Laccase from Trametes versicolor was immobilized on Amberlite IR-120 H beads. Maximum immobilization obtained was 78.7% at pH = 4.5 and temperature T = 45 °C. Kinetic parameters, Km and Vmax values, were determined respectively as 0.051 mM and 2.77 × 10?2 mM/s for free and 4.70 mM and 5.27 × 10?3 mM/s for immobilized laccase. The Amberlite–laccase system showed a 30% residual activity after 7 cycles. On the other hand, the loss of activity for free laccase after 7 days of storage at 4 °C was 18.5% in comparison to Amberlite–laccase system with a loss of 1.4%, during the same period. Improved operational, thermal and storage stabilities of the immobilized laccase were obtained compared to the free counterpart. Therefore, the use of low-cost matrices, like Amberlite for enzyme immobilization represents a promising product for enzymatic industrial applications.  相似文献

Extracellular laccase produced by the wood-rotting fungus Cerrena unicolor was immobilized covalently on the mesostructured siliceous cellular foams (MCFs) functionalised using various organosilanes with amine and glycidyl groups. The experiments indicated that laccase bound via glutaraldehyde to MCFs modified using 2-aminoethyl-3-aminopropyltrimethoxysilane remains very active. In the best biocatalyst activity was about 42,700 U mL?1 carrier (66,800 U mg?1 bound protein), and hence significantly higher than ever reported before. Optimisation of the immobilization procedure with respect to protein concentration, pH of coupling mixture and the enzyme purity afforded the biocatalyst with activity of about 90,980 U mL?1. For the best preparation, thermal- and pH-stability, and activity profiles were determined. Experiments carried out in a batch reactor showed that kcat/Km for immobilized enzyme (0.88 min?1 μM?1) was acceptable lower than the value obtained for the native enzyme (2.19 min?1 μM?1). Finally, potentials of the catalysts were tested in the decolourisation of indigo carmine without redox-mediators. Seven consecutive runs with the catalysts separated by microfiltration proved that adsorption of the dye onto the carrier and enzymatic oxidation contribute to the efficient decolourisation without loss of immobilized enzyme activity.  相似文献

Cross-linked Sepharose beads were treated with laccase–TEMPO system for oxidation of the primary alcohol groups on the sugar moieties. Optimal activation conditions using Trametes versicolor laccase were at pH 5 and 22 °C, giving an aldehyde content of 55 μmol g−1 Sepharose with 28 units g−1 of laccase and 12.5 mM TEMPO. The activated Sepharose was used for immobilization of trypsin as model protein. Highest degree of immobilization was obtained at pH 10.5 but the activity yield was only 31% of that loaded on the gel. The yield of gel bound trypsin activity was increased to 76% (corresponding to about 43 U g−1 Sepharose) when the immobilization was performed in the presence of trypsin inhibitor, benzamidine. The immobilization yields were comparable to that obtained on the matrix activated using sodium periodate (containing 72 μmol aldehyde per g Sepharose). Recycling and storage of the immobilized trypsin preparations showed high stability of the enzyme bound to laccase–TEMPO activated gel.  相似文献

The white-rot fungus Pleurotus ostreatus strain 32 is an excellent producer of the industrially important enzyme laccase. Laccase was the only ligninolytic activity detected in the supernatant when the fungus was grown in liquid culture with or without shaking. Growth and laccase production in static cultivation were superior to that in agitated cultivation, and N-limited culture is of benefit to laccase production. When using cellobiose and peptone as carbon and nitrogen source, a higher activity level was obtained. 2,2′-Azino-di-(3-ethylbenzothialozin-6-sulfonic acid) (ABTS) (1 mM) was shown to be the best inducer of laccase production, reaching maximum values of about 400 U/ml. Cu2+ (1 mM) also had a positive effect on laccase production, activity being enhanced to 360 U/ml. In addition, anthraquinone dye SN4R can be effectively decolorized by crude laccase (30 U/ml), the rate of which was 66%. The decolorization rate was increased by 90% with ABTS (0.16%) addition as a mediator of laccase.  相似文献

Abstract

The main objective of this study is the evaluation of the capability of laccase from Myceliophthora thermophila immobilized on fumed silica microparticles (fsMP) for the removal of endocrine disrupting chemicals (EDCs) in two enzymatic reactor configurations. This type of support can also be magnetized to allow the straightforward separation of the biocatalyst under a magnetic field. The support exhibited excellent biocompatibility with the enzyme, superior tolerance to pH and temperature as well as improved stability in comparison with the free enzyme, even in the presence of organic solvents and enzyme inhibitors. The technical feasibility of the removal of EDCs by immobilized laccase was assessed in two types of enzymatic reactors operated in sequential mode: a membrane reactor using fsMP-laccase and a reactor with magnetic separation using magnetized fsMP-laccase. The extent of transformation for the target compounds: bisphenol A (BPA) and 17β-estradiol (E2) was high and comparable to free laccase in both systems (up to 80%). The possibility of reusing the immobilized enzyme, especially for magnetized supports, offers an interesting approach in the development of enzyme based processes for the biotransformation of emerging pollutants.  相似文献

This paper studies the synthesis of triacylglycerols (TAGs) rich in palmitic acid (PA) at position 2, from palm oil stearin (POS), a vegetable oil highly rich in this acid (60%). The developed process consists of two steps: (1) obtaining PA enriched free fatty acids (FFAs), and (2) enrichment of POS in PA by acidolysis of this oil with PA enriched FFAs, catalyzed by lipase Novozym 435. In step (1) two PA enriched FFA mixtures were obtained: one by saponification of POS, and a PA concentrate (75.1% PA) obtained by crystallization at low temperature in solvents. The latter was obtained carrying out two crystallizations in acetone at −24 and −20 °C, from which PA was recovered in the solid phases with a total yield of 84%. These PA enriched FFA mixtures were used in step (2) of acidolysis of POS, along with commercial PA (98% PA). In this acidolysis step four factors were studied: temperature, hexane/reaction mixture ratio, FFA/POS molar ratio and the intensity of treatment (IOT = lipase amount × reaction time/POS amount). The best results (TAGs with 79% PA and 75% PA at position 2) were obtained with commercial PA, at 37 °C, 10 mL hexane/g reaction mixture, a FFA/POS molar ratio 3:1 (1:1, w/w) and an IOT = 9.6 g lipase × h/g POS (for example 48 h, 10 g lipase and 50 g POS). PA enriched TAGs were purified neutralizing the FFAs by KOH hydroethanolic solutions and extracting the TAGs with hexane. In this way 99% pure acylglycerols (TAG + DAG) were obtained; the recovery yield of this purification step was 95%. The experiments carried out with POS demonstrated that it is possible to use only this oil (60% PA, 23% PA at position 2) as a source of PA to obtain a TAG with 70.7% PA and 70.5% PA at position 2. This process consists of four steps: (1) saponification of POS, (2) crystallization of FFAs to obtain PA enriched FFAs (75.1% PA), (3) acidolysis of POS with these FFAs, catalyzed with Novozym 435, to produce PA enriched TAGs at position 2 (70.5% PA) and (4) purification of TAGs to obtain approximately 95% purity and yield. These PA enriched TAGs could be used to obtain structured TAGs rich in PA at position 2 and in oleic acid at positions 1 and 3 (OPO), which is the principal TAG of human milk fat.  相似文献

An integrated wetland system (IWS) including constructed wetlands (CWs) and modified natural wetlands (NWs) for wastewater treatment to replenish water to wetlands located at the Beijing Wetland School (BWS) in Beijing, China, is presented in this paper. The synergistic effects of CWs and NWs on treated water quality are investigated. The IWS is proved to be an effective wastewater treatment technique and a better alternative to alleviate the water shortage for conservation of wetlands based on the monitoring data obtained from October 2007 to 2008. The results show that CWs and NWs play different roles in removing contaminants from wastewater. The COD removal efficiency in CWs is higher than that in modified NWs, whereas the modified NWs can compensate for the deficiency of CWs where a stable and sufficient rhizosphere is not fully formed in the start-up period. All removal rates of COD, TN, and TP in CWs and modified NWs vary from 50 to 70%, while the total removal rate of COD, TN, and TP in IWS is about 85–90%. The operational results show that the maximum area loading of organic pollutants in modified NWs (65 kg/ha d) is slightly higher than the empirical one (60 kg/ha d) recommended by USEPA (2000) for free water surface wetlands.  相似文献

Elimination of recalcitrant chemicals during wastewater treatment is a difficult problem for both developing and industrialized countries. The biological elimination of very persistent xenobiotics such as endocrine disrupting chemicals from municipal and industrial sewage treatment plants is an ambitious challenge as existing physico-chemical methods, such as advanced oxidation processes, are energy-intensive and consume high amounts of chemicals. Through the entry into force of strict legislative measures, such as the Water Framework Directives (EU WFD in Directive 2000/60/EC of the European Parliament and of the Council establishing a framework for the Community action in the field of water policy, 2000) and REACH (REACH EU in European Community Regulation on chemicals and their safe use (EC 1907/2006), 2007), the market for wastewater treatment is exploding. For instance the European market potential for the membrane bioreactor technology is estimated to 57 M€ per year. Based on recent progresses in nanotechnology, new developments in catalysis and environmental applications can be foreseen for the near future. Indeed, because of high surface area-to-volume ratio in nano-systems, heterogeneous enzymatic or catalytic reactions can be greatly enhanced. In the LANCE project a nanoparticle (NP)-based technology is under development. Cheap and resistant oxidative enzymes, i.e. laccases are immobilized onto the surface of the particles in order to produce systems possessing a broad substrate spectrum for the degradation of cocktails of recalcitrant pollutants. One of the objectives is to produce NPs that are compatible with wastewater treatment and can be synthesised in a cost-effective and large-scale fashion, e.g. silica-based NPs using flame spray pyrolysis and emulsion-based techniques. The modified particles are applied in bioreactors where they are retained, i.e. membrane bioreactors or perfusion basket reactors to eliminate pollutants from the wastewater. Such reactors allow multi-cycle use of the NPs coated with active enzymes and thus contribute to decrease the treatment costs. The two-year activities of the LANCE project encompass the synthesis of various NP systems, the immobilization of selected low cost industrial laccases on the latter, and the technical and scientific proof of the “depollution” concept.  相似文献

This research was to study the effect of blast furnace dust (BFD) as a new reactive material for the degradation of chlorinated organic (COCs) and endocrine disrupting compounds (EDCs). When 100 g/L of BFD was used, the effective degradation of tetrachloroethylene (PCE) was obtained. The cis-DCE (0.93 mM) was dechlorinated to below detection limit within 120 h of reaction. Among various COCs and EDCs, they were degraded at least 90% except for sodium perchlorate. The metabolites of 4-tert-octylphenol (4-t-OP) were identified as 2,4,4-trimethyl-2-pentanol, hydroquinone and 2-tert-octylhydroquinone, respectively. The effective degradation of PCE and 4-t-OP was occurred at range of pH 4–7. A total of 100% of cis-DCE and 86% of 4-t-OP were degraded in fed-batch experiments after 264 h. A solution of highly enriched bacteria completely insolubilized 2.14 mM of Zn eluted from BFD within 168 h of culture. These researches will provide more information related to the application for other contaminated water and wastewater treatments.  相似文献

In this work, a laccase producer, Ganoderma lucidum, was separated and identified according to its morphological characteristics and phylogenetic data. A 4000 U/l and 8500 U/l of laccase activity was obtained in 500 ml flask by submerged culture and biomembrane-surface liquid culture (BSLC), respectively. Furthermore, the novel biomembrane-surface liquid co-culture (BSLCc) was developed by adding Saccharomyces cerevisiae to reactor in order to shorten the fermentation period and improve laccase production. Laccase activity obtained by BSLCc, 23 000 U/l, is 5.8 and 2.7 times of that obtained by submerged culture and BSLC, respectively. In addition, laccase production by BSLCc was successfully scaled-up to 100 l reactor, and 38 000 U/l of laccase activity was obtained on day 8. The mechanism of overproducing laccase by BSLCc was investigated by metabolism pathway analysis of glucose. The results show glucose limitation in fermentation broth induces the secretion of laccase. The addition of S. cerevisiae, on one hand, leads to an earlier occurrence of glucose limitation state, and thus shortens the fermentation time; on the other hand, it also results in the appearance of a series of metabolites of the yeast including organic acids, ethanol, glycerol and so forth in fermentation broth, and both polyacrylamide gel electrophoresis analysis and enzyme activity detection of laccase show that these metabolites contribute to the improvement of laccase activity.  相似文献

A packed-bed reactor (PBR) system using fungus whole-cell biocatalyst was developed for biodiesel fuel production by plant oil methanolysis. Lipase-producing Rhizopus oryzae cells were immobilized within 6 mm × 6 mm × 3 mm cuboidal polyurethane foam biomass support particles (BSPs) during batch cultivation in a 20-l air-lift bioreactor. Emulsification of the reaction mixture containing soybean oils and water improved the methanolysis reaction rate. Using a high flow rate for the reaction mixture in the PBR caused exfoliation of the immobilized cells from the BSPs, while the inefficient mixing of the reaction mixture at low flow rates allowed the BSPs to be covered with a hydrophilic layer of high methanol concentration, leading to a significant decrease in lipase activity. A high methyl ester content of over 90% was achieved at a flow rate of 25 l/h in the first cycle of repeated batch methanolysis and a high value of around 80% was maintained even after the tenth cycle. Comparison with methanolysis reaction in a shaken bottle suggested that the PBR enhances repeated batch methanolysis by protecting immobilized cells from physical damage and excess amounts of methanol. The process presented here is therefore considered to be promising for industrial biodiesel-fuel production.  相似文献

The properties of Trematosphaeria mangrovei laccase enzyme purified on Sephadex G-100 column were investigated. SDS–PAGE of the purified laccase enzyme showed a single band at 48 kDa. The pure laccase reached its maximal activity at temperature 65 °C, pH 4.0 with Km equal 1.4 mM and Vmax equal 184.84 U/mg protein. The substrate specificity of the purified laccase was greatly influenced by the nature and position of the substituted groups in the phenolic ring. The pure laccase was tested with some metal ions and inhibitors, FeSO4 completely inhibited laccase enzyme and also highly affected by (NaN3) at a concentration of 1 mM. Amino acid composition of the pure enzyme was also determined. Carbohydrate content of purified laccase enzyme was 23% of the enzyme sample. The UV absorption spectra of the purified laccase enzyme showed a single peak at 260–280 nm.  相似文献

To enhance laccase yield, the laccase gene from Bacillus vallismortis fmb-103 was cloned and heterologously expressed in Escherichia coli BL21 (DE3) cells. The auto-induction strategy was applied during fermentation, and the process was controlled, as follows: Cu2+ was added when the optical density at 600 nm (OD600) was 0.3, the fermentation temperature was adjusted to 16 °C when the OD600 was 0.9, and fermentation was stopped after 50 h. The yield of recombinant laccase was up to 3420 U/L, as assayed by 2,2-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid). Recombinant laccase was purified 4.47-fold by heating for 10 min at 70 °C and dialyzing against 50–60% ammonium sulfate, retained more than 50% activity after 10 h at 70 °C, and demonstrated broad pH stability. Malachite green was efficiently degraded by recombinant laccase, especially in combination with mediators. These results provided a basis for the future application of recombinant laccase to malachite green degradation.  相似文献

The objective of this work was to evaluate the post-treatment of an anaerobic recalcitrant effluent (anaerobically-treated weak black liquor, AnE) in an aerobic, upflow reactor packed with “biocubes” of Trametes versicolor immobilized onto small cubes of holm oak wood. The treated effluent (named anaerobic effluent; AnE) from an anaerobic fluidized bed reactor was fed to an up-flow aerobic fungal packed bed reactor (PBR). Two HRT were tested in this unit, namely 5 and 2.5 days; the PBR operated 60 days at 5-day HRT and 35 days at 2.5-day HRT. The aerobic packed bench scale reactor was a glass column 1.5 L total geometric volume containing 0.75 L biocubes of T. versicolor immobilized onto holm oak wood small cubes of 5 mm side. The reactor was operated at 25 °C. The pH of the AnE was adjusted to 4.5 before feeding; no carbohydrates or other soluble carbon source was supplemented. The fungal packed bed bioreactor averaged organic matter removals of 30% and 32% COD basis, during an experimental run of 60 days at 5-day HRT and 35 days at 2.5-day HRT, respectively. Colour and ligninoids contents were removed at higher percentages (69% and 54% respectively, average of both HRT). There was no significant difference between reactor performance at 5- and 2.5-day HRT, so, operation at 2.5-day HRT is recommended since reactor throughput is double. Activity of manganese peroxidase and laccase was found during the entire operation of the fungal PBR whereas lignin peroxidase activity practically disappeared in the second operation period. In general, enzyme activities were higher in the first period of operation (5-day HRT) than at 2.5-day HRT. To the best of our knowledge, this is one of the few works that demonstrated extended performance (3 months) of a fungal bioreactor for the treatment of a recalcitrant wastewater with no supplementation of glucose or other expensive, soluble carbohydrate.  相似文献

Ligninolytic enzyme production and polyphenolic compound extraction by liquid-state culture of Phanerochaete chrysosporium ATCC 24275 was investigated by employing apple pomace sludge and synthetic medium. Different physico-chemical and biological parameters namely viscosity, zeta potential and particle size, viability and enzyme production were investigated. The ligninolytic enzyme production was higher in apple pomace sludge (45 U/l of laccase, 220 U/l of MnP and 6.5 U/l of LiP) than in synthetic medium (17 U/l of laccase, 37 U/l of MnP and 6 U/l). These maximal activities were found during the stationary and decline phase. It was also found that enzyme production was strongly correlated with P. chrysoporium viability in both synthetic medium and apple pomace sludge. Moreover, physico-chemical parameters, such as particle size, zeta potential and viscosity were strongly correlated to the viability of P. chrysosporium and to the ligninolytic enzyme production. An increase in polyphenol content extracted by acetone (383–720 mg GAE/l) was observed during fermentation of apple pomace and it was found that the polyphenol content extracted by ethanol increased ~1.5 fold until 67 h of fermentation and later it decreased. It was found that antioxidant activity increased to 35% and eventually decreased based on the change in the polyphenol content.  相似文献

The extracellular laccase produced by the ascomycete Trichoderma atroviride was purified and characterized and its ability to transform phenolic compounds was determined. The purified laccase had activity towards typical substrates of laccases including 2,2′-azinobis-(3-ethylbenzthiazoline-6-sulphonate) (ABTS), dimethoxyphenol (2,6-DMP), syringaldazine and hydroquinone. The enzyme was a monomeric protein with an apparent molecular mass of 80 kDa and an isoelectric point of 3.5. The pH optima for the oxidation of ABTS and 2,6-DMP were 3 and 5, respectively, and the optimum temperature was 50 °C with 2,6-DMP. The laccase was stable at slightly acidic pH (4 and 5). It retained 80% of its activity after 4 h incubation at 40 °C. Under standard assay conditions, Km values of the enzyme were 2.5 and 1.6 mM towards ABTS and 2,6-DMP, respectively. This enzyme was able to oxidize aromatic compounds present in industrial and agricultural wastewater, as catechol and o-cresol, although the transformation of chlorinated phenols required the presence of ABTS as mediator.  相似文献

Polyamide matrices, such as membranes, gels and non-wovens, have been applied as supports for enzyme immobilization, although in literature the enzyme immobilization on woven nylon matrices is rarely reported. In this work, a protocol for a Trametes hirsuta laccase immobilization using woven polyamide 6,6 (nylon) was developed. A 24 full factorial design was used to study the influence of pH, spacer (1,6-hexanediamine), enzyme and crosslinker concentration on the efficiency of immobilization. The factors enzyme dosage and spacer seem to have played a critical role in the immobilization of laccase onto nylon support. Under optimized working conditions (29 U mL−1 of laccase, 10% of glutaraldehyde, pH = 5.5, with the presence of the spacer), the half-life time attained was about 78 h (18% higher than that of free enzyme), the protein retention was 30% and the immobilization yield was 2%. The immobilized laccase has potential for application in the continuous decolourization of textile effluents, where it can be applied into a membrane reactor.  相似文献

Laccase enzyme (L) from Trametes versicolor was entrapped in three hydrogel structures namely poly(acrylamide-N-isopropylacrylamide), P(AAm-NIPA), and semi-interpenetrating networks of poly(acrylamide)/alginate, P(AAm)/Alg, and poly(acrylamide-N-isopropylacrylamide)/alginate, P(AAm-NIPA)/Alg. The optimum temperatures for free and all immobilized systems were found to be 40 °C. For free and immobilized laccase systems of P(AAm-NIPA)-L, P(AAm)/Alg-L and P(AAm-NIPA)/Alg-L, Km values were found to be 6.7 × 10?3, 8.8 × 10?2, 5.5 × 10?2 and 1.8 × 10?2 mM; Vmax values were calculated as 1.8 × 10?3, 2.5 × 10?2, 1.5 × 10?2 and 6.1 × 10?3 mM min?1, respectively. For free and the same immobilized systems, the enzymes retained 42%, 91%, 79% and 86% of their initial activities at the end of 56 days of storage. After using the mentioned immobilized systems repeatedly 10 times, they retained 77%, 71% and 84% of their original activities, respectively. For free and the same immobilized systems, decolorization of Acid Orange 52 (AO52) in 6 h were found to be 63%, 50%, 48% and 66%, respectively. Addition of 2,2′-azino-bis-(3-ethylbenzothiazoline-6-sulfonic acid), ABTS, into the assay medium increased these values up to 73%, 73%, 74% and 75%, respectively.  相似文献

The potential application of dry biomass of a cyanobacterium Anacystis nidulans as a supplement in SSF for the production of laccase from Pleurotus ostreatus was evaluated. Experiments were carried out in solid culture using groundnut shell as a basic substrate supplemented with four independent nitrogen sources (ammonium sulphate, urea, yeast extract and dry powder of cyanobacteria). All the four supplements enhanced the enzyme yield, and yeast extract showed precedence over inorganic nitrogenous sources. However, when dry biomass of A. nidulans was used as an additive to groundnut shell (agricultural residues), it supported maximum cell growth (56.83 ± 5.56 mg/g dry substrate) and laccase production (49.21 ± 4.89 U/g dry substrate). Addition of 1 mM copper salt in the medium containing groundnut shell supplemented with yeast extract gave laccase activity of 32.64 ± 3.4 U/g dry substrate. When dry powder of cyanobacterial biomass was used as N-supplement, laccase production enhanced to 65.42 ± 6.48 U/g dry substrate. In addition to the enhancement to enzyme production inhibitory effects of high concentrations of copper was also diminished in the medium having dry cyanobacterial biomass. This study, forms the first report on the potential application of cyanobacterial biomass as an additive for production of laccase by Pleurotus ostraetus MTCC 1804 in solid state fermentation and has relevance in scale-up production of this fungal enzyme of commercial significance.  相似文献

A new laccase from Shiraia sp.SUPER-H168 was purified by ion exchange column chromatography and gel permeation chromatography and the apparent molecular mass of this enzyme was 70.78 kDa, as determined by MALDI/TOF-MS. The optimum pH value of the purified laccase was 4, 6, 5.5 and 3 with 2,6-dimethoxyphenol (DMP), syringaldazine, guaiacol and 2,2′-azinobis-(3-ethylbenzthiazoline-6-sulphonate) (ABTS) as substrates, respectively. The optimum temperature of the purified laccase was 50 °C using DMP, syringaldazine and guaiacol as substrates, but 60 °C for ABTS. Inhibitors and metal ions of SDS, NaN3, Ag+ and Fe3+ showed inhibition on enzyme activity of 10.22%, 7.86%, 8.13% and 67.50%, respectively. Fe2+ completely inhibited the purified laccase. The Kcat/Km values of the purified laccase toward DMP, ABTS guaiacol and syringaldazine were 3.99 × 106, 3.74 × 107, 8.01 × 104 and 2.35 × 107 mol?1 L S?1, respectively. The N-terminal amino acid sequence of the purified laccase showed 36.4% similarity to Pleurotus ostrestus. Approximately 66% of the Acid Blue 129 (100 mg L?1) was decolorized by 2.5 U of the purified laccase after a 120 min incubation at 50 °C.
