通过印度块菌子囊果石蜡切片的显微观察,对印度块菌子囊果包被及子实层的显微结构、子囊及子囊孢子的发育过程进行研究.结果表明:(1)印度块菌子囊果是由包被和子实层构成,子囊果的表面密被大量大小不一的疣状突起,包被由外皮层和内皮层构成;子实层由封闭的、大小不一的产孢组织构成,在产孢组织中包含有侧丝、产囊丝、子囊和子囊内的子囊孢子;(2)成熟印度块菌子囊果横切面上有明暗相间的迷宫状纹脉;(3)子囊卵形,由产囊丝顶端的细胞发育而来;(4)成熟子囊孢子红褐色,椭圆形至近球形不等,子囊孢子双层壁,外壁密布有刺状纹饰.子囊孢子在子囊内的发育过程中常出现败育现象,每个成熟子囊中含有1~5个子囊孢子(常见4个). 相似文献

对新近发现的块菌属一新种——攀枝花白块菌(Tuber panzhihuanense)子囊果中可培养细菌的多样性进行了研究。采用胰蛋白大豆培养基(TSA)对菌株进行分离。用毛细管电泳(HPCE)对所有获得的菌株的16S rDNA V3高变区进行筛选获得不同条带大小的菌株,对筛选出的菌株的16S rDNA进行测序,并进行细菌多样性分析和研究。结果显示,攀枝花块菌子囊果内可培养细菌在数量及种类上都表现出很高的多样性,所有细菌分属于5个门的11个属和20个种。在所分离到的变形菌门的细菌中,数量最多的菌株(4968%)属于γ Proteobacteria,其中假单胞菌属的Pseudomonas lurida为优势类群;其次为α Proteobacteria,占3742%,其中以固氮菌 Bradyrhizobium japonicum和Phyllobacterium spp.为优势类群。其余的菌株属于放线菌门(Actinobacteria) (322%)和厚壁菌门(Firmicutes) (774%),厚壁菌门中以芽孢杆菌属(Bacillus)为代表菌群。酸杆菌门中的Terriglobus roseus(194%)首次从块菌中分离获得。 相似文献

利用扫描电镜对成熟印度块菌子囊果的内部结构进行了观察,以系统揭示其子囊果内部组织特征,为块菌属的分类以及块菌属真菌子囊果的生理研究奠定基础。观察结果进一步证明,成熟印度块菌子囊果横切面上的白色迷宫状脉络是由不育的侧丝构成,而暗脉则是被侧丝缠绕并包裹着的可育的菌丝组织,即产孢组织,这些白色脉络和暗脉就构成了印度块菌子囊果横切面上迷宫状的纹脉;产孢组织中,可观察到正在发育的大大小小的子囊被缠绕在一起的大量产囊丝与侧丝包裹着,形成密密麻麻微小的类似蜂巢状结构;子囊孢子游离于子囊中,成熟子囊孢子表面有刺状纹饰,刺的顶端有小弯钩。单个子囊内含的子囊孢子大小与其内含的子囊孢子数目有关,子囊内所含的子囊孢子越多,子囊孢子就越小。 相似文献

Molecular and morphological data indicate that the genus Heydenia is closely related to the cleistothecial ascomycete Orbicula (Pyronemataceae, Pezizales). Observations on the disposition and the immediate surroundings of immature spores within the spore capsule suggest that the Heydenia fruiting bodies are teleomorphs producing early evanescent asci in stipitate cleistothecia. The once advocated identity of Heydenia with Onygena is refuted on molecular grounds. Onygena arietina E. Fischer is transferred to Heydenia. 相似文献

Species delimitation in the genus Geopora (Pyronemataceae) is complicated because of small number of differentiating characters, values of which tend to overlap among
the species. Current classification relies mainly on size and shape of ascospores and fruit-bodies, position of the apothecia
in the ground and length of excipular hairs. We measured ascospores in ca. 90 Geopora specimens. Sequences of internal transcribed spacer (ITS) rDNA gene were obtained to investigate phylogenetic relationships
in the genus. Maximum parsimony (MP) and Bayesian analyses reveal that the well-supported clades detected often do not correspond
to the species concepts based on morphological characters. Nine out of the ten lineages include specimens which were initially
identified as belonging to different species. The dimensions of ascospores vary to great extent within the lineages. The size
and shape of fruit-bodies, length of excipular hair and hymenium colour are mostly homogenous within each clade; however,
these characters coincide to a great extent among the lineages and the latter can be assessed only from fresh fruit-bodies.
Nuclear DNA content, and accordingly, ploidy level do not provide evidence for species distinction. Geopora arenicola, G. tenuis and G. sepulta were recognized as monophyletic species. Geopora foliacea and G. cervina could not be explicitly delimited and the G. cervina complex comprising three clades was introduced. 相似文献

The species concept within the genus Picoa Vittad. is here revisited in light of new molecular and ecological data obtained from samples collected throughout the Mediterranean basin. Two highly diverse widespread clades and four additional minor lineages were significantly supported by three genes dataset (ITS, 28s LSU and RPB2) inferences for 70 specimens. The two widespread clades occur in very different geographical and ecological areas associated with exclusive host plants in the genus Helianthemum. SEM study of spore surface morphology in these lineages revealed the existence of smooth ascospores in the majority of these clades. However the most frequent lineage in Europe and coastal North Africa displayed either smooth or verrucose spores. Hence this morphological criterion cannot be reliably used to discriminate between the different clades. In addition, SEM observations made on ascospores from several original collections of P. juniperi and P. lefebvrei supported the hypothesis that ornamentation depends on the degree of maturity in some of these lineages. Geographical and ecological, rather than morphological data are here suggested as the most useful characters to separate the different lineages in Picoa. Further studies focusing on these features are needed before the names P. juniperi and P. lefebvrei can be unambiguously linked with the genetic lineages observed. 相似文献
