Polyadenylated and nonpolyadenylated mRNa from the red algae Cyanidium caldarium Geitler and Porphyridium aerugineum Geitler were translated in a cell-free system. The α-and β-subunits of phycocyanin and allophycocyanin and also both subunits of ribulose-1,5-bisphosphate carboxylase-oxygenase (RuBPCase) were identified with the help of specific antibodies as translation products of non-polyadenylated mRNA. Both subunits of RuBPCase were synthesized with molecular weights in the range of the mature forms. This is in contrast to the findings with green algae and higher plants where the small subunits of RuBPCase are always made by polyadenylated mRNA in the form of a larger precursor molecule. We discuss our findings with regard to the systematic position of the algae used and the evolution of plastids.  相似文献

The suitability was evaluated of MNNG as a mutagen inducing increased frequencies of mutations in the cell populations of three strains of chlorococcal algae for the purposes of selection. MNNG has proved to be highly toxic to those algae as it produces severe physiological responses of the affected cells. The mutagenic effect of MNNG was relatively small in comparison with the recorded toxic effect. From these results it has been concluded that in reverse to NEU, MNNG can hardly be applied with such good an effect in the mutation breeding of chlorococcal algae that are suitable for mass cultivation.  相似文献

The blue-green algaPlectonema boryanum was chosen for a study of the properties of rRNA of procaryotic organisms. The process of rRNA species maturation was studied by labelling RNA with the isotope 32P.23 S and 16 S rRNA molecules each have their own precursors. Molecules with molecular weights of 1.65 X 106 and 1.24xlO6 were identified as precursors of23 S rRNA, while the molecules 0.87 X 106; 0.78 X 106 and 0.68 X 106 were found to be precursors of the 16 S rRNA. No common polycistronic precursor could be found. Experiments done with algae cultivated in light and in the dark confirmed the fact, that the rate of rRNA synthesis is reduced in the dark, without however stopping completely.  相似文献

We investigated concentrations of monomethylmercury (MMHg) at the base of benthic food webs in six lakes from polar desert (biologically poor and low annual precipitation) on Cornwallis Island (Nunavut, Canada, ~75°N latitude). Anthropogenic mercury emissions reach the Arctic by long-range atmospheric transport, and information is lacking on processes controlling MMHg entry into these simple lake food webs, despite their importance in determining transfer to lake-dwelling Arctic char. We examined the influences of diet (using carbon and nitrogen stable isotopes), water depth, and taxonomic composition on MMHg bioaccumulation in benthic invertebrates (Chironomidae and Trichoptera). We also estimated MMHg biomagnification between benthic algae and invertebrates. Similar MMHg concentrations of chironomid larvae in nearshore and offshore zones suggest that benthic MMHg exposure was homogeneous within the lakes. Chironomid δ13C values were also similar in both depth zones, suggesting that diet items with highly negative δ13C, specifically methanogenic bacteria and planktonic organic matter, were not important food (and therefore mercury) sources for profundal larvae. MMHg concentrations were significantly different among two subfamilies of chironomids (Diamesinae, Chironominae) and Trichoptera. Higher MMHg concentrations in Diamesinae were likely related to predation on other chironomids. We found high MMHg biomagnification between benthic algae and chironomid larvae compared with literature estimates for aquatic ecosystems at lower latitudes; thus, benthic processes may affect the sensitivity of polar desert lakes to mercury. Information on benthic MMHg exposure is important for evaluating and tracking impacts of atmospheric mercury deposition and environmental change in this remote High Arctic environment.  相似文献

比较河流浮游藻类和着生藻类群落的时空格局及其与环境因子关系的差异,有助于了解两类藻的区别与联系。然而,目前这方面的研究还不多。基于2019年秋季和2020年夏季金沙江上段干流17个样点藻类及水体理化指标的调查数据,分析了不同季节浮游藻类和着生藻类群落结构及其主要环境驱动因子,比较了两类藻的多样性格局及其与环境关系的异同。结果发现,调查河段的浮游藻类和着生藻类均以硅藻为主,其中浮游藻类以极小曲壳藻(Achnanthes minutissima)、钝脆杆藻(Fragilaria capucina)、适中舟形藻(Navicula accomoda)为主要优势种,着生藻类以极小曲壳藻(Achnanthes minutissima)、扁圆卵形藻(Cocconeis placentula)、橄榄绿色异极藻(Gomphonema olivaceum)为主要优势种。浮游藻类和着生藻类秋季平均密度分别为:2.41×10~5个/L、9.43×10~3个/cm~2,均明显高于夏季的平均密度(4.84×10~4个/L、4.84×10~3个/cm~2)。两类藻的群落格局表现出明显的季节变化,但只有着生藻分类单元...  相似文献

  • 1 Recent laboratory studies demonstrate that periphytic algae growing on the sediment surface reduce nutrient availability in the overlying water. Consequently, periphytic algae may competitively reduce growth of phytoplankton.
  • 2 The aim of this study was to quantify the competitive impact of sediment-attached periphytic algae on phytoplankton in the presence of all other factors simultaneously affecting nutrient dynamics in natural systems.
  • 3 In enclosure experiments, performed in three lakes of different productivity, the periphytic algal biomass was manipulated. When compared to enclosures with high biomass of periphytic algae, those with reduced biomass showed an increase in total phosphorus concentration in the water of 32–44%. Extrapolation of the experimental results to whole lakes predicts an increase in original total phosphorus concentration of between 1.5% and 8.0%. According to existing regressions between total phosphorus and phytoplankton chlorophyll, the potential increase in original phytoplankton biomass will be between 2.5% and 12.6%.
  • 4 With respect to the shallow parts of lakes, my results support the conclusions revealed from laboratory studies that periphytic algae have a significant impact on the phosphorus concentration in the overlying water. However, when considering whole-lake dynamics, the competitive impact of periphytic algae on phytoplankton biomass development is probably of minor importance.
  • 5 Rather, the main competitive advantage of growing on the sediment surface, compared to in the water, may be the exclusive access to nutrients in the sediment.

An aquatic biological survey was conducted in 1979–1980 to determine the effects of drainage from an active coal strip-mine on Trout Creek, in northwest Colorado, U.S.A. Sampling was conducted over four seasons at four stations for periphyton, benthic invertebrates and fish. Periphyton in Trout Creek changed in the relative abundance of algae divisions in no apparent relation to mining. Diatoms were the predominant division at all sites. Golden-brown algae were abundant in spring at the stations upstream and adjacent to the mine. Blue-green algae were relatively important at stations upstream and downstream of the mine in winter. Benthic invertebrates exhibited a progressive increase in density, biomass and number of taxa from the upstream station to the downstream station. Shannon-Wiener diversity index for bethic invertebrates decreased slightly downstream of mine drainage but remained indicative of a clean water community. Aquatic insects (especially Trichoptera) were the predominant invertebrates at all stations. Analysis of functional groups of benthic invertebrates revealed increased importance of collector species at the lower sites while shredders were most important upstream of the mine. Unlike the invertebrates, fish exhibited slightly lower biomass at the station adjacent to the mine. The decrease was due to fewer salmonids. However, salmonid density and biomass increased substantially at the station just downstream of the mine. Non-game species (suckers and minnows) increased in numbers downstream and were most abundant at the lowest station. This coal strip-mine had little discernable adverse effects on the periphyton and invertebrates of Trout Creek. Fish populations did not appear to be significantly affected by the mine. Apparently, the presence of settling ponds and a buffer zone of unmined land between the mine and the stream helped to minimize adverse effects.  相似文献

Nitrate reductase of the salt tolerant alga Dunaliella parva, in contrast to that of most green algae, can use NADPH as well as NADH as electron donor. Extracts of cells contained various amounts of latent nitrate reductase. The latent enzyme could be activated at 45°C but only in the presence of flavine adenine dinucleotide. The heat activated enzyme did not require flavine adenine dinucleotide for activity and was fully active with NADH, NADPH or reduced flavine mononucleotide as electron donors.  相似文献

One hundred and eighteen algal isolates comprising seven classes were obtained from a range of sites from polluted rivers running through Cu or Zn mining regions, and from unpolluted rivers. All the isolates were tested for photosynthetic activity when exposed to Cu, Cd or Zn. The tolerance levels of Bacillariophyceae, Charophyceae, Cyanophyceae and Chlorophyceae to Cu showed significant positive correlations with Cu concentrations in the field. However the distribution of metal sensitivities of the algae from the sites with the same metal concentration was broad. Both Bacillariophyceae and Charophyceae had a number of strains whose sensitivity to Cu differed more widely in relation to Cu levels in the environment than Cyanophyceae and Chlorophyceae. Cyanophyceae were sensitive to all three metals, whether or not isolates were obtained from polluted sites, whereas Chlorophyceae tended to have high tolerance even in isolates from unpolluted sites. For Cd and Zn the correlation between tolerance levels and concentrations in the field was not so clear as for Cu. The occurrence of Cu tolerance was shown in 4 diatom species and one Charophyceae, whereas metal resistance occurred in some Chlorophyceae. Cu-tolerant isolates tended also to be Zn-tolerant in Bacillariophyceae, and Cd-resistant isolates tended also to be Zn-resistant in Chlorophyceae.  相似文献

Despite certain environmental advantages over fossil diesel, land crop-derived biodiesels may not satisfy the increasing worldwide demand for transportation fuels. As an abundant photosynthesizer, algae could be an adequate surrogate for biodiesel production. Nevertheless, high production costs, scarce selected species, and inaccurate assumptions about production yields represent industrial uncertainties. In this study, a reliable approach to analyzing algal biodiesel production has been developed based on species-to-species variations in oil productivity and quality. This approach compares biodiesels from Chlorophyta strains with land crop feedstock according to (i) potential yields, (ii) oil quality, and (iii) compliance with biodiesel quality standards. Algal yields were assessed by (i) extrapolating the strain-specific laboratory results to commercial-scale growth systems; (ii) converting volumetric to areal biomass productivity; and (iii) estimating oil yields for each strain, as the product of their projected areal biomass productivity for each growth system, and the oil percentage in biomass as determined in the laboratory. Biodiesel fuel properties were estimated by using fatty acid methyl ester profile predictive models. The Chlorophyta strains in this study provided annual oil yields that were generally higher than those of land crops by one order of magnitude. Six strains yielding more than 40 mg oil l?1 day?1 were identified as adequate for sustaining biodiesel production. Trebouxiophyceae algae were the most productive. Critical biodiesel parameters from both feedstock types suggest that most microalgae-derived biodiesels meet international fuel quality standards with better values than those of land crops. Because some of the highly productive feedstock does not simultaneously meet all the standards for a high quality biodiesel, optimization solutions are discussed.  相似文献

The embryonic and postembryonic development times, time interval between clutches, and longevity of two common cyclopoid copepods — Eucyclops serrulatus and Cyclops strenuus — were studied at constant temperatures (5°, 10°, 15°, 20° and 25 °C), using algae and protozoans as food. Both, E. serrulatus and C. strenuus showed a considerable temperature tolerance and adaptability. Embryonic development times were similar in both species but postembryonic development times were shorter in E. serrulatus. Time intervals between successive clutches were relatively variable at all temperatures in both species. Longevity was shorter in E. serrulatus than in C. strenuus. The embryonic and postembryonic development of E. serrulatus was short relative to that of littoral copepods; the development of C. strenuus was similar or shorter than that of other species of the genus Cyclops.  相似文献

  1. Growth requirements of blue-green algae containing only the c-phycocyanin + chlorophyll a pigment system have been studied under blue light (380–540 nm) which approximates light conditions existing in subsurface waters in nature.

While a few species were capable of very slow photosynthetic growth on minimal medium with NO3 - as nitrogen source, most species were dependent on organic compounds for comparable growth under this condition. Some organisms did quite well with only Casamino Acids as a supplement, others did well with only glucose. One species, Agmenellum quadruplicatum strain PR-6, grew only when glucose and Casamino Acids were supplied simultaneously.

Interactions between scleractinian corals and benthic algae can be an important process structuring reef communities, yet interaction dynamics are not fixed and may be influenced by abiotic factors such as sedimentation, a process often underlying reef degradation. However, rates of sedimentation and the effects of trapped sediments may be influenced by water flow. The first goal of this study was to quantify gradients in sedimentation and flow along fringing and back reefs of the north shore of Moorea, French Polynesia, and determine whether such gradients correlate with changes in the frequency and outcomes of massive Porites–algal turf interactions. On the back reef, the frequency of Porites–algal turf interactions and the competitive success of algal turfs increased significantly with decreasing flow. Sedimentation, however, was not a significant driver of the observed patterns. Along fringing reefs, in the absence of a flow gradient, it was the gradient in sedimentation that best explained spatial variation in Porites–algal turf interaction frequencies and the competitive success of algal turfs. The second goal was to quantify the separate and combined effects of flow and sedimentation on Porites–algal turf interactions in a laboratory setting. The combined effects of low flow and sedimentation significantly increased the area of Porites tissue damaged when in contact with algal turf, while high flow attenuated the negative effects of sedimentation. Together, these results implicate flow and sedimentation as important drivers of biological interactions between massive Porites and algal turf.  相似文献

  • 1 The resistance and resilience of littoral zone communities to sedimentation will depend both on the extent to which sediment deposition affects productivity, and on interactions within the communities. A series of hypotheses were set up and tested to examine interactions and feedback mechanisms among deposited sediments, periphyton, macrophytes and grazers in a large oligotrophic lake subject to fluctuating sediment loadings.
  • 2 Although sediments incorporated into periphyton reduced light availability to macrophytes, periphytic algae were generally the dominant light absorbing component under natural conditions. When grazers were absent, both sediments incorporated in the periphyton and periphytic algal densities increased, and both were then important in reducing light available to macrophytes.
  • 3 Grazing rate and assimilation efficiency for the dominant grazer, the prosobranch gastropod Potamopyrgus antipodarum, increased with increasing sediment content under natural lake conditions to reach a maximum at 10 mg sediment cm?2.

A green algal phenol oxidase was firstly purified, confirmed to be a laccase, and a hetero-oligomeric quaternary structure is suggested. The operation of a laccase-mediator system is firstly described in algae. Laccases (EC 1.10.3.2) catalyze the oxidation of a multitude of aromatic substrates. They are well known in higher plants and fungi, while their presence in green algae appears uncertain. Extracellular laccase-like enzyme activity has previously been described in culture supernatants of the green soil alga Tetracystis aeria [Otto et al. in Arch Microbiol 192:759–768, (2010)]. As reported herein, the T. aeria enzyme was purified 120-fold by employing a combination of anion exchange and size exclusion chromatography. The purified enzyme was confirmed to be a laccase according to its substrate specificity. It oxidizes 2,2′-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS), syringaldazine, and 2,6-dimethoxyphenol (pH optima of pH ≤2.5, 7.0, and 6.5; K m values of 28.8, 40.5, and 1,830 µM; respectively), but not l-tyrosine or Fe2+. ABTS is by far the most efficient substrate. Two polypeptides, A (~110 kDa) and B (71 kDa), were co-purified by the applied procedure, both being highly N-glycosylated (≥~53 and ≥27 %, respectively). As suggested by various gel electrophoretic analyses, the native enzyme (apparent molecular mass of ~220 kDa) most probably is a hetero-oligomer with the composition AB 2 , wherein A is the catalytic subunit and B forms a disulfide-linked homo-dimer B 2 . The decolorization of anthraquinone (Acid Blue 62 and Remazol Brilliant Blue R) and diazo dyes (Reactive Black 5) was studied in the presence of redox-mediating compounds (ABTS and syringaldehyde), demonstrating the operation of the laccase-mediator system in algae for the first time. Thus, laccases from green algae may participate in the biotransformation of a wide spectrum of natural and xenobiotic compounds.  相似文献

Recent studies have evaluated the role of brain-derived neurotrophic factor (BDNF) in mood disorders; however, little is known about alterations in nerve growth factor (NGF) and glial cell line-derived neurotrophic factor (GDNF). The aim of this study was to evaluate differences among serum neurotrophic factors (BDNF, NGF and GDNF) in depressed patients and healthy controls and to verify the association between serum neurotrophic levels and clinical characteristics in a young, depressed population stratified by gender. This is a cross-sectional study with depressed patients and population controls 18–29 years of age. The concentrations of neurotrophic factors were determined by the ELISA method. The diagnosis of depression and the duration of the disease were assessed by the Structured Clinical Interview according to the diagnostic and statistical manual of mental disorders. Depression severity was measured with the 17-item Hamilton Rating Scale for Depression, and the severity of anxiety symptoms was measured using the Hamilton Anxiety Rating Scale. Serum BDNF and GDNF were lower in major depressive disorder (MDD) patients compared to controls (p ≤ 0.001). Serum NGF levels were higher in MDD patients versus controls (p ≤ 0.001). BDNF was associated with the duration of disease only in women (p = 0.005). GDNF was not associated with clinical characteristics in either gender. In women, NGF was associated with the severity of depressive symptoms (p = 0.009), anxiety (p = 0.011) and disease duration (p = 0.005). NGF was associated with disease duration in men (p = 0.026). Our results demonstrated that significant neurochemical differences in NGF and BDNF, but not in GDNF, were associated with the clinical features of MDD when patients were stratified by gender.  相似文献

It has been described that A disintegrin and metalloproteinase (ADAM10) may involve in the physiopathology of prion diseases, but the direct molecular basis still remains unsolved. In this study, we confirmed that ADAM10 was able to cleave recombinant human prion protein in vitro. Using immunoprecipitation tests (IP) and immunofluorescent assays (IFA), reliable molecular interaction between the native cellular form of PrP (PrPC) and ADAM10 was observed not only in various cultured neuronal cell lines but also in brain homogenates of healthy hamsters and mice. Only mature ADAM10 (after removal of its prodomain) molecules showed the binding activity with the native PrPC. Remarkably more prion protein (PrP)-ADAM10 complexes were detected in the membrane fraction of cultured cells. In the scrapie-infected SMB cell model, the endogenous ADAM10 levels, especially the mature ADAM10, were significantly decreased in the fraction of cell membrane. IP and IFA tests of prion-infected SMB-S15 cells confirmed no detectable PrP-ADAM10 complex in the cellular lysates and PrP-ADAM10 co-localization on the cell surface. Furthermore, we demonstrated that the levels of ADAM10 in the brain homogenates of scrapie agent 263K-infected hamsters and agent ME7-infected mice were also almost diminished at the terminal stage, showing time-dependent decreases during the incubation period. Our data here provide the solid molecular basis for the endoproteolysis of ADAM10 on PrP molecules and interaction between ADAM10 and PrPC. Obvious loss of ADAM10 during prion infection in vitro and in vivo highlights that ADAM10 may play essential pathophysiological roles in prion replication and accumulation.  相似文献

Co-existence growth relationships were studied in communities consisting of a lipid-producing alga Chlorella sp. HQ, another green alga and one cyanobacterium: I Scenedesmus obliquus and Microcystis aeruginosa; II Chlamydomonas reinhardtii and Anabaena flos-aquae; III Selenastrum capricornutum and Microcystis wesenbergii. The cyanobacteria and green algae except for Chlorella sp. HQ were commonly detected in Chinese reservoir and wastewater. The rate of increase of apparent cell number difference with other algae (k app), inhibition/stimulation ratio (ISR) and the parameters of logistic model and co-existence model were determined for Chlorella. Chlorella strains were the most competitive in Combination I, and were stimulated during 75% of the cultivation time for all three combinations. Anabaena growth exceeded those of Chlorella and Chlamydomonas on the 5th cultivation day under 1 : 1 : 1 inoculum ratio. Scenedesmus colonies consisted of fewer cells, whose average length significantly shortened after the 5th cultivation day under 1 : 1 : 1 inoculum ratio. The developed co-existence model can identify the concrete growth inhibitor or stimulator among three species compared with the single method of cell number monitoring. Good correlation was found between transformed and non-transformed co-existence model through a mn and b mn values. Allelopathy and nutrient competition are both possible mechanisms in the above growth relationships.  相似文献
