphosphonic acid in the same concentrations brought about similar but generally less severe reductions in leaf length and number. Phosphorous acid at the same concentration levels did not reduce the number and length of leaves. Our observation suggests that the intact organic phosphonates contribute to the inhibition of leaf growth caused by ethylene, which is released slowly from Ethephon in the culture medium and/or plant tissue, following uptake.  相似文献

Rubratoxin B can be produced in a semisynthetic medium by Penicillium rubrum under varying environmental and nutritional conditions. Maximum production (552.0 mg/500 ml) was obtained with P. rubrum NRRL A-11785 grown in stationary cultures of Mosseray's simplified Raulin solution supplemented with 2.5% malt extract broth at ambient temperature. Zinc is required at levels of at least 0.4 mg per liter. In the absence of iron sulfate, there was a 50-fold reduction in rubratoxin B production but not in growth. No toxin was produced by this isolate in 5- or 7-liter fermentors.  相似文献

The recalcitrance of xenobiotics may be caused by an absence of transforming enzymes or by their inability to enter microbial cells. A nondestructive method for differentiating between these two possibilities is described. The solid n-alkanes octadecane (C18) and hexatriacontane (C36) were encapsulated into phosphatidylcholine bilayers (liposomes). The uptake and metabolism rates of encapsulated and unencapsulated substrates were then compared. During 1 h at 25°C, a Pseudomonas isolate took up 1.3% of radiolabeled and unencapsulated C18 (solid state) versus 23.5% of labeled and encapsulated C18. Growth at 25°C occurred with an apparent ks of 2453 ± 148 mg/liter. Liposome encapsulation decreased this Ks to 60 ± 12 mg/liter. At 34°C, growth on C18 (liquid state) occurred with an apparent Ks of 819 ± 83 mg/liter and on the readily available carbon source succinate, Ks values were 80 ± 10 and 13 ± 7 mg/liter at 25 and 34°C, respectively. At 25°C, the isolate grew on C36 with an apparent Ks of 2,698 ± 831 mg/liter. Liposome encapsulation decreased the Ks more than 60-fold to 41 ± 7 mg/liter, resulting in the complete utilization of 400 mg of C36 per liter in 16 h. Since controls excluded the metabolic utilization of phosphatidylcholine, the results clearly identify transport limitation as the cause for C36 recalcitrance.  相似文献

Fourteen monosporal isolates of Hirsutella thompsonii grew vegetatively in various liquid media producing typical phialidic-like conidiophores. Hirsutella thompsonii var. synnematosa from Ivory Coast (HtIC) was the only pathotype which produced true conidia in submerged culture. HtIC began producing conidia after 3 days incubation reaching a peak ranging from 6.8 × 105 to 9.7 × 107 conidia/ml between 6 and 11 days. A concentration of 10 g/liter of corn steep liquor and 0.2% Tween 80 were essential for maximum conidiation. Submerged conidia had a smooth but somewhat rugose conidial walls, whereas aerially formed conidia were distinctly verrucose. Germination of submerged conidia ranged from 5.2 to 12.9% and were virulent causing 32.5% infection to adult citrus rust mites when sprayed on citrus foliage at 1.2 × 109 conidia/ml.  相似文献
