A polymorphism of the total protein (MY) and nonspecific esterase (EST) patterns was revealed in the musculocutaneous sac of earthworm Lumbricus rubellus by polyacrylamide gel electrophoresis. The revealed polymorphism is attributed to genetically determined codominant systems, each of which is controlled by the corresponding locus, My and Est, represented by two and four alleles, respectively. The allelic frequency distribution of both loci demonstrated genetic heterogeneity of four L. rubellus samples from Moscow and Moscow Region (311 individuals in total). Allelic frequencies of the revealed loci are proposed as genetic markers for monitoring L. rubellus generations in vermiculture.  相似文献

The island of Borneo is the diversity center of the Dipterocarpaceae, the most important family of tropical rainforest trees in Southeast Asia. However, changes in land use and climate have affected dipterocarp distributions on the island, raising concerns about the vulnerability (inter alia) of the endemic riparian species Shorea macrophylla. Thus, to aid conservation efforts, we have investigated the genetic diversity, structure, and demographic history of S. macrophylla. The species’ genetic diversity and structure in Kalimantan (part of Indonesia, covering 75% of the island) were explored by examining genotypes of 377 individuals representing 13 populations in three regions (Northeast, Central, and West Kalimantan) using 14 newly developed microsatellite loci. Higher genetic diversity was found, at all loci, in samples from Northeast Kalimantan than in samples from the other regions. Moderate genetic differentiation between populations was detected (FST 0.093). Bayesian clustering, principal coordinate, and neighbor joining tree analyses of the population structure consistently identified two genetically distinct groups, one in the Northeast and the other in the Central and West regions. The higher diversity of the diverged populations in Northeast Kalimantan indicates that the region may have hosted rainforest refugia during the ice age. Accordingly, analysis using DIY ABC software indicated that the Northeast and Central-West Kalimantan groups diverged 194,000 years ago. We conclude that global climate change during the Pleistocene strongly influenced the genetic diversity and structure of S. macrophylla populations in Kalimantan.  相似文献

The population genetic structure of white-spotted char Salvelinus leucomaenis (family Salmonidae) was determined based on variations in ten microsatellite DNA loci in samples from different parts of the species range in the Russian Far East. In a number of samples, variations in 21 allozyme loci were examined, of which five loci were found to be polymorphic. The overall diversity level at the examined markers was comparable to that observed in a closely related char species, Dolly Varden. The estimates of interpopulation genetic differentiation were highly statistically significant in most of the pairwise comparisons among the samples. The overall evaluation of the spatial genetic differentiation in white-spotted char constituted F ST = 0.203 and R ST = 0.202. Cluster analysis and multidimensional scaling based on microsatellite allele frequencies indicated the possible subdivision of the examined samples into two main groups, i.e., northern (represented by the regions of the north of Khabarovsk krai, Kamchatka, Yama Bay) and southern (with regions including Sakhalin, Primorye, and the Kuril Islands). The allozome data demonstrated a similar pattern of differentiation. The level of intra- and interpopulation genetic diversity in the southern group was higher than in the northern group. The isolation-by-distance test did not identify a significant correlation between genetic and geographic distances among the samples. The data obtained enabled the suggestion that the genetic structure of the populations of white-spotted char was shaped by the influence of historical geological climatic rearrangements of its range and the genetic drift because of relatively low population number and limited in the extent migration activity of its anadromous form.  相似文献

An analysis of 130 tissue specimens of the European (Capreolus capreolus) and the Siberian (C. pygargus) roe deer from nine regions of Russia and Ukraine using microsatellite loci analysis was conducted, aimed at the identification of hybrid animals. An optimized complex of 21 microsatellite loci was selected for the determination of species specificity and the search for interspecies hybrids. Hybrid animals were revealed in the Moscow and Volgograd regions. Their ratio in the total sample obtained in the European part of Russia was 11.9%.  相似文献

The genetic diversity and relationships among 47 pear cultivars and genotypes (Pyrus spp.), including 4 Japanese pears (Pyrus pyrifolia), 40 European pears (Pyrus communis), 1 Chinese pear (Pyrus bretschneideri) as well as 2 wild relatives (Pyrus salicifolia and Pyrus mazandaranica) were studied using 28 microsatellite primer pairs. A total of 174 alleles were produced at the 28 SSR loci with their sizes ranging from 81 to 290?bp. The number of observed alleles for each locus ranged from 3 (TsuENH014 and TsuENH046) to 12 (NB103a), with an average of 6.21 alleles per locus. In some SSR loci, more than two alleles were amplified in some cultivars and genotypes, suggesting that duplication has occurred in those accessions. This information suggests that at least two genomic regions exist for these loci in the pear genome. The observed heterozygosity (H o) values of amplified loci ranged from 0.17 (TsuENH006) to 0.97 (NB103a). Shannon's information index (I) value was observed to be highest (2.14) in the NB103a locus, while the TsuENH006 locus had the lowest value with an average of 1.37 among SSR loci. The Dice genetic similarity coefficient ranged from 0.29 (??Nijisseiki?? and P. mazandaranica) to 0.91 (??Chojuro?? and ??Nijisseiki??) among samples. The genetic diversity of four population of Norway spruce (Picea abies (L.) Karst.) in Moscow parks have been studied. For the first time, a comparison of the gene pool of planted stands in urban areas with the gene pool of natural populations and planted stands of the Moscow region investigated earlier revealed a significant reduction in the genetic diversity of planted stands of Moscow parks in comparison with conditionally native populations from the Moscow region. In three of the four urban planted stands, a decrease was revealed in the proportion of polymorphic loci (down to 0.41 and 0.50) compared to conditionally native forests (0.64). All plant stands do not differ from conditionally native in terms of average heterozygosity and average number of alleles per locus. However, the test for allelic frequency heterogeneity demonstrated a significant difference of all planted stands (from Moscow and the Moscow region) from conditionally native populations both by 3–11 loci and by all (17) polymorphic loci. The quality of the gene pool of three of the four Moscow planted stands was evaluated as unsatisfactory and one of them as critical. A reduction in viability of urban planted stands with a reduced gene pool diversity was revealed: two of the three plantations died. In our opinion, this is a consequence of the sharp deviations of the observed diversity of the gene pool of the studied plantations from the natural norm, i.e., an optimal state of the gene pool historical in this natural area. Thus, economic necessity of genetic control over the state of saplings in reforestation is obvious.  相似文献

A system of urban ecological genetic monitoring of vertebrates has been developed for the first time in Russia and in the world. As a model, two species of brown frogs, Rana arvalis Nilss. and R. temporaria L. (16 populations, seven isozyme loci) were used. An evaluation of the gene pool state in urban frog populations has shown that the diversity in the Moscow frog populations was lower than that in natural populations (up to 80 and 50% in respectively R. arvalis and R. temporaria). Mean heterozygosities per locus were higher in large natural populations than in small urban isolates: in R. arvalis, these values were 0.16 and 0.06; in R. temporaria, 0.34 and 0.18, respectively. The number of polymorphic loci was also higher in natural populations than in the urban ones: 4 versus 2 in R. arvalis and 5 versus 4 in R. temporaria. Using superoxiddismutase as an example, fixation of different alleles of the same locus in different small isolates was shown. The gene pool condition of all but one urban populations of brown frog was evaluated as unsatisfactory, and that of the R. arvalis populations, as critical. These changes of the gene pool are explained mainly by gene drift accompanied by inbreeding, which was caused by human-induced fragmentation of the range and a decrease in population size of the species. The results of this study was employed in the development of the Moscow governmental program on restoration of the gene pools of vanishing animal species on specially protected natural urban territories. The series of works on long-term monitoring and assessment of the state of natural populations of model species in anthropogenic landscapes of Moscow and Moscow region has laid a foundation for a new branch of science, gene urbanology.  相似文献

Common wild rice (Oryza rufipogon Griff.), the progenitor of Asian cultivated rice (O. sativa L.), is endangered due to habitat loss. The objectives of this research were to evaluate the genetic diversity of wild rice species in isolated populations and to develop a core collection of representative genotypes for ex situ conservation. We collected 885 wild rice accessions from eight geographically distinct regions and transplanted these accessions in a protected conservation garden over a period of almost two decades. We evaluated these accessions for 13 morphological or phenological traits and genotyped them for 36 DNA markers evenly distributed on the 12 chromosomes. The coefficient of variation of quantitative traits was 0.56 and ranged from 0.37 to 1.06. SSR markers detected 206 different alleles with an average of 6 alleles per locus. The mean polymorphism information content (PIC) was 0.64 in all populations, indicating that the marker loci have a high level of polymorphism and genetic diversity in all populations. Phylogenetic analyses based on morphological and molecular data revealed remarkable differences in the genetic diversity of common wild rice populations. The results showed that the Zengcheng, Gaozhou, and Suixi populations possess higher levels of genetic diversity, whereas the Huilai and Boluo populations have lower levels of genetic diversity than do the other populations. Based on their genetic distance, 130 accessions were selected as a core collection that retained over 90% of the alleles at the 36 marker loci. This genetically diverse core collection will be a useful resource for genomic studies of rice and for initiatives aimed at developing rice with improved agronomic traits.  相似文献

The genetic diversity, population structure, and linkage disequilibrium (LD) of peaches are greatly important in genome-wide association mapping. In the current study, 104 peach landrace accessions from six Chinese geographical regions were evaluated for fruit and phenological period. The accessions were genotyped with 53 genome-wide simple sequence repeat (SSR) markers. All SSR markers were highly polymorphic across the accessions, and a total of 340 alleles were detected, including 59 private alleles. Of the six regions studied, the northern part of China as well as the middle and lower reaches of the Changjiang River were found to be the most highly diverse genetically. Based on population structure analysis, the peaches were divided into five groups, which well agreed with the geographical distribution. Of the SSR pairs in these accessions, 18.07% (P?<?0.05) were in LD. The mean r 2 value for all intrachromosomal loci pairs was 0.0149, and LD decayed at 6.01?cM. The general linear model was used to calculate the genome-wide marker-trait associations of 10 complex traits. The traits include flesh color around the stone, red pigment in the flesh, flesh texture, flesh adhesion, flesh firmness, fruit weight, chilling requirement, flowering time, ripening time, and fruit development period. These traits were estimated by analyzing the 104 landraces. Many of the associated markers were located in regions where quantitative trait loci (QTLs) were previously identified. Peach association mapping is an effective approach for identifying QTLs and may be an alternative to QTL mapping based on crosses between different lines.  相似文献

The Siberian stone pine (Pinus sibirica Du Tour) is one of the main forest-forming coniferous species of the boreal ecosystems of Western Siberia. We used the isozyme method to analyze 11 ecotypes representing the latitudinal and longitudinal profiles within the species range, including samples from the geographic boundaries of the distribution. The genetic structure of the ecotypes is described on the basis of the variability for 26 isozyme loci encoding for 16 enzyme systems. The greatest genetic diversity was observed in the taiga ecotypes in the central part of the studied area, while the ecotypes along the species range boundaries were shown to be genetically depauperized. Approximately 8.1 % of the observed genetic diversity is attributed to differences between the studied ecotypes. We detected high levels of genetic diversity for the Fest-2, Pgm-1, Sod-4, and a few other loci, as well as a correlation between allele frequencies and geographical locations of the populations. The results of multivariate analysis of allelic frequencies as well as cluster analysis allowed us to discriminate three major groups of ecotypes: north-eastern, central and south-western. In view of our results, we compare two hypotheses: one which attributes the spatial distribution of genetic variations to the selectivity for some of the polymorphic allozyme loci, and the other based in the history of the formation of the range of the Siberian stone pine.  相似文献

Using bush snail Bradybaena fruticum Mull (20 populations) as a model, we were the first in Russia and in the world to develop a system of urban ecological genetic monitoring of the gene pool of an invertebrate species. The results of isozyme polymorphism studies in bush snail populations showed a dramatic (up to 70%) reduction in genetic diversity in small isolates from the urbanized environment as compared to large natural populations. In urban populations, genetic diversity parameters were demonstrated to be lower than in natural ones: the mean heterozygosity per locus was reduced up to 0.08 (0.15–0.20 in natural populations); the mean allele number, to 1.9 (2.7 in natural populations); and the number of polymorphic loci, to four, i.e., 2.2-fold (nine in natural populations). In Moscow region, the number of polymorphic loci also decreased to five in the population subjected to anthropogenic pressure. The changes in the population gene pool (as shown by the number of polymorphic loci) were different in Moscow and Moscow region. The percentage of populations with the number of polymorphic loci as low as four to six was 76.9 in Moscow and 23.1% in Moscow region. The gene pool quality of 80% of the urban snail populations was estimated as unsatisfactory, and in half of them, as critical. The main reason for these changes seems to be genetic drift accompanied by inbreeding, caused by fragmentation of the range and reduction in the abundance of populations of the species, due to the anthropogenic pressure. The results of the study were employed in the program of the Moscow government for restoring the gene pools of endangered animals species on the preserved territories of the city.  相似文献

Random amplified polymorphic DNA (RAPD) analysis based on polymerase chain reaction (PCR) with ten-nucleotide primers of arbitrary sequences (RAPD-PCR) was used to study the genetic characteristics of five samples of the striped field mouse (Apodemus agrarius Pallas, 1771) from the Russian Far East (Primorye, Khabarovsk krai, and Magadan oblast). Highly significant differentiation of the samples was demonstrated, the genetic diversity of each sample was estimated, and non-neutral loci were found. The genetic diversity was the highest in a population from the outskirts of Magadan and the lowest in populations from an island on the Amur River near Khabarovsk and from the village of Talon (Magadan oblast). These two last populations were found to be genetically close to each other. These results indicate that striped field mice have been repeatedly introduced to Magadan oblast. The Talon population is likely to have originated from the left bank of the Amur; the outskirts of Magadan are inhabited with a mixed population formed by mice from Primorye and other regions.  相似文献

Genetic variability of yellow potato cyst nematode G. rostochiensis from three Russian populations (Karelia, Vladimir oblast, and Moscow oblast) was investigated using two types of nuclear markers. Using RAPD markers identified with the help of six random primers (P-29, OPA-10, OPT-14, OPA-11, OPB-11, and OPH-20), it was possible to distinguish Karelian population from the group consisting of the populations from two adjacent regions (Moscow oblast and Vladimir oblast). Based on the combined matrix, containing 294 RAPD fragments, dendrogram of genetic differences was constructed, and the indices of genetic divergence and partition (P, H, and G st), as well as the gene flow indices N m between the nematode samples examined, were calculated. The dendrogram structure, genetic diversity indices, and variations of genetic distances between single individuals in each population from Karelia and Central Russia pointed to genetic isolation and higher genetic diversity of the nematodes from Karelia.Based on polymorphism of rDNA first intergenic spacer ITS1, attribution of all populations examined to the species G. rostochiensis was proved. Small variations of the ITS1 sequence in different geographic populations of nematodes from different regions of the species world range did not allow isolation of separate groups within the species. Possible factors (including interregional transportations of seed potato) affecting nematode population structure in Russia are discussed.  相似文献

This study presents the first examination of the genetic structure of Daphnia longispina complex populations in Eastern China. Only one species, D. galeata, was present across the eight investigated lakes; as identified by taxon assignment using allelic variation at 15 microsatellite loci. Three genetically differentiated D. galeata subgroups emerged independent of the type of statistical analysis applied. Thus, Bayesian clustering, discriminant analysis based on results from factorial correspondence analysis, and UPGMA clustering consistently showed that populations from two neighbouring lakes were genetically separated from a mixture of genotypes found in other lakes, which formed another two subgroups. Clonal diversity was high in all D. galeata populations, and most samples showed no deviation from Hardy-Weinberg equilibrium, indicating that clonal selection had little effect on the genetic diversity. Overall, populations did not cluster by geographical origin. Further studies will show if the observed pattern can be explained by natural colonization processes or by recent anthropogenic impact on predominantly artificial lakes.  相似文献

Allozyme variation was examined in 292 samples of the snorkel snail, Rhiostoma housei, collected from 12 localities within four regions of Thailand. Ten allozyme loci were screened across all the samples using horizontal starch gel electrophoresis, and eight of these loci, from eight enzyme systems, were found to be polymorphic. Within these eight loci the expected heterozygosity (Hexp) was moderate to high ranging from 0.015 to 0.148 with an overall mean of 0.081 ± 0.041, whilst a high genetic heterogeneity among samples was found (Fst = 0.798). R. housei exhibited a weak pattern of isolation by distance over the entire tested species range in Thailand. The high Fst and moderate to high Hexp suggest gene flow amongst populations is partially restricted resulting in fragmentation into local gene pools. Based on allele frequencies and an allozyme dendrogram, six groups were identified, and these entities may represent separate biological species. The large values of genetic distances and fixed allelic differences obtained for each combination of allopatric samples imply the presence of cryptic species under the name R. housei.  相似文献

Metal toxicity is a major abiotic stressor of plants. It has been established that changes in genetic variation occur very rapidly in plants in response to environmental stressors such as increased levels of metals. Quercus rubra (red oak) is a pioneer species in mining regions contaminated with metals in Northern Ontario (Canada). The objectives of the study were to (1) determine the level of genetic variation in Q. rubra populations from mining damaged ecosystems using RAPD marker system and (2) assess the level of gene expression of candidate genes for nickel resistance. Total gene diversity (HT) and the mean gene diversity among populations (HS) were 0.22 and 0.19, respectively. The percent of polymorphic loci within populations was high ranging from 61 % (Capreol) to 72 % (Daisy Lake) despite a high level of gene flow (2.4). The population differentiation (GST) value was low (0.17). No significant difference was found among the contaminated and reference sites for all the genetic parameters estimated. Hence, all the Q. rubra populations from the metal-contaminated and damaged ecosystems are genetically sustainable. Moreover, this study reveals that all populations were genetically closely related with genetic distance values varying from 0.17 to 0.35. A zinc finger protein of Arabidopsis thaliana (ZAT11) gene involved in nickel resistance was differentially expressed in samples analyzed. There was a 120 times higher of ZAT11 expression in samples from metal contaminated areas of Wahnapitae Dam compared to other metal contaminated and uncontaminated sites, but no association between soil metal levels and expression of ZAT11 was established.  相似文献

The interpopulation differentiation of the sockeye salmon Oncorhynchus nerka (Walbaum) from the Olyutorskiy and Karaginskiy districts and from the Kamchatka River basin was examined based on the allelic variation at eight microsatellite loci (Ots107, Oki1a, Oki1b, One104, One109, OtsG68, OtsG85, and Oki6). The genetic diversity of samples from the northern rivers was lower, compared to samples from the Kamchatka River basin. Significant heterogeneity was found in the allele-frequency distribution at microsatellite loci of sockeye salmon from the investigated localities. The degree of genetic similarity of populations corresponded to their geographic closeness. The differences between population groups greatly exceeded the level of interpopulation differentiation. The analyzed samples formed four relatively separate groups: Lake Azabachye, Kamchatka River basin, Karaginskiy area (including the Navyrinvayam River in the south of the Olyutorskiy district), and northern Olyutorskiy area. The identification likelihood estimates of eastern Kamchatkan sockeye salmon in mixed aggregations at the level of population groups were fairly high (67.2–81.8%), greatly exceeding the accuracy of identification of individual populations.  相似文献

Eucalyptus globulus Labill is a foundation tree species over its disjunct distribution in southeastern Australia. The quality of its pulp makes it the most important hardwood species in the world. The importance of E. globulus prompted the establishment of common gardens from seed collected across its geographic range. This enabled us to study the genetic structure of the species, its population boundaries, and gene flow using 444 trees from different open-pollinated families that were genotyped at 16 microsatellite loci. A Bayesian clustering method was used to resolve five genetically distinct groups across the geographical range. These groups were identified as regions, which varied in diameter from 38 to 294?km and contain 4 to 16 putative populations. For two of these regional groups, we used spatial autocorrelation analysis based on assignment of trees to their natural stands to examine gene flow within each region. Consistent significant local-scale spatial structure occurred in both regions. Pairs of individuals within a region showed significant genetic similarity that extended beyond 40?km, suggesting distant movement of pollen. This suggests that breeding populations in E. globulus are much bigger than traditionally accepted in eucalypts. Our results are important for the management of genetic diversity and breeding populations in E. globulus. Similar studies of a variety of eucalypts pollinated by insects and birds will determine whether the local-scale genetic structure of E. globulus is unusual.  相似文献

Levels of genetic diversity and population genetic structure of a collection of 230 accessions of seven tetraploid Triticum turgidum L. subspecies were investigated using six morphological, nine seed storage protein loci, 26 SSRs and 970 DArT markers. The genetic diversity of the morphological traits and seed storage proteins was always lower in the durum wheat compared to the wild and domesticated emmer. Using Bayesian clustering (K = 2), both of the sets of molecular markers distinguished the durum wheat cultivars from the other tetraploid subspecies, and two distinct subgroups were detected within the durum wheat subspecies, which is in agreement with their origin and year of release. The genetic diversity of morphological traits and seed storage proteins was always lower in the improved durum cultivars registered after 1990, than in the intermediate and older ones. This marked effect on diversity was not observed for molecular markers, where there was only a weak reduction. At K >2, the SSR markers showed a greater degree of resolution than for DArT, with their identification of a greater number of groups within each subspecies. Analysis of DArT marker differentiation between the wheat subspecies indicated outlier loci that are potentially linked to genes controlling some important agronomic traits. Among the 211 loci identified under selection, 109 markers were recently mapped, and some of these markers were clustered into specific regions on chromosome arms 2BL, 3BS and 4AL, where several genes/quantitative trait loci (QTLs) are involved in the domestication of tetraploid wheats, such as the tenacious glumes (Tg) and brittle rachis (Br) characteristics. On the basis of these results, it can be assumed that the population structure of the tetraploid wheat collection partially reflects the evolutionary history of Triticum turgidum L. subspecies and the genetic potential of landraces and wild accessions for the detection of unexplored alleles.  相似文献
