The fatty acid profiles and contents of protozoa from the rumen fluid of cattle varied according to the type of diet consumed by their host. Changing from a high-quality hay diet to a low-quality hay diet (DA) decreased the proportions of saturated acids and increased the proportions of the unsaturated acids 18:1 cis-9, 18:2 and 18:3 in neutral lipids (NL) and phospholipids (PL). Adding sucrose, urea and sulphur (SUS) to DA increased the proportions of branched chain acids in PL while addition of safflower oil increased polyunsaturated acids in PL and 18:1 trans-11 in NL. Diet did not alter the PL fatty acid content of protozoa but oil supplement of DA resulted in a 10-fold increase in the content of free fatty acids. The defaunating effect of oil supplement was partly reversed by SUS suggesting that factors other than the fatty acid content of cells are important in determining the toxicity of oil to rumen protozoa. The results indicate that the amounts of individual long-chain fatty acids taken up by rumen ciliates are largely determined by their concentrations in rumen digesta.  相似文献

Algivorous ciliates represent an important link in freshwater food webs, as they transfer energy and biochemical matter from their algal prey to mesozooplankton predators. However, it is still unknown how dietary composition influences the biochemical composition of ciliates. We analyzed the sterol composition of the algivorous ciliates Balanion planctonicum and Urotricha farcta and compared it to the sterol composition of their diet—the cryptomonad Cryptomonas phaseolus. The sterol composition of the ciliates did not resemble that of their algal diet. Ergosterol [(22E)-ergosta-5,7,22-trien-3β-ol] was the sterol in highest concentration in C. phaseolus, whereas stigmasterol [(24S)-24-ethylcholesta-5,22E-dien-3β-ol] was dominant in both B. planctonicum and U. farcta. Moreover, the occasionally higher sterol concentrations in the ciliates than in their algal diet suggest sterol accumulation by the ciliates. We conclude that dietary sterol composition influences the composition of the two algivorous ciliates studied, but species-specific differences in metabolism probably determine the ultimate sterol composition of the ciliates.  相似文献

The influence of diet on the fatty acid composition of the hepatopancreas of Mytilus trossulus was studied. Three groups of mollusks were fed monocultures of the microalgae Phaeodactylum tricornutum, Chaetoceros muelleri (Bacillariophyceae), and Nannochloropsis sp. (Eustigmatophyceae) for 10 days. After 10 days, the proportion of polyunsaturated fatty acids, mainly eicosapentaenoic and docosahexaenoic, increased in the total lipids of the hepatopancreas in all mollusk groups. The content of saturated fatty acids in the mussel tissues decreased and was not dependent on the amount in the algal diet. Toward the end of the experiment, the fatty acid composition of the hepatopancreas of mussels was similar irrespective of the fatty acid composition of their food. The fatty acid analysis of M. trossulus feces suggests a selective assimilation by mussels of predominantly the n-3 polyunsaturated fatty acids. The role of fatty acid metabolism in M. trossulus is discussed.  相似文献

A comparative study has been made of the major lipid fractions and their fatty acid compositions in the yolk of eggs from ostriches under wild and farmed conditions. There were no differences in the lipid contents and proportions of the lipid fractions between the two groups of yolks. In both groups of yolks triacylglycerol and phospholipid were the major fractions. In the eggs from the wild ostriches, all the lipid fractions displayed substantial concentrations of C18 polyunsaturated fatty acids, triacylglycerol being particularly rich in linolenic acid and phospholipid rich in linoleic acid; phospholipid displayed substantial concentrations also of C20 and C22 polyunsaturates. There were considerable differences in the fatty acid compositions between the yolks. Those from the farmed birds displayed lower proportions of C18 polyunsaturates, particularly linolenic acid, throughout the lipid fractions. Compensatory increases were displayed most obviously in the concentrations of oleic acid and palmitoleic acid as well as other acids. The distinctive and extensive changes in fatty acid composition, particularly relating to the polyunsaturates, are discussed with respect to overall dietary requirements and specificities for embryo metabolism and possible effects on reproductive performance.  相似文献

The biochemical factors that determine the food quality of protistsfor rotifers are poorly understood. We evaluated populationgrowth rates and egg production of the rotifer Keratella quadratafed with four protist species growing on either an algal ora bacterial diet. The cryptomonad Cryptomonas phaseolus, consideredas a good quality prey, and assays without prey served as controls.Population growth rates and egg numbers of K. quadrata werecorrelated with single biochemical compounds (fatty acids, aminoacids and sterols) of the protists. Feeding on the alga C. phaseolusor the algivorous ciliates resulted in enhanced population growthrates and high egg production by K. quadrata, whereas feedingon bacterivores supported only moderate egg production but nopopulation growth. The rotifers’ egg production was correlatedwith the protist biochemical composition, including polyunsaturatedfatty acids (PUFAs), the sterol desmosterol, ergosterol, stigmastanoland the amino acid leucine. No significant relationships wereobserved between population growth rates of the rotifers andthe protists’ biochemistry, suggesting that populationgrowth and reproduction of K. quadrata may have different nutritionalrequirements. To our knowledge, this is one of the first studiesto analyze a large variety of biochemical compounds to determinethe food quality of protists for a zooplankton predator andthe first study to analyze the biochemical quality of protiststo a rotifer species.  相似文献

We hypothesized that the polyunsaturated fatty acids of the butterfly were probably derived from the diet and that there might be a great loss of body fat during metamorphosis. To substantiate these hypotheses, we analyzed the fatty acid composition and content of the diet, the larva, and the butterfly Morpho peleides. Both the diet and the tissues of the larva and butterfly had a high concentration of polyunsaturated fatty acids. In the diet, linolenic acid accounted for 19% and linoleic acid for 8% of total fatty acids. In the larva, almost 60% of the total fatty acids were polyunsaturated: linolenic acid predominated at 42% of total fatty acids, and linoleic acid was at 17%. In the butterfly, linolenic acid represented 36% and linoleic acid represented 11% of total fatty acids. The larva had a much higher total fatty acid content than the butterfly (20.2 vs. 6.9 mg). Our data indicate that the transformation from larva to butterfly during metamorphosis drastically decreased the total fatty acid content. There was bioenhancement of polyunsaturated fatty acids from the diet to the larva and butterfly. This polyunsaturation of membranes may have functional importance in providing membrane fluidity useful in flight.  相似文献

Under natural conditions, Chroothece richteriana synthesizes a fairly high proportion of fatty acids. However, nothing is known about how environmental changes affect their production, or about the production of protective compounds, when colonies develop under full sunshine with high levels of UV radiation. In this study, wild colonies of C. richteriana were subjected to increasing temperature, conductivity, ammonium concentrations and photosynthetically active radiation (PAR), and UV radiations to assess the potential changes in lipid composition and mycosporine‐like amino acids (MAAs) concentration. The PERMANOVA analysis detected no differences for the whole fatty acid profile among treatments, but the percentages of α‐linolenic acid and total polyunsaturated fatty acids increased at the lowest assayed temperature. The percentages of linoleic and α‐linolenic acids increased with lowering temperature. γ‐linolenic and arachidonic acids decreased with increasing conductivity, and a high arachidonic acid concentration was related with increased conductivity. The samples exposed to UVB radiation showed higher percentages of eicosapentaenoic acid and total monounsaturated fatty acids, at the expense of saturated fatty acids. MAAs accumulation increased but not significantly at the lowest conductivity, and also with the highest PAR and UVR exposure, while ammonium and temperature had no effect. The observed changes are probably related with adaptations of both membrane fluidity to low temperature, and metabolism to protect cells against UV radiation damage. The results suggest the potential to change lipid composition and MAAs concentration in response to environmental stressful conditions due to climate change, and highlight the interest of the species in future research about the biotechnological production of both compound types.  相似文献

To explore the ruminal ecosystem, this paper is an attempt to illustrate the biochemistry and the metabolism of rumen protozoa. Different culture techniques (gnotobiotic, axenic, continuous culture) of ciliate protozoa are reviewed. The ability of ciliates to invade plant tissues, their association and the degradation of plant cell constituents (cellulose, hemicellulose, pectin, glycolipids), and the use of starch and soluble sugars for polysaccharide storage are determined. The interrelationships between ciliate protozoa and bacteria are discussed. These include the enzymic degradation of the engulfed plant particles, the bacterial nitrogen (amino acids, nucleic acids), and vitamins and trace element supplies as nutrient sources for protozoa. The composition of the rumen ciliate population, controlled by phylogenetic factors, geographical distribution, feeding habits and physiological status of the host, by competition between microorganisms and by the diet (nature of the diet, number of meals), is related. The transmission and establishment of the microfauna and its contribution to the microbial nitrogen available for the host are discussed.  相似文献

Our hypothesis that the trans fatty acids in hydrogenated fat inhibited the synthesis of polyunsaturated fatty acids in the phospholipid of arterial cells was tested with five groups each with six pregnant porcine fed from d 35 of gestation and during lactation. The basal diet contained 2% corn oil (control). The other four diets included the control + 10% butter or 10% hydrogenated fat plus two levels of Mg. Plasma, milk and aortic phospholipid fatty acids, phospholipid composition and calcium content of the aorta from the piglets were determined. At 48 +/- 2 d of age, the aorta phospholipid of piglets from porcine fed hydrogenated fat contained a significantly higher concentration of linoleic acid, less arachidonic acid, and less long chain polyunsaturated fatty acid (PUFA) than did piglets from porcine fed either butterfat or the control diet. Mg had no effect. These changes in composition in piglets from porcine fed hydrogenated fat indicate that trans fat inhibits the metabolic conversion of linoleic acid to arachidonic acid and to other n-6 PUFA. The aortic calcium content data showed a significant interaction of calcium concentration with age. We concluded: 1) that dietary trans fat perturbed essential fatty acid (EFA) metabolism which led to changes in the phospholipid fatty acid composition in the aorta, the target tissue of atherogenesis, 2) this inhibition of EFA to PUFA by the isomeric fatty acids in hydrogenated fat is a risk factor in the development of coronary heart disease.  相似文献

The effect of the supplementation of sulfur amino acids to a low casein or soy protein isolate diet on tissue lipid metabolism was investigated. Supplementation of methionine to a 8% casein diet produced a fatty liver in rats, however, supplementation of methionine to a 8.8% soy protein diet (corresponding to a 8% casein diet as to sulfur amino acids content) did not produce a fatty liver. At the level of 8% or less of soy protein in the diet, the accumulation of liver lipids and serum triglyceride was observed. An amino acid mixture simulating the composition of soy protein isolate caused significant accumulation of liver lipids, but serum triglyceride was not changed. Serum cholesterol in rats fed the soy protein diet was lower than that in rats fed the casein diet, but on feeding the amino acid mixtures simulating these protein diets, there was no difference between the two groups. The small differences between soy protein isolate and casein as to lipid metabolism might be due to the small differences in the contents of sulfur amino acids or the specific nature of the soy protein or casein. The supplemental effect of methionine and cystine was also studied. About 60% of total sulfur amino acids could be substituted by cystine for maximum growth.  相似文献

The chemical composition, mineral elements, vitamins, free fatty acids and amino acid content of the edible red alga Gracilaria lemaneiformis Bory, grown in the sea near Nan'ao island, Guangdong Province, were analyzed in the present study. Gracilaria lemaneiformis Bory showed a total sugar content of 14.65%. The protein content was 21%, of which approximately 41% was determined to be essential amino acids (EAA).The major amino acid components were glutamic acid, leucine, arginine, and alanine. Of the EAA assayed,methionine and cysteine appeared to be the most limiting amino acids compared with the EAA pattern provided by Food and Agricultural Organization of the United Nations. The total lipids content was 0.87% and comprised a high composition of unsaturated fatty acids (61%), mainly as linoleic acid and oleic acid, and a little amount of polyunsaturated fatty acid; palmitic acid was the main component (39%) of saturated acids.Relatively high levels of vitamin C, iodine, phosphorus, and zinc were also present in G. lemaneiformis. The nutritional composition between G. Iemaneiformis and Nostoc flagelliforme, a rare alga that is widely eaten in Chinese society, was compared. The results suggest that N. flagelliforme can be substituted for by G.Iemaneiformis, not only because of their similar shape, but also because of their approximate nutritional composition. Gracilaria Iemaneiformis may possibly serve as a potential healthy food in human diets in the future.  相似文献

Metabolomics is a high-throughput tool that quantifies and identifies the complete set of biofluid metabolites. This “omics” science is playing an increasing role in understanding the mechanisms involved in disease progression. The aim of this study was to determine whether a nontargeted metabolomic approach could be applied to investigate metabolic differences between obese rats fed a high-fat sucrose (HFS) diet for 9 weeks and control diet-fed rats. Animals fed with the HFS diet became obese, hyperleptinemic, hyperglycemic, hyperinsulinemic, and resistant to insulin. Serum samples of overnight-fasted animals were analyzed by 1H NMR technique, and 49 metabolites were identified and quantified. The biochemical changes observed suggest that major metabolic processes like carbohydrate metabolism, β-oxidation, tricarboxylic acid cycle, Kennedy pathway, and folate-mediated one-carbon metabolism were altered in obese rats. The circulating levels of most amino acids were lower in obese animals. Serum levels of docosahexaenoic acid, linoleic acid, unsaturated n-6 fatty acids, and total polyunsaturated fatty acids also decreased in HFS-fed rats. The circulating levels of urea, six water-soluble metabolites (creatine, creatinine, choline, acetyl carnitine, formate, and allantoin), and two lipid compounds (phosphatidylcholines and sphingomyelin) were also significantly reduced by the HFS diet intake. This study offers further insight of the possible mechanisms implicated in the development of diet-induced obesity. It suggests that the HFS diet-induced hyperinsulinemia is responsible for the decrease in the circulating levels of urea, creatinine, and many amino acids, despite an increase in serum glucose levels.  相似文献

The effect of dietary hydrogenated fat (Indian vanaspati) high in trans fatty acids (6 en%) on lipid composition, fluidity and function of rat intestinal brush border membrane was studied at 2 and 8 en% of linoleic acid. Three groups of weanling rats were fed rice-pulse based diet containing 10% fat over a ten week period: Group I (groundnut oil), Group II (vanaspati), Group III (vanaspati + safflower oil). The functionality of the brush border membrane was assessed by the activity of membrane bound enzymes and transport of D-glucose and L-leucine. The levels of total cholesterol and phospholipids were similar in all groups. The data on fatty acid composition of membrane phospholipids showed that, at 2 en% of linoleic acid in the diet, trans fatty acids lowered arachidonic acid and increased linoleic acid contents indicating altered polyunsaturated fatty acid metabolism. Alkaline phosphatase activity was increased while the activities of sucrase, gamma-glutamyl transpeptidase and transport of D-glucose and L-leucine were not altered by dietary trans fatty acids. However at higher intake of linoleic acid in the diet, trans fatty acids have no effect on polyunsaturated fatty acid composition and alkaline phosphatase activity of intestinal brush border membrane. These data suggest that feeding dietary fat high in trans fatty acids is associated with alteration in intestinal brush border membrane polyunsaturated fatty acid composition and alkaline phosphatase activity only when the dietary linoleic acid is low.  相似文献

The addition of ciliated protozoa to aquatic microcosms and bench-scale sewage treatment plants increases decomposition rates. This is surprising, inasmuch as protozoa consume bacteria, which are the primary decomposers. One possible mechanism of the increase in decomposition rate is the direct consumption of dissolved organic matter by protozoa that are feeding primarily on bacteria. This possibility was explored experimentally in two-stage continuous cultures, with glucose limitingEscherichia coli in the first stage andE. coli limitingTetrahymena pyriformis in the second. Glycine and histidine were the test nutrients. The results of adding them to the second stages suggested that direct uptake by ciliates does not affect the dynamics of dissolved amino acids in pelagic environments or activated sludge plants. Ciliates might, however, affect the dynamics of amino acid pools in environments high in nutrients and ciliates, perhaps including some microenvironments near decomposing material or in benthic sediments. Direct uptake of dissolved amino acids by ciliates probably does not affect ciliate or bacterial populations substantially.  相似文献

Summary. The free amino acid compositions in archaeobacteria, eubacteria, protozoa, blue-green alga, green alga, slime mold, plants and mammalian cells were analyzed, to investigate whether changes in their free amino acid compositions reflect biological evolution. Cell homogenates were treated with 80–90% ethanol to separate cellular proteins and free amino acids contained in the cells. Different patterns of the free amino acid compositions were observed in the various organisms. Characteristic differences were observed between plant and mammalian cells, and between archaeobacteria and eubacteria. The patterns of the free amino acid composition in blue-green alga, green alga, protozoa and slime mold differed from each other and from those of eubacteria and archaeobacteria. Rat hepatoma cells (R-Y121B) were cultured in Eagle's minimum essential medium (MEM) containing 5% serum or in a modified MEM lacking arginine, tyrosine and glutamine. No significant difference in the free amino acid composition was observed between the two cell groups cultured under two different conditions. Summary Dieary lipids strongly influence the pattern of torpor and the body lipid composition of mammalian hibernators. The object of the present study was to investigate whether these diet-induced physiological and biochemical changes also occur in species that show shallow, daily torpor. Deer mice, Peromyscus maniculatus, were fed with rodent chow (control diet) or rodent chow with either 10% sunflower seed oil (unsaturated diet) or 10% sheep fat (saturated diet). Animals on the unsaturated diet showed a greater occurrence of torpor (80–100% vs 26–43%), longer torpor bouts (4.5 vs 2.25 h), a lower metabolic rate during torpor (0.96 vs 2.25 ml O2·g-1·h-1), and a smaller loss of body mass during withdrawal of food (2.35 vs 3.90 g) than animals on the saturated diet; controls were intermediate. These diet-induced physiological changes were associated with significant alterations in the fatty acid composition of depot fat, leg muscle and brain total lipids, and heart mitochondrial phospholipids. Significant differences in the total unsaturated fatty acid (UFA) content between animals on saturated and unsaturated diet were observed in depot fat (55.7% vs 81.1%) and leg muscle (56.4% vs 72.1%). Major compositional differences between diet groups also occurred in the concentration of n6 and/or n3 fatty acids of brain and heart mitochondria. There are increasing interests in rearing edible insects in Africa, but information on how the feeds modify their fatty acids is largely lacking. In this work, the influence of artificial diets on the fatty acid contents and composition in the edible Ruspolia differens (Serville, 1838), in Uganda was assessed. R. differens was reared on the mixtures of six gradually diversified diets of two, three, four, six, eight and nine feeds. The diets were formulated from rice seed head, finger millet seed head, wheat bran, superfeed chicken egg booster, sorghum seed head, germinated finger millet, simsim cake, crushed dog biscuit pellet and shea butter. Fatty acid methyl esters were prepared using direct transesterification method, and analysed using gas chromatography. The contents of saturated, monounsaturated and polyunsaturated fatty acid differed significantly among the diets. The more diverse diets resulted in increased content of the polyunsaturated fatty acids. The n6:n3 ratio differed significantly among the diets and between the sexes, with R. differens fed on the four-feed diet having a higher n6:n3 ratio than those fed on other diets. Also, the fatty acid composition differed significantly among the diets, and diet diversification corresponded with the proportions of polyunsaturated fatty acids, especially linoleic acid. Overall, our results demonstrate that higher levels of essential fatty acids can be achieved by rearing R. differens on highly diversified diets. These findings are important in informing the design of future mass-rearing program for this edible insect.  相似文献

Intrauterine growth retardation (IUGR) induced by ligation of one uterine artery on day 17 of pregnancy in the rat lead to major abnormalities in the fatty acid content of neurons and oligodendrocytes but not in astrocytes. In neurons from IUGR rats, monounsaturated fatty acids were decreased; in the polyunsaturated series, ω-3 fatty acids were increased and Ω-6 fatty acids were decreased. In oligodendrocytes, monounsaturated fatty acids were also decreased, but the modifications in polyunsaturated fatty acids were the opposite of those in neurons: Ω-3 being decreased and w-6 increased. Although the animals received a normal diet after birth, the alterations were still present in adulthood. In addition, fatty acid composition of brain cells is a very indicative criterion of brain maturation.  相似文献
