Mouse interferon mRNA, extracted from NDV (Newcastle disease virus)-induced L-929 cells has been translated with high efficiency in Xenopus laevis oocytes and rabbit reticulocyte lysates. The translational efficiency of a crude RNA extract was 10 640 interferon units/mg RNA/hour for the Xenopus oocytes and 4 012 interferon units/mg RNA/hour for the reticulocyte lysates. The translation product fulfilled the usual criteria for mouse interferon, viz. species specificity and neutralization by specific anti-mouse interferon antiserum. Upon injection of crude interferon mRNA into Xenopus oocytes, interferon activity appeared both in the oocyte homogenates and the oocyte incubation medium. When analyzed by velocity sedimentation in formamidesucrose, the mouse interferon mRNA showed a rather sharp peak halfway between the 4 S and 18 S RNA markers, as could be expected from a mRNA which codes for a 20,000 dalton protein.  相似文献

A substance was isolated from mouse brain cortical tissue that inhibits both cell division and protein synthesis by cells in culture. The inhibitor was released from cerebral cortex tissue by mild protease treatment. A single exposure of cells to as little as 1.25 μg of the isolated material was sufficient to inhibit BHK-21 cell protein synthesis by 20%. Higher concentrations and continual exposure resulted in 87% reduction in protein synthesis. The inhibition was shown to be independent of amino acid uptake and most effective against primary mouse embryo fibroblasts and neonatal mouse brain cell suspensions. Cells previously adapted to culture or transformed cells derived from the nervous system were less affected by, or refractory to, the inhibitor. The substance was shown to be nondialyzable, relatively resistant to thermal inactivation and the inhibitor activity was not removed by chloroform extraction. Two active fractions were identified by Bio-Gel P-100 chromatography and the protein synthetic inhibitor was removed by affinity chromatography with Ulexeuropus agglutinin.  相似文献

We find an endonuclease of high specific activity in a purified mouse interferon preparation. The interferon was purified from Ehrlich ascites tumor cultures which were induced with Newcastle disease virus. It has a higher specific activity (1.5 × 109 NIH mouse reference standard interferon units/mg protein) than reported for any interferon preparation but is not homogeneous. We do not know if the endonuclease activity is due to a contaminating protein or to interferon. The endonuclease does not degrade in our conditions polyuridylic acid or double stranded reovirus RNA and does not inactivate the tRNA2Gln species from E. coli, or tRNAVal species or polysomes from mouse L cells. Endonuclease in as little as 0.5 ng protein of the interferon preparation degrades μg quantities of messenger RNA from mouse L cells, of encephalomyocarditis virus RNA and of in vitro-synthesized reo-virus messenger RNA at 37° in 1 hour. Further characteristics of the endonuclease and its possible relationship (if any) to interferon remain to be established.  相似文献

Aspirin, indomethacin, and phenbutazone at 50 μM concentration inhibit cyclooxygenase in cultured human foreskin fibroblasts as evidence by the suppression of the major prostaglandin species which aaccumulate in the culture medium. In contrast to data reported for mouse interferon on target mouse cells, these agents have no effect on the introduction of antiviral activity by human α and β interferons. Similarly, these agents have no effect on interferon induced inhibition of cell growth in vitro or on interferon induced natural killer cell activity.  相似文献

Fusion of cells of the NS-1 mouse myeloma line with spleen cells from BALBc mice immunized against ovalbumin produced hybrid cells which continuously secrete antibodies specific for ovalbumin. One of these cells was used to establish a cloned line. Studies of its antibody obtained either from ascites fluid or from medium from hybridoma cultures showed high titer and specificity against ovalbumin using the double antibody technique with rabbit anti-mouse immunobeads; the antibody proved to be of the IgG1 (kappa) subclass and type.  相似文献

The carbohydrate moiety of the “antifreeze” glycoprotein from Trematomus borchgrevinki was found to be β-D-galactosyl 1–3 N-acetyl galactosamine by gasliquid chromatography. The glycoprotein inhibited anti-T antibody from human serum and Arachis hypogoea lectin, but was inactive against Vicia graminea. Native “antifreeze” glycoprotein did not inhibit the agglutinins from Helix pomatia or Cepaea hortensis, although after Smith degradation showed a strong inhibition towards them. Inhibition of the latter agglutinin demonstrates the carbohydrate-protein linkage to be α-linked. The presence of the Thomsen-Friedenreich antigen (T-antigen) on the “antifreeze” glycoprotein and its relation to tumour cell surfaces is briefly discussed.  相似文献

Glycopeptides from brain inhibit rates of polypeptide chain elongation   总被引:3,自引:0,他引:3  
In previous reports, we have identified cell-surface glycopeptides from mouse cerebrum (BCSG) that inhibited protein synthesis and mitosis in several cell types. When baby hamster kidney (BHK)-21 cells were infected with vesicular stomatitis virus (a negative strand RNA virus), BCSG extensively inhibited viral protein synthesis. This inhibition was effective against both protein and glycoprotein synthesis and was independent of amino acid uptake by infected cells, synthesis of viral RNA, and degradation of viral proteins. Analysis of polyribosome profiles in uninfected BHK-21 cells indicated that the degree of cellular protein synthesis inhibition could not be attributed to activation of RNase or solely to a disruption of chain initiation. When added directly to a cell-free protein-synthesizing system derived from BHK-21 cells, BCSG was ineffective, but if the inhibitory material was first allowed to react with cells, cell-free protein synthesis was substantially reduced. When BCSG were reacted with cells for 5 min at 0 degrees C, the cells tested, BHK-21 (a BCSG-sensitive line) and murine fibrosarcoma 2237 (a BCSG-insensitive line), both effectively adsorbed the inhibitor from the medium.  相似文献

Mouse L cell interferon (IFN αβ) inhibited the differentiation of mouse 3T3-Li fibroblasts into adipocytes. IFN αβ also inhibited hexose monophosphate shunt (HMP) activity in these cells. HMP activity is required for the reducing power necessary for the conversion of 3T3-Li fibroblasts to adipocytes. Both IFN blockage of differentiation and HMP activity were reversed by the reducing agent 2-mercaptoethanol, probably through interaction with membrane receptors and not through direct inactivation of IFN. Several non-antiviral effects of IFN αβ on cellular function, including differentiation and immunoregulation, may be mediated at the biochemical level through blockage of HMP activity.  相似文献

A lysate of Staphylococcus aureus (S. aureus) induced high-titered (103.6 ~ 104.6 units/ml) mouse immune interferon (MuIFN-γ) in spleen cell cultures. mRNA was extracted from such cells, purified by oligo(dT) cellulose chromatography and sucrose gradient centrifugation, and injected in Xenopus laevis oocytes. Fractions containing RNA of 15 to 16 S gave rise to IFN activity, that was characterized as IFN-γ by virtue of its acid lability, neutralization by antiserum to partially purified MuIFN-γ, and lack of neutralization by antiserum to NDV-induced L-cell IFN.  相似文献

Rat glioma C6 cell polyribosomal preparations were tested in a heterologous in vitro system for their ability to direct the synthesis of nerve growth factor related polypeptides. Two major polypeptides of MW ~ 21,000 and ~ 43,000 respectively were found, both of which were immunoprecipitable with specific anti-mouse 2.5S nerve growth factor serum. After incubation of in vitro synthesized proteins with submaxillary gland extract the bulk of these protein species was converted into immunoprecipitable material of MW ~ 13,000, which comigrated in sodium dodecyl sulfate/polyacrylamide gel electrophoresis with mouse 2.5S nerve growth factor.  相似文献

A fraction enriched in plasma membranes from porcine polymorphonuclear leucocytes, isolated by sucrose density centrifugation was shown to possess considerable AMP hydrolysing activity (150 nmol/min per mg protein). However all of this activity could be inhibited using excess p-nitrophenyl phosphate in the incubation medium. Furthermore the hydrolysis of AMP by the membrane was unaffected by the 5′-nucleotidase inhibitor α,β-methyleneadenosine diphosphate and by the lectin concanavalin A, another potent inhibitor of 5′-nucleotidase. An antibody against mouse liver 5′-nucleotidase also did not inhibit the activity. These results suggest that the hydrolysis of AMP by porcine polymorph membranes is not accomplished by a specific 5′-nucleotidase and the necessity for distinguishing between true 5′-nucleotidase and non-specific phosphatase activity is discussed.  相似文献

Glycopeptides isolated from mouse cerebral cortex cell surfaces (BCSG) were shown to inhibit cell growth and protein synthesis in baby hamster kidney (BHK)-21 cells, whereas polyoma virus-transformed BHK-21 cells (pyBHK-21) were refractory to the inhibitory activity of the glycopeptides. Growth inhibition was shown to be reversible and non-lethal to BHK-21 cells. Despite that difference in sensitivity to the action of the glycopeptides, both cell lines could bind the inhibitor in a saturable fashion and in similar quantities. After trypsinization, BHK-21 cells appeared refractory to the inhibitor, whereas pyBHK-21 cells became sensitive. The data suggested the presence of a receptor for BCSG on the cell surface of both cell lines. Incubating BCSG with conditioned medium from pyBHK-21 cells resulted in loss of the glycopeptide's inhibitory activity. In contrast, medium conditioned by BHK-21 cells had no effect on the inhibitory activity of BCSG. We hypothesize that the refractoriness of pyBHK-21 cells to BCSG is related to their autonomous growth characteristics and failure to respond to topo-inhibitory growth control. BCSG may be a naturally occurring growth regulator whose function can be explored by use of the BHK-21/ pyBHK-21 model system.  相似文献

Proteins of gibbon ape lymphosarcoma virus (GaLV) and woolly monkey sarcoma virus, type 1, together with its associated virus (SSV-1SSAV-1) were analyzed by guanidine-agarose chromatography and the separation patterns were compared with those of mouse and feline type C viruses. GaLV contained five major proteins, including two glycoproteins, whereas lower mammalian viruses contained six major proteins, including two glycoproteins. The molecular weights of the five GaLV proteins closely resembled the molecular weights of the five equivalent lower mammalian viral proteins. SSV-1SSAV-1 showed a separation pattern similar to GaLV except it contained a low but detectable amount of an additional glycoprotein. Both GaLV and SSV-1SSAV-1 were deficient in a protein of molecular weight about 15,000 daltons which is found in all known type C viruses of avian, reptilian and lower mammalian species.  相似文献

A low-molecular-weight interferon-mediated ribosome-associated inhibitor of reovirus mRNA translation was purified from the 0.5 M KCl ribosomal salt-wash fraction of mouse L929 cells. The inhibitor possessed nucleolytic activity with reovirus [3H]mRNA as a substrate. Loss of translational inhibitory activity correlated with the thermal inactivation of the nuclease. A low-molecular-weight (<10K) component present in the Bio-Gel P150 chromatography fractions which contained the interferon-mediated nucleolytic activity was labeled in vivo with [14C]valine; a smaller component present in the same fractions was phosphorylated in vitro with [γ-32P]ATP. The <10K components were resolved from ~50K, ~30K and ~20K phosphorylatable proteins associated with ribosomes that possess the interferon-mediated inhibitor(s) of viral mRNA translation.  相似文献

Inositol hexaphosphate, and other polyphosphates, inhibit diphtheria toxin-mediated cytotoxicity by binding to the toxin at a highly cationic site called the P site and preventing toxin binding to cell surface receptors. The binding of diphtheria toxin to a solubilized cell surface glycoprotein (150,000 daltons) is also inhibited by these polyphosphates. Treatment of this 150,000 dalton diphtheria toxin-binding cell surface glycoprotein with papain yielded an 88,000 dalton and a 74,000 dalton diphtheria toxin-binding glycoprotein whose binding to toxin was no longer inhibited by inositol hexaphosphate. This result suggests a model of diphtheria toxin-receptor interaction in which the toxin receptor possesses one binding site which interacts with the P site of the toxin in a polyphosphate-sensitive fashion, and another binding site (located within the papain-derived 74,000–88,000 dalton glycoproteins) which can interact with the toxin at a site distinct from the P site (the X site) in a polyphosphate-insensitive fashion. This X site-receptor interaction may be involved in the binding of CRM proteins that bind to the toxin receptor but that do not bind polyphosphates, or it may be involved in the entry process of the toxin.  相似文献

We have investigated the interaction of three lectins, differing in their sugar specificities, with the surface of the three differentiation stages of Trypanosoma cruzi. The Scatchard constants for each lectin and parasite stage imply that differentiation of T. cruzi is accompanied by changes in the cell surface saccharides. Trypomastigotes obtained from two different sources do not differ appreciably as to the number and affinity of binding sites for the three lectins employed, suggesting a similar cell-surface saccharide composition. These conclusions are reinforced by sodium dodecyl sulfate-polyacrylamide gel electrophoretic analysis of the 131I-labeled surface glycoproteins, following isolation by affinity chromatography. The surface membrane of trypomastigotes, the infective stage to T. cruzi for mammalian cells, possesses a specific glycoprotein of apparent Mr 85 000 (Tc-85) which is absent from the other two stages and can be isolated by affinity chromatography on wheat germ agglutinin-Sepharose columns. This glycoprotein also binds to concanavalin A, but not to Lens culinaris lectin. The binding of Tc-85 to wheat germ agglutinin is unnafected by treatment of either the isolated glycoprotein or intact living trypomastigotes with neuraminidase. Since N-acetyl-d-glucosamine inhibits internalization of trypomastigotes by cultured mammalian cells, it is suggested that Tc-85 might be involved in adhesion and/or interiorization of T. cruzi into mammalian cells, possibly via recognition of an ubiquitous host-cell surface N-acetyl-d-glucosamine-specific receptor activity.  相似文献

Human diploid fibroblast (FS-4) cells were induced to produce interferon mRNA by exposure to poly(rI)·poly(rC) plus cycloheximide. The intracellular location of interferon mRNA was investigated by differential centrifugation of the cytoplasm into a membrane (pellet) and a free (supernatant) fraction, followed by injection of mRNA isolated from either fraction into X.laevis oocytes. When translation in FS-4 cells was prevented, most (85–90%) of the interferon mRNA activity was found in the free fraction. However, when translation was permitted, most (80–95%) of the interferon mRNA activity was found in the membrane fraction. These results are consistent with the predictions of the “signal hypothesis” (Blobel and Dobberstein, J. Cell Biol. 1975, 67:835) for secretory proteins.  相似文献

Trimethyloxonium ion inactivates acetylcholinesterase from the electric eel and acetylcholinesterase on the surface of human red blood cells. Tetramethylammonium ion, which is a competitive inhibitor of acetylcholinesterase, protects against this inactivation. Trimethyloxonium ion does not inactivate the system that transports choline into the red blood cell. We conclude that trimethyloxonium ion is an affinity-labeling reagent for acetylcholinesterase and that red blood cell acetylcholinesterase is probably not a component of the choline transport system.  相似文献

Ribavirin (1-β-D-ribofuranosyl-1,2,4-triazole-3-carboxamide) is a broad spectrum antiviral substance active against a wide range of both DNA and RNA viruses. It is, however, virtually inactive against polio virus. Its pharmacological mechanism of action was obscure. A possible common target for a chemotherapeutic agent in both DNA and RNA viruses is the “capping” reaction of mRNAs which interalia involves the formation of a guanine pyrophosphate structure at the 5′ terminus by mRNA guanylyl transferase. We have observed that Ribavirin triphosphate is a potent competitive inhibitor of the capping guanylation of viral mRNA. This finding could account for the antiviral potency of the drug against both DNA and RNA viruses and its ineffectiveness against a virus in which the mRNAs derived from them are not capped.  相似文献

Cell surface antigens of normal and anemic (WW) mouse erythroid cells have been examined in cytotoxicity assays with two rat antisera. When tested on fetal liver cells, a rat anti-erythroblast serum recognized antigen(s) present on erythroid cells early in development, while rat anti-adult red blood cell serum recognized antigen(s) present on mature erythroid cells. Each of these sera had different activity on normal (+/+ or W+) as compared to anemic (WW) erythroid cells.  相似文献
