Phaseolus vulgaris cv. Glamis plants grown at 7 and 28 W m–228 W m–2 in controlled environment cabinets showed copiousnodulation and high levels of acetylene reducing activity. Earlydifferences in nodulation were apparent before differences inphotosynthesis and were attributed to an effect of far-red lighton nodule development. Total plant nitrogen content was greater at 28 W m–2 thanat 7 W –2 but nitrogen content as a percentage of d. wtwas greater at the lower irradiance level. Total acetylene reducing activity (nmol. min–1 root–1)was greater at 28 W m–2 than at 7 W –2, but therewas no irradiance effect on specific activity (nmol. min–1g d. wt of pink nodules–1 or nmol. min–1 pink nodule–1). Transfer of 40-day-old plants from 7 W m–2 to 28 W m–2resulted in increased nodulesize(due toincreased size of infectedcells), accompanied by increased total, but not specific, acetylenereducing activity. Transfer of plants from 28 W m–2 to 7 W m–2 resultedin a fall of total acetylene reducing activity within 24 h,and senescence of large nodules. Specific acetylene reducingactivity was unaffected The results are interpreted as an effect of light on the productionof nitrogen fixing tissue, rather than on nitrogenase activity.  相似文献

Plants of Phaseolus vulgaris grown at 7 and 28 W m–2 showedno differences in rate of development of leaves or flowers.At 7 W m-Z plants had longer internodes, more succulent stemsand leaves, higher ratios of shoot:root and greater leaf areasthat those at 28 W m–2. These differences were establishedprior to detectable differences in photosynthesis and couldpartly be attributed to an increased proportion of far-red light. Although the final d. wt, carbon content, and fruit yield werehigher at 28 W m–2, plants at 7 W m–2 apparentlyhad similar relative growth rates and greater photosyntheticefficiency. Dry weight differences are most easily interpretedas resulting from the establishment of an earlier net carbongain at 28 W m–2 than at 7 W m–2.  相似文献

In the preceding article[Am. J. Physiol. 274 (Cell Physiol. 43):C1158-C1173, 1998], we describe the development of a kinetic model for the interaction of mitochondrial Ca2+ handlingand electrical activity in the pancreatic -cell. Here we describefurther results of those simulations, focusing on mitochondrialvariables, the rate of respiration, and fluxes of metabolicintermediates as a function of D-glucose concentration. Oursimulations predict relatively smooth increases of O2consumption, adenine nucleotide transport, oxidative phosphorylation,and ATP production by the tricarboxylic acid cycle asD-glucose concentrations are increased from basal to 20 mM.On the other hand, we find that the active fraction of pyruvatedehydrogenase saturates, due to increases in matrix Ca2+,near the onset of bursting electrical activity and that the NADH/NAD+ ratio in the mitochondria increases by roughly anorder of magnitude as glucose concentrations are increased. Themitochondrial ATP/ADP ratio increases by factor of <2 between theD-glucose threshold for bursting and continuousspiking. According to our simulations, relatively small changes inmitochondrial membrane potential (~1 mV) caused by uptake ofCa2+ are sufficient to alter the cytoplasmic ATP/ADP ratioand influence ATP-sensitive K+ channels in the plasmamembrane. In the simulations, these cyclic changes in the mitochondrialmembrane potential are due to synchronization of futile cycle ofCa2+ from the cytoplasm through mitochondria viaCa2+ uniporters and Na+/Ca2+exchange. Our simulations predict steady mitochondrial Ca2+concentrations on the order of 0.1 µM at low glucose concentrations that become oscillatory with an amplitude on the order of 0.5 µMduring bursting. Abrupt increases in mitochondrial Ca2+concentration >5 µM may occur during continuous electricalactivity.

The present study deals with structure and functioning of threeareas of Himalayan oak forest. Low- and mid-altitude oaks, namelyQuercus leucotrichophora, and Quercus floribunda, form predominantevergreen forests in Central and Western Himalaya. The totaltree basal cover ranged between 33·89 m2 ha–1 (Q.floribunda site) to 36·83 m2 ha–1 (Q. leucotrichophorasite). The density ranged between 570 and 760 individuals ha–1.Allometric equations relating biomass of different tree componentsto GBH (girth at breast height) were significant with the exceptionof leaf biomass in Q. leucotrichophora and Rhododendron arboreum.Total vegetation biomass (29·40–467·0 tha–1) was distributed as 377·1 t ha–1 intrees, 5·40 t ha–1 in shrubs and 1·23 tha–1 in herbs. Total forest floor biomass ranged between4·6 and 6·2 t ha–1. Of the total annuallitter fall (4·7–4·8 t ha–1), 77·5% was contributed by leaf litter, 17·8 % by wood litterand 4·7 % by miscellaneous litter. Turnover rate of treelitter varied from 0·66 to 0·70. Net primary productionof total vegetation ranged between 15·9 and 20·6t ha–1 yr–1, of which the contribution of trees,shrubs and herbs was 81·2 %, 8·6 % and 10·2%, respectively. A compartment model of dry matter on the basisof mean data across sites was developed to show dry matter storageand flow of dry matter within the system. Guttation was used as a non-destructive way to study the flowof water and mineral ions from the roots and compared with parallelmeasurements of root exudation. Guttation of the leaves of barley seedlings depends on age andon the culture solution. Best rates of guttation were obtainedwith the primary leaves of 6- to 7-day-old seedlings grown onfull mineral nutrient solution. The growing leaf tissue becomessaturated with K+ below 1.5 mM K+ in the medium, whereas K+concentration in the guttated fluid still increases furtheras K+ concentration in the medium is raised. At 3 mM K+ averagevalues of guttation were 1.4–2.4 mm3 h–1 per plantwith a K+ concentration of 10–20 mM; for exuding plantsthe flow was 4.2–7.6 mm3 h–1 per plant and K+ concentration35–55 mM. Abscisic acid (ABA) at 10–6 to 10–4 M 0–2h after addition to the root medium increased volume flow ofguttation and exudation and the amount of K+ exported. Threeh after addition of ABA both volume and amount of K+ were reduced.There was an ABA-dependent increase in water permeability (Lp)of exuding roots shortly after ABA addition. Later Lp was decreasedby 35 per cent and salt export by 60 per cent suggesting aneffect of ABA on salt transport to the xylem apart from itseffect on Lp. Benzyladenine (5 x 10–8 to 10–5 M)and kinetin (5 x 10–6 M) progressively reduced volumeflow and K+ export in guttation and exudation and reduced Lp. Guttation showed a qualitatively similar response to phytohormonesas found here and elsewhere using exuding roots. The aim of this work was to discover if the respiratory metabolismof the roots of Plantago lanceolata L. differed from that ofthe roots of Plantago major L. Measurements of oxygen uptakeand dry weight of excised root systems during growth of seedlingsprovided evidence that the two species differed in the amountof respiration needed to support a given increase in dry weight.Excised root systems were given a 6-h pulse in [U-14C]sucrosefollowed by a 16.5-h chase in sucrose. The detailed distributionof 14C amongst the major components of the roots at the endof the pulse and the chase revealed no significant differencebetween the two species. Patterns of 14CO2 production from [1-14C],[2-14C], [3,4-14C], and [6-14C]glucose of excised root systemsfrom plants of three ages were similar for the two species.It is suggested that there is no conclusive evidence for anysignificant inherent difference in the respiratory metabolismof the roots of the two species. The uptake of potassium, calcium, and magnesium ions by maizeand the interrelationships among the cations have been investigatedat 48 K: Ca: Mg ratios in culture solutions. Calcium was foundto stimulate K+ and Mg++ uptake at certain cation ratios butinhibit it at others. Potassium did the same for Ca++ uptake,and Mg++ for Ca++ and K+. The uptake of Mg++ was generally enhancedby K+. The sum of the cations in the plants expressed in meqwas fairly constant for treatments of the same K+ concentrationat the low to moderate levels of K+, but at considerably higher(> 24 meq l–1) K+ levels the constancy was not dependenton K+ concentration. Potassium depressed, but Mg++ stimulatedphosphorus accumulation. Calcium stimulated phosphate absorptionat certain cation ratios but had no effect at others. The plantyield increased with increasing K+ up to 24 meq l–1 ofK+ after which the yield tended to fall with further increasein K+. The yield was also increased by Ca++. Magnesium increasedthe yield at certain cation ratios and either depressed it orwas without effect at others.  相似文献

Membrane-bound ATPase associated with plasma membrane and solubleATPase associated with the cytoplasm were prepared from shootsof Salicornia pacifica var. utahensis by sucrose density gradientcentrifugation. The isolated ATPases were tolerant to high concentrationsof NaCl. The Km for membrane-bound ATPase was 1.75 mM and forsoluble ATPase, it was 1.4 mM. The relative effectiveness ofdivalent cations for stimulation of membrane-bound ATPase wasMg2+>Fe2+>Mn2+>Co2+>Cu2+. Soluble ATPase activitywas stimulated by Ba2+>Ca2+>Mg2+ and was inhibited byCu2+, Zn2+, Co2+ and Fe2+. The compounds N,N1-dicyclohexylcarbodiimide,NaF and ADP, did inhibit the ATPases but ouabain, triphenyltinhydroxide, sodium azide, indoleacetic acid and abscisic aciddid not inhibit the ATPases from s. pacifica var. utahensis. 1 Present address: Department of Biology, Kong-Ju National College,Kong-Ju, Korea. Northern red oak (Quercus rubra L.) seedlings were grown for63 d in a complete nutrient solution (pH 3.8) containing oneof three concentrations of Al (0, 0.75 or 2-0 mol m–3)and either 10 or 250 mmol m–3 Ca. Of all solution variables,the In of (Al3+)/(Ca2+), the solution activities ratio, wasmost closely correlated with declines in shoot and root growth.Ln (Al3+)/(Ca2+) also most closely predicted leaf and root [Mg],[Al], and [Al]/[Ca]. These three variables in turn were closelyrelated to growth. Toxic levels of (Al3+) and (Al3+)/(Ca2+)in solution are compared to levels in forest soils. We have used chlorotetracycline (CTC) and fluphenazine (FPZ)as fluorescent probes to visualize the distributional patternsof membrane calcium (mCa2+) and the Ca2+-receptor protein calmodulin(CaM) in various cell types of unfixed living isolated and unisolatedembryo sacs of Petunia hydrida L. and Nicotiana tabacum L. Ourresults indicate that in the young embryo sacs of Petunia, bothsynergids and the central cell sequester relatively higher amountsof mCa2+ and CaM than the egg cell and the antipodals. Much of the mCa2+ in the synergids is polarized in its distributionin that the mCa2+ is higher towards the micropylar end of thesynergids. Interestingly, in the mature embryo sacs of Petuniaonly one of the two synergids and the egg cell proper manifesta higher level of mCa2+. In vivo only one of the synergids inthe young as well as in the mature embryo sacs if Nicotianaconsistently show higher mCa2+. In the mature embryo sacs of Petunia the level of CaM is almostuniform in all the cell types except that one of the synergidsand the three antipodal cells show a slightly higher level ofCaM. The marine diatoms Lauderia annulata and Synedra planctonicaharvested during exponential growth were exposed to differentdoses of u.v.-B (286, 439 and 710 J m–2 d–1) for2 days. Uptake of 15N-nitrate was estimated before, during andafter u.v.-B radiation over 2 days. Exposure to high levelsof u.v.-B (710 J m–2 d–1) caused irreversible damageat the second daily irradiance. Lauderia cells were less affectedby u.v.-B stress than Synedra cells. 15N-nitrate uptake wasreduced under u.v.-B irradiance but could be reactivated within1 day following exposure to a low dose (286 J m–2 d–1).Higher levels of u.v.-B radiation (710 J m–2 d–1)led to irreversible damage. The pattern of 15N-incorporationinto several amino acids of Lauderia varied after 2 days ofu.v.-B radiation. 15N enrichment of glutamine increased markedlyafter u.v.-B stress (717 J m–2 d–1) whereas I5Nexcess of aspartic acid was significantly reduced. The distribution of 65Zn in zinc-tolerant and copper-tolerantplants of Agrostis spp. from toxic mine-tailings in Enflandand Wales was compared with zinc distribution in non-tolerantplants. Isotope was applied in culture solution in which theplants were growing. No differences could be demonstrated betweenthe plants were growing. No differences could be demonstratedbetween the plants by whole-plant radioautography, or by zincanalyses of the tops. Root/shoot ratios calculated from specificactivity values varied with population, the non-tolerant plantshaving the lowest and the zinc-tolerant plants the highest ratio.After solvent (80 per cent ethanol and water) extractions, theroot residue of zinc-tolerant plants contained a higher percentageof 65Zn than that of non-tolerant plants. Chemical fractionationof the roots revealed that the main high difference was thatthe amount of 65Zn in the pectate extract of the cell wall washigh in zinc-tolerant plants and low in non-tolerant plants.The 65Zn distribution in the copper-tolerant plants was similarto that in the non-tolerant plants, indicating that the tolerancemechanisms for the elements are different. Soluble protein andRNA preparations were made but they contained low levels of65Zn. An exception was the relatively high value for RNA fromzinc-tolerant A. stolonifera shoots. An anionic complex of 65Znin the soluble fraction was investigated. This complex accountedfor most of the radioactivity in A. tennis extracts of shootsbut the concentration of the complex was low in A. stoloniferashoots, and in root extracts of all plants examined.  相似文献

Cauliflower (Brassica oleracea L. botrytis) and broccoli (Brassicaoleracea L. italica) plants were grown in large pots in growthchambers for a range of temperatures (mean air temperaturesfrom 7.0-25.3 C) and irradi-ances (from 9.3-50.8 mol m–2d–1 or 4.7-25.4 MJ m–2 d–1). The extinctioncoefficient for PAR decreased with plant size reaching a valueof 0.55 in cauliflower and 0.45 in broccoli at plant leaf areasof 0.235 m2 and 0.227 m2, respectively. The leaf area expansionrate was unaffected by irradiance when compared at identicalleaf surface temperatures. The response of expansion rate tosurface temperature was fitted to a broken stick model witha base temperature of –0.7C and an optimum temperatureof 21.0C. The radiation conversion coefficient increased withair temperature below 13.8C and remained constant above this.The estimated radiation conversion coefficient above 13.8Cand for a PPFD of 20 mol m–2 d–1 was 0.77 g mol–1in cauliflower and 0.87 g mol–1 in broccoli. The radiationconversion coefficient declined with increasing irradiance levelfrom a maximum of 1.89 g mol–1 at near nil irradiancein cauliflower. Growth and ion accumulation were measured in callus culturesof Cicer arietinum L. cv. BG-203, grown on media supplementedwith 0–200 mol m–3 NaCl. Fresh and dry weights decreasedat concentrations ranging from 100–200 mol m–3,the reduction being greater during the third and fourth weeksof culture. Slight stimulation of growth was observed at 25and 50 mol m–3 NaCl. There was also a decrease in tissuewater content (fresh weight: dry weight) at 100–200 molm–3 NaCl. The concentration of Na+ and Cl in thetissue increased with increasing salinity of the medium. Mostof the accumulation of these ions occurred by the first weekwhile significant growth inhibition became apparent by onlythe third week of culture. Tissue K+ and Mg2+ decreased withincreasing salinization, the decrease being greater in K+ levels.Levels of Ca2+, however, were maintained throughout the experimentalrange. Ion contents and concentrations (K+, Na+, Cl, Ca2+, Mg2+,SO2–4, NO3, HPOJ2–4, amino and organic acids)of P. purpurea have been studied in relation to salinity variation.Cells were shown to accumulate large amounts of K+ and Clagainst their respective gradients of electrochemical potentialin all dilute and concentrated seawater media. Active influxof SOJ2–4, NO3, and HPOJ2–4 is also suggested,while Na+ is actively excluded from cells under hyposaline andhypersaline conditions. The relative proportions of individualcomponents of the internal osmotic potential were found to changeaccording to the external salt concentration. KCL forms themajor fraction of j} in concentrated seawater media while K+-aminoacids form the major fraction in dilute seawaters. Other intracellularsolutes comprise less than 15% oftj, in all media. Unidirectional fluxes of K+ and Cl were studied by radioisotopicmeans. Fluxes of K+ and Cl are reduced in hyposalinemedia, as is absolute KCL content per cell. Intracelhilar KCLcontent was also found to be markedly dependent upon externalK+ concentration, rather than water potential. Changes in KC1levels induced by salinity variation occur over a 6 h period.  相似文献

Total uptake of 3H-thymidine, 3H-thymine, and 3H-deoxyuridineand incorporation of these substances into DNA have been investigatedin excised roots in Vicia faba. Total uptake was found to behigher in excised roots than in intact ones. This was a consequenceof exogenous 3H-DNA precursors entering the excised roots throughthe cut surface. Where the cut surface was not immersed in the3H-thymidine solution 3H uptake was also higher than in intactroots. In this case 3H uptake scemed to be correlated with water-lossfrom the cut surface. Incorporation of 3H-thymidine, 3H-thymine,and 3H-deoxyuridine into DNA was found to be higher in excisedroots than in intact ones, probably as a result of a decreasein the size of the relevant endogenous precursor pools. It issuggested that this decrease resulted from the cessation ofthe supply of DNA precursors to these roots from the cotyledons.  相似文献

The extent to which Spartina alterniflora Loisel. excluded,secreted or accumulated the major seawater ions (Cl-, SO2-4,Na+, K+, Mg2+, and Ca2+) was investigated under varying salinitytreatments. From a quantitative viewpoint, ion exclusion wasmost prominent and accounted for 91–97% of the theoreticalmaximum ion uptake as a result of transpiration and growth.Of those ions taken up, approximately half was secreted fromthe shoots. Relative to K+, a disproportionate amount of Na+was excluded at the roots and secreted by the shoots. The concentrationwithin the tissues of S. alterniflora did not change with salinitytreatment for the majority of the ions examined, but Na+ wasmore than twice as concentrated at 40 g dm-3 than at lOgdm-3.Calculations of the flux of ions from salt marsh sediments tothe flood water via shoot secretion or stem/leaf turnover indicatethat these processes may be important to the ecology of S. alternifloraas mechanisms that limit the accumulation of salt within theroot zone.  相似文献

The present study deals with structure and function of fourareas of Himalayan chir pine forest. Tree layer was monospecificon all sites with varied density and basal cover in the rangeof 540–1630 individuals per ha and 25·0–47·2m2ha–1, respectively. Shrubs having low density were sparselydistributed. All allometric equations relating to biomass ofdifferent components, to circumference at breast height (cbh)were significant, with the exception of that for cone biomass.Total vegetation biomass (115–236 t ha–1) was distributedas 113–283 t ha–1 in trees. 0·56–0·82t ha–1 in shrubs and 1·63–2·57 t ha–1in herbs. Total forest floor biomass including herbaceous litterranged between 9·6 and 13·6 t ha–1. Of thetotal annual litter fall (4·26–7·38 t ha–1),60·3–75·1 per cent was distributed in leaflitter and 24·9–39·7 per cent in wood litter.Turnover rate of tree litter varied from 0·45 to 0·53,whereas rates for shrubs and herbs were assumed to 1. Net primaryproduction of total vegetation ranged between 9·91 and21·2 t ha–1 year–1, of which the contributionof trees, shrubs and herbs was 76·5– 88·1per cent 0·6–1·8 per cent and 11·3–21·5per cent, respectively. A compartment model of dry matter onthe basis of mean data across sites was developed to show drymatter storage and flow of dry matter within the ecosystem. 以江西铅山红芽芋脱毒苗为试材,研究不同因素对红芽芋脱毒苗球茎愈伤组织诱导及其再生体系的影响,以期对红芽芋脱毒苗的再生体系进行优化。结果表明,红芽芋脱毒苗球茎愈伤组织诱导的最佳培养基是MS+TDZ 2 mg·L-1+2,4-D 1 mg·L-1。红芽芋脱毒苗球茎愈伤组织分化的最佳培养基是MS+TDZ 2 mg·L-1+NAA 1 mg·L-1。红芽芋脱毒苗不定芽生根的最佳培养基是1/2MS+NAA 0.5 mg·L-1+PP333 0.5 mg·L-1。红芽芋再生苗最好的移栽基质为发酵后的腐锯木屑。红芽芋脱毒苗球茎愈伤组织再生苗移栽时最佳的PP333浓度为20~50 mg·L-1。本试验成功建立了红芽芋脱毒苗球茎愈伤组织的再生体系,为红芽芋脱毒苗转基因的研究和种质创新奠定了基础。  相似文献

This study investigated the relationship between seasonal changesin ambient UV-R, and sunscreen concentrations in phytoplanktonand krill. Concentrations of mycosprine-like amino acid (MAA)sunscreens were quantified in phytoplankton communities andin the krill Nyctiphanes australis over a 1-year period offthe Otago Coast, New Zealand. Ambient UV-B and UV-A ranged froma minimum mean daily dose of 2.19 x 104 kJ day–1 and 0.73x 106 kJ day–1 in June, to a maximum in January of 20.19x 104 kJ day–1 and 4.88 x 106 kJ day–1, respectively.Concentrations of MAAs (consisting almost entirely of Mycosporine-glycine)in the phytoplankton community were lowest in August (5.6 nmolµg–1 Chl) when UV-R irradiances were minimal andhighest in January (41.4 nmol µg–1 Chl) when UV-Rirradiances were maximal. Nyctiphanes australis was found tocontain five identified MAAs (mycosporine-glycine, shinorine,Porphyra-334, palythine and palythinol) and several unknownUV-R absorbing compounds. Concentrations ranged from 4.73 to15.51 nmol mg–1 dw, with little indication of a seasonalcycle that could be correlated with changes in either phytoplanktonMAA concentrations or ambient UV-R irradiances. The findingssuggest that krill are neither accumulating MAAs in responseto changes in MAA concentrations in their phytoplankton food,or that MAA concentrations in krill are increased in responseto higher ambient UV-R irradiances. Concentrations of MAAs inkrill body parts (carapace, legs, eyes, antennae, muscle) weresimilar (4.89–5.98 nmol mg–1 dw), with the exceptionof the carapace (2.03 nmol mg–1 dw).  相似文献
