The curves, obtained in the in vitro experiments, that show the dependence of the frequency of dicentrics (per a cell with dicentrics) upon dose and percentage of cells with dicentrics are proposed to be used in estimating a dose and body volume affected by partial irradiation (an extreme case of nonuniform exposure) by the analysis of chromosome aberrations in peripheral blood lymphocyte cultures.  相似文献

Unstable chromosome aberrations were scored in peripheral blood lymphocytes (PBL) serially collected from 21 breast cancer patients before and after radiotherapy (RT), chemotherapy (CT) and combined treatments. Local radiotherapy as treatment for mammary cancer induced unstable chromosome aberrations in peripheral blood lymphocytes. Only a fraction of these lymphocytes were exposed to irradiation during treatment and the chromosomal damage observed in PBL was equivalent to that induced by irradiation in vitro with 2 Gy at high dose rate, i.e., about 4% of the total dose delivered locally. Chemotherapy alone did not induce such anomalies. Apart from the observed interindividual variations in either the level or the fate of dicentrics with time, different features of chromosome damage were found when chemotherapy was given before or after local cobaltotherapy: secondary chemotherapy did not alter the frequency and the overdispersed distribution of dicentrics observed after first-line radiotherapy; in contrast, when CT was given before radiotherapy, a lower dicentric frequency was scored, the distribution of dicentrics was not always found to be overdispersed and there was a time-dependent decrease in dicentrics after in vivo exposure.  相似文献

To estimate the level of radiation exposure of personnel handling diagnostic X-ray machines, the yield of chromosomal aberrations was analysed in peripheral blood lymphocyte cultures. These occupationally exposed individuals showed higher frequencies of dicentrics as well as acentrics than normal controls. Absorbed radiation doses calculated by extrapolating reference in vitro dose-response curve for dicentrics ranged between 0.13 and 0.17 Gy. This implies exposure beyond the permissible limit of 0.05 Gy/year for the whole body. However, no obvious trend of increased aberrations as a function of either duration of employment or age was noticed. The increase in the aberration yields in this personnel underscores the need of adopting measures to avoid or minimise such overexposure.  相似文献

Cytogenetic analysis was performed on blood lymphocyte cultures from 12 moderate smokers (15-20 cigarettes/day) and 12 non-smokers. An increase in dicentrics was observed in the smokers using block-stained material but this was not significant. Analysis of banded material, enabling both symmetrical and asymmetrical aberrations to be scored, revealed a significant increase in total aberrations thus emphasizing the need to identify all chromosome rearrangements when examining the effects of low-level chronic exposures to clastogens.  相似文献

Peripheral blood lymphocyte culture system was used to construct reference dose-response curves for 60Co gamma-rays and 110 kV X-ray-induced chromosome aberrations at 6 dose points ranging from 0.25 to 4.0 Gy. Qualitative and quantitative differences between these two types of radiation for the yield of induced aberrations and their distribution pattern were analysed. Experimental data of aberration yields were compared after fitting them to five different dose-response models. The yields of chromosome aberrations in particular dicentrics, gave a good fit to linear-quadratic besides linear and power models. In this model, single-track events predominated over double-track events for both the qualities of radiation used. The pattern of distribution was mainly Poisson for dicentrics but gave a conflicting result for acentrics which was in excess.  相似文献

Changes in the numbers of peripheral lymphocytes with chromosome aberrations were observed in cynomolgus monkeys after fractionated or acute 60Co irradiation at the same total doses. Immediately after irradiation, the yield of dicentrics decreased when the dose was fractionated but remained constant for 20 to 80 days, depending on the dose, when irradiation was acute. Lymphopenia was greater than expected when the dose was fractionated. The kinetics of the loss of chromosome aberrations and the changes in peripheral lymphocyte count occurring in monkeys after acute irradiation were compared to those observed in accidentally irradiated men.  相似文献

The production of chromosome aberrations in vivo has been studied in lymphocytes from a patient undergoing a wholebody treatment with gamma-radiation up to a cumulative dose of 1.4 Gy. These results were compared with the observations performed on whole blood samples irradiated in vitro with doses from 0.05 up to 2 Gy of gamma-rays. The frequency of chromosome aberrations, particularly the dicentrics, was found to be similar in vivo and in vitro. The yield of dicentrics could be best related to the dose by using a linear-quadratic model in both cases, the ratio of the coefficients a/b being of 0.56 and 0.69 Gy, respectively in vivo and in vitro. These observations confirm that in vitro dose response curves may be used to evaluate accurately an in vivo absorbed dose.  相似文献

Peripheral blood lymphocytes were irradiated in vitro with (213)Bi alpha particles at doses of 0, 10, 20, 50, 100, 200 and 500 mGy. Chromosome analysis was performed on 47-h cultures using single-color fluorescence in situ hybridization (FISH) to paint chromosomes 1, 3 and 5. The whole genome was analyzed for unstable aberrations to derive aberration frequencies and determine cell stability. The dose response for dicentrics was 33.60 +/- 0.47 x 10(-2) per Gy. A more detailed analysis revealed that the majority of aberrations scored as dicentrics were part of complex/multiple aberrations, with the proportion of cells containing complexes increasing with dose. Cells containing aberrations involving painted chromosomes (FISH aberrations) were further classified according to cell stability and complexity. The majority of cells with FISH aberrations were unstable. The proportion of aberrant FISH cells with complex/multiple aberrations ranged from 56% at 10 mGy to 89% at 500 mGy. A linear dose response for genomic frequencies of translocations in stable cells fitted the data from 0 to 200 mGy with a dose response of 7.90 +/- 0.98 x 10(-2) per Gy, thus indicating that they are likely to be observed in peripheral blood lymphocytes from individuals with past or chronic exposure to high-LET radiation. Comparisons with the dose response for low-LET radiation suggest an RBE of 13.6 for dicentrics in all cells and 3.2 for translocations in stable cells. Since stochastic effects of radiation are attributable to genetic changes in viable cells, translocations in stable cells may be a better measure when considering the comparative risks of different qualities of radiation.  相似文献

To evaluate the effect of the chromosome arm number on the yield of dicentric chromosomes, frequencies of gamma-ray-induced chromosome aberrations were examined with peripheral lymphocytes from three different primate species, Saimiri sciureus (arm number, 77), Macaca fascicularis (arm number, 83) and Nycticebus coucang (arm number, 99). Irradiated blood samples were cultured by the same standard technique as that commonly used for human lymphocytes. The yields of dicentrics and dicentrics plus rings at doses of 100, 200 and 300 rad of gamma-irradiation were not significantly different among the three species, in spite of the difference in the chromosome arm number. Furthermore, dose-response relationships for these species were consistent with that for man. Statistical analysis indicated that the expected dicentric yields calculated from the arm number model were significantly different from the observed yields at 200 and 300 rad doses (P less than 0.01). From these results it can be pointed out that there is no correlation between the yield of dicentrics and the effective chromosome arm number, and that the chromosomal radiosensitivity of these primates is essentially the same as that of man, at least in the lymphocyte system.  相似文献

A study was made of the frequency of chromosome aberrations in human lymphocyte culture after gamma-irradiation (60Co) with doses ranging from 0.05 to 1.0 Gy at dose--rates of 0,005, 0.05 and 0.5 Gy/min. The frequency of structural changes in chromosomes at low doses was higher than it was expected in the case of extrapolating the effect produced by high to low doses of radiation; within the dose range from 0.1 to 0.5 Gy a plateau was registered for aberrations of the exchange type (dicentrics and rings). The abnormal character of the dose dependence of the yield of chromosome aberrations persisted with all three dose - rates under study.  相似文献

Literature data of long-term cytogenetic follow-up of people exposed to radiation as a results of different radiation accidents are considered for the purpose of discussing of some problems of biological dosimetry. The results obtained for mammals are also presented. Of particular interest is a decrease in the level of dicentrics and symmetrical translocations in peripheral blood lymphocytes with the time after acute exposure depending on the dose of irradiation. The frequency of dicentrics decreases in accordance to the exponential law passing the fast and slow phases of elimination. In different radiation situations the values of the parameter which defines the half-life period of lymphocytes characterizing 50% reduction of cells with dicentrics markedly vary. However a general regularity is a decrease in the parameter value as the exposure dose increases. The level of stable translocations estimates by the EISH method remains relatively constant at doses below 1-2 Gy. At higher doses their level in peripheral blood lymphocytes declines with time due to which the retrospective dose appears to be underestimated. The reasons of such regularity, the role of various factors affecting the scoring of translocations, criteria of analysis of the given chromosome aberrations are discussed in the context of common agreements between leading European laboratories on the use of FISH for improving biological dosimetric estimates.  相似文献

In a patient with Morbus Hodgkin, structural aberrations of the chromosome type in peripheral lymphocytes were analyzed during radiation therapy (accumulated target dose 44.6 Gy: 22 fractions of 1.8 Gy each and 2 fractions of 2.5 Gy each at the end of the therapy). The blood was sampled about 5 min after a fraction and/or 24, 48, or 72 h thereafter. The frequency of dicentric chromosomes:acentric fragments:centric ring chromosomes is 37:14:1 throughout the therapy. Independent of the time of blood sampling after a fraction, the distributions of dicentrics and acentrics are overdisperse and represent negative binomial distributions. The yields from these aberrations, as determined during the course of radiotherapy, are best fitted to a linear-quadratic function with a negative quadratic term. The two dose-effect curves (blood sampling about 5 min and 24 to 72 h after a fraction) of dicentrics and acentrics do not differ significantly. Up to an accumulated target dose of about 20 Gy the percentages of cells with chromosome aberrations increase to about 48 to 65% and, at this level, remain constant until the end of therapy.  相似文献

A dose dependence of the number of cells with chromosome aberrations was studied in PHA-stimulated donor's peripheral blood lymphocytes irradiated in vitro with doses of 10-400 cGy. In studying the number of chromosome aberrations and percentage of cells with micronuclei in parallel cultures no correlation was found between these indices within the groups exposed to a similar radiation dose.  相似文献

Data on lymphocyte kinetics may be derived in patients undergoing treatment by extracorporeal irradiation of the blood by studying chromosome aberrations induced in their lymphocytes; the aberrations are observed when the lymphocytes are stimulated by phytohaemagglutinin to divide in culture. Two methods of analysis are described, one depends on the total number of dicentrics scored, the other employs the dilution principle, regarding each lymphocyte bearing one or more aberrations as specifically labelled. Two parameters may be calculated: the mean residence time of lymphocytes in the blood and the size of the extravascular pool of lymphocytes in the body. In a patient with Hodgkin's disease, the mean residence time was found to be 4.7–7.5 min and the extravascular pool 800–1070 g.  相似文献

Summary Human peripheral blood was treated with ultrasound either before or after irradiation, and chromosome aberrations in lymphocytes of peripheral blood cultures compared to those resulting from an equivalent dose of irradiation given alone.When peripheral blood is sonicated at a high intensity (3 W/cm2) for 10 min after irradiation, there is an increase in aberration frequency as compared to control samples receiving the equivalent radiation dose alone. However, should the blood be sonicated at the same frequency and for the same time period before radiation there is no significant increase in total chromosome aberrations over the irradiated controls. On the contrary a significant decreases occurs in certain classes of aberration.When sonification with a lower intensity (20 mW/cm2) was used in combination with irradiation the reverse effect was noted. Ultrasound administered for 10 min after radiation caused no significant increase in aberrations. On the contrary increasing the period of sonification to one hour resulted in a lowering of all types of aberration, significant in the case of dicentrics and total chromosome aberrations, when compared to irradiation alone. Reversing the order of treatment again resulted in the opposite effect to that achieved with comparable experiments at high intensities of sonification. Ultrasound before radiation did not produce lower breakage rates. Instead, when the period of sonification was increased to one hour, the number of aberrant cells, fragments, and total aberrations rose significantly over controls.It is suggested that sonification produces chemical changes affecting cellular repair systems, which when combined with ionising radiation, results in an increased or decreased repair effect depending on the dose, duration, and order of treatment.Dedicated to Prof. Dr. Felix Mainx on the occasion of his 80th birthday  相似文献

A study was made of the frequency of chromatid aberrations in lymphocyte culture of subjects affected by the Chernobyl accident as a function of dose estimated by the incidence of dicentrics. The average number of chromatid aberrations was nearly the same within the dose range from 0 to 5 Gy exhibiting a tendency towards growth with dose. A high individual variability of the chromatid aberration frequency was observed.  相似文献

Data are presented from the cytogenetic analysis of the peripheral lymphocytes from 94 male and 15 female donors. However, a much smaller number of regular donors were selected for regular use. A total of 28674 cells were analysed and these acted as negative and positive controls in the in vitro human lymphocyte tests accumulated in this laboratory over a 6-year period. The significant observations were:. (1) in untreated and solvent control treated cultures (a) no chromosomal interchanges were observed in 21570 cells; (b) the incidence of dicentrics was less than 1 per 10000 cells; (c) other types of aberrations were seen with an increased frequency, chromosomal gaps being the most variable. (2) The reference clastogens mitomycin C and cyclophosphamide produced high and remarkably consistent yields of all types of aberrations. It is concluded that when screening compounds in vitro for new genotoxins, aberrations such as chromosomal interchanges and dicentrics (due to their rarity in negative control cultures) should be accorded greater significance than the several other types of aberrations routinely seen. These conclusions emphasize the value of maintaining and updating adequate historical control records.  相似文献

22-MeV neutrons affecting the culture of human peripheral blood lymphocytes have been studied for their cytogenetic peculiarities. Linear character of the dependence of chromosome aberrations yield within the dose range of 0.2-4.0 Gy is noted. Relative biological efficiency of 22-MeV neutrons reaches the highest values at low doses (18.8-2.4). Pair fragments and dicentrics prevail in spectrum of aberrations.  相似文献
