Three ectomycorrhizal (ECM) isolates of Rhizopogon luteolus, R. roseolus and Scleroderma citrinum were found to differ markedly in their in vitro tolerance to adverse conditions limiting fungal growth, i.e. water availability, pH and heavy metal pollution. S. citrinum was the most sensitive, R. luteolus intermediate and R. roseolus the most tolerant species. Pinus radiata D. Don seedlings were inoculated in the laboratory and in a containerised seedling nursery with spore suspensions of the three ECM species. Colonisation percentage was considerably lower under nursery conditions, probably due to competition by native fungi. The effects of nursery ECM inoculation on seedling growth depended on the fungal species. Only R. roseolus-colonised plants showed a significantly higher shoot growth than non-mycorrhizal plants. All three fungi induced significantly higher root dry weights relative to control plants. Despite the low mycorrhizal colonisation, mycorrhization with all three species improved the physiological status of nursery-grown seedlings, e.g. enhanced root enzyme activity, shoot nutrient and pigment content, net photosynthesis rate and water use efficiency. Of the three fungal species, R. roseolus was the most effective; this species was also the most adaptable and showed the greatest range of tolerance to adverse environmental conditions in pure culture. It is, therefore, proposed as a promising fungal species for ECM inoculation of P. radiata in the nursery.  相似文献

Ectomycorrhizal (ECM) syntheses between four ECM fungi, Laccaria amethystina, Hebeloma mesophaeum, Thelephora terrestris, and Tomentella sp., and Populus maximowiczii seedlings that are known to form ECM at a denuded area of Mt. Usu were performed in volcanic debris in a controlled growth chamber. The percentage of ECM colonization and seedling growth were determined 3 months after inoculation. Seedlings were successfully colonized by the inoculated ECM fungi with low contamination ratios. Seedling height and biomass were larger in the inoculated seedlings than in the control, although the effects of inoculation on seedling growth varied with the ECM fungus.  相似文献

The survival, development and mycorrhizal efficiency of a selected strain of Laccaria bicolor along with naturally occurring ectomycorrhizal fungi in a young plantation of Douglas fir was examined. Symbionts were identified and their respective colonization abilities were determined. Eight species of symbiotic fungi, which may have originated in adjacent coniferous forests, were observed on the root systems. Mycorrhizal diversity differed between inoculated (5 taxa) and control (8 taxa) seedlings. Ectomycorrhizal fungi which occurred naturally in the nursery on control seedlings (Thelephora terrestris and Suillus sp.) did not survive after outplanting. Both inoculated and naturally occurring Laccaria species, as well as Cenococcum geophilum, survived on the old roots and colonized the newly formed roots, limiting the colonization by other naturally occurring fungi. Other fungi, such as Paxillus involutus, Scleroderma citrinum and Hebeloma sp. preferentially colonized the old roots near the seedling's collar. Russulaceae were found mainly in the middle section of the root system. Mycorrhizal colonization by Laccaria species on inoculated seedlings (54%) was significantly greater than on controls (13%) which were consequently dominated by the native fungi. Significant differences (up to 239%) were found in the growth of inoculated seedlings, especially in root and shoot weight, which developed mainly during the second year after outplanting. Seedling growth varied with the species of mycorrhizae and with the degree of root colonization. Competitiveness and effectiveness of the introduced strain on improving growth performances of seedlings are discussed.  相似文献

Summary A greenhouse study was conducted to evaluate the effect of ectomycorrhizae on loblolly pine (Pinus taeda L.) growing in a Piedmont soil. Pine seedlings were inoculated with one of four species of fungi (Scleroderma aurantium, Pisolithus tinctorius, Thelophora terrestris, andRhizopogon roseolus). The seedlings were grown in pots containing a Cecil sandy clay loam amended to create a gradient of extractable P ranging from 5.9 to 52.5 g/g. After ten months, all colonized seedlings were significantly larger than control seedlings. However, of the four fungi,Scleroderma aurantium mediated a far superior shoot growth response to increasing levels of soil P; the seedlings were significantly larger than those colonized by any other fungus and also had the largest root systems and greatest degree of mycorrhizal colonization.  相似文献

Container-grown Pseudotsuga menziesii and Pinus pinaster seedlings were inoculated with water suspensions of spores of five ectomycorrhizal fungi commonly found in northeastern Spain. Pseudotsuga menziesii seedlings were inoculated with basidiospores of Melanogaster ambiguus, or Rhizopogon subareolatus, or with ascospores of Tuber maculatum. Pinus pinaster seedlings were inoculated with basidiospores of Melanogaster ambiguus, Rhizopogon roseolus or Scleroderma citrinum. The spore concentrations were 102–107 spores per seedling for Melanogaster ambiguus (in Pseu dotsuga menziesii) and Rhizopogon subareolatus, 103–107 for Melanogaster ambiguus (in Pinus pinaster), Rhizopogon roseolus, and Scleroderma citrinum, and 102–104 for Tuber maculatum. Melanogaster ambiguus colonized more short roots in a larger proportion of plants at 107 spores per seedling than at any other rate. The highest colonization by Rhizopogon subareolatus was obtained at 104 spores per seedling and higher, and all inoculated plants became infected at 106 spores per seedling and higher. Tuber maculatum colonized a high percentage of short roots at all rates tested; the proportion of infected plants was over 80% at 103–104 spores per plant, decreasing to 50% at 102 spores per plant. Rhizopogon roseolus colonized the highest number of short roots on nearly all the inoculated plants when applied at 105 spores per seedling and higher. Scleroderma citrinum colonized a high percentage of short roots on all inoculated plants when applied at 105 spores per seedling and higher. The abundance of sporocarps of Melanogaster ambiguus, Rhizopogon subareolatus, R hizopogon roseolus and Scleroderma citrinum and their colonization ability at relatively low rates allows these spores to be used as ectomycorrhizal inocula on a large scale. Effect of ectomycorrhizal fungi on chestnut ink disease   总被引:4,自引:0,他引:4
Seedlings of Castanea sativa were inoculated at transplanting time with four ectomycorrhizal (ECM) fungi, Laccaria laccata, Hebeloma crustuliniforme, H. sinapizans and Paxillus involutus. At the end of the first vegetative season, 7 months after sowing, half of the mycorrhizal and nonmycorrhizal seedlings were challenged with a zoospore suspension of Phytophthora cambivora and the other half with P. cinnamomi. Five months later, mycorrhizal plants infected with P. cambivora or P. cinnamomi showed no sign of pathogen infection. The ECM fungi increased plant biomass also in the presence of the pathogen. Mycorrhizal seedlings inoculated with the pathogens showed greater shoot and root development than nonmycorrhizal chestnut plants. All the fungi tested reduced the negative effect of the ink disease pathogens on the plant host in vivo. The mechanisms by which the ECM fungi protect chestnut seedlings are discussed. Seedlings of three Fagaceae species planted on acidic, infertile colluvial soil showed accelerated growth when inoculated with ectomycorrhizal fungi.

Abstract

We conducted a study with seedlings of Fagaceae species inoculated with ectomycorrhizal fungi to estimate their utility for growth in acidic soil conditions. We selected Quercus glauca, Quercus salicina and Castanopsis cuspidata as typical evergreen, broad-leaved, woody species of southwestern Japan. Seedlings were inoculated with Astraeus hygrometricus or Scleroderma citrinum, and planted in acidic, infertile colluvial soil collected from an abandoned site. Six months after planting, seedlings of the three species inoculated with A. hygrometricus were growing well, especially, Q. salicina. The growth of seedlings inoculated with S. citrinum was inferior to seedlings inoculated with A. hygrometricus. In contrast, seedlings without ectomycorrhizal fungi did not grow well. Differences in growth among the three types of seedlings were related to differences in the levels of nutrient acquisition. We concluded that Fagaceae seedlings inoculated with A. hygrometricus were best suited for acidic, infertile environments.  相似文献

This paper focuses on growth pattern of seedlings from tree-ring proxies and their performance at different altitudes under high disturbance regime. The disturbance is predominately anthropogenic. Four conifer species found during investigations, namely, Pinus wallichiana, Pinus roxburghii, Cedrus deodara and Abies pindrow. The parameters chosen for assessment of regeneration potential were age and growth rate. In addition, edaphic and topographic characteristics of thirty disturbed forest stands were also determined. The stands were deterministically selected because these were deteriorating with respect to tree population, biodiversity and degradation of soil. Seedling density varied in the order: P. wallichianaPinus roxburghii > C. deodara > Abies pindrow while the growth rate was exhibited in the order Pinus roxburghii > P. wallichiana > Abies pindrow > C. deodara. These findings suggest that Pinus roxburghii is the most suitable species (sub tropical species) at lower elevations (~1600–1900 a.s.l.) with regard to its regenerative ability and consequently potentially useful for the conservation of these forests. P. wallichiana showed highly significant (P < 0.01) correlations with PC1 (primarily controlled by total dissolved salts and water holding capacity) only whereas Abies pindrow was highly correlated (P < 0.001 and P < 0.01) with PC1 and PC2 respectively (function of oxidative-reductive potential and water holding capacity) while C. deodara and Pinus roxburghii did not show correlation with any of the PCA components. In the light of current findings, Pinus roxburghii was found to be effectively adapted to the environment of the study area. Growth rate of species steadily decreased in response to unfavorable conditions. The other pine species i.e. P. wallichiana (moist temperate species) also showed some degree of adaptability under the influence of predefined environmental attributes. Hence pines are most likely the species that can be able to retain forest cover in future and are the best choice for restoration of these forests. However, the other conifers i.e. C. deodara and Abies pindrow are also able to withstand the physiographic characteristics and the extent of perturbation and persisting possibly beyond their tolerance limits but they need to be preserved and retained in these forests. Conifers are still giving positive response with respect to growth rate and their adaptive compatibility to such an extent that this forest can recover if management and conservation strategies are implemented. Therefore, it is strongly recommended that seedlings and saplings should be conserved to assure future forest cover.  相似文献

The effectiveness of ectomycorrhizal inoculation at the tree nursery seedling production stage on growth and survival was examined in jack pine (Pinus banksiana) and white spruce (Picea glauca) planted in oil sands reclamation sites. The seedlings were inoculated with Hebeloma crustuliniforme strain # UAMH 5247, Suillus tomentosus strain # UAMH 6252, and Laccaria bicolor strain # UAMH 8232, as individual pure cultures and in combinations. These treatments were demonstrated to improve salinity resistance and water uptake in conifer seedlings. The field responses of seedlings to ectomycorrhizal inoculation varied between plant species, inoculation treatments, and measured parameters. Seedling inoculation resulted in higher ectomycorrhizal colonization rates compared with non-inoculated control, which had also a relatively small proportion of roots colonized by the nursery contaminant fungi identified as Amphinema byssoides and Thelephora americana. Seedling inoculation had overall a greater effect on relative height growth rates, dry biomass, and stem volumes in jack pine compared with white spruce. However, when examined after two growing seasons, inoculated white spruce seedlings showed up to 75 % higher survival rates than non-inoculated controls. The persistence of inoculated fungi in roots of planted seedlings was examined at the end of the second growing season. Although the inoculation with H. crustuliniforme triggered growth responses, the fungus was not found in the roots of seedlings at the end of the second growing season suggesting a possibility that the observed growth-promoting effect of H. crustuliniforme may be transient. The results suggest that the inoculation of conifer seedlings with ectomycorrhizal fungi could potentially be carried out on a large scale in tree nurseries to benefit postplanting performance in oil sands reclamation sites. However, these practices should take into consideration the differences in responses between the different plant species and fungal strains.  相似文献

Few studies have focused on analyzing the effect of native inoculated ectomycorrhizal (ECM) fungal strains on seedlings under field conditions in temperate forests. However, it is crucial to verify that the positive effects of ECM under nursery conditions also occur in the field, favoring their performance. We evaluated the short-term effect of ECM on three-year-old seedlings of Pinus hartwegii and Abies religiosa in central Mexico by subjecting them to four treatments: inoculation with Inocybe splendens, inoculation with Suillus brevipes (both native strains), inoculation with forest soil, and non-inoculated plants. Percentage of ECM colonization, plant growth (shoot height and stem diameter), and physiological (osmotic potential, stomatal conductance, CO2 assimilation and water use efficiency) responses were evaluated. We found that these two ECM species were partial (P. hartwegii) or totally (A. religiosa) replaced after one and a half years in the field. P. hartwegii seedlings increased their water use efficiency during the dry season, but in A. religiosa seedlings, a clear strategy for avoiding water stress was not detected. This ECM replacement had a negative effect on the physiological performance Of A. religiosa. Our results emphasize the importance of selecting compatible fungal-host species combinations for nursery inoculation and of using sources of inoculum adapted to the environmental conditions of the transplant site, ensuring root colonization prior to field transplanting to minimize seedling mortality due to water stress.  相似文献

Influences of soil P fertilization on temporal changes in ergosterol content and ornithine decarboxylase (E.C. 4.1.1.17, ODC) activity were monitored in rhizosphere soil, non-rhizosphere soil and Pinus contorta roots ectomycorrhizal with Hebeloma crustuliniforme grown in a loamy sand. With addition of mycorrhizal inoculum to loamy sand, ODC activity mg-1 root increased between 10% and 2 fold within 21 weeks of pianting. Inoculation also decreased root mass per seedling. Inoculation increased mycelia mass per root mass by up to 2 fold but no differences were observed for total seedling mass until 35 weeks. Intramatrical mycelia were detrimental to early plant growth, but inoculated seedlings had 1.7 times more root mass and 1.3 times more shoot mass at 35 weeks. Rhizosphere soil contained up to 5 times more mycelia and up to 6 times greater ODC activity than non-rhizosphere soil. Inoculation increased rhizophere metabolic activity and intramatrical mycelia mass. Their sensitivity to fungal inoculation, P fertilization and temporal trends may make the methods useful in studies of rhizosphere ecology and root-microbe relationships.  相似文献

Many trees species form symbiotic associations with ectomycorrhizal (ECM) fungi, which improve nutrient and water acquisition of their host. Until now it is unclear whether the species richness of ECM fungi is beneficial for tree seedling performance, be it during moist conditions or drought. We performed a pot experiment using Pinus sylvestris seedlings inoculated with four selected ECM fungi (Cenococcum geophilum, Paxillus involutus, Rhizopogon roseolus and Suillus granulatus) to investigate (i) whether these four ECM fungi, in monoculture or in species mixtures, affect growth of P. sylvestris seedlings, and (ii) whether this effect can be attributed to species number per se or to species identity. Two different watering regimes (moist vs. dry) were applied to examine the context-dependency of the results. Additionally, we assessed the activity of eight extracellular enzymes in the root tips. Shoot growth was enhanced in the presence of S. granulatus, but not by any other ECM fungal species. The positive effect of S. granulatus on shoot growth was more pronounced under moist (threefold increase) than under dry conditions (twofold increase), indicating that the investigated ECM fungi did not provide additional support during drought stress. The activity of secreted extracellular enzymes was higher in S. granulatus than in any other species. In conclusion, our findings suggest that ECM fungal species composition may affect seedling performance in terms of aboveground biomass.  相似文献

American elm (Ulmus americana) seedlings were either non-inoculated or inoculated with Hebeloma crustuliniforme, Laccaria bicolor and a mixture of the two fungi to study the effects of ectomycorrhizal associations on seedling responses to soil compaction and salinity. The seedlings were grown in the greenhouse in pots containing non-compacted (0.4 g cm?3 bulk density) and compacted (0.6 g cm?3 bulk density) soil and subjected to 60 mM NaCl or 0 mM NaCl (control) treatments for 3 weeks. All three fungal inocula had similar effects on the responses of elm seedlings to soil compaction and salt treatment. In non-compacted soil, ectomycorrhizal fungi reduced plant dry weights, root hydraulic conductance, but did not affect leaf hydraulic conductance and net photosynthesis. When treated with 60 mM NaCl, ectomycorrhizal seedlings had several-fold lower leaf concentrations of Na+ compared with the non-inoculated plants. Soil compaction reduced Na+ leaf concentrations in non-ectomycorrhizal plants and decreased dry weights, gas exchange and root hydraulic conductance. However, in ectomycorrhizal plants, soil compaction had little effect on the leaf Na+ concentrations and on other measured growth and physiological parameters. Our results demonstrated that ECM associations could be highly beneficial to plants growing in sites with compacted soil such as urban areas.  相似文献

Although increasing concentrations of atmospheric CO2 are predicted to have substantial impacts on plant growth and functioning of ecosystems, there is insufficient understanding of the responses of belowground processes to such increases. We investigated the effects of different dark septate root endophytic (DSE) fungi on growth and nutrient acquisition by Pinus sylvestris seedlings under conditions of N limitation and at ambient and elevated CO2 (350 or 700 μ1 CO2 l?1). Each seedling was inoculated with one of the following species: Phialocephala fortinii (two strains), Cadophora finlandica, Chloridium paucisporum, Scytalidium vaccinii, Meliniomyces variabilis and M. vraolstadiae. The trial lasted 125 days. During the final 27 days, the seedlings were labeled with 14CO2 and 15NH 4 + . We measured extraradical hyphal length, internal colonization, plant biomass, 14C allocation, and plant N and 15N content. Under elevated CO2, the biomass of seedlings inoculated with DSE fungi was on average 17% higher than in control seedlings. Simultaneously, below-ground respiration doubled or trebled, and as a consequence carbon use efficiency by the DSE fungi significantly decreased. Shoot N concentration decreased on average by 57% under elevated CO2 and was lowest in seedlings inoculated with S. vaccinii. Carbon gain by the seedlings despite reduced shoot N concentration indicates that DSE fungi increase plant nutrient use efficiency and are therefore more beneficial to the plant under elevated CO2.  相似文献

Pinus sylvestris seedlings, grown in a vertical petri dish system, were inoculated with five different mycorrhizal fungi. Half of the root system in the petri dish was subjected to periodic flooding, and mycorrhizal colonization was studied. Thelephora terrestris, Laccaria laccata, and Hebeloma crustuliniforme were not sensitive to flooding, whereas Suillus flavidus and S. bovinus were highly sensitive. The latter failed to colonize the root even when flooded for only 2 min per day four times a week.  相似文献

The application of ectomycorrhizal (ECM) fungi on forest nursery production is regarded as part of good management practice. However, before employing large scale inoculations in a nursery the interaction between ECM symbionts, growth substrate and fertilisation input should be studied to select the most suitable nursery practices for promoting plant growth and ECM colonisation. In this study, seedlings of Quercus ilex were inoculated with Paxillus involutus, Hebeloma mesophaeum or Cenococcum geophilum and grown in three different substrates commonly used in forest nurseries: peat-based compost, forest soil or composted pine bark. The effect of various fertilisation regimes was also studied. The choice of substrate had a significant effect on plant growth and ECM colonisation. The most appropriate combination of substrate and ECM fungus for Q. ilex growth and nutrition was peat and H. mesophaeum. Plants grown on a peat-based compost and inoculated with H. mesophaeum had a significantly greater biomass and leaf phosphorus concentration without fertilisation. Composted pine bark was found not to be suitable for growth or for mycorrhization. If the appropriate growth substrate is selected, it is possible to replace the use of chemical fertilisers by inoculation with selected ECM fungi. This results in a significant increase in plant development, and thus ECM fungi can be recommended as a more environmental friendly biotechnological approach to plant management in the nursery.  相似文献

White spruce [Picea glauca (Moench) Voss] seedlings were inoculated with Hebeloma crustuliniforme and treated with 25 mM NaCl to examine the effects of salinized soil and mycorrhizae on root hydraulic conductance and growth. Mycorrhizal seedlings had significantly greater shoot and root dry weights, number of lateral branches and chlorophyll content than non-mycorrhizal seedlings. Salt treatment reduced seedling growth in both non-mycorrhizal and mycorrhizal seedlings. However, needles of salt-treated mycorrhizal seedlings had several-fold higher needle chlorophyll content than that in non-mycorrhizal seedlings treated with salt. Mycorrhizae increased N and P concentrations in seedlings. Na levels in shoots and roots of salt-treated mycorrhizal seedlings were significantly lower and root hydraulic conductance was several-fold higher than in non-mycorrhizal seedlings. A reduction of about 50% in root hydraulic conductance of mycorrhizal seedlings was observed after removal of the fungal hyphal sheath. Transpiration and root respiration rates were reduced by salt treatments in both groups of seedlings compared with the controls, however, both transpiration and respiration rates of salt-treated mycorrhizal seedlings were as high as those in the non-mycorrhizal seedlings that had not been subjected to salt treatment. The reduction of shoot Na uptake while increasing N and P absorption and maintaining high transpiration rates and root hydraulic conductance may be important resistance mechanisms in ectomycorrhizal plants growing in salinized soil.

As the main source of inocula, ectomycorrhizal (ECM) fungal propagules are critical for root colonization and seedling survival in deforested areas. It is essential to know factors that may affect the diversity and composition of ECM fungal community on roots of seedlings planted in deforest areas during reforestation. We quantitatively evaluated the effect of host plant and soil origin on ECM fungal propagule community structure established on roots of Castanopsis fargesii, Lithocarpus harlandii, Pinus armandii, and Pinus massoniana growing in soils from local natural forests and from sites deforested by clear-cut logging in the 1950s and 1960s. ECM root tips were sampled in April, July, and October of 2006, and ECM fungal communities were determined using ECM root morphotyping, internal transcribed spacer (ITS)-RFLP, and ITS sequencing. A total of 36 ECM fungal species were observed in our study, and species richness varied with host species and soil origin. Decreased colonization rates were found in all host species except for L. harlandii, and reduced species richness was found in all host species except for P. armandii in soil from the deforested site, which implied the great changes in ECM fungal community composition. Our results showed that 33.3% variance in ECM fungal community composition could be explained by host plant species and 4.6% by soil origin. Results of indicator species analysis demonstrated that 14 out of 19 common ECM fungal species showed significant preference to host plant species, suggesting that the host preference of ECM fungi was one of the most important mechanisms in structuring ECM fungal community. Accordingly, the host plant species should be taken into account in the reforestation of deforested areas due to the strong and commonly existed host preference of ECM fungi.  相似文献

In common with many plants native to low P soils, jarrah (Eucalyptus marginata) develops toxicity symptoms upon exposure to elevated phosphorus (P). Jarrah plants can establish arbuscular mycorrhizal (AM) and ectomycorrhizal (ECM) associations, along with a non-colonizing symbiosis described recently. AM colonization is known to influence the pattern of expression of genes required for P uptake of host plants and our aim was to investigate this phenomenon in relation to P sensitivity. Therefore, we examined the effect on hosts of the presence of AM and ECM fungi in combination with toxic pulses of P and assessed possible correlations between the induced tolerance and the shoot P concentration. The P transport dynamics of AM (Rhizophagus irregularis and Scutellospora calospora), ECM (Scleroderma sp.), non-colonizing symbiosis (Austroboletus occidentalis), dual mycorrhizal (R. irregularis and Scleroderma sp.), and non-mycorrhizal (NM) seedlings were monitored following two pulses of P. The ECM and A. occidentalis associations significantly enhanced the shoot P content of jarrah plants growing under P-deficient conditions. In addition, S. calospora, A. occidentalis, and Scleroderma sp. all stimulated plant growth significantly. All inoculated plants had significantly lower phytotoxicity symptoms compared to NM controls 7 days after addition of an elevated P dose (30 mg P kg?1 soil). Following exposure to toxicity-inducing levels of P, the shoot P concentration was significantly lower in R. irregularis-inoculated and dually inoculated plants compared to NM controls. Although all inoculated plants had reduced toxicity symptoms and there was a positive linear relationship between rank and shoot P concentration, the protective effect was not necessarily explained by the type of fungal association or the extent of mycorrhizal colonization.  相似文献
