As shown by A. Rapoport (1952), when a very brief stimulation or “instantaneous input” is applied to a random net, the subsequent events are determined by the parameters of the net as follows: If the axon densitya is sufficiently large and the fraction γ of the neurons initially stimulated exceeds a certain value γ1 (theover-all threshold of the net for instantaneous stimulation), excitation will spread through the net until a steady state is reached in which a fraction γ2 ⩾ γ1 of the neurons is firing (“ignition phenomenon”). If γ < γ1 the activity in the net dies out. However, if the axon density is too small, the activity will ultimately die out, no matter how large the fraction of initially stimulated neurons. Thus there exists a limiting valueA of the axon density below which the net cannot “ignite”. ThisA is a function ofh, theindividual threshold of the neurons constituting the net (we assume hereh≥2, since forh=1 the situation is essentially different). Geometrically γ1 and γ2 are determined as the two intersection points of a straight line with a sigmoid curve. Whena<A the two curves do not intersect and fora=A they are tangent. In this paper the “tangency case” is investigated and the general features of the functionA(h) are determined. It is shown thatA increases monotonically withh (as one would expect). For all values ofh>1 we haveA(h)>h, but the fractionA(h)/h and the derivativedA(h)/dh approach unity ash increases. An analytical expression of the functionA(h) valid for very large values ofh is derived.  相似文献

Agronomic traits, photosynthetic pigments, gas exchange, and chlorophyll (Chl) fluorescence parameters of red stem buckwheat (Fagopyrum dibotrys Hara) mutants induced by γ-radiation were compared with green control at seedling stage. Plant height, number of first-class branches, and rhizome biomass were inhibited significantly (p<0.01). Chl a, Chl b, and Chl a+b contents decreased with elevated dose of γ-rays, while increasing carotenoid content indicated that buckwheat was capable of adjusting to the radiation damage. Decrease in net photosynthetic rate was the result of both stomatal and non-stomatal limitations. Fluorescence parameters, such as F0, Fm, Fv/Fm, Fv/F0, ΦPS2, electron transport rate, and photochemical quenching declined significantly (p<0.01) as compared with control due to photoinhibition, while non-photochemical quenching increased to enhance thermal dissipation. Lower parameters implied that leaf tissue was damaged significantly by high dose of γ-radiation and therefore leaf senescence was accelerated.  相似文献

The conditions under which the output,γ b (t), of a biological system is related to the input,γ a (t), by an integral equation of the typeγ b (t) = ∫ 0 t γ a (ω)w(t−ω)dω, where ω(t) is a transport functioncharacteristic of the system, are analyzed in detail. Methods of solving this type of integral equation are briefly discussed. The theory is then applied to problems in tracer kinetics in which input and output are sums of exponentials, and explicit formulae, which are applicable whether or not the pool is uniformly mixed, are derived for “turnover time” and “pool” size.  相似文献

The effect of high salt stress on PS II heterogeneity was investigated in wheat (Triticum aestivum) leaves. On the basis of antenna size, PS II has been classified into three forms, i.e., α, β, and γ centers while on the basis of electron transport properties of the reducing side of the reaction centers, two distinct forms of PS II have been suggested, i.e., QB reducing centers and QB non-reducing centers. The chlorophyll a (Chl a) fluorescence transients, which can quantify PS II behavior, were recorded using PEA to derive OJIP in vivo with high time resolution and further analyzed according to JIP test. Our results showed that with an increase in the salt concentration during growth, the number of QB non-reducing centers increased. In antenna size heterogeneity the number of β and γ centers increased while the number of α centers decreased. A change in the energetic connectivity between the PS II units was also observed. Recovery studies showed that antenna heterogeneity was completely recovered from damage at 0.5 M NaCl concentration and partially recovered at 1 M NaCl concentration while reducing side heterogeneity showed no recovery at all after 0.5 M onwards.  相似文献

Summary A Clostridium sp., isolated from flooded soil amended with lindane (γ-BHC), decomposed methoxychlor, γ-BHC and heptachlor in that order under anaerobic condition. During the bacterial degradation of ring-labelled C14-γ-BHC, there was a net loss of radioactivity from the reaction mixture. Release of C14O2 during the degradation of C14-γ-BHC was negligible.  The mouse Fcgr1 gene encoding the high-affinity IgG receptor (FcγRI) exists as two known alleles, FcγRI-BALB and FcγRI-NOD, and these alleles exhibit functional differences. To determine whether other alleles exist in mouse strains, Fcgr1 coding regions from 35 strains of mice were sequenced and a further five alleles were identified. The FcγRI-BALB and NOD alleles are now designated the "a" and "d" alleles, respectively. Analysis of the five new alleles revealed that although no polymorphisms were observed in the two leader exons, nucleotide and subsequent amino acid changes were observed in the exons encoding the extracellular domains, and transmembrane and cytoplasmic tail. The cDNA of the seven alleles (a–g) were isolated and transiently transfected into COS cells, and IgG-binding studies were performed. Receptors encoded by four of the five new alleles (b, c, f, g) bound IgG2a with high affinity, displaying IgG binding characteristics similar to the a allele (previously FcγRI-BALB). The d allele (previously FcγRI-NOD) and the e allele [derived from Mus spretus (SPRET/Ei)] encoded receptors which showed broader specificity by binding monomeric IgG2a, IgG2b, and IgG3. Coinjection strategy for visual identification of transgenic mice   总被引:10,自引:0,他引:10  
Transgenic mice were generated by coinjection of a dominant marker gene that induces fur and eye pigmentation (a tyrosinase minigene) plus an unrelated DNA construction that has a γ-glutamyl transferase (γGT) promoter linked to aras oncogene. Mice transgenic for γGT-ras could be identified in the first and all subsequent generations by simple visual inspection for pigmentation. Furthermore, the γ-glutamyl transferase promoter was active in kidney but not skin of the transgenic mice, indicating that the cointegrated DNA was active and independently expressed. These results confirm that the tyrosinase minigene can be used for coinjections to allow rapid visual identification of transgenic mice.  相似文献

A symbiotic green sulfur bacterium, strain CaD, was isolated from an enrichment culture of the phototrophic consortium “Chlorochromatium aggregatum”. The capability of the epibiont to grow in pure culture indicates that it is not obligately symbiotic. Cells are Gram-negative, nonmotile, rod-shaped and contain chlorosomes. Strain CaD is obligately anaerobic and photolithoautotrophic, using sulfide as electron donor. Acetate and peptone are photoassimilated in the presence of sulfide and hydrogencarbonate. Photosynthetic pigments contain bacteriochlorophylls a and c, and γ-carotene and OH-γ-carotene glucoside laurate as the dominant carotenoids. In cells from pure cultures, chlorosomes are equally distributed along the inner face of the cytoplasmic membrane. In contrast, the distribution of the chlorosomes in symbiotic epibiont cells is uneven, with chlorosomes being entirely absent at the site of attachment to the central bacterium. The symbiotic epibiont cells display a conspicuous additional layered structure at the attachment site. The G + C content of genomic DNA of strain CaD is 46.7 mol%. On the basis of 16S rRNA sequence comparison, the strain is distantly related to Chlorobium species within the green sulfur bacteria phylum (≤94.6% sequence homology). The novel isolate is therefore described as a novel species within the genus Chlorobium, Chlorobium chlorochromatii.  相似文献

The chlorinated insecticide γ-hexachlorocyclohexane (γ-HCH) is sequentially metabolized by the products of linA, linB, linC, linD, linE, and linF genes to β-ketoadipate, which is subsequently mineralized. Two or more copies of these genes are present in the bacterium Pseudomonas aeruginosa ITRC-5 that was isolated earlier by selective enrichment on technical-HCH. At least one copy of linA, linB, linC, linD, and possibly linE is lost from ITRC-5 upon its growth on γ-HCH. All the lin genes, however, are lost when the bacterium was grown in Luria–Bertani (LB) medium. The loss of lin genes is accompanied with the loss/rearrangement of insertion sequence IS6100 genes. Concomitant to the loss of lin genes, the degradation of HCH-isomers by “γ-HCH grown cells” is slower, when compared with “technical-HCH grown cells”, and is completely lost by “LB-grown cells”. The selective loss of lin genes during different growth conditions has not been reported before and is expected to help in understanding the dynamism of degradative genes.  相似文献

From 1999 to 2003, a range of carbon fluxes was measured and integrated to establish a carbon balance for a natural evergreen forest of Castanopsis kawakamii (NF) and adjacent monoculture evergreen plantations of C. kawakamii (CK) and Chinese fir (Cunninghamia lanceolata, CF) in Sanming Nature Reserve, Fujian, China. Biomass carbon increment of aboveground parts and coarse roots were measured by the allometric method. Above- and belowground litter C inputs were assessed by litter traps and sequential cores, respectively. Soil respiration (SR) was determined by the alkaline absorbance method, and the contribution from roots, above- and belowground litters was separated by the DIRT plots. Annual SR averaged 13.742 t C ha−1 a−1 in the NF, 9.439 t C ha−1 a−1 in the CK, and 4.543 t C ha−1 a−1 in the CF. For all forests, SR generally peaked in later spring or early summer (May or June). The contribution of root respiration ranged from 47.8% in the NF to 40.3% in the CF. On average, soil heterotrophic respiration (HR) was evenly distributed between below- (47.3∼54.5%) and aboveground litter (45.5%–52.7%). Annual C inputs (t C ha−1 a−1) from litterfall and root turnover averaged 4.452 and 4.295, 4.548 and 2.313, and 2.220 and 1.265, respectively, in the NF, CK, and CF. As compared to HR, annual net primary production (NPP) of 11.228, 13.264, and 6.491 t C ha−1 a−1 in the NF, CK, and CF brought a positive net ecosystem production (NEP) of 4.144, 7.514, and 3.677 t C ha−1 a−1, respectively. It suggests that native forest in subtropical China currently acts as an important carbon sink just as the timber plantation does, and converting native forest to tree plantations locally during last decades might have caused a high landscape carbon loss to the atmosphere.  相似文献

 The 20S proteasome is a multi-subunit protease responsible for the production of peptides presented by major histocompatibility complex (MHC) class I molecules. Recent evidence indicates that an interferon-γ (IFN-γ)-inducible PA28 activator complex enhances the generation of class I binding peptides by altering the cleavage pattern of the proteasome. In the present study, we determined the primary structures of the mouse PA28 α- and β-subunits. The deduced amino acid sequences of the α- and β-subunits were 49% identical. We also determined the primary structure of the mouse PA28 γ-subunit (Ki antigen), a protein of unknown function structurally related to the α- and β-subunits. The amino acid sequence identity of the γ-subunit to the α- and β-subunits was 40% and 32%, respectively. Interspecific backcross mapping showed that the mouse genes coding for the α- and β-subunits (designated Psme1 and Psme2, respectively) are tightly linked and map close to the Atp5g1 locus on chromosome 14. Thus, unlike the LMP2 and LMP7 subunits, the IFN-γ-inducible subunits of PA28 are encoded outside the MHC. The gene coding for the γ-subunit (designated Psme3) was mapped to the vicinity of the Brca1 locus on chromosome 11. A computer search of the DNA databases identified a γ-subunit-like protein in ticks and Caenorhabditis elegans, the organisms with no adaptive immune system. It appears that the IFN-γ-inducible α- and β-subunits emerged by gene duplication from a γ-subunit-like precursor. Received: 11 March 1997  相似文献

 One hundred and eighty dihaploid clones used for protoplast fusions, and 144 tetraploid German potato cultivars were analysed for their cytoplasms using 11 homologous mt DNA-probes, and were classified as mitochondrial (mt) types α, β, γ, δ, and ɛ according to their RFLP patterns. From the 4x cultivars, 79 had the typical mt-type β of Solanum tuberosum being different from the 46 cvs which had the mt-α type and 19 others with mt-γ. A dendrogram shows their relationships to other Solanum species. The distantly related mt-ɛ was only found in di-haploids, and particularly in clones deriving from Solanum phureja and Solanum andigena. Accessory mt types will be actualized on website (http://www.edv.agrar.tu-muenchen.de/pbpz/ mm/mt/al1.htm). In order to evaluate the genetic potential of novel plastid-mitochondrial configurations we have analyzed four representative populations, which derive from different fusion-combination classes: [α (+) β], [α (+) γ], [α (+) δ] and [α (+) ɛ]. On the mitochondrial expression level, hybrids from an [α (+) ɛ] fusion could be distinguished by in-organello translation from [α (+) β] hybrids, and other di-haploids, by an additional translation product of 15 kDa. In fusion parents with mt-α and -γ an additional atp6 reading frame is detectable in sub-stoichiometric amounts by the use of specific PCR primers. The gene differs from the original 211 bp 3′ from the stop codon. Novel RFLP-patterns in 10% of the somatic hybrids were due to a high-rate replication of this pre-existing parental genome region. A second characteristic for somatic hybrids was the partial addition of parental mt sub-genomes. The major part of them revealed a new organization in their mt genomes at the mt-type characteristic loci rpl5, rps14, cob, rps10, coxI and rpl2, which contain recombination-specific repeats homologous to Petunia spp. and Nicotiana. Increasing therapeutic applications for recombinant human interferon-gamma (rhIFN-γ), an antiviral pro-inflammatory cytokine, has broadened interest in optimizing methods for its production. We herein describe a unicellular eukaryotic system, Leishmania tarentolae, a Trypanosomatidae protozoan parasite of gecko Tarentola annularis, which has recently been introduced as a candidate for heterologous gene expression. In this study, the hIFN-γ cDNA was amplified from phyto-hemagglutinin-stimulated peripheral blood mononuclear cells of a healthy blood donor using RT–PCR. In order to express, the rhIFN-γ protein, the resulting cDNA was cloned in two expression cassettes (each containing one copy of hIFN-γ cDNA) and integrated into the small subunit of ribosomal RNA gene of L. tarentolae genome by electroporation. Transformed clones were selected in the presence of appropriate antibiotics. Western blotting of rhIFN-γ and ELISA confirmed the expression and production of 9.5 mg of rhIFN-γ protein/l respectively.  相似文献
