Separation of the life cycle of flowering plants into two distinct growth phases, vegetative and reproductive, is marked by the floral transition. The initial floral inductive signals are perceived in the leaves and transmitted to the shoot apex, where the vegetative shoot apical meristem is restructured into a reproductive meristem. In this study, we report cloning and characterization of the maize (Zea mays) flowering time gene delayed flowering1 (dlf1). Loss of dlf1 function results in late flowering, indicating dlf1 is required for timely promotion of the floral transition. dlf1 encodes a protein with a basic leucine zipper domain belonging to an evolutionarily conserved family. Three-dimensional protein modeling of a missense mutation within the basic domain suggests DLF1 protein functions through DNA binding. The spatial and temporal expression pattern of dlf1 indicates a threshold level of dlf1 is required in the shoot apex for proper timing of the floral transition. Double mutant analysis of dlf1 and indeterminate1 (id1), another late flowering mutation, places dlf1 downstream of id1 function and suggests dlf1 mediates floral inductive signals transmitted from leaves to the shoot apex. This study establishes an emergent framework for the genetic control of floral induction in maize and highlights the conserved topology of the floral transition network in flowering plants.  相似文献

Higher plants use multiple perceptive measures to coordinate flowering time with environmental and endogenous cues. Physiological studies show that florigen is a mobile factor that transmits floral inductive signals from the leaf to the shoot apex. Arabidopsis FT protein is widely regarded as the archetype florigen found in diverse plant species, particularly in plants that use inductive photoperiods to flower. Recently, a large family of FT homologues in maize, the Zea CENTRORADIALIS (ZCN) genes, was described, suggesting that maize also contains FT-related proteins that act as a florigen. The product of one member of this large family, ZCN8, has several attributes that make it a good candidate as a maize florigen. Mechanisms underlying the floral transition in maize are less well understood than those of other species, partly because flowering in temperate maize is dependent largely on endogenous signals. The maize indeterminate1 (id1) gene is an important regulator of maize autonomous flowering that acts in leaves to mediate the transmission or production of florigenic signals. This study finds that id1 acts upstream of ZCN8 to control its expression, suggesting a possible new link to flowering in day-neutral maize. Moreover, in teosinte, a tropical progenitor of maize that requires short-day photoperiods to induce flowering, ZCN8 is highly up-regulated in leaves under inductive photoperiods. Finally, vascular-specific expression of ZCN8 in Arabidopsis complements the ft-1 mutation, demonstrating that leaf-specific expression of ZCN8 can induce flowering. These results suggest that ZCN8 may encode a florigen that integrates both endogenous and environmental signals in maize.  相似文献

Previous studies indicated that the lethal leaf spot 1 lesion mimic locus of maize ( ZmLls1 ) encodes a novel cell protective function in plants. Here we show that the accelerated cell death 1 ( acd1 ) locus of Arabidopsis thaliana corresponds to gene At3g44880 on chromosome 3. Proof that the Acd1 gene is an orthologue of ZmLls1 is provided by in vivo complementation of the acd1 mutant by the ZmLls1 gene. The Atlls1 lesion mimic phenotype was delayed in a chlorophyll a oxygenase (CAO) mutant chlorina1 background which is deficient in chlorophyll b synthesis. The interpretation that the cell protective function of LLS1 is linked with the removal of a phototoxic chlorophyll intermediate is supported by the recent report that the maize Lls1 gene encodes pheophorbide a oxygenase (PaO). Western blot analysis demonstrates that the LLS1 protein is present constitutively in all photosynthetic plant tissues. A transient increase in Lls1 gene expression by about 50-fold upon physical wounding of maize leaves indicates that the function of Lls1 is regulated in response to stress. We show that the LLS1 protein is also present at low levels in non-photosynthetic tissues including etiolated leaves suggesting that the ability to degrade chlorophyll exists in a standby mode in plant cells.  相似文献

We have identified a limited number of proteins secreted into the nectar of tobacco plants. Nectarin I is the most highly expressed nectar protein and has a monomer molecular mass of 29 kDa. The other major nectar proteins are expressed at lower levels and have monomer molecular masses of 41, 54, and 65 kDa respectively. Nectarin I was purified and antiserum was raised against the protein. Under nondenaturing conditions, Nectarin I has an apparent molecular mass of >120 kDa. The expression of Nectarin I was restricted to nectary tissues and to a much lower level in the ovary. No Nectarin I was found in petals, stems, leaves, or roots or other floral tissues. The expression of Nectarin I was also developmentally regulated. It is expressed in nectary tissues only while nectar is being actively secreted. Subsequently, the N-terminus of purified Nectarin I was sequenced. Sequence identity showed Nectarin I is related to wheat germin. Although hydrogen peroxide is readily detectable in tobacco floral nectar, we were unable to demonstrate any oxalate oxidase activity for Nectarin I. A partial cDNA encoding the mature Nectarin I N-terminus was isolated and used to probe a Nicotiana plumbaginifolia genomic library. The Nectarin I gene was isolated and the translated sequence was consistent with both N-terminal and internal cyanogen bromide-derived amino acid sequence. The gene contains a single 386 nt intron and encodes a mature protein of 197 amino acids.  相似文献

During leaf development, the formation of dorsal-ventral and proximal-distal axes is central to leaf morphogenesis. To investigate the genetic basis of dorsoventrality and proximodistality in the leaf, we screened for mutants of Arabidopsis thaliana (L.) Heynh. with defects in leaf morphogenesis. We describe here the phenotypic analysis of three mutant alleles that we have isolated. These mutants show varying degrees of abnormality including dwarfism, broad leaf lamina, and aberrant floral organs and fruits. Genetic analysis revealed that these mutations are alleles of the previously isolated mutant asymmetric leaves1 ( as1). In addition to the leaf phenotypes described previously, these alleles display other phenotypes that were not observed. These include: (i) some rosette leaves with petiole growth underneath the leaf lamina; (ii) leaf vein branching in the petiole; and (iii) a leaf lamina with an epidermis similar to that on the petiole. The mutant phenotypes suggest that the ASYMMETRIC LEAVES1 ( AS1) gene is involved in the control of cell differentiation in leaves. As the first step in determining a molecular function for AS1, we have identified the AS1 gene using map-based cloning. The AS1 gene encodes a MYB-domain protein that is homologous to the Antirrhinum PHANTASTICA ( PHAN) and maize ROUGH SHEATH2 ( RS2) genes. AS1 is expressed nearly ubiquitously, consistent with the pleiotropic mutant phenotypes. High levels of AS1 expression were found in tissues with highly proliferative cells, which further suggests a role in cell division and early cell differentiation.  相似文献

Heterotrimeric GTP binding proteins (G proteins) are important signal transducers in lower eukaryotes and in animal cells. In plants, the occurrence of GTP binding proteins has been reported, but their biological function remains unclear. Two genes coding for G protein alpha subunits have been cloned: GPA1 in Arabidopsis and TGA1 in tomato. To gain some insights into the function of GPA1, we describe an extensive immunolocalization of GPalpha1, the gene product of GPA1, during Arabidopsis development. Our results show that the GPalpha1 is present through all stages of development and in all organs examined, with the exception of mature seeds. It is expressed in roots, floral stem, rosette leaves, cauline leaves, flowers, and seed pods. Interestingly, the level of GPalpha1 protein is higher in immature organs than in mature organs. GPalpha1 is present at a high level in the root meristem and elongation zone, in the shoot and floral meristems, and in the leaf primordium and floral organ (sepal, petal, stamen, and gynoecium) primordia. During flower development, dividing microspores, but not mature pollen, show high levels of GPalpha1. During pollination, GPalpha1 is present in the growing pollen tubes. The protein is also present in nectaries and developing ovules and, after fertilization, in developing embryos. In mature tissue, GPalpha1 is preferentially found in the vascular system but is also present in other cell types. The complexity of the GPalpha1 localization pattern suggests that GPalpha1 might be involved in different signaling pathways depending on the developmental stage.  相似文献

采集了处于营养生长向生殖行长转化期(6~8月)的红玉苹果(Malus domestica Borkh.)茎顶,构建了其cDNA文库,并从中分离得到了一个具有锌指结构的EST序列,又通过5`RACE的方法,从cDNA文库中找到了其上游779bp的cDNA片段.最后用PCR的方法获得了苹果锌指蛋白的全长cDNA,并命为MdZF1.该cDNA序列已在GenBank登录,登录号为AB116545.MdZF1的锌指结构域与玉米的开花转换基因ID1有高度同源性.通过对苹果不同组织、器官的Northem和RT-PCR分析表明MdZF1在根、茎、叶、顶芽以及花器官(萼片、花瓣、雄蕊、雌蕊)中都有表达.Southern分析表明MdZF1的基因组中是以单拷贝存在的.  相似文献

In higher plants, leaves initiate in constant spatial and temporal patterns. Although the pattern of leaf initiation is a key element of plant shoot architecture, little is known about how the time interval between initiation events, termed plastochron, is regulated. Here, we present a detailed analysis of plastochron2 (pla2), a rice (Oryza sativa) mutant that exhibits shortened plastochron and precocious maturation of leaves during the vegetative phase and ectopic shoot formation during the reproductive phase. The corresponding PLA2 gene is revealed to be an orthologue of terminal ear1, a maize (Zea mays) gene that encodes a MEI2-like RNA binding protein. PLA2 is expressed predominantly in young leaf primordia. We show that PLA2 normally acts to retard the rate of leaf maturation but does so independently of PLA1, which encodes a member of the P450 family. Based on these analyses, we propose a model in which plastochron is determined by signals from immature leaves that act non-cell-autonomously in the shoot apical meristem to inhibit the initiation of new leaves.  相似文献

Nitrate reductase (NR; EC 1.6.6.1) activity increased at the beginning of the photoperiod in mature green maize (Zea mays L.) leaves as a result of increased enzyme protein level and protein dephosphorylation. In vitro experiments suggested that phosphorylation of maize leaf NR affected sensitivity to Mg2+ inhibition, as shown previously in spinach. When excised leaves were fed 32P-labeled inorganic phosphate, NR was phosphorylated on seryl residues in both the light and dark. Tryptic peptide mapping of NR labeled in vivo indicated three major 32P-phosphopeptide fragments, and labeling of all three was reduced when leaves were illuminated. Maize leaf NR mRNA levels that were low at the end of the dark period peaked within 2 h in the light and decreased thereafter, and NR activity generally remained high. It appears that light signals, rather than an endogenous rhythm, account primarily for diurnal variations in NR mRNA levels. Overall, regulation of NR activity in mature maize leaves in response to light signals appears to involve control of gene expression, enzyme protein synthesis, and reversible protein phosphorylation.  相似文献
