When infested with the fungus Rhizoctonia leguminicola, certain forages, e.g., red clover hay, can cause a "slobber syndrome" of varying severity when consumed by ruminants. The causative agent has been presumed to be slaframine [(1S,6S,8aS)-1-acetoxy-6-aminooctahydroindolizine], which is produced by R. leguminicola. In one serious outbreak of the slobber syndrome in horses, the red clover forage involved was carefully examined and found to contain R. leguminicola and slaframine. An identical hay sample is shown here by ion-exchange chromatographic and gas chromatographic-mass spectrometric analysis of appropriate hay extracts to also contain swainsonine [(1S,2R,8R,8aR)-1,2,8-trihydroxyoctahydroindolizine], a potent alpha-mannosidase inhibitor. Swainsonine has previously been isolated from pure cultures of R. leguminicola and from higher plants, namely the Darling pea (Swainsona canescens) and spotted locoweed (Astragalus lentiginosus). Consumption of Darling pea and spotted locoweed by livestock results in a severe neurological condition resembling that observed in hereditary mannosidosis in cattle and humans. Our findings indicate that swainsonine may be viewed as a mycotoxin when present in moldy forages consumed by livestock. The extent to which slaframine and swainsonine mycotoxicosis pose threats to animal husbandry and, indeed, to humans, if these alkaloids were to enter the human food chain, deserves serious consideration.  相似文献

The occurrence of the mycotoxin penitrem A in refrigerated cream cheese that was involved in the intoxication of two dogs is described. Penitrem A and the fungus Penicillium crustosum were isolated from the cream cheese and identified. This is believed to be the first definitive case of the natural occurrence of penitrem A.  相似文献

The direct-acting mutagens in diesel particulate extracts were identified. It is concluded that the major mutagens are in all probability 1,6- and 1,8-dinitropyrene (DNP). 1-Nitropyrene (NP) and 3-nitrofluoranthene (NF) were also present. The DNP isomers contributed 43% of the total mutagenic activity of the crude extracts, whereas 1-NP (or 3-NF) was responsible for less than 10% of the activity. The quantities of 1,6- and 1,8-DNP were 1.2 and 3.4 ppm of the crude extracts, respectively, and the induction of both DNPs in the diesel particulate matter corresponded to about 1.7-4.8% by weight of the 1-NP content (70.5 ppm in the crude extracts).  相似文献

Fusarium tricinctum strain (KF 260) isolated from wheat in Poland, produced on maize cultures a compound (C11H14O5, MW226) toxic toArtemia salina. The compound, identified by various spectroscopical tecniques as 5-hydroxy-4-methoxy-6,8a-dimethyl-6, 7-dihydro-2H, 8aH-pyrano/2, 3-b/pyran-2-one, was identical to chlamydosporol, a mycotoxin recentlydiscovered in cultures ofF._ chlamydosporum isolated from rice. The compound showed inhibitory effect on human lymphocyte proliferation at concentration of 25 μg/mL. LD50 onA. salina was 56 μg/mL.  相似文献

Evolutionary pressure has selected individuals with traits that allow them to survive to reproduction, without consideration of the consequences for the post-child rearing years and old age. In the 21st century, society is populated increasingly by the elderly and with the falling birth rate and improved health care this trend is set to continue for the foreseeable future. To minimize the potential burden on health services one would hope that 'growing old gracefully' should also mean 'growing old healthily'. However, for too many the aging process is accompanied by increasing physical and mental frailty producing an elevated risk of physical and psychological stress in old age. Stress is a potent modulator of immune function, which in youth can be compensated for by the presence of an optimal immune response. In the elderly the immune response is blunted as a result of the decline in several components of the immune system (immune senescence) and a shifting to a chronic pro-inflammatory status (the so-called 'inflamm-aging' effect). We discuss here what is known of the effects of both stress and aging upon the innate immune system, focusing in particular upon the age-related alterations in the hypopituitary-adrenal axis. We propose a double hit model for age and stress in which the age-related increase in the cortisol/sulphated dehydroepiandrosterone ratio synergizes with elevated cortisol during stress to reduce immunity in the elderly significantly.  相似文献

Solvent extracts of soybeans inoculated with 10 strains of S. sclerotiorum were toxic on injection but nontoxic on per os administration to mice. Weaning Swiss mice were percutaneously infected with 30 cercariae of Schistosoma mansoni and submitted to a shifting either from a deficient to a balanced diet or vice-versa, for 24 weeks. The nutritional status was weekly evaluated by measurements of growth curves and food intake. Hepatic fibrosis and periovular granulomas were studied by histological, morphometric and biochemical methods. All mice fed on a deficient diet failed to develop periportal "pipestem" fibrosis after chronic infection. An unexpected finding was the absence of pipestem fibrosis in mice on normal diet, probably related to the sample size. The lower values for nutritional parameters were mainly due to the deficient diet, rather than to infection. Liver/body weight ratio was higher in "early undernutrition" group, after shifting to the balanced diet. Volume density and numerical density of egg granulomas reached lowest values in undernourished animals. The amount of collagen was reduced in undernourished mice, attaining higher concentrations in well-fed controls and in "late undernutrition" (balanced diet shifted to a deficient one), where collagen deposition appeared increased in granulomas. That finding suggested interference with collagen degradation and resorption in "late" undernourished animals. Thus, host nutritional status plays a role in connective tissue changes of hepatic schistosomiasis in mice.  相似文献

We examined the blastogenic response to phytohaemaglutinin (PHA) in HLA-B8, DR3 positive and negative subjects in the presence or absence of the immunosuppressive Fusarium mycotoxin. HLA-B8, DR3 haplotype was associated with a depression of the response to mitogen in the absence of the mycotoxin, whereas in the presence of deoxynivalenol we could not detect significant differences among individuals either possessing or lacking this haplotype.  相似文献

Taste signal is received in taste buds and transmitted via sensory afferent nerves to the brainstem. Although a signaling pathway involving phospholipase C-β2 has been shown to transduce taste signals of bitterness, sweetness and umami in taste receptor cells (Type II cells), these taste receptor cells appear to be different from the presynaptic cells (Type III cells) containing afferent synapses associated with nerve processes. To elucidate the neurotransmission system in the taste receptor cells expressing phospholipase C-β2, we searched for candidate molecules involved in the neurotransmission, and identified synaptophysin. Synaptophysin was expressed in the taste receptor cells expressing phospholipase C-β2, as well as in the presynaptic cells harboring synaptic structures with taste nerves and containing serotonin. Synaptophysin-immunoreactive signals were not limited to gustducin-positive bitter taste receptor cells, and sweet/umami taste receptor cells were indicated to also express synaptophysin. Expression of synaptophysin was already initiated 6 days after cell division, almost in synchrony with the initiation of phospholipase C-β2 expression. Synaptophysin-containing cells co-expressed vesicular-associated membrane protein 2, a v-SNARE molecule which is important for exocytosis. In addition, majority of the synaptophysin-expressing cells also expressed cholecystokinin, a neuropeptide expressed in taste buds. These results suggest that the taste receptor cells have a neurotransmission system involving synaptophysin, which occurs alternatively or additionally to a recently shown hemichannel system.  相似文献

Determining what constitutes protective immunity to TB is critical for the development of improved diagnostics and vaccines. The comparison of the immune system between contacts of TB patients, who later develop TB disease (progressors), versus contacts who remain healthy (non-progressors), allows for identification of predictive markers of TB disease. This study provides the first comprehensive analysis of the immune system of progressors and non-progressors using a well-characterised TB case-contact (TBCC) platform in The Gambia, West Africa. 22 progressors and 31 non-progressors were analysed at recruitment, 3 months and 18 months (time to progression: median[IQR] of 507[187-714] days). Immunophenotyping of PBMC, plasma cytokine levels and RT-MLPA analysis of whole blood-derived RNA was performed to capture key immune system parameters. At recruitment, progressors had lower PBMC proportions of CD4+ T cells, NKT cells and B cells relative to non-progressors. Analysis of the plasma showed higher levels of IL-18 in progressors compared to non-progressors and analysis of the RNA showed significantly lower gene expression of Bcl2 but higher CCR7 in progressors compared to non-progressors. This study shows several markers that may predict the onset of active TB at a very early stage after infection. Once these markers have been validated in larger studies, they provide avenues to prospectively identify people at risk of developing TB, a key issue in the testing of new TB vaccines.  相似文献

Renal biopsy of two children and a maternal relative, diagnosed with severe progressive tubulointerstitial nephritis, has shown the presence of distorted mitochondria. Mitochondrial DNA from the blood of these patients was analysed. No major deletions were found, but an A to G mutation was detected in position 5656. Sprouted barley grains, the waste product of malt extract production, were incriminated as the cause of a lethal (96% mortality) neurotoxic syndrome in sheep fed the grains. Clinical manifestations, comprising tremors, lameness, abnormal gait, paralysis and death indicated a tremorgenic mycotoxicosis. Whilst 50% of the flock died within 17 days, mortality continued over more than 5 months. Pathological findings were limited to neuronal degeneration and necrosis in the midbrain. Germinating grains were shown to be contaminated with growth of Aspergillus clavatus.  相似文献

Our results demonstrate high concentrations of the UV absorbing mycotoxin citrinin in the outer layer of spores from three citrinin-producing strains of Penicillium verrucosum, which is released in an aqueous environment. An important function of the toxin could be to act as a sun protectant in order to create favorable conditions during the initial germination process. When spores from these strains of P. verrucosum were examined by confocal microscopy, a clearly visible fluorescent layer associated with the cell wall was observed. The strains were grown on agar plates, and the mycelial mat was washed with saline. This suspension contained at least 95% of the spores and particulate material, which was removed by filtration after counting the conidia. An aliquot of this filtrate was extracted and citrinin was purified by high pressure liquid chromatography. The absorbance at 319 nm was used to calculate the amount of UV absorbing material released from the spores. Based on the spore numbers in the suspension of the saline extract, we estimated that this material released was 1.4–4.1 pg per spore or 8–24% of the spore weight. Citrinin (and minor amounts of ochratoxin A and some other unidentificable fluorescent compounds) were observed in the filtrate when subjected to thin layer chromatography. In previous investigations, it was necessary to have Fe(II) and cysteine present in order to assay the catalytic activity of purified hog kidney myo-inositol oxygenase. In the present study it was found that, if this purified nonheme iron enzyme is slowly frozen in solution with glutathione and stored at -20 degrees C, it is fully active in the absence of activators if catalase is present to remove adventitious H2O2. With this simpler assay system it was possible to clarify the effects of several variables on the enzymic reaction. Thus, the maximum velocity is pH-dependent with a maximum around pH 9.5, but the apparent Km for myo-inositol (air atmosphere) remains constant at 5.0 mM throughout a broad pH range. The enzyme is quite specific for its substrate myo-inositol, is very sensitive to oxidants and reductants, but is not affected by a variety of complexing agents, nucleotides, sulfhydryl reagents, etc. In other experiments it was found that L-myo-inosose-1, a potential intermediate in the enzymic reaction, is a potent competitive inhibitor (Ki = 62 microM), while other inososes and a solution thought to contain D-glucodialdehyde, another potential intermediate, are weak inhibitors. Also, both a kinetic deuterium isotope effect (kH/kD = 2.1) and a tritium isotope effect (kH/kT = 7.5) are observed for the enzymic reaction when [1-2H]- and [1-3H]-myo-inositol are used as reactants. These latter results are considered strong evidence that the oxygenase reaction proceeds by a pathway involving L-myo-inosose-1 as an intermediate rather than by an alternative pathway that would have D-glucodialdehyde as the intermediate.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献
