We recorded neurons from the hippocampus of freely behaving rats during an auditory fear conditioning task. Rats received either paired or unpaired presentations of an auditory conditioned stimulus (CS) and an electric shock unconditioned stimulus (US). Hippocampal neurons (place and theta cells) acquired responses to the auditory CS in the paired but not in the unpaired group. After CS-US pairing, rhythmic firing of theta cells became synchronized to the onset of the CS. Conditioned responses of place cells were gated by their location-specific firing, so that after CS-US pairing, place cells responded to the CS only when the rat was within the cell's place field. These findings may help to elucidate how the hippocampus contributes to context-specific memory formation during associative learning.  相似文献

Male hooded rats were habituated, classically conditioned with 30 CS-UCS (light-footshock) pairings, and subsequently tested for corticosterone response or instrumental hurdle-jump acquisition. In Experiment 1, corticosterone levels were lowest during chamber placement alone (during habituation), higher during presentations of the CS after conditioning with a low shock intensity, even higher during classical conditioning with the low shock intensity, and highest during classical conditioning or CS presentations involving a high shock intensity. Injections of the synthetic glucocorticoid dexamethasone before both conditioning and hurdle-jump acquisition sessions (Experiment 2) did not affect acquisition occurring during early trials, but produced slow hurdle-jump speeds late in the session. This agrees with previous findings of glucocorticoid facilitation of fear extinction, but does not indicate a simple suppression of fear by dexamethasone. When dexamethasone was given only prior to the classical conditioning session (Experiment 3) hurdle-jump acquisition was poor only on the early trials, and corticosterone levels after 60 min of CS presentations were higher than control values. These results agree with the proposal of a state-dependent effect of dexamethasone on memory retrieval.  相似文献

Many fear conditioning studies use electric shock as the aversive stimulus. The intensity of shocks varies throughout the literature. In this study, shock intensities ranging from 0 to 1.5 mA were used, and the effects on the rats assessed by both behavioural and biochemical stress parameters. Results indicated a significant difference with respect to defaecation and freezing behaviour between controls and those animals that received a shock. Significant differences in corticosterone levels were also noted between controls and those groups that received a shock. No significant differences were found between the shock groups with regards to the stress parameters measured in our fear conditioning paradigm, indicating that the two shock groups were similarly stressed. Increased significance levels were noted for freezing behaviour as well as a lower standard error of means was found in the highest shock intensity group. We therefore recommend using the higher shock intensity (1.5 mA) as the rats in the higher shock intensity group were more homogeneously fear-conditioned and therefore the results should be more reproducible and robust than in the lower shock intensity group. This would allow for fewer rats to be used in order to gain an accurate impression of the conditioning paradigm employed.  相似文献

One group (N = 14) of human volunteers received three sessions of discriminated avoidance and punishment with the skin resistance response (SRR) as the instrumental behavior. Each session consisted of three 7 minute periods of Sidman avoidance (response-stimulus [R-S] and stimulus-stimulus [S-S] = 40 sec) of a 0.5 second, 15 Hz square wave shock mixed with three periods of punishment with the same shock. The avoidance and punishment periods were differentially signaled by red and green lights, and a circle appeared superimposed on the discriminative stimuli during a criterion SRR. A second group (N = 14) was yoked to the first by recording shock events on magnetic tape and shocking the yoked subject in the same temporal sequence. In both groups the visual feedback stimulus was consistently related to the subject's ongoing electrodermal behavior. As in previous research, the contingent shock subjects made significantly more SRRs during avoidance than during punishment in all three sessions. For yoked subjects the discrimination did not appear until the third session. The findings imply that the discrimination is at least partly independent of the avoidance and punishment contingencies, and they raise questions about the role of the feedback stimulus.  相似文献

The present experiments were designed to determine if repeated presentations of an empty sipper tube (the conditioned stimulus or CS) with the response-independent delivery of a sucrose solution (the unconditioned stimulus or US) from a second spout results in the development of Pavlovian conditioned responding. In Experiment 1, rats in the experimental condition received paired CS-US presentations whereas subjects in the control condition were exposed to random presentations of CS and US. In Experiment 2, an additional control condition (CS alone) was included and, to encourage generalized responding between the US and CS, the CS tube was filled with water for all groups. The results of both experiments indicate that the CS-directed responding in the paired CS-US condition was Pavlovian in nature. Thus, the present procedure serves as an autoshaping task in which conditioned licking is generated.  相似文献

Heart rate (HR), freezing score, and motor component were estimated during acoustic startle (ASR) habituation (two sessions with 24-hour interval, 10 trials in a session). It was shown that rats previously exposed to the experimental context (once for 5 min 24 h before training) demonstrated HR decrease in response to the first stimulus and tachycardia in response to the 4th-10th stimuli during the first session. The interstimulus HR declined from the 4th to the 6th trials. The same profile of cardiovascular response was observed in this group at the beginning of the second session with the following (to the 7th trial) habituation of tachycardia. These rats didn't demonstrate the intertrial HR decrease during the second session. Nonadapted rats responded by bradycardia to the 1st and 2nd trials of the first session. The response didn't change for tachycardia with continuation of stimuli presentation. Tachycardia appeared only in response to the 7th-10th trials of the second session and didn't habituate. The intertrial HR level decreased in this group only during the second session. The results are discussed in terms of contextual latent inhibition of the cardiac acoustic startle response.  相似文献

Summary By changing the conditioned discrimination paradigm of Quinn et al. (1974) from an instrumental procedure to a classical (Pavlovian) one, we have demonstrated strong learning in type flies. About 150 flies were sequestered in a closed chamber and trained by explosing them sequentially to two odors in air currents. Flies received twelve electric shock pulses in the presence of the first odor (CS+) but not in the presence of the second odor (CS–). To test for conditioned avoidance responses, flies were transported to a Tmaze choice point, between converging currents of the two odors. Typically, 95% of trained flies avoided the shock-associated odor (CS+).Acquisition of learning was a function of the number of shock pulses received during CS+ presentation and was asymptotic within one training cycle. Conditioned avoidance increased with increasing shock intensity or odor concentration and was very resistant to extinction. Learning was best when CS+ presentations overlap shock (delay conditioning) and then decreased with increasing CS-US interstimulus intervals. Shocking flies immediately before CS+ presentation (backward conditioning) produced no learning. Nonassociative control procedures (CS Alone, US Alone and Explicitly Unpaired) produced slight decreases in avoidance responses, but these affected both odors equally and did not alter our associative learning index (A).Memory in wild-type flies decayed gradually over the first seven hours after training and still was present 24 h later. The mutantsamnesiac, rutabaga anddunce showed appreciable learning acquisition, but their memories decayed very rapidly during the first 30 min. Animals need to associate different environmental stimuli with each other regardless of whether they temporally overlap or not. Drosophila melanogaster displays olfactory trace conditioning, where an odor is followed by electric shock reinforcement after a temporal gap, leading to conditioned odor avoidance. Reversing the stimulus timing in olfactory conditioning results in the reversal of memory valence such that an odor that follows shock is later on approached (i.e. relief conditioning). Here, we explored the effects of stimulus timing on memory in another sensory modality, using a visual conditioning paradigm. We found that flies form visual memories of opposite valence depending on stimulus timing and can associate a visual stimulus with reinforcement despite being presented with a temporal gap. These results suggest that associative memories with non-overlapping stimuli and the effect of stimulus timing on memory valence are shared across sensory modalities.  相似文献

MultiCS conditioning is an affective associative learning paradigm, in which affective categories consist of many similar and complex stimuli. Comparing visual processing before and after learning, recent MultiCS conditioning studies using time-sensitive magnetoencephalography (MEG) revealed enhanced activation of prefrontal cortex (PFC) regions towards emotionally paired versus neutral stimuli already during short-latency processing stages (i.e., 50 to 80 ms after stimulus onset). The present study aimed at showing that this rapid differential activation develops as a function of the acquisition and not the extinction of the emotional meaning associated with affectively paired stimuli. MEG data of a MultiCS conditioning study were analyzed with respect to rapid changes in PFC activation towards aversively (electric shock) paired and unpaired faces that occurred during the learning of stimulus-reinforcer contingencies. Analyses revealed an increased PFC activation towards paired stimuli during 50 to 80 ms already during the acquisition of contingencies, which emerged after a single pairing with the electric shock. Corresponding changes in stimulus valence could be observed in ratings of hedonic valence, although participants did not seem to be aware of contingencies. These results suggest rapid formation and access of emotional stimulus meaning in the PFC as well as a great capacity for adaptive and highly resolving learning in the brain under challenging circumstances.  相似文献

The several functions that a stimulus can assume were investigated in a Pavlovian conditioning procedure. The subjects were six rhesus monkeys; the response under observation was heart rate. The conditioning began with a temporal separation of zero between a signal and a regularly repeating electric shock; the signal was then moved to a series of earlier locations in the inter-shock interval. After six sessions at each location, two sessions followed in which only the shock was delivered periodically. The findings included: (1) A two-phased conditioned cardiac rate response seen at the first location became more multiphasic and irregular during longer intervals between signal and shock; (2) the location where the conditioned response peaked became increasingly variable as the signal was moved back, but this variability maintained a constant proportion to the signal-shock interval; and (3) heart rate during a presignal period, and during a comparable period in shock only sessions, was generally deceleratory early in training and acceleratory thereafter. Sessions with the signal showed heart rate in the presignal period to have become acceleratory earlier in training than sessions with shock only. The data pertain to stimulus control over heart rate as a function of: (A) the temporal proximity of a signal to an aversive stimulus; and, (B) the presence or absence of the signal. The use of appropriate response units in cardiac conditioning is also discussed.  相似文献

Three experiments were conducted using a conditioned taste aversion procedure with rats to examine the effect of nonreinforced presentations of a conditioned stimulus (CS) on its ability to compete with a target stimulus for manifest conditioned responding. Two CSs (A and B) were presented in a serial compound and then paired with the unconditioned stimulus. CS A was first paired with the US and then presented without the US (i.e., extinction) prior to reinforced presentation of the AB compound. Experiment 1 showed that A was poor at competing with B for conditioned responding when given conditioning and extinction prior to reinforcement of AB relative to a group that received both A and B for the first time during compound conditioning. That is, an extinguished A stimulus allowed greater manifest acquisition to B. Experiment 2 found that extinction treatment produced a poor CR to the pretrained and extinguished CS itself following compound conditioning. Experiment 3 found that interposing a retention interval after extinction of A and prior to compound conditioning enhanced A's ability to compete with B. The results of these experiments are discussed with regard to different theories of extinction and associative competition.  相似文献

目的:观察Orexin受体1对摄食条件反射的调控研究。方法:将40只大鼠随机分为四组,分别为NS/NS组,SB/SB组,NS/SB组,SB/NS组,每组10只大鼠,给予大鼠八组"条件刺激-无条件刺激(CS-US)"训练,给予无条件刺激后立即进行条件刺激,每组进行训练前30 min给予SB或生理盐水(NS)。获得性训练后,给予2组反射消失性训练,即给予8次条件刺激。条件刺激是给予大鼠10 s,2kHZ声音刺激,无条件刺激是直接给予大鼠食物。结果:给予条件刺激后,四组大鼠摄食行为均明显增加,摄食间隔均明显缩短,当条件刺激强度增加时,摄食行为也增加,而预先给予SB,与NS/NS组相比,其余组大鼠摄食行为相对减少(P0.05),摄食间隔增加。消失性训练中,与NS/NS,NS/SB组相比,SB/NS组和SB/SB组大鼠摄食行为明显减少(P0.05)。与其他三组大鼠相比,SB/NS组大鼠摄食间隔缩短。预先给予SB使后两次刺激后摄食间隔明显增加(P0.05)。结论:OX1R信号通路调控摄食反射的产生和消失。  相似文献

In this experiment we present a technique to measure learning and memory. In the trace fear conditioning protocol presented here there are five pairings between a neutral stimulus and an unconditioned stimulus. There is a 20 sec trace period that separates each conditioning trial. On the following day freezing is measured during presentation of the conditioned stimulus (CS) and trace period. On the third day there is an 8 min test to measure contextual memory. The representative results are from mice that were presented with the aversive unconditioned stimulus (shock) compared to mice that received the tone presentations without the unconditioned stimulus. Trace fear conditioning has been successfully used to detect subtle learning and memory deficits and enhancements in mice that are not found with other fear conditioning methods. This type of fear conditioning is believed to be dependent upon connections between the medial prefrontal cortex and the hippocampus. One current controversy is whether this method is believed to be amygdala-independent. Therefore, other fear conditioning testing is needed to examine amygdala-dependent learning and memory effects, such as through the delay fear conditioning.  相似文献

Habituation appears to play a role in the decline in wheel running within an interval. Aoyama and McSweeney [Aoyama, K., McSweeney, F.K., 2001. Habituation contributes to within-session changes in free wheel running. J. Exp. Anal. Behav. 76, 289-302] showed that when a novel stimulus was presented during a 30-min interval, wheel-running rates following the stimulus increased to levels approximating those earlier in the interval. The present study sought to assess the role of habituation in the decline in running that occurs over a briefer interval. In two experiments, rats responded on fixed-interval 30-s schedules for the opportunity to run for 45 s. Forty reinforcers were completed in each session. In the first experiment, the brake and chamber lights were repeatedly activated and inactivated after 25 s of a reinforcement interval had elapsed to assess the effect on running within the remaining 20 s. Presentations of the brake/light stimulus occurred during nine randomly determined reinforcement intervals in a session. In the second experiment, a 110 dB tone was emitted after 25 s of the reinforcement interval. In both experiments, presentation of the stimulus produced an immediate decline in running that dissipated over sessions. No increase in running following the stimulus was observed in the first experiment until the stimulus-induced decline dissipated. In the second experiment, increases in running were observed following the tone in the first session as well as when data were averaged over several sessions. In general, the results concur with the assertion that habituation plays a role in the decline in wheel running that occurs within both long and short intervals.  相似文献

Three experiments examined spontaneous recovery from extinction of appetitive conditioned responding (CR) as a function of training history. Rats first received reinforced and non-reinforced conditioning trials. Groups of rats were equated for total number of reinforced trials but differed in the number of non-reinforced trials, or in the order of reinforced and non-reinforced trials. CR was then extinguished in all groups. Subsequently, the extent of recovery of CR was assessed in a test session performed either 1 or 17 days after the last extinction session. After a 17-day delay, rats that had received all reinforced trials immediately prior to the first extinction session showed stronger recovery than did rats having received all reinforced trials at the beginning of training, or interspersed among non-reinforced trials. No significant spontaneous recovery was observed after a 1-day test delay. These results, which may be of clinical relevance with respect to relapse after therapy, are explained in terms of the training schedules generating differences in strength of inhibitory associations, and a relatively long, but not a short, test delay attenuating these associations.  相似文献

In repeated short-term tests (four sessions, each of 45-minute duration), and one longer test (a 23.5-hour session), behavior of rats was evaluated in a long, narrow shuttlebox. One side of the box was exposed to an electric field at various strengths, while a visually identical opposite side was shielded from exposure. In the short-term tests, rats generally remained shielded from electric fields of 90 kV/m and greater during the first session, and maintained this response in subsequent sessions. In the longer test, this same preference response was demonstrated at field strengths of 75 kV/m and greater; however, at 25 and 50 kV/m, rats exhibited a statistically significant preference for the exposed region of the shuttlebox, but only during the light portion of a 12-hour light: 12-hour dark cycle. Exposed animals made more traverses than sham-exposed controls between the two ends of the shuttlebox during the first hour of the test. The experimental data support the hypothesis that the observed behavioral effects are the result of direct interaction of the electric field with the animal, and not the result of secondary factors such as electric shock, corona discharge, audible noise, ozone, or vibration of the experimental apparatus.  相似文献

The possibility that acoustic startle stimuli could support a conditional response (freezing) to contextual stimuli was investigated. Rats were exposed to three acoustic startle stimuli on the first day, and one on the second day. On day 1, 20 rats received naloxone pretreatment and another 20 received saline (placebo) pretreatment. Half of each group received a high-intensity acoustic stimulus, the other half a low-intensity acoustic stimulus. Both the higher stimulus intensity and the naloxone pretreatment led to greater freezing behavior during the 3-minute test period before the single startle stimulus on day 2. These findings support the notion that increased actual or perceived intensity of the acoustic startle stimulus increases conditioning to contextual stimuli as indexed by freezing behavior.  相似文献

The present study investigated whether rats' rates of licking or pressing a lever for 1% liquid sucrose delivered by a continuous reinforcement schedule would decrease (contrast) or increase (induction) when the upcoming period would allow access to 32% sucrose and whether such changes would be influenced by how long each substance was available. In Experiment 1, different groups of rats licked a spout or pressed a lever for 1% sucrose in the first half of the session and, in different conditions, for 1% or 32% sucrose in the second half. Across conditions, halves of the session were 3, 6, 12, or 24 min long. Upcoming 32% sucrose significantly decreased rates of licking at each duration whereas it increased rates of lever pressing except when access duration was 3 min. Experiment 2 replicated Experiment 1 with the exception that rats that licked did so from the same spout in both halves of the session and rats that pressed a lever collected the sucrose reinforcers in the different halves at different locations. In these procedures, upcoming 32% sucrose significantly increased rates of licking. Significant, but small, increases in rates of lever pressing were still observed. The present results suggest that continuous reinforcement or duration of access to sucrose are not primary determinants of whether contrast or induction is observed. Rather, they suggest that the type of behavior (licking versus pressing a lever) and the location at which the substances are collected and consumed play a large role in which effect occurs.  相似文献

Suppression of operant behavior by exposure to pain reactions of conspecifics was examined with pigeons. Three groups of pigeons were trained on a VI schedule, and were then exposed to the pain reactions of an adjoining bird to electric shocks. Although every subject showed suppression of responding, the suppression decreased with repeated exposures. Following this assessment, a conditioning group received conditioned suppression training in which the pain reaction of the adjoining bird was the CS and an electric shock was the US; a shock exposure group received the electric shock without any explicit CS; and, a no-shock group did not receive any shock. After these treatments, every group was exposed to the pain reactions of the adjoining bird (test 1). The conditioning group and the shock exposure group showed clear suppression in responding, but the no-shock group did not.The no-shock group then received the shock exposure treatment and the conditioned suppression training succesively, and the shock exposure group received the conditioned suppression training. Results of tests with the pain reaction of the adjoining bird supported the results of the test 1, however, suppression caused by the shock exposure was not so clear in the no-shock group.The present results demonstrated that conspecific behavior can become a CS by conditioned suppression training, and, the behavior to an aversive stimulus can acquire aversive properties for other conspecifics when they have shared the exposure to the same aversive stimulus.  相似文献
