Biochemical variation at 14 blood loci was reviewed, and specific features compared experimentally in sheep Ovis aries, mouflon Ovis musimon, goat Capra hircus, aoudad Ammotragus lervia and in 2 stillborn aoudad × goat hybrids. Variation at 3 loci was also studied in dall sheep Ovis dalli, bighorn sheep Ovis canadensis and rocky mountain goat Oreamnos americanus. Haemoglobin C production in an anaemic Hb AB mouflon and in mouflon × sheep hybrids was examined. Mouflon differ from domestic sheep in that synthesis of both Hb βAHb βBchains is switched off during Hb C production. The mouflon × sheep hybrids switched off one or both chains depending on whether they had inherited sheep or mouflon Hb β chain genes. In general aoudad showed a closer affinity to goats than to sheep.  相似文献

A total of 20990 Finnish Ayrshire cows were phenotyped for the major milk proteins by isoelectric focusing in polyacrylamide gels. The predominant alleles in the Finnish Ayrshire were αS1-casein B (0.999), αS2-casein A (0.991), β-casein A1 (0.509) and α2 (0.490), α22-casein A (0.612) and β-lactoglobulin B (0.716). The K-casein E allele (0.307) was also rather common in the Finnish Ayrshire. A new K-casein variant (K-casein F) was demonstrated in two Finnish Ayrshire cows, a dam and a daughter.  相似文献

Abstract: Angiotensin IV (Val-Tyr-Ile-His-Pro-Phe) has been reported to interact with specific high-affinity receptors to increase memory retrieval, enhance dopamine-induced stereotypy behavior, and induce c- fos expression in several brain nuclei. We have isolated a decapeptide (Leu-Val-Val-Tyr-Pro-Trp-Thr-Gln-Arg-Phe) from sheep brain that binds with high affinity to the angiotensin IV receptor. The peptide was isolated using 125I-angiotensin IV binding to bovine adrenal membranes to assay receptor binding activity. This peptide is identical to the amino acid sequence 30–39 of sheep βA- and βB-globins and has previously been named LVV-hemorphin-7. Pharmacological studies demonstrated that LVV-hemorphin-7 and angiotensin IV were equipotent in competing for 125I-angiotensin IV binding to sheep cerebellar membranes and displayed full cross-displacement. Using in vitro receptor autoradiography, 125I-LVV-hemorphin-7 binding to sheep brain sections was identical to 125I-angiotensin IV binding in its pattern of distribution and binding specificity. This study reveals the presence of a globin fragment in the sheep brain that exhibits a high affinity for, and displays an identical receptor distribution with, the angiotensin IV receptor. This globin fragment, LVV-hemorphin-7, may therefore represent an endogenous ligand for the angiotensin IV receptor in the CNS.  相似文献

The alternative sigma factor σE is activated by unfolded outer membrane proteins (OMPs) and plays an essential role in Salmonella pathogenesis. The canonical pathway of σE activation in response to envelope stress involves sequential proteolysis of the anti-sigma factor RseA by the PDZ proteases DegS and RseP. Here we show that σE in Salmonella enterica sv. Typhimurium can also be activated by acid stress. A σE-deficient mutant exhibits increased susceptibility to acid pH and reduced survival in an acidified phagosomal vacuole. Acid activation of σE-dependent gene expression is independent of the unfolded OMP signal or the DegS protease but requires processing of RseA by RseP. The RseP PDZ domain is indispensable for acid induction, suggesting that acid stress may disrupt an inhibitory interaction between RseA and the RseP PDZ domain to allow RseA proteolysis in the absence of antecedent action of DegS. These observations demonstrate a novel environmental stimulus and activation pathway for the σE regulon that appear to be critically important during Salmonella –host cell interactions.  相似文献

Summary
Using agarose gel isoelectric focusing and immunoblotting with rabbit anti-rabbit C6, a genetic polymorphism has been found in the sixth component of complement (C6) in six breeds of pigs. The C6 locus was highly polymorphic. Family data indicated that pig C6 pheno-types were inherited by means of five codomonant alleles named C6 A, C6 B, C6 C, C6 D and C6 E at a single autosomal locus. C6 deficiency in two of 241 individuals tested was found, which suggested the presence of a null allele in pig populations. Marked breed differences among the gene frequencies and heterozygosities at C6 locus were observed.  相似文献

Summary. Milk samples from 189 Merinoland Sheep, 145 Black Faced Mutton Sheep, 89 East Friesian Milk Sheep, 36 Rhön, 36 Pleven, 23 Tsigaja, 25 Black Razka and 86 Hungarian Merino x Pleven (F1) sheep were analysed by polyacrylamide gel electrophoresis under acid conditions and isoelectric focusing in ultrathin layer polyacrylamide gels with carrier ampholytes. Six different β-lactoglobulin (β-Lg) phenotypes (A, AB, B, AC, BC and C) were observed by both methods. The occurrence of three codominant alleles (β-LgA, β-LgB, β-Lgc) at an autosomal locus (β-Lg) was supported by family and population data on genetic equilibrium. Differences in gene frequencies between the breeds were observed.  相似文献

A method for genotyping K-casein ( A, B, E ), β-casein ( A 1, A 2, A 3, A5, B ) and β-lactoglobulin ( A, B ) simultaneously by the use of allele discrimination by primer length combined with automated detection of fragments with a sequencing instrument is described. Seven different mutations within the milk protein genes were analysed in order to distinguish between the alleles examined. The samples were amplified in two separate multiplex polymerase chain reactions (PCRs), which were then pooled and separated according to size in a single lane on the gel. By using stringent PCR conditions, we have been able to achieve allele-specific amplifications and minimize amplification of mismatched primer for all seven mutations.  相似文献

A gene ( sigF ) encoding a new sigma factor was isolated from Streptomyces aureofaciens using a degenerate oligonucleotide probe designed from the GLI(KDNE)A motif lying within the well-conserved region 2.2 of the eubacterial σ70 family. Homologues were present in other Streptomyces spp., and that of the genetically well-studied Streptomyces coelicolor A3(2) was also cloned. The nucleotide sequences of the two sigF genes were determined and shown to encode primary translation products of 287 ( S. coelicolor ) and 295 ( S. aureofaciens ) amino acid residues, both showing greatest similarity to σB of Bacillus subtilis . However, while σB is involved in stationary-phase gene expression and in the general stress response in B. subtilis , σF affects morphological differentiation in Streptomyces , Disruption of sigF did not affect vegetative growth but did cause a whi mutant phenotype. Microscopic examination showed that the sigF mutant produced spores that were smaller and deformed compared with those of the wild type, that the spore walls were thinner and sensitive to detergents and that in sigF mutant spores the chromosome failed to condense. σF is proposed to control the late stages of spore development in Streptomyces .  相似文献

By means of starch gel electrophoresis the polymorphism of the 6-phosphogluconate dehydrogenase (6PGD) has been found in cattle leucocytes. The phenotypes (A, AB and B) and allele frequencies ( 6PGD A= 0.425; 6PGD B= 0.575) are demonstrated in families of German Friesian cattle.  相似文献

A genetic polymorphism of the pig amylase 2 system is described. It is controlled by two codominant genes, Am 2A and Am 2B. A comparison of some breeds from Byelorussian breeding farms gives the following frequencies for the alleles Am 2A and Am 2B , respectively: 0.35 and 0.865 in Large White (n = 682), 0.257 and 0.743 in Byelorussian Black and White (n = 400), 0.540 and 0.460 in Hampshire (n = 51).
While studying the genotype distribution according to the Hardy-Weinberg law, a genetic imbalance in Byelorussian Black and White pigs was established (χ2= 56.4). Genetic relationship between the Am 1 and Am 2 loci was not found.  相似文献

The RsbT serine kinase has two known functions in the signal transduction pathway that activates the general stress factor σB of Bacillus subtilis . First, RsbT can phosphorylate and inactivate its specific antagonist protein, RsbS. Second, upon phosphorylation of RsbS, RsbT is released to stimulate RsbU, a PP2C phosphatase, thereby initiating a signalling cascade that ultimately activates σB. Here we describe a mutation that separates these two functions of RsbT. Although the mutant RsbT protein had essentially no kinase activity, it still retained the capacity to stimulate the RsbU phosphatase in vitro and to activate σB when overexpressed in vivo . These results support the hypothesis that phosphatase activation is accomplished via a long-lived interaction between RsbT and RsbU. In contrast, RsbT kinase activity was found to be integral for the transmission of external stimuli to σB. Thus, one route by which environmental stress signals could enter the σB network is by modulation of the RsbT kinase activity, thereby controlling the magnitude of the partner switch between the RsbS–RsbT complex and the RsbT–RsbU complex.  相似文献

Genetic polymorphism was found in the isozymes of leucine aminopeptidase in the canine plasma. Analysis of parentage records of the dogs examined revealed that the phenotypic variation of leucine aminopeptidase isozymes was controlled by one autosomal locus, designated Lap , with two codominant gene alleles, Lap A and Lap B, though individuals of homozygous B type were not found in the specimen used. The gene frequencies were 0.97 for Lap A and 0.03 for Lap B in most of the dog breeds examined. Higher frequencies for Lap B (0.11–0.25) were obtained in breeds such as Pointer, Doberman pinscher, Boxer, Borzoi and San'in-Shiba (a Japanese native breed), while in German shepherd, Collie, and two of the Japanese native breeds, Akita and Kishu, phenotype of the isozyme seemed to be fixed to homozygous A ( Lap A/ Lap A).  相似文献

The induction of stress proteins is an important component of the adaptional network of a non-growing cell of Bacillus subtilis . A diverse range of stresses such as heat shock, salt stress, ethanol, starvation for oxygen or nutrients etc. induce the same set of proteins, called general stress proteins. Although the adaptive functions of these proteins are largely unknown, they are proposed to provide general and rather non-specific protection of the cell under these adverse conditions. In addition to these non-specific general stress proteins, all extracellular signals induce a set of specific stress proteins that may confer specific protection against a particular stress factor. In B. subtilis at least three different classes of heat-inducible genes can be defined by their common regulatory characteristics: Class I genes, as exemplified by the dnaK and groE operons, are most efficiently induced by heat stress. Their expression involves a σA-dependent promoter, an inverted repeat (called the CIRCE element) highly conserved among eubacteria, and probably a repressor interacting with the CIRCE element. The majority of general stress genes (class II, more than 40) are induced at σB-dependent promoters by different growth-inhibiting conditions. The activation of σB by stress or starvation is the crucial event in the induction of this large stress regulon. Only a few genes, including lon clpC clpP , and ftsH, can respond to different stress factors independently of σB or CIRCE (class III). Stress induction of these genes occurs at promoters presumably recognized by σA and probably involves additional regulatory elements which remain to be defined.  相似文献
