Few useful microsatellites are available for population studies of the European corn borer, Ostrinia nubilalis (Hübner). An enrichment strategy was used to develop microsatellite markers for O. nubilalis, and over 500 positive clones were isolated. Seventy-five contained unique microsatellites, 10 of which were polymorphic with discernable polymerase chain reaction products. The 10 loci were surveyed for variability in 72 wild individuals from central Iowa. Five loci showed no deviation from Hardy-Weinberg proportions, and all were successfully cross-amplified in the related Asian corn borer, Ostrinia furnacalis. These loci represent a significant addition to microsatellites appropriate for population studies of O. nubilalis.  相似文献

Binding of three Bacillus thuringiensis insecticidal crystal proteins (ICPs) to the midgut epithelium of Ostrinia nubilalis larvae was characterized by performing binding experiments with both isolated brush border membrane vesicles and gut tissue sections. Our results demonstrate that two independent ICP receptors are present in the brush border of O. nubilalis gut epithelium. From competition binding experiments performed with I-labeled and native ICPs it was concluded that CryIA(b) and CryIA(c) are recognized by the same receptor. An 11-fold-higher binding affinity of CryIA(b) for this receptor correlated with a 10-fold-higher toxicity of this ICP compared with CryIA(c). The CryIB toxin did not compete for the binding site of CryIA(b) and CryIA(c). Immunological detection of ingested B. thuringiensis ICPs on gut sections of O. nubilalis larvae revealed binding only along the epithelial brush border membrane. CryID and CryIE, two ICPs that are not toxic to O. nubilalis, were not bound to the apical microvilli of gut epithelial cells. In vitro binding experiments performed with native and biotinylated ICPs on tissue sections confirmed the correlation between ICP binding and toxicity. Moreover, by performing heterologous competition experiments with biotinylated and native ICPs, it was confirmed that the CryIB receptor is different from the receptor for CryIA(b) and CryIA(c). Retention of activated crystal proteins by the peritrophic membrane was not correlated with toxicity. Furthermore, it was demonstrated that CryIA(b), CryIA(c), and CryIB toxins interact in vitro with the epithelial microvilli of Malpighian tubules. In addition, CryIA(c) toxin also adheres to the basement membrane of the midgut epithelium.  相似文献

We investigated vertical and horizontal transmission as means by which entomopathogenic microsporidia may be isolated in their hosts. Ostrinia nubilalis larvae were challenged with microsporidia isolated from other stalk-boring and row crop Lepidoptera and were susceptible to seven species. Two species were horizontally transmitted. A Nosema sp. from Eoreuma loftini was transmitted among O. nubilalis larvae but not among larvae of the E. loftini host. This species was also vertically transmitted to the offspring of infected O. nubilalis females. An rDNA sequence showed the E. loftini isolate to be Nosema pyrausta, a naturally occurring species in O. nubilalis. Our results suggest that both horizontal and vertical transmission provide physiological barriers to host switching in the microsporidia, thus restricting the natural host range.  相似文献

The efficacy of methanolic seed extracts of Annona squamosa (As) and Prosopis juliflora (Pj) individually and in combination with spore-crystal mixture of Bacillus thuringiensis kurstaki (Btk) was tested against third instar larvae of Helicoverpa armigera. The combination of botanicals with Btk significantly decreased LC50 values in less time as compared to individual treatments. As and Pj in combination with Btk significantly affected feeding and dietary utilization of H. armigera significantly than in individual treatments. Physiological effects found in larvae fed on Btk and botanicals included extended larval and pupal durations, decreased adult longevity and fecundity, difficulty in moulting between various instars, formation of larval–pupal intermediates and abnormal pupae. As was found to have a higher interaction with Btk than Pj. Further, Btk showed strong synergistic action with As and Pj, which are the promising candidates for development of alternative bioinsecticide in IPM programs.  相似文献

The American bollworm, H. armigera, evolved 31-fold resistance to selection pressure of B. thuringiensis endotoxin Cry1Ac within six generations. The Cry1Ac selected larvae of H. armigera showed cross-resistance to Cry1Aa and Cry1Ab both in terms of mortality and growth reduction. Studies on mechanisms of resistance to Cry1Ac showed that proteases of resistant insects degraded Cry1Ac faster than those of susceptible insects, which led to the relative unavailability of toxin of about 58 kDa for binding and perforation of midgut epithelial membrane of the target insect. Besides, resistant and susceptible populations of H. armigera differed in the binding of their receptors with Cry1Ac toxin. These results suggest the possibility of both mechanisms existing in imparting resistance. These findings mandate the necessity of B. thuringiensis resistance management for usage of B. thuringiensis either as a conventional insecticide or through transgenic crops.  相似文献

The anatomy of the neurosecretory cells in the brain-subesophageal ganglion complex of female European corn borer moth Ostrinia nubilalis (Lepidoptera: Pyralidae) was studied using histological and cobalt backfilling techniques. Histological staining revealed the presence of 2 median and one lateral neurosecretory cell groups in the brain. These brain neurosecretory cells are made up of mainly type A cells with a few type B cells in the median group. Three type C neurosecretory cell clusters occupy the apparent mandibular, maxillary, and labial neuromeres at the ventral median aspect of the subesophageal ganglion. Axonal pathways of the neurosecretory cell groups were delineated by retrograde cobalt filling from the corpora cardiaca. Fibers of the 3 brain neurosecretory cell groups merged to form a distinct axonal tract that exits the brain via the fused nervi corporis cardiaci-1 + 2. Cobalt backfilling from the corpora cardiaca filled 4 groups of cell bodies in the subesophageal ganglion. The presence in the subesophageal ganglion of extensive dendritic arborizations derived from the brain suggests interactions between neurosecretory cell groups in the 2 head ganglia.  相似文献

Intra-specific variation in susceptibility of Helicoverpa armigera (Hübner) and Helicoverpa punctigera (Wallengren) in Australia to the Cry1Ac and Cry2Ab delta-endotoxins from Bacillus thuringiensis (Berliner) (Bt) was determined to establish a baseline for monitoring changes that might occur with the use of Bt cotton. Strains of H. armigera and H. punctigera were established from populations collected primarily from commercial farms throughout the Australian cotton belts. Strains were evaluated for susceptibility using two bioassay methods (surface treatment and diet incorporation) by measuring the dose response for mortality (LC50) and growth inhibition (IC50). The variation in LC50 among H. armigera (n=17 strains) and H. punctigera (n=12 strains) in response to Cry1Ac was 4.6- and 3.2-fold, respectively. The variation in LC50 among H. armigera (n=19 strains) and H. punctigera (n=12 strains) to Cry2Ab was 6.6- and 3.5-fold, respectively. The range of Cry1Ac induced growth inhibition from the 3rd to 4th instar in H. armigera (n=15 strains) was 3.6-fold and in H. punctigera (n=13 strains) was 2.6-fold, while the range of Cry2Ab induced growth inhibition from neonate to 3rd instar in H. armigera (n=13 strains) was 4.3-fold and in H. punctigera (n=12 strains) was 6.1-fold. Variation in susceptibility was also evaluated for two age classes (neonates and 3rd instars) in laboratory strains of H. armigera and H. punctigera. Neonates of H. punctigera had the same or higher sensitivity to Bt than 3rd instars. Neonates of H. armigera were more sensitive to Cry2Ab than 3rd instars, while being less sensitive to Cry1Ac than 3rd instars. Differences in the two methods of bioassay used affected relative sensitivity of species to Bt toxins, highlighting the need to standardize bioassay protocols.  相似文献

Ultrastructural examination of diapause and nondiapause larval brains of the European corn borer disclosed anatomical differences that may be related to the insect's "blood-barrier". The perineurial type I cells are quite closely appressed in the diapause brain, but thrown into extensive folds with large intercellular spaces in the nondiapause brain. The perineurial type II cells of diapause and nondiapause larvae are basically similar in general ultrastructure, and most likely form the basis for the "blood-brain barrier." Horseradish peroxidase penetration studies indicated that the outer margin of the perineurial type II cells constitute the limits of infiltration into the brain. An enzymatic component of the "blood-brain barrier" is postulated in this insect. The localization of ATPase in the perineurial type II cells indicates that energy-requiring regulatory mechanisms may be localized here. Metabolic studies with isolated brains, coupled with recent evidence from mammalian systems, suggest that glial cells may be of importance in an enzymatic "blood-brain barrier."  相似文献

Monoamine oxidase (MAO) activity was demonstrated in intact brains of larvae from the European corn borer, Ostrinia nubilalis using 2-(2′-benzothiazolyl)-5-stryl-3-(4′-phthalhydrazidyl) tetrazolium chloride (BSPT). With tryptamine as the substrate, MAO specific activity was restricted to mitochondria within perineurial cells. The basic BSPT methodology was modified by the substitution of 2% dimethylsulfoxide for dimethylformamide in the incubation medium. This yielded increased permeability of the brain to the incubation medium, presumably by disrupting the integrity of the blood-brain barrier. A discussion of possible reasons for the previous inability to demonstrate insect neuronal MAO activity is presented.  相似文献

For a molecular genetic study on Hungarian populations of European corn borer L5 stage larvae were collected from 14 places of three different regions of the country (uni- and bivoltine ecotypes). Additionally, the study included larvae from Egypt, too (multivoltine ecotype). Molecular examinations of European corn borer larvae using the study of mitochondrial cytochrome b (cyt b) revealed that by single strand conformation polimorphism (SSCP) the populations found in Hungary represented the same haplotype. Even the Egyptian sample showed no genetic divergence. Some minor deviatons were found in the case of a sample from Székkutas, but that did not prove the genetic divergence of the bivoltine ecotype either, since the other samples of South-East Hungary did not display this kind of genetic variation. On the basis of our investigations it can be said that the univoltine and bivoltine generations, have uniform genetic complements.  相似文献

We examined whether Macrocentrus cingulum (Hymenoptera: Braconidae) of Asian origin could serve as a biological control agent of the maize pest Ostrinia nubilalis (Lepidoptera: Crambidae) in Europe. M. cingulum is already present in Europe, where it does not parasitize O. nubilalis but Ostrinia scapulalis, a related species feeding on wild dicotyledons. In contrast, M. cingulum have been imported from Europe and Asia into North America (where O. nubilalis had been accidentally introduced from Europe), and does parasitize O. nubilalis there. We conducted laboratory infestations to assess host acceptability (parasitoid’s propensity to oviposit) and suitability (parasitoid’s ability to develop) of European O. nubilalis for M. cingulum of European and Asian origin, and of Ostrinia furnacalis (their original host) for Asian M. cingulum. Asian M. cingulum parasitized European O. nubilalis as readily as O. furnacalis, and developed equally well in terms of: % female cocoons, time to first emergence from the cocoon, total number of adult offspring, % female offspring and adult longevity. Adult female parasitoids were significantly larger when emerging from O. nubilalis, mixed-sex and male cocoons were significantly more and less frequent, respectively. The acceptability of O. nubilalis was significantly lower for European than for Asian M. cingulum, and its suitability was zero. Asian M. cingulum appears a potential candidate for introduction as a biological control agent of a major maize pest, European O. nubilalis, provided environmental impact studies, economic analyses, and foreseeable interactions with other biological control agents such as the egg parasitoid Trichogramma brassicae (Hymenoptera: Trichogrammatidae) are satisfying.  相似文献

Helicoverpa armigera, an important polyphagous insect pest in agriculture, attacks more than 200 plant species of more than 30 families. Our previous study showed that the choice feeding percentages of H. armigera larvae to tobacco leaf discs treated with sweet, bitter, and hot taste substances were higher than the control leaf discs, while numb and salty substances could significantly inhibit their feeding. To quantitatively determine the synergistic effect of numb and salty substances, in this paper, the antifeeding activities of numb and salty substances and their mixtures blended in different doses or volume ratios were assayed on H. armigera larvae. The first bioassay was designed to elucidate the relative feeding preference of the larvae to the leaves from several common host species, each paired with tobacco leaf discs, and the result indicated that the most preferred host leaf by the larvae was tobacco leaf, followed by cotton and peanut leaves, suggesting that tobacco leaf was the most suitable matrix for the antifeeding bioassay, and the larval consumption of maize, pepper, or tomato leaves were significantly lower than that of tobacco leaf. The second bioassay was to test the choice feeding response of H. armigera larvae to tobacco leaf discs treated with Zanthoxylum bungeanum extracts obtained with different solvents, and the result showed that the antifeeding activity of the alcohol extracts was the strongest (93.38%), and the leaf consumption in the treatment and the control showed extremely significant difference (t = 4.23, t0.01 = 3.25, P = 0.0022), followed by the dichloromethane extracts (47.64%), while the other three solvents (water, acetone, and n-hexane) could not extract the active antifeeding components from Z. bungeanum. The larval consumption of tobacco leaf discs treated with the alcohol extracts of Z. bungeanum and NaCl solution were significantly less than their corresponding controls. The mean larval consumption of the treated leaf discs decreased with ever-increasing dosage, and the consumption of tobacco leaf discs coated with different doses of alcohol extracts of Z. bungeanum or NaCl solution showed extremely significant difference (Falcohol extract of Z. bungeanum = 3.88, F0.01 = 3.58, P = 0.0064; FNaCl solution = 54.29, F0.01 = 3.58, P = 0.0000), with maximum antifeeding effects at a dosage of 30 μL per 1.5 cm ID leaf disc. We further tested the larval consumption of tobacco leaf discs treated with alcohol extracts of Z. bungeanum in saturated NaCl solution mixed in different volume ratios, and the result showed that the choice antifeeding percentages of the treatments with 15 μL or more Z. bungeanum alcohol extracts were higher than 90%, among which the mixture with 25:15 volume ratio of Z. bungeanum alcohol extracts and saturated NaCl solution exhibited the strongest antifeeding activity, and the mean consumed leaf area of tobacco leaf discs coated with this blend was only 0.10 mm2. In the further test on feeding dose-response of the 25:15 mixture, the mean leaf consumption decreased linearly with ever-increasing dosage, with a regression equation y = ?3.9356x + 120.78(R2 = 0.9998), and the 30 μL dose could completely inhibit H. armigera feeding.  相似文献

The susceptibility of Trichoplusia ni larvae to several Bacillus thuringiensis insecticidal crystal proteins (ICPs) was tested. Neonatal larvae proved to be susceptible to solubilized trypsin-treated CryIA(a), CryIA(b), and CryIA(c) (50% lethal concentrations [LC(50)s], 570, 480, and 320 ng/cm, respectively) but showed little susceptibility to CryIB and CryID (LC(50)s, 5,640 and 2,530 ng/cm, respectively). The toxicity of ICPs was correlated to binding to the epithelial brush border of the midgut, as revealed by immunocytochemical staining with monoclonal antibodies. In vitro binding experiments with iodinated ICPs and brush border membrane vesicles indicated that CryIA(b) and CryIA(c) share the same high-affinity binding site, whereas CryIA(a) binds to a different one. The affinities of CryIA(b) and CryIA(c) for the binding site were similar (K(d) = 3.6 and 4.7 nM, respectively), and the mean binding-site concentration was 0.71 pmol/mg of vesicle protein. Selection of a population with increasing concentrations of CryIA(b) produced 31-fold resistance in seven generations. The realized heritability (h) was 0.19. The increase of homozygosity (for resistance factors) as selection proceeded was reflected in the increase in the slopes of the dose-mortality curves. Resistance was specific for CryIA(b) and did not extend to CryIA(a) or even to CryIA(c). This result was not predicted by the binding-site model, in which CryIA(b) and CryIA(c) bind to the same high-affinity binding site. This result may suggest a more complicated relationship between in vitro binding of ICPs to specific sites in the epithelial membrane of the midgut and the in vivo toxic effect.  相似文献

A detailed morphometric analysis of the antennae of male and female Trichoplusia ni (Hübner) is presented, based on light and scanning electron microscopic observations. These measurements of general antennal dimensions and the number, distribution, dimensions, and surface features of the antennal sensilla are applied in the consideration of olfactory transduction processes and in the derivation of a mathematical representation of the antennal surface. The surface area and the number of sensilla on the antennal flagellum are found to vary with the weight of the pupa and adult moth. The long Type 1 sensilla trichodea are hypothesized to be innervated by pheromone-sensitive olfactory cells on the basis of morphological comparisons with sensilla of other insects.  相似文献

In the past five decades, agricultural pests are controlled by synthetic pesticides which caused ill effects on non-target organisms and environment; also insect pests developed resistance and minor pests became major pests. By keeping this in mind, the present study was selected. Antifeedant and larvicidal activities and growth inhibitory effects of hexane, chloroform and ethyl acetate leaf extracts of Fleuggea leucopyrus (Koen.) Willd. against Helicoverpa armigera (Hbn.) were studied. Antifeedant and larvicidal activities were evaluated at 0.5, 1, 2.5 and 5% concentrations at laboratory conditions. Hexane and chloroform extracts at 5% concentration recorded significant antifeedant activity of 82.41 and 74.45%, respectively. Hexane extract recorded the least LC50 and LC90 values of 1.37 and 4.80%, respectively. Hexane extract recorded maximum larvicidal activity of 85.78% with LC50 and LC90 values of 1.69 and 4.94%, respectively. Ethyl acetate extract also had notable amount of larvicidal activity of 81.77% at 5% concentration. Hexane extract at 5 and 2.5% and ethyl acetate extract at 5% concentrations completely prevented the adult emergence of H. armigera. This plant could be further exploited to identify the active principle(s) responsible for the activities and to develop a novel pesticidal formulation. This is the first report of F. leucopyrus is studied for its bioefficacy against H. armigera.  相似文献

This study provides comprehensive documentation of silk production in the pest moth Helicoverpa armigera from gland secretion to extrusion of silk thread. The structure of the silk glands, accessory structures and extrusion apparatus are reported. The general schema of the paired silk glands follows that found for Lepidoptera. Morphology of the duct, silk press, muscle attachments and spigot are presented as a three-dimensional reconstruction and the cuticular crescent-shaped profile of the silk press is demonstrated in both open and closed forms with attendant muscle blocks, allowing advances in our knowledge of how the silk press functions to regulate the extrusion of silk. Growth of the spigot across instars is documented showing a distinctive developmental pattern for this extrusion device. Its shape and structure are related to use and load-bearing activity.  相似文献

Summary The identity of a cell line derived from hemocytes ofMalacosoma disstria was investigated serologically by using complement fixation, double diffusion, immunoelectrophoresis in agarose, and acrylamide gel electrophoresis. TheM. disstria cell line antiserum gave a specific cross-reaction with its homologous antigen and withM. disstria larval antigen, although cross-reactions with cell lines fromAedes aegypti, Bombyx mori, andChoristoneura fumiferana and with larval antigens ofB. mori andC. fumiferana were also produced. Antisera againstA. aegypti andB. mori cell lines showed very similar cross-reaction with both the cell line antigens. Also, these two antisera gave a strong reaction withB. mori larval antigen, but no reaction withA. aegypti larval antigen. These tests confirm the identity of ourM. disstria cell line. Also, they indicate: (a) that theA. aegypti andB. mori cell lines tested are similar, and (b) that they are closely related toB. mori, but not at all related toA. aegypti. A comparative study of different steps in the mode of action of the individual Bacillus thuringiensis kurstaki BNS3 Cry1Aa and Cry1Ac δ-endotoxins on E. kuehniella larvae was performed in order to investigate the origin of the difference in the response of this larvae to each of the latter. Proteolytic activation was shown to be one of the main steps impaired in E. kuehniella tolerance to Cry1Aa. The absence of two proteinase activities as well as an altered activity level observed in the case of Cry1Aa would be the consequence of proteinase-mediated tolerance of E. kuehniella to this toxin. In situ binding and histopathological effect analyses allowed concluding that the binding of the toxin to BBMV receptors is the key step in E. kuehniella tolerance to Cry1Aa toxin. The latter was slightly bound to apical membranes of epithelial cells that remained intact, whereas Cry1Ac was tightly bound to completely damaged cells basal membranes.  相似文献

There is a great demand for food with increasing population of the world. Many chemical fertilisers and pesticides are used to increase crop productivity. Over use of pesticides and fertilisers has affected the environment and non-target organisms severely; minor pests have become major destructive pests. Hence, eco-friendly approaches are needed. The present study reports the effect of hexane, chloroform and ethyl acetate extract of Couroupita guianensis on the eggs of Helicoverpa armigera. All the extracts showed ovicidal activity, and among them hexane extract showed 64.28% ovicidal activity with LC50 value of 2.62% and regression of (R 2) 83.5%. All the original data showed normality and homogeneity. The active hexane extract was fractionated; finally, eight fractions were obtained and studied at different concentrations. Among the fractions, fraction 8 showed 75% ovicidal activity with R 2 of 90%. It showed LC50 value of 322.94?ppm with χ 2 value at 0.5%. C. guianensis could be used in pest management programme.  相似文献
