Influences of inbreeding and genetics on telomere length in mice   总被引:2,自引:0,他引:2  
We measured telomere lengths of blood leukocytes in several inbred and outbred mammalian species, using a telomere-specific fluorescent probe and flow cytometry. Humans, non-human primates, and three outbred populations of Peromyscus mice (Peromyscus leucopus, Peromyscus maniculatus, and Peromyscus polionotus) have short telomeres. Two common strains of laboratory mice, C57BL/6J and DBA/2J, have telomeres several times longer than most other mammals surveyed. Moreover, the two inbred laboratory mouse strains display significantly different telomere lengths, suggesting the existence of strain-specific genetic determinants. To further examine the effects of inbreeding, we studied three Peromyscus leucopus inbred lines (GS109, GS16A1, and GS16B), all derived from the outbred P. leucopus stock. Telomeres of all three inbred lines are significantly lengthened relative to outbred P. leucopus, and the three lines display strain-specific significantly different telomere lengths, much like the C57BL/6J and DBA/2J strains of M. musculus. To further characterize the genetic inheritance of telomere length, we carried out several crosses to obtain hybrid F1 mice between parental strains displaying the phenotype of long and short telomeres. In all F1 mice assayed, peripheral blood leukocyte telomere length was intermediate to that of the parents. Additionally, we generated F2 mice from a cross of the (P. leucopus outbred × GS16B)F1. Based on the distribution of telomere length in the F2 population, we determined that more than five loci contribute to telomere length regulation in Peromyscus. We concluded that inbreeding, through unknown mechanisms, results in the elongation of telomeres, and that telomere length for a given species and/or sub-strain is genetically determined by multiple segregating loci.  相似文献

During 1980-1982, white-footed mice (Peromyscus leucopus) were captured in Newtown, Connecticut, an area where Rickettsia rickettsii, the etiologic agent of Rocky Mountain spotted fever, is thought to be enzootic. An indirect microimmunofluorescence test identified specific antibodies to this organism in 16 of 237 (7%) sera: titration end points for 14 samples were relatively high (1:128-1:2048). Antibodies were detected in mice during 1980 and 1981 with monthly prevalences varying from 8 to 22%. These results suggest that P. leucopus may be involved in the ecology of R. rickettsii and that these rodents can be included along with other mammals to monitor spotted fever rickettsial infections in nature.  相似文献

Two groups of adult Mongolian gerbils (Meriones unguiculatus) of mixed ages and sex were used to study the effect of bilateral Harderian gland adenectomy on development of nasal dermatitis. One group of gerbils underwent bilateral Harderian gland adenectomies, while the other group underwent sham surgeries, leaving the Harderian gland intact. All animals in both groups were fitted with Elizabethian collars to prevent self-grooming, allowing a buildup of nasolacrimal or Harderian gland secretions near the medial canthus of the eye and at the external nares. Twenty-six of 27 animals with intact Harderian glands developed nasal and facial lesions within 20 days. None of the 27 Harderian gland adenectomized animals developed nasal or facial lesions. Apparently, accumulation of Harderian gland secretions is involved in the pathogenesis of nasal dermatitis in the Mongolian gerbil.  相似文献

Effects of short-day photoperiod, pinealectomy, and melatonin on sexual maturation were tested in Peromyscus leucopus from either Connecticut (CT) or Georgia (GA). Laboratory reared-stocks from CT and GA were exposed to short daylength (photoperiod) from birth or 25 days of age. At 12 wk of age, delay in sexual maturation was indicated in most CT mice by decreased testis length, combined testes weight, and seminal vesicle weight. Conversely, GA animals did not delay sexual maturation when exposed to short-day photoperiod from either birth or 25 days of age. These results indicate that responses to short daylengths differ for juvenile CT and GA populations. In a second experiment, pinealectomized or sham-operated CT males were exposed to short-day (9L:15D) or long-day (16L:8D) photoperiod from birth. Pinealectomy blocked the effect of short daylength on reproduction. Therefore, the pineal must be involved in the delay of sexual maturation observed for short-day CT mice. The effects of melatonin, a pineal gland hormone, were tested with chronic s.c. implants or daily injections. In CT mice given either melatonin implants or afternoon injections, sexual maturation was delayed. GA mice were insensitive to all melatonin treatments. Further, no differences in circadian organization (phase angle, duration of activity, period under constant dark) between GA and CT animals were apparent. Collectively, these studies indicate that melatonin is involved in the mechanism responsible for delay of sexual maturation in CT mice. Short-day insensitivity of GA Peromyscus leucopus probably results from a deficiency in the melatonin effector pathway and is not due to a disruption of circadian organization.  相似文献

1. We surveyed the primary humoral immune responsiveness of six small mammal species (Peromyscus leucopus, Microtus pinetorum, Perognathus hispidus, Neotoma floridana, Onychomys leucogaster, Mus musculus) collected from wild populations in central Oklahoma using sheep red blood cells (SRBC) as the immunogen and a splenic plaque-forming cell (PFC) assay. 2. Individuals within each wild species examined produced antibodies to a single intraperitoneal injection of SRBC, however, considerable interspecific and intraspecific variation in responsiveness was indicated. 3. Overall, primary immune responsiveness varied from 0 to 5013 PFC/10(6) cells. Spleen weights, total splenic nucleated cell yields, PFC/spleen, PFC/mg spleen, and PFC/10(6) cells were significantly different among species. 4. Mean cell yield in M. pinetorum was greater than in P. leucopus and CD-1 laboratory mice (included as positive controls). Number of PFC/10(6) cells was greater in CD-1 laboratory mice than P. hispidus and P. leucopus. The coefficient of variation for PFC/10(6) cells in CD-1 laboratory mice was 38% compared to 109, 129, and 56% for M. pinetorum, P. hispidus, and P. leucopus, respectively. 5. Interspecific and intraspecific differences among wild species may be a reflection of disparate life histories and other environmental selection pressures.  相似文献

Summary A sexual dimorphism of the hamster Harderian gland at the ultrastructural level has been reported. The effect of testosterone on the fine structure of the gland from castrated male golden hamsters is reported here. Harderian glands from the following three groups of animals were examined at regular intervals up to 60 days after castration: (1) castrated; (2) castratedsham-injected, receiving 0.1 ml sesame oil per day; (3) castrated-testosterone injected, receiving 2mg testosterone propionate in 0.1 ml sesame oil per day. In groups 1 and 2, clusters of cylindrical tubules, typical of the male gland, decreased in number and disappeared almost completely 2 weeks after castration. Membranous structures, typical of the female gland, prevailed in these two groups throughout the remaining period of experiment. On the other hand, these changes were prevented in the group of castrated animals maintained on testosterone propionate. It is concluded that castration modified the ultrastructure of the male hamster Harderian gland toward the female type and that daily administration of testosterone propionate prevented this change.  相似文献

One hundred ten white-footed mice (Peromyscus leucopus) were captured on horse farms in south-central Maryland, examined for ticks, and tested for specific antibodies to Ehrlichia risticii, the causative agent of Potomac horse fever. Peromyscus leucopus were consistently infested with immature American dog ticks (Dermacentor variabilis), with monthly prevalences as high as 80%. Sera from all 97 P. leucopus tested for antibodies to E. risticii were negative. This indicates that P. leucopus is not a reservoir of E. risticii, and suggests that immature D. variabilis do not acquire E. risticii in feeding upon white-footed mice.  相似文献

1. Secretion pattern of Harderian gland of neonatal rats maintained under (a) diurnal lighting conditions; (b) continuous light or (c) continuous darkness was studied at light microscopy level. 2. All animals were placed in especially designed cages at 13:00 hr on day 1 and studied on day 7 at 13:00 and 23:00 hr, respectively. 3. Acini with intraluminal secretion were counted in glands from each animal and the results were separately grouped for male and female animals. 4. A diurnal rhythm in secretion pattern of rat Harderian gland in neonatal period was demonstrated. 5. A statistically significant difference was observed in the gland secretion pattern between males and females at both, 13:00 and 23:00 hr when the animals were kept under diurnal lighting conditions. 6. Under continuous light or continuous darkness, the diurnal rhythm in secretion pattern was lost and no significant differences were seen when data from males were compared to those from female neonates. 7. Results are discussed in terms of the possible function of Harderian gland as element of an extraretinal photoreceptor system involved in the regulation of pineal function in neonatal rats.  相似文献

Immunoreactive melatonin levels were measured in the retina and Harderian gland of adult male rats throughout a 24 hour period. The animals were maintained under a light:dark cycle of 14:10 (lights on at 0600h). In intact animals, immunoreactive melatonin values in both organs exhibited a 24h rhythm with peak levels being measured at 0800h, 2 hours after lights on. Pinealectomy significantly increased peak levels at 0800h in both the retina and the Harderian gland. Gonadectomy abolished the peak retinal melatonin levels at 0800h. Likewise, continual light exposure for 1 week depressed the melatonin peak in the retina but not in the Harderian gland.  相似文献

The effects of age and food restriction on the porphyrin concentration in Harderian glands were studied in male Fisher 344 rats. Harderian gland porphyrin concentrations increased with age; this was statistically significant in 20 month old animals compared with 3 month old animals. Food restriction (by 40%) prevented the age-associated rise in porphyrins; thus, in 20 month old food restricted rats had porphyrin concentrations similar to those found in young animals. In a second experiment, we correlated the age-associated rise in Harderian gland porphyrin concentrations with an increase in mRNA levels for 5-aminolevulinate synthase (ALV-S). Both the porphyrin concentration and ALV-S mRNA rose at 12 and 18 months of age, but decreased by 24 months of age. It is concluded that, a) porphyrin biosynthesis in the Harderian glands increases up to 20 months of age but decreases in rats that are 24 months old, and b) food restriction prevents the porphyrin rise associated with age in the Harderian gland of male Fisher 344 rats.  相似文献

Trypanosoma peromysci Watson, 1912 (Sarcomastigophora: Kinetoplastida), is described from a new host and locality. One of 20 (5.0%) Peromyscus leucopus collected from Pottawatomie and Riley counties in Kansas was found to harbor the parasite. Morphometric and statistical analysis confirmed the trypanosome to be indistinguishable from T. peromysci, the only difference being a greater mean flagellar length than reported previously. This is the first reported occurrence of T. peromysci in the white-footed mouse (Peromyscus leucopus noveboracensis Fischer, 1829) and also the first record of its occurrence in Kansas.  相似文献

Some of the studies on late effects of neutron and gamma radiation previously carried out with the C57BL6 X BALB/c F1 hybrids of Mus musculus have been repeated with the white-footed mouse, Peromyscus leucopus, a cricetid rodent of a different subfamily, with differing physiological characteristics and a different spectrum of pathologies. Among the more important findings were the following: For both species, the life shortening per rad at low doses of either radiation was the same percentage of the life span. The limiting values of the relative biological effectiveness for life shortening from all causes of death were about the same for the two species, ranging from 8 to 16, depending on the method of calculation. Fractionated neutron exposures failed to produce significant life shortening in Peromyscus over that observed at single doses. Tumor-related deaths accounted for at least 70 to 75% of the radiation-specific excess mortality in Peromyscus.  相似文献

The complete M segment sequences of hantaviruses amplified from tissues of a patient with hantavirus pulmonary syndrome in the northeastern United States and from white-footed mice, Peromyscus leucopus, from New York were 99% identical and differed from those of Four Corners virus by 23%. The serum of this patient failed to recognize a conserved, immunodominant epitope of the Four Corners virus G1 glycoprotein. Collectively, these findings indicate that P. leucopus harbors a genetically and antigenically distinct hantavirus that causes hantavirus pulmonary syndrome.  相似文献

Summary Manipulation of circulating levels of thyroid hormones modifies Harderian gland structure and porphyrin concentrations in male and female golden hamsters. Specifically, thyroxine (T4) and triiodothyronine (T3) induce the morphological conversion of the Harderian glands of females to approximate those of the male. Further, porphyrin concentrations are markedly decreased by this treatment. This effect occurs in ovariectomized animals as well, indicating that the gonads are not involved. Suppression of thyroid function by potassium perchlorate (KClO4) drastically reduces Harderian gland weight in both males and females. However, KClO4 decreases porphyrin levels in the Harderian glands of females and increases it in the male. Concurrently, KClO4 also induces a morphological conversion of the Harderian glands of males to the female type. This effect is evident in photoperiods of either 14:10 (h) or 8:16 (h).  相似文献

Harderian gland (HG) weight and lysosomal enzyme activity were evaluated after 21-day-old female rats were singly caged in a long (LP; 14:10 LD) or short (SP; 8:16 LD) photoperiod and fed on one of two dietary regimens (fed ad libitum or 50% underfed) for 50 days; an additional fed and an underfed group of animals in LP were injected every afternoon with 100 micrograms melatonin. Absolute HG weights were significantly lower in all underfed groups compared to their respective fed controls or to the LP fed control group. Absolute HG weights of underfed rats in SP were significantly lower than the underfed rats in LP. Relative HG weights (mg/100 g body wt) were significantly higher in the underfed saline or melatonin-treated groups compared to their respective fed controls; however, HG of the underfed SP group were not different from SP-fed controls. No significant differences in HG acid phosphatase, hexosaminidase, and beta-glucuronidase activities were observed in any of the treatment groups maintained in LP. Acid phosphatase, hexosaminidase, and beta-glucuronidase activities were significantly elevated in HG of underfed animals maintained in SP compared to their respective fed controls or to the LP-underfed group. Both the underfed control and the underfed-melatonin treated groups had lower pineal protein values than their respective fed groups; underfed animals in 8:16 LD had similar pineal protein values compared to those of the fed control group in SP. Significant effects of photoperiod and underfeeding with no interaction between these variables were observed on pineal acid phosphatase. The fed group maintained in 8:16 LD had significantly higher acid phosphatase activity than the fed group kept in 14:10 LD. In conclusion, underfeeding resulted in severely reduced body weights and absolute Harderian gland weights. Increased activity in certain lysosomal enzymes occurred in both the pineal and Harderian gland and in some instances this was dependent upon the light cycle and dietary regimen to which the animals were exposed.  相似文献

Of 198 deermice (Peromyscus spp) collected from various localities in the southwestern United States and northern Mexico, 106 (54%) had eimerian oocysts in their feces when examined. These included 50 of 106 (47%) Peromyscus truei, 34 of 54 (63%) Peromyscus maniculatus, 4 of 17 (24%) Peromyscus leucopus, and 18 of 21 (86%) Peromyscus eremicus. The following Eimeria were identified from infected mice: Eimeria arizonensis and Eimeria langebarteli from P. truei; E. arizonensis, Eimeria peromysci, and Eimeria delicata from P. maniculatus; E. arizonensis and Eimeria lachrymalis n. sp. from P. eremicus; and E. langebarteli from P. leucopus. Of the 106 Peromyscus found positive for Eimeria, 97 (91.5%) harbored only a single eimerian species at the time of examination. Sporulated oocysts of E. lachrymalis n. sp. were ellipsoid, 27-35 X 17-21 (30.8 +/- 1.7 X 19.1-0.9) micron, possessed a smooth wall and one polar granule, but lacked a micropyle and an oocyst residuum. Sporocysts were teardrop-shaped, 9-13 X 6-10 (10.9 +/- 0.9 X 7.9 +/- 0.5) micron, and had a Stieda body and sporocyst residuum, but no substieda body. Prepatent periods in experimental infections were 3-6 days after inoculation (DAI) for E. arizonensis (hosts: P. eremicus, P. maniculatus, P. truei); 4-5 DAI for E. peromysci (host: P. maniculatus); 6-9 DAI for E. langebarteli (hosts: P. truei, P. leucopus); and 8-10 DAI for E. lachrymalis (host: P. eremicus). Patency in these infections lasted 6-11 days for E. arizonensis, 5-10 days for E. peromysci, 14-40+ days for E. langebarteli, and 19-50+ days for E. lachrymalis. Eimeria lachrymalis appears to produce occult infections in P. eremicus that can be reactivated upon inoculation of the host with E. arizonensis.  相似文献

We have derived and mated separate strains of transgenic mice that carry either the v-Ha-ras or the c-myc gene driven by the mouse mammary tumor virus (MMTV) promoter/enhancer. Mice carrying the MMTV/v-Ha-ras transgene manifest two distinct disturbances of cell growth. The first, a benign hyperplasia of the Harderian lacrimal gland, is diffuse, involves the entire gland, and likely requires only the abnormal action of the v-Ha-ras gene. The second involves the focal development of malignancies of mammary, salivary, and lymphoid tissue and likely requires additional somatic events. When the MMTV/v-Ha-ras and MMTV/c-myc strains are crossed to yield hybrid mice, their joint action results in a dramatic and synergistic acceleration of tumor formation. Since these tumors arise stochastically and are apparently monoclonal in origin, additional somatic events appear necessary for their full malignant progression, even in the presence of activated v-Ha-ras and c-myc transgenes.  相似文献

Cross-transmission experiments were performed using oocysts of an Eimeria arizonensis-like coccidian from Peromyscus leucopus and Peromyscus truei, an E. arizonensis-like coccidian from Reithrodontomys fulvescens, Eimeria baiomysis and Eimeria taylori from Baiomys taylori, Eimeria albigulae from Neotoma albigula, and Eimeria onychomysis from Onychomys spp., between representatives of the above host genera. The E. arizonensis-like coccidian from R. fulvescens infected Reithrodontomys megalotis, Reithrodontomys montanus, and Peromyscus leucopus. Oocysts of E. arizonensis from P. leucopus could be transmitted to both P. leucopus and R. megalotus. Oocysts of E. baiomysis and E. taylori infected only B. taylori. Oocysts of E. arizonensis from P. truei infected P. truei but not Neotoma mexicana or Onychomys leucogaster. Oocysts of E. albigulae from N. albigula were infective for N. mexicana but not for P. truei or O. leucogaster. Oocysts of E. onychomysis from Onychomys spp. infected O. leucogaster but not N. mexicana or P. truei. These results demonstrate that Peromyscus and Reithrodontomys, genera known to be related very closely evolutionarily, are capable of sharing E. arizonensis, whereas morphologically similar coccidians (E. albigulae, E. baiomysis, and E. onychomysis) from more distantly related hosts, are probably distinct and more stenoxenous. This also is the first report of coccidians infecting species of Reithrodontomys.  相似文献

BACKGROUND: Polymorphous low grade adenocarcinoma (PLGA) is a histologically low grade tumor of minor salivary gland origin. It is important to differentiate PLGA from other salivary gland tumors with myoepithelial differentiation, such as pleomorphic adenoma, adenoid cystic carcinoma and epithelial myoepithelial carcinoma. Here we report 2 cases of PLGA originating in the palate and describe the cytomorphologic and immunocytochemical features. CASES: The patients were a 55-year-old woman and a 63-year-old man. Both presented with a mass in the palate. Clinically the mass appeared malignant, and resection was performed. Cytologically the tumor cells were composed of sheet clusters, pseudopapillary epithelial clusters, naked cells and stromal components. Immunocytochemically the tumor cells showed strong expression of carcinoembryonic antigen (CEA) and vimentin. CONCLUSION: PLGA may be difficult to distinguish from other salivary gland tumors with myoepithelial differentiation. However, the cytopathologist should be aware of the distinctive cytomorphologic features of PLGA, demonstrating immunopositivity to CEA and vimentin.  相似文献
