Pisum arvense plants were subjected to 5 days of nitrogen deprivation. Then, in the conditions that increased or decreased the root glutamine and asparagine pools, the uptake rates of 0.5 mM NH4 + and 0.5 mM K+ were examined. The plants supplied with 1 mM glutamine or asparagine took up ammonium and potassium at rates lower than those for the control plants. The uptake rates of NH4 + and K+ were not affected by 1 mM glutamate. When the plants were pre-treated with 100 μM methionine sulphoximine, an inhibitor of glutamine synthesis, the efflux of NH4 + from roots to ambient solution was enhanced. On the other hand, exposure of plants to methionine sulphoximine led to an increase in potassium uptake rate. The addition of asparagine, glutamine or glutamate into the incubation medium caused a decline in the rate of NH4 + uptake by plasma membrane vesicles isolated from roots of Pisum arvense, whereas on addition of methionine sulphoximine increased ammonium uptake. The results indicate that both NH4 + and K+ uptake appear to be similarly affected by glutamine and asparagine status in root cells. The kinetics of NH4 + and NO3 uptake in young Douglas fir trees (Pseudotsuga menziesii [Mirb.] Franco) were studied in solutions, containing either one or both N species. Using solutions containing a single N species, the Vmax of NH4 + uptake was higher than that of NO3 uptake. The Km of NH4 + uptake and Km of NO3 uptake differed not significantly. When both NH4 + and NO3 were present, the Vmax for NH4 + uptake became slightly higher, and the Km for NH4 + uptake remained in the same order. Under these conditions the NO3 uptake was almost totally inhibited over the whole range of concentrations used (10–1000 μM total N). Summary The purpose of this study was to investigate the phytotoxicity of nitrapyrin 2-chloro-6-(trichloromethyl)pyridine to sunflower (Helianthus annuus L.) under different N regimes and to see if N forms affect the phytotoxicity of nitrapyrin. Sunflower was grown in pot culture for 21 days and was fertilized with (NH4)2SO4, NH4NO3 and NaNO3 to provide 0, 100 and 200 ppm N and with nitrapyrin application of 0 and 20 ppm. All N-treated sunflower plants in all N regimes and regardless of titrapyrin treatment produced more root and shoot dry weights and contained a significantly higher N than untreated check. Nitrapyrin toxicity appeared as a curling of leaf margin and a tendril type of stem growth, the visible toxicity symptoms decreased in the order: (NH4)2SO4>NH4NO3>NaNO3. Furthermore nitrapyrin addition suppressed sunflower growth in each N regime, the suppressing effect being greater with (NH4)2SO4 and NH4NO3 than as with NaNO3. Although, shoot growth from plants receiving nitrapyrin was not significantly affected by any N regime, root growth of nitrapyrin-treated plants was somewhat restricted by NH4 +−N nutrition relative to NO3 −N nutrition.  相似文献

In short-term water culture experiments with different 15N labeled ammonium or nitrate concentrations, citrus seedlings absorbed NH4 + at a higher rate than NO3 . Maximum NO3 uptake by the whole plant occurred at 120 mg L–1 NO3 -N, whereas NH4 + absorption was saturated at 240 mg L–1 NH4 +-N. 15NH4 + accumulated in roots and to a lesser degree in both leaves and stems. However, 15NO3 was mostly partitioned between leaves and roots.Adding increasing amounts of unlabeled NH4 + (15–60 mg L–1 N) to nutrient solutions containing 120 mg L–1 N as 15N labeled nitrate reduced 15NO3 uptake. Maximum inhibition of 15NO3 uptake was about 55% at 2.14 mM NH4 + (30 mg L–1 NH4 +-N) and it did not increase any further at higher NH4 + proportions.In a long-term experiment, the effects of concentration and source of added N (NO3 or NH4 +) on nutrient concentrations in leaves from plants grown in sand were evaluated. Leaf concentration of N, P, Mg, Fe and Cu were increased by NH4 + versus NO3 nutrition, whereas the reverse was true for Ca, K, Zn and Mn.The effects of different NO3 -N:NH4 +-N ratios (100:0, 75:25, 50:50, 25:75 and 0:100) at 120 mg L–1 total N on leaf nutrient concentrations, fruit yield and fruit characteristics were investigated in another long-term experiment with plants grown in sand cultures. Nitrogen concentrations in leaves were highest when plants were provided with either NO3 or NH4 + as a sole source of N. Lowest N concentration in leaves was found with a 75:25 NO3 -N/NH4 +-N ratio. With increasing proportions of NH4 + in the N supply, leaf nutrients such as P, Mg, Fe and Cu increased, whereas Ca, K, Mn and Zn decreased. Yield in number of fruits per tree was increased significantly by supplying all N as NH4 +, although fruit weight was reduced. The number of fruits per tree was lowest with the 75:25 NO3 -N:NH4 +-N ratio, but in this treatment fruits reached their highest weight. Rind thickness, juice acidity, and colour index of fruits decreased with increasing NH4 + in the N supply, whereas the % pulp and maturity index increased. Percent of juice in fruits and total soluble solids were only slightly affected by NO3 :NH4 + ratio.  相似文献

Although many studies support the importance of the external mycelium for nutrient acquisition of ectomycorrhizal plants, direct evidence for a significant contribution to host nitrogen nutrition is still scarce. We grew nonmycorrhizal seedlings and seedlings mycorrhizal with Paxillus involutus (Batsch) Fr. in a sand culture system with two compartments separated by a 45-m Nylon mesh. Hyphae, but not roots, can penetrate this net. Nutrient solutions were designed to limit seedling growth by nitrogen. Hyphal density in the hyphal compartment, host N status and shoot growth of mycorrhizal seedlings significantly increased in response to NH4 + addition to the hyphal compartment. Labeling the compartment only accessible to hyphae with 15NH4 + showed that the increase in N uptake in the mycorrhizal seedlings was a result of hyphal N acquisition from the hyphal compartment. These results indicate that hyphae of P. involutus may actively forage into N-rich patches and improve host N status and growth. In the mycorrhizal seedlings, which received additional NH4 + via their external mycelium, the increase in NH4 + supply less negatively affected Ca and Mg uptake than in nonmycorrhizal seedlings, where the additional NH4 + was directly supplied to the roots. This was most likely due to the close link of NH4 + uptake and H+ extrusion, which, in the nonmycorrhizal seedlings, lead to a strong acidification in the root compartment, and subsequently reduced Ca and Mg uptake, whereas in the mycorrhizal seedlings the site of intensive NH4 + uptake and acidification was in the hyphal and not in the root compartment. Our data support the idea that the ectomycorrhizal mycelium connected to an N-deficient host may actively forage for N. The mycelium may also be important as a biological buffer system ameliorating negative influence of high NH4 + supply on cation uptake.  相似文献

Wheat plants grown during 10 days in the absence of N were pretreated with 1.0 eq m-3 of methionine, asparagine or glutamine and/or 1.0 eq m-3 MSX4 or 0.17 eq m-3 DON. Net NH4 + uptake was measured both in the presence or in the absence of the amino acid or enzyme inhibitor used in the pretreatment. The effect of met, asn and gln on net K+ uptake was also studied using K+-depleted plants. Changes in the contents of root free NH4 +, asn, gln and the activities of GS, PEP-carboxylase, NAD+-GDH and NADH-GDH were determined. Net NH4 + uptake in gln and asn pretreated plants was markedly, and sometimes completely suppressed provided uptake was measured in the presence of the amides. On the other hand, the met pretreated plants absorbed only 35% less NH4 + than the control. When NH4 + uptake was measured in the absence of the amino acids, only those plants pretreated with asn showed a marked suppression of net uptake during the first 120 min. None of the 3 amino acids tested significantly inhibited K+ uptake. Free NH4 + concentration in roots of N-starved plants increased after 4 h incubation with gln, asn or MSX in the absence of external NH4 +. Nevertheless, no correlation was observed between root NH4 + concentration and the extent of net NH4 + uptake suppression. The inhibitory effect exerted by asn decreased when it was supplied together with MSX or DON. Pretreatments with gln or asn in the absence of external NH4 + significantly increased the level of asn in the roots, while that of gln remained unchanged. The effects of metabolic and protein synthesis inhibitors on NH4 + uptake by Pisum arvense plants at low (0.05 mM) and high (1 mM) external ammonium concentration were studied. In short-time experiments cycloheximide decreased the ammonium uptake rate at low level of NH4 + and increased the absorption of NH4 + from uptake medium containing high ammonium concentration. Arsenate and azide supplied into uptake solutions at low ammonium concentration strongly decreased or completely suppressed the NH4 + uptake rate, respectively. When the experiments were carried out at high level of ammonium only azide decreased the uptake rate of NH4 + and arsenate stimulated this process. Dinitrophenol very strongly repressed the uptake rate of NH4 + at both ammonium concentrations. After removing dinitrophenol from both solutions, neither at low nor high external ammonium level the recovery of NH4 + uptake rate was achieved within 150 min or 3 h, respectively. The recovery of NH4 + uptake rate after removing azide was observed within 90 min and 3 h at low and high ammonium concentrations, respectively. The regulation of NH4 + uptake by some inhibitors at low external ammonium level was investigated using plasma membrane vesicles isolated from roots by two-phase partitioning. Orthovanadate completely suppressed the uptake of NH4 + by vesicles and quinacrine decreased the NH4 + uptake which 55 suggests that ammonium uptake depends on activities of plasma membrane-bound enzymes. On the other hand, it was found that dinitrophenol completely reduced the NH4 + uptake by vesicles. The various effects of inhibitors on ammonium uptake dependent on external ammonium concentration suggest the action of different ammonium transport systems in Pisum arvense roots. The author studied the effect of different nickel concentrations (0, 0.4, 40 and 80 μM Ni) on the nitrate reductase (NR) activity of New Zealand spinach (Tetragonia expansa Murr.) and lettuce (Lactuca sativa L. cv. Justyna) plants supplied with different nitrogen forms (NO3 –N, NH4 +–N, NH4NO3). A low concentration of Ni (0.4 μM) did not cause statistically significant changes of the nitrate reductase activity in lettuce plants supplied with nitrate nitrogen (NO3 –N) or mixed (NH4NO3) nitrogen form, but in New Zealand spinach leaves the enzyme activity decreased and increased, respectively. The introduction of 0.4 μM Ni in the medium containing ammonium ions as a sole source of nitrogen resulted in significantly increased NR activity in lettuce roots, and did not cause statistically significant changes of the enzyme activity in New Zealand spinach plants. At a high nickel level (Ni 40 or 80 μM), a significant decrease in the NR activity was observed in New Zealand spinach plants treated with nitrate or mixed nitrogen form, but it was much more marked in leaves than in roots. An exception was lack of significant changes of the enzyme activity in spinach leaves when plants were treated with 40 μM Ni and supplied with mixed nitrogen form, which resulted in the stronger reduction of the enzyme activity in roots than in leaves. The statistically significant drop in the NR activity was recorded in the aboveground parts of nickel-stressed lettuce plants supplied with NO3 –N or NH4NO3. At the same time, there were no statistically significant changes recorded in lettuce roots, except for the drop of the enzyme activity in the roots of NO3 -fed plants grown in the nutrient solution containing 80 μM Ni. An addition of high nickel doses to the nutrient solution contained ammonium nitrogen (NH4 +–N) did not affect the NR activity in New Zealand spinach plants and caused a high increase of this enzyme in lettuce organs, especially in roots. It should be stressed that, independently of nickel dose in New Zealand spinach plants supplied with ammonium form, NR activity in roots was dramatically higher than that in leaves. Moreover, in New Zealand spinach plants treated with NH4 +–N the enzyme activity in roots was even higher than in those supplied with NO3 –N.  相似文献

Three-year-old Scots pine (Pinus sylvestris) trees were grown on a sandy forest soil in pots, with the objective to determine their NH4/NO3 uptake ratio and proton efflux. N was supplied in three NH4-N/NO3-N ratios, 3:1, 1:1 and 1:3, either as 15NH4+14NO3 or as 14NH4+15NO3. Total N and 15N acquisition of different plant parts were measured. Averaged over the whole tree, the NH4/NO3 uptake ratios throughout the growing season were found to be 4.2, 2.5, and 1.5 for the three application ratios, respectively. The excess cation-over-anion uptake value (Ca-Aa) appeared to be linearly related to the natural logarithm of the NH4/NO3 uptake ratio. Further, this uptake ratio was related to the NH4/NO3 ratio of the soil solution. From these relationship it was estimated that Scots pine exhibits an acidifying uptake pattern as long as the contribution of nitrate to the N nutrition is lower than 70%. Under field circumstances root uptake may cause soil acidification in the topsoil, containing the largest part of the root system, and soil alkalization in deeper soil layers.  相似文献

Responses to inoculation with N2-fixing bacteria were studied in relation to genotypic differences in pearl millet, effect of nitrogen levels, and FYM additions in India. In some experiments, inoculation increased mean grain yield up to 33% over the uninoculated control, whereas in the remaining 11 experiments there was no significant increase. Increased grain yields, >10% over the uninoculated controls were observed in 46% of the experiments withAzospirillum lipoferum (18.7% average increase) and withAzotobacter chroococcum (13.6% average increase). Yield increases were nil or reduced in three experiments withAzos. lipoferum and four experiments withAztb. chroococcum. In two experiments continued inoculation for two or three years resulted in increased grain, plant biomass yield, and N uptake. Interactions of bacterial cultures with cultivars or years were not observed. The counts of the inoculated strains increased two to three-fold when inoculation was continued for three years. Repeated inoculations increased the mean cumulative N uptake from season 1 to season 3 by 19 kg ha–1. Repeated inoculations withAztb. chroococcum andAzos. lipoferum increased mean grain yield of a succeeding crop by 14.4% and 9.8%, respectively, over the uninoculated control. Inoculation increased the efficiency of N-assimilation by pearl millet. Marginal increase in nitrogenase activity, associated with the inoculated plants was observed during later stages of plant growth. Increased leaf nitrate reductase activity (NRA) was observed after inoculation with these bacteria. The responses to inoculation are mainly attributable to increased plant N assimilation which could be the effect of growth promoting substances secreated by the bacteria; and thus the contribution from BNF may be small.CRISAT, journal article 732.  相似文献

The effect of external inorganic nitrogen and K+ content on K+ uptake from low-K+ solutions and plasma membrane (PM) H+-ATPase activity of sorghum roots was studied. Plants were grown for 15 days in full-nutrient solutions containing 0.2 or 1.4 mM K+ and inorganic nitrogen as NO3-, NO3-/NH4+ or NH4+ and then starved of K+ for 24, 48 and 72 h. NH4+ in full nutrient solution significantly affected the uptake efficiency and accumulation of K+, and this effect was less pronounced at the high K+ concentration. In contrast, the translocation rate of K+ to the shoot was not altered. Depletion assays showed that plants grown with NH4+ more efficiently depleted the external K+ and reached higher initial rates of low-K+ uptake than plants grown with NO3-. One possible influence of K+ content of shoot, but not of roots, on K+ uptake was evidenced. Enhanced K+-uptake capacity was correlated with the induction of H+ extrusion by PM H+-ATPase. In plants grown in high K+ solutions, the increase in the active H+ gradient was associated with an increase of the PM H+-ATPase protein concentration. In contrast, in plants grown in solutions containing 0.2 mM K+, only the initial rate of H+-pumping and ATP hydrolysis were affected. Under these conditions, two specific isoforms of PM H+-ATPase were detected, independent of the nitrogen source and deficiency period. No change in enzyme activity was observed in NO3--grown plants. The results suggest that K+ homeostasis in NH4+-grown sorghum plants may be regulated by a high capacity for K+ uptake, which is dependent upon the H+-pumping activity of PM H+-ATPase.  相似文献

Summary Non-nodulatedalnus glutinosa plants were grown for 6 weeks in nutrient solutions using 3 combined-N treatments (NO3; NO3/NH4; and NH4) at a total N level of 4 meq.l–1, and growth was ccmpared with nodulated plants at zero N (N2 fixation). Of the combined-N sources, 100 per cent NH4 resulted in the highest dry matter yields when the solution pH was adjusted daily atc. 6. The dry matter yield was lowest with NO3.During the first 3 weeks, the yield of the N2-fixing plants was as high as that of the NH4 plants, but fell relatively behind during the second 3-week period. These effects could be attributed to higher initial N contents and higher shoot:root ratios, respectively, in the N2-fixing plants. Specific rates of N acquisition in the root were of a comparable order of magnitude for the combined-N and zero-N treatments.When NO3 was taken up, it was almost completely reduced in the roots. Regardless of N source there was a large excess of cations (C) relative to inorganic anions (A) in the plants, which was presumed to be balanced by an equivalent amount of organic anions (C-A). The relatively small differences in generation of organic anions for the various modes of N supply indicated the relative importance of the proton pump when NH4 or N2 was the N source. Proton or hydroxyl-ion effluxes, calculated on the basis of plant analyses, were generally in good agreement with measured excretion values. The acidity generation with N2 fixation amounted toc. 0.5 meq H+.mmol–1 Norg, which was distinctly higher than the range of 0.1–0.2 mentioned by Raven and Smith43 for dinitrogen-fixing plants.Without pH adjustment, specific rates of cation uptake and carboxylate generation were strongly depressed as the acidity increased, when NO3/NH4, NH4 and N2 were the N sources. Growth ofAlnus glutinosa appeared to be still normal at a pH ofc. 2.8. During the final 3 weeks, only the NH4 plants ceased growing at a pH of 2.6.  相似文献

New information on N uptake and transport of inorganic and organic N in arbuscular mycorrhizal fungi is reviewed here. Hyphae of the arbuscular mycorrhizal fungus Glomus mosseae (Nicol. and Gerd.) Gerd. and Trappe (BEG 107) were shown to transport N supplied as 15N-Gly to wheat plants after a 48 h labelling period in semi-hydroponic (Perlite), non-sterile, compartmentalised pot cultures. Of the 15N supplied to hyphae in pot cultures over 48 h, 0.2 and 6% was transported to plants supplied with insufficient N or sufficient N, respectively. The increased 15N uptake at the higher N supply was related to the higher hyphal length density at the higher N supply. These findings were supported by results from in vitro and monoxenic studies. Excised hyphae from four Glomus isolates (BEG 84, 107, 108 and 110) acquired N from both inorganic (15NH4 15NO3, 15NO3 or 15NH4 +) and organic (15N-Gly and 15N-Glu, except in BEG 84 where amino acid uptake was not tested) sources in vitro during short-term experiments. Confirming these studies under sterile conditions where no bacterial mineralisation of organic N occurred, monoxenic cultures of Glomus intraradices Schenk and Smith were shown to transport N from organic sources (15N-Gly and 15N-Glu) to Ri T-DNA transformed, AM-colonised carrot roots in a long-term experiment. The higher N uptake (also from organic N) by isolates from nutrient poor sites (BEG 108 and 110) compared to that from a conventional agricultural field implied that ecotypic differences occur. Although the arbuscular mycorrhizal isolates used contributed to the acquisition of N from both inorganic and organic sources by the host plants/roots used, this was not enough to increase the N nutritional status of the mycorrhizal compared to non-mycorrhizal hosts. Developing tumors induced by Agrobacterium tumefaciens, strain C58, on stems of Ricinus communis L. var. gibsonii cv. Carmencita were shown to be strong metabolic sinks for sucrose and amino acids, thus causing higher nutrient demand in the host plant. However, NO3 uptake and, to a lesser extent, also H2PO4 uptake were strongly inhibited. Correspondingly, NO3 concentration was lower in tumorised than in the control plants. NO3 reductase activity was the same in both plant types, but it was completely suppressed in the tumors. The electrical membrane potential difference of root cells was unaffected in tumorised plants when soil-grown, but significantly lowered when grown hydroponically. Consistent with the low NO3 uptake rate, NO3 -dependent membrane depolarisation at the onset of NO3 /2H+-cotransport was nearly zero. In the phloem sap, sucrose and amino acid concentrations were considerably lower in tumorised than in control plants, and lower below than above the tumor. The qualitative pattern of amino acids of the phloem sap of stems was almost the same in tumorised and control plants. It is concluded that neither the overall amino acid concentration nor special amino acids nor ammonium in the transport phloem suppress NO3 uptake in the roots. Aminocyclopropane-carboxylate, the precursor of ethylene, which is produced in the tumors in high amounts, was low in the stems and the same in both plant types. Thus, ACC and ethylene were ruled out as directly interfering with nutrient uptake in the roots. Root morphology was strongly affected during tumor development. Root fresh weight decreased to 50% of the controls and lateral root development was almost completely prevented. This suggests that the high tumor ethylene production, together with an increasing concentration of phenolic compounds, severely inhibits the basipetal auxin flow to the roots. Auxin accumulation and retention was confirmed by specifically enhanced expression of the auxin-responsive promoter of the soybean gene GH3:GUS in tumors induced in transgenic Trifolium repens L. Hence, root development is poorer and anion uptake inhibited in tumorised plants. This may be aggravated by abscisic acid accumulation in the tumor and its basipetal export into the roots. Moreover, sucrose depletion of the sieve tubes leads to energy shortage at the root level for maintaining energy-dependent anion uptake.  相似文献

Summary A microplot field experiment was conducted in the presence or absence of P and N application to evaluate the influence of the seed inoculation of mustard (cv. Baruna T59) withAzospirillum lipoferum on N2-fixation in rhizosphere, association of the bacteria with the roots and grain yield and N uptake. Inoculation significantly increased the N content in rhizosphere soil particularly at early stage (40 days) of plant growth, which was accompanied by the increased association of the bacteria (A. lipoferum) in rhizosphere soil, root surface washing and surface-sterilized macerated root. A significant increase in grain yield and N uptake was also observed due to inoculation. Application of P particularly at the 20 kg. ha–1 level further enhanced the beneficial effect ofAzospirillum lipoferum inoculation, while N addition markedly reduced such an effect.  相似文献

The carbon and nitrogen partitioning characteristics of wheat (Triticum aestivum L.) and maize (Zea mays L.) grown hydroponically at a constant pH on either 4 mM or 12 mM NO3 - or NH4 + nutrition were investigated using either 14C or 15N techniques. Greater allocation of 14C to amino-N fractions occurred at the expense of allocation of 14C to carbohydrate fractions in NH4 +-compared to NO3 --fed plants. The [14C]carbohydrate:[14C]amino-N ratios were 1.5-fold and 2.0-fold greater in shoots and roots respectively of 12 mM NO3 --compared to 12 mM NH4 +-fed wheat. In both 4 mM and 12 mM N-fed maize the [14C]carbohydrate:[14C]amino-N ratios were approximately 1.7-fold and 2.0-fold greater in shoots and roots respectively of NO3 --compared to NH4 +-fed plants. Similar results were observed in roots of wheat and maize grown in split-root culture with one root-half in NO3 --and the other in NH4 +-containing nutrient media. Thus the allocation of carbon to the amino-N fractions occurred at the expense of carbohydrate fractions, particularly within the root. Allocation of 14N and 15N within separate sets of plants confirmed that NH4 --fed plants accumulated more amino-N compounds than NO3 --fed plants. Wheat roots supplied with 15NH4 + for 8 h were found to accumulate 15NH4 + (8.5 g 15N g-1 h-1) whereas in maize roots very little 15NH4 + accumulated (1.5 g 15N g-1 h-1)It is proposed that the observed accumulation of 15NH4 + in wheat roots in these experiments is the result of limited availability of carbon within the roots of the wheat plants for the detoxification of NH4 +, in contrast to the situation in maize. Higher photosynthetic capacity and lower shoot: root ratios of the C4 maize plants ensure greater carbon availability to the root than in the C3 wheat plants. The effect of two N-forms (NH4 + and NO3 ) and NaCl on pattern of accumulation of some essential inorganic nutrients was examined in sunflower (Helianthus annuus L.) cv. Hisun 33. Eight-day-old plants of were subjected for 21 d to Hoagland's nutrient solution containing 8 mM N as NH4 + or NO3 ·, and salinized with and addition of NaCl to the growth medium had no significant effect on total leaf N. However, root N of NH4-supplied plants decreased significantly with increase in NaCl concentration, whereas that of NO3-supplied plants remained unaffected. There was no significant effect of NaCl on leaf or root P, but the NO3-supplied plants had significa concentration of leaf P than that of NH4-supplied plants at varying salt treatments. Salinity of the rooting med did not show any significant effect on Na+ concentrations of leaves or roots of plants subjected to two differen N. NH4-treated plants generally had greater concentrations of Cl in leaves and roots and lower K+ content in leaves than NO3-supplied plants. Ca2+ concentrations of leaves and roots and Mg2+ concentrations of leaves decreased in NH4-supplied plants due to NaCl, but they remained unaffected in NO3-treated plants.  相似文献

After growth for 17 to 36 days on nutrient solutions with NH4NO3 as nitrogen source (pH 4.2) dry matter of sorghum genotype SC0283 was much less affected by Al (1.5 and 3.0 ppm) than that of genotype NB9040. In the absence of Al both cultivars released protons into the nutrient solution as a result of an excess of cationic nutrients taken up. When Al was present, this proton efflux per unit dry weight increased drastically, especially with the sensitive genotype NB9040. Chemical analysis of plant material and continuous analyses of NO 3 and NH 4 + in the nutrient solution indicated, that the Al-induced shift in H+-balance of both genotypes could almost completely be attributed to a decreased NO 3 /NH 4 + uptake ratio. In vivo nitrate reductase activity (NRA) was reduced in the shoot of NB9040 and to a lesser degree in SC0283. Al-induced decrease in NRA was accompanied by similar percentual decreases in NO 3 tissue concentrations. Therefore this decrease is interpreted as being indirect,i.e., the consequence of the reduced NO 3 uptake of the plants. A direct repression of NRA by Al seems also unlikely because nitrate reductase activity of the roots (where cellular Al-concentrations should be higher than in shoots) was not affected in Al-treated plants of either genotype.  相似文献

为探讨氮沉降对九寨沟藓类植物的影响,该研究以当地优势藓类植物锦丝藓(Actinothuidium hookeri)和塔藓(Hylocomium splendens)为对象,以NH4NO3为氮源,设置对照(0 kg N·hm-2·a-1)、低浓度(20 kg N·hm-2·a-1)、高浓度(50 kg N·hm-2·a-1)3种处理,开展为期6个月的氮沉降模拟实验。结果表明:(1)氮沉降处理导致两种藓类植物的活性氧、丙二醛、叶绿素、脯氨酸和可溶性蛋白含量显著增加,同时锦丝藓过氧化氢酶、过氧化物酶、超氧化物歧化酶、抗坏血酸过氧化物酶活性增加。(2)对于生长旺期和生长末期的塔藓,氮沉降导致其过氧化物酶、过氧化氢酶、抗坏血酸过氧化物酶活性降低。(3)锦丝藓的综合隶属函数值随氮沉降浓度增大而增加,在生长旺期和生长末期,塔藓综合隶属函数值对氮沉降的响应存在差异。综上认为,两种藓类植物对氮沉降处理的生理响应存在差异,高浓度氮沉...  相似文献

Growth, chemical composition, and nitrate reductase activity (NRA) of hydroponically cultured Rumex crispus, R. palustris, R. acetosa, and R. maritimus were studied in relation to form (NH4 +, NO3 -, or both) and level of N supply (4 mM N, and zero-N following a period of 4mM N). A distinct preference for either NH4 + or NO3 - could not be established. All species were characterized by a very efficient uptake and utilization of N, irrespective of N source, as evident from high concentrations of organic N in the tissues and concurrent excessive accumulations of free NO3 - and free NH4 +. Especially the accumulation of free NH4 + was unusually large. Generally, relative growth rate (RGR) was highest with a combination of NH4 + and NO3 -. Compared to mixed N supply, RGR of NO3 -- and NH4 +-grown plants declined on average 3% and 9%, respectively. Lowest RGR with NH4 + supply probably resulted from direct or indirect toxicity effects associated with high NH4 + and/or low Ca2+ contents of tissues. NRA in NO3 - and NH4NO3 plants was very similar with maxima in the leaves of ca 40 μmol NO2 - g-1 DW h-1. ‘Basal’ NRA levels in shoot tissues of NH4 + plants appeared relatively high with maxima in the leaves of ca 20 μmol NO2 - g-1 DW h-1. Carboxylate to organic N ratios, (C-A)/Norg, on a whole plant basis varied from 0.2 in NH4 + plants to 0.9 in NO3 - plants. After withdrawal of N, all accumulated NO3 - and NH4 + was assimilated into organic N and the organic N redistributed on a large scale. NRA rapidly declined to similar low levels, irrespective of previous N source. Shoot/root ratios of -N plants were 50–80% lower than those from +N plants. In comparison with +N, RGR of -N plants did not decline to a large extent, decreasing by only 15% in -NH4 + plants due to very high initial organic-N contents. N-deprived plants all exhibited an excess cation over anion uptake (net proton efflux), and whole-plant (C-A)/Norg ratios increased to values around unity. Possible difficulties in interpreting the (C-A)/Norg ratio and NRA of plants in their natural habitats are briefly discussed.  相似文献
