目的:研究Rab23在皮肤鳞状细胞癌(SCC)中的表达及意义。方法:用免疫组化S-P法分别检测30例皮肤SCC、15例正常皮肤组织标本中Rab23的表达。结果:Rab23在皮肤SCC和正常皮肤中阳性率分别为90%和13.3%,二者差异有统计学意义(P〈0.05)。结论:Rab23在皮肤SCC中高表达可能在皮肤SCC的发生发展过程中发挥作用。  相似文献

目的:观察Rab23在鳞癌细胞系中的表达及其对鳞癌细胞侵袭的影响。方法:western blot检测Rab23在四个头颈部鳞癌细胞系中的表达,对鳞癌细胞系HSQ-89用脂质体转染小干扰RNA,对Sa3用脂质体转染Rab23质粒,分别干涉或过表达Rab23,用铺有基质胶的transwell小室检测细胞侵袭能力,细胞划痕试验检测迁移能力。结果:Rab23在四个鳞癌细胞系中均有表达,在Hacat中不表达,HSQ-89细胞经Rab23干涉后细胞侵袭能力减弱,而Sa-3细胞过表达Rab23后细胞侵袭能力增强。干涉或过表达Rab23后,鳞癌细胞的迁移能力无差异。结论:Rab23可促进鳞癌细胞侵袭。  相似文献

Hedgehog信号通路在胚胎发育过程中发挥着重要作用, 同时与多种肿瘤的发生密切相关。Rab23蛋白在Hedgehog信号通路中扮演着十分重要的角色。目前关于文昌鱼Rab23同源基因的研究仅局限于佛罗里达文昌鱼(Branchiostoma floridae)基因组中的注释。文章首次克隆了中国文昌鱼(Branchiostoma belcheri) Rab23b基因 (AmphiRab23b)cDNA全序列 , 对其演译的蛋白序列进行了序列比对、进化树分析以及基因时空表达分析。研究结果显示, 文昌鱼AmphiRab23b基因的 cDNA总长为2 062 bp (包括UTR区), 其中开放阅读框 (Open reading frame, ORF) 714 bp, 编码237个氨基酸; 虽然在进化树中不属于脊椎动物Rab23进化支, 但是AmphiRab23具有保守的Rab23_lke结构域, 暗示该基因在进化过程中可能在功能上是保守的。时空表达的研究结果进一步显示, AmphiRab23b基因在胚胎发育中的神经板和消化道中表达, 与其脊椎动物同源基因的表达模式相似, 这说明该基因可能对文昌鱼神经系统和消化道的发育有重要作用。  相似文献

目的:探讨PTEN和β-cat在尖锐湿疣、Bowen's病和皮肤鳞癌中的表达及意义。方法:采用免疫组化法检测蛋白PTEN和β-cat在正常皮肤、尖锐湿疣、Bowen's病、皮肤鳞状细胞癌(Scc)中的表达。结果:在正常皮肤、尖锐湿疣、Bowen's病、皮肤鳞状细胞癌(Scc)中PTEN蛋白的高表达率分别为100%,80.95%,46.67%,36.67%;β-cat蛋白的异常表达率分别为16.67%,30.16%,53.33%,70.00%,与正常皮肤组织相比,Bowen's病及Scc中PTEN蛋白的高表达率表达下降(P<0.05,P<0.01);β-cat蛋白的异常表达率明显升高(P<0.05,P<0.01)。在Scc中两者的表达呈负相关(r=-0.416,P<0.05)。结论:蛋白PTEN、β-cat在Scc的发生、发展中有重要的意义。  相似文献

目的:研究人类mut-s同系物6(hMSH6)和P53蛋白在散发性结肠癌(SCC)中的表达及与SCC病理特征的关系。方法:采用免疫组化Max-Vision二步法检测48例SCC癌组织、距癌灶3~9 cm的癌旁组织及≥10 cm的结肠组织以及45例正常结肠组织中hMSH6、P53的表达情况。结果:癌组织、距癌灶3~9 cm的癌旁组织中,hMSH6的阳性表达率低于距癌灶≥10 cm结肠组织及正常结肠组织,P53的阳性表达率高于距癌灶≥10 cm结肠组织及正常结肠组织,差异比较有统计学意义(P0.05);hMSH6阳性表达与癌灶部位及浸润深度有关(P0.05);P53阳性表达与组织学分级有关(P0.05);hMSH6与P53阳性表达呈负相关(r=-0.403,P0.05)。结论:hMSH6、P53的异常表达参与了SCC发生及发展的过程,hMSH6、P53可作为评价SCC生物学行为的指标。  相似文献

目的:探讨蕈样肉芽肿、湿疹、正常皮肤中p53、Cox-2蛋白的表达差异性及其临床意义。方法:运用免疫组织化学法检测30例蕈样肉芽肿、20例湿疹及15例正常人皮肤组织中p53、Cox-2蛋白的表达情况。结果:蕈样肉芽肿表皮中及真皮中p53、Cox-2的阳性表达率显著高于湿疹及正常皮肤,差异均有统计学意义(P0.05)。各时期蕈样肉芽肿表皮、真皮中p53、Cox-2的表达比较差异均无统计学意义(P0.05)。结论:p53、Cox-2在蕈样肉芽肿和湿疹、正常皮肤的表达有显著差异,提示p53和Cox-2的表达异常在蕈样肉芽肿的发生中起着重要作用,p53、Cox-2可能作为鉴别诊断MF、皮肤炎症性疾病有价值的参考指标。  相似文献

构建Rab9 GTPase siRNA表达载体,观察siRNA对哺乳动物细胞内Rab9 GTPase表达的抑制作用,为应用Rab9 GTPase siRNA表达载体进行抗麻疹病毒感染研究奠定基础。根据Rab9 GTPase基因的mRNA序列设计合成2对靶向Rab9 GTPase基因的寡核苷酸,克隆到表达载体pSUPER.neo EGFP,通过双酶切和序列分析对重组表达载体进行鉴定。将鉴定为阳性的重组表达载体转染B95a细胞株,通过逆转录聚合酶链反应(RT-PCR)和免疫印迹技术(Western-blot)检测B95a细胞内Rab9 GTPase mRNA和蛋白的表达水平。双酶切和序列分析表明成功构建了Rab9 GTPase siRNA表达载体,RT-PCR和Western-blot技术实验表明重组表达载体可显著抑制B95a细胞内Rab9 GTPase mRNA和蛋白的表达(最高抑制率分别为89.4%±0.5%和87.6%±0.7%),而对照组则没有变化。结果表明,成功构建了Rab9 GTPase siRNA表达载体,该表达载体可有效地抑制Rab9 GTPase在哺乳动物细胞内的表达。  相似文献

目的:探讨新疆卡波氏肉瘤组织中膜型1型基质金属蛋白酶(Menberane 1 Matrix metalloproteinases,MT1-MMP),Rho蛋白解离抑制因子β(Rho GDP dissociation inhibitor GDI beta,ARHGDIB)的表达及临床意义.方法:应用免疫组织化学Envision二步法检测新疆17例卡波氏肉瘤和17例健康人正常皮肤组织内MT1-MMP和ARHGDIB的表达水平.结果:免疫组化显示MT1-MMP和ARHGDIB在卡波氏肉瘤组织阳性表达率为82.4%和100%,在正常皮肤组织中阳性率为6.9%和17.7%,差异均有统计学意义(P<0.01).结论:新疆卡波西肉瘤组织中MT1-MMP和ARHGDIB蛋白呈异常表达,有可能在KS的发展过程中发挥了重要作用.  相似文献

[目的]Rab19属于Ras-GTPase基因家族成员,探讨Rab19表达在胃癌诊断和预后中的价值。[方法]从肿瘤基因组图谱(The Cancer Genome Atlas,TCGA)数据库中下载胃癌数据集表达谱及临床病理特征资料。评估Rab19 mRNA在GC中的表达水平。通过Kaplan-Meier和Cox分析,确定Rab19在胃癌中的预后价值。使用TCGA数据集进行基因集富集分析(GSEA)以预测胃癌中Rab19作用的分子机制。[结果]Rab19在胃癌中过表达(P 0.0001),且Rab19的表达与肿瘤分级和瘤体大小都有显著相关(P 0.05)。生存分析结果显示Rab19高表达与胃癌患者生存时间长相关(P 0.05)。Rab19表达可作为胃癌患者的独立预后因子(P 0.005)。此外,过氧化物酶体、状腺癌、泛酸和Co A生物合成、鞘脂代谢、细胞凋亡、硒氨基酸代谢、Notch信号传导途径和子宫内膜癌相关的基因集与Rab19的高表达表型存在差异。[结论]Rab19在胃癌细胞中明显上调,有望成为胃癌的诊断和预后中的有效生物标志物。  相似文献

通过对6种藓类植物,即褶叶青藓(Brachythecium salebrosum(Web.et Mohr.)B.S.G.)、湿地匐灯藓(Plagiomnium acutum(Lindb.)Kop.)、侧枝匐灯藓(Plagiomnium maximoviczii(Lindb.)Kop.)、大凤尾藓(Fissidensnobilis Griff.)、大羽藓(Thuidium cymbifolium(Doz.et Molk.)B.S.G.)和大灰藓(Hypnum plumaeforme Wils.)嫩茎和老茎的石蜡切片和显微观察发现,同一藓类植株的嫩茎和老茎,茎结构稳定,不同种藓类植物茎横切面具有不同特征.植物体茎横切面形状、表层细胞的层数、细胞大小和细胞壁厚薄、皮层细胞大小和形状、中轴的有无以及比例等特征可以作为藓类植物的分科分类依据之一.  相似文献

The levels of endogenous phytohormones and respiratory rate in nine sorts of flowers such as Cymbidium faberi Rolfe, Nopalxochia ackermannii Kunth and others were investigated both at full bloom and senescence and meanwhile the effect of exogenous phytohormones on prolonging the blossoms and promoting ethylene production were tested. There is a high content of endogenous ethylene in all the long-lived flowere, about 3–16 folds higer than the short-lived ones. There is a high level of ABA at full blooming flowers of short-lived flowers, in which there is no or only some cytokinins in it, but the ratio of CTK (6BA+zeatin)/ABA is smaller(l.7). The endogenous ABA reached a much higher level at senescence in all nine sorts of flowers, so it is reasonable to consider that it is ABA which plays an important role of regulation in controlling flower's senescence. There is a much higher level of GA3 and zeatin in the long-lived flowers which is not demonstrated in the shortlived ones. The respiratory rate is one of the factors controtling the longevity of flowers, but it does not play a decided role. Application of 6BA and zeatin prolongs distinctly orchid’s longevity, however exogenous IAA through the promotive action on ethylene production, evidently extends the longevity of the flowers of the Nopalxochia ackermannii Kunth.  相似文献

Summary Anthers were cultured from two sets of seven lines of hexaploid wheat (Triticum aestivum L.) with different cytoplasms, the euplasmic nucleus donors, Siete Cerros 66 and Penjamo 62, as well as their six alloplasmic lines derived from wild relative species of the genera Triticum and Aegilops. Significant cytoplasmic and nuclear effects but no cytoplasmic-nuclear interaction were found for embryogenic anther response, with the best performance of Penjamo 62 in Ae. kotschyi cytoplasm. Plant regeneration was not affected significantly by the cytoplasmic background of the lines cultured. The possible genetic implications of the observed cytoplasmic and nuclear influences on the in vitro haploid induction of wheat are discussed.  相似文献

Summary Embryogenic tissues of sugi (Cryptomeria japonica) were induced on a modified Campbell and Durzan (CD) medium containing 1 μM 2,4-dichlorophenoxyacetic acid (2,4-D) and 600 mg l−1 glutamine, and subcultured in the medium of the same composition for over 1 yr. This resulted in a mixed culture of embryogenic and non-embryogenic cells. When embryogenic cells were isolated and cultured independently, their capacity to form embryogenic aggregates was lost. Thus, the non-embryogenic cells present within a mixed culture system were essential to the formation of embryogenic aggregates. When embryogenic tissues were isolated and cultured independently on a high glutamine-containing (2400 mg l−1) medium, dry weights and endogenous levels of glutamine increased, and the tissue could generate a large number of embryogenic aggregates. Amino acid analysis of embryogenic and non-embryogenic cells from the maintenance culture indicated a higher level of glutamine was present in the latter. The high endogenous level of glutamine in the non-embryogenic portion of mixed cell masses may be the supplier of glutamine for maintaining the embryogenic property of the tissues.  相似文献

Seed germination of an aurea mutant of tomato ( Lycopersicon esculentum Mill.) is promoted by continuous irradiation with red, far-red or long-wavelength far-red (758 nm) light as well as by cyclic irradiations (5 min red or 5 min far-red/25 min darkness). Far-red light applied immediately after each red does not change the germination behaviour. Seed germination of the isogenic wild-type, cv. UC-105, is promoted by continuous and cyclic red light while it is inhibited by continuous and cyclic far-red light and by continious 758 nm irradiation. Far-red irradiation reverses almost completely the promoting effect of red light. The promoting effect (in the aurea mutant) and the inhibitory effect (in the wild-type) of continuous far-red light do not show photon fluence rate dependency above 20 nmol m−2 s−1. It is concluded that phytochrome controls tomato seed germination throgh low energy responses in both the wild type and the au mutant. The promoting effect of continuous and cyclic far-red light in the au mutant can be attributed to a greater sensitivity to Pfr.  相似文献
