We describe the isolation of 10 tetranucleotide microsatellite loci from the yellow-streaked greenbul using an enrichment protocol. All loci were highly variable with the number of alleles ranging from 8 to 13, and observed heterozygosity ranging from 0.652 to 0.870. All loci were in Hardy-Weinberg equilibrium; however, loci Pfl12 and Pfl54 showed significant linkage disequilibrium. All 10 loci successfully amplified and were polymorphic in at least one of four related Phyllastrephus species. These loci should prove to be widely applicable to studies of phylogeography, hybridization and paternity in African greenbuls.  相似文献

We describe the isolation of 10 microsatellite loci from the Great White Pelican using an enrichment protocol. All loci were variable with the number of alleles ranging from 2 to 19, and observed heterozygosity ranging from 0.261 to 0.913. Two loci were out of Hardy–Weinberg equilibrium, although in each case this was restricted to one of the two populations screened. None of the loci were linked. Eight to 10 loci PCR-amplified in three related species: Brown, American White and Pink-backed Pelicans. These loci should prove to be widely applicable to studies of phylogeography and demography in pelicans globally.  相似文献

Eight microsatellite markers were developed for the lepidopteran species Tortrix viridana using an enrichment protocol. The loci were highly variable with number of alleles ranging from four to 38. Six of the eight loci were in Hardy-Weinberg equilibrium. The other two were linked to the Z-chromosome. Values of observed heterozygosity ranged for the autosomal loci from 0.510 to 0.957. All loci will be useful to study dispersal and the autosomal loci, as well for phylogeographical studies.  相似文献

We describe the isolation of six tetranucleotide microsatellites from the Cape sugarbird (Promerops cafer) using an enrichment protocol. All loci were highly variable with number of alleles ranging from nine to 26 and values of observed heterozygosity ranging from 0.534 to 0.931. All loci were in Hardy–Weinberg equilibrium with the exception of Pro66 and Pro86. Further analysis of Pro86 indicated it was Z‐linked. All loci amplified and were variable in the congeneric Gurneys sugarbird (Promerops gurneyi). These loci will be used in mating system and phylogeographic studies of Cape sugarbirds in South Africa.  相似文献

Three southern African nectarivorous passerine birds, Gurney's Sugarbird Promerops gur-neyi , the Malachite Sunbird Nectarinia famosa and the Black Sunbird Nectarinia amethystina , were tested to determine their hexose and sucrose preferences. All three species preferred sucrose when offered a choice of 0.25 M solutions of glucose, fructose and sucrose. However, when the concentrations were increased to 0.73 M, the three species showed no preference for any of the three sugars. The choice made at low concentrations (equivalent to the lower limit of the range of nectar concentrations of preferred nectar-producing plants) may reflect preference for the sugar with the highest energy reward. We also examined the proposition that birds offered a choice of different concentrations of one sugar would show ranked preferences and maximize their rate of energy return by selectivity. In contrast to expectations, Gurney's Sugarbird and the Malachite Sunbird showed no preference for the highest concentrations. We suggest that dietary choices in these species indicate the birds had either reached a limit where they had sufficient energy intake or were affected by post-ingestion constraints.  相似文献

We investigated the mechanism of coexistence of the rare Amani Sunbird (Hedydipna pallidigastra) and the widespread Collared Sunbird (H. collaris), within Brachystegia woodland in the Arabuko‐Sokoke Forest, Kenya. We compared how prey abundance and search strategies affect resource exploitation by the two species. We used foraging theory to direct our measures of feeding activities as influenced by sunbird species, tree species and foraging height. We evaluated invertebrate abundance among tree species at different heights within trees. The Collared Sunbird primarily used the understory, and the Amani Sunbird primarily used the upper‐canopy. Overall, the rate of prey attacks per flight of the Amani Sunbird was 2.8 times greater than that of the Collared Sunbird. The Amani Sunbird, however, used increased search and attack rates in the understory compared with the mid‐ and upper‐canopies, but the Collared Sunbird foraged similarly throughout all strata. We hypothesize that the increased foraging rate of the Amani in the understory reflects increased foraging costs due to interference from the Collared Sunbird in that stratum. Furthermore, the Collared Sunbird exploits rich patches by moving frequently from place to place. The Amani Sunbird forages slowly, with reduced travel rates, and with a greater number of prey captures within a patch. Arthropod density did not differ among the vegetative strata, but was higher in Brachystegia spiciformis and Hymenaea verrucosa than in six other tree species. We hypothesize that the Amani Sunbird appears dependent upon continued tall B. spiciformis trees within the canopy of the Arabuko‐Sokoke Forest.  相似文献

In this paper we describe the isolation and characterization of six polymorphic microsatellite loci from the orchid Serapias vomeracea. This species is widely distributed in the Mediterranean region. Microsatellite loci were isolated from an enriched library and primer pairs were designed for 18 loci. Primer pairs for six loci amplified well and were tested on samples from southern Italy. Levels of genetic variability detected at these six loci are high, with numbers of alleles per locus ranging from 3 to 6, and observed heterozygosity (HO) ranging from 0.35 to 0.86. All primer pairs tested amplified DNA from four other Serapias species, indicating that the primers are useful for population genetic studies throughout the genus.  相似文献

Twenty‐four microsatellite loci were isolated from three species of Antarctic seals (Subfamily Monachinae, Tribe Lobodontini). Eleven loci were cloned from Weddell seal, Leptonychotes weddellii, seven from leopard seal, Hydrurga leptonyx, and six from crabeater seal, Lobodon carcinophagus. Variability was assessed in Weddell seals collected in McMurdo Sound, leopard seals from Bird Island, South Georgia, and crabeater seals sampled in the eastern Ross Sea. All loci were variable in the three species used for cloning and 22 of these loci amplified variable products in the Ross seal, Ommatophoca rossii. Cross‐species amplification was largely successful, with an average of 19 loci amplifying products in other phocids.  相似文献

The isolation and characterization of nine polymorphic microsatellite loci (eight tetranucleotide and one dinucleotide) from the canary rockfish Sebastes pinniger are described. Polymorphism at these loci revealed from six to 28 alleles, with expected heterozygosities ranging between 0.42 and 0.88, enabling high‐resolution genetic population structure investigation for this overfished species in the northeastern Pacific. They also amplify in 13 other congeneric species, providing highly variable loci for research on other rockfishes.  相似文献

Microsatellite markers were isolated from red‐flanked bushrobin, Tarsiger cyanurus, for parentage analysis. From an enriched genomic library using the fast isolation by AFLP of sequences containing repeats (FIASCO) method, six polymorphic loci were obtained. Seven to 21 alleles were identified in an analysis of 19 red‐flanked bushrobin, with observed heterozygosity ranging from 0.842 to 1.00. All loci showed Hardy–Weinberg equilibrium and linkage equilibrium. Total exclusion probability using six loci was 0.998 for neither parent known, and 0.999 for one parent known. We also tested the utility of these loci on four species of Turdidae. Although one of the loci failed to amplify in any species, the other primers successfully amplified in at least two of the species we tested.  相似文献

Eight microsatellite loci were isolated from an enriched genomic library of the great leaf-nosed bat, Hipposideros armiger. The polymorphism of these loci was tested on a population of 48 individuals from Anhui Province, China. All loci revealed the polymorphism ranging from three to 12 alleles. The observed heterozygosity values were from 0.213 to 0.875 and expected heterozygosity from 0.232 to 0.820. No significant linkage disequilibrium was detected. Two loci significantly deviated from Hardy–Weinberg equilibrium after Bonferroni correction. In addition, successful cross-amplification also suggested that these microsatellite markers will facilitate research on the population genetics and gene flow of H. armiger and other related species.  相似文献

We describe the cloning and characterization of eight novel tetranucleotide microsatellite loci in the ornate chorus frog (Pseudacris ornata). We also screened 26 loci from GenBank that were isolated from other Pseudacris species and obtained consistent product from five of these dinucleotide loci. All loci are polymorphic. In our sample of 26 frogs from a natural population, polymorphism ranged from 1 to 22 alleles per locus with expected heterozygosities ranging from 0 to 0.958. These loci enable high-resolution studies of P. ornata. Moreover, cross-species amplification success suggests they will also be useful for other chorus frog species.  相似文献

Pteronotus psilotis, a mormoopid bat, is an insectivorous, gregarious and strict cave-dwelling species that is found areas between the sea level and an elevation of about 1000 masl. This species is present in diverse habitats ranging from rain forest to dry deciduous forest. Nine microsatellite loci were developed for Wagner’s mustached bat, Pteronotus psilotis using the next-generation sequencing approach, and their utility for population genetics studies was assessed. All loci were polymorphic (7–15 alleles) and characterized in 30 individuals from three P. psilotis populations, with the levels of observed and expected heterozygosity ranging from 0.280 to 0.867 and 0.584 to 0.842, respectively. One locus showed significant departures from Hardy–Weinberg expectations after Bonferroni correction. Cross-amplification in 11 other bat species was tested, for which eight microsatellites were successfully amplified, and of these seven were polymorphic. The development of these new microsatellite loci will contribute to investigations of genetic population structure, genetic diversity and gene flow in P. psilotis populations, as well as in other closely related bat species.  相似文献

Highly polymorphic microsatellite markers can supply demographic information on founder events and range expansion following initial introduction of invasive insect species. Six microsatellite loci were isolated from a partial DNA library in order to study the invasion patterns of a seed chalcid, Megastigmus spermotrophus, introduced to Europe and New Zealand. Allelic diversity at all described loci was high, ranging from 17 to 30 alleles per locus. All six loci were successfully amplified in 15 congeneric species.  相似文献

Eight microsatellite loci were developed for the jack mackerel (Trachurus murphyi), a fish of significant commercial importance in the Southeast Pacific. Genetic variation at these loci was examined in 15 samples from the locality of Talcahuano (Chile). All eight were highly polymorphic, with a number of alleles per locus ranging from 4 to 22 and an observed heterozygosity from 0.429 to 1. These markers will be useful to address issues of population genetics, ecology, conservation and fisheries management related to that species.  相似文献

All loci were highly variable with the number of alleles ranging from six to 20 and observed heterozygosity ranging from 0.410 to 0.940. Although all loci were in Hardy-Weinberg equilibrium, locus Mst95 showed significant homozygote excess in both surveyed populations, possibly a consequence of the presence of null alleles at this locus. These loci will be used to determine the extent to which spot-throat populations are isolated in order to help set conservation priorities for this ancient African lineage.  相似文献

Corallium rubrum, an endemic Mediterranean gorgonian coral, has undergone an intensive exploitation leading to the extinction of local commercial banks and changes in the structure and dynamics of coastal populations. Management and conservation of this species requires a better understanding of the genetic structuring and connectivity among populations. With this aim, seven microsatellite loci have been isolated. All loci were polymorphic with allele numbers ranging from five to 26 and observed heterozygosity ranging from 0.18 to 0.68. Significant deviations from Hardy–Weinberg expected genotype frequencies due to heterozygote deficiency were detected at all loci.  相似文献
